Abstract:An assay for cadmium in whole blood and urine using deuterium background-correction electrothermal atomic absorption spectroscopy (D(2)-ETAAS) was developed. Cadmium (in a 1- to 2-ml sample) was bound to 15 mg anion-exchange resin, interfering ions were removed in a 2-ml Bio-Spin column, and cadmium was extracted into 100 microl 1M nitric acid for analysis. Cadmium in the sample extract was concentrated 7-fold for blood and 10-fold for urine over the starting material. These steps produced cadmium atomic absorption traces with high signal to background ratios and allowed analysis against aqueous standards. At approximately 0.1 ng Cd/ml, mean intra- and interassay coefficients of variation were 11-12%. Cadmium recovery for 0.1 to 0.6 ng added cadmium was 107+/-4% for blood and 94+/-4% for urine (mean+/-SE, n=3). The mean detection limit (mean + 3 x SD of blank) was 0.008 ng/ml for blood and 0.003 ng/ml for urine. Samples from "unexposed" animals including humans ranged from 0.051+/-0.000 to 0.229+/-0.035 ng/ml. Values were approximately 10-fold lower than those obtained by the method of Stoeppler and Brandt using Zeeman background-correction ETAAS. This new high-sensitivity, low-volume assay will be useful for epidemiological studies, even those involving children, and will provide a means to help determine the contribution of cadmium to disease incidence in the general population.

Abstract:Estimation of heavy metals in water has been paid significant attention because of the toxicity of such metals on the whole ecological system. A microelectrode array (MEA) of individually addressable microelectrodes was fabricated for cadmium, lead and copper analysis in water pollution. The ratio of interelectrode distance to the electrode diameter of the MEA was sufficient enough to get a collective current response while maintaining the excellent features of single microelectrodes. Cyclic voltammograms made in various scan rates confirmed that a high spatial resolution MEA was constructed with no diffusion layers overlapping. Then the mercury film Au-MEA was utilized to detect cadmium, lead and copper ions using differential pulse anodic stripping voltammetry (DPASV). The sensitivity of 130nA/μg, 52nA/μg and 46nA/μg for Cd, Pb and Cu ions, separately, were achieved using this microsensing chip. Calibrations were made via partial least-squares regression (PLSR) modeling. Well correlation was found between the lab-determined values and the values predicted from PLSR calibration.

Abstract:The aim of this work is to propose a biomonitoring method for the simultaneous determination of Cd and Pb in whole blood by simultaneous electrothermal atomic absorption spectrometry for assessment of environmental levels. A volume of 200 microL of whole blood was diluted in 500 microL of 0.2% (wv(-1)) Triton) X-100+2.0% (vv(-1)) HNO3. Trichloroacetic acid was added for protein precipitation and the supernatant analyzed. A mixture of 250 microg W+200 microg Rh as permanent and 2.0% (wv(-1)) NH4H2PO4 as co-injected modifiers were used. Characteristic masses and limits of detections (n=20, 3s) for Cd and Pb were 1.26 and 33 pg and 0.026 microg L(-1) and 0.65 microg L(-1), respectively. Repeatability ranged from 1.8 to 6.8% for Cd and 1.2 to 1.7% for Pb. The trueness of method was checked by the analysis of three Reference Materials: Lyphocheck Whole Blood Metals Control level 1 and Seronorm Trace Elements in Whole Blood levels 1 and 2. The found concentrations presented no statistical differences at the 95% confidence level. Blood samples from 40 volunteers without occupational exposure were analyzed and the concentrations ranged from 0.13 to 0.71 microg L(-1) (0.32+/-0.19 microg L(-1)) for Cd and 9.3 to 56.7 microg L(-1) (25.1+/-10.8 microgL(-1)) for Pb.

