Abstract:The salt-gradient operation mode used in ion-exchange simulated moving bed chromatography (SMBC) can improve the efficiency of protein separations. A detailed model that takes into account any kind of adsorption/ion-exchange equilibrium, salt gradient, size exclusion, mass transfer resistance, and port periodic switching mechanism, was developed to simulate the complex dynamics. The model predictions were verified by the experimental data on upward and downward gradients for protein separations reported in the literature. All design and operating parameters (number, configuration, length and diameter of columns, particle size, switching period, flow rates of feed, raffinate, desorbent and extract, protein concentrations in feed, different salt concentrations in desorbent and feed) can be chosen correctly by numerical simulation. This model can facilitate the design, operation, optimization, control and scale-up of salt-gradient ion-exchange SMBC for protein separations.

Abstract:Simulated moving bed chromatography (SMBC) is attracting increased interest for separations of pharmaceuticals and fine chemicals. The SMBC process is a mixed discrete and continuous process. It is a non-linear non-ideal multivariable-coupling hybrid system. A detailed model that takes account of non-linear isotherms, mass transfer resistance, extra-column effect, and discrete event of port periodic switching, was developed to simulate the complex dynamics. Abstract:The adsorption of levulinic acid in fixed beds of basic polymeric adsorbents at 22 degrees C was studied under various operating conditions. A general rate model which considers pore diffusion and parallel pore/surface diffusion was solved numerically by orthogonal collocation on finite elements to describe the experimental breakthrough data. The adsorption isotherms, and the pore and surface diffusion coefficients were determined independently in batch adsorption studies. The external film resistance and the axial dispersion coefficient were estimated by the Wilson-Geankoplis equation and the Chung-Wen equation, respectively. Simulation elucidated that the model which considers parallel diffusion successfully describes the breakthrough behavior and gave a much better prediction than the model which considers pore diffusion. The results obtained in this work are applicable to design and optimizes the separation process.
Minimising Biomass/adsorbent Interactions in Expanded Bed Adsorption Processes: a Methodological Design Approach

Abstract:Expanded bed adsorption (EBA) is an integrated technology for the primary recovery of proteins from crude feedstock. Interactions between solid matter in the feed suspension and fluidised adsorbent particles influence bed stability and therefore have a significant impact on protein adsorption in expanded beds. In order to design efficient and reliable EBA processes a strategy is needed, which allows to find operating conditions, where these adverse events do not take place. In this paper a methodological approach is presented, which allows systematic characterisation and minimisation of cell/adsorbent interactions with as little experimental effort as possible. Adsorption of BSA to the anion exchanger Streamline Q XL from a suspension containing S. cerevisiae cells was chosen as a model system with a strong affinity of the biomass towards the stationary phase. Finite bath biomass adsorption experiments were developed as an initial screening method to estimate a potential interference. The adhesiveness of S. cerevisiae to the anion exchanger could be reduced significantly by increasing the conductivity of the feedstock. A biomass pulse response method was used to find optimal operation conditions showing no cell/adsorbent interactions. A good correlation was found between the finite bath test and the pulse experiment for a variety of suspensions (intact yeast cells, E. coli homogenate and hybridoma cells) and adsorbents (Streamline Q XL, DEAE and SP), which allows to predict cell/adsorbent interactions in expanded beds just from finite bath adsorption tests. Under the optimised operating conditions obtained using the prior methods, the stability of the expanded bed was investigated during fluidisation in biomass containing feedstock (up to 15% yeast on wet weight basis) employing residence time distribution analysis and evaluation by an advanced model. Based on these studies threshold values were defined for the individual experiments, which have to be achieved in order to obtain an efficient EBA process. Breakthrough experiments were conducted to characterise the efficiency of BSA adsorption from S. cerevisiae suspensions in EBA mode under varying operating conditions. This allowed to correlate the stability of the expanded bed with its sorption efficiency and therefore could be used to verify the threshold values defined. The approach presented in this work provides a fast and simple way to minimise cell/adsorbent interactions and to define a window of operation for protein purification using EBA.

