Abstract:Na+/K+-ATPases (NKA) in the basolateral membrane of the intestinal enterocytes create a Na+-gradient that drives both ion-coupled fluid uptake and nutrient transport. Being dependent on the same gradient as well as on the environmental salinity, these processes have the potential to affect each other. In salmonids, L-lysine absorption has been shown to be higher in freshwater (FW) than in seawater (SW) acclimated fish. Using electrophysiology (Ussing chamber technique), the aim was to explore if the decrease in L-lysine transport was due to allocation of the Na+-gradient towards ion-driven fluid uptake in SW, at the cost of amino acid transport. Intestinal NKA activity was higher in SW compared to FW fish. Exposure to ouabain, an inhibitor of NKA, decreased L-lysine transport. However, exposure to bumetanide and hydrochlorothiazide, inhibitors of Na+, K+, 2Cl--co-transporter (NKCC) and Na+, Cl--co-transporter (NCC) respectively, did not affect the rate of intestinal L-lysine transport. In conclusion, L-lysine transport is Na+-dependent in rainbow trout and the NKA activity and thus the available Na+-gradient increases after SW acclimation. This increased Na+-gradient is most likely directed towards osmoregulation, as amino acid transport is not compromised in SW acclimated fish.

Abstract:Although iono-regulatory processes are critical for survival of crustaceans during the molt cycle, the mechanisms involved are still not clear. The Na+/K+/2Cl− cotransporter (NKCC), a SLC12A family protein that transports Na+, K+ and 2Cl− into cells, is essential for cell ionic and osmotic regulation. To better understand the role of NKCC in the molt osmoregulation, we cloned and characterized a NKCC gene from the mud crab, Scylla paramamosain (designated as SpNKCC). The predicted SpNKCC protein is well conserved, and phylogenetic analysis revealed that this protein was clustered with crustacean NKCC. Expression of SpNKCC was detected in all the tissues examined but was highest in the posterior gills. Transmission electron microscopy revealed that posterior gills had a thick type of epithelium for ion regulation while the anterior gills possessed a thin phenotype related to gas exchange. During the molting cycle, hemolymph osmolality and ion concentrations (Na+ and Cl−) increased significantly over the postmolt period, remained stable in the intermolt and premolt stages and then decreased at ecdysis. Meanwhile, the expression of SpNKCC mRNA was significantly elevated (26.7 to 338.8-fold) at the ion re-establishing stages (postmolt) as compared with baseline molt level. This pattern was consistent with the coordinated regulation of Na+/K+-ATPase α-subunit (NKA α), carbonic anhydrase cytoplasmic (CAc) isoform and Na+/H+ exchanger (NHE) genes in the posterior gills. These data suggest that SpNKCC may be important in mediating branchial ion uptake during the molt cycle, especially at the postmolt stages.

Abstract:Recent research has shown that nitric oxide (NO) produced by nitric oxide synthases (NOS) is an inhibitor of ion transporter activity and a modulator of epithelial ion transport in fish, but little is known on changes in the NOS/NO system during osmotic stress. We hypothesized that the NOS/NO system responds to salinity changes as an integrated part of the acclimation process. Expression and localization of nos1/Nos1 and nos2/Nos2 were investigated in gill, kidney, and intestine of freshwater (FW)- and seawater (SW)-transferred trout using quantitative PCR, Western blotting, and immunohistochemistry, along with expressional changes of major ion transporters in the gill. The classical branchial ion transporters showed expected expressional changes upon SW transfer, there among a rapid decrease in Slc26a6 mRNA, coding a branchial Cl-/[Formula: see text] exchanger. There was a major downregulation of nos1/ nos2/Nos2 expression in the gill during SW acclimation. A significant decrease in plasma nitrite supported an overall decreased Nos activity and NO production. In the middle intestine, Nos1 was upregulated during SW acclimation, whereas no changes in nos/Nos expression were observed in the posterior intestine and the kidney. Nos1 was localized along the longitudinal axis of the gill filament, beneath smooth muscle fibers of the intestine wall and in blood vessel walls of the kidney. Nos2 was localized within the epithelium adjacent to the gill filament axis and in hematopoietic tissues of the kidney. We conclude that downregulation of branchial NOS is integrated to the SW acclimation process likely to avoid the inhibitory effects of NO on active ion extrusion.