Abstract:Microelectrode array (MEA) was used for in situ heavy metal determination in aquatic environments due to its characteristics of high sensitivity, large current density and fast mass transfer rate, while interferences caused by background factors and complex external environment were inevitable in analysis leading to inaccurate results. Hence, two algorithms, partial least square regression and local optimum method were employed in different situations for electrochemical analysis to eliminate interference. Partial least square regression was superior to local optimum method thanks to convenient operations and good outcomes for in situ determination, which was proved to be effective in low interference samples. Nevertheless, inaccurate results were obtained in samples of complicated composition because of negative impacts on working electrode. Local optimum method was proposed aiming at those situations, by which severe interference was ignored and better results were acquired to prove the reasonability and validity of the algorithm. The determination of cadmium and lead was improved effectively combined PLS with local optimum method.
Determination of cadmium in blood, plasma, and urine by electrothermal atomic absorption spectrophotometry after isolation by anion-exchange chromatography

Abstract:The determination of cadmium in whole blood, urine, or plasma by atomic absorption using electrothermal atomization is described. In preparation for atomic absorption analysis, cadmium was concentrated on an anion-exchange column, significantly lowering the limit of detection and allowing for the first time the accurate and precise determination of plasma cadmium concentrations in persons/animals with low-level cadmium exposures. Recovery of 109Cd from spiked whole blood, plasma, and urine into supernatants of nitric acid-deproteinated samples averaged 99, 100, and 95%, respectively. Anion-exchange isolation of the anionic chlorocadmium complex removed 99.8% of the major elements associated with a deproteinated whole blood sample. The recovery of 109Cd from the anion-exchange column was 92.2 +/- 0.9% (mean +/- SE, N = 35). The separation of cadmium from constituents in blood, urine, or plasma in this manner allowed comparison of unknown samples to aqueous standards with a defined acid matrix using commercially available acids. The mean intra-assay coefficient of variation (CV) was 12 +/- 3% (mean +/- SE, N = 6) for blood, plasma, and urine samples having cadmium concentrations of 0.1-0.8 microgram/liter. The interassay CV was 13% (N = 7) for a blood sample containing 0.6 microgram Cd/liter. The recovery of known amounts of cadmium added to blood, plasma, and urine in the range of 0.2 to 5.0 micrograms Cd/liter was 97 +/- 6% (mean +/- SE, N = 4).

Abstract:The determination of Cd in whole blood and urine by electrothermal atomic absorption spectrometry using Pd-based modifiers and in situ decontamination was studied. The performance of Pd, Pd + NH4NO3, and Pd + Mg(NO3)2 as modifiers were compared with respect to maximum pyrolysis and atomization temperatures, background attenuation, linear calibration range and matrix interference. Samples were diluted (1 + 4) with 0.1% Triton X-100 (blood) or 0.2% HNO3 (urine), and introduced onto the graphite tube platform already containing the in situ dried and decontaminated modifier. The influence of modifier volume and concentration, as well as that of the dilution ratio was investigated. The proposed procedure involves the use of Pd(NO3)2 + (Mg(NO3)2 as modifier, a pyrolysis temperature of 700 degree C and an in situ decontamination step of 1200 degrees C. The atomization temperature was 1700 degrees C. The detection limit (n = 10, k = 3) was 0.22 micrograms l-1 for both samples. Typical relative standard deviation values (n = 3) were below 3%. Using matrix-matched standards, good agreement was observed between experimental and reference values in the analysis of certified reference materials. Interlaboratory comparison data have confirmed the validity of the proposed analytical procedure.
Bismuth-Coated Thick-Film Carbon Electrodes for the Detection of Lead and Cadmium by Anodic Stripping Voltammetry

Abstract:This paper reported the detection of trace heavy metal ions, lead and cadmium, with in-situ bismuth-coated thick-film carbon electrodes by anodic stripping voltammetry. The influence of carbon electrodes pretreatment, bismuth ion concentrations, preconcentration time upon the stripping responses were investigated. Experimental results indicate that anodic stripping voltametric measurements of lead and cadmium on bismuth-coated carbon electrodes yield well-defined peaks, whose peak potential and height are in a manner analogous to those of in-situ plated mercury-film electrodes. Using bismuth instead of mercury a-voids the pollution. Due to the low cost of thick-film process, the thick-film carbon electrode could be used as disposable device.

