Abstract:Lipid droplets (LDs) are ubiquitous intracellular structures whose formation, growth, and maintenance are highly regulated (Wang et al., 2013; Ranall et al., 2011; Goodman, 2009). Lipid metabolism and droplet dynamics are of considerable interest to agriculture, biofuel production, viral pathology, nutrition, and cancer biology (Walther and Farese, 2009; Liu et al., 2010). Accumulation of fatty acids and neutral lipids in nonadipose tissues is cytotoxic (Kourtidis et al., 2009). BODIPY 493/503 (4,4-Difluoro-1,3,5,7,8-Pentamethyl-4-Bora-3a,4a-Diaza-s-Indacene) is the standard dye to study LDs within adipocytes. BODIPY 493/503 contains a nonpolar structure that, upon binding to neutral lipid, emits a green fluorescence signal with a narrow wavelength range, making it an ideal fluorophore for multi-labeling experiments. The hydrophobic nature of the dye molecules promotes rapid entry into the nonpolar environment of LDs (Listenberge and Brown, 2007). Gocze and Freeman showed that the lipid fluorescent variability is significantly lower when using BODIPY493/503 compared to Nile Red, suggesting that it may be more specific for the LD (Gocze and Freeman, 1994). Here, we describe a BODIPY 493/503 assay for the detection of neural fat stores in cultured cells (Figure 1) (Wang et al., 2013).

Abstract:Lipid droplets (LDs) with own interface architecture not only play crucial roles in protecting cell from lipotoxicity and lipoapoptosis but also closely relate with many diseases such as fatty liver and diabetes. Thus, as one of important applied biomaterials, fluorescent probes with ultrahigh selectivity for in-situ and high-fidelity imaging LDs in living cells and tissues are critical to elucidate relevant physiological and pathological events as well as detect related diseases. However, available LDs probes only utilizing their waterless neutral cores but ignoring the unique phospholipid monolayer interfaces exhibit low selectivity. Particularly, current probes cannot discriminate neutral cores of LDs from intracellular other lipophilic microenvironments, so that extensively cloud-like background fluorescence diffusing in cytoplasm, which severely limited their applications. Herein, in order to design LDs probes with ultrahigh selectivity, the exceptionally interfacial architecture of LDs is considered adequately and thus an interface-targeting strategy is proposed for the first time. According to the novel strategy, we have developed two amphipathic fluorescent probes (N-Cy and N-Py) by introducing different cations into a lipophilic fluorophore (NBD). Consequently, their cationic moiety precisely locates the interfaces through electrostatic interaction and simultaneously NBD entirely embeds into the waterless core via hydrophobic interaction. Thus high-fidelity and background-free fluorescence imaging of LDs is expectably realized in living cells in-situ. Moreover, LDs in turbid tissues like skeletal muscle slices have been clearly imaged (up to 82 μm depth) in two-photon microscope. Importantly, using N-Cy, we not only intuitively monitored the variations of LDs in number, size and morphology, but also clearly revealed their abnormity in hepatic tissues resulted from fatty liver. Therefore, these unique probes provide excellent imaging tools for elucidating LDs-related physiological and pathological processes, and the interface-targeting strategy possesses universal significance for designing probes with ultrahigh selectivity.
Quantitative high-content/high-throughput microscopy analysis of lipid droplets in subject-specific adipogenesis models