Regulation of apical H⁺-ATPase activity and intestinal HCO₃⁻ secretion in marine fish osmoregulation

Abstract:The absorption of Cl(-) and water from ingested seawater in the marine fish intestine is accomplished partly through Cl(-)/HCO(3)(-) exchange. Recently, a H(+) pump (vacuolar-type H(+)-ATPase) was found to secrete acid into the intestinal lumen, and it may serve to titrate luminal HCO(3)(-) and facilitate further Cl(-)/HCO(3)(-) exchange, especially in the posterior intestine, where adverse concentration gradients could limit Cl(-)/HCO(3)(-) exchange. The H(+) pump is expressed in all intestinal segments and in gill tissue of gulf toadfish (Opsanus beta) maintained in natural seawater. After acute transfer of toadfish to 60 ppt salinity, H(+) pump expression increased 20-fold in the posterior intestine. In agreement with these observations was a fourfold-increased H(+)-ATPase activity in the posterior intestine of animals acclimated to 60 ppt salinity. Interestingly, Na(+)-K(+)-ATPase activity was elevated in the anterior intestine and gill, but not in the posterior intestine. Apical acid secretion by isolated intestinal tissue mounted in Ussing chambers fitted with pH-stat titration systems increased after acclimation to hypersalinity in the anterior and posterior intestine, titrating >20% of secreted bicarbonate. In addition, net base secretion increased in hypersalinity-acclimated fish and was ∼70% dependent on serosal HCO(3)(-). Protein localization by immunohistochemistry confirmed the presence of the vacuolar-type H(+)-ATPase in the apical region of intestinal enterocytes. These results show that the H(+) pump, especially in the posterior intestine, plays an important role in hypersaline osmoregulation and that it likely has significant effects on HCO(3)(-) accumulation in the intestinal lumen and, therefore, the continued absorption of Cl(-) and water.
Fundamental differences between wildlife and biomedical research.

Abstract:Non-human animals have starred in countless productions of biological research. Whether they play the lead or supporting role depends on the nature of the investigation. These differences in the roles of animals affect nearly every facet of animal involvement, including: the choice of species, the sample size, the source of individuals, and the settings in which the animals are used. These roles establish different baselines for animal use that require substantially different ethical considerations. Efficient and appropriate oversight of wildlife research benefits the animals and their investigators. Toward that end, Institutional Animal Care and Use Committee (IACUCs) must appreciate the profound differences between biomedical and wildlife research and recognize the value of the state and federal permitting processes required for wildlife studies. These processes assure us that potential impacts beyond the level of the individual are minimal or are justified. Most importantly, IACUCs must recognize that they, and their investigators, have an obligation to use appropriate guidelines for evaluating wildlife research.

Abstract:Animals living in estuaries suffer from rapid and continuous salinity fluctuations, while the global warming and extreme precipitation aggravate this situation. Osmoregulation is important for estuarine animals adapt to salinity fluctuations. The present study investigated the effects of low salinity stress on osmoregulation and gill transcriptome in two populations of mud crab from Hangzhou Bay and Zhangzhou Bay of China, respectively. Crabs were transferred from salinity 25 ppt to 5 ppt for 96 h. Edematous swelling in gill filaments was caused by low salinity stress and was more serious in Zhangzhou Bay population. Gill Na+/K+-ATPase activity increased (p < 0.01) in both populations under the low salinity stress and was significantly higher (p < 0.01) in Hangzhou Bay population than in Zhangzhou Bay population. According to transcriptome analysis, there were 191 genes differentially expressed under the low salinity stress in gill tissue of both populations. Several ion transport and energy metabolism related pathways, as well as the arginine and proline metabolism pathway, were enriched by these genes. On the other hand, 272 genes were identified to differentially express between two populations under the low salinity stress, but not under the control salinity. The enrichment analysis showed that these genes were mainly related to ion transport, energy metabolism, osmolytes metabolism and methyltransferase activity. In conclusion, the present study suggested that mud crab exploited a combination of extracellular anisosmotic regulation and intracellular isosmotic regulation for osmoregulation under the low salinity stress. Hangzhou Bay population showed a greater osmoregulatory capacity, which is probably due to the enhanced ion transport, energy supply, and osmolytes regulation. Meanwhile, epigenetic modification might also contribute to an inherent osmoregulation ability for Hangzhou Bay population to response to salinity fluctuation rapidly.

