Abstract:In this paper, a new type of high performance packed column, the submicroparticle packed column (SPPC), has been proposed. SPPC has the advantages of a microparticle packed column, high efficiency and high analytical speed, and a classical packed column, high column capacity, low cost, and easy operation. In some respects, its properties are similar to that of a capillary, which can analyze high boiling point components or wide boiling range samples at a relatively low column temperature. The height equivalent to an effective plate can be as low as 0.155 mm, and the plate number per second is 95. Moreover, the extra-column effect of the chromatographic system and its diminutiveness and the influences on column efficiency (such as average carrier gas velocity, make-up gas flow, and temperature) are also discussed in detail. SPPC is suitable for fast and trace analysis in many fields.

Abstract:Monolithic capillary columns based on polyacrylamides with UV initiated technology were made,and the length and boundaries of the column bed were made precisely through adjustment of UV radiation.β-cyclodextrin(β-CD) was bonded to the porous polymers of columns,which served as chiral selector.FITC labeled chiral amino acids,which had important impacts on metabonomics in vivo,were separated and detected by laser induced fluorescence(LIF) detection using liquid chromatography mode under an alkaline condition.Theoretical number of the column was about 125 000 plates/m.The influence of the mobile phase on the chromatographic performances was analyzed.
Multi-particle Frits for Packed Capillary Columns in Electrochromatographic Use

Abstract:Further development of capillary electrochromatography (CEC) is to some extent restricted by column technology. Packed capillary columns are most commonly used in CEC because of higher sample capacity and higher sensitivity than open tubular columns, and better reproducibility and uniformity than monolithic columns. Fritting is the core component for the fabrication of packed capillary columns. Therefore various fritting technologies attract continuous attention from analytical community. A convenient fritting method was developed in this study. In contrast to the reported single-particle fritting, this method applies a multi-particle fritting technology to hold stationary phase in capillary columns. Several large porous particles smaller than the column internal diameter were tapped in and immobilized in inlet and outlet of the packing material bed due to direct and indirect keystone effect. Except the smallest permeability by the sintered frit and the lowest breaking pressure by the single-particle frit, EOF mobility and peak dispersion of the multi-particle frit were at the same levels with the formers. In addition, all the three frits avoided bubble formation. It demonstrated that the multi-particle frits met the general requirements of permeability, mechanical resistance and efficiency for packing capillary microcolumns as well as the sintered frits and the single-particle frits. Thus, peak efficiency around 4.3 μm plate height (approximately 232,560 plates per meter) was obtained by multi-particle fritted capillary columns with electrokinetic packing, compared to 6.5 μm plate height by single-particle fritted columns with slurry pressure packing. In particular, the multi-particle fritting was simpler, faster, and more reproducible than the sintered fritting, and the multi-particle fritting was suitable to pack capillary columns of various capillary bore sizes owing to rich commercial resources of fritting particles of various diameters. Multi-particle fritted columns offered satisfactory run-to-run and column-to-column reproducibilities for retention factor, plate height and peak area (precisions of 4.1-9.3% for 10 replicated columns, and precisions of 0.9-3.1% for 30 replicated trials). They also had consistent CEC performance (differences of 1.1-3.9% between the first and 31 st day) during a lifetime more than one month. Application examples in packing capillary columns of 75 and 100 μm i.d. and CEC analysis of sixteen polycyclic aromatic hydrocarbons prove the diversity and reliability of the multi-particle fritting in column technology.

Abstract:Single-walled carbon nanohorns have received great interest for their unique properties and diverse potential applications. Herein, we demonstrated the feasibility of single-walled carbon nanohorns incorporated poly( styrene-divinylbenzene) monolith as the stationary phase for capillary electrochromatography, which were prepared by one-step in situ copolymerization. Single-walled carbon nanohorns were dispersed in styrene to give a stable and homogeneous suspension. The monolithic column gave effective separation for a wide range of aromatic compounds, which was based on hydrophobicity and p-p electrostatic stacking of single-walled carbon nanohorns. The precisions of migration time and peak area varied in the ranges of 1.4-1.9% for intraday trials and 1.7-3.5% for interday trials, and 3.2-6.7% for intraday trials and 4.1-7.4% for interday trials, and 3.6-7.2% for inter-column trials and 5.2-21.3% for inter-column trials, respectively, indicating the good reproducibility of single-walled carbon nanohorns embedded monolithic columns.

