Abstract:BACKGROUND: As a potential source of biomass, Jerusalem artichoke has been studied for bioethanol production; however, thus far it has not been investigated for the production of other liquid biofuels, such as biodiesel. This work aims to develop a novel approach for biodiesel production from Jerusalem artichoke tuber using heterotrophic microalgae.RESULTS: In this study, Chlorella protothecoides utilized hydrolysate of Jerusalem artichoke tuber as carbon source and accumulated lipid in vivo, with lipid content as high as 44% by dry mass, and a carbon source to lipid conversion ratio of about 25% in a 4-day scale cultivation. The lipids were extracted and then converted into biodiesel by transesterification. Cetane acid methyl ester, linoleic acid methyl ester and oleic acid methyl ester were the dominating components of the biodiesel produced. Unsaturated fatty acids methyl ester constituted over 82% of the total biodiesel content.CONCLUSION: This work suggests the feasibility of an alternative method of producing biodiesel from Jerusalem artichoke tuber using microalgae cultivation, and a cost reduction of carbon source feed in algal oil production can be expected. Abstract:Microbial oil is drawing increasing interest worldwide as an alternative non-food oil feedstock for biodiesel industry. Nowadays researchers have been increasingly focused on the improvement of microbial oil production process. Oleaginous yeast Rhodosporidium toruloides (R. toruloides) is considered an important candidate due to its excellent capabilities of lipid accumulation, broad adaptabilities to various carbon substrates, and the potential of co-production of some pigments. In present work, the individual effects of non-ionic, cationic, and anionic surfactant on cell growth and lipid accumulation of R. toruloides were investigated for the first time. Interesting results were noticed when some anionic surfactants were supplemented. The most significant effect was observed with addition of 0.2 % (w/v) sodium lignosulfonate, that biomass concentration, lipid concentration, and lipid yield was increased by 25.1, 44.9, and 15.7 %, respectively. The fatty acid compositions of R. toruloides lipids remained unchanged, which is similar to that of vegetable oils, and is considered potential feedstock for biodiesel preparation.

Abstract:The characteristic of Rhodotorula gutini As2.107 for producing lipid was studied. The biomass was 11 g/L and the lipid concentration reached 4.5 g/L with the optimal fermentation medium containing glucose 5%, (NH4)2SO40.25%, MgSO40.2%, CaCl20.01%, FeCl30.007%. The optimal fermentation temperature, rotation speed and inoculation size were 30°C, 180 rpm and 5% respectively. The acid value, saponification value and the average molecular weight were analyzed as 26.31 mgKOH/g oil, 231.86 mgKOH/g oil and 818.78. The components of the microbial lipid were palmitic acid 17.8%, steric acid 10.27%, oleic acid 53.02%, linoleic acid 5.29%, which was similar with that of vegetable oil. Based on the above results, biodiesel production with the microbial lipid of Rhodotorula gutini As2.107 was investigated. The results indicated that the maximum methyl esters (ME) yield reached 93% by using alkali as the catalyst, with the molar ratio of methanol to oil 20:1, 1.5% KOH and reaction temperature 60°C, after the high acid value of the microbial lipid was first lowed.

Abstract:Rhodosporidium toruloides AS 2.1389 has been considered a promising oleaginous strain due to its flexible substrate adaptability, high lipid content, and co-production of some pigments. In previous work, R. toruloides has shown good potential to directly convert crude glycerol into intracellular lipids. However, the difference in nutritional demand between cell growth and lipid accumulation was found to be a dilemma. Therefore, a glycerol-based two-stage process was proposed in the present work to separately meet the nutritional demand of both the cell proliferation phase and lipid accumulation phase. Factors that affect microbial conversion of glycerol into lipid were investigated, statistically analyzed, and optimized. As a result, 26.5 g L-1 biomass with 10 g L-1 lipid was obtained in the two-stage process. Lipid yield (0.20 g g(-1)) and productivity (0.083 g L-1 h(-1)) achieved were significantly higher than the previously optimized batch culture. In R. toruloides lipids, the dominant fatty acid compositions are palmitic acid (28.5%), stearic acid (12.9%), oleic acid (41.3%), and linoleic acid (12.8%). Phospholipids accounts for 0.63% in total lipid. Lipasecatalyzed methanolysis could achieve up to 95% biodiesel yield. The characterization of R. toruloides lipid suggests its great application potential for biodiesel and specialty-type lipid products.
Efficient Production of L-Lactic Acid from Hydrolysate of Jerusalem Artichoke with Immobilized Cells of Lactococcus Lactis in Fibrous Bed Bioreactors

