Abstract:Intracellular calcium concentration ([Ca2+]i) in articular chondrocytes changes during mechanical challenges associated with joint movements, because of the fluctuation of the extracellular osmotic environment during joint loading. Matrix synthesis by chondrocytes is modulated by loading patterns, possibly mediated by variations in intracellular composition, including [Ca2+]i. The present study has employed the Ca(2+)-sensitive fluoroprobe Fura-2 to determine the effects of hypertonic shock on intracellular Ca2+ concentration ([Ca2+]i) and to characterise the mechanisms involved in the response for isolated bovine articular chondrocytes. In cells subjected to a hypertonic shock, [Ca2+]i rapidly increased by approximately 300%, reaching a maximal value within 50 s following the hypertonic shock with a recovery of more than 90% towards the initial [Ca2+]i within 5 min. The effect was inhibited by removal of extracellular Ca2+ ions, but not by thapsigargin, indicating that the rise in [Ca2+]i is only a result of influx from the extracellular medium. The rise was insensitive to inhibitors of L-type voltage-activated Ca2+ channels, TRPV channels or stretch-activated cation channels. Non-specific inhibitors of Ca2+ channels like CdCl2, NiCl2, LaCl3 and ZnCl2 significantly attenuated the response, although the extent in which CdCl2 and NiCl2 (both of them inhibitors of annexin-mediated Ca2+ fluxes) inhibited the response was significantly greater. The rise was also sensitive to KBR7943, inhibitor of NCE reverse mode and trifluoperazine, inhibitor of the activity of annexins. Hypertonic shock also produced also hyperpolarisation of chondrocytes (Em measured by means of Di-BA-C4(3), a membrane potential sensitive dye), which was inhibited by TEA-Cl and BaCl, but was not affected by changing the extracellular solution to Ca(2+)-free HBS. Inhibition of hyperpolarisation completely abolished the [Ca2+]i rise following hypertonic shock. Treatment with retinoic acid, which can increase the activity of annexins as Ca2+ transport pathways caused a significant increase in [Ca2+]i. The recovery of [Ca2+] was inhibited by benzamil and was dependent on extracellular Na+, but was unaffected by Na-orthovanadate, an inhibitor of plasma Ca(2+)-ATPase. We conclude that in response to hypertonic shock, NCE reverse mode and annexins are the pathways responsible for the [Ca2+]i increase, while forward mode operation of NCE is responsible for the subsequent extrusion of Ca2+ and recovery of [Ca2+]i towards initial values.

Abstract:Activation and inactivation of ion channels involve volume changes from conformal rearrangements of channel proteins. These volume changes are highly susceptible to changes in ambient pressure. Depending on the pressure level, channel function may be irreversibly altered by pressure. The corresponding structural changes persist through the post-decompression phase. High-pressure applications are a useful tool to evaluate the pressure dependence as well as pressure limits for reversibility of such alterations. Mammalian cells are only able to tolerate much lower pressures than microorganisms. Although some limits for pressure tolerance in mammalian cells have been evaluated, the mechanisms of pressure-induced alteration of membrane physiology, in particular of channel function, are unknown. To address this question, we recorded fast inward sodium (INa) and slowly activating L-type calcium (ICa) currents in single mammalian muscle fibers in the post-decompression phase after a prolonged 3-h, high-pressure treatment of up to 20 MPa. INa and ICa peak amplitudes were markedly reduced after pressure treatment at 20 MPa. This was not from a general breakdown of membrane integrity as judged from in situ high-pressure fluorescence microscopy. Membrane integrity was preserved even for pressures as high as 35 MPa at least for pressure applications of shorter durations. Therefore, the underlying mechanisms for the observed amplitude reductions have to be determined from the activation (time-to-peak [TTP]) and inactivation (τdec) kinetics of INa and ICa. No major changes in INa kinetics, but marked increases, both in TTP and τdec for ICa, were detected after 20 MPa. The apparent molecular volume changes (activation volumes) ΔV ‡ for the pressure-dependent irreversible alteration of channel gating approached zero for Na+ channels. For Ca2+ channels, ΔV‡ was very large, with approx 2.5-fold greater values for channel activation than inactivation (approx 210 Å3). We conclude, that in skeletal muscle, high pressure differentially and irreversibly affects the gating properties and the density of functional Na+ and Ca2+ channels. Based on these results, a model of high pressure-induced alterations to the channel conformation is proposed.

