Abstract:The NH2 termini of light-harvesting complex I (LHI) polypeptides alpha and beta of Rhodobacter capsulatus are thought to be involved in the assembly of the LHI complex. For a more detailed study of the role of the NH2-terminal segment of the LHI alpha protein in insertion into the intracytoplasmic membrane (ICM) of R. capsulatus, amino acids 6 to 8, 9 to 11, 12 and 13, or 14 and 15 of the LHI alpha protein were deleted. Additionally, the hydrophobic stretch of the amino acids 7 to 11 was lengthened by insertion of hydrophobic or hydrophilic amino acids. All mutations abolished the ability of the mutant strains to form a functional LHI antenna complex. All changes introduced into the LHI alpha protein strongly reduced the stability of its LHI beta partner protein in the ICM. The effects on the mutated protein itself, however, were different. Deletion of amino acids 6 to 8, 9 to 11, or 14 and 15 drastically reduced the amount of the LHI alpha protein inserted into the membrane or prevented its insertion. Deletion of amino acids 12 and 13 and lengthening of the stretch of amino acids 7 to 11 reduced the half-life of the mutated LHI alpha protein in the ICM in comparison with the wild-type LHI alpha protein. Under the selective pressure of low light, revertants which regained a functional LHI antenna complex were identified only for the mutant strain deleted of amino acids 9 to 11 of the LHI alpha polypeptide [U43 (pTPR15)]. The restoration of the LHI+ phenotype was due to an in-frame duplication of 9 bp in the pufA gene directly upstream of the site of deletion present in strain U43(pTPR15). The duplicated nucleotides code for the amino acids Lys, Ile, and Trp. Membranes purified from the revertants were different from that of the reaction center-positive LHI+ LHII- control strain U43(pTX35) in doubling of the carotenoid content and increase of the size of the photosynthetic unit. By separating the reaction center and LHI complexes of the revertants by native preparative gel electrophoresis, we confirmed that the higher amount of carotenoids was associated with the LHI proteins.

Abstract:Employing double-isotope derivative techniques, noradrenaline and adrenaline have been determined in plasma and in urine and dopamine in urine in 21 patients with essential hypertension as well as in 32 controls. Plasma noradrenaline rose with age in both groups of subjects. No differences were observed in plasma noradrenaline and plasma adrenaline in the resting supine position and in urinary excretion of noradrenaline and dopamine in hypertensive patients as compared to control subjects. Urinary excretion of adrenaline was somewhat lower in the hypertensives than in the controls. Treatment with alprenolol, a beta-adrenergic blocking agent, did not influence noradrenaline and adrenaline in plasma in the basal state or the urinary excretion of the three catecholamines. The combined treatment with alprenolol and hydralazine was followed by a significant rise in plasma noradrenaline. It is concluded that the adrenergic activity evaluated by circulating catecholamines is normal in most patients with essential hypertension.
Oxygen-insensitive synthesis of the photosynthetic membranes of Rhodobacter sphaeroides: a mutant histidine kinase

Abstract:Two new loci, prrB and prrC, involved in the positive regulation of photosynthesis gene expression in response to anaerobiosis, have been identified in Rhodobacter sphaeroides. prrB encodes a sensor histidine kinase that is responsive to the removal of oxygen and functions through the response regulator PrrA. Inactivation of prrB results in a substantial reduction of photosynthetic spectral complexes as well as in the inability of cells to grow photosynthetically at low to medium light intensities. Together, prrB and prrA provide the major signal involved in synthesis of the specialized intracytoplasmic membrane (ICM), harboring components essential to the light reactions of photosynthesis. Previously, J. K. Lee and S. Kaplan (J. Bacteriol. 174:1158-1171, 1992) identified a mutant which resulted in high-level expression of the puc operon, encoding the apoproteins giving rise to the B800-850 spectral complex, in the presence of oxygen as well as in the synthesis of the ICM under conditions of high oxygenation. This mutation is shown to reside in prrB, resulting in a leucine-to-proline change at position 78 in mutant PrrB (PRRB78). Measurements of mRNA levels in cells containing the prrB78 mutation support the idea that prrB is a global regulator of photosynthesis gene expression. Two additional mutants, PRRB1 and PRRB2, which make two truncated forms of the PrrB protein, possess substantially reduced amounts of spectral complexes. Although the precise role of prrC remains to be determined, evidence suggests that it too is involved in the regulatory cascade involving prrB and prrA. The genetic organization of the photosynthesis response regulatory (PRR) region is discussed.
Comparative Study of Spectral and Functional Properties of Wild Type and Double Mutant H(L173)L/I(L177)H Reaction Centers of the Purple Bacterium Cereibacter sphaeroides

