Abstract:When adherently growing cells cultured on the surface of the micro-electrodes, impedance detection can provide information related to cellular physiological functions. To establish electric cell-substrate impedance sensing (ECIS) system for detecting of culture morphology and proliferation of cultured mammal cells in a real time, non-invasive and quantitative way, a cell based biosensor with 20 channel gold micro-electrode arrays (MEA) was designed in this study. Based on micromechanical systems, diameters of these electrodes were fabricated about 20~50 micron on silicon wafe. After human osteoblasts Saos-2 cell lines were cultured on the surface of the electrodes about 24 h later, the impedance increase was mainly focused on 102～104 Hz of the frequency spectroscopy. The impedance detecting result was well according with analysis of the impedance models of the theoretic analysis. Therefore, this cell-based impedance biosensor with micro-electrodes can be used for studies of cell physiologies and drug screenings.

Abstract:In this study, cell biosensor chip with micro-electrode arrays was used to monitor the culture behaviour of mammalian cancer cells and evaluate the chemosensitivity of anti-cancer drugs using electrochemical impedance spectroscopy. The electrode arrays were fabricated by semiconductor technology with diameter from 50 pm to 10 pm of each electrode. The human oesophageal cancer cell lines (KYSE 30) were cultured on the surface of the electrode with the help of fibronectin. Morphology changes of cells adhesion, spreading, and proliferation can be detected by impedimetric analysis in a real time and non-invasive way. Anti-caner drug of cisplatin was added to cells for potential drug screening applications. The experimental results show that drug's chemosensitivity effects can be detected by this cell based biosensor.

Abstract:Activation of invasion and metastasis is recognized as one of the hallmarks of cancer. There are 90% of cancer-related deaths due to metastasis and given that it is worthy of note to study cancer progression and metastasis. Owing to restricted tools used to underpin the study of tumor invasion process, an on-site platform was developed to monitor this event in vitro. We used interdigital gold electrodes to monitor the dynamic process of cancer cells invading into extracellular matrix in situ continuously. Influences of collagen concentration and number of cancer cells on the measured impedance was exhibited. In addition, the parameters used to demonstrate the experiment results were optimized. The change of impedance magnitude indicated the cell-matrix interaction during invasion process. The potential further use of this platform would be complementary in cell studies when concerning metastasis.
Distinguishing cancerous from non-cancerous cells through analysis of electrical noise

Abstract:Since 1984, electric cell-substrate impedance sensing (ECIS) has been used to monitor cell behavior in tissue culture and has proven sensitive to cell morphological changes and cell motility. We have taken ECIS measurements on several cultures of non-cancerous (HOSE) and cancerous (SKOV) human ovarian surface epithelial cells. By analyzing the noise in real and imaginary electrical impedance, we demonstrate that it is possible to distinguish the two cell types purely from signatures of their electrical noise. Our measures include power-spectral exponents, Hurst and detrended fluctuation analysis, and estimates of correlation time; principal-component analysis combines all the measures. The noise from both cancerous and non-cancerous cultures shows correlations on many time scales, but these correlations are stronger for the non-cancerous cells.

Abstract:In this paper, a novel microelectrical impedance spectroscopy (μEIS) with three-dimensional interdigitated electrodes (3D-IDE) is developed to differentiate normal and cancerous cells. The device utilizes a microfluidic tunnel structure, which forces cells to be squeezed. Thus, the enlarged contact area between cells and electrodes allows the device to measure the intrinsic electrical signal of the cells with a higher sensitivity than a noncontact case. The cell squeezing is realized by fabricating smaller microfluidic channel than cell size. The electrical impedances are measured through 3D-IDE. The device well distinguishes normal human breast cell (MCF-10A) and early-stage human breast cancer cell (MCF-7) with a phase difference of 1.42° at 500 kHz. The proposed device also features a high repeatability because the phase change is as small as 0.27° (which is sufficiently smaller than the phase difference between normal and cancer cell) before and after the each cell assay.

