Abstract:Freeform fabrication has emerged as a key direction in printing biologically-relevant materials and structures. With this emerging technology, complex structures with microscale resolution can be created in arbitrary geometries and without the limitations found in traditional bottom-up or top-down additive manufacturing methods. Recent advances in freeform printing have used the physical properties of microparticle-based granular gels as a medium for the submerged extrusion of bioinks. However, most of these techniques require post-processing or crosslinking for the removal of the printed structures (Miller et al., 2015; Jin et al., 2016) [1], [2]. In this communication, we introduce a novel method for the one-step gelation of silk fibroin within a suspension of synthetic nanoclay (Laponite) and polyethylene glycol (PEG). Silk fibroin has been used as a biopolymer for bioprinting in several contexts, but chemical or enzymatic additives or bulking agents are needed to stabilize 3D structures. Our method requires no post-processing of printed structures and allows for in situ physical crosslinking of pure aqueous silk fibroin into arbitrary geometries produced through freeform 3D printing.

Abstract:We present a silk biomaterial platform with highly tunable mechanical and degradation properties for engineering and regeneration of soft tissues such as, skin, adipose, and neural tissue, with elasticity properties in the kilopascal range. Lyophilized silk sponges were prepared under different process conditions and the effect of silk molecular weight, concentration and crystallinity on 3D scaffold formation, structural integrity, morphology, mechanical and degradation properties, and cell interactions in vitro and in vivo were studied. Tuning the molecular weight distribution (via degumming time) of silk allowed the formation of stable, highly porous, 3D scaffolds that held form with silk concentrations as low as 0.5% wt/v. Mechanical properties were a function of silk concentration and scaffold degradation was driven by beta-sheet content. Lyophilized silk sponges supported the adhesion of mesenchymal stem cells throughout 3D scaffolds, cell proliferation in vitro, and cell infiltration and scaffold remodeling when implanted subcutaneously in vivo.

Abstract:Hydrogel materials are of great value in medical field due to their similarity to human soft tissue.With silk fibroin as raw materials,organic solvent induction method can be employed to shorten the gelling time of silk fibroin to several hours.Silk fibroin/collagen composite hydrogel prepared with carbodiimide (EDC) as activation cross-linking agent has a storage modulus of up to 10 kPa.In vivo experiments indicate that silk fibroin hydrogel created by enzymatic crosslinking has good biocompatibility and biodegradability.We first explain the gelling mechanism of silk fibroin hydrogels,and then systematically introduce the research progress on preparation and modification methods of various silk fibroin hydrogels.Besides,potential applications of silk fibroin hydrogels as novel biomaterial in clinical applications are analyzed.Basic theoretical and technological issues to be solved for clinical applications of silk fibroin hydrogels are also discussed.

Abstract:In order to use a simple and ecofriendly method to prepare porous silk scaffolds, aqueous silk fibroin solution (ASF) was extracted from silk gland of 7-day-old fifth instar larvae of Bombyx mori (B. mori). SDS-page analysis indicated that the obtained fibroin had a molecular weight higher than 200 kDa. The fabrication of porous scaffolds from ASF was achieved by using the freeze-drying method. The pore of porous scaffolds is homogenous and tends to become smaller with an increase in the concentration of ASF. Conversely, the porosity is decreased. The porous scaffolds show impressive compressive strength which can be as high as 6.9 +/- 0.4 MPa. Furthermore, ASF has high cell adhesion and growth activity. It also exhibits high ALP activity. This implies that porous scaffolds prepared from ASF have biocompatibility. Therefore, the porous scaffolds prepared in this study have potential application in tissue engineering due to the impressive compressive strength and biocompatibility.
Fabrication and characterization of porous tubular silk fibroin scaffolds.

