Abstract:Photosynthetic organisms rely on a network of light-harvesting protein-pigment complexes to efficiently absorb sunlight and transfer excitation energy to reaction center proteins for charge separation. In photosynthetic purple bacteria, these complexes are embedded in the cell membrane, where lipid composition affects their clustering and inter-complex energy transfer. However, the lipid bilayer's impact on intra-complex excitation dynamics is less understood. Recent experiments compared photo-excitation dynamics in detergent-isolated light harvesting complex 2 (LH2) to LH2 embedded in membrane discs mimicking the biological environment, revealing differences in spectra and intra-complex energy transfer rates. We use available quantum chemical and spectroscopy data to develop a complementary theoretical study on the excitonic structure and intra-complex energy transfer kinetics of the LH2 from photosynthetic purple bacteria Rhodoblastus acidophilus in two conditions: LH2 in a membrane environment and detergent-isolated LH2. Dark excitonic states crucial for B800-B850 energy transfer within LH2 are found to be more delocalised in the membrane model. Using non-perturbative and generalised Förster calculations, it is shown that the increased quantum delocalisation leads to a B800 to B850 transfer rate 30% faster than in the detergent-isolated complex, consistent with experimental results. We identify the main energy transfer pathways in each environment and show how differences in the B800 to B850 transfer rate stem from changes in LH2's electronic properties when embedded in the membrane. By considering quasi-static variations of electronic excitation energies in LH2, we show that the broadening of the B800 to B850 transfer rate distribution is affected by lipid composition. We argue that the variation in broadening could indicate a speed-accuracy trade-off, common in biological systems.

Abstract:The phycobilisomes function as the primary light-harvesting antennae in cyanobacteria and red algae, effectively harvesting and transferring excitation energy to both photosystems. Here we investigate the direct energy transfer route from the phycobilisomes to photosystem I at room temperature in a mutant of the cyanobacterium Synechocystis sp. PCC 6803 that lacks photosystem II. The excitation dynamics are studied by picosecond time-resolved fluorescence measurements in combination with global and target analysis. Global analysis revealed several fast equilibration time scales and a decay of the equilibrated system with a time constant of ≈220 ps. From simultaneous target analysis of measurements with two different excitations of 400 nm (chlorophyll a) and 580 nm (phycobilisomes) a transfer rate of 42 ns -1 from the terminal emitter of the phycobilisome to photosystem I was estimated.

Spatial Patterns of Light-Harvesting Antenna Complex Arrangements Tune the Transfer-to-Trap Efficiency of Excitons in Purple Bacteria

Abstract:In photosynthesis, the efficiency with which a photogenerated exciton reaches the reaction center is dictated by chromophore energies and the arrangement of chromophores in the supercomplex. Here, we explore the interplay between the arrangement of light-harvesting antennae and the efficiency of exciton transport in purple bacterial photosynthesis. Using a Miller-Abrahams-based exciton hopping model, we compare different arrangements of light-harvesting proteins on the intracytoplasmic membrane. We find that arrangements with aggregated LH1s have a higher efficiency than arrangements with randomly distributed LH1s in a wide range of physiological light fluences. This effect is robust to the introduction of defects on the intracytoplasmic membrane. Our result explains the absence of species with aggregated LH1 arrangements in low-light niches and the large increase seen in the expression of LH1 dimer complexes in high fluences. We suggest that the effect seen in our study is an adaptive strategy toward solar light fluence across different purple bacterial species.
Comparative Study of Spectral and Functional Properties of Wild Type and Double Mutant H(L173)L/I(L177)H Reaction Centers of the Purple Bacterium Cereibacter sphaeroides