Abstract:Expanded bed adsorption (EBA) is a special chromatography technique with perfect classification of adsorbent particles in the column, thus the performance of protein adsorption in expanded beds is particular, obviously nonuniform and complex along the column. Detailed description of the complex adsorption kinetics of proteins in expanded bed is essential for better analyzing of adsorptive mechanisms, the design of chromatographic processes and the optimization of operation parameters of EBA processes. In this work, a theoretical model for the prediction of protein adsorption kinetics in expanded beds was developed by taking into account the classified distribution of adsorbent particles along the bed height, the nonuniform behaviors of axial liquid dispersion, the axial variation of local bed voidage as well as the axial changes of target component mass transfer. The model was solved using the implicit finite difference scheme combining with the orthogonal collocation method, and then applied to predict the breakthrough behaviors of bovine serum albumin (BSA) on Streamline DEAE and lysozyme on Streamline SP along the bed height in expanded beds under various conditions. In addition, the experiments of front adsorption of BSA on Streamline DEAE at different axial column positions were carried out to reveal the adsorption kinetics of BSA along the bed height in a 20 mm I.D. expanded bed, and the influences of liquid velocity and feed concentration on the breakthrough behaviors were also analyzed. The breakthrough behaviors predicted by the present model were compared with the experimental data obtained in this work and in the literature published. Process Design in Expanded Bed Adsorption - Integrating Target Adsorption and Biomass Influence

Abstract:Expanded bed adsorption (EBA) is an innovative chromatography technology that allows the introduction of particle containing feedstock without the risk of blocking the bed. Compared with normal chromatography, two factors should be considered in EBA process design: (1) the fluid side transport can not be neglected due to the increased void fraction in the bed; (2) the influence of suspended biomass in the feedstock should be seriously considered. With the appropriate methods of predicting the sorption efficiency and characterizing biomass influences in EBA, a parallel process design strategy is proposed that considers the efficient adsorption of target protein, together with the negative effects of biomass in the feedstock. The strategy was applied to the EBA separation of recombinant formate dehydrogenases from E. coli homogenate. Good results were obtained with a purification factor of 6 and a yield of 85%. In addition, the process should be scaled-up.

Abstract:Abstract Expanded bed adsorption (EBA) emerged in the early 1990s in an attempt to integrate the clarification, capture and initial product concentration/purification process. Several mathematical models have been put forward to describe its operation. However, none of the models developed specifically for EBA allows simultaneous prediction of bed hydrodynamics, mass transfer/adsorption and (unwanted) interactions and fouling. This currently limits the development and early optimization of EBA‐based separation processes. In multiphase reactor engineering, the use of multiphase computational fluid dynamics has been shown to improve fundamental understanding of fluidized beds. To advance EBA technology, a combination of particle, equipment and process scale models should be used. By employing a cascade of multiscale simulations, the various challenges EBA currently faces can be addressed. This allows for optimal design and selection of equipment, materials and process conditions, and reduces risks and development times of downstream processes involving EBA. © 2018 The Authors. Abstract:Multi-column periodic counter-current chromatography (PCC) has been developed for continuous antibody capture, but the complexity of continuous processes makes experimental optimization time consuming and costly. In this work, with twin-column continuous system as a typical case, mathematical models were established and used to evaluate the impacts of operating parameters for process development. The model fitted well with the experimental breakthrough curves and process performance under varying protein concentrations and residence times. Three important operating parameters, residence time for interconnected feeding (RTC), breakthrough percentage control for interconnected feeding (s) and disconnected feeding time (tDC), were evaluated systematically. The profiles of productivity and resin capacity utilization showed three phases as a function of RTC, which resulted in different optimization strategies towards s and tDC. Based on the model prediction, a working window of RTC and s can be determined for process development. Finally, a model-based design approach was proposed to determine the optimum operating conditions and to design a suitable continuous process for high productivity and capacity utilization. With the model-based design approach developed, the best performance of 12.8 g/L/h productivity and 91.9% capacity utilization was found for MabSelect SuRe resin under 1 mg/mL feeding IgG concentration at RTC = 2 min, s = 65% and tDC = 26 min.