Abstract:Neutral lipids packed in lipid droplets (LDs) are essential as a source of fuel for organisms, and specialized storing cells, the adipocytes, provide a buffer for energy variations. Many modern-society-disorders are connected with excess accumulation or deficiency of LDs in adipose tissue. Intracellular LD number and size distribution reflect the tissue conditions, while the associated mechanisms and genes rs are still poorly understood. Large-scale genetic screens using human in vitro differentiated primary adipocytes require cell samples donated from many patients. The heterogeneity appearing between donors highlighted the need for high-throughput methods robust to individual variations. Previous image analysis algorithms failed to handle individual LDs, but focused on averages, hiding population heterogeneity. We present a new high-content analysis (HCA) technique for analysis of fat cell metabolism using data from a large-scale RNAi screen including images of more than 500 k in vitro differentiated adipocytes from three donors. The RNAi-based suppression of Perilipin 1 (PLIN1), a protein involved in the adipocyte lipid metabolism, served as a positive control, while cells treated with randomized RNA served as negative controls. We validate our segmentation by comparing our results to those of previously published methods: We also evaluate the discriminative power of different morphological features describing LD size distribution. Classification of cells as containing few large or many small LDs followed by calculating the percentage of cells in each class proved to discriminate the positive PLIN1-suppressed phenotype from the untreated negative control with an area under the receiver operating characteristic curve of 0.98. The results suggest that this HCA method offers improved segmentation and classification accuracy, and can, thus, be utilized to quantify changes in LD metabolism in response to treatment in many cell models relevant to a variety of diseases. © 2017 International Society for Advancement of Cytometry.

Abstract:Background Lipid droplets (LDs) are an important organelle as the main energy storage site in cells. LDs viscosity controls the material and energy exchange between it and other organelles. Furthermore, the LDs metabolic abnormalities, cell dysfunction, some diseases may be attributed to the singular LDs viscosity. Currently, the fluorescent probes for sensing the variations of LDs viscosity are still scarce and expose some drawbacks of low fluorescence quantum yield, low sensitivity and LDs polarity interference. Thus, the development of high performance probes is significant to detect LDs viscosity. Results We hereby provide a lipophilic fluorescent probe ( TPE-BET) with high fluorescence quantum yield ( Φ f , 0.91 in glycerol) for imaging LDs viscosity in living cells. With the increase of viscosity from 0.54 cp to 934 cp, the fluorescence at λ ex /λ em = 405/520 nm and the fluorescence quantum yield of TPE-BET linearly increased by 64.9 and 128.5 folds, respectively. Meanwhile, the outstanding LDs staining capability of TPE-BET may provide a high spatial resolution for LDs imaging. The cell imaging of TPE-BET not only successfully observed the viscosity variations of LDs in cell stress models, e.g., ferroptosis, inflammation and mitophagy, but also revealed the increased viscosity and extracellular delivery of LDs in heavy metal cell injury models (Hg/As) for the first time, which may supply concrete evidence for understanding the structure and function of LDs. Significance This represents a new fluorescent probe TPE-BET with high fluorescence quantum yield for imaging LDs viscosity, which may decrease the dose of probe and excitation light intensity along with the improvement on signal noise ratio (S/N). The imaging results of TPE-BET clarified that LDs viscosity may be an appraisal index on cell differentiation, state evaluation and drug screening.

Abstract:Lipid droplets (LDs) are ubiquitous intracellular organelles present in various cell types, ranging from adipocytes to cancer cells. Despite their involvement in numerous diseases, current methodologies for monitoring LD dynamics rely primarily on static imaging with a limited selection of fluorescent dyes. In this study, we introduce two novel BODIPY-derived fluorophores designed to facilitate real-time monitoring of LD dynamics. By incorporating triphenylamino (TPA) moieties into the BODIPY core both asymmetrically and symmetrically, we modulated the fluorescence response to the green (GFP) and deep-red (Cy5) confocal channels, respectively, thus allowing dual-channel fluorescence capability for high-resolution, dynamic imaging of LDs within live cells, providing new insights into their behavior.

Abstract:Lipids are essential for various cellular functions, including energy storage, membrane flexibility, and signaling molecule production. Maintaining proper lipid levels is important to prevent health problems such as cancer, neurodegenerative disorders, cardiovascular diseases, obesity, and diabetes. Monitoring cellular lipid droplets (LDs) in real-time with high resolution can provide insights into LD-related pathways and diseases owing to the dynamic nature of LDs. Fluorescence-based imaging is widely used for tracking LDs in live cells and animal models. However, the current fluorophores have limitations such as poor photostability and high background staining. Herein, we developed a novel fluorogenic probe based on a push-pull interaction combined with aggregation-induced emission enhancement (AIEE) for dynamic imaging of LDs. Probe 1 exhibits favorable membrane permeability and spectroscopic characteristics, allowing specific imaging of cellular LDs and time-lapse imaging of LD accumulation. This probe can also be used to examine LDs in fruit fly tissues in various metabolic states, serving as a highly versatile and specific tool for dynamic LD imaging in cellular and tissue environments.