Abstract:Ultrasound-assisted extraction is compared with slurry sampling and microwave-assisted acid digestion for the determination of cadmium, lead and copper in biological samples and cadmium in sediment samples by electrothermal atomic absorption spectrometry (ETAAS). The main advantage of using an ultrasound-assisted extraction in comparison to slurry sampling is a doubling of the tube lifetime for all the chosen analytes, most likely because the sample matrix was not introduced into the atomiser, avoiding the build up of carbonaceous residues or silicates on the graphite platform. Consequently the analytical variable costs were remarkably diminished. In comparison to microwave-assisted digestion, ultrasound-assisted extraction resulted typically in a two-fold better precision and better detection limits. For the extraction, slurries in nitric acid were sonicated for 2–5 min using a titanium probe. Subsequently, the solid phase was separated by centrifugation, and 20 µl of the liquid phase was introduced into a graphite tube previously treated with a W–Rh permanent modifier. For slurry sampling, the slurries were sonicated for 20 s before being delivered to the W–Rh treated platform. Nitric acid concentration, sonication time, sonication power, sample mass and particle size were optimised for each analyte to obtain quantitative recoveries. For cadmium the optimum conditions (sediments and biological materials) were, 0.2–3.0% m/v slurry, 0.5% HNO3, 2 min sonication at 15 W. For copper and lead (biological materials only) the optimum conditions were 0.2–2.0% m/v slurry, 5.0% HNO3, 5 min 35 W. The particle size chosen for all analytes was <30 µm. For biological materials, the extraction recoveries were: Cd, 95–105%; Cu, 90–101%; and Pb, 88–98%. However, only cadmium was quantitatively extracted (based on the certified values of the reference materials) from sediment samples (recoveries 92–95%). For Pb and Cu the maximum recoveries attained were 54% and 60% respectively. Detection limits, based on integrated absorbance for the extracts of a 2.5% m/v biological material were: Cd, 3.0 ng g−1; Cu, 88 ng g−1; and Pb, 55 ng g−1. For 0.5% m/v sediment the cadmium detection limit was 15 ng g−1.

Abstract:Environmental pollution from trace elements has been increasing in recent decades and has become an important concern for environmental agencies. The trace elements arsenic, cadmium, chromium, copper, mercury, and lead are among the elements that cause the greatest environmental impact and carry the highest risk to human health. Electrothermal atomic absorption spectrometry (ETAAS) has long been employed in trace element determination. In the last few years the main constraints of spectroscopy absorption methods have been overcome. These advances have increased the possibilities and the utility of ETAAS for trace element determination at μg L−1 levels in difficult matrices such as soils and sediments, giving greater accuracy and precision, lower economic cost, and easier sample pretreatment than other methods. The main advances come from sample manipulation for matrix destruction and preconcentration, the use of new lab-on-valve FIA systems, the solid sampling, the use of new, more efficient modifiers and in situ trapping methods for analyte stabilization and pre-concentration, and the progress in the capacity to control the atomization temperature and to correct background spectral interferences. All of them have permitted an improvement in the sensitivity, decreasing the detection limits and manipulation process, and increasing the accuracy and precision of the analyses. Moreover, the new technology in the optic and detector systems have given rise to high-resolution continuum source ETAAS (HR-CS ETAAS) spectrometers that solve most of the constraints presented by the more conventional line source ETAAS (LS ETAAS) spectrometers. HR-CS ETAAS enables a rapid detection of several elements at once, facilitates direct determination from solid sampling, and reduces the matrix interferences and background noise. Here we give an overview of the recent advances and the different possibilities of using ETAAS, drawing on studies from the last decade on methods to analyze As, Cd, Cu, Hg, and Pb in soils and sediments.

Abstract:A new zonal wavefront reconstruction method for lateral shearing interferometry was presented. The proposed algorithm allows shear amounts equal to arbitrary integral multiple of the sample intervals. High spatial resolution reconstruction is achieved with only two difference wavefronts measured in orthogonal shear directions. The presented algorithm was generalized to be applicable for general aperture shape by using zero padding and Gerchberg-type iterative methods. The capability of the presented algorithm was demonstrated by some numerical examples. Also, the reconstruction error was analyzed theoretically and numerically.
Determination of arsenic, selenium and cadmium in marine biological tissue samples using a stabilised temperature platform furnace and comparing deuterium arc with Zeeman-effect background correction atomic absorption spectrometry

