Abstract:Understanding the behavior of skeletal muscle is critical to implementing computational methods to study how the body responds to compressive loading. This work presents a novel approach to studying the fully nonlinear response of skeletal muscle in compression. Porcine muscle was compressed in both the longitudinal and transverse directions under five stress relaxation steps. Each step consisted of 5% engineering strain over 1 s followed by a relaxation period until equilibrium was reached at an observed change of 1 g/min. The resulting data were analyzed to identify the peak and equilibrium stresses as well as relaxation time for all samples. Additionally, a fully nonlinear strain energy density-based Prony series constitutive model was implemented and validated with independent constant rate compressive data. A nonlinear least squares optimization approach utilizing the Levenberg-Marquardt algorithm was implemented to fit model behavior to experimental data. The results suggested the time-dependent material response plays a key role in the anisotropy of skeletal muscle as increasing strain showed differences in peak stress and relaxation time (p < 0.05), but changes in equilibrium stress disappeared (p > 0.05). The optimizing procedure produced a single set of hyper-viscoelastic parameters which characterized compressive muscle behavior under stress relaxation conditions. The utilized constitutive model was the first orthotropic, fully nonlinear hyper-viscoelastic model of skeletal muscle in compression while maintaining agreement with constitutive physical boundaries. The model provided an excellent fit to experimental data and agreed well with the independent validation in the transverse direction.
Effect of skeletal muscle immobilization on regional anisotropic viscohyperelastic properties change in a rat model

Abstract:Passive mechanical properties in three different zones (distal, proximal, and medial) of biceps brachii immobilized in short position and its free contralateral were investigated. For that, in vitro equibiaxial relaxation tests were performed with samples harvested from skeletal muscles of immobilized rats during one or two weeks. From data obtained in two plane axes of loading, a viscohyperelastic anisotropic model described by a strain energy function coupled with second order Maxwell's model, was used to identify the material parameters. It has been shown that the zone influences the material parameters of the hyperelasticity behavior while the immobilization acts rather on the viscoelasticity response. Based on measurements of histological parameters from flexor carpi ulnaris muscles, we observed that immobilization produced contractile tissue atrophy and connective tissue thickening. The muscle atrophy caused by immobilization could lead to a more linear mechanical behavior along the axis of the muscle fibers. Furthermore, fibrosis, quantified by histological analysis on flexor carpi ulnaris muscles, could result in highly non-linear behavior along the other axis. These structural changes also contribute to an increase in relaxation following immobilization along both axes (+11.7% and +15.5% on average with p < 0.001 and p < 0.01 respectively for each axis). Statement of significance To our best knowledge, this work is the first to investigate the anisotropic, viscohyperelastic and heterogeneous properties within a passive skeletal muscle immobilized in a short position. For that, from a rat model, equibiaxial relaxation tests were performed and material parameters were found from a strain energy function coupled with a Maxwell's model. We showed that excising zone of samples greatly influences the parameters of the hyperelastic behavior while the immobilization influences the viscoelasticity response instead. Furthermore, histological analysis permitted to quantify fibrosis in the skeletal muscle and bear out, at least in part, changes in relaxation behavior following immobilization.