Abstract:Hydrolysate of Jerusalem artichoke was applied for the production of l-lactic acid by immobilized Lactococcus lactis cells in a fibrous bed bioreactor system. Preliminary experiments had indicated that the high quality hydrolysate, which was derived from the 40 min acid treatment at 95 °C and pH 1.8, was sufficient to support the cell growth and synthesis of l-lactic acid. With the addition of 5 g/l yeast extract, the fermentative performance of free cell system was evidently improved. After the basal settlement of hydrolysate based fermentation, the batch mode and the fed-batch mode fermentation were carried out in the free cell system and the fibrous bed bioreactor system, respectively. In all cases the immobilized cells presented the superior ability to produce l-lactic acid. The comparison of batch mode and fed-batch mode also indicated that the growth-limiting feeding strategy could reduce the lag phase of fermentation process and enhance the production of l-lactic acid. The achieved maximum concentration of l-lactic acid was 142 g/l in the fed-batch mode. Subsequent repeated-batch fermentation of the fibrous bed bioreactor system had further exhibited the persistence and stability of this system for the high production of l-lactic acid in a long term. Our work suggested the great potential of the fibrous bed bioreactor system and hydrolysate of J. artichoke in the economical production of l-lactic acid at industrial scale.

Abstract:Oleaginous microorganisms are receiving significant attention worldwide for their utility in biodiesel production and the potentiality to produce some specialty-type lipids. There is an increasing interest in isolation/adaption of robust microbe strains and design of innovative fermentation processes to make microbial lipid production a more efficient and economically feasible bio-process. Currently, the genus Rhodosporidium has been considered an important candidate, for the reason that several strains belonging to this genus have shown excellent capabilities of lipid accumulation, broad adaptabilities to various substrates, and co-production of some carotenoids. This paper reviews the current trends in the exploitation of Rhodosporidium species for microbial lipid production, including the utilization of various (single or mixed, pure or waste-derived) substrates, progress of genetic modification and metabolic engineering, innovations in fermentation mode, lipid characterizations and their potential applications. Finally, the constraints and perspectives of cultivating Rhodosporidium species for lipid production are also discussed.

Abstract:To valorize oleaginous Schizochytrium sp. biomass, a novel sequential process was addressed and developed to simultaneously produce high value DHA-enriched glycerides and low value bulky biodiesels. In this process, Schizochytrium sp. biomass was first treated by alkaline protease; then the resulting biomass was directly subjected to enzymatic ethanolysis catalyzed by liquid enzymes from C. antarctica (CALA), T. lanuginosus (TL), R. miehei (RM) and A. oryzae (PLA). For all four liquid lipases, the alkaline protease treatment on Schizochytrium sp. biomass significantly enhanced total lipid recovery, biodiesel yield and yield of DHA-enriched glycerides. Under optimized conditions, CALA and TL achieved the highest biodiesel yields 76.28-76.66 wt% based on microalgae TFAs while PLA had the lowest (55.25 wt%). Moreover, CALA attained the highest DHA (77.93 wt%) and LC PUFAs (93.08 wt%) contents in glyceride fraction, suggesting a superior performance to concentrate DHA and LC PUFAs in glycerides. C-13 NMR analysis illustrated that non-regiospecificity and unfavorability towards LC PUFAs of CALA made it possible to transesterify saturated fatty acids located in all 3 positions of microalgal oil; thus LC PUFAs were highly enriched in glycerides. Overall, the sequential process using enzymes and ethanol as green media was successfully established for the co-production of high-value microalgal bioproducts rich in LC PUFAs and sustainable biodiesels in a comprehensive manner.

Abstract:With the development of biodiesel industry, the byproduct glycerol will become a considerable resource as feedstock for production of many other chemicals. In present work, microbial conversion of crude glycerol to triacylglycerols (microbial lipid) was proposed and investigated using the oleaginous yeast Rhodosporidium toruloides (R. toruloides) by one-stage batch fermentation. Compared with glucose and refined glycerol, the crude glycerol could obtain significantly higher biomass concentration and lipid yield. The highest biomass concentration of R. toruloides obtained from crude glycerol in a 5L fermenter reached 26.7g/L with an intracellular lipid content of 70%. Further study was performed to investigate the individual effect of five representative compounds which were present in crude glycerol as impurities. It was found that within the general concentration ranges, only methanol displayed somewhat inhibitive effect, while others showed positive influence on lipid production. These results indicated that crude glycerol could be directly converted to triacylglycerols by R. toruloides without purification. Contrarily, certain amount of salt and soap could promote the accumulation of biomass and lipid.