Abstract:The preparation of a novel carbon monolithic column for high performance liquid chromatography is described. A phenolic resin rod with embedded 10-microm silica beads was prepared by acid-catalyzed polymerization of a resorcinol/iron(III) complex and formaldehyde. This rod was carbonized and graphitized under inert atmosphere with a programmed temperature cycle from room temperature to 1250 degrees C. Subsequently, the silica beads along with iron catalysts were removed, leaving a porous carbon rod. Imaging of this monolithic rod by scanning and transmission electron microscopies revealed a highly interconnected bimodal porous structure. The porosity and pore size distribution of the mesopores were characterized by N2 absorption/desorption. Graphene sheets were found in the TEM images of the carbon rod, and the graphite index was characterized by Raman spectrum and X-ray diffraction. A monolithic column prepared with the aforementioned carbon rod was evaluated using a mixture of alkylbenzenes. It exhibited an excellent separation power and a low hydraulic resistance. The bundle-of-capillaries model was used to characterize the hydrodynamics of this monolith. Its permeability was found to agree well with the theoretical one.

Abstract:A 3-mm length of a porous monolithic polymer was prepared in a 0.32-mm inner-diameter fused-silica capillary by an in-situ thermo-polymerization method and used as an on-column frit for a packed capillary HPLC column. The on-column frit can resist high pressure up to 400 bar. A 5-μm packing material was packed in the capillary with the on-column frit by a slurry method. At pressure driving mode, separation of samples was performed using the capillary HPLC column. The in-situ frit preparation method has the advantages of easy preparation, easy control of the location of the frit and a mild preparing reaction condition.

Abstract:Graphene oxide functionalized silica microspheres (GO@SiO2) were synthesized based on condensation reaction between amino from aminosilica particles and carboxyl groups from GO. Reduction of GO@SiO2 with hydrazinium hydroxide generated graphene modified silica particles (G@SiO2). GO@SiO2 and G@SiO2 packed capillary columns for capillary electrochromatography were thereafter fabricated by pressure slurry packing with single‐particle frits. GO of 0.3 mg/mL in dispersion solution for GO@SiO2 synthesis was considered as a compromise between retaining and column efficiency whereas GO@SiO2 of 20 mg/mL in slurries for column packing was chosen for a homogenous and tight bed. Optimum mobile phases were acquired considering both electroosmotic flow and resolution at an applied voltage of −6 kV as the following: acetonitrile/phosphate buffer (10 mM, pH 7.0), 75:25 (v/v) for polycyclic aromatic hydrocarbons and 50:50 (v/v) for aromatic compounds. A comparison was made between electrochromatographic performances for three PAHs (naphthalene, fluorene and phenanthrene) and three aromatic compounds of various polarities (toluene, aniline and phenol) on bare aminosilica, GO@SiO2 and G@SiO2 packed columns, which proved the contribution of alone or combinational actions of solvophobic effect and π‐π electron stacking as well as hydrogen bonds to retaining behaviors by GO@SiO2 and G@SiO2. Well over‐run, over‐day and over‐column precisions (retention time: 0.3–1.4, 1.1–3.8 and 2.8–5.2%, respectively; peak area: 2.6–6.5, 4.8–8.3 and 6.5–12.6%, respectively) of GO@SiO2 packed columns were a powerful proof for good reproducibility. Analytical characteristics of GO@SiO2 packed capillary columns in CEC analysis of fresh water were evaluated with respect to linearity (R2 = 0.9961–0.9989) over the range 0.1 to 100 mg/L and detection limits of 9.5 for naphthalene, 12.6 for fluorene and 16.2 μg/L for phenanthrene. Further application to fresh water increased the visibility of the proposed material, where good spike recoveries in the range 89–96% were offered.