Abstract:Previously, we found that in the reaction center (RC) of the purple bacterium Cereibacter sphaeroides , formation of heterodimeric primary electron donor (P) caused by the substitution of His-L173 by Leu, was compensated by the second mutation Ile-L177 – His. Significant changes in the spectral properties, pigment composition, and redox potential of P observed in the H(L173)L RC, are restored to the corresponding characteristics of the native RC in the RC H(L173)L/I(L177)H, with the difference that the energy of the long-wavelength Q Y optical transition of P increases significantly (by ~75 meV). In this work, it was shown using light-induced difference FTIR spectroscopy that the homodimeric structure of P is preserved in the RC with double mutation with partially altered electronic properties: electronic coupling in the radical-cation of the P + dimer is weakened and localization of the positive charge on one of its halves is increased. Results of the study of the triple mutant RC, H(L173)L/I(L177)H/F(M197)H, are consistent with the assumption that the observed changes in the P + electronic structure, as well as considerable blue shift of the Q Y P absorption band in the RC H(L173)L/I(L177)H, are associated with modification of the spatial position and/or geometry of P. Using femtosecond transient absorption spectroscopy, it was shown that the mutant H(L173)L/I(L177)H RC retains a sequence of reactions P* → P + B A − → P + H A − → P + Q A − with electron transfer rates and the quantum yield of the final state P + Q A − close to those observed in the wild-type RC (P* is the singlet-excited state of P; B A , H A , and Q A are molecules of bacteriochlorophyll, bacteriopheophytin, and ubiquinone in the active A-branch of cofactors, respectively). The obtained results, together with the previously published data for the RC with symmetrical double mutation H(M202)L/I(M206)H, demonstrate that by introducing additional point amino acid substitutions, photochemical activity of the isolated RC from C. sphaeroides could be maintained at a high level even in the absence of important structural elements – axial histidine ligands of the primary electron donor P.

Form follows function -- how PufX increases the efficiency of the light-harvesting complexes of Rhodobacter sphaeroides

Abstract:Some species of purple bacteria as, e.g., Rhodobacter sphaeroides contain the protein PufX. Concurrently, the light harvesting complexes 1 (LH1) form dimers of open rings. In mutants without PufX, the LH1s are closed rings and photosynthesis breaks down, because the ubiquinone exchange at the reaction center is blocked. Thus, PufX is regarded essential for quinone exchange. In contrast to this view, which implicitly treats the LH1s as obstacles to photosynthesis, we propose that the primary purpose of PufX is to improve the efficiency of light harvesting by inducing the LH1 dimerization. Calculations with a dipole model, which compare the photosynthetic efficiency of various configurations of monomeric and dimeric core complexes, show that the dimer can absorb photons directly into the RC about 30% more efficient, when related to the number of bacteriochlorophylls, but that the performance of the more sophisticated dimeric LH1 antenna degrades faster with structural perturbations. The calculations predict an optimal orientation of the reaction centers relative to the LH1 dimer, which agrees well with the experimentally found configuration. For the increased required rigidity of the dimer additional modifications of the LH1 subunits are necessary, which would lead to the observed ubiquinone blockage, when PufX is missing.