Abstract:Silk fibroin (SF) has been one of promising resources of biotechnology and biomedical materials due to its unique properties. Here, different sizes of porous tubular scaffolds were fabricated from a SF aqueous solution with the addition of poly(ethylene glycol diglycidyl ether) (PGDE). The scaffolds were generally flexible and transparent at the wet state with a pore size of 81-128 mu m and porosity of 90-96%, depending on the concentrations of SF and PGDE. The mechanical properties measurement showed that the tubular SF scaffolds had satisfying tensile and compression properties, especially the excellent deformation-recovery ability. FT-IR spectra indicated that the SF in the tubular scaffolds was in a beta-sheet structure, and no PGDE characteristic band was observed, suggesting that the PGDE could be removed from the scaffolds by soaking in deionized water. The cell compatibility of scaffolds was evaluated, and no obvious cytotoxicity to mouse L-929 fibroblasts was detected. (C) Koninklijke Brill NV, Leiden, 2009
Extraction of silk protein from middle silk gland of B.mori for preparation of 3-D scaffold

Abstract:In this paper, 3-D scaffolds were prepared using native protein solution extracted from middle silk gland of B.mori silkworm (SS). The distribution of pore in 3-D scaffold was homogeneous, and the pore size decreased with increase in the concentration of SS. By changing the concentration from 1.7%, 3.5% to 7%, the porosity rate of scaffolds was 95%, 94% and 91%, respectively. The water absorbency apparently decreases and the water retention rate increases with increase in the concentration of SS. The scaffolds prepared from SS with concentration of 3.5% and 7% have higher strength, on the contrary, the scaffolds prepared from SS with low concentration 1.7% shows higher elasticity. FTIR spectra indicated that SS mainly adopted beta-sheet conformation. This paper proposed one green way to extract silk protein from the silk gland of B.mori silkworm and prepare its corresponding scaffolds. It seems meaning to provide implication to develop silk-based biomaterials.

Abstract:To mimic the natural fibrous structure of the tissue extracellular matrix, a nano-fibrous silk fibroin (SF)/sodium alginate (SA) composite scaffold was fabricated by a thermally-induced phase-separation method. The effects of SF/SA ratio on the structure and the porosity of the composite scaffolds were examined. Scanning electron microscopy and porosity results showed that the 5SF/1SA and 3SF/1SA scaffolds possessed an excellent nano-fibrous structure and a porosity of more than 90%. Fourier transform infrared, X-ray diffraction, and differential scanning calorimetry results indicated the physical interaction between SF and SA molecules and their good compatibility in the 5SF/1SA and 3SF/1SA scaffolds, whereas they showed less compatibility in the 1SF/1SA scaffold. Cell culture results showed that MG-63 cells can attach and grow well on the surface of the SF/SA scaffolds. The nano-fibrous SF/SA scaffold can be potentially used in tissue engineering.

Abstract:Novel tissue engineering scaffold materials of nano-hydroxyapatite (nHA)/silk fibroin (SF) biocomposite were prepared by freeze-drying. The needle-like nHA crystals of about 10 nm in diameter by 50-80 nm in length, which were uniformly distributed in the porous nHA/SF scaffolds, were prepared by a co-precipitation method with a size. The as-prepared nHA/SF scaffolds showed good homogeneity, interconnected pores and high porosity. XRD and FT-IR analysis suggested that the silk fibroin was in beta-sheet structure, which usually provides outstanding mechanical properties for silk materials. In this work, composite scaffolds containing as high as 70% (w/w) nHA were prepared, which had excellent compressive modulus and strength, higher than the scaffolds at low nHA content level and other porous biodegradable polymeric scaffolds often considered in bone-related tissue engineering reported previously. The cell compatibility of composite scaffolds was evaluated through cell viability by MTT assay. All these results indicated that these nHA/SF scaffold materials may be a promising biomaterial for bone tissue engineering.

Abstract:Silk fibroin has been explored as a suitable biomaterial due to its biocompatibility, tunable degradability, low toxicity, and mechanical properties. To harness silk fibroin's innate properties, it is purified from native silkworm cocoons by removing proteins and debris that have the potential to cause inflammatory responses. Typically, within the purification and fabrication steps, chemical solvents, energy-intensive equipment, and large quantities of water are used to reverse engineer silk fibroin into an aqueous solution and then process into the final material format. Gentler, green methods for extraction and fabrication have been developed that reduce or remove the need for harmful chemical additives and energy-inefficient equipment while still producing mechanically robust biomaterials. This review will focus on the alternative green processing and fabrication methods that have proven useful in creating silk fibroin materials for a range of applications including consumer and medical materials.This article has not yet been cited by other publications.

