Abstract:The synthesis of L-ascorbyl flurbiprofenate was achieved by esterification and transesterification in nonaqueous organic medium with Novozym 435 lipase as biocatalyst. The conversion was greatly influenced by the kinds of organic solvents, speed of agitation, catalyst loading amount, reaction time, and molar ratio of acyl donor to L-ascorbic acid. A series of solvents were investigated, and tert-butanol was found to be the most suitable from the standpoint of the substrate solubility and the conversion for both the esterification and transesterification. When flurbiprofen was used as acyl donor, 61.0% of L-ascorbic acid was converted against 46.4% in the presence of flurbiprofen methyl ester. The optimal conversion of L-ascorbic acid was obtained when the initial molar ratio of acyl donor to ascorbic acid was 5 : 1. kinetics parameters were solved by Lineweaver-Burk equation under nonsubstrate inhibition condition. Since transesterification has lower conversion, from the standpoint of productivity and the amount of steps required, esterification is a better method compared to transesterification.
Picogram per liter quantification of pyrethroid and organophosphate insecticides in surface waters: a result of large enrichment with liquid–liquid extraction and gas chromatography coupled to mass spectrometry using atmospheric pressure chemical ionization

Abstract:Insecticides such as pyrethroids and organophosphates are extensively used globally. Once released into surface water bodies, they can pose a major threat to aquatic ecosystems already at trace concentrations. Therefore, selected pyrethroids and organophosphates are listed as priority substances within the European Water Framework Directive with chronic quality criteria in the picogram per liter range. Previously applied analytical methods were unable to detect pyrethroids and organophosphates at ecotoxicological relevant concentrations, thereby hindering the assessment of surface water quality. In this work, we developed an ultra-sensitive method for the analysis of 12 pyrethroid and two organophosphate insecticides in surface waters. This method is based on the liquid–liquid extraction of surface water samples with n-hexane to achieve large enrichment factors (4000×) and subsequent chemical analysis by gas chromatography coupled to tandem mass spectrometry using atmospheric pressure chemical ionization, a soft ionization technique. Quality control parameters including the method limits of quantification (12.5–125 pg L−1), intra-day precision (1–22%), intra-day accuracy (84–133%), and absolute recoveries covering liquid–liquid extraction (67–114%) showed that the method is sensitive and robust and therefore suitable for the analysis of pyrethroids and organophosphates in surface waters. The developed method was applied to Swiss surface water samples and detected pyrethroids and organophosphates below the ecotoxicological relevant concentrations, exemplifying the suitability of the proposed method for aquatic monitoring.Graphical abstract

Abstract:In this study, a simple, rapid, precise, and environmentally friendly microextraction named dispersive micro-solid phase extraction based on a graphene/polydopamine composite as a sorbent was investigated for the analysis of four pyrethroids (fenpropathrin, cyhalothrin, etofenprox and bifenthrin) in water samples. The graphene/polydopamine composite was successfully synthesized using a one-step method, and was characterized by scanning electron microscopy, transmission electron microscopy, energy-dispersive spectroscopy and X-ray photoelectron spectroscopy. The simplicity and rapidity of dispersive micro-solid phase extraction and the high surface area and adsorptivity of the graphene/polydopamine composite were combined in the proposed method. Several main parameters, including the amount of the sorbent, extraction time, ionic strength and desorption conditions, were independently optimized and the results were compared to find the best extraction setup for dispersive micro-solid phase extraction. Under the optimal conditions, good linearity was observed for all the target analytes, with the coefficient of determination ranging from 0.9997 to 0.9999. The extraction recoveries obtained using the proposed method ranged from 76.81% to 85.29%, and the limits of detection varied from 1.5 to 3 μg L-1. In addition, the relative standard deviation values for the intra-day precision were between 0.41% and 3.00%, while the inter-day precision showed relative standard deviation values ranging from 1.61% to 5.59%. Overall, the figures of merit of the entire procedure showed that this technique could produce satisfactory results in the detection of pyrethroids in water samples or other organic pollutants in the future.
Positive AMPA Receptor Modulation Rapidly Stimulates BDNF Release and Increases Dendritic mRNA Translation