Abstract:When a living tissue is subjected to cyclic stretching, the stress-strain curves show a shift down with the increase in the number of cycles until stabilization. This phenomenon is referred to in the literature as a preconditioning and is performed to obtain repeatable and predictable measurements. Preconditioning has been routinely performed in skin tissue tests; however, its effects on the mechanical properties of the material such as viscoelastic response, tangent modulus, sensitivity to strain rate, the stress relaxation rate, etc….remain unclear. In addition, various physical interpretations of this phenomenon have been proposed and there is no general agreement on its origin at the microscopic or mesoscopic scales. The purpose of this study was to investigate the effect of the cyclical stretching and the stress-relaxation tests on the mechanical properties of the porcine skin. Cyclic uniaxial tensile tests at large and constant strain were performed on different skin samples. The change in the reaction force, and skin's tangent modulus as a function of the number of cycles, as well as the strain rate effect on the mechanical behavior of skin samples after cycling were investigated. Stress-relaxation tests were also performed on skin samples. The change in the reaction force as a function of relaxation time and the strain rate effect on the mechanical behavior of skin samples after the stress-relaxation were investigated. The mechanical behavior of a skin sample under stress-relaxation test was modeled using a combination of hyperelasticity and viscoelasticity. Overall, the results showed that the mechanical behavior of the skin was strongly influenced by cycling and stress relaxation tests. Indeed, it was observed that the skin's resistance decreased by about half for two hours of cycling; the tangent modulus degraded by nearly 30% and skin samples became insensitive to the strain rates and accumulated progressively an inelastic deformation over time during cycling. Finally, the hysteresis loops became very narrow at the end of cycling and after relaxation process.

Abstract:Virtual finite element human body models have been widely used in biomedical engineering, traffic safety injury analysis, etc. Soft tissue modeling like skeletal muscle accounts for a large portion of a human body model establishment, and its modeling method is not enough explored. The present study aims to investigate the compressive properties of skeletal muscles due to different species, loading rates and fiber orientations, in order to obtain available parameters of specific material laws as references for building or improving the human body model concerning both modeling accuracy and computational cost. A series of compressive experiments of skeletal muscles were implemented for human gastrocnemius muscle, bovine and porcine hind leg muscle. To avoid long-time preservation effects, all experimental tests were carried out in 24 h after that the samples were harvested. Considering computational cost and generally used in the previous human body models, one-order hyperelastic Ogden model and three-term simplified viscoelastic quasi-linear viscoelastic (QLV) were selected for numerical analysis. Inverse finite element analysis was employed to obtain corresponding material parameters. With good fitting records, the simulation results presented available material parameters for human body model establishment, and also indicated significant differences of muscle compressive properties due to species, loading rates and fiber orientations. When considering one-order Ogden law, it is worthy of noting that the inversed material parameters of the porcine muscles are similar to those of the human gastrocnemius regardless of fiber orientations. In conclusion, the obtained material parameters in the present study can be references for global human body and body segment modeling.

Abstract:Using shear wave elastography, we measure the changes in the wave speed with the stress produced by a striated muscle during isometric voluntary contraction. To isolate the behaviour of an individual muscle from complementary or antagonistic actions of adjacent muscles, we select the flexor digiti minimi muscle, whose sole function is to extend the little finger. To link the wave speed to the stiffness, we develop an acousto-elastic theory for shear waves in homogeneous, transversely isotropic, incompressible solids subject to uniaxial stress. We then provide measurements of the apparent shear elastic modulus along, and transversely to, the fibre axis for six healthy human volunteers of different age and sex. The results display a great variety across the six subjects. We find that the slope of the apparent shear elastic modulus along the fibre direction changes inversely to the maximum voluntary contraction (MVC) produced by the volunteer. We propose an interpretation of our results by introducing the S (slow) or F (fast) nature of the fibres, which harden the muscle differently and accordingly, produce different MVCs. This work opens the way to measuring the elastic stiffness of muscles in patients with musculoskeletal disorders or neurodegenerative diseases.

Abstract:Optical chemical sensing of pH and water content in organic solvents is proposed, using multiinformation dyes (MIDs) based on the support matrixes for the dyes. In this investigation, four kinds of merocyanine-type dyes having a polymerizable olefin unit as the MIDs were synthesized. These dyes were copolymerized with hydrophilic monomer molecules to obtain dye-immobilized optical chemical sensor (optode) membranes. In this case, selection of the monomer molecule gave optode membranes having different color change properties, because different monomer molecules provided different chemical environments around the immobilized dye. These optode membranes were used for the measurement of pH and water content in organic solvents. These membranes offered two-dimensional sensing information in one spectrum when they were employed for water content sensing in organic solvents, in which the maximum wavelength represents the water content and the absorbance at this wavelength represents the pH of the water present. These polymer membranes have a long lifetime, which can be adequate for practical use.
Anisotropic and viscoelastic tensile mechanical properties of aponeurosis: Experimentation, modeling, and tissue microstructure

