Abstract:Macrocyclic hosts with a well-defined cavity are particularly appealing for supramolecular chemistry, but they are still rare. In this research, we reported a new class of macrocyclic hosts, namely methylene-bridged naphthotubes, with well-defined cavities. They were synthesized through TFA-catalyzed Friedel-Crafts reactions between alkoxy-substituted bisnaphthalenes and paraformaldehyde. A configurational selection was observed. Three-membered macrocycles possess a single cavity, while four-membered macrocycles adopt a double-cavity conformation or a self-filling conformation depending on the alkoxy length. The small homologue shows quite strong binding affinities (up to 108 M-1 ) to organic cations, which is better than structurally similar but flexible macrocycles. This demonstrates the advantage of a well-defined cavity in molecular recognition. Moreover, these naphthotubes can be used as chirality sensors for chiral organic cations through different chirality transfer mechanisms.

Abstract:Macrocyclic compounds play an important role in supramolecular chemistry accounting for their unique recognition and self-assembly properties and potential applications in biomedicine and material science. Azo compounds display promising capability in fields of molecular switches, polymers, smart materials and molecular machines because of their photoactive and electroactive properties. Introducing azo groups into macrocycles gives them signal moieties, additional recognition sites, photo-responsive properties, and so on. Herein, we comprehensively review the structures of azo-containing macrocyclic compounds reported up to now. Then we describe representative works on azo macrocycles with their molecular recognition, self-assembly and application emphasized.

Abstract:pi-Conjugated macrocycles have been highly attractive due to their challenging synthesis, fascinating aesthetic structure and unique physical and chemical properties. Although some progress has been made in synthesis, the study of pi-macrocycles with different structural characteristics and supramolecular interactions still faces major challenges. In this paper, two new single-bond linked macrocycles (MS-4T/MS-6T) were reported, and the corresponding vinyl-bridged heterocycloarenes (MF-4T/MF-6T) were synthesized by the periphery fusion strategy. Further studies have indicated that the structure of these four macrocycles is determined by both size and curvature, showing unique variations from nearly planar to bowl and then to saddle. Interestingly, the nearly planar MS-4T with a small size and the rigid saddle-shaped MF-6T show no obvious response to fullerenes C-60 or C-70, while the bowl-shaped MS-6T and MF-4T demonstrate a strong binding affinity towards fullerenes C-60 and C-70. What's more, two kinds of co-crystals with capsule-like configurations, MS-6T@C-60 and MS-6T@C-70, have been successfully obtained, among which the former shows a loose columnar arrangement while the latter displays a unique three-dimensional honeycomb arrangement that is extremely rare in supramolecular complexes. This work systematically studies the pi-conjugated macrocycles and provides a new idea for the development of novel host-guest systems and further multifunctional applications.

Abstract:A synthetic strategy for the preparation of a series of polyimidazolium macrocycles from the corresponding dicarbene-derived metallacycles is described. Photodimerization of terminal cinnamic esters (UV-irradiation, λ=365 nm) produces the closed metallacycles with perfect stereoselectivity and high yields. Subsequent removal of the template from the photodimerization product results in polyimidazolium macrocycles. The size and shape of the receptor can be tuned easily by changing the length and breadth of the internal bridging groups of the ligands. Preliminary investigation shows the potential of the macrocycle as iodide sensor.

Abstract:Two o-carborane-based π-conjugated macrocycles are synthesized via oxidative coupling of acetylene-terminated o-carborane precursors. The single-crystal X-ray diffraction analysis unambiguously revealed their unique porous supramolecular structures, which display micropore characteristics. The novel macrocycles possess aggregation-induced emission both in aggregate state and solid state. This work provides a new design strategy for developing o-carborane macrocyclic molecules with excellent photophysical properties. Based on the obvious fluorescence quenching phenomenon of macrocycle by 1,4-dinitrobenzene and 2,4,6-trinitrophenol, the macrocyclic compounds have promising potential as sensors for nitroaromatics.