Abstract:Arsenic, selenium and cadmium are determined in three marine biological tissue samples from an inter-calibration exercise and NBS Standard Reference Material oyster tissue using graphite furnace atomic absorption spectrometry. Deuterium arc background correction is plagued by severe spectral interferences, which cause substantial signal distortion. Highly time-resolved signal display is a useful diagnostic tool for this type of interference. Zeeman-effect background correction appears to work satisfactorily for the routine determination of these elements in marine biological tissue samples. Calibration against acid standard solutions can be used for selenium and cadmium, when stabilised temperature platform furnace conditions are applied, but not for arsenic. Agreement with accepted average values of the inter-calibration exercise is generally good. Standard reference materials are only useful in part for methods development because their analytical behaviour is sometimes different from that of the samples investigated.

Abstract:The estimation of environmental and occupational exposure to metals is based on the determination of metal concentrations in biological material. This paper describes methods for determining lead and cadmium in blood, and cadmium, copper, nickel and chromium in urine. The methods are evaluated in view of their linearity, precision, repeatability, reproducibility and detectability. The reliability of the methods was verified by determining lyphilized standards of known metal concentrations (Behring). The reproducibility expressed by the value of relative standard deviation ranged from 1.2% to 7.8%. In the case of lead and cadmium determinations, the reliability of results was confirmed due to the participation of our laboratory in the UK NEQAS external control project, the Queen Elizabeth Hospital, Birmingham. The results of external quality control (MR VIS = 19 for lead, as compared with the mean = 41 for all the project participants and MR VIS = 29 for cadmium, mean = 62) prove undoubtedly correctness of the methods applied. A small volume of sample to be analyzed is their major advantage, and due to adopted analytical parameters the methods can be used to determine metals in biological material in order to assess occupational or environmental exposure.

Abstract:Toxicity, mobility, bioavailability and biofunctions of chromium, cadmium, mercury and lead are heavily dependent upon their specific chemical forms, leading to a high demand to metal speciation analysis rather than total quantification. Simultaneous speciation analysis of multiple metal(loid)s is attractive to a large sample capacity containing unstable analytes due to its economic and environmental advantages over the conventional single elemental strategies. In this work, an analytical method integrating online solid phase extraction into high performance liquid chromatography interfaced with inductively coupled plasma mass spectrometry (ICP-MS) to simultaneously preconcentrate and quantify Cr, Cd, Hg and Pb forms in pg L-1 levels in water was developed. Cr (III + VI), Cd(II), Hg(II), Pb(II), methylmercury (MeHg), ethylmercury (EtHg), and trimethyl (TML) and triethyl lead (TEL) were captured by the C18 adsorbent (equilibrated with 10 mL of 1.0 mM 2-hydroxyethanethiol at 10 mL min-1) and eluted by mobile phase (5.0 mM Cys at pH 2.0), then completely separated on the C18 column within 8.0 min and eventually determined by ICP-MS. Low limits of detection (0.001-0.007 ng L-1) and quantification (0.003-0.023 ng L-1), good relative standard deviations (<4%) and high enrichment factors (827-2656 folds) were obtained with good linearities. Three reference materials of total cadmium (GBW08602), total mercury (GBW08603) and total lead (GBW08601) in water were analyzed by the developed method to validate the accuracy with good agreement with certified values and satisfactory recoveries (92-100%). This method was proved feasible by the determination of Cr, Cd, Hg and Pb compounds in drinking water, river water, pond water and tap water.

Abstract:The applicability of nitric acid, palladium nitrate and a mixture of palladium and magnesium nitrate as matrix modifiers was estimated for the accurate and reproducible determination of cadmium (Cd), lead (Pb) and arsenic (As) in sediments of the Sava River by electrothermal atomic absorption spectrometry, ETAAS. Decomposition of the samples was done in a closed vessel microwave-assisted digestion system using nitric, hydrochloric and hydrofluoric acids, followed by the addition of boric acid to convert the fluorides into soluble complexes. The parameters for the determination of Cd, Pb and As in sediments were optimized for each individual element and for each matrix modifier. In addition, two sediment reference materials were also analyzed. In determination of Cd and Pb, nitric acid was found to be the most appropriate matrix modifier. The accurate and reliable determination of Cd and Pb in sediments was possible also in the presence of boric acid. The use of a mixture of palladium and magnesium nitrate efficiently compensated for matrix effects and enabled the accurate and reliable determination of As in the sediments. Quantification of Cd and As was performed by calibration using acid matched standard solutions, while the standard addition method was applied for the quantification of Pb. The repeatability of the analytical procedure for the determination of Cd, Pb and As in sediments was ?5 % for Cd, ?4 % for Pb and ?2 % for As. The LOD values of the analytical procedure were found to be 0.05 mg/kg for Cd and 0.25 mg/kg for Pb and As, while the LOQ values were 0.16 mg/kg for Cd and 0.83 mg/kg for Pb and As. Finally, Cd, Pb and As were successfully determined in sediments of the Sava River in Slovenia.