Abstract:Previously, we found that in the reaction center (RC) of the purple bacterium Cereibacter sphaeroides , formation of heterodimeric primary electron donor (P) caused by the substitution of His-L173 by Leu, was compensated by the second mutation Ile-L177 – His. Significant changes in the spectral properties, pigment composition, and redox potential of P observed in the H(L173)L RC, are restored to the corresponding characteristics of the native RC in the RC H(L173)L/I(L177)H, with the difference that the energy of the long-wavelength Q Y optical transition of P increases significantly (by ~75 meV). In this work, it was shown using light-induced difference FTIR spectroscopy that the homodimeric structure of P is preserved in the RC with double mutation with partially altered electronic properties: electronic coupling in the radical-cation of the P + dimer is weakened and localization of the positive charge on one of its halves is increased. Results of the study of the triple mutant RC, H(L173)L/I(L177)H/F(M197)H, are consistent with the assumption that the observed changes in the P + electronic structure, as well as considerable blue shift of the Q Y P absorption band in the RC H(L173)L/I(L177)H, are associated with modification of the spatial position and/or geometry of P. Using femtosecond transient absorption spectroscopy, it was shown that the mutant H(L173)L/I(L177)H RC retains a sequence of reactions P* → P + B A − → P + H A − → P + Q A − with electron transfer rates and the quantum yield of the final state P + Q A − close to those observed in the wild-type RC (P* is the singlet-excited state of P; B A , H A , and Q A are molecules of bacteriochlorophyll, bacteriopheophytin, and ubiquinone in the active A-branch of cofactors, respectively). The obtained results, together with the previously published data for the RC with symmetrical double mutation H(M202)L/I(M206)H, demonstrate that by introducing additional point amino acid substitutions, photochemical activity of the isolated RC from C. sphaeroides could be maintained at a high level even in the absence of important structural elements – axial histidine ligands of the primary electron donor P.

Abstract:The study of how photosynthetic organisms convert light offers insight not only into nature's evolutionary process, but may also give clues as to how best to design and manipulate artificial photosynthetic systems -- and also how far we can drive natural photosynthetic systems beyond normal operating conditions, so that they can harvest energy for us under otherwise extreme conditions. In addition to its interest from a basic scientific perspective, therefore, the goal to develop a deep quantitative understanding of photosynthesis offers the potential payoff of enhancing our current arsenal of alternative energy sources for the future. In the following Chapter, we consider the trade-off between dynamics, structure and function of light harvesting membranes in Rps. Photometricum purple bacteria, as a model to highlight the priorities that arise when photosynthetic organisms adapt to deal with the ever-changing natural environment conditions.

Abstract:The ring-like peripheral light-harvesting complex 2 (LH2) expressed by many phototrophic purple bacteria is a popular model system in biological light-harvesting research due to its robustness, small size, and known crystal structure. Furthermore, the availability of structural variants with distinct electronic structures and optical properties has made this group of light harvesters an attractive testing ground for studies of structure–function relationships in biological systems. LH2 is one of several pigment-protein complexes for which a link between functionality and effects such as excitonic coherence and vibronic coupling has been proposed. While a direct connection has not yet been demonstrated, many such interactions are highly sensitive to resonance conditions, and a dependence of intra-complex dynamics on detailed electronic structure might be expected. To gauge the sensitivity of energy-level structure and relaxation dynamics to naturally occurring structural changes, we compare the photo-induced dynamics in two structurally distinct LH2 variants. Using polarization-controlled 2D electronic spectroscopy at cryogenic temperatures, we directly access information on dynamic and static disorder in the complexes. The simultaneous optimal spectral and temporal resolution of these experiments further allows us to characterize the ultrafast energy relaxation, including exciton transport within the complexes. Despite the variations in PPC molecular structure manifesting as clear differences in electronic structure and disorder, the energy-transport and—relaxation dynamics remain remarkably similar. This indicates that the light-harvesting functionality of purple bacteria within a single LH2 complex is highly robust to structural perturbations and likely does not rely on finely tuned electronic- or electron-vibrational resonance conditions.

Abstract:Femtosecond transient absorption spectroscopy has been used to investigate the photoinduced energy and electron transfer processes in photosystem I (PS I) particles from cyanobacteria, green algae, and higher plants. At room temperature, the kinetics observed in all three species are very similar: Following 590 nm excitation, an equilibration process(es) with a 3.7-7.5 ps lifetime was observed, followed by a 19-24 ps process that is associated with trapping. In all three species long-wavelength pigments (pigments that absorb at longer wavelengths than the primary electron donor) were observed. The difference spectrum associated with reduction of the primary electron acceptor [Ao(-)-Ao) difference spectrum] was obtained for all three species. The (Ao(-)-Ao) difference spectra obtained from measurements using detergent-isolated PS I particles from spinach and Chlamydomonas reinhardtii are similar but clearly membrane fragments. In all three species the reduced primary electron acceptor (Ao(-)) is reoxidized extremely rapidly, in about 20 ps. The difference spectrum associated with Ao reduction appears to contain contributions from more than a single chlorophyll pigment.
Contour tracking of an unknown planar object by regulating force for mobile robot navigation