Electrical impedance characterization of normal and cancerous human hepatic tissue

Abstract:The four-electrode method was used to measure the ex vivo complex electrical impedance of tissues from 14 hepatic tumors and the surrounding normal liver from six patients. Measurements were done in the frequency range 1-400 kHz. It was found that the conductivity of the tumor tissue was much higher than that of the normal liver tissue in this frequency range (from 0.14 +/- 0.06 S m(-1) versus 0.03 +/- 0.01 S m(-1) at 1 kHz to 0.25 +/- 0.06 S m(-1) versus 0.15 +/- 0.03 S m(-1) at 400 kHz). The Cole-Cole models were estimated from the experimental data and the four parameters (rho(0), rho(infinity), alpha, f(c)) were obtained using a least-squares fit algorithm. The Cole-Cole parameters for the cancerous and normal liver are 9 +/- 4 Omega m(-1), 2.2 +/- 0.7 Omega m(-1), 0.5 +/- 0.2, 140 +/- 103 kHz and 50 +/- 28 Omega m(-1), 3.2 +/- 0.6 Omega m(-1), 0.64 +/- 0.04, 10 +/- 7 kHz, respectively. These data can contribute to developing bioelectric applications for tissue diagnostics and in tissue treatment planning with electrical fields such as radiofrequency tissue ablation, electrochemotherapy and gene therapy with reversible electroporation, nanoscale pulsing and irreversible electroporation.

Abstract:It is commonly accepted that electrical impedance provides relevant information about the physiological condition of living tissues. Currently, impedance measurements are performed with relatively large surface electrodes not suitable for studies in micro tissue due to their poor spatial resolution and high ESEI that they cause on the surface of skin. A minimally invasive electrical impedance measurement technique of living tissues using needle shaped electrode is described in this paper. A series of experiments in ovum gallinaceums was performed to demonstrate the feasibility of the probes and the measurement system to decrease diagnostics uncertainty. Ovum impedance measurements with the minimally invasive probe showed that the effect of the outer layer membrane (the stratum corneum of skin) was substantially reduced compared to regular non-invasive impedance. Minimally invasive probes resulted in significant changes of all measured parameters.

Abstract:In this paper, impedance measurement of electrolyte–insulator–semiconductor (EIS) structure with high spatial resolution was proposed to monitor cell adhesion. The light addressing ability of this work overcomes the geometrical restrict of cell culture on conventional impedance detection devices such as interdigitated electrode (IDE) and electric cell-substrate impedance sensing (ECIS). Instead of studying cells on predetermined sites of IDE and ECIS, cells cultured anywhere on EIS sensor surface can be addressed and selected as target cells. Principle and primary models for high resolution impedance detection were described and tested by experiments. The EIS sensor was investigated in terms of its intrinsic characteristics, like impedance behavior, voltage characteristic, frequency dependency and photovoltaic effect. Optimized working condition was studied for cell experiments. Cell adhesion under treatment of 0.1% Triton X-100 was monitored using rat kidney cells as the source. Results showed good sensitivity (10% change of impedance) and resolution (40μm) for cell adhesion impedance detection and suggested this work should be suitable for monitoring cell impedance. Further improvements on sensitivity, spatial resolution were discussed as well as the further applications for single cell monitoring and cell adhesion imaging.

Abstract:Frequency dependent cellular micro-impedance estimates obtained from a gold two-electrode configuration using phase sensitive detection have become increasingly used to evaluate cellular barrier model parameters. The results of this study show that cellular barrier function parameter estimates optimized using measurements obtained from this biosensor are highly susceptible to both time dependent and systematic instrumental artifacts. Based on a power spectral analysis of experimentally measured microelectrode voltages, synchronous, 60 Hz, and white Gaussian noise were identified as the most significant time dependent instrumental artifacts. The reduction of these artifacts using digital filtering produced a corresponding reduction in the optimized model parameter fluctuations. Using a series of instrumental circuit models, this study also shows that electrode impedance voltage divider effects and circuit capacitances can produce systematic deviations in cellular barrier function parameter estimates. Although the implementation of an active current source reduced the voltage divider effects, artifacts produced by coaxial cable and other circuit capacitive elements at frequencies exceeding 1 kHz still remained. Reducing time dependent instrumental fluctuations and systematic errors produced a significant reduction in cellular model barrier parameter errors and improved the model fit to experimental data

