Abstract:
In addition to being associated with energy supply, lipid droplets (LDs) play vital roles in many other important physiological and some pathological processes. Fluorescent probes are useful tools for studying the biological functions of LDs. In this study, we develop three new highly fluorescent LD-specific bioprobes, which can be excited by one- and two-photon, with favorable biocompatibility. They have little crosstalk with some commonly used commercially-available green- and red-emissive probes, such as BODIPY 493/503, MitoTracker Red CMXRos (MTR) and LysoTracker Red DND-99 (LTR), thus allowing the simultaneous multicolor imaging. Notably, one of the probes, BIP-1, has superior photostability and allows excellent 3D imaging of cellular LDs. BIP-1 can be used to monitor the number and morphological changes of LDs, and the interactions of LDs with other organelles (e.g. mitochondria) under specific physiological conditions (e.g. starvation). Overall, BIP-1 is a promising alternative fluorescent bioprobe for live-cell LD-specific imaging and long-term tracking during LD-related research.

What problem does this paper attempt to address?
This paper aims to develop new fluorescent probes for three - dimensional imaging and metabolic monitoring of lipid droplets (LDs). Specifically, the authors designed and synthesized three novel fluorescent probes (BIP - 1, BIP - 2 and BIP - 3) based on the benzo[4,5]imidazo[1,2 - a]pyridine structure. These probes have the following characteristics: 1. **High fluorescence performance**: Under excitation at 405 nm, these probes can emit cyan fluorescence (about 475 nm) and have a relatively high quantum yield. 2. **Good biocompatibility**: These probes have low toxicity to cells and are suitable for long - term live - cell imaging. 3. **Low cross - interference**: There is almost no cross - interference with commonly used commercial green and red fluorescent probes (such as BODIPY 493/503, MitoTracker Red CMXRos (MTR) and LysoTracker Red DND - 99 (LTR)), allowing simultaneous multicolor imaging. 4. **Excellent photostability**: In particular, the BIP - 1 probe has excellent photostability and is suitable for 3D imaging and long - term tracking. Through these properties, the BIP - 1 probe can be used to monitor the number and morphological changes of lipid droplets, as well as the interactions between lipid droplets and other organelles (such as mitochondria), especially under specific physiological conditions (such as starvation). Overall, the BIP - 1 probe provides a promising fluorescent bioprobe tool for lipid - droplet - related research.