Abstract:Brain-derived neurotrophic factor (BDNF) stimulates local dendritic mRNA translation and is involved in formation and consolidation of memory. 2H,3H,6aH-pyrrolidino[2″,1″-3′,2′]1,3-oxazino[6′,5′-5,4]-benzo[e]1,4-dioxan-10-one (CX614), one of the best-studied positive AMPA receptor modulators (also known as ampakines), increases BDNF mRNA and protein and facilitates long-term potentiation (LTP) induction. Several other ampakines also improve performance in various behavioral and learning tasks. Since local dendritic protein synthesis has been implicated in LTP stabilization and in memory consolidation, this study investigated whether CX614 could influence synaptic plasticity by upregulating dendritic protein translation. CX614 treatment of primary neuronal cultures and acute hippocampal slices rapidly activated the translation machinery and increased local dendritic protein synthesis. CX614-induced activation of translation was blocked by K252a [(9S,10R,12R)-2,3,9,10,11,12-hexahydro-10-hydroxy-9-methyl-1-oxo-9,12-epoxy-1H-diindolo[1,2,3-fg:3′,2′,1′-kl]pyrrolo[3,4-i][1,6]benzodiazocine-10-carboxylic acid methyl ester], CNQX, APV, and TTX, and was inhibited in the presence of an extracellular BDNF scavenger, TrkB-Fc. The acute effect of CX614 on translation was mediated by increased BDNF release as demonstrated with a BDNF scavenging assay using TrkB-Fc during CX614 treatment of cultured primary neurons and was blocked by nifedipine, ryanodine, and lack of extracellular Ca2+ in acute hippocampal slices. Finally, CX614, like BDNF, rapidly increased dendritic translation of an exogenous translation reporter. Together, our results demonstrate that positive modulation of AMPA receptors rapidly stimulates dendritic translation, an effect mediated by BDNF secretion and TrkB receptor activation. They also suggest that increased BDNF secretion and stimulation of local protein synthesis contribute to the effects of ampakines on synaptic plasticity.

Abstract:An ultrasound/microwave-assisted solid-liquid-solid dispersive extraction method was developed for the determination of 16 pyrethroid insecticides in Dendrobium officinale. The extraction and purification were performed in one step. The pyrethroids were extracted with 8: 2 acetone: hexane and purified with a mixture of primary secondary amine, Florisil, C18, and graphitized carbon black. The extraction temperature, extraction time, and ultrasound power were optimized. Trans-cyfluthrin-d(6) was used as the internal standard to improve the accuracy. Under the optimized conditions, the recoveries of the pyrethroids were from 72.2 to 113.9% with the limits of detection between 0.29 and 9.38 mu g kg(-1). The optimized method was successfully used for the determination of pyrethroids in commercial D. officinale samples and may allow monitoring pyrethroids in herbal products.
Simultaneous Determination of Seven Pyrethroid Pesticide Residues in Aquatic Products by Gas Chromatography

Abstract:Aquatic products are good sources of essential nutrients, but the presence of pyrethroids (PYRs) as contaminants may pose risks to consumer health as the intricate matrices of PYRs usually obstruct chemical hazard detection. In this study, a gas chromatographic method was established and validated for simultaneously detecting residues of seven PYRs (cyhalothrin, permethrin, cyfluthrin, cypermethrin, tau-fluvalinate, fenvalerate, deltamethrin) in aquatic products. The aim of this method was to assess better the potential risks of pesticide residues in aquatic products. The PYRs in aquatic products were extracted with acetonitrile, purified with n-hexane saturated with acetonitrile, and cleaned up on a Cleanert® Alumina N column. Statistical analysis and orthogonal array experimental design were used to optimize the key parameters. To validate the proposed method, commonly consumed aquatic products (such as carp, crucian carp, whiteleg shrimp, river crab, sea cucumber, and scallop) were obtained from local supermarkets in Shanghai. Satisfied linearity of the calibration curves was achieved in a matrix-matched standard solution, with a correlation coefficient (R2) larger than 0.995. The average recoveries at five fortification levels varied from 77.0% to 117.2%, with relative standard deviations (RSDs) below 11.1%. Concerning electron capture detection, the limits of detection (LOD) and limits of quantification (LOQ) were 2.0–5.0 μg/kg and 5.0–10.0 μg/kg, respectively. These results demonstrate the high stability and sensitivity of this method for simultaneously detecting PYRs in aquatic products, having great practicability and which can be popularized easily.