Abstract:SUMMARY In this paper, a novel contour-tracking control method of an unknown planar object by lateral force regulation for wheeled mobile robot navigation is presented. The robot is required to follow the contour of an unknown object toward the goal position. Based on mobile robot dynamic equations, a force-control algorithm is proposed to maintain constant contact with a planar object. Measured contact force from an object is used not only to regulate a contact force in a lateral direction, but also to control the orientation angle of the robot to avoid collision with an object. Simulation and experiment of contour-tracking tasks of a wheeled mobile robot are conducted. Experimental results show that the contact force is well-regulated, and the robot arrives at the goal position successfully.

Abstract:We perform a quantum mechanical study of the peptides that are part of the LH2 complex from Rhodopseudomonas acidophila, a non-sulfur purple bacteria that has the ability of producing chemical energy from photosynthesis. The electronic structure calculations indicate that the transmembrane helices of these peptides are characterized by dipole moments with a magnitude of ~150 D. When the full nonamer assembly made of eighteen peptides is considered, then a macrodipole of magnitude 704 D is built up from the vector sum of each monomer dipole. The macrodipole is oriented normal to the membrane plane and with the positive tip toward the cytoplasm thereby indicating that the electronic charge of the protein scaffold is polarized toward the periplasm. The results obtained here suggest that the asymmetric charge distribution of the protein scaffold contributes an anisotropic electrostatic environment which differentiates the absorption properties of the bacteriochlorophyll pigments, B800 and B850, embedded in the LH2 complex.

Abstract:Phycobilisomes (PBs) play an important role in cyanobacterial photosynthesis. They capture light and transfer excitation energy to the photosynthetic reaction centres. PBs are also central to some photoprotective and photoregulatory mechanisms that help sustain photosynthesis under non-optimal conditions. Amongst the mechanisms involved in excitation energy dissipation that are activated in response to excessive illumination is a recently discovered light-induced mechanism that is intrinsic to PBs and has been the least studied. Here, we used single-molecule spectroscopy and developed robust data analysis methods to explore the role of a terminal emitter subunit, ApcE, in this intrinsic, light-induced mechanism. We isolated the PBs from WT Synechocystis PCC 6803 as well as from the ApcE-C190S mutant of this strain and compared the dynamics of their fluorescence emission. PBs isolated from the mutant (i.e., ApcE-C190S-PBs), despite not binding some of the red-shifted pigments in the complex, showed similar global emission dynamics to WT-PBs. However, a detailed analysis of dynamics in the core revealed that the ApcE-C190S-PBs are less likely than WT-PBs to enter quenched states under illumination but still fully capable of doing so. This result points to an important but not exclusive role of the ApcE pigments in the light-induced intrinsic excitation energy dissipation mechanism in PBs.

Proteomic analysis of the developing intracytoplasmic membrane in Rhodobacter sphaeroides during adaptation to low light intensity

Abstract:Although the primary photochemical events in the facultative photoheterotrophic purple bacterium Rhodobacter sphaeroides are now well understood, comparatively little is known about how their photosynthetic apparatus is assembled. Here we present a proteomic analysis of the intracytoplasmic membrane (ICM) assembly process during adaptation to lowered light intensity, in which the size of the photosynthetic units is greatly expanded by addition of the light-harvesting 2 (LH2) peripheral antenna complex. When the isolated ICM-derived chromatophore vesicles were subjected to clear native gel electrophoresis (CNE), four pigmented bands appeared; the top and bottom bands contained the reaction center - light-harvesting 1 (RC-LH1) core complex and LH2 peripheral antenna, respectively, while the two bands of intermediate migration contained associations of the LH2 and core complexes. Proteomic analysis revealed a large array of other proteins associated with the CNE gel bands - in particular, several F(1)F(O)-ATP synthase subunits gave unexpectedly high spectral counts, given the inability to detect this coupling factor, as well as the more abundant cytochrome bc (1) complex, by atomic force microscopy. Significant levels of general membrane assembly factors were also found, as well as numerous proteins of unknown function including high counts for RSP6124 that were correlated with LH2 levels. When combined with further AFM and spectroscopic studies, these proteomic approaches are expected to provide a much-improved understanding of the overall assembly process.
Catecholamines in plasma and urine in patients with essential hypertension determined by double-isotope derivative techniques.