Abstract:The life history of Atlantic salmon ( Salmo salar ) includes an initial freshwater phase (parr) that precedes a springtime migration to marine environments as smolts. The development of osmoregulatory systems that will ultimately support the survival of juveniles upon entry into marine habitats is a key aspect of smoltification. While the acquisition of seawater tolerance in all euryhaline species demands the concerted activity of specific ion pumps, transporters, and channels, the contributions of Na + /HCO 3 − cotransporter 1 (Nbce1) to salinity acclimation remain unresolved. Here, we investigated the branchial and intestinal expression of three Na + /HCO 3 − cotransporter 1 isoforms, denoted nbce1.1 , -1.2a , and -1.2b . Given the proposed role of Nbce1 in supporting the absorption of environmental Na + by ionocytes, we first hypothesized that expression of a branchial nbce1 transcript ( nbce1.2a ) would be attenuated in salmon undergoing smoltification and following seawater exposure. In two separate years, we observed spring increases in branchial Na + /K + -ATPase activity, Na + /K + /2Cl − cotransporter 1 , and cystic fibrosis transmembrane regulator 1 expression characteristic of smoltification, whereas there were no attendant changes in nbce1.2a expression. Nonetheless, branchial nbce1.2a levels were reduced in parr and smolts within 2 days of seawater exposure. In the intestine, gene transcript abundance for nbce1.1 increased from spring to summer in the anterior intestine, but not in the posterior intestine or pyloric caeca, and nbce1.1 and -1.2b expression in the intestine showed season-dependent transcriptional regulation by seawater exposure. Collectively, our data indicate that tissue-specific modulation of all three nbce1 isoforms underlies adaptive responses to seawater.

Abstract:1.1. Steady-state drinking rates and intestinal absorption of water and electrolytes in vivo were studied by means of indwelling Foley catheters and a volume indicator (polyethylene glycol).2.2. Drinking rates of 47, 95 and 129 ml/kg body wt. per day were measured in one-third-, half- and full-strength sea water and 66, 78 and 80 per cent of these loads were absorbed from the intestine respectively. Fresh-water fish did not drink.3.3. Net intestinal salt absorption rates (mM/kg body wt. per day) in one-third, half- and full-strength sea water were: sodium, 4, 19·1 and 56; chloride, 5·5, 23·8 and 67·6; potassium, 0·1, 0·4 and 1·3; calcium, 0·08, 0·11 and 0·43; magnesium, 0·06, 0·37 and 3·13; and sulfate, 0·0, 0·0 and 1·0. Relative absorption rates from ingested loads for sodium and chloride were: sodium, 66, 91 and 99 per cent and chloride, 85, 94 and 98 per cent respectively.4.4. In the intestine, ingested media were granually depleted of mono-valent ions and water and enriched in magnesium and sulfate.5.5. In plasma-hyperosmotic media, relative absorption of water and monovalent ions were significantly higher (P<0·01). Integumentary water permeability appears to decrease in such media.
Esophageal expression of Na+/H+ exchanger 2 and -3 (nhe2 and -3) gene transcripts in Atlantic salmon during smoltification and seawater acclimation