Abstract:Lipid droplets (LDs) and lysosomes are vital organelles that play crucial roles in various physiological and pathological processes. However, simultaneous two-color visualization of these two organelles using a single probe for cell homeostasis monitoring remains a challenge due to the lack of rational design strategies. To address this issue, we have developed an aggregation-induced emission (AIE) fluorescent probe named TPE-NDI-Mor with an electron donor (D)–acceptor (A) structure, which can stain both LDs and lysosomes with high selectivity through green and red fluorescence imaging, respectively. A detailed mechanism study revealed that TPE-NDI-Mor , with a twisted intramolecular charge transfer (TICT) effect, shows a high affinity for a polar microenvironment. Additionally, the probe also demonstrates good stability, high anti-interference performance and a large Stokes shift, making it suitable for visualizing cell homeostasis and further disease diagnosis by tracking the dynamic changes of LDs and lysosomes.

Abstract:Lipid droplets (LDs) are involved in a variety of complex life activities in cells, which are closely related to human diseases including obesity, diabetes and cancer. LDs are generally considered as a kind of organelle with extremely low water content and high viscosity. Herein, two novel probes (E)-2-(3-(2-(7-(diethylamino)-2-oxo-2H-chromen-3-yl)vinyl)-5,5-dimethylcyclohex-2-en-1-ylidene)malononitrile (LDP-1) and (E)-2-(3-(4-(diethylamino)styryl)-5,5-dimethylcyclohex-2-en-1-ylidene)malononitrile (LDP-2) were synthesized, which exhibited large Stokes shifts of 4758 cm−1 and 3986 cm−1 respectively. The two probes emitted strongly in hydrophobic and high viscosity environments. Moreover, bio-probes LDP-1 and LDP-2 exhibited low biological toxicity, and good selectivity. The two probes were also fit for selectively monitoring LDs distribution in living HeLa cells.

Abstract:AML with eosinophilia belongs to the morphologic cytogenetic entity M1/M2 t(8;21) with the involvement of the genes AML1/ETO. These eosinophils differ only slightly from normal eosinophils with one rare exception i.e. Auer rods in eosinophils which has only been found on peroxidase staining: This subtype belongs to the good prognosis group of AML. AML with inv(16) (mostly M4Eo) per se is a morphologic-cytogenetic entity with inv(16),--rarely t(16;16), and the genes MYL 11/CBF beta involved. The eosinophils show special abnormalities. AML with inv(16) also belongs to the good prognosis group of AML.

Abstract:Lipid droplets (LDs) are involved in various physiological processes and associated with cancer development, and are regarded as a potential tumor marker for cancer diagnosis. Monitoring LDs is of prior importance to understand their involvement in biological mechanisms and the early detection of cancers. Highly sensitive and specific noninvasive fluorescent probes are particularly desirable for imaging LDs and cancer diagnosis. Herein, according to the high-viscosity and low-polarity microenvironment in LDs, we developed four easily prepared LDs-specific probes based on noncharged merocyanines. Among them, LD-1 absorbs and emits in the near-infrared (NIR) region with a large Stokes shift. Importantly, LD-1 displayed high sensitivity to high viscosity and low polarity, which allowed it to show high LDs-targeting ability. Abstract:Lipid droplets (LDs) are cellular organelles that are essential for maintaining lipid and energy homeostasis. Once regarded as merely inert fat particles, they are now recognized as highly dynamic, mobile organelles required for preventing lipotoxicity and for interacting and cooperating with various organelles. Despite the progress made in understanding the role of LDs, a number of fundamental questions remain unanswered. Effective imaging agents for observing the morphology and dynamic physiological processes of LDs in cells could help address this knowledge gap. Such probes are expected to aid in our understanding of LDs and facilitate the development of new and effective therapeutics. In this review, we have provided a brief introduction to the formation and physiological functions of LDs in an attempt to highlight the importance of these underappreciated organelles. Recent examples of LD-based fluorescent probes are discussed, including the fluorescence phenomenon used in their design. To date, both intramolecular charge transfer (ICT) and aggregation-induced emission (AIE) fluorescence mechanisms have been exploited to create LD probes. However, alternative strategies can be envisioned. We hope the readers will be enlightened as to the importance of these key organelles, will be poised to exploit existing probes to explore various biological applications, and be inspired to create new LD fluorescent sensors that will further our understanding of LDs and their associated physiology.