Proteomic analysis of the developing intracytoplasmic membrane in Rhodobacter sphaeroides during adaptation to low light intensity

Abstract:Although the primary photochemical events in the facultative photoheterotrophic purple bacterium Rhodobacter sphaeroides are now well understood, comparatively little is known about how their photosynthetic apparatus is assembled. Here we present a proteomic analysis of the intracytoplasmic membrane (ICM) assembly process during adaptation to lowered light intensity, in which the size of the photosynthetic units is greatly expanded by addition of the light-harvesting 2 (LH2) peripheral antenna complex. When the isolated ICM-derived chromatophore vesicles were subjected to clear native gel electrophoresis (CNE), four pigmented bands appeared; the top and bottom bands contained the reaction center - light-harvesting 1 (RC-LH1) core complex and LH2 peripheral antenna, respectively, while the two bands of intermediate migration contained associations of the LH2 and core complexes. Proteomic analysis revealed a large array of other proteins associated with the CNE gel bands - in particular, several F(1)F(O)-ATP synthase subunits gave unexpectedly high spectral counts, given the inability to detect this coupling factor, as well as the more abundant cytochrome bc (1) complex, by atomic force microscopy. Significant levels of general membrane assembly factors were also found, as well as numerous proteins of unknown function including high counts for RSP6124 that were correlated with LH2 levels. When combined with further AFM and spectroscopic studies, these proteomic approaches are expected to provide a much-improved understanding of the overall assembly process.

Abstract:PprI, a DNA damage response factor from the extraordinary radioresistant bacteriumDeinococcus radi-odurans, plays a central regulatory role in multiple DNA damage repair. In this study, a fusion DNA fragment carrying kanamycin resistance gene with theD. radiodurans groEL promoter was cloned by PCR amplification and reversely inserted into thepprI locus in the genome of the wild-type strain R1. The resultingpprI-deficient strain, designated YR1, was very sensitive to ionizing radiation. Meanwhile, the recombinant DNA fragment was cloned into the shuttle vector pRADZ3, and resulted in plasmid pRADK with kanamycin resistance inD. radiodurans. The fragments containing completepprI gene and 3′-terminal deletionpprI † were cloned into plasmid pRADK. The resulted plasmids designated pRADKpprI and pRADKpprI† were then transformed to YR1. Results show that YR1 carrying pRADKpprI was able to fully restore the extreme radioresistance to the same level as the wild-typeD. raiodurans Rl, whereas YR1 pRADKpprI† failed to do so. Construction of DNA repair switch PprI function-deficient and function-complementary mutants inD. radiodurans is not only useful to elucidating the relationship between domains and functions of Pprl protein, but also opens the door to the further studies of the biological functions of PprI proteinin vivo.

Abstract:Purple photosynthetic bacteria (PPB) are versatile microorganisms capable of producing various value-added chemicals, e.g., biopolymers and biofuels. They employ diverse metabolic pathways, allowing them to adapt to various growth conditions and even extreme environments. Thus, they are ideal organisms for the Next Generation Industrial Biotechnology concept of reducing the risk of contamination by using naturally robust extremophiles. Unfortunately, the potential of PPB for use in biotechnology is hampered by missing knowledge on regulations of their metabolism. Although Rhodospirillum rubrum represents a model purple bacterium studied for polyhydroxyalkanoate and hydrogen production, light/chemical energy conversion, and nitrogen fixation, little is known regarding the regulation of its metabolism at the transcriptomic level. Using RNA sequencing, we compared gene expression during the cultivation utilizing fructose and acetate as substrates in case of the wild-type strain R. rubrum DSM 467 T and its knock-out mutant strain that is missing two polyhydroxyalkanoate synthases PhaC1 and PhaC2. During this first genome-wide expression study of R. rubrum , we were able to characterize cultivation-driven transcriptomic changes and to annotate non-coding elements as small RNAs. Abstract:Rhodobacter ( Rba .) capsulatus has been a favored model for studies of all aspects of bacterial photosynthesis. This purple phototroph contains PufX, a polypeptide crucial for dimerization of the light-harvesting 1–reaction center (LH1–RC) complex, but lacks protein-U, a U-shaped polypeptide in the LH1–RC of its close relative Rba. sphaeroides . Here we present a cryo-EM structure of the Rba. capsulatus LH1–RC purified by DEAE chromatography. The crescent-shaped LH1–RC exhibits a compact structure containing only 10 LH1 αβ-subunits. Four αβ-subunits corresponding to those adjacent to protein-U in Rba. sphaeroides were absent. PufX in Rba. capsulatus exhibits a unique conformation in its N-terminus that self-associates with amino acids in its own transmembrane domain and interacts with nearby polypeptides, preventing it from interacting with proteins in other complexes and forming dimeric structures. These features are discussed in relation to the minimal requirements for the formation of LH1–RC monomers and dimers, the spectroscopic behavior of both the LH1 and RC, and the bioenergetics of energy transfer from LH1 to the RC.