Abstract:Microbial lipids with fatty acid compositional profiles similar to vegetable oils are of great potential for the production of biofuels and oleochemicals. However, the costs of microbial lipids remain prohibitively high for broader applications. Microalgae known as the third-generation biomass resources may be explored to reduce the feedstock costs of lipid production. Here, hydrolysates of the microalga Arthrospira platensis were converted into lipids by the oleaginous yeast Cryptococcus curvatus ATCC 20509. Results indicated that hydrothermal pretreatment at 120 °C followed by enzymatic hydrolysis led to recovery of up to 97% total reducing sugar from microalgal biomass. While the as-prepared hydrolysates were ineffective for lipid production due to its nitrogen-rich nature, a high lipid content of 64.5% and a lipid yield of 0.233 g/g were obtained upon phosphate removal with calcium hydroxide. The lipid samples were found similar to palm oils in terms of fatty acid compositional profiles. This work demonstrated that A. platensis biomass can be readily converted into hydrolysates for high-yield lipid production, suggesting an alternative feedstock to cellulosic residues for microbial lipid technology and biorefinery.This article has not yet been cited by other publications.

Abstract:Dry acid pretreatment and biodetoxification (DryPB) has been considered as an advanced technology to treat lignocellulosic materials for improved downstream bioconversion. In this study, the lipid production from DryPB corn stover was investigated by the oleaginous yeast Rhodosporidium toruloides using a new process designated prehydrolysis followed by simultaneous saccharification and lipid production (PreSSLP). The results found that prehydrolysis at 50 °C and then lipid production at 30 °C improved lipid yield by more than 17.0% compared with those without a prehydrolysis step. The highest lipid yield of 0.080 g/g DryPB corn stover was achieved at a solid loading of 12.5%. The fatty acid distribution of lipid products was similar to those of conventional vegetable oils that are used for biodiesel production. Our results suggested that the integration of DryPB process and PreSSLP process can be explored as an improved technology for microbial lipid production from lignocellulosic materials.
Orthogonal Optimization Of A Newly Isolated Microalga Desmodesmus Sp.-Chx1 As A Potential Candidate For Biodiesel Production

Abstract:This study aimed to optimize lipid productivity of an indigenous microalgal isolate Desmodesmus sp.-CHX1 when grown in modified BG-11 medium. An orthogonal L-9(3)(4) test was applied to select the optimum cultivation parameters including glucose concentration, nitrate concentration, seeding cell concentration and initial pH. Results showed that glucose concentration and seeding cell concentration were the main variables that influenced both biomass and lipid productivities. 160.8mg L-1 d(-1) of lipid productivity was much higher than those reported previously in the literatures, when Desmodesmus sp.-CHX1 was grown under the optimized conditions of 3g L-1 of glucose concentration, pH=10, 10% of seeding cell concentration and 2g L-1 of nitrate concentration. The presence of C16/C18 fatty acids accounted for 70% of total fatty acid as the major constituents. Both of high lipid productivity and ratio of C16/C18 fatty acids indicated that Desmodesmus sp.-CHX1 was a suitable feedstock for biodiesel production.
A novel strategy for integrated utilization of Jerusalem artichoke stalk and tuber for production of 2,3-butanediol by Klebsiella pneumoniae

Abstract:Jerusalem artichoke stalk and tuber can serve as a low cost feedstock for the production of 2,3-butanediol. However, like other lignocellulosic materials, the sugar concentration in the hydrolysate prepared from Jerusalem artichoke stalk is too low to be utilized effectively by microorganisms. In this paper a novel strategy was proposed to increase the sugar concentration by adding Jerusalem artichoke tuber into the hydrolysate of the stalk. The sugar was then biotransformed into high-valued 2,3-butanediol by Klebsiella pneumoniae. Fed-batch simultaneous saccharification and fermentation (SSF) was effectively performed and 901.2 mmol/l (80.5 g/l) target products (2,3-butanediol plus acetoin) was obtained in 68 h by a stage-shift aeration strategy. The concentration, yield and productivity of target products were 16.9%, 16.8% and 23.4%, respectively, higher than the best results obtained with SSF operated under constant aeration. This showed that adding tuber to the stalk hydrolysate was a useful strategy for increasing the production of 2,3-butanediol from Jerusalem artichoke via fermentation.
Culture Strategies for Lipid Production Using Low-cost Carbon Sources by Rhodosporidium Toruloides