Abstract:Atlantic salmon ( Salmo salar) undergo smoltification, a springtime transformation associated with the development of physiological, morphological, and behavioral traits that enable individuals to thrive in pelagic marine habitats. In particular, the capacity for salmon to maintain hydromineral balance in seawater is attained through the coordinated development of ion- and water-transporting processes in key organs such as the gill, kidney, and gastrointestinal tract. To combat dehydration, Atlantic salmon drink ambient seawater that is initially desalinated in the esophagus to enable subsequent solute-linked water absorption by the intestine. The molecular processes underlying esophageal desalination of imbibed seawater and their regulation during smoltification and following seawater exposure remain largely unresolved. The current study explored whether Na + /H + exchangers in the esophagus may contribute to the removal of Na + from luminal seawater. We first determined that both nhe2 and -3 were robustly expressed in the esophagus of smolts; nhe2 was also highly expressed in gill and stomach. Next, we sampled salmon undergoing smoltification and observed springtime elevations in branchial Na + /K + -ATPase activity, Na + /K + /2Cl - cotransporter 1 and cystic fibrosis transmembrane regulator 1 transcription that coincided with increased seawater tolerance. There were no parallel springtime changes in esophageal nhe2 or -3 expression; but nhe3 expression was enhanced in smolts after 2 days of seawater exposure. In a subsequent experiment, we observed elevated nhe2 and -3 expression in smolts that were long-term acclimated (2 weeks) to seawater versus freshwater. Collectively, our results indicate that Na + /H + exchangers contribute to the desalination processes in the esophagus. The next challenge will be to determine whether essential “seawater-adapting hormones,” such as growth hormone and cortisol, regulate the expression of Na + /H + exchangers in support of seawater acclimation. Abstract:Understanding how freshwater species respond to changes in salinity is essential for comprehending their distribution and ecological niches. We investigated the osmoregulatory mechanisms of the freshwater catfish Rhamdia quelen , which could justify occasional incursions and residence in brackish waters and their resistance to salt treatments in aquaculture. Specimens were exposed to freshwater (control) and dilute seawater (15 ‰) for 6 h. It was measured: plasma osmolality and Na + , K + , and Cl − concentrations; gills and kidneys Na + /K + -ATPase (NKA), V(H + )-ATPase (VAT), and carbonic anhydrase (CA) enzymes activities; gills enzymatic immunolocalization and Heat Shock Protein (HSP70) abundance. Our results showed that R. quelen presented hypoosmotic/ionic regulation in 15 ‰. However, there was no significant effect of the increased salinity on enzymatic activities in the gills and kidneys, or in the abundance of HSP70. All enzymatic activities, except for CA, were higher in the kidneys compared to the gills. The immunolocalization of NKA and VAT was observed in ionocytes of the interlamellar region of the gills, and the pavement cells, while CA was detected in both ionocytes and red blood cells. The lack of change in the abundance of HSP70 may be attributed to the protein's high constitutive abundance in the gill cells. The results indicate a complex regulatory network in response to high salinity, showing that brief periods of salty water exposure could be enough for the species to move or spread in brackish waters or survive aquaculture disease treatments.

Abstract:Based on 12 patients with posttraumatic erectile dysfunction due to pelvic fractures, ruptures of urethra and straddle-trauma, the diagnostic and therapeutic means in the management of erectile failure are discussed. The most frequent etiology encountered in posttraumatic erectile failure were combined arterial-neurogenic lesions followed by insufficiencies of the venae profundae penis as a sequelae of pelvic fractures or symphyseal ruptures. Promising therapeutic alternatives represent injections of cavernous bodies with vasoactive drugs, penile implants and also in suitable cases penile revascularization procedures.
A sodium binding system alleviates acute salt stress during seawater acclimation in eels

