Abstract:Nanoparticles (NPs) can exert toxicity to aquatic animals through water and dietary exposure. However, little is known about the contribution of the two exposure pathways to the total toxicity of NPs to aquatic animals. In the present study, we fed Daphnia magna with ZnO NPs-accumulated-algae or directly exposed them to ZnO NPs suspension at the same Zn concentration. The body Zn concentrations were the same in both treatments, but the surface elastic modulus in the water-borne group was lower than that in the dietary-borne group, indicating more severe surface damage in the water-borne treatment. Total egg numbers of the first brood in both Zn treatments were significantly lower than the control, but they were not significantly different between the two treatments. However, the first brood in the water-borne treatment was distinctly delayed than that in the dietary-borne group. The dietary-borne treatment suppressed the expression of Hem and SOD genes. The water-borne treatment significantly suppressed the expression of Pro, SOD, VTG, but increased the GST expression. In particular, the expression of Pro, a gene functions in modulation of cell multiplication, was lower in the water-borne treatment, which might be related to the suppression of body growth in D. magna. The expression of SOD in the water-borne treatment decreased by about four times in comparison with the dietary-borne treatment, and the SOD enzyme activity of the water-borne treatment was significantly lower than that of the dietary-borne treatment. These results indicate that the two exposure routes have different targets and responding toxic mechanisms.
Altered Life History Traits and Transcripts of Molting- and Reproduction-Related Genes by Cadmium in Daphnia Magna.

Abstract:Cadmium (Cd) is a non-essential element and can be toxic to aquatic organisms at low concentrations. Despite its well-known toxicity to Daphnia magna, the effects of Cd on physiological parameters (heart rate and thoracic limb activity) and molting- and reproduction-related genes are relatively understudied. In this study, D. magna were exposed to 0 (control), 25, 50 and 75 μg L−1 of Cd for 7 d and 21 d to determine the toxicity of Cd. The results showed that the Cd body burden in D. magna was significantly increased with elevated Cd concentrations, up to 13.4 μg Cd/g dry weight (dw) after exposure to 75 μg L−1 for 21 d. After 21 d of exposure, the body length and body weight of D. magna were significantly decreased in all Cd treatments compared to the control. The heart rate and thoracic limb activity were reduced by 4.3–11.7 and 5.0–10.3%, respectively. The levels of malondialdehyde (MDA) were increased by ~24–37% and the activity of catalase (CAT) was inhibited by ~50% compared to the control. The reproductive parameters (i.e., size of the first brood, the total number of offspring per female and the number of offspring per brood) were remarkably reduced, causing adverse effects on the population dynamics. In addition, the transcripts of genes (cyp314, cyp18a1, ecra, usp, hr3, cut, cht and cht3) related to the molting of D. magna were altered, whereas the transcripts of genes (vtg1, vtg2 and vmo1) related to reproduction were down-regulated. This study helps better understand the effects of Cd at different biological levels.
Effects of cadmium on consumption, assimilation and biochemical parameters of Daphnia magna: possible implications for reproduction

Abstract:1. The effects of cadmium on consumption, assimilation rates and biochemical parameters of Daphnia magna were determined. 2. The consumption and assimilation rates of 14 days 1.0 ppb Cd treated animals tended to decrease slightly, the decline of these rates at 5.0 ppb Cd (14 days), however, was highly significant (P less than 0.001). 3. The assimilation efficiencies of daphnids exposed to cadmium did not significantly differ from control. 4. No notable changes in the biochemical composition of daphnids could be noticed after 7, 14 and 21 days of cadmium exposure. 5. It seems as if not one metabolic process in particular was depressed due to cadmium, but metabolic activities seemed to be inhibited on the whole. 6. Results are discussed in relation with data of a previous study on the reproduction of D. magna under cadmium stress.

Abstract:The effect of sublethal concentrations of heavy metals on cladoceran growth and reproduction is a cornerstone of modern ecotoxicology. However, the literature contains assays across numerous concentrations, on numerous species and genotypes, and conditions are far from consistent. We undertook a systematic review of the sublethal effects of copper and cadmium concentrations on Cladocera spp. life history (reproduction, maturation age, and somatic growth rate). Using meta-analysis, we tested the hypothesis that the effects of increasing Cu and Cd concentrations on traits may vary by species. We also evaluated where possible whether the effect of metal concentrations on traits vary by water hardness, exposure duration, or whether the metals were delivered in aqueous solution or via food. We surveyed > 200 papers, resulting in a set of 32 experimental studies representing 446 trials where the results were presented compared with Daphnia magna—the most commonly assayed species. We found qualitatively similar effects of Cu and Cd on life history traits that included reduction in reproduction and somatic growth rate and delay of maturation. Cladocera species showed marked variations in their susceptibility to metals, and D. magna was found to be the least sensitive species to sublethal changes in reproduction. The effects were largely consistent for aqueous vs. dietary food. Water hardness, where data were available, had no detectable effect. Available data indicate that exposure duration had no effect on the toxicity of Cu but did for D. magna reproductive response to Cd. Our study highlights the importance of including species identity when considering toxicological testing and regulation development.