Abstract:Asynchronous simulated moving bed chromatography(ASMBC),known also as the "VARICOL" process,is more efficient and flexible than the well-known and traditional simulated moving bed chromatography(SMBC).A detailed model of ASMBC,taking account of non-linear competitive isotherms,mass transfer parameters,and complex port switching schedule parameters,was developed to simulate the complex dynamics of ASMBC.The simulated performance is in close agreement with the experimental data of chiral separation reported in the literature.The simulation results show that ASMBC can achieve the performance similar to SMBC with fewer columns and can achieve better performance than SMBC with the same total column number.All design and operation parameters can be chosen correctly by numerical simulation.This detailed ASMBC model and the numerical technique are useful for design,operation,optimization and scale-up of ASMBC.

Abstract:A novel mixed-mode expanded bed adsorbent with anion-exchange properties was explored with benzylamine as the functional ligand. The cellulose composite matrix, densified with stainless steel powder, was prepared with the method of water-in-oil suspension thermal regeneration. High activation levels of the cellulose matrix were obtained with allyl bromide because of the relative inertness of the allyl group under the conditions of the activation reaction. After the formation of the bromohydrin with N-bromosuccinimide and coupling with benzylamine, the activated matrix was derived to function as a mixed-mode adsorbent containing both hydrophobic and ionic groups. The protein adsorption capacity was investigated with bovine serum albumin as a model protein. The results indicated that the prepared adsorbent could bind bovine serum albumin with a high adsorption capacity, and it showed salt tolerance. Effective desorption was achieved by a pH adjustment across the isoelectric point of the protein. The interactions between the cell and adsorbent were studied, and the bioadhesion was shielded by the adjustment of the salt concentration above 0.1M. Stable fluidization in the expanded bed was obtained even in a 2% (dry weight) yeast suspension. The direct capture of target proteins from a biomass-containing feedstock without extra dilution steps could be expected with the mixed-mode adsorbent prepared in this work, and this would be especially appropriate for expanded bed adsorption applications. Abstract:Chromatographic separation processes such as simulated moving bed (SMB) are widely used in the petrochemical, fine chemical, sugar, and pharmaceutical industries. Their separation efficiency can be improved by optimizing the components' adsorptivity in the different unit sections through, e.g., the use of solvent, temperature, or PH gradients. In this work, the salt gradient ion-exchange SMB used to separate two proteins, beta-lactoglobulins A and B, is theoretically analyzed, where the protein adsorption is described by the steric mass action model. Detailed model-based sequential optimization studies have been carried out for both closed-loop isocratic SMB as well as closed and open-loop gradient ion-exchange SMB. The separation efficiency is described by the throughput. Different gradient cases were analyzed to find out the influence factors. The results show that the gradient SMB is more efficient than the isocratic SMB in terms of startup time, throughput, and desorbent-to-throughput ratio. Moreover, it can be known that using open-loop gradient ion-exchange SMB to separate proteins is more effective than using closed-loop gradient SMB. The influencing factors such as the mass transfer efficiency and the maximum flow rate on separation efficiency are discussed according to different cases.

Abstract:Separation mechanism of liquid-solid fluidized bed separator (LSFBS) was studied by using theoretical analysis and experimental researches.Characteristics of autogenous medium fluidized bed in LSFBS were analyzed and the influence of autogenous medium size on the separation performance of LSFBS was studied,taking quartz particles of various sizes as autogenous medium.According to the separation results,a better segregation could be achieved by using finer autogenous medium.Particularly,the optimum of Ep value of 0.15~0.2 mm quartz was 0.053 while that of 0.25~0.45 mm quartz increased to 0.095.Finally,the influence of size distribution of feed and operating conditions on the separation efficiency was analyzed.Reducing feed size range and enlarging lower-size limit of feed could improve the device performance.Under a certain bed pressure,a smaller upward current velocity could generate a higher solid volume fraction and bed effective density.A smaller upward current velocity could also decrease the influence of size and shape on the movement of particles.All of these changes could induce a better separation performance for LSFBS.