Abstract:BackgroundTeleosts transiting from freshwater (FW) to seawater (SW) environments face an immediate osmotic stress from ion influxes and water loss, but some euryhaline species such as eels can maintain a stable plasma osmolality during early SW exposure. The time course changes in the gene expression, protein abundance, and localization of key ion transporters suggested that the reversal of the ion transport systems was gradual, and we investigate how eels utilize a Na-binding strategy to slow down the ion invasion and complement the transporter-mediated osmoregulation.ResultsUsing an electron probe micro-analyzer, we localized bound Na in various eel tissues in response to SW transfer, suggesting that the Na-binding molecules were produced to sequester excess ionic Na+ to negate its osmotic potential, thus preventing acute cellular dehydration. Mucus cells were acutely activated in digestive tract, gill, and skin after SW transfer, producing Na-binding molecule-containing mucus layers that fence off high osmolality of SW. Using gel filtration HPLC, some molecules at 18 kDa were found to bind Na in the luminal secretion of esophagus and intestine, and higher binding was associated with SW transfer. Transcriptome and protein interaction results indicated that downregulation of Notch and β-catenin pathways, and dynamic changes in TGFβ pathways in intestine were involved during early SW transition, supporting the observed histological changes on epithelial desquamation and increased mucus production.ConclusionsThe timing for the activation of the Na-binding mechanism to alleviate the adverse osmotic gradient was temporally complementary to the subsequent remodeling of branchial ionocytes and transporting epithelia of the digestive tract. The strategy to manipulate the osmotic potential of Na+ by specific binding molecules is similar to the osmotically inactive Na described in human skin and muscle. The Na-binding molecules provide a buffer to tolerate the salinity changes, which is advantageous to the estuary and migrating fishes. Our data pave the way to identify this unknown class of molecules and open a new area of vertebrate osmoregulation research.

Abstract:We used the AAMR's Adaptive Behavior Scale to ascertain current menstrual status in a population-based sample of 157 women with Down's syndrome (DS) and 187 women with other intellectual disability, all 40 years of age or older. The age-adjusted likelihood of menopause was twice as high in women with DS syndrome as in women with other intellectual disability (OR = 2.3; 95% CI = 1.1-4.9). Treated thyroid conditions did not influence menstrual status and did not modify the relationship between DS and menstrual status. These findings support the hypothesis that women with DS experience menopause at an earlier age and that this may be associated with accelerated aging.

Effect of salinity on flavin-containing monooxygenase expression and activity in rainbow trout (Oncorhynchus mykiss)

Abstract:In order to obtain more information about the physiological role(s) of flavin-containing monooxygenases (FMOs) in euryhaline teleost fishes, two experimental series were performed using adult and juvenile rainbow trout (Oncorhynchus mykiss). Cannulated adult trout were exposed to freshwater or 21% seawater for 48 h, whereas juvenile trout were acclimated to one of four different salinities: freshwater, 7%, 14%, or 21% during a 2-week period. FMO expression and activity were determined in red blood cells (RBC), liver, gill, kidney, gut, heart and brain. Furthermore, the content of trimethylamine oxide (TMAO; an FMO metabolite and an osmolyte) as well as urea were determined in various tissues. FMO expression and activity increased significantly and in a salinity dependent manner in osmoregulatory organs (gills, kidney and gut) in both juveniles and adult trout and, furthermore, in RBC in adults. No significant changes were observed in liver or heart. Urea content increased significantly and in a salinity dependent manner in all tissues, whereas TMAO was accumulated primarily in muscle tissue. Salinity dependent adjustment of FMO expression and activity primarily in osmoregulatory organs as well as regulation of TMAO content in muscle is consistent with previous studies showing an association of FMO with osmoregulation in euryhaline teleosts. However, the lack of a parallel increase of TMAO with urea in other tissues of fish at high salinity indicates other mechanisms of protection from intracellular urea may exist in non-muscular tissues.
Salinity acclimation affects the somatotropic axis in rainbow trout

