Abstract:Background: Salinity is one of the main influencing factors in the culture environment and is extremely important for the survival, growth, development and reproduction of aquatic animals. Methods: In this study, a comparative transcriptome analysis (maintained for 45 days in three different salinities, 30 psu (HC group), 18 psu (MC group) and 3 psu (LC group)) was performed by high-throughput sequencing of economically cultured Penaeus monodon. P. monodon gill tissues from each treatment were collected for RNA-seq analysis to identify potential genes and pathways in response to low salinity stress. Results: A total of 64,475 unigenes were annotated in this study. There were 1,140 upregulated genes and 1,531 downregulated genes observed in the LC vs. HC group and 1,000 upregulated genes and 1,062 downregulated genes observed in the MC vs. HC group. In the LC vs. HC group, 583 DEGs significantly mapped to 37 signaling pathways, such as the NOD-like receptor signaling pathway, Toll-like receptor signaling pathway, and PI3K-Akt signaling pathway; in the MC vs. HC group, 444 DEGs significantly mapped to 28 signaling pathways, such as the MAPK signaling pathway, Hippo signaling pathway and calcium signaling pathway. These pathways were significantly associated mainly with signal transduction, immunity and metabolism. Conclusions: These results suggest that low salinity stress may affect regulatory mechanisms such as metabolism, immunity, and signal transduction in addition to osmolarity in P. monodon. The greater the difference in salinity, the more significant the difference in genes. This study provides some guidance for understanding the low-salt domestication culture of P. monodon.
Effects of temperature and salinity on tissue structure, antioxidant capacity and heat shock protein gene expression of Penaeus monodon

Abstract:Abstract In order to explore the influence of temperature and salinity on stress response of P. monodon , four experimental groups were set, respectively were normal temperature and normal salt (Control group), normal temperature and low salt(Group A), high temperature and low salt(Group B), high temperature and normal salt(Group C). The stress experiment was conducted for 96 hours, and the activities of acid phosphatase(ACP), alkaline phosphatase(AKP), total superoxide dismutase (T-SOD) and total antioxidant capacity (T-AOC) in hepatopancreas, the changes of tissue structure and the expression of related heat shock protein genes were measured．The results showed that the branchial structures of the three stress groups were found to be damaged to varying degrees. In Group A, AKP and ACP levels increased initially and then decreased, while T-SOD levels decreased initially and then steadily increased. In Group B and Group C, the four enzymes showed similar trends.There was no significant difference in the expression level of Hsps in the low salt group. The expression levels of Hsp-10, Hsp-60 and Hsp-90 genes showed a similar trend, and the maximum expression levels of the four genes were all at 96h. The expression levels of Hsps in muscle tissue were similar to those in hepatopancreas tissue. However, the highest expression levels of Hsp-70 and Hsp-90 in high temperature and low salt group were at 24h. Through this work on P. monodon stress responses to high temperature and low salinity, we have gained a deeper understanding of the physiological responses to environmental stressors, which will be useful in the development of protective measures for the shrimps under aquaculture conditions that replicate these stressors.
Effects of Low Salinity Stress on Osmoregulation and Gill Transcriptome in Different Populations of Mud Crab Scylla Paramamosain.

Abstract:Animals living in estuaries suffer from rapid and continuous salinity fluctuations, while the global warming and extreme precipitation aggravate this situation. Osmoregulation is important for estuarine animals adapt to salinity fluctuations. The present study investigated the effects of low salinity stress on osmoregulation and gill transcriptome in two populations of mud crab from Hangzhou Bay and Zhangzhou Bay of China, respectively. Crabs were transferred from salinity 25 ppt to 5 ppt for 96 h. Edematous swelling in gill filaments was caused by low salinity stress and was more serious in Zhangzhou Bay population. Gill Na+/K+-ATPase activity increased (p < 0.01) in both populations under the low salinity stress and was significantly higher (p < 0.01) in Hangzhou Bay population than in Zhangzhou Bay population. According to transcriptome analysis, there were 191 genes differentially expressed under the low salinity stress in gill tissue of both populations. Several ion transport and energy metabolism related pathways, as well as the arginine and proline metabolism pathway, were enriched by these genes. On the other hand, 272 genes were identified to differentially express between two populations under the low salinity stress, but not under the control salinity. The enrichment analysis showed that these genes were mainly related to ion transport, energy metabolism, osmolytes metabolism and methyltransferase activity. In conclusion, the present study suggested that mud crab exploited a combination of extracellular anisosmotic regulation and intracellular isosmotic regulation for osmoregulation under the low salinity stress. Hangzhou Bay population showed a greater osmoregulatory capacity, which is probably due to the enhanced ion transport, energy supply, and osmolytes regulation. Meanwhile, epigenetic modification might also contribute to an inherent osmoregulation ability for Hangzhou Bay population to response to salinity fluctuation rapidly.

