Abstract:Protocols in the academic life science laboratory are heavily reliant on the manual manipulation of tools, reagents and instruments by a host of research staff and students. In contrast to industrial and clinical laboratory environments, the usage of automation to augment or replace manual tasks is limited. Causes of this 'automation gap' are unique to academic research, with rigid short-term funding structures, high levels of protocol variability and a benevolent culture of investment in people over equipment. Automation, however, can bestow multiple benefits through improvements in reproducibility, researcher efficiency, clinical translation, and safety. Less immediately obvious are the accompanying limitations, including obsolescence and an inhibitory effect on the freedom to innovate. Growing the range of automation options suitable for research laboratories will require more flexible, modular and cheaper designs. Academic and commercial developers of automation will increasingly need to design with an environmental awareness and an understanding that large high-tech robotic solutions may not be appropriate for laboratories with constrained financial and spatial resources. To fully exploit the potential of laboratory automation, future generations of scientists will require both engineering and biology skills. Automation in the research laboratory is likely to be an increasingly critical component of future research programs and will continue the trend of combining engineering and science expertise together to answer novel research questions.

Abstract:Data science has revolutionized chemical research and continues to break down barriers with new interdisciplinary studies. The introduction of computational models and machine learning (ML) algorithms in combination with automation and traditional experimental techniques has enabled scientific advancement across nearly every discipline of chemistry, from materials discovery, to process optimization, to synthesis planning. However, predictive tools powered by data science are only as good as their data sets and, currently, many of the data sets used to train models suffer from several limitations, including being sparse, limited in scope and requiring human curation. Likewise, computational data faces limitations in terms of accurate modeling of nonideal systems and can suffer from low translation fidelity from simulation to real conditions. The lack of diverse data and the need to be able to test it experimentally reduces both the accuracy and scope of the predictive models derived from data science. This Account contextualizes the need for more complex and diverse experimental data and highlights how the seamless integration of robotics, machine learning, and data-rich monitoring techniques can be used to access it with minimal human labor. We propose three broad categories of data in chemistry: data on fundamental properties, data on reaction outcomes, and data on reaction mechanics. We highlight flexible, automated platforms that can be deployed to acquire and leverage these data. The first platform combines solid- and liquid-dosing modules with computer vision to automate solubility screening, thereby gathering fundamental data that are necessary for almost every experimental design. Using computer vision offers the additional benefit of creating a visual record, which can be referenced and used to further interrogate and gain insight on the data collected. The second platform iteratively tests reaction variables proposed by a ML algorithm in a closed-loop fashion. Experimental data related to reaction outcomes are fed back into the algorithm to drive the discovery and optimization of new materials and chemical processes. The third platform uses automated process analytical technology to gather real-time data related to reaction kinetics. This system allows the researcher to directly interrogate the reaction mechanisms in granular detail to determine exactly how and why a reaction proceeds, thereby enabling reaction optimization and deployment.

Abstract:Automation transformed various aspects of our human civilization, revolutionizing industries and streamlining processes. In the domain of scientific inquiry, automated approaches emerged as powerful tools, holding promise for accelerating discovery, enhancing reproducibility, and overcoming the traditional impediments to scientific progress. This article evaluates the scope of automation within scientific practice and assesses recent approaches. Furthermore, it discusses different perspectives to the following questions: Where do the greatest opportunities lie for automation in scientific practice?; What are the current bottlenecks of automating scientific practice?; and What are significant ethical and practical consequences of automating scientific practice? By discussing the motivations behind automated science, analyzing the hurdles encountered, and examining its implications, this article invites researchers, policymakers, and stakeholders to navigate the rapidly evolving frontier of automated scientific practice.

Abstract:The automated chemicals flow in organic electrochemistry will accelerate the scale-up process. In traditional flow electrochemistry, the reaction parameters (concentration, current density, flow rate, temperature, pressure) can be precisely adjusted. However, control over these parameters in the automated electrochemical flow system will be innovative. Multi- objective algorithms that link hardware and digital chemical operations on software can establish the autonomous system for flow electro-organic reactions. Electrochemical transformations are controlled by the application of electrical current, so the chemical process in an electrochemical microreactor can be organised and started or stopped via computer- controlled power supply. Such control over chemical transformations is not possible in conventional chemical reactions. Due to advantage of this unique feature of electrochemical process, new methods for acquisition of real-time reaction data are possible with the help of online analysis and self-optimising machine learning algorithms. Herein, we build our opinions and describe the perspective of automation in flow organic electrochemistry.