Abstract:Aponeuroses are stiff sheath-like components of the muscle-tendon unit that play a vital role in force transmission and thus locomotion. There is clear importance of the aponeurosis in musculoskeletal function, but there have been relatively few studies of aponeurosis material properties to date. The goals of this work were to: 1) perform tensile stress-relaxation tests, 2) perform planar biaxial tests, 3) employ computational modeling to the data from 1 to 2, and 4) perform scanning electron microscopy to determine collagen fibril organization for aponeurosis tissue. Viscoelastic modeling and statistical analysis of stress-relaxation data showed that while relaxation rate differed statistically between strain levels (p = 0.044), functionally the relaxation behavior was nearly the same. Biaxial testing and associated modeling highlighted the nonlinear (toe region of ~2-3% strain) and anisotropic (longitudinal direction linear modulus ~50 MPa, transverse ~2.5 MPa) tensile mechanical behavior of aponeurosis tissue. Comparisons of various constitutive formulations showed that a transversely isotropic Ogden approach balanced strong fitting (goodness of fit 0.984) with a limited number of parameters (five), while damage modeling parameters were also provided. Scanning electron microscopy showed a composite structure of highly aligned, partially wavy collagen fibrils with more random collagen cables for aponeurosis microstructure. Future work to expand microstructural analysis and use these data to inform computational modeling would benefit this work and the field.
Simulation of active skeletal muscle tissue with a transversely isotropic viscohyperelastic continuum material model

Abstract:Human body models with biofidelic kinematics in vehicle pre-crash and crash simulations require a constitutive model of muscle tissue with both passive and active properties. Therefore, a transversely isotropic viscohyperelastic continuum material model with element-local fiber definition and activation capability is suggested for use with explicit finite element codes. Simulations of experiments with New Zealand rabbit's tibialis anterior muscle at three different strain rates were performed. Three different active force-length relations were used, where a robust performance of the material model was observed. The results were compared with the experimental data and the simulation results from a previous study, where the muscle tissue was modeled with a combination of discrete and continuum elements. The proposed material model compared favorably, and integrating the active properties of the muscle into a continuum material model opens for applications with complex muscle geometries.

Abstract:Precise characterization of the passive elastic properties of human skeletal muscle could provide valuable information for functional assessment and medical diagnosis. Using normalized elastic modulus-length curve based on a piecewise exponential model, a non-invasive ultrasonography (US) method was developed for estimating three physiologically meaningful parameters, termed as passive-elastic coefficient k, slack length l0 and slack elastic modulus G0, to quantify the passive elastic properties of human gastrocnemius (GM) muscle, and correlate different findings with healthy and atrophic subjects. Dynamic B-mode US and shear wave elastography (SWE) videos of right GM muscles were performed in 32 healthy subjects and 18 atrophic patients with their ankle angles from 40° plantarflexion to 30° dorsiflexion by an isokinetic dynamometer. During muscle passive stretching induced by ankle rotation, consecutive muscle lengths were measured by automatically tracking the distal muscle-tendon junction positions in B-mode imaging frames using Hough transform and an active contour method. The corresponding elastic moduli were obtained using SWE imaging frames. The Gauss-Newton algorithm was used to estimate the parameters k, l0 and G0 from normalized elastic modulus-length curve. In-vivo experimental results showed that the measured muscle elastic modulus-length data fitted well to the proposed model with three estimated parameters for all subjects (0.942 < R2 < 0.997). The passive-elastic coefficient k was significantly higher for atrophic subjects compared with normal subjects (105.5 ± 45.3 versus 48.4 ± 16.0, p < 0.001). The proposed parameters allow further characterization of muscle essential mechanical properties and have a potential to become effective indexes for muscular disease diagnosis.