Abstract:Lipid droplets (LDs) are crucial dynamic organelles within cells that play an indispensable role in cellular energy supply, membrane formation, and signal transduction. Therefore, detecting and tracking LDs with high selectivity and resolution is particularly important. In this study, we describe a "trident" anchoring strategy (based on polarity, lipophilicity, and viscosity sensitivity) to achieve highly specific and dynamic imaging of LDs. A series of carbazole-substituted fluorescent protein (Car-FP) mimics conjugated with different electron-nature substituents (A-H) were synthesized and characterized. Spectroscopic properties indicate that the electron-donating triphenylamine (TPA)-conjugated H exhibits high sensitivity to polarity, viscosity response, and lipophilicity. Colocalization experiments in cells also demonstrate that H selectively localizes at LDs in living cells, displaying a bright yellow fluorescence signal (lambda ex = 488 nm; lambda em = 577 nm) with a larger Stokes shift (89 nm) than the commercial LD dye bodipy 493/503 (10 nm). Super-resolution fluorescence imaging (SIM) and lifetime imaging (FLIM) are employed to visualize the distribution and aggregation in cell models of nonalcoholic fatty liver disease as well as zebrafish and mice models, demonstrating the potential applications of H in LD biology research and diagnosis of related diseases.
Push-Pull Fluorescent Dyes with Trifluoroacetyl Acceptor for High-Fidelity Sensing of Polarity and Heterogeneity of Lipid Droplets

Abstract:Imaging and sensing of lipid droplets (LDs) attracted significant attention due to growing evidence for their important role in cell life. Solvatochromic dyes are promising tools to probe LDs' local polarity, but this analysis is biased by their non-negligible emission from intracellular membranes and capacity to emit from both the apolar core and polar interface of LDs. Here, we developed two push-pull solvatochromic dyes based on naphthalene and fluorene cores bearing an exceptionally strong electron acceptor, the trifluoroacetyl group. The latter was found to boost the optical properties of the dyes by shifting their absorption and emission to red and increasing their extinction coefficient, photostability, and sensitivity to solvent polarity (solvatochromism). In contrast to classical solvatochromic dyes, such as parent aldehydes and reference Nile Red, the new dyes exhibited strong fluorescence quenching by millimolar water concentrations in organic solvents. In live cells, the trifluoroacetyl dyes exhibited high specificity to LDs, whereas the parent aldehydes and Nile Red showed a detectable backgrounds from intracellular membranes. Experiments in model lipid membranes and nanoemulsion droplets confirmed the high selectivity of new probes to LDs in contrast to classical solvatochromic dyes. Moreover, the new probes were found to be selective to the LDs oil core, where they can sense lipid unsaturation and chain length. Their ratiometric imaging in cells revealed strong heterogeneity in polarity within LDs, which covered the range of polarities of unsaturated triglyceride oils, whereas Nile Red failed to properly estimate the local polarity of LDs. Finally, the probes revealed that LDs core polarity can be altered by fatty acid diets, which correlates with their chain length and unsaturation.

Abstract:In addition to being associated with energy supply, lipid droplets (LDs) play vital roles in many other important physiological and some pathological processes. Fluorescent probes are useful tools for studying the biological functions of LDs. In this study, we develop three new highly fluorescent LD-specific bioprobes, which can be excited by one- and two-photon, with favorable biocompatibility. They have little crosstalk with some commonly used commercially-available green- and red-emissive probes, such as BODIPY 493/503, MitoTracker Red CMXRos (MTR) and LysoTracker Red DND-99 (LTR), thus allowing the simultaneous multicolor imaging. Notably, one of the probes, BIP-1, has superior photostability and allows excellent 3D imaging of cellular LDs. BIP-1 can be used to monitor the number and morphological changes of LDs, and the interactions of LDs with other organelles (e.g. mitochondria) under specific physiological conditions (e.g. starvation). Overall, BIP-1 is a promising alternative fluorescent bioprobe for live-cell LD-specific imaging and long-term tracking during LD-related research.