Abstract:A novel inorganic silica-organic polymer mixed-based capillary electrochromatography monolithic column was prepared by using precursor 3-(trimethoxysilyl) propyl methacrylate with double-functional group hydrolysis/polycondensation in acidic aqueous solution, followed by double-bond co-polymerization with organic monomer hexadecyl mathacrylate under the thermal initiated condition, and the combination of preparation of silica-based monolithic column and organic polymer monolithic column was suggested. A simple operation of controlling sol mixture using mercury infusion when the mixture infused into capillary was raised to acquire smooth interface of stationary phase. The preparation conditions of monolithic column were optimized. The structure of the monolithic columns was characterized by SEM images and nitrogen absorption method, and the results showed that the median pore diameter of the materials was in the range of 3 to 7 nm with average surface area of 103.4 M2 g−1. The influence of buffer concentration, pH value, and organic modifier content on electroosmotic flow (EOF) velocity has been investigated. Seven neutral aromatic hydrocarbons were baseline separated with column efficiency of naphthalene up to 54000 plates m−1. The reproducibility for separation time and retention factor was given in terms of RSD values at 0.9% and 0.6% (n = 5) for run-to-run injections and 1.2% and 0.8% (n = 3) for day-to-day operations. The RSD values of separation time and capacity factor of different columns (n = 3) were 8.7 and 5.7%, respectively. The studies on performance of columns indicated that the columns possess typical reverse retention mechanism.

Abstract:In this work, GO bonded monolith (pAS-GO@PS-DVB) as the stationary phase for capillary electrochromatography was fabricated, which was achieved by a simple one-step in-situ copolymerization of styrene and vinylized GO in the presence of divinylbenzene as a cross-linker. GO functionalization was primarily completed using p-aminostyrene based on condensation reaction between amino and carboxyl groups. The characterization by infrared spectroscopy, X-ray photoelectron spectroscopy, X-ray diffraction and scanning electron microscopy proved the covalent bonding of GO on the monolith. The average pore diameter via Barrett-Joyner-Halenda, specific surface area and pore volume via Brunauer-Emmett-Teller equation by nitrogen adsorption/desorption were determined to be 112.4 nm, 485.8 m(2) g(-1) and 1.4 cm(3) g(-1), respectively. The pAS-GO@PS-DVB monolithic column gave effective separation for a wide range of aromatic compounds, which was based on hydrogen bonding and pi-pi interactions of GO with polar and/or non-polar organic compounds. The reproducibility in terms of the precisions of migration time, peak height and peak area was estimated below 6% using thiourea and other aromatic compounds. Furthermore, the differences of migration time, peak height and peak area between the first-week analysis and the forth-week analysis were less than 19%, indicating good stability of the proposed monolithic column in one month. The applicability of the pAS-GO@PS-DVB monolith was also demonstrated by baseline separation of three phenols and three anilines. (C) 2016 Elsevier B.V. All rights reserved.

Abstract:A new type of monolithic trapping columns with high mechanical strength was prepared by thin-layer sol-gel coating method and applied to trapping intact proteins for on-line capillary liquid chromatography. Monolithic trapping columns were fabricated by entrapping C8 reversed-phase particles into the capillary columns through a sol-gel network, which was formed by hydrolysis and polycondensation of methyltriethoxysilane. Hundreds times of trapping/untrapping for intact proteins were carried out. The trapping columns showed long-term stability up to 300 bar. Recovery, loading capacity and reproducibility of trapping columns were evaluated using four proteins. The recovery of four protein mixtures for the C8 monolithic trapping columns was 99.3% on average. The loading capacity of 5 mm × 320 μm i.d. C8 trapping columns for the protein mixtures was 30 μg. Day-to-day relative standard deviation (RSD) values for recoveries of protein mixtures on the same C8 trapping column ranged from 2.34 to 5.87%, column-to-column RSD values were from 3.01 to 6.81%. The C8 trapping columns were used to trap normal mouse liver intact proteins in a capillary liquid chromatography system. Results demonstrated high efficiency of the monolithic trapping columns for trapping intact proteins for proteomic analysis in on-line capillary liquid chromatography system.