Abstract:Based on the logarithmic rate and piecewise linearization theory, a thermodynamically consistent elasto-viscoplastic constitutive model is developed in the framework of finite deformations to describe the nonlinear time-dependent biomechanical performances of soft biological tissues, such as nonlinear anisotropic monotonic stress-strain responses, stress relaxation, creep and ratchetting. In the proposed model, the soft biological tissue is assumed as a typical composites consisting of an isotropic matrix and anisotropic fiber aggregation. Accordingly, the free energy function and stress tensor are divided into two parts related to the matrix and fiber aggregation, respectively. The nonlinear biomechanical responses of the tissues are described by the piecewise linearization theory with hypo-elastic relations of fiber aggregation. The evolution equations of viscoplasticity are formulated from the dissipation inequalities by the co-directionality hypotheses. The anisotropy is considered in the hypo-elastic relations and viscoplastic flow rules by introducing some material parameters dependent on the loading direction. Then the capability of the proposed model to describe the nonlinear time-dependent deformation of soft biological tissues is verified by comparing the predictions with the corresponding experimental results of three tissues. It is seen that the predicted monotonic stress-strain responses, stress relaxation, creep and ratchetting of soft biological tissues are in good agreement with the corresponding experimental ones.

Abstract:Cellular viscoelastic modulus in skeletal muscle tissue responds dynamically to chronic stressors, such as age and exercise. Passive tissue mechanics may also be sensitive to acute stimuli such as mechanical loading and/or activation-induced muscle fatigue. These insights are largely derived from preclinical studies of age and acute muscle activation. Therefore, we sought to understand the relative responsiveness of muscle cellular passive mechanics to chronic (resistance training) and acute (muscle fatigue) stressors in healthy young males and females categorized as Resistance Trained or Untrained. We measured passive mechanics to test the hypothesis that Young's Modulus and stress would be greater in fibers from trained versus untrained participants and both would be reduced following fatigue. We further assessed the translation of these findings to composite tissue in a sub-set of volunteers where muscle tissue bundles, containing both fibers and extracellular matrix, were analyzed in addition to single fibers. We report a main effect of training such that cellular passive mechanical measures were increased in single fibers from trained versus untrained participants. We likewise report reductions in passive mechanical measures following fatiguing exercise. Surprisingly, both training and acute fatigue only impacted muscle fiber passive measures in males, whereas females showed a more variable response across conditions. Last, we provide preliminary evidence supporting the translation of per-individual cellular differences to the tissue level. Together, these data suggest males respond more dynamically to acute and chronic stressors of muscle tissue mechanics, potentially linking cellular response and sex-dependent differences in musculotendinous injury risk.

Structural Changes of Active Skeletal Muscles: Modelling, Validation and Numerical Experiments

Abstract:The purpose of this study was to report numerical validation of a 3D finite element model of contracting muscle. The model was based on continuum theory for fibre-reinforced composite materials. Here we simulated contractions for an idealized medial gastrocnemius muscle in man, using the model. Simulations were performed to test the force-length relation of the whole muscle, to evaluate the changes in internal fascicle geometry during contractions, and to assess the importance of material formulations for the aponeurosis and tendon. The simulation results were compared to previously published experimental values. The force-length profile for the whole muscle showed a realistic profile. As the muscle contracted the fascicles curved into S-shaped trajectories and curled around 3D paths, both of which matched previous experimental findings. As the fascicles shortened they increased in their cross-sectional area, but this increase was asymmetric with the smaller increase occurring within the fascicle-plane: the Poisson's ratio in this plane matched that previously shown from ultrasound imaging. The distribution of strains in the aponeurosis and tendon was shown to be a function of their material properties. This study demonstrated that the model could replicate realistic patterns of whole muscle-force, and changes to the internal muscle geometry, and so will be useful for testing mechanisms that affect the structural changes within contracting muscle.