Abstract:This study aimed to investigate whether different salinity stresses (0, 2, 4, 6, or 8 ppt NaCl) affect the histoarchitecture, serum biochemical indices, antioxidant responses, and transcriptome analysis of the red swamp crayfish Procambarus clarkii. Transmission electron microscopy results showed that the degree of damage to the nuclei and mitochondria in the hepatopancreas increased with increasing salinity. Scanning electron microscopy results showed that the degree of gill wrinkles was enhanced under salinity stress. Serum biochemical indices demonstrated that the cholesterol significantly decreased while the triglyceride, aspartate transaminase, and alanine transaminase contents significantly increased with increasing salinity. The antioxidant parameters, including catalase, total antioxidant capacity, and glutathione peroxidase, significantly decreased, while the malondialdehyde content significantly increased under salinity stress. Transcriptome analysis revealed that the expression pattern of immune-related genes showed a downward trend. These findings enrich our knowledge about the salinity stress response of farmed organisms and provide a theoretical base for salinity domestication and saline soil cultivation of P. clarkii, which might contribute to income improvement, employment generation, food security, and environmental safety.

Abstract:With global warming on the rise, hypersalinity events are occurring with increasing frequency due to accelerated evaporation rates of seawater. Hypersalinity affects the distribution, growth, and physiological processes of aquatic organisms, especially for osmoconforming bivalves that lack complete means of osmoregulation. In this way, knowledge concerning comprehensive metabolic regulation is limited in hypersalinity-stressed bivalves. The hard clam, Mercenaria mercenaria, is an important pond-farmed bivalve in China, with an annual output value of 150 million USD. This species can be farmed in salt pans because it is highly resistant to hypersalinity. In this study, we investigated the comprehensive pattern of metabolic regulation in hard clams under hypersalinity stress. We identified 979 metabolites in the gills of hard clams, 295 of which were differentially accumulated after different periods of exposure to hypersalinity (8 h, 1 day, and 5 days), compared with the control. Critical hypersalinity-responsive metabolites, including acyl carnitine, oxidized lipids, and several antioxidants were identified using K-means analysis. During hypersalinity exposure, the activities of hexokinase and pyruvate kinase, and the pyruvic acid content significantly increased, suggesting that glycolysis was enhanced. Alanine and glycine were identified as key free amino acids involved in osmoregulaion. After 5 days of hypersalinity exposure, the accumulation of alanine and lactic acid marked the initiation of anaerobic glycolysis. The up-regulation of amino acid catabolism and inhibition of ammonia excretion led to ammonia accumulation in the gills. Glutamine synthetase and glutamate dehydrogenase activities significantly increased to reduce ammonia toxicity. Many antioxidant metabolites, such as allantoin, cinnamic acid, and trigonelline, were dramatically up-regulated to relieve hypersalinity-induced oxidative stress. The accumulation of lysophosphatides suggested that cell membranes damaged by prolonged oxidative stress and/or hyperosmosis were dissolved by phospholipase A1. This process helped to maintain overall cellular homeostasis. Potential biomarkers of hypersalinity stress in hard clams were identified. These results will facilitate the assessment of the physiological status of hard clam in aquaculture and provide novel insights into the metabolic regulatory mechanisms of bivalves under hypersalinity stress.