Abstract:There is a growing literature on how natural resources affect both economic performance and political regimes. In this article the authors add to this literature by focusing on how natural resource wealth affects the incentives of governments to uphold contracts with foreign investors across all sectors. They argue that although all states suffer reputation costs from reneging on contracts, governments in natural-resource-dependent economies are less sensitive to these costs, leading to a greater probability of expropriation and contract disputes. Specifically, leaders weigh the benefits of reneging on contracts with investors against the reputation costs of openly violating agreements with firms. The authors’ theoretical model predicts a positive association between resource wealth and expropriation. Using a data set from the political risk insurance industry, the authors show that resource dependent economies have much higher levels of political risk.
Hydrostatic pressure enhances chondrogenic differentiation of human bone marrow stromal cells in osteochondrogenic medium

Abstract:This study demonstrated the chondrogenic effect of hydrostatic pressure on human bone marrow stromal cells (MSCs) cultured in a mixed medium containing osteogenic and chondrogenic factors. MSCs seeded in type I collagen sponges were exposed to 1 MPa of intermittent hydrostatic pressure at a frequency of 1 Hz for 4 h per day for 10 days, or remained in identical culture conditions but without exposure to pressure. Afterwards, we compared the proteoglycan content of loaded and control cell/scaffold constructs with Alcian blue staining. We also used real-time PCR to evaluate the change in mRNA expression of selected genes associated with chondrogenic and osteogenic differentiation (aggrecan, type I collagen, type II collagen, Runx2 (Cbfa-1), Sox9, and TGF-beta1). With the hydrostatic pressure loading regime, proteoglycan staining increased markedly. Correspondingly, the mRNA expression of chondrogenic genes such as aggrecan, type II collagen, and Sox9 increased significantly. We also saw a significant increase in the mRNA expression of type I collagen, but no change in the expression of Runx2 or TGF-beta1 mRNA. This study demonstrated that hydrostatic pressure enhanced differentiation of MSCs in the presence of multipotent differentiation factors in vitro, and suggests the critical role that this loading regime may play during cartilage development and regeneration in vivo.
Pressure does not affect fibrotic function of dermal fibroblast through an in vitro 3D hydrogel culture model

Abstract:Pressure therapy has been used for the prevention and treatment of hypertrophic scars for decades. However, the cellular and molecular mechanisms of this treatment modality have not been fully elaborated, leading to long‐lasting controversies regarding its clinical effectiveness. In this current study, we adopted an in vitro 3D culture and compression model to explore the effect of pressure force on fibroblasts, in order to further explain the working mechanism of compression force during pressure treatment. Human dermal fibroblasts were cultured in the 3D culture hydrogel and treated with 1.5 atm of external compression force through a syringe tube device, for 4, 8, and 20 h respectively. RNA‐seq identified 437 differentially regulated genes after an 8‐h compression intervention compared with control cells, among which 256 genes were up‐regulated and 181 genes were down‐regulated. Further q‐PCR analysis confirmed that early growth response 1(EGR1) and c‐fos were down‐regulated after an 8‐h compression intervention. However, the down‐regulation of EGR1 and c‐fos at the mRNA level does not lead to altered protein synthesis through western blot, for both 8 and 20‐h time points after pressure intervention. Genes closely related to the fibrotic function of fibroblasts including type I collagen (COL1), type III collagen (COL3), transforming growth factor β1(TGF‐β1), matrix metallopeptidase 1 (MMP1), matrix metallopeptidase 1 (TIMP1), connective tissue growth factor (CTGF), α smooth muscle actin (α‐SMA), and fibronectin 1 (FN1), were also unaffected after pressure treatment for 8 h. The current study indicated that in our 3D hydrogel culture model, pressure does not directly affect the fibrotic function of dermal fibroblast in vitro. Indirect regulation including reducing oedema, blood perfusion, and tension could be a more possible mechanism of pressure therapy.