Benzo(a)pyrene-DNA adduct formation and removal in mouse epidermis in vivo and in vitro: relationship of DNA binding to initiation of skin carcinogenesis.

Abstract:An antiserum specific for the major benzo(a)pyrene (BP) adduct formed with deoxyguanosine in vivo has been used by enzyme-linked immunosorbent assay to monitor the formation and removal of DNA-bound products in BALB/c mouse epidermis exposed topically to initiating doses of BP and in BALB/c mouse keratinocytes exposed in vitro to BP or its activated derivatives. In mouse epidermal DNA, formation of antibody-recognizable products increased proportionally between doses of 50 and 250 nmol of BP, giving 2.3 to 6.0 fmol/micrograms of DNA, respectively, and reached a plateau of 10 to 11 fmol/micrograms of DNA at doses between 1000 and 1500 nmol. Antibody-recognizable adducts comprised roughly one-half of the total BP-DNA binding, since a 250-nmol dose of [3H]BP yielded 6 fmol/micrograms of DNA by enzyme-linked immunosorbent assay and 12.9 fmol/micrograms of DNA by radiolabeling. Removal of trans-(7R)-N2-(10-[7 beta, 8 alpha, 9 alpha-trihydroxy-7,8,9,10-tetrahydrobenzo(a)pyrene]-yl)-deoxyguanosine adducts was monitored in epidermal DNA of mice exposed to 500 nmol of BP and, although no correction was approximated for DNA turnover in the skin, about one-half of the adducts formed by 24 hr were removed 3 days later, and only 10% remained at the end of a week. BP-DNA binding and removal were also studied in cultured mouse keratinocytes, where proliferating basal cells and terminally differentiating cells can be selectively studied by modulating the Ca2+ concentration of the medium. BP dose-response studies showed that, in cells of different maturation states, BP-DNA adduct levels were similar. Adduct formation greater than 10 to 11 fmol/micrograms (the highest obtained in vivo) was associated with extensive cytotoxicity and cell death. The kinetics of adduct removal was followed in culture under conditions in which dilution by DNA synthesis or cell loss could be monitored. Results of these experiments suggested that initial removal of BP-DNA adducts was more rapid in the differentiating population although, in both populations, 50% of the adduct was removed by 24 hr. The formation of foci resistant to Ca2+-induced terminal differentiation has been associated previously with carcinogen treatment in cultured keratinocytes. Exposure to BP or the antidiol-epoxide, at concentrations producing low cytotoxicity, yielded frequencies of differentiation-altered foci proportional to the dose of the compound used and to the number of DNA adducts formed.
Sensory and affective dimensions of advanced cancer pain