Abstract:Immobilized metal affinity chromatography (IMAC) ensures the specific purification of proteins containing histidine tags through high affinity with transition metal chelators, which has various applications in biological protein separation. Most chromatographic separations currently use a fixed bed. In this form, internal flow pressure drops very sharply, accompanied by uneven solution flow, pore blockages, etc., all of which greatly reduce separation efficiency. Therefore, this study uses hollow fiber membranes (HFMs) with micron-scale inner diameters as a base, thus reducing operating pressure and significantly enhancing mass transmission. Batch adsorption experiments were performed using flat plate membranes to obtain the reaction's thermodynamic and kinetic model parameters for use in a dynamic column breakthrough simulation. The numerical simulation was based on a single HFM model and established a mathematical model for computational fluid dynamics (CFD) in ANSYS Fluent software. Model accuracy was validated by combining the simulation with experiments. The effects of different module and process parameters on the breakthrough curve were investigated by varying parameters such as flow rate, initial feed concentration, and HFM inner diameter. Design parameters and operating conditions contributing to module utilization were subsequently obtained.
CFD modelling and simulation of hydrodynamics in liquid-solid circulating fluidized beds

Abstract:Hydrodynamics in liquid-solid circulating fluidized beds (LSCFBs) are modelled and simulated by computational fluid dynamic (CFD) technique. Turbulence model and kinetic theory descriptions of granular flow are incorporated in the governing equations to model the high Reynolds number two-phase flows with strong particle-particle interactions in LSCFBs. Liquid-solid upflows under wide operating conditions, different particle properties and different bed dimensions are investigated by time-dependent simulations using commercial software, FLUENT 4.5.6. The model predictions show good agreements with the experimental data and reasonable trends when different fluids/solids and bed dimensions are applied. Stronger nonuniformities in flow structures are found in larger particle systems, with larger fluid/solids density ratios and/or larger diameter columns.

Abstract:Carbon capture, utilization, and sequestration (CCS) is one of the key technologies to reduce the emission of carbon dioxide, including that from exiting flue gas of fossil fuel-fired power plants. In this study, the two-fluid model approach was used to simulate the hydrodynamics of the entire circulating fluidized bed (CFB) loop of the integrated carbonation and regeneration sections using the mass and momentum equations and constitutive relations for both the gas and solid phases. Then, the verified models of the non-reactive simulation of the CFB loop were combined with species, energy equations, and carbonation regeneration rate of reactions for both phases to simulate the entire circulating fluidized bed carbon capture unit. The predicted extent of CO 2 capture in this system showed the potential of continuous carbon capture using amine-based solid sorbents through the circulating fluidized bed CO 2 capture unit.

This review includes discussion of several modeling techniques, such as CFD, ANN, and ANFIS in the (G)LSCFB system, which can help to minimize the cost and time required for experimental studies. Additionally, the (G)LSCFB is versatile and useful for designing new bioreactors, especially for simultaneous and independent reaction and regeneration of biocatalyst particles due to its two distinct zones. The article describes the applications of (G)LSCFBs in various fields, including wastewater treatment , polymerization of phenol , lactic acid production, protein recovery, heavy metal removal, and linear alkylbenzene (LAB) production. Abstract:A detailed study on the hydrodynamics of liquid–solid circulating fluidized bed (LSCFB) reactors is crucial in the efficient design and scale-up of these reactors. In this paper, an axisymmetric CFD model is developed to simulate the flow field in a LSCFB riser. The model is based on Eulerian–Eulerian approach incorporating the kinetic theory of granular flow. The predicted results agree well with our earlier experimental data. Furthermore, it is found that the dispersed k–ε multiphase turbulence model is more accurate and computationally efficient than other k–ε multiphase turbulence models. Also, the model predicts the residence time of both liquid and solid phases in the riser by using a Pulse technique. Finally, the proposed CFD model is used to investigate the effects of the liquid stream velocity and the solids circulation rate on the performance of the LSCFB. It is demonstrated that the proposed CFD model can be a robust tool for the scale-up and design of industrial LSCFB reactors.