Abstract:A 4.4-kb HindIII fragment, encoding an unusual rubredoxin (denoted RubA), a homolog of the Synechocystis sp. PCC 6803 gene slr2034 and Arabidopsis thaliana HCF136, and the psbEFLJ operon, was cloned from the cyanobacterium Synechococcus sp. PCC 7002. Inactivation of the slr2034 homolog produced a mutant with no detectable phenotype and wild-type photosystem (PS) II levels. Inactivation of the rubA gene of Synechococcus sp. PCC 7002 produced a mutant unable to grow photoautotrophically. RubA and PS I electron transport activity were completely absent in the mutant, although PS 11 activity was similar to80% of the wild-type level. RubA contains a domain of similar to50 amino acids with very high similarity to the rubredoxins of anaerobic bacteria and archaea, but it also contains a region of about 50 amino acids that is predicted to form a flexible hinge and a transmembrane a-helix at its C terminus. Overproduction of the water-soluble rubredoxin domain in Escherichia coli led to a product with the absorption and EPR spectra of typical rubredoxins. RubA was present in thylakoid but not plasma membranes of cyanobacteria and in chloroplast thylakoids isolated from spinach and Chlamydomonas reinhardtii. Fractionation studies suggest that RubA might transiently associate with PS I monomers, but no evidence for an association with PS I trimers or PS II was observed. PS I levels were significantly lower than in the wild type (similar to40%), but trimeric PS I complexes could be isolated from the rubA mutant. These PS I complexes completely lacked the stromal subunits PsaC, PsaD), and PsaE but contained all membrane-intrinsic subunits. The three missing proteins could be detected immunologically in whole cells, but their levels were greatly reduced, and degradation products were also detected. Our results indicate that RubA plays a specific role in the biogenesis of PS I.
Effects of Chlorophyll Availability on Fluoreseenee Components of Photosystems in the ORF469-Deletion Mutant of Cyanobacterium

Abstract:PCI-amplified ORF 469 fragment from Synechocystis sp. PCC 6803 was cloned into pUC11K and a construct was made in which part of ORF 6803 was deleted and replaced by erythromycin resistance cassette. Transformation of wild type strain of Synechocystis sp. PCC 6803 with this construct yielded a mutant in which ORF 469 was deleted. In the resulting mutant, the light-independent pathway of chlorophyll biosynthesis was inactivated and availability of chlorophyll was fully dependent on light. When propagated the mutant in dark, the chlorophyll was non-detectable and protochlorophyllide with 645 nm fluorescence emission peak was accumulated. Meanwhile, the fluorescence emission peaks (excited at 435 nm) of thylakoids at 685 nm, 695 nm and 725 nm, which represented relative chlorophyll-binding proteins, disappeared. Upon return of dark-grown ORF 469 mutant to the light, greening occurred and chlorophyll was synthesized to assembly fluorescence emission components in photosystems. Newly synthesized chlorophyll combined the fluorescenee component of 685 nm at first, then 725 nm and 695 nm at last, which indicates a pecking order for biogenesis of chlorophyll-binding proteins when availability of chlorophyll is limited. The mutant lacking ORF 469 in Synechocystis sp. PCC 6803 was suggested as an excellent cyanobacterial system for studies on the interactions between chlorophyll and chlorophyll-binding proteins in photosystems.
Proteomic characterization of the Rhodobacter sphaeroides 2.4.1 photosynthetic membrane: identification of new proteins