Hypersalinity tolerance of mummichogs ( Fundulus heteroclitus ): A branchial transcriptomic analysis

Abstract:Along the east coast of North America, mummichogs ( Fundulus heteroclitus ) are subjected to a broad range of salinities in their nearshore habitats. However, there is a paucity of information regarding the molecular and cellular processes that mummichogs (and other highly osmotolerant fishes) engage to survive environmental salinities greater than seawater (SW). To reveal branchial processes underlying their extraordinarily broad salinity tolerance, we performed an RNA-Seq analysis to identify genes differentially expressed by mummichogs residing in 3, 35, and 105 ppt conditions. We identified a series of DEGs previously associated with both freshwater (FW)- and SW-type ionocytes; however, the heightened expression of anoctamin 1a , a Ca 2+ -activated Cl − channel, in 35 and 105 ppt indicates that an undescribed Cl − -secretion pathway may operate within the SW-type ionocytes of mummichogs. Concerning FW-adaptive branchial processes, we identified claudin 5a as a gene whose product may limit the diffusive loss of ions between cellular tight junctions. Further, in response to hypersaline conditions, we identified DEGs linked with myo -inositol synthesis and kinase signaling. This study provides new molecular targets for future physiological investigations that promise to reveal the mechanistic bases for how mummichogs and other euryhaline species tolerate hypersaline conditions.

Abstract:Marine bivalves in intertidal zones and land-based seawater ponds are constantly subjected to a wide range of salinity fluctuations due to heavy rainfall, intense drought, and human activities. As osmoconformers, bivalves rely primarily on rapid release or accumulation of free amino acids (FAAs) for osmoregulation. Euryhaline bivalves are capable of withstanding hyposaline and hypersaline environments through regulation of physiology, metabolism, and gene expression. However, current understanding of the molecular mechanisms underlying osmoregulation and salinity adaptation in euryhaline bivalves remains largely limited. In this study, RNA-seq, WGCNA and flow cytometric analysis were performed to investigate the physiological responses of hard clams ( Mercenaria mercenaria ) to acute short-term hyposalinity (AL) and hypersalinity (AH), and chronic long-term hyposalinity (CL) and hypersalinity (CH) stress. We found that amino acids biosynthesis was significantly inhibited and aminoacyl-tRNA biosynthesis was augmented to decrease intracellular osmolarity during hyposaline exposure. Under CH, numerous autophagy-related genes ( ATGs ) were highly expressed, and the autophagy activity of gill cells were significantly up-regulated. A significant decrease in total FAAs content was observed in gills after NH 4 Cl treatment, indicating that autophagy was crucial for osmoregulation in hard clams during prolonged exposure to hypersaline environments. To prevent premature or unnecessary apoptosis, the expression of cathepsin L was inhibited under AL and AH, and inhibitors of apoptosis was augmented under CL and CH. Additionally, neuroendocrine regulation was involved in salinity adaption in hard clams. This study provides novel insights into the physiological responses of euryhaline marine bivalves to hyposaline and hypersaline environments.

Abstract:Scylla paramamosain is a high-quality cultivar for saline-alkaline water aquaculture as a euryhaline crustacean species. However, salinity impacts the respiratory metabolism, growth, and survival of marine crustaceans. The metabolic response of crabs adapting to multiple low salinity environments has not been thoroughly studied yet, especially in inland saline-alkaline water. In this study, we analyzed metabolites in the gill and hemolymph of crabs cultured in three different low salinity environments. The results showed that membrane composition (lipids and lipid molecules) and free amino acids played an essential role in the osmoregulation of crabs, and the energy consumption accompanied as well. Meanwhile, S. paramamosain relied on ion transport and energy metabolism under acute/short-term low salinity conditions for osmoregulation. In contrast, amino acids and energy metabolism occupied a leading position in long-term low salinity. Furthermore, taurine and hypotaurine play a vital role in crabs adapting to inland saline-alkaline water. This is the first study to identify the crucial metabolites and key pathways as biomarkers to differentiate the metabolic mechanisms of S. paramamosain under multiple low salinity stress modes based on GC-MS technology, which provided novel insight into the metabolic response of S. paramamosain adapting to inland low salinity saline-alkaline water, and provided theoretical guidance for the aquaculture of S. paramamosain in the inland saline-alkaline water.