Abstract:Employing double-isotope derivative techniques, noradrenaline and adrenaline have been determined in plasma and in urine and dopamine in urine in 21 patients with essential hypertension as well as in 32 controls. Plasma noradrenaline rose with age in both groups of subjects. No differences were observed in plasma noradrenaline and plasma adrenaline in the resting supine position and in urinary excretion of noradrenaline and dopamine in hypertensive patients as compared to control subjects. Urinary excretion of adrenaline was somewhat lower in the hypertensives than in the controls. Treatment with alprenolol, a beta-adrenergic blocking agent, did not influence noradrenaline and adrenaline in plasma in the basal state or the urinary excretion of the three catecholamines. The combined treatment with alprenolol and hydralazine was followed by a significant rise in plasma noradrenaline. It is concluded that the adrenergic activity evaluated by circulating catecholamines is normal in most patients with essential hypertension.

Abstract:The light-harvesting (LH) and reaction center (RC) core complex of purple bacterium Roseiflexus castenholzii, B880-B800-RC, are different from those of the typical photosynthetic unit, (B850-B800)(x)-B880-RC. To investigate the excitation flowing dynamics in this unique complex, two-dimensional electronic spectroscopy is employed. The obtained time constants for the exciton relaxation in B880, exciton relaxation in B800, B800 -> B880 energy transfer (EET), and B880 -> closed RC EET are 43 fs, 177 fs, 1.9 ps, and 205 ps, respectively. These time constants result in an overall EET efficiency similar to that of the typical photosynthetic unit. Analysis of the oscillatory signals reveals that while several vibronic coherences are involved in the exciton relaxation process, only one prominent vibronic coherence, with a frequency of 27 cm(-1) and coupled to the B880 electronic transition, may contribute to the B800 -> B880 EET process.
Selective expression of light-harvesting complexes alters phospholipid composition in the intracytoplasmic membrane and core complex of purple phototrophic bacteria

Abstract:Phospholipid-protein interactions play important roles in regulating the function and morphology of photosynthetic membranes in purple phototrophic bacteria. Here, we characterize the phospholipid composition of intracytoplasmic membrane (ICM) from Rhodobacter (Rba.) sphaeroides that has been genetically altered to selectively express light-harvesting (LH) complexes. In the mutant strain (DP2) that lacks a peripheral light-harvesting (LH2) complex, the phospholipid composition was significantly different from that of the wild-type strain; strain DP2 showed a marked decrease in phosphatidylglycerol (PG) and large increases in cardiolipin (CL) and phosphatidylcholine (PC) indicating preferential interactions between the complexes and specific phospholipids. Substitution of the core light-harvesting (LH1) complex of Rba. sphaeroides strain DP2 with that from the purple sulfur bacterium Thermochromatium tepidum further altered the phospholipid composition, with substantial increases in PG and PE and decreases in CL and PC, indicating that the phospholipids incorporated into the ICM depend on the nature of the LH1 complex expressed. Purified LH1-reaction center core complexes (LH1-RC) from the selectively expressing strains also contained different phospholipid compositions than did core complexes from their corresponding wild-type strains, suggesting different patterns of phospholipid association between the selectively expressed LH1-RC complexes and those purified from native strains. Effects of carotenoids on the phospholipid composition were also investigated using carotenoid-suppressed cells and carotenoid-deficient species. The findings are discussed in relation to ICM morphology and specific LH complex-phospholipid interactions.