Abstract:Dispersive liquid–liquid microextraction (DLLME) based on Fe3O4 magnetic nanoparticle-assisted in situ ionic liquid was developed as a novel method for the pretreatment of pyrethroid pesticides (flumethrin, bifenthrin and silafluofen) in urine samples. [N4,4,4,4]Cl and Na[N(CN)2] formed a ionic liquid ([N4,4,4,4][N(CN)2]), which provides an alternative to highly toxic solvents; moreover, high extraction recoveries and enrichment factors were achieved. The extraction parameters affecting the extraction recoveries were investigated. Under optimum conditions, the analytical method showed a good linear response, with correlation coefficient (R2) values ranging from 0.9994 to 0.9996. The enrichment factor, limit of detection and recovery were 108–125, 2.7–5.0 μg L−1 and 88.0–101.1%, respectively. Therefore, this green and simple method is suitable for the microextraction of pyrethroid pesticides in urine samples. It is valuable to establish a sensitive method for monitoring trace levels of pyrethroids, which could contribute to the use of rapid and green sample preparation procedures in urine samples.

Abstract:A fast and efficient method for the determination of trace level of carbamate pesticides using a lower-density-than-water solvent for ultrasound-assisted emulsification microextraction coupled to on-column derivatization and analysis by GC-MS has been developed and studied. In this approach, a soft plastic Pasteur pipette was employed as a convenient extraction device. Fifty microliters of extraction solvent, of lower density than water, was injected into the sample solution held in the pipette. The latter was immediately immersed in an ultrasound water bath to form an emulsion. After 2 min extraction, the emulsion was fractionated into two layers by centrifugation. The upper layer (organic extract) could be collected conveniently by squeezing the bulb of the pipette, now held upside down, to move it into the narrow stem of the device, facilitating its retrieval for analysis. The extract was then combined with trimethylphenylammonium hydroxide and directly injected into a gas chromatography-mass spectrometry (GC-MS) system for on-column derivatization and analysis. The on-column derivatization provided an added convenience (since a separate step was not necessary). Parameters affecting the derivatization and extraction were investigated. Under the most favorable conditions, the method demonstrated high extraction efficiency with low limits of detection of between 0.01 and 0.1 μg/L, good linearity in the range of 0.05-50 μg/L, to 0.5-100 μg/L, and good repeatability (RSD below 9.2%, n=5). The proposed method was evaluated by determining carbamate pesticides in river water samples.
Development of an enzyme-linked immunosorbent assay for the pyrethroid cypermethrin

Abstract:A competitive enzyme-linked immunosorbent assay (ELISA) for the detection of cypermethrin was developed. Two haptens, the trans- and cis-isomers of 3-[(+/-)-cyano-3-(2,2-dichloroethenyl)-2,2-dimethylcyclopropanecarbonyloxy]methyl]phenoxyacetic acid, were conjugated with thyroglobulin as immunogens. Four antisera were generated and screened against six different coating antigens. The assay that was the most sensitive for cypermethrin was optimized and characterized. The IC(50) for cypermethrin was 13.5 +/- 4.3 microg/L, and the lower detection limit (LDL) was 1.3 +/- 0.5 microg/L. This ELISA had relatively low cross-reactivities with other major pyrethroids, such as deltamethrin, phenothrin, resmethrin, fluvalinate, and permethrin. Methanol was found to be the best organic cosolvent for this ELISA, with an optimal sensitivity observed at a concentration of 40% (v/v). The assay parameters were unchanged at pH values between 5.0 and 8.0, whereas higher ionic strengths strongly suppressed the absorbances. To increase the sensitivity of the overall method, a C(18) sorbent-based solid-phase extraction was applied to various domestic and environmental water samples. The water samples, fortified with cypermethrin, were analyzed according to this method. Good recoveries and correlation with spike levels were observed.