Abstract:The present study was designed to explore the extent to which advanced cancer pain is explicable in terms of both physical pain intensity and affect. Most notably, it expanded on previous findings by more clearly elucidating the relationship between several discrete emotional states and the total experience of cancer pain. One hundred and eleven patients with cancer pain attending a Pain and Symptom Control Clinic were studied. Visual Analogue Scales (VASs) were used to quantify overall pain intensity and the accompanying affect. Then, correlations were calculated to evaluate the relationships both between and within these two variables. Overall, the participants rated both the pain intensity and the negative affect associated with that pain as high. Of the examined affective components of pain, frustration and exhaustion were found to be the most significant. Abstract:Background and aims: Reliable lead screening methods are necessary to support early identification of lead exposure in children. Sample collection using dried blood spots (DBS) offers advantages compared to traditional venipuncture and capillary collection. Here, we describe and compare three lead DBS inductively coupled plasma-mass spectrometry (ICP-MS) methods for lead screening. Materials and methods: Lead was extracted from Whatman 903 protein saver cards punches and analyzed by ICP-MS across three independent clinical laboratories. Each laboratory evaluated the performance of aqueous and matrix-matched DBS calibrators using external quality control samples (WI State of Laboratory of Hygiene Program). Leftover patient samples (n = 39) were used for an interlaboratory comparison of lead DBS. Lead DBS results were compared to whole blood methods. Results: The DBS ICP-MS methods using matrix-matched DBS calibrators had superior performance to the aqueous calibrations. There was a strong correlation between lead measured in DBS (matrix-matched) and whole blood for the three methods evaluated. Conclusion: Lead can be measured accurately by ICP-MS in DBS samples when matrix-matched calibrators are used. External quality control programs are valuable to assess the performance of DBS methods. DBS lead ICP-MS methods are a robust analytical option for lead screening even though the limitations of DBS are well recognized.

Abstract:A new procedure for the preconcentration of trace amounts of free Pb and Cd by disposable pipette extraction (DPX) is proposed herein. Recycled cork is used as a biosorbent and the procedure is completely free of organic solvents. The cork was reduced to a powder and characterized by scanning electron microscopy. Several parameters that influence the preconcentration of Pb and Cd with DPX-cork, such as the sample pH, number of extraction cycles, biosorbent mass, and percentage of acid in the desorption step, were studied. The tolerance of DPX-cork with respect to 10 potential interfering ions was also evaluated. Coupled with the high-resolution continuum source graphite furnace atomic absorption spectrometry (HR-CS GF AAS) technique, the sample preparation method allowed the analytical limits required for quality control (according to the permitted limits established by legislation) to be obtained. The limits of detection for the extraction of a 3.5-mL water sample were 200 ng L−1 for Pb and 100 ng L−1 for Cd. The relative standard deviations were around 7.5% for Pb and 8.0% for Cd. The optimized method was successfully applied to the determination of Cd and Pb in two certified reference materials (water and wastewater) and five water samples (collected from a mangrove, a creek, and the sea).

Abstract:Hydrolases containing two metal ions connected by a bridging ligand catalyze reactions important in carcinogensis, tissue repair, post-translational modification, control and regulation of biochemical pathways, and protein degradation. The aminopeptidase from Aeromonas proteolytica serves as a paradigm for the study of such bridged bimetallic proteases since its three-dimensional structure is known to very high resolution and its catalytic reaction is amenable to spectroscopic examination. Herein, we report the X-ray crystal structure at 1.9 A resolution of AAP complexed with 1-butaneboronic acid (BuBA). This structure suggests that this complex represents a snapshot of the proteolytic reaction in an arrested form between the Michaelis complex and the transition state. Comparison of the structure with spectroscopic and other data allows us to conclude that the apparently structurally symmetrical dizinc site is actually asymmetric electrostatically.
Direct determination of tin in whole blood and urine by GF AAS

Abstract:Objective: The aim of this work was to develop a procedure for the determination of tin in whole blood and urine by GF AAS with a minimum sample pre-treatment, using Pd/Mg as chemical modifier. Design and methods: The analyses of tin were conducted using an atomic absorption spectrometer with Zeeman background correction. The laboratory staff volunteered blood and urine samples for the experimental studies and application of the methodology. Results: Samples were just diluted with 0.2% v/v Triton X-100, and pyrolysis and atomization temperatures of 1300 and 2200 °C were used. External calibration was performed with matrix matched calibration solutions. Limits of detection of 2.7 and 0.8 μgL(-1) were reached for blood and urine, respectively. The method was applied to the determination of Sn in blood and urine of eleven subjects not occupationally exposed, working in a laboratory of toxicology in a large Brazilian city, and the results ranged from 7.4 to 11.2 μgL(-1) and ≤0.8 to 2.2 μgL(-1), for blood and urine, respectively. Accuracy was assessed by analysis of standard reference materials for tin in blood (Contox I, lot TM144-1097, Kaulson Laboratories, USA) and urine (Seronorm, lot 0511545, Sero AS, Norway). Conclusions: Results showed good agreement between experimental and reference values according to the Student's t test at 95% of confidence.