Effects of hydrostatic pressure on matrix synthesis in different regions of the intervertebral disk

Abstract:The intervertebral disk is routinely subjected to compressive loads that alter with posture and muscle activity and can produce pressures > 2 MPa in human lumbar disks in vivo (A. Nachemson and G. Elfstrom. Scand. J. Rehabil. Med. 2, Suppl. 1:1-40, 1979; A. Nachemson and J. M. Morris. J. Bone Jt. Surg. Am. Vol. 46A: 1077-1092, 1964). We measured the effect of load on hydrostatic pressures in bovine caudal disks. With increase in applied load, pressure increased linearly in the nucleus and inner annulus. The resting pressure measured after slaughter (0.19 +/- 0.05 MPa) and the pressure at failure (34 MPa, estimated from the vertebrae/disk segment failure load of 7,430 +/- 590 N) define the limits that can occur in vivo. Because hydrostatic pressure influences matrix synthesis in articular cartilage, we have examined the effects of pressures in the range 1-10 MPa applied for 20 s or 2 h on proteoglycan synthesis in bovine caudal and human lumbar intervertebral disks in vitro. In the nucleus pulposus and inner annulus of bovine disks, application of hydrostatic pressure in the range of 1-7.5 MPa for only 20 s stimulated matrix synthesis over the following 2 h at atmospheric pressure. The maximum stimulation in the bovine disks was seen in the inner annulus after application of 2.5 MPa, where proteoglycan synthesis rates doubled. Exposure to 2.5 MPa also stimulated synthesis in the nucleus pulposus of human disks taken at surgery, whereas 7.5 MPa inhibited synthesis in five out of six specimens. With 2-h continuous exposure to the same levels of pressure, no stimulation was seen in the nucleus of bovine disks, and significant stimulation was only observed at 5.0 MPa in the inner annulus. Exposure to 10 MPa for either 20 s or 2 h inhibited proteoglycan synthesis in these regions of the disks. In contrast, in the outer annulus, where loading does not lead to a rise in hydrostatic pressure in vivo, there was no significant response to hydrostatic pressure over the range of 1-10 MPa in bovine or human disks.

Abstract:Many studies have illustrated the effect of mechanical loading on articular cartilage and the corresponding changes in chondrocyte metabolism, yet the mechanism through which the cells respond to loading still is unclear. The purpose of this study was to evaluate the change in shape of chondrocytes under a statically applied uniaxial compressive load. Isolated chondrocytes from rat chondrosarcoma were embedded in 2% agarose gel. Strains of 5, 10, and 15% were applied, and images of the cell were recorded from initial loading to equilibrium (15 minutes). A finite-element model was used to model the experimental setup and to estimate the mechanical properties of the chondrocyte at equilibrium. The transient behavior of the composite in the experiment was analyzed with use of a standard linear viscoelastic model. We found that all cells decreased in cross-sectional area under each of the applied compressive strains. In the finite-element model, the elasticity of the chondrocyte was similar to that of the surrounding agarose gel (4.0 kPa) and had a Poisson's ratio of 0.4. Viscoelastic analysis showed that the chondrocytes contributed a significant viscoelastic component to the behavior of the composite in comparison with the agarose gel alone. If a decrease in cell volume proportional to the decrease in cross-sectional area is assumed, the decrease observed was greater than would be predicted by a passive cellular response due to an equivalent osmotic pressure. This indicates that the chondrocyte may be altering its intracellular composition by cellular processes in response to mechanical loading.

The Effect of the Static Pressure on Expression of MMP-13 in Condylar Chondrocytes

Abstract:Objective: To investigate the effect of the static pressure on the mRNA and protein expression of MMP-13 in condylar chondrocytes. Methods: Condylar chondrocytes were dissected and cultured from the TMJ of 3-week-old SD rat in vitro. The chondrocytes of P3 were identified and were subjected to different static pressure of 0, 50, 100, 150 and 200 kPa respectively for 2 h, while the monolayer of condylar chondrocytes were subjected to static pressure of 200 kPa for different time (0, 1, 2, 4, 8 and 12 h). Expression of MMP-13 was examined by Western blot and quantitative real-time PCR. Results:The static pressure with a magnitude from 50 to 200 kPa can reduce the expression of MMP-13 of condylar chondrocytes, and has a significant difference (P<0.05). Under static pressure of 200 kPa, with the compression time ex-tended, the expression of MMP-13 first increased and then decreased, and up to the highest level at 4 h(P<0.05). Conclu-sion:The results indicated TMJ condylar chondrocytes has a certain adaptability to static compression.