Abstract:In this study, we set out to examine the role of the somatotropic axis in the ion-regulation process in rainbow trout. Specifically, our objective was to examine whether plasma insulin-like growth factor-binding proteins (IGFBPs) are modulated by gradual salinity exposure. To this end, freshwater (FW)-adapted rainbow trout were subjected to gradual salinity increases, up to 66% seawater, over a period of 5 days. During this acclimation process, minimal elevations in plasma Ca 2+ and Cl − were seen in the salinity-acclimated groups compared with FW controls. There were no changes in plasma Na + levels, and only a minor transient change in plasma cortisol levels was seen with salinity exposure. The salinity challenged animals responded with elevations in plasma growth hormone (GH) and IGF-I levels and gill Na + -K + -ATPase activity. We identified IGFBPs of 21, 32, 42, and 50 kDa in size in the plasma of these animals, and they were consistently higher with salinity. Despite the overall increase in IGFBPs with salinity, transient changes in individual BPs over the 5-day period were noted in the FW and salinity-exposed fish. Specifically, the transient changes in plasma levels of the 21-, 42-, and 50-kDa IGFBPs were different between the FW and salinity groups, while the 32-kDa IGFBP showed a similar trend (increases with sampling time) in both groups. Considered together, the elevated plasma IGFBPs suggest a key role for these binding proteins in the regulation of IGF-I during salinity acclimation in salmonids.
Esophageal desalination is mediated by Na⁺, H⁺ exchanger-2 in the gulf toadfish (Opsanus beta)

Abstract:Esophageal desalination is a crucial step in the gastrointestinal water absorption pathway, as this pre-intestinal processing establishes the osmotic conditions necessary for water absorption. Previous work has shown that esophageal Na(+) absorption is amiloride sensitive; however, it is as yet unclear if Na(+), H(+) exchangers (NHE) or Na(+) channels (ENaC) are responsible. The purpose of the current study was therefore to investigate the roles that NHE isoforms may play in this process in a marine teleost, the gulf toadfish (Opsanus beta), as well as what role NHE isoforms may play in the downstream intestinal Na(+) transport. A combination of symmetrical current clamp and asymmetrical voltage clamp experiments showed the esophagus to contain both an ion absorptive current (I(sc)=0.83±0.68) and serosal side negative transepithelial potential (TEP=-4.9±0.6). (22)Na uptake (J(Na)(m→s)) was inhibited by 0.5 mM EIPA, with no effect of 0.1 mM amiloride, 1 mM furosemide or 1 mM thiazide. A Cl(-) free saline reduced J(Na)(m→s) by 40% while also reducing conductance and reversing TEP. These results suggest that both transcellular and paracellular components contribute to esophageal Na(+) transport, with transcellular transport mediated by NHE. The NHE1, NHE2 and NHE3 genes were amplified and tissue distribution analysis by real-time PCR showed high NHE2 expression levels in the esophagus and stomach. Little NHE3 expression was observed throughout the gastrointestinal tract, and NHE2 expression was absent from the intestine. Hypersalinity (60 ppt) had no effect on the expression profile of NHE2, slc4a2, scl26a6, CAc or V-type ATPase (β-subunit), suggesting that esophageal desalination is less flexible in response to osmotic stress than the intestine.
Hypersalinity tolerance of mummichogs ( Fundulus heteroclitus ): A branchial transcriptomic analysis

Abstract:Along the east coast of North America, mummichogs ( Fundulus heteroclitus ) are subjected to a broad range of salinities in their nearshore habitats. However, there is a paucity of information regarding the molecular and cellular processes that mummichogs (and other highly osmotolerant fishes) engage to survive environmental salinities greater than seawater (SW). To reveal branchial processes underlying their extraordinarily broad salinity tolerance, we performed an RNA-Seq analysis to identify genes differentially expressed by mummichogs residing in 3, 35, and 105 ppt conditions. We identified a series of DEGs previously associated with both freshwater (FW)- and SW-type ionocytes; however, the heightened expression of anoctamin 1a , a Ca 2+ -activated Cl − channel, in 35 and 105 ppt indicates that an undescribed Cl − -secretion pathway may operate within the SW-type ionocytes of mummichogs. Concerning FW-adaptive branchial processes, we identified claudin 5a as a gene whose product may limit the diffusive loss of ions between cellular tight junctions. Further, in response to hypersaline conditions, we identified DEGs linked with myo -inositol synthesis and kinase signaling. This study provides new molecular targets for future physiological investigations that promise to reveal the mechanistic bases for how mummichogs and other euryhaline species tolerate hypersaline conditions.