Abstract:Scylla paramamosain is an important marine aquaculture crustacean on the southeastern coast of China. Given the difficulties of overcrowded coastal aquaculture space and insufficient production, inland saline-alkaline water culture has the potential to alleviate this bottleneck. This study separated 600 crabs into four groups: normal salinity (12-18 ppt, NS), coastal low salinity (3-5 ppt, CS), inland low salinity saline-alkaline (1.5 ppt, IS), and acute low salinity (23 ppt down to 3 ppt, AS), followed by a transcriptomic analysis of the gills. CS-vs-NS, IS-vs-NS, and AS-vs-NS obtained 1154, 1012, and 707 DEGs, respectively. GO analysis showed that the DEGs of the three comparison groups were mainly involved in cellular process, metabolic process, biological regulation, organelle, membrane, extracellular region, binding, and catalytic activity. The findings demonstrate that a significant number of genes are engaged in controlling metabolic processes in the osmoregulation process, and that cell membrane catalysis and different enzymes play a vital part in the environmental adaption process. KEGG enrichment analysis revealed that IS possesses a considerable number of signaling pathways that play important roles in osmoregulation when compared to NS. The cAMP signaling pathway increased the expression of CaM and Na + /K + -ATPase. These findings show that cations like Ca 2+ , Na + , and K + are critical for S. paramamosain to adapt to inland low salinity saline-alkaline water, and that the cAMP signaling pathway regulates their levels. This research provides a theoretical foundation for improving the saline-alkaline aquaculture technology of S. paramamosain .

A laboratory simulation of the effects of acute salinity decrease on osmoregulation and Hsps expression in the swimming crab, Portunus trituberculatus: implications for aquaculture

Abstract:Groups of swimming crabs, Portunus trituberculatus, were subject to sharp decreases of water salinity from 30 to 25, 20 and 15 psu, respectively, under laboratory conditions to simulate events that could occur during culture. Their osmoregulation and heat shock proteins (Hsps) expression were analysed. After the salinity stress, hemolymph osmolality, osmotic enzymes (Na+/K+-ATPase and carbonic anhydrase) activities in gills and Hsps (Hsp70 and Hsp90) expression in gills and muscles were measured. Dramatic changes in all parameters (p < 0.05) were detected after acute salinity decrease to 15 psu, while a decrease to 25 psu resulted in only slight changes except in the muscle Hsps levels. Hemolymph osmolalities in all treatments were significantly reduced after the salinity decrease (p < 0.05), and the greater the salinity change, the more the osmolality decrease. The osmotic enzymes activities and Hsps concentrations increased simultaneously after the salinity decrease. The osmotic enzymes activities were elevated at the end of the study with a significant difference at 15 psu but not at 25 psu compared with controls. The muscle Hsps levels were reduced in all treatments and the gill Hsps levels increased except that at 25 psu. It can be concluded that acute salinity decrease can have a negative impact on P. Trituberculatus but a salinity change of less than 10 psu is likely to be harmless for crab farming.

Abstract:Salinity stress can trigger a series of physiological changes. However, the mechanism underlying the response to acute salinity stress in Macrobrachium rosenbergii remains poorly understood. In this study, osmoregulation, physiological metabolism, antioxidant capacity, and apoptosis were examined over 96 h of acute salinity stress. Hemolymph osmolality increased with increasing salinity. After 48 h of salinity exposure, the glucose, triglycerides, total protein, and total cholesterol contents in two salinity stress groups (13 and 26‰ salinity) were significantly lower than those in the 0‰ salinity group. The highest levels of these parameters were detected at 6 h; however, superoxide dismutase (SOD), total antioxidant capacity (T-AOC), and malondialdehyde (MDA) were the lowest at 96 h in the 13‰ salinity group. The activity of immunity-related enzyme alkaline phosphatase (AKP) showed a decreasing trend with increasing salinity and remained at a low level in the 26‰ salinity group throughout the experiment. No significant differences were observed in aspartate aminotransferase (AST), alanine aminotransferase (ALT), or lysozyme (LZM) among the three treatments at 96 h. After 96 h of salinity treatments, the gill filament diameter significantly decreased, and a more pronounced terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL)-positive signal was detected in the 13‰ and 26‰ groups compared to that in the 0‰ group. Expression levels of apoptosis-related genes, including Cysteine-aspartic acid protease 3 (Caspase 3), Cysteine-aspartic acid protease 8 (Caspase 8), Cytochrome c (Cyt-c), tumor suppressor gene (P53), Nuclear factor kappa-B (NF-κB), and B cell lymphoma 2 ovarian killer (Bok) were significantly higher in the 26‰ salinity group than in the other groups at 24 h, but lower than those in the 0‰ salinity group at 96 h. Cyt-c and P53 levels exhibited a significantly positive relationship with MDA, AST, and LZM activity during salinity stress. In the 13‰ salinity group, Bok expression was significantly correlated with SOD, T-AOC, AKP, acid phosphatase, and LZM activity, whereas in the 26‰ group, the AST content was positively correlated with Caspase 8, Cyt-c, and P53 expression. A significant negative relationship was observed between Caspase 3 expression and catalase (CAT) activity. These findings provide insight into the mechanisms underlying the response to acute salinity stress and will contribute to improving M. rosenbergii aquaculture and management practices.