Abstract:A novel all-aqueous process is described to form three-dimensional porous silk fibroin (SF) scaffolds, which not only avoided the use of organic solvents or harsh chemicals, but also can form scaffolds with various sizes and in large quantities. The scaffolds show a rough surface on the pores and the pores are highly interconnected. The porosity of the scaffolds, which varied between a large range (67.6~99.3%), can be controlled by the SF concentrations and the salt/fibroin ratio. The results of measurements indicated that this novel process can improve and enforce the transformation in SF structure from a random coil to a β-sheet. Swelling studies showed that the scaffold has excellent properties of hydrophilicity. The cell culture experiments demonstrated that the scaffolds facilitated the human osteosarcoma cells attachment and proliferation in vitro.

Abstract:The regulation of the biodegradation rate of 3D‐regenerated silk fibroin scaffolds and the avoidance of premature collapse are important concerns for their effective applications in tissue engineering. In this study, bromelain, which is specific to sericin, was used to remove sericin from silk, and high molecular weight silk fibroin was obtained after the fibroin fibers were dissolved. Afterwards, a 3D scaffold was prepared via freeze‐drying. The Sodium dodecyl sulfate–polyacrylamide gel electrophoresis results showed that the average molecular weight of the regenerated silk fibroin prepared by using the bromelain‐degumming method was approximately 142.2 kDa, which was significantly higher than that of the control groups prepared by using the urea‐ and Na2CO3‐degumming methods. The results of enzyme degradation in vitro showed that the biodegradation rate and internal three‐dimensional structure collapse of the bromelain‐degumming fibroin scaffolds were significantly slower than those of the two control scaffolds. The proliferation activity of human umbilical vein vascular endothelial cells inoculated in bromelain‐degumming fibroin scaffolds was significantly higher than that of the control scaffolds. This study provides a novel preparation method for 3D‐regenerated silk fibroin scaffolds that can effectively resist biodegradation, continuously guide cell growth, have good biocompatibility, and have the potential to be used for the regeneration of various connective tissues.

Abstract:The effectiveness of cell culture and tissue regeneration largely depends on the structural and physiochemical characteristics of tissue-engineering scaffolds. Hydrogels are frequently employed in tissue engineering because of their high-water content and strong biocompatibility, making them the ideal scaffold materials for simulating tissue structures and properties. However, hydrogels created using traditional methods have low mechanical strength and a non-porous structure, which severely restrict their application. Herein, we successfully developed silk fibroin glycidyl methacrylate (SF-GMA) hydrogels with oriented porous structures and substantial toughness through directional freezing (DF) and in situ photo-crosslinking (DF-SF-GMA). The oriented porous structures in the DF-SF-GMA hydrogels were induced by directional ice templates and maintained after photo-crosslinking. The mechanical properties, particularly the toughness, of these scaffolds were enhanced compared to the traditional bulk hydrogels. Interestingly, the DF-SF-GMA hydrogels exhibit fast stress relaxation and variable viscoelasticity. The remarkable biocompatibility of the DF-SF-GMA hydrogels was further demonstrated in cell culture. Accordingly, this work reports a method to prepare tough SF hydrogels with aligned porous structures, which can be extensively applied to cell culture and tissue engineering.

Abstract:Hydrogels are widely utilized in regenerative medicine for drug delivery and tissue repair due to their superior biocompatibility and high similarity to the extracellular matrix. For minimally invasive therapies, in situ forming gel scaffolds are desirable, but technical challenges remain to be overcome to achieve the balance between tissue-like strength and cell-sized porosity, especially for intracranial and osteological therapies. Here, a new method-inspired by the liquid crystalline spinning process in natural silk fibers-is reported for preparing injectable silk gel scaffolds with favorable preclinical efficacy and unique characteristics including 1) in situ gelling for minimally invasive surgeries, 2) controllable porosity for efficient cellular infiltration and desirable degradation, 3) resilient and tunable mechanical properties that are compatible with the modulus regime of native soft tissues, and 4) all-aqueous processing that avoids toxic solvents and enables facile loading of bioactive agents. Moreover, hierarchically structured heterogeneous silk gel scaffolds with variable porosity and bioactive agent gradients within 3D matrices can be achieved for sustained drug release and guided tissue regeneration. Preclinical efficacy studies in rodent models show efficient bacterium and glioma inhibition and positive effects on bone regeneration and vascularization.
Silk Protein Composite Bioinks and Their 3D Scaffolds and In Vitro Characterization