Abstract:The development of sustainable biofuels is inserted in the biorefinery concept through the use and valorization of raw material. Volatile fatty acids (VFAs) and rich-carbon wastes, present in different raw materials, have gradually been used as the carbon source for lipid production by oleaginous yeasts. In this context, it is essential to develop practical culture strategies for oleaginous yeast to use these wastes, acetic acid and the carob industry by-product. In this study, new approaches in the cultivation of the yeast Rhodosporidium toruloides were tested, in order to maximize the accumulation of biomass and lipids. Different concentrations of carob kibbles sugar rich extract and acetic acid waste were used, as low-cost carbon sources, to select the best culture strategy for the bioconversion of these wastes into microbial oils. Microbial oils were produced by R. toruloides using 3–18 g/l acetic acid and 9–70 g/l carob extract, with different carbon-to-nitrogen ratios (C/Ns). Batch and fed-batch cultures were compared in terms of biomass and lipid production and lipid productivity. The highest biomass of 19.14 ± 0.54 g/l was obtained in batch cultures with 70 g/l of carob extract. A lipid content of 20% (w/w) was achieved using 15 g/l of acetic acid as the carbon source in a fed-batch mode. To improve biomass production as well as lipid accumulation, the strategy used was the combination of both residues in fed-batch sequential mode of the cultures in a 2L STR bioreactor. This strategy improved the lipid yield to 40% (w/w). These results provide viable culture strategies for lipid accumulation by R. toruloides using diverse waste-derived VFA’s and sugars.

Abstract:Lipids produced by oleaginous yeasts are considered as sustainable sources for the production of biofuels and oleochemicals. The red yeast Rhodosporidium toruloides can accumulate lipids to over 70% of its dry cell mass. To facilitate lipid extraction, a recombinant β-1,3-glucomannanase, MAN5C, has been applied to partially breakdown R. toruloides cell wall. In this study, R. toruloides NP11 was engineered for secretory expression of MAN5C to simplify the lipid extraction process. Specifically, a cassette contained a codon-optimized gene MAN5C was integrated into the genome of R. toruloides by Agrobacterium -mediated transformation. The engineered strain NP11-MAN5C was found with proper expression and secretion of active MAN5C, yet no notable compromise in terms of cell growth and lipid production. When NP11-MAN5C cell cultures were extracted with ethyl acetate without any pretreatment, 20% of total lipids were recovered, 4.3-fold higher than that of the parental strain NP11. When the cells were heat-treated followed by extraction with ethyl acetate in the presence of the culture broth supernatants, up to 93% of total lipids were recovered, confirming beneficial effects of MAN5C produced in situ. This study provides a new strategy to engineer oleaginous yeasts for more viable lipid extraction and down-stream processes.

Abstract:Rhodosporidium toruloides AS 2.1389 was tested using different concentrations of acetic acid as a low-cost carbon source for the production of microbial lipids, which are good raw materials for biodiesel production. It grew and had higher lipid contents in media containing 4-20 g/L acetic acid as the sole carbon source, compared with that in glucose-containing media under the same culture conditions. At acetic acid concentrations as high as 20 g/L and the optimal carbon-to-nitrogen ratio (C/N) of 200 in a batch culture, the highest biomass production was 4.35 g/L, with a lipid content of 48.2%. At acetic acid concentrations as low as 4 g/L, a sequencing batch culture (SBC) with a C/N of 100 increased biomass production to 4.21 g/L, with a lipid content of 38.6%. These results provide usable culture strategies for lipid production by R. toruloides AS 2.1389 when using diverse waste-derived volatile fatty acids.

Abstract:Tremendous amounts of herbal extraction residues (HERs) derived from the traditional Chinese herb medicine industry are routinely generated annually. However, HERs are generally treated by landfill or incineration, resulting in a great waste of resources and serious environmental issues. Here, the residue of the production of Xuanmai Ganjie Capsule (XGCR) was described for lipid production. Part of the sugar polymers (44.0%) contained in the raw XGCR could be directly hydrolyzed using a fed-batch hydrolysis process at 54% solids loading. More interestingly, sugar yield approached the theoretical maximal value when the residue was pretreated by 1% (w/w) dilute sulfuric acid before enzymatic hydrolysis. Cutaneotrichosporon oleaginosum exhibited high oleaginicity with lipid titer, content, and yield of 8.5 g/L, 40.7%, and 80.2 g/kg of residue supplied, respectively. The fatty acid profiles and the prediction of biodiesel parameters indicated the microbial lipid prepared from the XGCR was a suitable feedstock for biodiesel industry. This study strongly supported that the XGCR-to-lipid route can be a promising way for turning HERs into treasure.