Abstract:Toxicity tests of chemicals have mainly focused on the partial life-cycle evaluation of model animals. Limited information is available for the evaluation of effects of chemicals from a whole-life-stage exposure perspective. The objective of this study was to perform a whole-life-stage characterization in the basic biology of Daphnia magna (D. magna) and evaluate the effects of a known organophosphate ester (OPE) contaminant, tris(1,3-dichloro-2-propyl) phosphate (TDCIPP), on growth, reproduction, survival, and transcription of genes. The whole-life-stage characterization in growth, reproduction, and survival of D. magna was conducted, and representative sampling time points for the three developmental stages were identified (day 6, day 32, and day 62). Transcriptomic profiles for these three stages were compared, and stage-specific PCR arrays of D. magna were developed. The whole-life-stage exposure to environmentally relevant or greater concentrations of TDCIPP significantly inhibited growth and reproduction of D. magna and decreased survival at the later stage of the exposure experiment (≥32 days). Such adverse effects were not observed in the early stage of the exposure (<32 days), suggesting that short-term toxicity tests, such as the standard 21-day test, might underestimate the environmental risk of TDCIPP. Furthermore, expressions of genes selected at day 6, day 32, and day 62 were significantly changed after TDCIPP exposure, and the changes in the expressions of partial genes were correlated to the inhibitory effects on growth, reproduction, and survival.

Abstract:Cadmium (Cd) is being frequently detected in marine organisms. However, dose-dependent effects of Cd challenged unraveling the toxicological mechanisms of Cd to marine organisms and developing biomarkers. Here, the dose-dependent effects of Cd on clams Ruditapes philippinarum following exposure to 5 doses of Cd (3, 9, 27, 81, 243 μg/L) were investigated using benchmark dose (BMD) method. By model fitting, calculation of BMD values was performed on transcriptomic profiles, metals concentrations, and antioxidant indices. Cd exposure induced not only significant Cd accumulation in clams, but also marked alterations of essential metals such as Ca, Cu, Zn, Mn, and Fe. Gene regulation posed little influence on essential metal homeostasis, indicated by poor enrichment of differentially expressed genes (DEGs) associated with metal binding and metal transport in lower concentrations of Cd-treated groups. BMD analysis on biological processes and pathways showed that peptide cross-linking was the most sensitive biological process to Cd exposure, followed by focal adhesion, ubiquitin mediated proteolysis, and apoptosis. Occurrence of apoptosis was also confirmed by TUENL-positive staining in gills and hepatopancreas of clams treated with Cd. Furthermore, many DEGs, such as transglutaminases (TGs), metallothionein (MT), STEAP2-like and laccase, which presented linear or monotonic curves and relatively low BMD values, were potentially preferable biomarkers in clams to Cd. Overall, BMD analysis on transcriptomic profiles, metals concentrations and biochemical endpoints unraveled the sensitiveness of key events in response to Cd treatments, which provided new insights in exploring the toxicological mechanisms of Cd in clams as well as biomarker selection.