Abstract:Salinity is an important environmental factor affecting the growth and survival of marine organisms. Marine shellfish possess the efficient osmoregulatory mechanism to cope with either hypo- or hyperosmotic stress. However, little is known about the molecular and metabolic adaptation involved in the osmotic regulation of these intertidal habitants. The Manila clam Ruditapes philippinarum is a worldwide cultured intertidal shellfish, which usually encounters a great risk of salinity stress in its coastal habitats. In the present study, the integrated analysis of transcriptomics and metabolomics was performed to investigate the transcriptional and metabolic profiles of the clams in response to abrupt salinity decrease. The comparative transcriptome analysis uncovers the changes of genes involved in ion channel activation, amino acid metabolism, and energy metabolism in the salinity-stressed condition. The metabolomics analysis reveals that the glucose-succinate pathway may play a critical role in energy metabolism during the osmotic stress. The multi-omics analysis reveals that neural adaptation to salinity stress may be potentially regulated by arachidonic acid metabolism and glutathione metabolism. The important metabolites (12-HPETE, PGEs, and spermine) may be served as the signaling molecules in response to acute salinity stress in the intertidal shellfish. These findings suggest that clams may get through the osmotic stress by ion channel activation, neural adaptation, and anaerobic metabolism to maintain their energy homeostasis for maximum survival. The present study provides great insights into the molecular and metabolic strategies of intertidal shellfish in response to salinity stress, and uncovers the new potential signaling metabolites to assess the health states of molluscs in aquaculture.

Abstract:In recent years, saline-alkaline aquaculture development has become an important measure for China to expand its fishery development space to ensure food safety. Previous studies have verified that salinity and alkalinity positively influence the quality of Chinese mitten crabs (Eriocheir sinensis). However, the regulatory mechanism of E. sinensis endures saline-alkaline stress which remains obscure. This study investigated the metabolic changes in puberty-molting E. sinensis gills exposed to freshwater (FW), sodium chloride salinity of 5 ppt (SW), and carbonate alkalinity 10.00 mmol/L (AW) for 50 days using untargeted liquid chromatography-mass spectrometry metabolomics (LC-MS). A total of 5802 (positive-ion mode) and 6520 (negative-ion mode) peaks were extracted by LC-MS, respectively. A total of 188 (50 upregulated and 138 downregulated), 141 (94 upregulated and 47 downregulated), and 130 (87 upregulated and 43 downregulated) significantly regulated metabolites (SRMs) were observed in the FW-SW, FW-AW, and SW-AW treatments, respectively, wherein 42 generic SRMs were also found by Venn diagram analysis. Seven of the top 10 SRMs with the highest (variable importance in projection) VIP values were similarly identified in FW-SW and SW-AW. Integrated analysis of key metabolic pathways revealed glycerophospholipid, choline in cancer, phenylalanine, and butanoate metabolism. Overall, significant differences were observed in the metabolites and key metabolic pathways of E. sinensis gill exposed to salinity and alkalinity stress. These results will be helpful in understanding the environmental adaptability of aquatic crustaceans to saline-alkaline water.
Effect of salinity on the physiological response and transcriptome of spotted seabass (Lateolabrax maculatus)