Abstract:Impedimetric analysis on adherently growing cells by micro-electrodes provides information related to cell number, cell adhesion and cellular morphology. In this study, cell-based biosensor with micro-electrode arrays (MEAs) was used to monitor the culture behavior of mammalian cancer cells and evaluate the chemosensitivity of anti-cancer drugs using electrochemical impedance spectroscopy. The platinum electrode arrays were fabricated by semiconductor technology to a 10 x 10 pattern, with diameter of 80 microm of each electrode. The human oesophageal cancer cell lines (KYSE 30) were cultured on the surface of the electrodes with the pre-coated fibronectin, the connecting protein for tumor cells metastasis and adhesion in extracellular matrix. Morphology changes during cells adhesion, spreading, and proliferation can be detected by impedimetric analysis in a real time and non-invasive way. Cisplatin was added to cells for potential drug screening applications. The experimental results show that this well-known anti-cancer drug has characteristic chemosensitivity effects on KYSE 30 cells which can be detected by MEA. Thus, this cell-based chip provides a useful analytical method for cancer research.

Abstract:Here, we reported a study on the detection and electrical characterization of both cancer cell line and primary tumor cells. Dielectrophoresis (DEP) and electrical impedance spectroscopy (EIS) were jointly employed to enable the rapid and label-free differentiation of various cancer cells from normal ones. The primary tumor cells that were collected from two colorectal cancer patients, cancer cell lines (SW-403, Jurkat, and THP-1), and healthy peripheral blood mononuclear cells (PBMCs) were trapped first at the level of interdigitated microelectrodes with the help of dielectrophoresis. Correlation of the cells dielectric characteristics that was obtained via electrical impedance spectroscopy (EIS) allowed evident differentiation of the various types of cell. The differentiations were assigned to a "dielectric phenotype" based on their crossover frequencies. Finally, Randles equivalent circuit model was employed for highlighting the differences with regard to a series group of charge transport resistance and constant phase element for cancerous and normal cells.

Abstract:The aim of this study was to detect expression of different cytokines in epithelial ovarian carcinoma (EOC) cells and normal ovarian surface epithelial (OSE) cells in vitro and the levels of those with elevated expression in the EOC patients, and to analyze the contribution of cytokine profiles to tumor immune deficiency. Cytokine antibody array was used to detect cytokine profiles in two cell lines of EOC (SKOV3 and CaoV3), primarily cultured EOC and OSE cells. The levels of leukemia inhibitory factor (LIF), interleukin-10 (IL-10), IL-4, and transforming growth factor-beta 1 (TGF-beta 1) in peritoneal fluids and sera in the patients with EOC and benign gynecological tumors were detected by enzyme-linked immunosorbent assay. The levels of LIF, IL-10, and IL-4 were detected two times higher in the culture supernatants of the EOC cell lines than those in OSE cells by cytokine antibody array. Both LIF and IL-10 levels were more increased in ascites of EOC patients than in those in benign gynecological tumor patients (P < 0.05). The level of IL-4 was not detectable in any samples of ascites or sera. No difference of TGF-beta 1 value was detected between patients with EOC and benign gynecological tumors. Epithelium ovarian carcinoma cells can produce more LIF, IL-10 and IL-4 than OSE cells, and contribute to the elevated levels of those cytokines in EOC patients, which probably participates in the development of immune deficiency in the peritoneal cavity of EOC patients.

Abstract:In this work, cell adhesion and electroporation effects have been studied through impedance measurements. Chinese Hamster Ovary (CHO-K1) cells have been plated and grown adherent to multi-electrode arrays and then stimulated to obtain the electroporation. We used Electrochemical Impedance Spectroscopy (EIS) measurements to analyze the impedance variation that occurs with cell adhesion and, consequently, to obtain the intrinsic electrical parameters of the electrode-cell interface through a lumped parameter model. The whole model allows to estimate the correction factor β and the sealing resistance Rseal, both dependent on the cell coverage and adhesion. The electroporation has been performed after cell adhesion, using a custom stimulation bench, able to evaluate the cell-electrode coupling and to stimulate an individually addressable site. By performing EIS measurements, before and after the electroporation, cell status, adhesion and membrane resistance can be detected. From measurements, an impedance decrease as a function of pulse amplitude can be noted. The proposed equivalent electrical model allows to evaluate the variation in membrane conductance due to pores formation. Moreover, average pore radius between 0.5 nm and 2.2 nm, increasing with pulse amplitude, was estimated.