Abstract:This paper describes the use of silk protein, including fibroin and sericin, from an alkaline solution of Ca(OH)2 for the clean degumming of silk, which is neutralized by sulfuric acid to create calcium salt precipitation. The whole sericin (WS) can not only be recycled, but completely degummed silk fibroin (SF) is also obtained in this process. The inner layers of sericin (ILS) were also prepared from the degummed silk in boiling water by 120 °C water treatment. When the three silk proteins (SPs) were individually grafted with glycidyl methacrylate (GMA), three grafted silk proteins (G-SF, G-WS, G-ILS) were obtained. After adding I2959 (a photoinitiator), the SP bioinks were prepared with phosphate buffer (PBS) and subsequently bioprinted into various SP scaffolds with a 3D network structure. The compressive strength of the SF/ILS (20%) scaffold added to G-ILS was 45% higher than that of the SF scaffold alone. The thermal decomposition temperatures of the SF/WS (10%) and SF/ILS (20%) scaffolds, mainly composed of a β-sheet structures, were 3 °C and 2 °C higher than that of the SF scaffold alone, respectively. The swelling properties and resistance to protease hydrolysis of the SP scaffolds containing sericin were improved. The bovine insulin release rates reached 61% and 56% after 5 days. The L929 cells adhered, stretched, and proliferated well on the SP composite scaffold. Thus, the SP bioinks obtained could be used to print different types of SP composite scaffolds adapted to a variety of applications, including cells, drugs, tissues, etc. The techniques described here provide potential new applications for the recycling and utilization of sericin, which is a waste product of silk processing.

Abstract:Silk fibroin (SF) microspheres show bright prospects for biomedical applications, such as microcarriers, drug delivery, tumor embolization agents, and microscaffolds. However, the chemistry-independent preparation of SF microspheres, which is critical to biomedical applications, has been challenging. In this study, the SF microspheres with silk I crystal type were generated by using electrostatic spraying and freezing-induced assembly. The SF solution was sprayed into liquid nitrogen to form frozen microspheres with tunable size. Annealing can crystallize frozen SF to form silk I crystal type, providing a green approach to harvest water-insoluble microspheres. The SF microspheres can retain a monolithic shape in water for up to 30 days, while having a 77 % degradation ratio in PBS in 14 days, showing high stability in water and rapid degradation under physiological conditions. The biomedical application prospects of the silk I microspheres were demonstrated by cell culture and small molecule drugs (doxorubicin). The microspheres can support the growth and expansion of mammalian cells, and provide a sustainable release for DOX with 10 days. This strategy offers a green approach that avoids the use of organic solvents and cross-linkers for designing SF microsphere biomaterials.
Microstructured silk fiber scaffolds with enhanced stretchability

Abstract:Despite extensive research, current methods for creating three-dimensional (3D) silk fibroin (SF) scaffolds lack control over molecular rearrangement, particularly in the formation of β-sheet nanocrystals that severely embrittle SF, as well as hierarchical fiber organization at both micro- and macroscale. Here, we introduce a fabrication process based on electrowriting of aqueous SF solutions followed by post-processing using an aqueous solution of sodium dihydrogen phosphate (NaH2PO4). This approach enables gelation of SF chains via controlled β-sheet formation and partial conservation of compliant random coil structures. Moreover, this process allows for precise architecture control in microfiber scaffolds, enabling the creation of 3D flat and tubular macro-geometries with square-based and crosshatch microarchitectures, featuring inter-fiber distances of 400 μm and ~97% open porosity. Remarkably, the crosslinked printed structures demonstrated a balanced coexistence of β-sheet and random coil conformations, which is uncommon for organic solvent-based crosslinking methods. This synergy of printing and post-processing yielded stable scaffolds with high compliance (modulus = 0.5–15 MPa) and the ability to support elastic cyclic loading up to 20% deformation. Furthermore, the printed constructs supported in vitro adherence and growth of human renal epithelial and endothelial cells with viability above 95%. These cells formed homogeneous monolayers that aligned with the fiber direction and deposited type-IV collagen as specific marker of healthy extracellular matrix, indicating that both cell types attach, proliferate, and organize their own microenvironment within the SF scaffolds. These findings represent a significant development in fabricating organized stable SF scaffolds with unique microfiber structures and mechanical and biological properties that make them highly promising for tissue engineering applications.