Abstract:Mechanical stimulation plays in an important role in regulating stem cell differentiation and their release of extracellular vesicles (EVs). In this study, effects of low magnitude hydrostatic pressure (HP) on the chondrogenic differentiation and microvesicle release from human embryonic stem cells (hESCs) and human bone marrow stem cells (hBMSCs) are examined. hESCs were differentiated into chondroprogenitors and then embedded in fibrin gels and subjected to HP (270 kPa, 1 Hz, 5 days per week). hBMSC pellets were differentiated in chondrogenic media and subjected to the same regime. HP significantly enhanced ACAN expression in hESCs. It also led to a significant increase in DNA content, sGAG content and total sGAG/DNA level in hBMSCs. Furthermore, HP significantly increased microvesicle protein content released from both cell types. These results highlight the benefit of HP bioreactor in promoting chondrogenesis and EV production for cartilage tissue engineering.

Light and low-frequency pulsatile hydrostatic pressure enhances extracellular matrix formation by bone marrow mesenchymal cells: an in-vitro study with special reference to cartilage repair

Abstract:Ovine bone marrow mesenchymal cells (BMMCs) were seeded on to non-woven filamentous plasma-treated polyester scaffolds and cultured in a chondrogenic medium for 4 weeks. Thereafter a pulsatile hydrostatic pressure (PHP) was applied to these cell-scaffolds constructs at an amplitude of 0.1 MPa and frequency of 0.25 Hz, for 30 min a day, over a period of 10 days. Samples (n = 6) were removed 24 h after PHP stimulation at days 1, 4, 7, and 10 for biochemical analysis. Similar analyses were conducted, at the same time points, on control samples that were not subjected to a PHP. The results showed that the glycosaminoglycan (GAG) content did not significantly increase until after the application of a PHP for 7 days. The GAG content was 1.5 and 2.7 times higher in the PHP group than in the control group at days 7 and 10 respectively (p<0.01). The deoxyribonucleic acid (DNA) content was 1.5 times greater in the PHP group than in the control group at day 10 (p<0.01). GAG synthesis amounts, expressed as the total GAG contents per microgram of DNA, were 1.6 and 1.8 times higher in the PHP group than in the control group at days 7 and 10 respectively (p<0.01). The total collagen content in the medium did not change until after PHP application for 10 days, when it was 1.9 times higher than the control (p < 0.05). The results suggest that a light PHP applied at a low frequency has a cumulative stimulatory effect on the BMMCs' metabolic activities including cell proliferation and synthesis of the extracellular matrix.
HMGB2 promotes chondrocyte proliferation under negative pressure through the phosphorylation of AKT

Abstract:Cells in articular cartilage are zonal arranged. Cells in superficial zone cartilage are generally small and proliferative. Appropriate negative pressure stimulation is beneficial to cell survival and tissue repair. Whether negative pressure has promotive impact on the proliferation activity of the superficial zone chondrocytes is of interest. In this study, we isolated superficial chondrocytes from the mandibular condylar cartilage of rats. After negative pressure treatment, the cells were collected for RNA-sequencing, quantitative real-time PCR and western blotting assays, aiming to detect the proliferative responses of chondrocytes to negative pressure and explore the potential molecular mechanisms. Data from RNA-sequencing analysis indicated that the superficial chondrocytes responded to the 4 h -10 kPa treatment by a significant increase in proliferation. In addition, the expression of high-mobility group box 2 (HMGB2) and the phosphorylation of AKT were obviously promoted. Knockdown of HMGB2 decreased AKT phosphorylation and diminished the negative pressure-induced proliferation of chondrocytes, as shown by decreased expression of Ki67 and cyclin-dependent kinase 6 (CDK6). In contrast, overexpression of HMGB2 enhanced AKT phosphorylation and further promoted proliferative activity. Moreover, LY294002, an AKT inhibitor, suppressed the proliferative activity of chondrocytes under negative pressure, while SC79, an activator of AKT phosphorylation, enhanced the proliferation of chondrocytes. Our data demonstrated that HMGB2 exhibits a promotion impact on chondrocyte proliferation under negative pressure via the phosphorylation of AKT. These results provide a new perspective for superficial zone chondrocytes proliferation under negative pressure, which should be benefit for cartilage regeneration.
The expression and significance of HIF-2αin condyle chondrocytes under different stress loadings