Abstract:Lipid droplets (LDs) are closely associated various diseases, such as liver diseases and cancers. In the last decade, a mountain of fluorescent probes has been developed for LDs, while the ones bearing near-infrared (NIR) emission, superb specificity, and high stability are still lacking. In this work, we prepare a phenothiazine-derived probe that shows a suitable hydrophobicity with ClogP as 6.25 and a strong emission at 681 nm in the low polarity solvents such as 1,4-dioxane, enabling the NIR imaging of LDs. The probe is verified to showcase high chemostability, superb pH tolerance, and excellent anti-photobleaching capacity. Using the probe, the changes in the polarity, size, and number of LDs can be monitored in living cells under different stimuli. Moreover, a variety of morphological dynamics of LDs is visualized over a period of time, including fusion, fission, transfer of lipid, LD growth, and lipolysis. Utilizing its NIR feature, in vivo tumor imaging is achieved and the therapeutic efficacy of cisplatin is primarily assessed in a triple-negative breast cancer mice model using LDs as the biomarker for the first time. These results suggest that NIR-LD is potential to act as a potent imaging agent for LDs-related diseases.

Abstract:BACKGROUND There have been a few case reports of motor neuron disease in association with Huntington disease (HD). OBJECTIVE To describe a patient presenting with prominent fasciculations, chorea, and possible amyotrophic lateral sclerosis (ALS) in whom genetic testing revealed HD mutation. DESIGN Case report. SETTING University of Texas Southwestern Medical Center, Dallas. Patient A 69-year-old man with chorea and fasciculations. INTERVENTIONS Genetic and electrophysiologic testing. MAIN OUTCOME MEASURES Genetic test result, electrophysiologic test result, and physical examination. RESULTS A 69-year-old man with long-standing depression and failing memory presented with muscle twitches of 8 months' duration. He was found to have choreoathetoid movements and distal weakness on neurological examination. Electrophysiologic studies revealed evidence of motor neuron disease. Genetic test showed CAG repeat of 40 on chromosome 4, confirming the diagnosis of HD. CONCLUSION Motor neuron disease can rarely occur in patients with HD and could be one of its presenting features.

Abstract:As essential subcellular organelles, lipid droplets (LDs) are involved in various the pathophysiological processes of metabolic diseases, including fatty liver and diabetes. Therefore, detecting LDs is conducive for studying the pathogeny and the early diagnosis of associated diseases. In this work, a series of fluorescent probes L1 - L3 based on triphenylamine derivatives were designed and synthesized for tracking LDs in living cells. Due to the different terminal moieties of the three probes, L1 and L3 are aggregation-induced emission (AIE)-active, while L2 is a dual-state emission (DSE) molecule. In addition, confocal cell imaging experiments showed that probes L1 - L3 specifically target intracellular LDs sites without the post washing. Importantly, probe L3 allows real-time monitoring of the dynamic processes and changes in the number of LDs. In summary, probe L3 is an ideal probe for studying LDs-related pathophysiological processes, and this work provides more ideas for the future design and synthesis of high-performance fluorescent probes for targeting LDs.

Abstract:Investigating the interactions between lipid droplets (LDs) and other organelles are greatly significant to the in-depth understand the physiology and pathology of LDs. Despite of commercial LDs probes available, the ER and other membrane structures are also stained due to their amphiphilicity, which limits their further applications. To address this issue, we herein report a highly hydrophobic fluorescent probe to specifically label intracellular LDs with high signal/noise ratio, avoiding undesirable staining of ER and thus achieving to real-time monitor the interactions between LDs and other organelles. The probe showed strong emission in lipid environments but no fluorescence in water and liposomes, enabling the probe with high signal/noise ratio. Using this probe, it was found that the contacts of LDs with ER and mitochondria clearly increased under the condition of starvation.