Abstract:Electrochemical impedance spectroscopy (EIS) is widely accepted as an effective and non-destructive method to assess cell health during cell-culture. However, there is a lack of compact devices compatible with microfluidic integration and microscopy that could provide the real-time and non-invasive monitoring of cell-cultures using EIS. In this paper, we reported the design and characterization of a modular EIS testing system based on a patented technology. This device was fabricated using easily processable methodologies including screen-printing of the impedance electrodes and molding or micromachining of the cell culture chamber with an easy assembly procedure. Accordingly, to obtain processable, biocompatible and sterilizable electrode materials that lower the impact of interfacial impedance on TEER (Transepithelial electrical resistance) measurements, and to enable concomitant microscopy observations, we optimized the formulation of the electrode inks and the design of the EIS electrodes, respectively. First, electrode materials were based on carbon biocompatible inks enriched with IrOx particles to obtain low interfacial impedance electrodes approaching the performances of classical non-biocompatible Ag/AgCl second-species electrodes. Secondly, we proposed three original electrode designs, which were compared to classical disk electrodes that were optically compatible with microscopy. We assessed the impact of the electrode design on the response of the impedance sensor using COMSOL Multiphysics. Finally, the performance of the impedance spectroscopy devices was assessed in vitro using human airway epithelial cell cultures.

Abstract:In this paper, a quantitative modeling and wound-healing analysis of fibroblast and human keratinocyte cells is presented. Our study was conducted using a continuous cellular impedance monitoring technique, dubbed Electric Cell-substrate Impedance Sensing (ECIS). In fact, we have constructed a mathematical model for quantitatively analyzing the cultured cell growth using the time series data directly derived by ECIS in a previous work. In this study, the applicability of our model into the keratinocyte cell growth modeling analysis was assessed first. In addition, an electrical "wound-healing" assay was used as a means to evaluate the healing process of keratinocyte cells at a variety of pressures. Two innovative and new-defined indicators, dubbed cell power and cell electroactivity, respectively, were developed for quantitatively characterizing the biophysical behavior of cells. We then employed the wavelet transform method to perform a multi-scale analysis so the cell power and cell electroactivity across multiple observational time scales may be captured. Numerical results indicated that our model can well fit the data measured from the keratinocyte cell culture for cell growth modeling analysis. Also, the results produced by our quantitative analysis showed that the wound healing process was the fastest at the negative pressure of 125mmHg, which consistently agreed with the qualitative analysis results reported in previous works.

Abstract:Electrical cell-substrate impedance sensing is increasingly being used for label-free and real-time monitoring of changes in cell morphology and number during cell growth, drug screening, and differentiation. In this study, we evaluated the feasibility of using ECIS to monitor C2C12 myoblast differentiation using a fabricated indium tin oxide (ITO) electrode-based chip. C2C12 myoblast differentiation on the ITO electrode was validated based on decreases in the mRNA level of MyoD and increases in the mRNA levels of myogenin and myosin heavy chain (MHC). Additionally, MHC expression and morphological changes in myoblasts differentiated on the ITO electrode were comparable to those in cells in the control culture dish. From the monitoring the integration of the resistance change at 21.5 kHz, the cell differentiation was label-free and real-time detectable in 30 h of differentiation (p < 0.05).

Abstract:A PDMS-glass based micro-device was designed and fabricated with 12 coplanar impedance sensors integrated for electrical cell–substrate impedance sensing (ECIS). The sensitivity and frequency characteristics of the sensors were investigated both theoretically (equivalent circuit model) and experimentally for the commonly used micro-electrode dimension scale (20–80μm). The experimental results matched well with the theoretical model analysis and revealed that, within this micro-electrode dimension scale, as the electrode width decreased or as the total electrode length decreased the sensitivity of sensor increased over the whole sensing frequency range, whilst electrode to electrode distance had no influence on sensitivity. Through our frequency characteristics analysis, the whole frequency range could be divided into four parts. New functions describing the dominant components in each frequency range were defined and validated experimentally, and could be used to explain the phenomenon of an ECIS sensing frequency window. The contribution to the impedance measurement of cells growing on the edges of the electrodes was determined for the first time. Finally, novel proposals for ECIS sensor design and ECIS measurements were presented.