Abstract:Objective To explore the expression of HIF-2αand its downstream catabolic factors (MMP3, MMP13, ADAMTs-4) in the mandibular condyle chondrocytes ( MCC) under different stress loadings. Methods The third generation of MCC were cultured in 5% hypoxia. The uniaxial pressure of 0,1000,2000,4000 units strain ( ust) were exerted on the MCC for 2 h. Then the specimens were collected and dyed by trypan blue staining. The morphology and number of cells were observed under a light microscope, and the cell mortality was calculated. Westernblot and RT-PCR were performed to detect the changes of expression of HIF-2α and its down-stream catabolic factors (MMP3, MMP13, ADAMTs-4). Results With the increase of pressure, the cell mortality increased (P<0. 05). The expression of HIF-2αand its downstream catabolic factors (MMP3, MMP13, ADAMTs-4) increased with the increase of pressure (P<0. 05). Conclusion The activation of HIF-2αand its downstream catabolic factors by overload pressure may be respon-sible for the damage of condyle chondrocytes.

Abstract:Objective: Recent experimental evidence has suggested that pressure may play an important role in the pathogenesis of arthritic diseases such as temporomandibular disorders (TMDs), rheumatic diseases and osteoarthritis. This study examines the effects of hydrostatic pressure (HP) on cytoskeleton and protein production of bone morphogenetic protein-2 (BMP-2), transforming growth factor-beta (TGF-beta) and the SRY HMG box related gene 9 (SOX-9) in synovial fibroblasts (SFs) of rat temporomandibular joint (TMJ).Methods: SFs derived from rat TMJ were grown to confluence in Dulbecco's modified Eagle medium supplemented with 15% fetal calf serum. The monolayer of SFs was subjected to different HPs (0, 30, 60, and 90 kPa) by an in-house designed pressure chamber for 12 h. Changes of cell morphology were observed by fluorescent microscope. Production of TGF-P, BMP-2 and SOX-9 was examined by immunocytochemical assay and western blot.Results: Compared with the untreated control, the cellular actin configuration of SFs became elongated and more intense F-actin stress fiber staining was observed after HP loading. Exposure of SFs to HP for 12 h resulted in significant up-regulation of BMP-2 by 46, 54, and 66% at 30, 60, and 90 kPa, respectively, whilst TGF-P increased by 11, 19, and 28% at 30, 60, and 90 kPa, respectively. HIP also induced the increase of SOX-9 by 72% at 30 kPa and 83% at 60 kPa, but only 54% at 90 kPa.Conclusions: The obtained data suggest that HP induced the alteration of cytoskeleton and bone-morphogenetic-related proteins' production of SFs, which may influence the pathological condition of TMDs. Effect of Cydic Pressure on ERK1/2 in Rat Chondrocytes

Abstract:Objective To investigate the effect of cyclic pressure on extracellular signal-regulated kinases 1 and 2(ERK1,ERK2) in rat chondrocytes.Methods The rat chondrocytes were divided into four groups.Groups of B,C and D were mechanically stimulated with 150 kPa,0.1 Hz system for 0.5 h,1.0 h and 2.0 h,respectively.Group A was not treated with cyclic pressure as the control.The phosphorylation of ERK1 and ERK2(pERK1 and pERK2) was detected by immunofluorescence and Western blot.Results The immunofluorescence intensity of pERK1 and pERK2 increased after loading in groups of B,C and D,of which the increase of group C was the greatest.The expressions of pERK1 and pERK2 following cyclic pressure are higher in groups of B,C and D than those in group A(P0.01).Conclusion ERK1 and ERK2 are activated following cyclic pressure in rat chondrocytes,in which ERK1 and ERK2 may play an important role in machanical signal transduction of chondrocytes.

