Abstract:An improved experiment setup for Stimulated Photoacoustic Raman Spectroscopy (PARS) was reported. The employed two laser beams, unlike general arrangement of propagating in the same direction, counterpropagated with each other. This arrangement can eliminate the limitation that the small Raman shift could not be measured with general arrangement. Therefore, this improved setup was especially useful for measuring the PARS spectrum with small Raman shift, such as the pure rotational Raman spectrum. In the experiment, taking CH4 molecules as a research object, one laser was fixed at 532.1 nm while another tunable dye laser scanned. The PARS spectrum of CH4 molecules for the nu(1), nu(2) and nu(3) bands was recorded in the two wavelength ranges of 625 similar to 642 nm and 573 similar to 589 nm, and the results agreed well with those obtained by Spontaneous Raman Scattering (ORS) technique, confirming the validity of the improved experimental setup.

Abstract:To overcome the shortcoming of traditional Raman spectral decomposition method that requires a priori information of pure spectra, a new Raman spectral decomposition algorithm based on multivariate curve resolution and Lorentz function constraint was proposed. This method was based on the bilinear model for mixture spectra, and only the mixture spectral matrix was needed to perform the analysis. It first decomposes the mixture spectra with principal component resolution, and then the evolving factor analysis was performed to find the distribution of the mixture spectral eigenvalues evolves along with the pixel direction, so that it could determine the number of components in the mixture, the range of their peaks and the initial estimation 14 their spectra. Finally an alternative least squares method with Lorentz function constraint was proposed to optimize the pure spectra, during the process, it first fits the pure spectra with Lorentz function, and then based on the fitted spectra, it regresses the spectra and the coefficient matrix with non-negative alternative least squares method. The above optimization process was repeatedly performed until the fit error for the mixture spectra was within the threshold. In the experimental part, the proposed algorithm was adopted to decompose the Raman spectra of the circulating fluid in an adsorption tower of a p-xylene unit, with which the relative pure spectra was obtained in the mixture fluid. Finally, with the repeated quantitative analysis experiments for its main component of p-xylene, we prove the effectiveness for the proposed algorithm.
A Method for the Quantitative Analysis of a Key Component in Complex Mixtures Using Raman Spectroscopy Based on Peak Decomposition

Abstract:Raman spectroscopy has been increasingly used in industrial production processes for analytical purposes. However, the quantitative analysis of complex mixtures based on their Raman spectra remains a problem, especially when not all of the spectra of pure components in the mixture are available. In this work, a method for the quantitative analysis of a key component in complex mixtures based on peak decomposition is introduced. The proposed method only requires the spectrum of the main component in the mixture before analysis. The spectra of the mixture are decomposed to the contributions of the main component and a group of parametric Lorentzian functions. Then, the representative peaks of the key component are extracted. Finally, a calibration model is established. This method is applied to a complex mixture containing m -xylene, o -xylene, p -xylene, ethylbenzene, methylbenzene, three trimethylbenzenes, non-aromatic hydrocarbons, etc. to determine the concentration of o -xylene in the mixture. Experiments demonstrate that the proposed method performs better than the commonly-used partial least squares (PLS), especially when the number of training samples is very small.
Precise measurement of the depolarization ratio from photoacoustic Raman spectroscopy

Abstract:Abstract A new method for the accurate determination of the Raman depolarization ratio is reported with an improved setup for photoacoustic Raman spectroscopy (PARS). The precise measurement is achieved by measuring the dependence of the acoustic signal intensity on the cross‐angle between the polarizations of two incident laser beams. We demonstrate this sensitive and simple method with several gaseous molecules, such as CH 4 and H 2 . The measured results of depolarization ratios agree well with the theoretical values with an upper error limit of ± 0.005, which is comparable to that with polarization‐resolved CARS spectroscopy. Abstract:In this Letter, photoacoustic-guided Raman spectroscopy (PARS) is proposed for a fast depth-resolved Raman measurement with accurate depth localization. The approach was experimentally demonstrated to receive both photoacoustic and Raman signals from a three-layer agar phantom based on a developed synergic photoacoustic-Raman probe, showing strong depth correlation and achieving magnitude of faster operation speed due to photoacoustic time-of-flight measurement and guidance, compared with the conventional depth-resolved Raman spectroscopy method. In addition, further combination with advanced optical-focusing techniques in biological-scattering medium could potentially enable the proposed approach for cancer diagnostics with both tight and fast optical focusing at the desired depth of tumor.

Abstract:Two kinds of spectrum, the Raman-Induced Kerr Effect Spectrum(RIKES)and the Photoacoustic Raman Spectrum(PARS),were measured simultaneously for the ν 1 vibrational band of methane. The difference between the profiles of the two spectra obtained with linearly polarized pump and Stokes laser beams lies in that, the intensity of RIKES spectrum is stronger at the small Raman shift side, and is weaker at the large Raman shift side compared to the PARS spectrum. It was confirmed that this difference is caused by the interference between the resonant and non-resonant parts of the third order non-linear susceptibility.