Autonomous discovery in the chemical sciences part I: Progress

Abstract:This two-part review examines how automation has contributed to different aspects of discovery in the chemical sciences. In this first part, we describe a classification for discoveries of physical matter (molecules, materials, devices), processes, and models and how they are unified as search problems. We then introduce a set of questions and considerations relevant to assessing the extent of autonomy. Finally, we describe many case studies of discoveries accelerated by or resulting from computer assistance and automation from the domains of synthetic chemistry, drug discovery, inorganic chemistry, and materials science. These illustrate how rapid advancements in hardware automation and machine learning continue to transform the nature of experimentation and modelling. Part two reflects on these case studies and identifies a set of open challenges for the field.

Abstract:Laboratories in chemistry, biochemistry, and materials science are at the leading edge of technology, discovering molecules and materials to unlock capabilities in energy, catalysis, biotechnology, sustainability, electronics, and more. Yet, most modern laboratories resemble factories from generations past, with a large reliance on humans manually performing synthesis and characterization tasks. Robotics and automation can enable scientific experiments to be conducted faster, more safely, more accurately, and with greater reproducibility, allowing scientists to tackle large societal problems in domains such as health and energy on a shorter timescale. We define five levels of laboratory automation, from laboratory assistance to full automation. We also introduce robotics research challenges that arise when increasing levels of automation and when increasing the generality of tasks within the laboratory. Robots are poised to transform science labs into automated factories of discovery that accelerate scientific progress.

Hence, there is a strong interest in automating the synthesis process, resulting in a growth in synthesis automation, especially of systems and configuration. Here, we systematically summarize recently developed automated systems for organic synthesis. This review will be useful for computational scientists aiming to develop novel tools and also for non-specialists and students to understand the frontier of automated synthesis.</p>

In addition, autonomous robotic systems are rapidly accelerating the pace of discovery by performing tedious tasks with unprecedented speed and precision. Abstract:Medicinal chemistry plays a fundamental and underlying role in chemical biology, pharmacology, and medicine to discover safe and efficacious drugs. Small molecule medicinal chemistry relies on iterative learning cycles composed of compound design, synthesis, testing, and data analysis to provide new chemical probes and lead compounds for novel and druggable targets. Using traditional approaches, the time from hypothesis to obtaining the results can be protracted, thus limiting the number of compounds that can be advanced into clinical studies. This challenge can be tackled with the recourse of enabling technologies that are showing great potential in improving the drug discovery process. In this Perspective, we highlight recent developments toward innovative medicinal chemistry strategies based on continuous flow systems coupled with automation and bioassays. After a discussion of the aims and concepts, we describe equipment and representative examples of automated flow systems and end-to-end prototypes realized to expedite medicinal chemistry discovery cycles.

Abstract:This paper presents the first overview of recent developments in techniques and methods that enable closed-loop optimization, also sometimes called ‘self optimization’, as well as discovery in different areas of molecular sciences. The closed-loop experimental platforms offer tremendous new opportunities by significantly increasing productivity, as well as enabling completely new types of experiments to be performed. Such experiments involve three main enabling technology areas: automated experimental systems, analytical instruments connected to automated chemoinformatics software and optimization or decision-making algorithms. We review the most exciting developments concerning robotic experiments, 3D printed lab-ware, experimental systems with multiple analytical instruments and advanced optimization algorithms based on machine learning approaches. A range of different chemical problems is described, which show the breadth of potential applications of this emerging experimental approach.