Abstract:There is no clear consensus regarding how cells respond to hydrostatic pressure. This is largely attributable to the high heterogeneity among cell types and the diverse custom-made devices used in previous studies. The aim of this work was to develop a facile device that could mimic various pressure environments and then delineate the cellular response to pressure stimulus. The device described here achieved both stable and periodic pressurization without oxygen deprivation. The biological utility of the device was assessed using human umbilical vein endothelial cells. We found more stereoscopic nuclear morphology and re-distribution of lamin A/C under high hydrostatic pressure compared to control cells. Mass spectrometry-based proteomics analysis showed significant changes in mitochondria-related pathways. Western blot analysis confirmed that high hydrostatic pressure induced a tendency toward mitochondrial fusion. Increased mitochondrial activity was observed as well. In conclusion, this device can be readily applied in biological research and extend our understanding of cellular mechano-sensation and the associated changes in mitochondrial behaviors.

Cell Senescence Reduced the Mechanotransduction Sensitivity of Porcine Angular Aqueous Plexus Cells to Elevation of Pressure

Abstract:PURPOSE:To investigate the effect of pressure/flow on the barrier function and protein expression of normal and senescent porcine aqueous humor plexus (AAP) cells, which are the porcine equivalent of human Schlemm's canal endothelial cells.METHODS:AAP cells were grown for 2 weeks in physiological (5% O2) or hyperoxic conditions (40% O2) to model cell senescence. Control and senescent AAP cells were subjected to control and elevated hydrostatic pressure gradient of 10 mm Hg for 72 hours. Hydraulic conductivity (HC) and transendothelial electric resistance (TEER) were measured. The expressions of senescence-associated β-galactosidase and DNA damage marker 8-hydroxy-2'-deoxyguanosine (8-OHdG) were monitored, and the protein expression profile was analyzed by Western blot.RESULTS:After 14 days of hyperoxia, AAP cells stained positive for 8-OHdG and β-galactosidase. Pressure elevation/flow resulted in significant increase of HC in control cells (from 1.37 ± 0.12 to 1.64 ± 0.18 μL/mm Hg/min/cm(2), P < 0.05), but not in senescent cells (1.15 ± 0.17 and 1.08 ± 0.10 μL μL/mm Hg/min/cm(2)). TEER changes were consistent with the HC results. Western blot analysis showed that the expression level of myosin light chain, claudin-5, and VE-cadherin significantly reduced under pressure elevation in control cells but not in senescent cells.CONCLUSIONS:AAP cells are mechano-sensitive; however, cell senescence rendered the cells less responsive to mechanical stimulus, which may have pathological consequences.
Effects of tensile strain and fluid flow on osteoarthritic human chondrocyte metabolism in vitro

Abstract:This study examined the hypothesis that tensile strain and fluid flow differentially influence osteoarthritic human chondrocyte metabolism. Primary high-density monolayer chondrocytes cultures were exposed to varying magnitudes of tensile strain and fluid-flow using a four-point bending system. Metabolic changes were quantified by real-time PCR measurement of aggrecan, IL-6, SOX-9, and type II collagen gene expression, and by determination of nitric oxide levels in the culture medium. A linear regression model was used to investigate the roles of strain, fluid flow, and their interaction on metabolic activity. Aggrecan, type II collagen, and SOX9 mRNA expression were negatively correlated to increases in applied strain and fluid flow. An effect of the strain on the induction of nitric oxide release and IL-6 gene expression varied by level of fluid flow (and visa versa). This interaction between strain and fluid flow was negative for nitric oxide and positive for IL-6. These results confirm that articular chondrocyte metabolism is responsive to tensile strain and fluid flow under in vitro loading conditions. Although the articular chondrocytes reacted to the mechanically applied stress, it was notable that there was a differential effect of tensile strain and fluid flow on anabolic and catabolic markers.