Abstract:Ultra time-resolved fluorescence spectra were used to study the energy transfer processes and mechanism of complex PEC/PC/APC at 77 K, which was reconstructed with phycobiliproteins (PEC, PC and APC) of Anabaena variabilis , and has intact light-harvesting system and single terminal emitter. The energy transfer relationships between different chromophores especially between rod and core were also discussed based on fluorescence decay kinetic under different detected wavelengths. As a result, we got the possible energy transfer pathways and transfer time constants to be 29 ps between two PEC trimers, 12 ps between PEC and C-PC, 51 ps between rod and core.
Interplay between excitation kinetics and reaction-center dynamics in purple bacteria

Abstract:Photosynthesis is arguably the fundamental process of life, since it enables energy from the Sun to enter the food chain on the Earth. It is a remarkable non-equilibrium process in which photons are converted to many-body excitations, which traverse a complex biomolecular membrane, where they are captured and fuel chemical reactions within a reaction center (RC) in order to produce nutrients. The precise nature of these dynamical processes—which lie at the interface between quantum and classical behavior and involve both noise and coordination—is still being explored. Here, we focus on a striking recent empirical finding concerning an illumination-driven transition in the biomolecular membrane architecture of the purple bacteria Rsp. photometricum. Using stochastic realizations to describe a hopping rate model for excitation transfer, we show numerically and analytically that this surprising shift in preferred architectures can be traced to the interplay between the excitation kinetics and the RC dynamics. The net effect is that the bacteria profit from efficient metabolism at low illumination intensities while using dissipation to avoid an oversupply of energy at high illumination intensities.

Abstract:Purple photosynthetic bacterium Thermochromatium (Tch.) tepidum is a moderate thermophile growing in an optimal temperature range of 48-50 ℃.Its light-harvesting complex 2 (LH2) possesses heterogeneous compositions of apoprotein and carotenoid (Car), but the high-resolution crystallographic structure remains unknown.We have attempted an ultrafast time-resolved spectroscopic study of the isolated LH2 complex from Tch.tepidum.The spectral dynamics and population kinetics of n-dodecyl-β-D-moltoside (DDM) and lauryldimethylamine oxide (LDAO) preparations of LH2 reveal efficient S2-state mediated Car-to-Car and Car-to-bacteriochlorophyll (BChl) singlet excitation energy transfer (EET) occurring in a time scale of ～100 fs, as well as the Qy-state mediated B800-to-B850 singlet EET for the DDM preparation proceeding with a time constant of ～1.2 ps.These ultrafast EET processes suggest that the Cars with 11 and 12 conjugated C=C double bonds (NC=C) coexist in the LH2 complex, and that the B800-B850 mutual orientation in LH2 differs considerably from those in the LH2s from some of the previously investigated bacterial species.In addition, anhydrorhodovibrin (NC=C=12) as a minor Car composition is found to act as an efficient trap of excitation energy, which is considered to be an important photoprotection mechanism.Furthermore, based on the results of ultrafast Car band shift in response to BChl excitation, we propose that, compared to other Car compositions, (OH-)spirilloxanthin (NC=C=13) locates in closer proximity to BChl.Our results may facilitate to understand the light-harvesting and the photoprotection mechanisms of Tch.tepidum living under harsh natural conditions.
Photosynthetic energy transfer: missing in action (detected spectroscopy)?

Abstract:In recent years, action-detected ultrafast spectroscopies have gained popularity. These approaches offer some advantages over their coherently-detected counterparts, enabling spatially-resolved and operando measurements with high sensitivity. However, there are also fundamental limitations connected to the different process of signal generation in action-detected experiments. Specifically, state mixing by nonlinear interactions during signal emission leads to a large static background which can obscure the excited-state dynamics. This could severely limit the applicability of action-detected spectroscopy to study energy transfer in larger systems. Here we perform fluorescence-detected two-dimensional electronic spectroscopy (F-2DES) of the light-harvesting II (LH2) complex from purple bacteria. We demonstrate that the B800-B850 energy transfer process in LH2 is barely discernable in F-2DES, representing a ~6.2% rise of the lower cross-peak intensity. This is in stark contrast to measurements using coherently-detected 2DES where the lower cross-peak reveals energy transfer with 100% contrast. We explain the weak presence of excited-state dynamics using a disordered excitonic model with realistic experimental conditions. We further derive a general formula for the presence of excited-state signals in multi-chromophoric aggregates, dependent on the aggregate geometry and size, and the interplay of excitonic coupling and disorder. We find that, dependent on the excitonic state structure, the excited state dynamics in F-2DES can be visible even in large aggregates. Our work shows that the signatures of energy transfer in F-2DES can be used to directly infer the excitonic structure in multichromophoric systems.