Abstract:Recent developments in automated flow chemistry for pharmaceutical compound synthesis have garnered significant attention. Automation in synthesis represents a cutting-edge frontier in the field of chemistry, offering highly efficient, rapid, and reproducible synthetic methods that significantly shorten reaction time and reduce costs. In the realm of pharmaceutical compound synthesis, automated flow chemistry demonstrates unique importance. By utilizing flow chemistry, reactions can be performed under continuous flow conditions, enabling precise reaction control, higher yields, and increased product purity. Additionally, automated flow synthesis overcomes several challenges encountered in traditional batch synthesis, such as decreased generation of chemical waste, optimization of reaction conditions, and enhanced operational safety. This review highlights the recent developments in automated flow synthesis of various pharmaceutical compounds, including large biopharmaceutical molecules, small organic drug molecules, and carbohydrates. It covers automated iterative synthesis and the use of machine learning to enhance synthesis efficiency. Furthermore, it explores the practical application of high-throughput synthesis and screening technologies. Finally, the review offers concise perspectives on potential future developments in the field.

Abstract:Machines can revolutionize the field of chemistry and material science, driving the development of new chemistries, increasing productivity, and facilitating reaction scale up. The incorporation of automated systems in the field of polymer chemistry has however proven challenging owing to the demanding reaction conditions, rendering the automation setup complex and costly. There is an imminent need for an automation platform which uses fast and simple polymerization protocols, while providing a high level of control on the structure of macromolecules via precision synthesis. In this work, we combine an oxygen tolerant, room temperature polymerization method with a simple liquid handling robot to automatically prepare precise and high order multiblock copolymers with unprecedented livingness even after many chain extensions. We report the highest number of blocks synthesized in such a system, demonstrating the capabilities of this automated platform for the rapid synthesis and complex polymer structure formation.

Abstract:Aspen HYSYS is a typical tool used in some Master in Chemical Engineering courses at the University of Castilla‐La Mancha like "Analysis and Optimization of Chemical Processes" and "Dynamic of Process: Regulation of Chemical Plants." Automation is the process of linking commercial software to third‐parties applications built in EXCEL‐Visual Basic for Applications. The capability of automation as a powerful tool for simulating unprecedented complex processes suggests that it could be a relevant background complement to the classical one offered by a university. In this sense, a seminar about automation based on the study of two cases: a refrigeration process and a production one taken from literature, has been proposed at the University of Castilla‐La Mancha. The analysis of the survey performed for the subject evaluation resulted to be very positive. Students consider the methodology of the course and the potentiality of automation for developing their research and professional skills appropriately. However, more examples are required to better understand the automation concept and its potential application to other situations. Finally, they declare that the concept of automation requires a lot of expertise and deep knowledge of programming and the correct application of numerical methods for solving complex problems.

Digitising chemical synthesis in automated and robotic flow

Abstract:Continuous flow chemical synthesis is already known to have many attributes that give it superiority over batch processes in several respects. To expand these advantages with those from automation will only drive such enabling technologies further into the faster producing, more efficient 21st century chemical world. In this report we present several examples of algorithmic chemical search, along with flow platforms that link hardware and digital chemical operations on software. This enables organic syntheses to be automatically carried out and optimised with as little human intervention as possible. By applying such enabling technologies to the production of small organic molecules and pharmaceutical compounds in end-to-end multistep processes, a range of reaction types can be accessed and, thus, the flexibility of these single, compact flow designs may be revealed. Automated systems can allow several reactions to take place on the same setup, enabling direct comparison of reactions under different conditions. Moreover, the production of new and known target compounds can be made faster and more efficient, the recipes of which can then be stored as digital files. Some of the automating software has employed machine-powered learning to assist the chemist in developing intelligent algorithms and artificial intelligence (AI) driven synthetic route planning. This ultimately produces a continuous flow platform that can design its own viable pathway to a particular molecule and then carry it out on its own, allowing the chemists, at the same time, to apply their expertise to other pressing challenges in their fields.

Abstract:Automated laboratory experiments have the potential to propel new discoveries, while increasing reproducibility and improving scientists' safety when handling dangerous materials. However, many automated laboratory workflows have not fully leveraged the remarkable advancements in robotics and digital lab equipment. As a result, most robotic systems used in the labs are programmed specifically for a single experiment, often relying on proprietary architectures or using unconventional hardware. In this work, we tackle this problem by proposing a novel robotic system architecture specifically designed with and for chemists, which allows the scientist to easily reconfigure their setup for new experiments. Specifically, the system's strength is its ability to combine together heterogeneous robotic platforms with standard laboratory equipment to create different experimental setups. Finally, we show how the architecture can be used for specific laboratory experiments through case studies such as solubility screening and crystallisation.