Effects of cadmium on fecundity and defence ability of Drosophila melanogaster

Abstract:Cadmium (chemical symbol, Cd) is an extremely common pollutant that poses a toxicity threat to organisms. Therefore, we tested Drosophila melanogaster fecundity, Cd accumulation, and activity of two enzymes following Cd stress and used quantitative real-time polymerase chain reaction (qPCR) to quantify the mRNA expression levels of several genes involved in fecundity and defence. D. melanogaster was placed in a medium containing different concentrations of Cd (13, 26, and 52 mg L-1), following which, inductively coupled plasma atomic emission spectroscopy showed that Cd accumulation in Drosophila increased with the increase in its dietary intake. We also observed that Cd at these concentrations significantly prolonged the mating latency in females and reduced the number of eggs laid. However, the same Cd concentrations did not affect male fecundity. Acetylcholinesterase activity was only detected at 52 mg L-1 Cd in both sexes, whereas glutathione S-transferase activity was inhibited at 26 and 52 mg L-1 Cd in females. The results of qPCR indicated that exposure to 13-52 mg L-1 Cd affected the expression of reproduction-related genes, including downregulation of enok and upregulation of dally and dpp. The same level of exposure also induced transcriptional responses from three defence-related genes (hsp70, gstd2, and gstd6). Taken together, the results revealed that Cd exposure might negatively affect the expression of genes associated with D. melanogaster reproduction and trigger the transcription of defence-related genes. We suggest that further analyses of fecundity and defence responses may help develop indicators of Cd toxicity and improve our understanding of antitoxin defences.
Multigenerational effects of cadmium on the lifespan and fertility of Drosophila melanogaster

Abstract:Although the damage and tolerance mechanisms of Cd stress are known, the data on genetic risk are limited. The aim of this study was to assess the chronic toxicity of Cd, genetic responses, and multigenerational effects in five generations of Drosophila melanogaster. For each generation, lifespan and fertility were statistically analysed and the expression of apoptosis- (p53 and caspase-3) and epigenesis-related (dDnmt2 and dMBD2/3) genes was examined. Lifespan and fertility significantly declined under Cd stress and these effects were maintained for two generations and one generation, respectively, when Cd stress was removed. The expression of p53 and caspase-3 was significantly up-regulated after exposure, suggesting that apoptosis contributes to the resistance mechanism. Their altered expression was retained for two generations. Furthermore, high expression of dDnmt2 and dMBD2/3 accompanied Cd exposure, which was passed on to three generations, suggesting that genetic modifications in apoptosis-related genes are carried to the offspring through epigenetic regulation.

Abstract:With water eutrophication and global warming, cyanobacteria blooms have occurred frequently, and the interaction between M. aeruginosa and Daphnia has been widely paid attention by researchers. However, the effects of toxic M. aeruginosa on the SOD activity of Daphnia are poorly known. Six D. similoides sinensis clones collected from Lake Junshan and the offspring of two clones were employed. The effects of toxic M. aeruginosa on the life history traits and SOD activities of D. similoides sinensis in the mother and their offspring were studied. Toxic M. aeruginosa could significantly inhibit the life history traits (e.g., body lengths, offspring numbers at first reproduction, cumulative offspring numbers, and the intrinsic rate of population) and induce higher SOD activities of D. similoides sinensis . Compared with the mother, the effects of toxic M. aeruginosa on the life history traits and SOD activities of D. similoides sinensis in the offspring showed obvious differences. Moreover, the adaptability of the offspring to M. aeruginosa indicated also the differences between two clones. Our results suggested that the mother exposed to toxic M. aeruginosa could enhance the fitness of their offspring to Microcystis by maternal effect and was also affected by the D. similoides sinensis genotypes.

Abstract:Cadmium (Cd) is a heavy metal that accumulates easily in organisms and causes several detrimental effects, including tissue damage. Cd contamination from anthropogenic terrestrial sources flows into rivers, and through estuaries to the ocean. To evaluate the toxic effects of Cd on estuary crustaceans, we exposed the mud crab Scylla paramamosain to various Cd concentrations (0, 10.0, 20.0, and 40.0mg/L) for 24h. We also exposed mud crabs to a fixed Cd concentration (20.0mg/L) for various periods of time (0, 6, 12, 24, 48, and 72h). We observed that after exposure to Cd, the surfaces of the gill lamellae were wrinkled, and the morphologies of the nuclei and mitochondria in the hepatopancreas were altered. We analyzed the expression profiles of 36 stress-related genes after Cd exposure, including those encoding metallothioneins, heat shock proteins, apoptosis-related proteins, and antioxidant proteins, with quantitative reverse transcription PCR. We found that exposure to Cd altered gene expression, and that some genes might be suitable bioindicators of Cd stress. Gene expression profiles were organ-, duration-, and concentration-dependent, suggesting that stress-response genes might be involved in an innate defense system for handling heavy metal exposure. To the best of our knowledge, this study is the first one of histopathology and stress-response gene expression pattern of Scylla paramamosain after Cd exposure. Our work could increase our understanding of the effect of environmental toxins on estuary crustaceans.