Abstract:Photoacoustic (PA) measurements encode the information associated with both physical microstructures and chemical contents in biological tissues. A two-dimensional physio-chemical spectrogram (PCS) can be formulated by combining the power spectra of PA signals acquired at a series of optical wavelengths. The analysis of PCS, or namely PA physio-chemical analysis (PAPCA), enables the quantification of the relative concentrations and the spatial distributions of a variety of chemical components in the tissue. This study validated the feasibility of PAPCA in characterizing liver conditions during the progression of non-alcoholic fatty liver disease. A catheter based setup facilitating measurement in deep tissues was also tested.

Abstract:Recently, photoacoustic (PA) spectroscopy has emerged as a valuable tool for the study of various kinds of materials. Herein, we present the results of PA spectral studies of chemical materials. First, the PA study on luminescent materials in condensed states is reported. Combining with the luminescence technique, the energy transfer efficiency and the intrinsic luminescence quantum yield are determined for a europium (III) complex in the glassy state, smectic A phase, and the isotropic liquid. Second, neodymium (III) compounds with l -glycine, l -phenylalanine, and l -tryptophan are synthesized and their PA spectra are reported. The nephelauxetic ratio and Sinha parameter are calculated based on the PA spectra. The environmental effect on the f–f transitions of the neodymium(III) ion is also studied.

Abstract:Photoacoustic imaging is an emerging technique which inherits the merits of optical imaging and ultrasonic imaging. However, classical photoacoustic imaging mainly makes use of the time-domain parameters of signals. In contrast to previous studies, we theoretically investigate the spectral characteristics of the photoacoustic signal from stochastic distributed particles. The spectral slope is extracted and used for describing the spectral characteristics of the photoacoustic signal. Both Gaussian and spherical distributions of optical absorption in particles are considered. For both situations, the spectral slope is monotonically decreased with the increase of particle size. In addition, the quantitative relationship between the spectral slope and the imaging system factors, including the laser pulse envelope, directivity of ultrasound transducer, and signal bandwidth, are theoretically analyzed. Finally, an idealized phantom experiment is performed to validate the analyses and examine the instrument independent of the spectral slope. This work provides a theoretical framework and new experimental evidence for spectrum analysis of the photoacoustic signal. This could be helpful for quantitative tissue evaluation and imaging based on the spectral parameters of the photoacoustic signal.

Abstract:The development and applications of photoacoustic spectroscopy gas sensors is present. The theory of the photoacoustic gas sensors and the advantages of different photoacoustic sensors are discussed. This work focuses on the one-dimension PA cell and has given the model simulation of it. Based on recent instances, the important and difficult points in the research of photoacoustic sensors are given out and some possible methods to improve them are discussed. And the conclusion is that the MEMS photoacoustic sensors have many advantages and will be the future studying trending.
Quantification of Optical Absorption Coefficients from Acoustic Spectra with Photoacoustic Tomography

Abstract:Optical absorption is closely associated with many physiologically important parameters, such as the concentration and oxygen saturation of hemoglobin, and it can be used to quantify the concentrations of non-fluorescent molecules. We introduce a method to quantify the absolute optical absorption based upon the acoustic spectra of photoacoustic (PA) signals. This method is self-calibrating and thus insensitive to variations in optical fluence. Factors such as the detection system bandwidth and acoustic attenuation can affect the quantification but can be canceled by measuring the acoustic spectra at two optical wavelengths. This method has been implemented on various PA systems, including optical-resolution PA microscopy, acoustic-resolution PA microscopy, and reconstruction based PA array systems. We quantified the optical absorption coefficients of phantom samples at various wavelengths. We also quantified the oxygen saturation of hemoglobin in live mice.

Abstract:Driven by applications in chemical sensing, biological imaging and material characterisation, Raman spectroscopies are attracting growing interest from a variety of scientific disciplines. The Raman effect originates from the inelastic scattering of light, and it can directly probe vibration/rotational-vibration states in molecules and materials. Despite numerous advantages over infrared spectroscopy, spontaneous Raman scattering is very weak, and consequently, a variety of enhanced Raman spectroscopic techniques have emerged. These techniques include stimulated Raman scattering and coherent anti-Stokes Raman scattering, as well as surface- and tip-enhanced Raman scattering spectroscopies. The present review provides the reader with an understanding of the fundamental physics that govern the Raman effect and its advantages, limitations and applications. The review also highlights the key experimental considerations for implementing the main experimental Raman spectroscopic techniques. The relevant data analysis methods and some of the most recent advances related to the Raman effect are finally presented. This review constitutes a practical introduction to the science of Raman spectroscopy; it also highlights recent and promising directions of future research developments.