Abstract:The Rhodobacter sphaeroides intracytoplasmic membrane (ICM) is an inducible membrane that is dedicated to the major events of bacterial photosynthesis, including harvesting light energy, separating primary charges, and transporting electrons. In this study, multichromatographic methods coupled with Fourier transform ion cyclotron resonance mass spectrometry, combined with subcellular fractionation, was used to test the hypothesis that the photosynthetic membrane of R. sphaeroides 2.4.1 contains a significant number of heretofore unidentified proteins in addition to the integral membrane pigment-protein complexes, including light-harvesting complexes 1 and 2, the photochemical reaction center, and the cytochrome bc(1) complex described previously. Purified ICM vesicles are shown to be enriched in several abundant, newly identified membrane proteins, including a protein of unknown function (AffyChip designation RSP1760) and a possible alkane hydroxylase (RSP1467). When the genes encoding these proteins are mutated, specific photosynthetic phenotypes are noted, illustrating the potential new insights into solar energy utilization to be gained by this proteomic blueprint of the ICM. In addition, proteins necessary for other cellular functions, such as ATP synthesis, respiration, solute transport, protein translocation, and other physiological processes, were also identified to be in association with the ICM. This study is the first to provide a more global view of the protein composition of a photosynthetic membrane from any source. This protein blueprint also provides insights into potential mechanisms for the assembly of the pigment-protein complexes of the photosynthetic apparatus, the formation of the lipid bilayer that houses these integral membrane proteins, and the possible functional interactions of ICM proteins with activities that reside in domains outside this specialized bioenergetic membrane.

Abstract:With the method of Polymerase Chain Reaction and homologous genetic recombination in vivo, the key gene encoding bacterial-type phytoene desaturase (Crtl) which controls the carotenoids biosynthesis pathway in the non-photosynthetic and extremely radioresistant bacterium Deinococcus radiodurans was deleted from the genome. The colorless mutant obtained was designated as M61. Survival rates of mutant strain and wild type strain were investigated under different doses of gamma-radiation and hydrogen peroxide. The results showed that the radioresistant activity of M61 reduced rapidly under ionization radiation, and it became more sensitive to the treatment of hydrogen peroxide especially to high concentration of hydrogen peroxide compared to that of wild type R1. Reverse Phase High Performance Liquid Chromatography (RP-HPLC) was used to investigate the carotenoid composition of wild type R1 and mutant M61. HPLC results exhibited that the deficient of crtl gene had important effect on pigment biosynthesis pathway, leading to inhibition of the biosynthesis of lycopene and other carotenoids in D. radiodurans. All the results indicated that crtl gene was a key gene controlling the biosynthesis of red carotenoid including lycopene in D. radiodurans. The roles of carotenoids in protecting the bacterial cell from damage by ionization radiation and hydrogen peroxide suggest that the carotenoids contribute to the defense system in D. radiodurans. This study is important for elucidating the radioresistant and antioxidant mechanism in which carotenoids are involved, and it will supply some ideas to the further investigation on the biosynthesis pathway and functions of carotenoids in D. radiodurans.
Characterization of Psai and Psal Mutants of Synechococcus Sp Strain Pcc 7002: A New Model for State Transitions in Cyanobacteria