Abstract:Salinity fluctuations significantly impact the reproduction, growth, development, as well as physiological and metabolic activities of fish. To explore the osmoregulation mechanism of aquatic organisms acclimating to salinity stress, the physiological and transcriptomic characteristics of spotted seabass (Lateolabrax maculatus) in response to varying salinity gradients were investigated. In this study, different salinity stress exerted inhibitory effects on lipase activity, while the impact on amylase activity was not statistically significant. Notably, a moderate increase in salinity (24 psu) demonstrated the potential to enhance the efficient utilization of proteins by spotted seabass. Both Na+/K+-ATPase and malondialdehyde showed a fluctuating trend of increasing and then decreasing, peaking at 72 h. Transcriptomic analysis revealed that most differentially expressed genes were involved in energy metabolism, signal transduction, the immune response, and osmoregulation. These results will provide insights into the molecular mechanisms of salinity adaptation and contribute to sustainable development of the global aquaculture industry.

Abstract:Salinity is an important factor affecting the survival, growth, and metabolism of marine crustaceans. Low-salt stress will result in the death of swimming crabs. This paper investigates the metabolic response in the gills of Portunus trituberculatus under short-term low-salt stress by comparing the metabolic molecules in the four salinity treatment groups (24‰, 16‰, 12‰, and 8‰) by GC-MS technique. In this study, nine common differential metabolites such as pyruvate, malic acid, and phosphoethanolamine were found in the gill tissues of crabs. KEGG enrichment analysis revealed that six metabolic pathways, including the citric acid cycle, pyruvate metabolism, and the HIF-1 signaling system, were significantly impacted by low salt stimulation. According to the findings, salinity 12‰ is a critical node in crab adaptation to low salinity. In the process of adaptation to short-term low-salinity environment, amino acids participated in osmotic regulation, and organic acids such as pyruvate and malic acid were involved in energy metabolism to ensure their energy supply. This research further enriched the theory of osmotic regulation and metabolic mechanism of adaptation to low salt in crustaceans, with the goal of providing guidance for the improvement of culture technique in Portunus trituberculatus.

Abstract:Salinity, a key limiting factor, affects the distribution and survival of marine species. The Hong Kong oyster ( Crassostrea hongkongensis ), a euryhaline species found along the coast of the South China Sea, has become a major aquaculture bivalve species. To determine the molecular mechanism by which oysters respond to coastal waters with varying salinity levels, we used RNA-seq to sequence the gill samples of oysters exposed to normal (25 ‰, S25), low (5 ‰, S5) and high (35 ‰, S35) salinity conditions for one month. The results revealed different expression transcriptome levels among oysters living under low and high salinity conditions. Using high-throughput sequencing, we identified 811 up-regulated genes and 769 down-regulated genes. As determined by KEGG pathway mapping, the differentially expressed genes (DEGs) were significantly enriched in the prion diseases, histidine metabolism, arginine and proline metabolism, and beta-alanine metabolism pathways in both the S5 vs. S25 and S35 vs. S25 group comparison. Several DEGs including heat shock 70 kDa protein 12B-like, poly (ADP-ribose) polymerase ( PARP ), and tripartite motif-containing protein 2 ( TRIM 2), and low-density lipoprotein receptor-like, as well as KEGG pathways, including arginine and proline metabolism, apoptosis, PPAR signaling pathway, the thyroid hormone signaling pathway, were concerning response to salinity stress. Additionally, eight DEGs involved in salinity adaptation were selected for RT-qPCR validation, and the results confirmed the credibility of the transcriptome sequencing data. Overall, we designed a one-month, medium-term experiment to examine the responses of C. hongkongensis exposed to different levels of salinity stress and performed transcriptome analysis using high-throughput sequencing. Our results enhance current understanding of the molecular mechanisms of salinity stress responses in C. hongkongensis and provided insights into the osmotic biology of oysters.