Abstract:The controlled synthesis of organometallic supramolecular macrocycles cages remains interesting and challenging work in the field of supramolecular chemistry. Here, two tetranuclear rectangular macrocycles and an octuclear cage were designed and synthesized utilizing a rigid and functionalized pillar linker, 2,6-bis(pyridin-4-yl)-1,7-dihydrobenzo [1,2-d:4,5-d′]diimidazole (BBI4PY) based on three half-sandwich rhodium building blocks bearing different sizes. X-ray crystallography in combination with 1H NMR spectroscopy elucidated that the two building blocks with shorter spacers only result in rectangular macrocycles. However, the building block of bulkier size to avoid the π-π stacking interactions between two ligands BBI4PY led to the formation of an octuclear cage complex. The latter cage contains two types of metal ions, namely Rh3+ and Cu2+, showing significant characteristics of heterogeneous metal-assembling compounds. In addition, the cage accommodates two free isopropyl ether solvent molecules, thus displaying host–guest behavior.

Abstract:Steric crowding of the 3-position of tris(pyrazolyl)borate and -methane ligands has produced tetrahedral metal complexes with controlled reactivity. As an alternative, we propose to incorporate the tris(pyrazolyl)methane chelate in a macrobicyclic structure in order to create a cavity with well-defined dimensions and shape. Acid-catalyzed equilibration of excess of the new pyrazole 3-(1H-pyrazol-3-yl)benzenemethanethiol acetate with HC(3,5-Me(2)pz)(3) followed by hydrolysis affords a functionalized tris(pyrazolyl)methane, which reacts with 1,3,5-tris(bromomethyl)benzene in K(2)CO(3)/DMF to give the title compound. Abstract:In this study, a twisted macrocycle was synthesized via ring closure of a double-helical aromatic oligoamide foldamer with two disulfide bridges. Single-crystal X-ray structure and NMR spectroscopy demonstrate the twisted conformation of macrocycle both in the solid state and in solution. As a result of the rearrangement of hydrogen bonding preference, the twisted conformation could be transformed to boxlike through protonation of the pyridine segments of macrocycle. In addition, the NMR titration experiments revealed that flat aromatic guests (e. g., coronene and perylene) could bind to the boxlike macrocycle with a 1 : 1 binding stoichiometry. The addition of base to the host-guest complexes resulted in conformational reversal of the macrocycle from boxlike to twisted as well as the release of guest.

Abstract:Trifluoroacetic acid (TFA) catalyzed condensation reaction between tetraaminooxacalix[4]arene and N-alkylcarbazole-3,6-dicarbaldehyde in CH2Cl2 afforded a single product in 87%∼89% yield. Well-defined yet undissolvable 1H NMR spectra suggested formation of robust and discrete structures in solution. X-ray single crystal analysis further revealed a giant twisted double-layer chiral macrocycle in the solid state, which was formed from [4 + 8] condensation of the two reactants via 16 imine bonds. DFT calculations discovered that only the [4 + 8] twisted product is thermodynamically favorable, which accounts for its highly selective and efficient formation out of a library of many other combinations.

Abstract:While a plenty of macrocyclic hosts have been developed in supramolecular chemistry, those that combine chiral luminescent properties and host-guest recognition abilities are still uncommon. Herein, two pairs of enantiomeric macrocycles were synthesized via Suzuki-Miyaura [2+2] cyclization reactions using Tröger's base and diphenyl maleimide as the building blocks. The diphenyl maleimide units impart these macrocycles with highly strong fluorescence, achieving quantum yields up to 100% in apolar solvents. Furthermore, the chiral, V-shaped Tröger's base units provide the macrocycles with circularly polarized luminescence (|glum| = 1.68 × 10-3) and well-define cavity for hosting electron-deficient or positively charged guests with Ka up to 1.7 × 106 M-1.