Abstract:Ultrasound-assisted magnetic nanofluid-based liquid-phase microextraction was coupled to GC-MS in a method for simultaneous determination of the pyrethroid insecticides permethrin, deltamethrin, and cypermethrin. A highly efficient extraction solvent called "magnetic nanofluid (MNF)" is introduced for preconcentration of pyrethroids. The MNF consists of magnetic multiwalled carbon nanotubes (MMWCNTs) and deep eutectic solvent. Following microextraction, the MNF was detached by an external magnet from the medium without the need for centrifugation. 2-Pyridinecarboxamide, choline chloride, and anhydrous ferric chloride were used for the synthesis of the deep eutectic solvent. The effects of pH value, volume of MNF, sonication time, sample volume, and ionic strength, type and amount of back extraction solvent were investigated. Under optimum conditions of MNF volume of 50 μL, time of 5 min, 100 μL acetone as back extraction solvent, NaCl concentration of 1 mol.L−1 and sample volume of 30 mL, the detection limits are 2.8, 2.7 and 2.0 ng·mL−1 for permethrin, deltamethrin and cypermethrin, respectively. The linear response ranges are between 0.01 and 250 ng·mL−1, and relative standard deviations (for n = 7) are 3.5, 3.2 and 2.8%. Abstract:An analytical method was developed for the simultaneous measurement of 18 pyrethroids (allethrin, bifenthrin, cyfluthrin, cypermethrin, cyphenothrin, deltamethrin, empenthrin, fenpropathrin, furamethrin, imiprothrin, metofluthrin, permethrin, phenothrin, prallethrin, profluthrin, resmethrin, tetramethrin and transfluthrin) in indoor air. The pyrethroids were collected for 24 h using a combination of adsorbents (quartz fiber filter disk and Empore C18 disk), with protection from light, and then extracted with acetone, concentrated, and analyzed by GC/MS. They could be determined accurately and precisely (detection limits: ca. 1 ng/m(3)). The collected pyrethroid samples could be stored for up to one month at 4 degrees C in a refrigerator.

Abstract:Abstract Ion mobility spectrometry with corona discharge ion source was used for the measurements of selected pesticides cybutryne, terbutryn, dichlorvos, and quinoxyfen in water. The analyses were carried out by (i) direct injection of the aqueous solution of the pesticides and (ii) extraction using solid phase microextraction by commercial SPME arrows. The instrumental and the experimental extraction parameters including the drift tube temperature, injection port temperature, and the SPME fiber type were studied and optimized to achieve the maximum signal intensity. The limit of detections obtained by the direct injection were about 0.1–1.0 mg L −1 . Using the SPME arrow-ion mobility spectrometry method, the detection limits were improved 7–50 times and obtained as 0.01, 0.015, 0.05, and 0.1 mg L −1 for terbutryn, dichlorvos, cybutryne, and quinoxyfen, respectively. The recovery rate for analysis in surface water (real samples) was approximately 90–110% for both the direct injection and solid phase microextraction method. The capability of ion mobility spectrometry for the simultaneous detection of a mixture of the studied pesticides was assessed, and it was found that terbutryn, dichlorvos, and quinoxyfen can be detected simultaneously without peak overlapping.