Abstract:Many different techniques, such as UV/vis absorption, IR spectroscopy, fluorescence and Raman spectroscopy are routinely applied in chemical (micro-)analysis and chemical imaging, and a large variety of instruments is commercially available. Up to now, opto- or photoacoustic (PA) and other optothermal (OT) methods are less common and only a limited number of instruments reached a level of application beyond prototypes in research laboratories. The underlying principle of all these techniques is the detection of local heating due to the conversion of light into heat by optical absorption. Considering the versatility, robustness and instrumental simplicity of many PA techniques, it is surprising that the number of commercial instruments based on such approaches is so sparse. The impetus of this review is to summarize basic principles and possible applications described in the literature, in order to foster routine application of these techniques in industry, process analysis and environmental screening. While the terms OT and PA methods cover a very wide range of methods and physical phenomena, this review will concentrate on techniques with applications for analytical measurements.

Photon Absorption Remote Sensing (PARS): A Comprehensive Approach to Label-free Absorption Microscopy Across Biological Scales

Abstract:Label-free optical absorption microscopy techniques have evolved as effective tools for non-invasive chemical specific structural, and functional imaging. Yet most modern label-free microscopy modalities target only a fraction of the contrast afforded by an optical absorption interaction. We introduce a comprehensive optical absorption microscopy technique, Photon Absorption Remote Sensing (PARS), which simultaneously captures the dominant light matter interactions which occur as a pulse of light is absorbed by a molecule. In PARS, the optical scattering, attenuation, and the transient radiative and non-radiative relaxation processes are collected at each optical absorption event. This provides a complete representation of the absorption event, providing unique contrast presented here as the total absorption (TA) and quantum efficiency ratio (QER) measurements. By capturing a complete view of each absorption interaction, PARS bridges many of the specificity challenges associated with label-free imaging, facilitating recovery of a wider range of biomolecules than independent radiative or non-radiative modalities. To show the versatility of PARS, we explore imaging across a wide range of biological specimens, from single cells to in-vivo imaging of living subjects. These examples of label-free histopathological imaging, and vascular imaging illustrate some of the numerous fields where PARS may have profound impacts. Overall PARS may provide comprehensive label-free contrast in a wide variety of biological specimens, providing otherwise inaccessible visualizations, and representing a new a source of rich data to develop new AI and machine learning methods for diagnostics and visualization.

Abstract:Photoacoustic spectrum analysis (PASA) has been found to have the ability to characterize microstructures in phantoms. The ability of PASA technique which is subjected to the ultrasound detector had been reported in identify tissue with hundreds of microns in size. This paper demonstrated the feasibility of micro-ring, an ultrasonic detector with ultra-broad bandwidth, in characterizing microspheres' sizes ranging from few microns to 100 microns using PASA technique and the results were compared with a commercial hydrophone. In order to further verify the capability of micro-ring, spectrum of single micro-spheres with sizes of tens of microns were measured and compared to simulation result. Our work proves that micro-ring based PASA technique has the ability of differentiating the particles with different size in phantoms.

Abstract:The Rosencwaig-Gersho theory of the photoacoustic effect has been extended to the case of three-layer solids. The general expression for the complex amplitude of the periodic temperature at the solid-gas boundary was obtained. The phase and relative magnitude of the photoacoustic signal varying with chopping frequency for varies optical absorption coefficients, thermal conductivity coefficients and material depths were numerically calculated and compared with the experimental results.

Abstract:We demonstrate a photoacoustic method for measurement of sound speed in solid medium.The pulsed photoacoustic signals generated on the surface of a solid medium are muti-reflected by the opposite interfaces of the medium,and it results in a photoacoustic pulses sequence.We calculate the transmitting time of sound between the opposite interfaces of the medium from the measured muti-reflected photoacoustic signals.The sound speed is computed with the measured transmitting time and the width of the object.This method is experimentally verified on a brass cylinder and an aluminum plate.This measurement method is suitable for experimental teaching for undergraduates in physics.

Abstract:Optical-resolution photoacoustic microscopy (OR-PAM) is one of the major implementations of photoacoustic (PA) imaging. With tightly focused optical illumination and high-frequency ultrasound detection, OR-PAM provides micrometer-level resolutions as well as high sensitivity to optical absorption contrast. Traditionally, it is assumed that the detected PA signal in OR-PAM has a linear dependence on the target's optical absorption coefficient, which is the basis for quantitative functional and molecular PA imaging. In this paper, we demonstrate that, due to the limited detection bandwidth and detection view, OR-PAM can have a strong nonlinear dependence on the optical absorption, especially for weak optical absorption (<10 cm<sup>-1</sup>). We have investigated the nonlinear dependence in OR-PAM using numerical simulations, analyzed the underlining mechanisms, proposed potential solutions, and experimentally confirmed the results on phantoms. This work may correct a traditional misunderstanding of the OR-PAM signals and improve quantitative accuracy for functional and molecular applications.