Abstract:A deoxycholic acid-based macrocycle CDTB was synthesized and showed selective recognition of Hg2+ ion utilizing 1,2,3-triazole motifs as binding sites. The resultant [CDTB.Hg2+] complex could further discriminate amino acids enantioselectively through the difference in fluorescence enhancement on account of the chiral scaffolds of deoxycholic acid and the macrocylic structure. This line of research not only enriches the library of cascade recognition, but also opens a door for chiral natural products in the field of the enantioselective discrimination. Abstract:The Friedel–Crafts reaction was employed to prepare the shape-persistent macrocycle (TPAP)3 with direct cyclization from three monomers of 2-(4′-(Phenyl(4″-methylphenyl)amino) phenyl)-propan-2-ol (TPAP), which contains two reactive groups of the t-hydroxyl and the p-proton on the triphenylamine. Under a very low monomer concentration (ca. 1%), the yield of the shape-persistent macrocycle (TPAP)3 is up to 34%. FT-IR, NMR, MALDI-TOF and MS were applied to determine the structure of macrocycle. It can be a good candidate as the hole-transporting layer because its photoluminescent spectrum appears no emission peaks in the range of 400–600nm. The geometry of (TPAP)3, optimized by the density functional theory (DFT) calculations using Gaussian03 software at the B3LYP/6-31G(d) level, showed that (TPAP)3 possesses an approximately 10Å diameter and 8.4Å depth molecular inner cavity. Owing to three nitrogenous lone electron pairs and six conjugated large π bonds of the macrocyclic electron-excessive phenylene almost directing into the cavity, the macrocycle is a big electron-rich donor which may be favorable for selective extraction of special diameter metal ions, high effective and selective catalysis and the formation of 1:1 π-donor–π-acceptor (C60) complexes through the host–guest interactions.
Smart macrocyclic molecules: induced fit and ultrafast self-sorting inclusion behavior through dynamic covalent chemistry.

Abstract:A family of macrocycles with oligo(ethylene glycol) chains, 4O, 5O, and 6O, was developed to construct a series of new incorporated macrocycles through dynamic covalent chemistry. These flexible macrocycles exhibited excellent "self-sorting" abilities with diamine compounds, which depended on the "induced-fit" rule. For instance, the host macrocycles underwent conformational modulation to accommodate the diamine guests, affording [1+1] intramolecular addition compounds regardless of the flexibility of the diamine. These macrocycles folded themselves to fit various diamines with different chain length through modulation of the flexible polyether chain, and afforded intramolecular condensation products. However, if the chain of the diamine was too long and rigid, oligomers or polymers were obtained from the mixture of the macromolecule and the diamine. All results demonstrated that inclusion compounds involving conformationally suitable aromatic diamines were thermodynamically favorable candidates in the mixture due to the restriction of the macrocycle size. Furthermore, kinetic and thermodynamic studies of self-sorting behaviors of both mixed 4O-5O and 4O-6O systems were investigated in detail. Finally, theoretical calculations were also employed to further understand such self-sorting behavior, and indicated that the large enthalpy change of H2NArArNH2@4O is the driving force for the sorting behavior. Our system may provide a model to further understand the principle of biomolecules with high specificity due only to their conformational self-adjusting ability.

As macrocyclic host molecules, they formed a 1:1 complex with fullerenes C-60 and C-70 with association constants ranging from 3 x 10(4) to 1 x 10(5) M-1.

Abstract:A series of macrocycles, including crown ethers, cyclodextrins, calixarenes, pillararenes and cucurbiturils, are well known to be able to associate various organic/inorganic/biological guest molecules and ions in their well-defined cyclic cavities to form stable host-guest complexes and supramolecular systems through the cooperative contributions of various non-covalent interactions. When one or more functional groups are attached to the cavity of macrocycles or guest molecules, enhanced and/or controlled host-guest associations may take place, leading to not only improved host-guest binding abilities but also fascinating properties. In this review, some representative contributions in the construction of controllable macrocyclic supramolecular assemblies in aqueous solution are presented with an emphasis on the stimuli-responsive control manner and wide applications of this property.

Abstract:By a one-pot reaction strategy and a stepwise fragment-coupling approach, three types of giant cage molecules with a triangular prism shape and an approximate cavity volume of 930 angstrom(3) were synthesized. Through efficient aromatic nucleophilic substitution reactions of chlorotriazine with functionalized amines or aliphatic substitution reactions of the linking imino moieties with allyl bromide, the giant cages were successfully functionalized on the surface with multiple hydroxy, 2-pyridyl, or allyl groups. As evidenced by variable-temperature H-1 NMR spectroscopy, the D-3h- and C-3v-symmetric cage molecules underwent very slow conformational changes or deformation in solution because of their giant size and their flexible building units. Their giant cavities, different electronic interiors, and facile multiple exterior surface functionalization could render the molecular cages powerful three-dimensional macrocyclic hosts that might find wide applications in supramolecular science.