Abstract:Phenoxy acid herbicides are widely used because of their excellent efficiency and low cost. However, owing to their strong polarity and water solubility, these herbicides do not degrade easily in a water environment and persist for a long time in water bodies. These herbicides readily enter water bodies via surface runoff, infiltration, and other migration routes, thus affecting water quality safety. Therefore, it is of great significance to establish a sensitive and simple method for the quantitative analysis of phenoxy acid herbicides in environmental water. Given the low concentration of such contaminants in environmental water, appropriate detection methods are important. Ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) has high sensitivity and accuracy, thus being well suited for the phenoxy acid herbicides analysis. Sample preparation techniques are also important for the extraction and enrichment of contaminants in environmental water. Dispersive solid phase extraction (DSPE) has attracted considerable attention owing to its low cost, ease of operation, and low solvent consumption. In general, the selectivity and efficiency of solid phase extraction are largely dependent on the characteristics of the solid adsorbent materials. Ionic metal-organic frameworks (iMOFs) have excellent ion-exchange properties and show selective absorptivity to ionic compounds. In this work, a metal-organic framework (MOF) MIL-101-NH2 was synthesized by a facile hydrothermal method. Then, a cationic MOF mixed matrix membrane (MMM) was fabricated by soaking the MOFs in a polyvinylidene fluoride (PVDF) solution and further functionalization with quaternary amine groups. A method was developed for the determination of seven phenoxy acid herbicides in water by UPLC-MS/MS based on DSPE. The prepared material was characterized by Fourier transform infrared spectroscopy and scanning electron microscopy to determine its functional groups and morphology. The results showed that there were quaternary amine groups in the material, and that the functionalization did not have any obvious effect on the chemical and crystal structures of MIL-101-NH2. The prepared MIL-101-$NMe_{3}^{+}$-PVDF MMM was used as an adsorbent for DSPE to enrich the seven phenoxy acid herbicides in water. It is well known that the key factors influencing extraction efficiency are the adsorption and elution conditions. To establish the optimum extraction conditions, the influence of some important factors, including the adsorbent amount, sample pH, extraction time, elution solvent, elution volume, and elution time, was investigated in detail. Gradient elution was carried out with 0.01% formic acid aqueous solution and acetonitrile as the mobile phase. The target analytes were separated on an ACQUITY UPLC BEH C18 column (100 mm×2.1 mm, 1.7 μm), and multiple reaction monitoring (MRM) was conducted in the negative electrospray ionization mode. The external standard method was used for quantitative analysis. The established method was verified in terms of the linear ranges, limits of detection (LODs), limits of quantification (LOQs), recoveries, and precision. Under the optimal conditions, the seven phenoxy acid herbicides showed good linear relationships in their respective concentration ranges, and the correlation coefficients were all higher than 0.997. The LODs and LOQs were 0.00010-0.00090 μg/L and 0.00033-0.00300 μg/L, respectively. The recoveries were tested at three spiked levels of 0.005, 0.05, and 0.2 μg/L. The average recoveries of the seven compounds were in the range of 80% to 102%. The intra-day and inter-day relative standard deviations were within 1.4% to 9.4% and 4.2% to 12.6%, respectively. The established method was applied to the analysis of the phenoxy acid herbicides in tap water and reservoir water. Three levels of spiked samples were adopted to investigate the accuracy of the method. The results demonstrated that our method is applicable to the detection of trace phenoxy acid herbicides in water samples. In summary, this method has the advantages of simplicity, rapidity, and sensitivity, and it is suitable for the detection of the seven phenoxy acid herbicides in environmental water.
Evaluation of Graphene Aerogel Monolith-Based Solid-Phase Extraction for the Separation of Pyrethroids from Water Samples

Abstract:Graphene aerogel monolith was synthesized directly in the solid-phase extraction cartridge through template-free “sol-cryo” method and used for the separation of pyrethroids insecticides from water samples, followed by gas chromatography–mass spectrometry analysis. Graphene sheets, the basic components of graphene aerogel, extracted the analytes from water samples through π–π stacking interaction and hydrophobic interaction. Then the targets were desorbed in ethyl acetate for subsequent analysis. Several parameters such as adsorption property, the type of the elution solvent and elution volume, the loading volume and its pH were investigated to achieve the best solid-phase extraction performance. Low limits of detection in the range of 0.83–9.31 ng L −1 and acceptable precision (RSDs 3.8–11.7%) were achieved under the optimized conditions. The developed method was applied to extract pyrethroids from real water samples with satisfactory recoveries between 76.2 and 106.0%. Abstract:Residues of pyrethroid insecticides tend to accumulate in bed sediments due to their strong hydrophobicity. Rather than the total or bulk sediment concentration, it is the freely dissolved concentration (C-free) that drives toxicity to benthic invertebrates. In this study we developed thin film-based samplers for in situ ambient monitoring of pyrethroids at trace levels in sediment. Out of five common polymer materials, polyethylene (PE) and silicone rubber (SR), were identified to offer superior enrichment for pyrethroids from sediment. To circumvent the slow equilibrium process, C-13-permethrin and bifenthrin-d(5) were preloaded onto the films as performance reference compounds (PRC5). The PRC-preloaded film samplers were deployed at five sites in Southern California under field conditions for 7 d and retrieved for analysis. The sediment porewater C(free )of eight pyrethroids derived from PRC-PE films ranged from 173 to 903 ng/L accounting for 18.2-36.1% of the corresponding total porewater concentrations. The PRC-SR film samplers yielded C-free values closely mimicking those from the PRC-PE samplers, cross-validating the two sampling devices. Additionally, a significant positive association was found between the observed mortality from toxicity tests using Hyalella azteca and the C-free of bifenthrin (r = 0.628, p = 0.02). A significant linear correlation (R-2 = 0.99) between C-free derived from in situ monitoring and that of ex situ measurement under equilibrium conditions was also observed. Results from this study demonstrated that the film-based samplers may be used for in situ ambient monitoring to detect biologically relevant contamination of pyrethroids in bed sediments, which may contribute to improved risk assessment for this class of widely used insecticides. Fluorometric assay of cyanophoric pyrethroids based on benzothiazole modified carbon quantum dots