Abstract:The effect of glucocorticoid on the increase of cytosolic free calcium [Ca2+]i) induced by high-K+ was investigated in pheochomocytoma PC12 cells. The main result was as follows: (1) When the cells were preincubated at 37 degrees C for various time intervals in the presence of 10(-5) mol/L of corticosterone and stimulated with 55 mmol/L KCl, an inhibitory effect of corticosterone on delta [Ca2+]i was observed in a time-dependent manner with maximal effect at 5th min and no effect at 25th min. (2) When PC12 cells were preincubated with various concentration of corticosterone at 37 degrees C for 5 min, the inhibitory effect of corticosterone on delta [Ca2+]i induced by 55 mmol/L KCl was concentration-dependent. The effect was found to be maximal at 10(-5) mol/L of corticosterone and disappeared at 10(-9) mol/L of corticosterone. (3) Other steroids such as cortisol, dexamethasone, progesterone, testosterone, 17 beta-estradiol and aldosterone also inhibited the rise of [Ca2+]i evoked by 55 mmol/L KCl in PC12 cells to different extents, i.e., in the rank of P = F > Dex > T > Ald = E2. Cholesterol was ineffective at the concentration up to 10(-5) mol/L for inhibiting [Ca2+]i evoked by 55 mmol/L KCl. (4) Corticosterone could not inhibit the [Ca2+]i rise induced by changing the concentration of extracellular calcium from Ca(2+)-free to 3 mmol/L.

Abstract:Purpose: TGF-β plays important roles in proliferation, differentiation, and transition of fibroblasts. The objective of present study was to investigate whether the release of TGF-β was correlated to the magnitude of hydrostatic pressure (HP) in synovial fibroblasts (SFs) from rat temporomandibular joint, and explore its potential relationship with activation of MAP kinase pathway. Methods: SFs isolated from double condyles of rat temporomandibular joint were treated with hydrostatic pressures (HP) of 30, 60, and 90 kPa HP for 12 hours. The production of transforming growth factor-beta (TGF-β) in SFs suffered with different HP was determined by ELISA from 5 minutes to 12 hours in absence and presence of ERK1/2 inhibitor (PD98059). Furthermore, the activation of ERK1, JNK and p38 MAPKs in response to the treatment of hydrostatic pressure were examined by Western blot. Results: Compared with untreated control, the level of TGF-β dramatically increased in synovial with hydrostatic pressure of 30 kPa from 5 minutes to 4 hours, while co-treatment of PD98059 can attenuate the increase. The TGF-β production was slightly increased in synovial with higher pressure (60 and 90 kPa), while PD98059 can also reduce the increase. Hydrostatic pressure can significantly up-regulated the phosphorylation of ERK1/2 in SFs, whilst significant activations of JNK and p38 were not observed. Conclusions: TGF-β is generally thought to play an important role in bone development, remodeling and osteogenesis. According to our findings, SFs treated with PD98059 caused a decrease in TGF-β production stimulated by HP, indicating that the ERK1/2 pathway is essential for the enhancement of TGF-β production induced by HP.View Large Image Figure ViewerDownload Hi-res image Download (PPT)

Abstract:Osteoarthritis (OA) is the most common joint disease in older adults and is characterized by a gradual degradation of articular cartilage due to decreased cartilage matrix gene expression and increased expression of genes involved in protein degradation, apoptosis and inflammation. Due to the high water content of cartilage, one of the main physical stimuli sensed by chondrocytes is hydrostatic pressure. We previously showed that high pressure above 20 MPa induced gene expression changes in chondrocyte precursor cells similar to what is observed in OA. Micro-RNAs are small non-coding RNAs essential to many physiological and pathological process including OA. As the micro-RNA miR-155 has been found increased in OA chondrocytes, we investigated the effects of high pressure on the expression of the miR-155 host gene Mir155hg . The chondrocyte progenitor cell line ATDC5 was pressurized under hydrostatic pressure up to 25 MPa and the expression of Mir155hg or the resulting micro-RNAs were measured; pharmacological inhibitors were used to identify the signaling pathways involved in the regulation of Mir155hg . We found that Mir155hg is strongly and rapidly up-regulated by high, but not moderate, pressure in chondrocyte progenitor cells. This up-regulation likely involves the membrane channel pannexin-1 and several intracellular signaling molecules including PKC and Src. MiR-155-5p and -3p were also up-regulated by pressure though somewhat later than Mir155hg , and a set of known miR-155-5p target genes, including Ikbke , Smarca4 and Ywhae , was affected by pressure, suggesting that Mir155hg may have important roles in cartilage physiology.