Abstract:Scaffolds used for bone tissue engineering need to have a variety of features to accommodate bone cells. The scaffold should mimic natural bone, it should have appropriate mechanical strength, support cell differentiation to the osteogenic lineage, and offer adequate porosity to allow vascularization and bone in-growth. In this work, we aim at developing a new process to fabricate such materials by creating a porous composite material made of silk fibroin and cellulose as a suitable scaffold of bone tissue engineering. Silk fibroin and cellulose are both dissolved together in N,N-dimethylacetamide/LiCl and molded to a porous structure using NaCl powder. The hydrogels are prepared by a sequential regeneration process: cellulose is solidified by water vapor treatment, while the remaining silk fibroin in the hydrogel is insolubilized by methanol, which leads to a cellulose framework structure embedded in a silk fibroin matrix. Finally, the hydrogels are soaked in water to dissolve the NaCl for making a porous structure. The cellulose composition results in improving the mechanical properties for the hydrogels in comparison to the silk fibroin control material. The pore size and porosity are estimated at around 350 μm and 70%, respectively. The hydrogels support the differentiation of MC3T3 cells to osteoblasts and are expected to be a good scaffold for bone tissue engineering.

Abstract:<span>Silk fibroin (SF) from <i>Bombyx mori</i> is a natural polymer with exceptional biocompatibility, low immunogenicity, and ease of processability. SF-based hydrogels have been identified as one of the most attractive candidate scaffolds for tissue engineering and can be fabricated through various physical or chemical crosslinking approaches. However, conventional SF hydrogels may suffer from several major drawbacks, such as structural inhomogeneity, poor mechanical properties or utilization of cytotoxic reagents. Herein, a dually crosslinked SF-based composite hydrogel with enhanced strength and elasticity was fabricated by inducing the formation of uniform and small β-sheet structures by sonication in a restricted enzymatic precrosslinked network. The composite hydrogel not only demonstrated concentration-dependent stiffness variation but also exhibited time-dependent changes in toughness behavior. Moreover, subsequent experimental results revealed that the hydrogels exhibit other advantages, including high water retention capacity and long-term stability under physiological conditions. Finally, a three-dimensional (3 D) construct of the cell-laden hydrogel was fabricated, confirming that the composite hydrogel could provide a biocompatible microenvironment with dynamically changing mechanical properties. The combination of physical and enzymatic crosslinking strategies contributes to a biocompatible composite hydrogel with unique mechanical properties that holds great potential for use in tissue engineering and regenerative medicine.</span>

Abstract:Although three-dimensional fibroin scaffolds have been prepared with freeze–drying method, these scaffolds still cannot meet the requirements of tissue engineering. In this article, a new process is described to form fibroin-based porous scaffolds with controllable structure and morphological features. When collagen was added to fibroin solution, the viscosity of the blend solution increased because of the interaction between fibroin and collagen, and then it restrained the unwanted fibroin leaf formation in freezing process that generally appeared in the previous fibroin scaffold preparation. With methanol treatment, the fibroin/collagen scaffolds became water-stable, following the transition from random and α-helix to β-sheet conformation. The aqueous-fibroin porous scaffolds had highly homogeneous and interconnected pores with pore sizes ranging from 127 to 833μm, depending on the fibroin concentration. The porosity of scaffolds was >90%, and the yield strength and modulus were up to 354±25kPa and 30±0.1MPa, respectively, when the blend solution, containing 20% collagen, maintained 4% fibroin concentration. Adhesion, spreading and proliferation of HepG2 cells on fibroin and fibroin/collagen blend scaffolds were also observed to investigate the biocompatibility. Scanning electron microscopy (SEM) and MTT analyses demonstrated that the adding of collagen evidently facilitated HepG2 attachment and proliferation in vitro. These new fibroin based three-dimensional scaffolds provided much more excellent properties due to the greatly improved control of pore size, the uniform pore distribution, the hydrophility, the mechanical properties and the biocompatibility compared with those of reported three-dimensional fibroin scaffolds.