Abstract:Summary form only given, as follows. Reconfigurable processors, like the Field Programmable Gate Arrays (FPGAs), open new computational paradigms where the processor is able to tailor its internal structure to better implement a given application. A typical FPGA consists of an array of configurable logic blocks and a mesh of interconnections fully programmable by the user to perform a given application. By just changing its internal connectivity, the FPGA can implement a totally different new function. However in most of the applications, the FPGA is configured only once and used as coprocessor to carry out some highly complex or time-consuming computation. The reason for such limitation is the small communication bandwidth between the FPGA chip and the external memory, usually ROM, where the configuration data is stored. The Optically Programmable Gate Array (OPGA), an enhanced version of a conventional. FPGA, can overcome this problem. The OPGA utilizes a holographic memory accessed by an array of VCSELs to program its logic. The on-chip logic has been complemented with an array of photodetectors to detect the configuration template recorded in the memory. Combining spatial and shift multiplexing to store the configuration pages in the memory, the OPGA module is very compact and has extremely short configuration time allowing for dynamic reconfiguration. The reconfiguration capability of the OPGA can be applied to solve more efficiently problems in pattern recognition and database searches.
Viscoelastic properties of the passive mechanical behavior of the porcine carotid artery: influence of proximal and distal positions

Abstract:The viscoelastic properties of porcine carotid tissue are investigated in this work. Experimental uniaxial stress relaxation tests along the longitudinal and circumferential directions of the vessel were performed for carotid strips extracted from 10 vessels. Directional and local differences--distal versus proximal position--in the tissue behavior were investigated. The experimental tests reveal a highly anisotropic, non-linear viscoelastic response and local dependence of the samples. The carotid artery shows anisotropic relaxation behavior for both proximal and distal samples. The highest stress relaxation was found in the circumferential tensile test for the highest applied strain at the distal position. For the circumferential direction, the relaxation stress was higher than in the longitudinal being at its highest in the distal position. These facts show that the stress relaxation is higher in the distal than in the proximal position. However, there are no differences between both positions in the longitudinal direction. In addition, a constitutive law that takes into account the fundamental features, including non-linear viscoelasticity, of the arterial tissue is proposed. The present results are correlated with the purely elastic response and the microstructural analysis of the tissue by means of histological quantification presented in a previous study.
Along- and cross-muscle fiber shear moduli in skeletal muscle

Abstract:The material properties of muscle play a central role in how muscle resists joint motion, transmits forces internally, and repairs itself. While many studies have evaluated muscle's tensile material properties, few have investigated muscle's shear properties. The objective of this study was to quantify the shear moduli of skeletal muscle both along (along-muscle fiber) and perpendicular (cross-muscle fiber) to the direction of muscle fibers. We collected data from the extensor digitorum longus, tibialis anterior, and soleus harvested from both hindlimbs of 12 rats. These muscles were chosen to further evaluate the consistency of shear moduli across muscles with different architectures. We applied strains and measured stress in three configurations: parallel, perpendicular, and across the muscle fibers to characterize the along- and cross-muscle fiber tensile and shear material parameters. We found no significant difference between the shear modulus measured parallel to the fibers (along-muscle fiber) and the shear modulus in the plane perpendicular to the fibers (cross-muscle fiber). Although the shear moduli were not significantly different, there was a greater difference with increasing strain, suggesting that there is greater anisotropy at larger strains. We also found no significant difference in moduli between the muscles with differing muscle architecture. These results characterize the shear behavior of skeletal muscle and are relevant to understanding the role of shear in force transmission and injury.