Abstract:A fluorometric assay based on benzothiazole carbon quantum dots (benzothiazole-CDs) was developed for the determination of cyanophoric pyrethroids (CPs, e.g. , cypermethrin, deltamethrin and fenpropathrin). Benzothiazole-CDs were synthesized by amidation and condensation reaction of N, S-CDs based on a hydrothermal method. CPs could be hydrolyzed to generate cyanide ions under alkaline conditions, which reacted with the benzothiazole-CDs and destroyed the big conjugate system of the CDs inducing the quenching of fluorescence. Thus, CPs could be determined based on the fluorescence change. In order to obtain the optimum fluorescence conditions for the assay of CPs, the reaction time, temperature, the pH value of the detection system and the concentration of benzothiazole-CDs were optimized. The results demonstrated that under the optimum conditions, the concentration of CPs showed good linearity with the fluorescence quenching efficiency in the range of 0.01–2 mg L −1 and the correlation coefficient ( R 2 ) was between 0.991 and 0.995. The limits of detection were between 2.7 and 2.8 μg L −1 . The assay was applied to the detection of CPs in real samples including underground water, rice and apples. The recoveries were between 74.2 and 103.5% and the relative standard deviations (RSDs) were between 1.4 and 9.7%.

Pyrethroids, pyrethrins, and piperonyl butoxide in sediments by high-resolution gas chromatography/high-resolution mass spectrometry

Abstract:A new method for determination of pyrethroids, pyrethrins, and piperonyl butoxide (PBO) by high-resolution gas chromatography/high-resolution mass spectrometry (HRGC/HRMS) was developed for aquatic sediments. The method detection limits for pyrethroids (15 compounds), pyrethrins, and PBO ranged from 0.16 to 1.50 ng/g sediment, which was suitable for detecting these chemicals at environmentally relevant concentrations (low ng/L range) that are toxicologically significant to benthic organisms. Recovery of the analytes from a low level spiked sediment ranged from 89.7% to 135%. Resmethrin showed the lowest recovery at 23.5% and pyrethrins showed the highest recovery at 154%. To confirm the utility of this new method for environmental applications, sediment samples collected from five tributaries of the San Francisco Bay, California were analyzed. Individual pyrethroids were detected in concentrations of up to 17.6 ng/g, while PBO was detected in all sediment samples in concentrations of 0.010-0.215 ng/g. Pyrethrins were not found in the sediment samples.
Magnetic adsorbent based on mesoporous silica nanoparticles for magnetic solid phase extraction of pyrethroid pesticides in water samples

Abstract:Mesoporous KIT-6-magnetite composite (Fe3O4@SiO2@KIT-6) was successfully prepared and characterized by transmission electron microscopy, scan electron microscopy, elemental mapping, energy dispersive X-ray spectrum, Fourier transform infrared spectroscopy, thermal gravimetric analysis, X-ray powder diffraction, Brunauer Emmett Teller analysis and magnetic property analysis. It was then applied as adsorbent for magnetic solid phase extraction of fenpropathrin, lambda-cyhalothrin, S-fenvalerate and bifenthrin from water samples before analyzed by high performance liquid chromatography-ultraviolet detector. A few factors impacting extraction were studied. The optimal results were acquired by using acetonitrile to eluent the targets adsorbed on 40 mg of Fe3O4@SiO2@KIT-6 adsorbent in 100 mL water samples at a pH of 7 at 25 ℃ within 10 min. The experimental limit of detections of fenpropathrin, cyhalothrin, S-fenvalerate and bifenthrin are 0.01 × 10-3, 0.006 × 10-3, 0.01 × 10-3 and 0.005 × 10-3 mg∙L-1, respectively. Good relative recoveries (86.58-98.80%) and reproducibility values are also obtained. The kinetic experiment shows that the adsorption follows pseudo-second order, while the isotherm experiment follows Langmuir isotherm model. Trace pyrethroid pesticides in water samples can be detected by the developed magnetic solid phase extraction technique followed by high performance liquid chromatography, which is simple, easy, environmental friendly and efficient.