Abstract:The life history of the Chinese mitten crab (Eriocheir japonica sinensis) includes two migrations: a feeding migration and a reproductive migration. Ambient salinity is one of the most critical factors during migration. In this study, the salinity adaptation mechanism of Chinese mitten crabs was simulated using continuous salinity changes. The expression of six key genes [Na+/K+-ATPase α subunit (NAK-α), V-type H+-ATPase subunit A (VHA-A), Zinc transporter (ZnT), Cl- channel protein 2 (CLCN2), ubiquitin/ribosomal S27 fusionprotein (S27), and glutathione S-transferase (GST)] and the activities of three enzymes [Na+/K+-ATPase (NAK), V-type H+-ATPase (VHA), and glutathione S-transferase (GST)] were evaluated in ten groups exposed to a range of salinity changes during mariculture based on the transcriptome data obtained from short term salinity-induced crabs (ES) compared to control group in freshwater crabs (EF). The results revealed that different genes exhibited different roles in physiological regulation. In total, 3,599 unigenes were significantly and differentially expressed in a comparison between the EF and ES treatments A novel modulation of gene expression and the corresponding enzyme activity of NAK and VHA exhibited similar patterns. As genes related to osmoregulation, NAK and VHA showed similar patterns of both gene expression and enzyme activity in mariculture. During the gradual change in salinity from 0‰ to 25‰ and back to 0‰, the gene expression and enzyme activities of NAK and VHA initially increased (0‰→10‰), weakened (10‰→20‰) and then increased again (20‰→25‰→0‰). S27 could serve as a reference gene in the expression analysis of Chinese mitten crabs under salinity stress. ZnT and CLCN2 were involved in osmoregulation as functional proteins. Our findings provide insights into the regulation mechanisms employed during the migration of the Chinese mitten crab.

Abstract:Background: Salinity is one of the main influencing factors in the culture environment and is extremely important for the survival, growth, development and reproduction of aquatic animals. Methods: In this study, a comparative transcriptome analysis (maintained for 45 days in three different salinities, 30 psu (HC group), 18 psu (MC group) and 3 psu (LC group)) was performed by high-throughput sequencing of economically cultured Penaeus monodon . P. monodon gill tissues from each treatment were collected for RNA-seq analysis to identify potential genes and pathways in response to low salinity stress. Results: A total of 64,475 unigenes were annotated in this study. There were 1,140 upregulated genes and 1,531 downregulated genes observed in the LC vs. HC group and 1,000 upregulated genes and 1,062 downregulated genes observed in the MC vs. HC group. In the LC vs. HC group, 583 DEGs significantly mapped to 37 signaling pathways, such as the NOD-like receptor signaling pathway, Toll-like receptor signaling pathway, and PI3K-Akt signaling pathway; in the MC vs. HC group, 444 DEGs significantly mapped to 28 signaling pathways, such as the MAPK signaling pathway, Hippo signaling pathway and calcium signaling pathway. These pathways were significantly associated mainly with signal transduction, immunity and metabolism. Conclusions: These results suggest that low salinity stress may affect regulatory mechanisms such as metabolism, immunity, and signal transduction in addition to osmolarity in P. monodon . The greater the difference in salinity, the more significant the difference in genes. This study provides some guidance for understanding the low-salt domestication culture of P. monodon .

Abstract:Salinity is an important ecological factor affecting the osmolality of aquatic animals. Solenaia oleivora is an endemic and economically important freshwater shellfish in China. However, its osmotic response and osmoregulatory mechanisms under high salinity stress are still unclear. In this study, S. oleivora was exposed to saline water (salinity: 2.2‰) for 3 h, 6 h, 12 h, 24 h, and 48 h, and then the changes in osmolality, ion concentrations, free amino acid (FAA) content, Na+/K+-ATPase (NKA) activity, and gill histology were analyzed. The hemolymph osmolality increased from 3 h after salinity stress and stabilized between 24–48 h. Na+ in the hemolymph increased from 24 h after salinity stress, and Cl− increased from 3 h. The content of total FAAs in the hemolymph increased after salinity stress. The content of alanine, glycine, glutamine, proline, and other FAAs increased after salinity stress. NKA activity in the gill, hepatopancreases, adductor muscle, and axe foot decreased during 3–48 h of salinity stress. The gill filament space increased and the number of gill cilia decreased after salinity stress. Principal component analysis (PCA) showed that the first two principal components (PC1 and PC2) cumulatively explained 77.6% of the total variation. The NKA activity was positively associated with PC1, while the ion concentration and most FAAs were negatively associated with PC1. Thus, these results indicated that S. oleivora is an osmoconformer, and inorganic ions, FAA, NKA, and gill structure changes play an important role in its osmoregulation.