Abstract:A novel constitutive model that describes stress relaxation in transversely isotropic soft collagenous tissues such as ligaments and tendons is presented. The model is formulated within the nonlinear integral representation framework proposed by Pipkin and Rogers (J. Mech. Phys. Solids. 16:59–72, 1968). It represents a departure from existing models in biomechanics since it describes not only the strain dependent stress relaxation behavior of collagenous tissues but also their finite strains and transverse isotropy. Axial stress–stretch data and stress relaxation data at different axial stretches are collected on rat tail tendon fascicles in order to compute the model parameters. Toward this end, the rat tail tendon fascicles are assumed to be incompressible and undergo an isochoric axisymmetric deformation. A comparison with the experimental data proves that, unlike the quasi-linear viscoelastic model (Fung, Biomechanics: Mechanics of Living Tissues. Springer, New York, 1993) the constitutive law can capture the observed nonlinearities in the stress relaxation response of rat tail tendon fascicles.

Abstract:Background: The single-axis extension test is relatively little used to study the mechanical properties of human skin in vivo. A campaign of tests was carried out with an original, modern machine developed in our laboratory. It can perform extension or compression tests using servo-controlled position or force in different directions. The load can either be of the extension or monotonous compression type, creep or relaxation. The results obtained were used to develop a viscoelastic model. The elastic modulus calculated helps us to determine the main directions of anisotropy on the forearm. Methods: We use a new in vivo single-axis extension machine (patent no. FR03/09220 application in progress). With it, we can carry out monotonous, creep and relaxation tests on the forearm. An associated finite elements model enables conversion to the intrinsic parameters of the skin under stress and strain from external stress applied in force and displacement. From the tests, we can propose a viscoelastic model and the identification of his parameters. We carried out tests in four directions with respect to the axis of the forearm of 63 people of different ages. The present report is limited to a brief presentation of the experimental set-up used, and a more complete presentation of the viscoelastic model and how it is defined and also the work on the anisotropy in the elastic domain. Results: The viscoelastic model proposed has only four intrinsic parameters: elasticity parameters E(e) and E(ve) and viscosity parameters epsilon(ve) and A. Skin being considered as orthotropic, we were able to determine the average main direction of 63 people, which is of 5.33+/-5.78 around the longitudinal axis of the arm. An average modulus E(1) (ave)=6.57E(5) (Pa) can be found in the direction close to the axis of the arm and E(2) (ave)=1.30E(5) (Pa) in the perpendicular direction and a G(12)=1.32E(5) (Pa) shear modulus. Conclusions: The parameters obtained with the viscoelastic model are independent of the type of load, the same coefficients enable a correct representation in creep and relaxation tests. The main directions vary from one person to another, Young's modulus in these directions could be an indicator for dermatologists and cosmeticians.
Anisotropic Mechanics of Vascular Smooth Muscle Cells Exposed to Dynamic Loads

Abstract:Vascular smooth muscle cells (VSMCs) are the most prevalent cells in the arterial wall. In vivo, arteries are exposed to dynamic biaxial loads; thus, when characterizing VSMC mechanics, it is important to determine their anisotropic and time-dependent mechanical properties. In this work, we use cellular microbiaxial stretching to apply complex deformations to single micropatterned VSMCs and measure the resulting changes in cell stress. Previously, cellular microbiaxial stretching has been used to measure VSMC mechanical properties in response to extensional strain. Here, we measure changes in cell stress in response to both extension and compression. Additionally, we measure immediate temporal changes in stress in response to cyclically applied deformations. We find that the VSMCs display clear hysteresis when incrementally stretched and compressed and demonstrate cycle-dependent stress-relaxation when exposed to cyclic step change extension and compression. Finally, we demonstrate that a Hill-type active fiber model is capable of replicating all observed hysteresis and cycle-dependent stress-relaxation, suggesting that the temporal stress-strain behavior of the cell is regulated by acto-myosin contraction and relaxation, rather than passive viscoelasticity. This study improves upon previous studies of cellular mechanical properties by considering cellular architecture and more complex deformations when measuring the time-dependent mechanical properties of VSMCs. These findings have important implications for modeling in mechanobiology as VSMCs are mechanosensitive and actively respond to changes in their mechanical environment to maintain vascular function.