Abstract:Pesticides are used in the agricultural production process to ensure the yield and quality of agricultural products. However, in recent years, environmental pollution issues caused by pesticide residues have sparked widespread concern in society. It is important to develop convenient and efficient approaches to detect and monitor pesticide residues. In this study, targeting benzoylurea insecticides (BUs), polyamidoamine dendrimer-functionalized silica nanocomposite with polydopamine coating (SiO2-PAMAM-PDA) was designed and successfully synthesized. First, monodisperse silica nanoparticles were prepared by the hydrolysis of tetraethyl orthosilicate (TEOS) in mixed solution of ethanol, water and ammonia. The silane coupling agent (3-aminopropyl)triethoxysilane was then employed to introduce amino groups into the silica. Silica with the zeroth generation of polyamidoamine (PAMAM) modification (SiO2-PAMAM-G0) was obtained through Michael addition reaction of methyl acrylate. Ethylenediamine was added to polymerize with methyl acrylate using an amidation reaction to form the first-generation PAMAM (SiO2-PAMAM-G1). Finally, by polymerizing dopamine under alkaline conditions (pH=8.5), the SiO2-PAMAM-G1 was coated with PDA. Thus, the final product named SiO2-PAMAM-PDA was obtained. The composite was characterized using a transmission electron microscope (TEM) and an increase in surface roughness indicated the successful grafting of PDA coating. Dopamine structure contains abundant benzene rings and amino and hydroxyl groups. It could bind with BUs through multiple secondary interactions, such as hydrogen bond and π-π stacking interaction. Therefore, the introduction of PDA could effectively enhance the affinity of the material toward benzoylurea insecticides. The prepared nanocomposites were used as sorbents in a dispersive micro solid-phase extraction approach (D-μ-SPE). The established approach was employed to extract and enrich the BUs in water samples before high-performance liquid chromatography (HPLC) analysis. Diflubenzuron, triflumuron, hexaflumuron, and teflubenzuron were chosen as target analytes. The following was a typical D-μ-SPE procedure. The prepared adsorbents measuring 40 mg were first dispersed in an 8-mL sample solution containing 150 g/L NaCl. The dispersion was assisted by 120-s vortexing to ensure full contact between the SiO2-PAMAM-PDA and the targets. Next, the adsorbents were separated from the liquid phase by 4-min centrifugation (5000 r/min). Thereafter, the adsorbed benzoylureas were eluted using 1 mL acetonitrile as desorption solvent by 120-s vortexing. Separated by centrifugation, the eluate was dried under a mild nitrogen stream. The solid remains were redissolved in 0.1 mL of acetonitrile, filtered by filter membrane (0.22 μm), and then analyzed by HPLC. The experimental conditions in the D-μ-SPE process could have a great impact on the extraction efficiency. Experimental conditions were optimized using a single factor optimization approach to further enhance the extraction recoveries. The optimized conditions included adsorbent amount, extraction time, desorption solvent type, desorption solvent volume, desorption time, and NaCl addition amount. Under the optimal conditions, a linearity range of 10-500 μg/L and limits of detection (LODs, S/N=3) of 1.1-2.1 μg/L were obtained. The extraction recoveries and relative standard deviations (RSDs) of the four BUs were 82.8%-94.1% and 2.1%-8.0%, respectively. The established approach was compared with reported approaches targeting benzoylurea insecticides. It was discovered that this approach consumed less sample, material, organic solvent, and pretreatment time. It provided a more rapid and green choice for the determination of benzoylurea pesticides. To determine the applicability, the proposed approach was applied to analyze the four benzoylurea insecticides in three river water samples. The real water samples were pretreated using the developed approach ahead of instrumental analysis, and no benzoylurea pesticides residue was detected. Next, standard addition experiments were performed under three spiking levels, including 15, 50, and 200 μg/L. The established approach had good accuracy and feasibility with satisfactory recoveries (69.5%-99.4%) and RSDs (0.2%-9.5%).