Abstract:The psaI and psaL genes were characterized from the cyanobacterium Synechococcus sp. strain PCC 7002. The gene organization was different from that reported for other cyanobacteria with psaI occurring upstream and being divergently transcribed from the psaL gene. Mutants lacking PsaI or PsaL were generated by interposon mutagenesis and characterized physiologically and biochemically. Mutant strains PR6307 (delta psaI), PR6308 (psaI-) and PR6309 (psaL-) had doubling times similar to that of the wild type under both high- and low-intensity white light, but all grew more slowly than the wild type in green light. Only monomeric photosystem I (PS I) complexes could be isolated from each mutant strain when Triton X-100 was used to solubilize thylakoid membranes; however, approximately 10% of the PS I complexes from the psaI mutants, but not the psaL mutant, could be isolated as trimers when n-dodecyl beta-D-maltoside was used. Compositional analyses of the mutant PS I complexes indicate that the presence of PsaL is required for trimer formation or stabilization and that PsaI plays a role in stabilizing the binding of both PsaL and PsaM to the PS I complex. Strain PR6309 (psaL-) was capable of performing a state 2 to state 1 transition approximately three times more rapidly than the wild type. Because the monomeric PS I complexes of this mutant should be capable of diffusing more rapidly than trimeric complexes, these data suggest that PS I complexes rather than phycobilisomes might move during state transitions. A "mobile-PS I" model for state transitions that incorporates these ideas is discussed.

Abstract:As a subfamily member of SF1 superfamily, the RecO helicases are highly conserved in evolution and are required for maintaining genome stability in all organisms. Loss of RecO helicase function leads to a breakdown in the maintenance of genome integrity, in particular hyper-recombination. Named after the recQ gene of Escherichia coli, lower eukaryotic species generally only contain a single RecQ family representative; for example, Sgsl in the budding yeast, Saccharomyces cerevisiae, and Rqhl in the fission yeast, Schizosaccharomyces pombe. There are, however, multiple members in most higher organisms, with five being present in humans. Defects in three of these human RecQ helicases give rise to defined clinical disorders associated with cancer predisposition and variable aspects of premature aging. Deinococcus radiodurans encodes two recQ genes with unusual domain: DR1289 and DR2444, whose functions, however, remain obscure currently. DR1289 contains three tandem copies of the C-terminal helicase-RNase D (HRDC) domain, instead of the single copy present in all other bacteria except Neisseria that similarly possesses three copies. DR2444 contains a HRDC domain and a domain homologous to cystathionine gamma-lyase; this is the first example of an HRDC domain that is not associated with either a helicase or a nuclease. In this study, a fusion DNA fragment carrying kanamycin resistance gene with the D. radiodurans groEL promoter, chloramphenicol resistance gene with KAT promoter was cloned by PCR amplification and reversely inserted into the recQ locus in the genome of the wild-type strain RI. Three resulting recQ-deficient strains, designated deltaDR1289, deltaDR2444 and deltarecQ (double mutation), were constructed. Results show that deltaDR1289 and delta recQ were very sensitive to ionizing radiation and H2O2, while delta DR2444 and wild strain R1 were not. The phenotype of delta DR1289 was similar to many RecQ helicase mutants. Therefore, it was presumed that DR1289 was the necessary gene in maintaining the extreme resistance to DNA damaging agents, whereas DR2444 was not. Further research based on genetic and biochemical approaches should help to gain a better understanding of the genes involved in DNA repair.

