Determination of nine mental drugs in human plasma using solid-phase supported liquid-liquid extraction and HPLC-MS/MS

Abstract:<p>A rapid and reliable method using solid-phase supported liquid–liquid extraction (SLE) coupled with high-performance liquid chromatography tandem mass spectrometry (HPLC-MS/MS) was established for the analysis of nine mental drugs in human plasma. The plasma sample was loaded onto a 96-well SLE plate and the analytes were separated on an Agilent Poroshell 120 EC-C<sub>18</sub> column using a mixture of methanol and water containing 0.1% formic acid using a gradient elution within 10 min of running time. Quantification was performed using internal isotope in positive ion multiple-reaction-monitoring mode and an electrospray ionization source. Under the optimized conditions, good extraction efficiency for the targets was obtained with negligible matrix interference in the subsequent HPLC-MS/MS analysis. A good linearities was obtained in the range of 1 ~ 100 ng·mL<sup>−1</sup> for aripiprazole, olanzapine, sertralin, moclobemide, paroxetine, maprotiline and iproniazid; 0.1 ~ 10 ng·mL<sup>−1</sup> for risperidone and fluoxetine with a correlation coefficient above 0.995. The method was fully validated with an accuracy of 77.24 ~ 127.71%, matrix effect (ME) of 80 ~ 120%, precision, recovery and stability. This method can quickly and sensitively determine the concentration of nine mental drugs in human plasma, provide support for the clinical monitoring of depression patients, and promote the treatment of depression.</p>