Abstract:The molecular recognition of methylazacalix[4]pyridine (MACP-4; 1) towards various diols was investigated by using (1)H NMR spectroscopic and X-ray diffraction analysis. As a unique macrocyclic host molecule that undergoes conformational inversions very rapidly in solution, MACP-4 has been shown to self-regulate its conformation, through the formation of different conjugations of the four bridging nitrogen atoms with their adjacent pyridine rings, to form a cavity that best fits the guest species through intermolecular hydrogen-bond, C-H...pi, and pi-pi interactions between the host and guest. As a consequence, depending upon the diol structure and geometry, MACP-4 forms a 1:1 molecular sandwich, 2:1 molecular capsule, and 1:2 butterfly-layered complex with the guests. As a result of favorable enthalpy and entropy effects, MACP-4 exhibits excellent selectivity in the recognition of resorcinol, thus resulting in a very stable 1:1 sandwich complex with resorcinol with a binding constant of 6000 (M-1). The dynamic (1)H NMR spectroscopic study demonstrated that the 1,3-alternate conformation of the macrocyclic ring of the MACP-4resorcinol complex (13) is stable at low temperature (T<243 K), and its conformational inversion requires a larger activation energy (DeltaG++=(45.5+/-2.2) kJ mol(-1)). In the presence of an excess amount of resorcinol, however, the conformational inversion of the MACP-4resorcinol complex proceeds more readily with a decreased activation energy (DeltaG++=(33.5+/-1.5) kJ mol(-1)) owing most probably to the favorable enthalpy effect of the [3...1...3]++transition state.

Abstract:By performing Friedel-Crafts reaction with a mixture of 1,3,5-tri(furan-2-yl) benzene (TFB) and acetone, we successfully synthesized a cylinder-shaped macrocycle. The isolated yield of 36% is relatively high, considering that twelve C-C bonds are formed simultaneously. The structure of this macrocycle was confirmed using H-1 NMR spectroscopy, mass spectrometry and X-ray crystallography. This macrocycle is able to recognize a variety of cationic guests driven by cation-dipole and CH-pi interactions.

Abstract:In the arena of molecular recognition and sensing, the development of polypyrrolic macrocycles capable of anion chelation has gained significant attention over the last two decades. The host-guest chemistry of calixpyrroles, in particular, has led to a better understanding of the guest recognition by tailoring the size, geometry, and binding modes of the receptors. Calix[4]pyrrole (C4P), a very well-known member of the calix[n]pyrrole family, has smaller binding cavity to bind the larger anions. In order to modulate the molecular recognition and sensing properties of the C4P-based receptor, various structural modifications have successfully been realized. However, the core expansion of C4P has attracted vast attention due to the fact that it offers larger cavity size compared to the parent C4P-system. This opens the door towards the development of synthetically challenging higher ordered or expanded calixpyrroles for the effective and selective binding of large-sized guest entities. These rigid/preorganized macrocyclic architectures holds the multiple binding sites for enhanced binding affinities. Supramolecular synthetic chemists have expanded research on expanded calixpyrroles, and have achieved several milestones in this regard. This comprehensive review article covers both the synthetic approaches and recognition characteristics of the expanded calixpyrrole architectures. Apart from the expanded calixpyrroles, information on the synthetic approaches and recognition properties of contracted macrocyclic systems have also been revealed. To our knowledge, no comprehensive account of the synthetic challenges and peculiar recognition properties on the expanded and contracted calixpyrroles appeared in the literature until we almost finished this particular write-up. The authors believe that the current account will serve as a guide for the synthetic supramolecular chemists seeking novel design concepts for the construction of selective and efficient higher ordered macrocyclic systems with real-world applications.