Abstract:BackgroundMicrobial lipids (triacylglycerols, TAG) have received large attention for a sustainable production of oleochemicals and biofuels. Rhodosporidium toruloides can accumulate lipids up to 70% of its cell mass under certain conditions. However, our understanding of lipid production in this yeast is still much limited, especially for growth with mixed substrates at the level of metabolic network. In this work, the potentials of several important carbon sources for TAG production in R.toruloides are first comparatively studied in silico by means of elementary mode analysis followed by experimental validation.ResultsA simplified metabolic network of R.toruloides was reconstructed based on a combination of genome and proteome annotations. Optimal metabolic space was studied using elementary mode analysis for growth on glycerol, glucose, xylose and arabinose or in mixtures. The in silico model predictions of growth and lipid production are in agreement with experimental results. Both the in silico and experimental studies revealed that glycerol is an attractive substrate for lipid synthesis in R. toruloides either alone or in blend with sugars. A lipid yield as high as 0.53 (C-mol TAG/C-mol) has been experimentally obtained for growth on glycerol, compared to a theoretical maximum of 0.63 (C-mol TAG/C-mol). The lipid yield on glucose is much lower (0.29 (experimental) vs. 0.58 (predicted) C-mol TAG/C-mol). The blend of glucose with glycerol decreased the lipid yield on substrate but can significantly increase the overall volumetric productivity. Experimental studies revealed catabolite repression of glycerol by the presence of glucose for the first time. Significant influence of oxygen concentration on the yield and composition of lipids were observed which have not been quantitatively studied before.ConclusionsThis study provides for the first time a simplified metabolic model of R.toruloides and its detailed in silico analysis for growth on different carbon sources for their potential of TAG synthesis. Experimental studies revealed the phenomenon of catabolite repression of glycerol by glucose and the importance of oxygen supply on the yield and composition of lipids. More systematic studies are needed to understand the mechanisms which should help to further optimize the lipid production in this strain of industrial interest.

Abstract:Rhodotorula mucilaginosa TJY15a which was isolated from surface of marine fish could accumulate a large amount of lipid from hydrolysate of cassava starch. The cells contained 47.9% (w/w) oil during batch cultivation, whereas 52.9% (w/w) of lipid was obtained during the fed-batch cultivation. At the end of the fed-batch cultivation, all the starch were converted into reducing sugar and only 0.34gdm−3 of reducing sugar was left in the fermented medium. Therefore, the marine-derived R. mucilaginosa TJY15a was another candidate for single cell oil production. The fatty acids from R. mucilaginosa TJY15a were mainly composed of palmitic acid (C16:0), palmitoleic acid (C16:1), stearic acid (C18:0), oleic acid (C18:1) and linolenic acid (C18:2), suggesting that the fatty acids could be used as feedstock for biodiesel production.

Abstract:Environmental enrichment (EE) and exercise have been implicated in influencing behaviour and altering neuronal processes associated with cellular morphology in both ‘normal’ and injured states of the CNS. Using a rodent model of Huntington's disease, we investigated whether prolonged EE or involuntary exercise can induce morphological and cellular changes within embryonic striatal transplants. Adult rats were trained on the Staircase test – requiring fine motor control to reach and collect reward pellets – prior to being lesioned unilaterally in the dorsal neostriatum with quinolinic acid. The lesioned animals received E15 whole ganglionic eminence cell suspension grafts followed by housing in EE or standard cages. Half of the animals in standard cages received daily forced exercise on a treadmill. The grafted animals showed significant functional recovery on both the Staircase test and in drug‐induced rotation. Neither the housing conditions nor the training had an impact on the behaviour, with the exception of the treadmill reducing the ipsilateral drug‐induced rotation observed amongst the lesioned animals. However, the animals housed in the EE had significantly increased striatal brain‐derived neurotrophic factor (BDNF) levels, and graft neurons in these animals exhibited both greater spine densities and larger cell volumes. Animals on forced exercise regime had reduced BDNF levels and grafted cells with sparser spines. The study suggests that the context of the animal can affect the plasticity of transplanted cells. Appropriately exploiting the underlying, and yet unknown, mechanisms could lead the way to improved anatomical and potentially functional integration of the graft.