Abstract:This work set out to study the effects of cadmium (Cd) in sediments on the antioxidant enzyme activities in the digestive gland of Asian clam Corbicula fluminea and to explore the potential for applying these responses to evaluate the Cd-contaminated sediment. Additionally, diffusive gradients in thin-films (DGT) technique was used to predict the response of its antioxidant defense system. The sediments, collected from Taihu Lake, were spiked with Cd at different concentrations (0.72, 0.91, 1.62, 2.59, 11.2, 20.4 and 40.6 mg kg(-1), dry weight). Asian clam was cultivated for 28 days. Concentrations of Cd in the body of Asian clam had a good relationship with concentrations of Cd in overlying water and sediments, as measured by DGT. Cd affected these biochemical parameters significantly, especially for superoxide (SOD), peroxidase (POD) and glutathione (GSH), which began to show higher sensitivity and have a significant difference in low dosage (0.91 mg Cd kg(-1)) compared with blank. Contents of MDA and MT, which were induced by Cd, increased with the increasing Cd concentration in sediments and reached peak values at 11.2 and 20.4 mg kg(-1), respectively, after 28 days exposure. All of these results suggested that biochemical responses cooperated in detoxifying and maintaining cellular metabolic homeostasis. The R(2) of regression analysis between the contents of MT and the concentrations of Cd measured by DGT, in sediments and soft body were 0.71, 0.94 and 0.88 after 28 days exposure. This suggested that DGT measurement could predict the response of MT. Cd accumulation, GSH and MT were indispensable biomarkers and the MDA content and DGT appeared to be promising biomarkers. The results clearly indicated that Cd could induce oxidative stress in the digestive gland of Asian clam. The combination of biomonitors with DGT can obtain different information about Cd bioavailability and confirm the significance of applying a suite of biomarkers rather than a selective index to assess the sublethal effect. It also offered theoretical methods for the prediction of sediment Cd pollution.
Chronic Toxicity Testing with Daphnia magna in Three Generations

Abstract:This work shows the informativeness and predicted value of multigenerational testing on Daphnia magna Straus on exposure of 75 days in comparison with standard bioassays for chronic toxicity on exposure of 21–24 days. D. magna were exposed in water polluted with strontium chloride (1 and 2 mg Sr/L). No acute toxicity (death within 96 hours) and chronic toxicity (mortality and fertility within 24 days) were observed. In 50 days of the experiment, mortality of crustaceans was more than 30%, and fertility decreased by 2 times (P < 0.05). In 75 days of the experiment, mortality of the experimental daphnia was above 90%, and most of the individuals did not reproduce. The effects intensified in subsequent generations F2 and F3. In the third generation, the number of juveniles in 24 days of exposure (2 mg Sr/L) was only 9% of the control value (P = 0.03); then the juveniles did not appear. We also noted an increase in the number of abortive eggs and dead juveniles in generations F2 and F3 compared with F1. The totality of the observed effects testifies the delayed toxic effect of strontium. Delayed mortality and reduced fertility of the remaining individuals in a series of generations threaten the existence of the population of organisms. Therefore, chronic toxicity testing using D. magna is a method for detecting and predicting loss of biodiversity in the environment.
Importance of metallothioneins in the cadmium detoxification process in Daphnia magna

Abstract:Good knowledge of the relationship between toxic metals and biological systems, particularly the sub-cellular fraction, could be a suitable early indicator of toxic effects. These effects and the sub-cellular behaviour of cadmium were studied with a widely used species in freshwater toxicity bioassays, Daphnia magna. In spite of this very commonplace usage in ecotoxicological studies, very few data are available on its toxicant metabolism and in particular metal homeostasis. Combining multi-tools analysis, a soluble protein was found: it is heat-stable, rich in sulfhydryl groups (differential pulse polarography), characterised by a molecular mass of approximately 6.5 kDa, with a G-75 chromatographic profile corresponding to the rabbit metallothioneins monomer, with few if any aromatic-containing amino acids, it binds metals (e.g. Cd, Cu), and its concentration increases with Cd exposure. This evidence led us to hypothesise that metallothioneins (MTs) are present in D. magna. Up to 75% of the Cd body burden with Cd exposure is bound to the MTs fraction. The increase in the Cd concentration in the surrounding medium and concomitantly in daphnids induces sub-cellular reorganisation of essential metals such as Cu and Zn. The rate of metals in the soluble cellular fraction and associated with MTs increases with the Cd body burden. Monitoring sub-cellular distribution of metals after exposure in the natural environment could be very useful for ecotoxicological assessment.