Abstract:Semi-packed columns are microfabricated gas chromatography columns which have a large surface area and high aspect ratio. In this paper, a new semi-packed column with high-density elliptic cylindrical posts (SCHECP) made by a micro-electro-mechanical system (MEMS) technique was reported. Compared to a semi-packed column with cylindrical posts (SCCP) under the same effective width, the surface area and aspect ratio of SCHECP were improved by 71.19% and 76.47%, respectively. To compare the performance of these two semi-packed columns, SCHECP and SCCP were fabricated. A 10-nm thick alumina film was deposited as the stationary phase by atomic layer deposition technique to ensure the uniformity and repeatability of the stationary-phase film. A contrast experiment was conducted, and the results showed that compared with SCCP, better separation performance was realized in SCHECP due to the increase in surface area and aspect ratio. The number of theoretical plates of nonane was increased by 541.84%, and the tailing factor was decreased by 54.31%.

Abstract:Columns for open tubular capillary electrochromatography, coated with a mixed-mode (RP/ion-exchange) stationary phase, were prepared by using the sol-gel method. The synthetic procedure was optimized by changing the ratios of tetraethoxysilane, octyltriethoxysilane, and 3-aminopropyltriethoxysilane in the initial sol. SEM studies reveal that a coating with about 400 nm thickness can be obtained. The inner surface properties of these capillaries were probed by measuring the EOF as a function of pH. The surface of this stationary phase contains octyl, amine, and residual silanol moieties; the amine and silanol groups determine the net charge on the inner surface of the capillary and can produce a switchable EOF (anodal/cathodal). The performances of the columns were evaluated by open tubular capillary electrochromatography using a wide range of compounds (polycyclic aromatic hydrocarbons, aromatic acids, and aromatic amines).

Abstract:Newly developed porous polymer monolithic capillary columns modified with gold nanoparticles coated with exchangeable functionalities allow easy switching of separation modes by a simple ligand exchange process. These columns are prepared from a poly(glycidyl methacrylate-co-ethylene dimethacrylate) monolith through reaction of its epoxide moieties with cysteamine to afford a monolith rich in surface thiol groups. Gold nanoparticles prepared via in situ reduction of chloroauric acid within the column become attached to the surface of the pores of the monolith. Alternatively, a solution of colloidal gold nanoparticles can be pumped through the thiol modified column to achieve their attachment. While the first approach is faster, it affords a lower coverage of nanoparticles than the second method, while both methods preserve the excellent hydrodynamic properties that are typical of the monolithic columns. Functionalization of the surface of the bound gold nanoparticles is then carried out using low molecular weight thiol-containing surface ligands. The dynamic nature of the bond between gold and these surface ligands enables the replacement of one surface ligand by another through a simple solution exchange process. This novel approach expands the application range of monoliths as a single column can now be used in different separations modes. Applications of the columns with exchangeable chemistries are demonstrated with the capillary electrochromatographic separation of peptides and the nano-high-pressure liquid chromatography (HPLC) separation of proteins in both reversed phase and ion exchange modes.

Abstract:Packed capillary columns have become the standard front-end separation device for mass spectrometry-based proteomics. The development of simple, fast, and robust capillary column technology, especially that with mass-fabrication capacity, can greatly improve analytical throughput and reproducibility in omics research. In this technical note, we report a centrifugal packing technology, which has the capability to mass fabricate high quality capillary columns with a 2886 columns/day fabrication throughput. The centrifugally packed columns presented significantly improved efficiency (reduced plate height hmin = 1.6, 37%-40% improvement compared with slurry packed columns), advanced kinetic performance limit, and excellent column-to-column reproducibility (2.0% RSD for retention time, 50 columns). Such columns enabled ∼5300 HeLa proteins identified in single-shot proteomic analysis, displaying both intercolumn and inter-run retention time stability (retention time RSD = 0.94% between nine replicates on three columns for probing peptide sequence). The mass-fabrication technology reported in this technical note may support disposable use of high quality chromatographic columns in large-scale bioanalysis.