Abstract:The rubA gene was insertionally inactivated in Synechococcus sp. PCC 7002, and the properties of photosystem I complexes were characterized spectroscopically. X-band EPR spectroscopy at low temperature shows that the three terminal iron-sulfur clusters, F-X, F-A, and F-B, are missing in whole cells, thylakoids, and photosystein (PS) I complexes of the rubA mutant. The flashinduced decay kinetics of both P700(+) in the visible and A(1)(-) in the near-UV show that charge recombination occurs between P700degrees and A(1)(-) in both thylakoids and PS I complexes. The spin-polarized EPR signal at room temperature from PS I complexes also indicates that forward electron transfer does not occur beyond A, In agreement, the spin-polarized X-band EPR spectrum of P700(+) A(1)(-) at low temperature shows that an electron cycle between A(1)(-) and P700degrees occurs in a much larger fraction of PS I complexes than in the wild-type, wherein a relatively large fraction of the electrons promoted are irreversibly transferred to [F-A/F-B]. The electron spin polarization pattern shows that the orientation of phylloquinone in the PS I complexes is identical to that of the wild type, and out-of-phase, spin-echo modulation spectroscopy shows the same P700degrees to A(1)(-) center-to-center distance in photosystem I complexes of wild type and the rubA mutant. In contrast to the loss of F-X, F-B, and F-A, the Rieske iron-sulfur protein and the non-heme iron in photosystem 11 are intact. It is proposed that rubredoxin is specifically required for the assembly of the Fx iron-sulfur cluster but that Fx is not required for the biosynthesis of trimeric P700-A(1) cores. Since the PsaC protein requires the presence of Fx for binding, the absence of F-A and F-B, may be an indirect result of the absence of F-X.

Abstract:The CO2 fixation ability of Rhodopseudomonas palustris DH was enhanced by introducing the recombinant plasmid pMG-CBBM containing the form II ribulose-1,5-bisphosphate carboxylase/oxygenase (RubisCO) gene (cbbM) isolated from Rps. palustris NO. 7. Sequencing of a 3.0-kb PstI fragment containing the cbbM gene revealed an open reading frame encoding 461 amino acids, homologous to known cbbM genes, with a ribosome binding site upstream of cbbM and a terminator downstream of cbbM, without promoter. pMG-CBBM, a Rhodopseudomonas/Escherichia coli shuttle expression plasmid, was derived from the Rhodopseudomonas/E. coli shuttle cloning vector pMG105, by inserting the promoter of the pckA gene and the cbbM gene into its multiple cloning site. Plasmid pMG-CBBM was transformed into Rps. palustris DH by electroporation, and was stably maintained when transformants were grown either photoheterotrophically or photolithoautotrophically in the absence of antibiotics. This is the first report of an expression plasmid containing a Rps. palustris-specific promoter that allows stable expression of a foreign gene in the absence of antibiotic selection.
Selective expression of light-harvesting complexes alters phospholipid composition in the intracytoplasmic membrane and core complex of purple phototrophic bacteria

Abstract:Phospholipid-protein interactions play important roles in regulating the function and morphology of photosynthetic membranes in purple phototrophic bacteria. Here, we characterize the phospholipid composition of intracytoplasmic membrane (ICM) from Rhodobacter (Rba.) sphaeroides that has been genetically altered to selectively express light-harvesting (LH) complexes. In the mutant strain (DP2) that lacks a peripheral light-harvesting (LH2) complex, the phospholipid composition was significantly different from that of the wild-type strain; strain DP2 showed a marked decrease in phosphatidylglycerol (PG) and large increases in cardiolipin (CL) and phosphatidylcholine (PC) indicating preferential interactions between the complexes and specific phospholipids. Substitution of the core light-harvesting (LH1) complex of Rba. sphaeroides strain DP2 with that from the purple sulfur bacterium Thermochromatium tepidum further altered the phospholipid composition, with substantial increases in PG and PE and decreases in CL and PC, indicating that the phospholipids incorporated into the ICM depend on the nature of the LH1 complex expressed. Purified LH1-reaction center core complexes (LH1-RC) from the selectively expressing strains also contained different phospholipid compositions than did core complexes from their corresponding wild-type strains, suggesting different patterns of phospholipid association between the selectively expressed LH1-RC complexes and those purified from native strains. Effects of carotenoids on the phospholipid composition were also investigated using carotenoid-suppressed cells and carotenoid-deficient species. The findings are discussed in relation to ICM morphology and specific LH complex-phospholipid interactions.