Abstract:Cadmium (Cd) can adversely affect aquatic life, altering reproductive and molting processes in crustaceans. The objective of this study was to evaluate the influence of Cd on reproduction and molting in the crab Callinectes danae. Adult females were obtained from environments with different levels of pollution: low (LC), medium (MC), and high contaminated (HC) areas. Animals from LC, MC, and HC areas were exposed to 0, 0.5, and 2 mg L-1 of CdCl2 for 3 h. Cd bioaccumulation, oxidative stress (evaluated by antioxidant enzymes activity), and lipid peroxidation (LPX) were analyzed in mature ovaries (stage II), gills, and hepatopancreas. The expression levels of crustacean hyperglycemic hormone (CHH) and molt-inhibiting hormone (MIH) genes were quantified in the eyestalks, while 17β-estradiol (E2) and melatonin concentration were measured in the hemolymph. Cd bioaccumulated mainly in the hepatopancreas and gills, with increased E2, LPX, and antioxidant enzymes in HC compared to the LC region. Decreased CHH and MIH transcripts were observed in the animals from HC regions compared to LC and MC areas. Physiological differences were recorded, especially for bioaccumulation, oxidative stress, and hormone levels, in animals sampled in HC areas compared to LC and MC regions. In conclusion, the physiological damage triggered by Cd could be reduced due to higher levels of melatonin and antioxidant enzymes in HC areas.
Developmental Toxicity in Daphnia Magna Induced by Environmentally Relevant Concentrations of Carbon Black: from the Perspective of Metabolomics and Symbiotic Bacteria Composition.

Abstract:The level of carbon black (CB) pollution in the environment is rapidly increasing, owing to the increase in natural and industrial emissions. The water environment has become an important sink for CB. However, studies on CB mainly focused on its impact on air pollution and phytoremediation applications, and the toxicity mechanism of CB in aquatic organisms is relatively limited. Thus, Daphnia magna was used as a model organism to explore the developmental toxicity of environmentally relevant concentrations of CB under a full life-cycle exposure. The toxicity mechanism of CB in aquatic organisms was investigated based on metabolomic and symbiotic microbial analyses. It was found that compared with the control group, the body length of exposed D. magna decreased, while the mortality and intestinal inflammation increased with increasing concentration of CB. The normal reproductive regularity of D. magna was disturbed, and the deformity and body length of the offspring increased and decreased, respectively, after CB exposure. Metabolomic analysis showed that the urea cycle metabolic pathway of exposed D. magna was increased significantly, suggesting a perturbation of N metabolism. In addition, two eicosanoids were increased, suggesting possible inflammation in D. magna. The levels of seven phospholipid metabolites decreased that might be responsible for offspring malformations. Microbiological analysis showed that the composition of the symbiotic microbial community of D. magna was disturbed, including microorganisms involved in carbon cycling, nitrogen cycling, and biodegradation of pollutants, as well as pathogenic microorganisms. Overall, this study found that the inflammatory related metabolites and symbiotic bacterial, as well as reproductive related metabolites, were disrupted after D. magna exposed to different concentrations of CB, which revealed a possible developmental toxicity mechanism of CB in D. magna. These findings provide a scientific basis for analyzing the risks of CB in aquatic environments.
Effects of exposure to cadmium, microplastics, and their mixture on survival, growth, feeding, and life history of Daphnia magna

Abstract:There is concern that microplastics can act as a vector for cadmium, altering the bioavailability and subsequent toxicity of cadmium to ecologically important species such as Daphnia magna. The toxicity of cadmium to D. magna has been well‐described; however, what is not known, and what this study aimed to address was how the addition of polyethylene microplastics altered cadmium toxicity. Using high‐throughput feeding assays and size assessments, this study quantified effects of exposure to cadmium, microplastics, or their mixture on daphnids from neonate to adult. Exposure to cadmium inhibited feeding efficiency while exposure to microplastics inhibited growth rates of juveniles. Daphnia magna co‐exposed to cadmium and microplastics showed significant decreases in both feeding and pre‐reproductive growth rate. There were no differences in life history traits across any treatments. The alterations of feeding and growth while maintaining reproductive endpoints (time to first brood, reproductive frequency, the number of neonates released at each reproductive event, and the size of neonates produced) might be the result of a shift in energy allocation away from somatic growth, allowing individuals to maintain reproductive output despite lower nutritional reserves. Our findings suggest that co‐contamination of microplastics and cadmium has additive effects on feeding and growth rates, resulting in a greater energy allocation shift.This article is protected by copyright.
