Abstract:<p>Organic synthesis is a vital process that is a mainstay of drug discovery. However, traditional manual-based approaches to organic synthesis might not be economical, especially in a research environment where budgets are increasingly restricted and the effective use of manpower and materials is crucial. Hence, there is a strong interest in automating the synthesis process, resulting in a growth in synthesis automation, especially of systems and configuration. Here, we systematically summarize recently developed automated systems for organic synthesis. This review will be useful for computational scientists aiming to develop novel tools and also for non-specialists and students to understand the frontier of automated synthesis.</p>

Abstract:Continuous flow chemical synthesis is already known to have many attributes that give it superiority over batch processes in several respects. To expand these advantages with those from automation will only drive such enabling technologies further into the faster producing, more efficient 21st century chemical world. In this report we present several examples of algorithmic chemical search, along with flow platforms that link hardware and digital chemical operations on software. This enables organic syntheses to be automatically carried out and optimised with as little human intervention as possible. By applying such enabling technologies to the production of small organic molecules and pharmaceutical compounds in end-to-end multistep processes, a range of reaction types can be accessed and, thus, the flexibility of these single, compact flow designs may be revealed. Automated systems can allow several reactions to take place on the same setup, enabling direct comparison of reactions under different conditions. Moreover, the production of new and known target compounds can be made faster and more efficient, the recipes of which can then be stored as digital files. Some of the automating software has employed machine-powered learning to assist the chemist in developing intelligent algorithms and artificial intelligence (AI) driven synthetic route planning. This ultimately produces a continuous flow platform that can design its own viable pathway to a particular molecule and then carry it out on its own, allowing the chemists, at the same time, to apply their expertise to other pressing challenges in their fields.

Synthesis of Many Different Types of Organic Small Molecules Using One Automated Process.

Abstract:Small-molecule synthesis usually relies on procedures that are highly customized for each target. A broadly applicable automated process could greatly increase the accessibility of this class of compounds to enable investigations of their practical potential. Here we report the synthesis of 14 distinct classes of small molecules using the same fully automated process. This was achieved by strategically expanding the scope of a building block-based synthesis platform to include even C(sp3)-rich polycyclic natural product frameworks and discovering a catch-and-release chromatographic purification protocol applicable to all of the corresponding intermediates. With thousands of compatible building blocks already commercially available, many small molecules are now accessible with this platform. More broadly, these findings illuminate an actionable roadmap to a more general and automated approach for small-molecule synthesis.

Abstract:Automated chemical synthesis carries great promises of safety, efficiency and reproducibility for both research and industry laboratories. Current approaches are based on specifically-designed automation systems, which present two major drawbacks: (i) existing apparatus must be modified to be integrated into the automation systems; (ii) such systems are not flexible and would require substantial re-design to handle new reactions or procedures. In this paper, we propose a system based on a robot arm which, by mimicking the motions of human chemists, is able to perform complex chemical reactions without any modifications to the existing setup used by humans. The system is capable of precise liquid handling, mixing, filtering, and is flexible: new skills and procedures could be added with minimum effort. We show that the robot is able to perform a Michael reaction, reaching a yield of 34%, which is comparable to that obtained by a junior chemist (undergraduate student in Chemistry).

Abstract:The new era with prosperous artificial intelligence (AI) and robotics technology is reshaping the materials discovery process in a more radical fashion. Here we present authentic intelligent robotics for chemistry (AIR-Chem), integrated with technological innovations in the AI and robotics fields, functionalized with modules including gradient descent-based optimization frameworks, multiple external field modulations, a real-time computer vision (CV) system, and automated guided vehicle (AGV) parts. AIR-Chem is portable and remotely controllable by cloud computing. AIR-Chem can learn the parametric procedures for given targets and carry on laboratory operations in standalone mode, with high reproducibility, precision, and availability for knowledge regeneration. Moreover, an improved nucleation theory of size focusing on inorganic perovskite quantum dots (IPQDs) is theoretically proposed and experimentally testified to by AIR-Chem. This work aims to boost the process of an unmanned chemistry laboratory from the synthesis of chemical materials to the analysis of physical chemical properties, and it provides a vivid demonstration for future chemistry reshaped by AI and robotics technology.

Mehr et al. report a software platform that uses natural language processing to translate the organic chemistry literature directly into editable code, which in turn can be compiled to drive automated synthesis of the compound in the laboratory. Abstract:Pairing prediction and robotic synthesis Progress in automated synthesis of organic compounds has been proceeding along parallel tracks. One goal is algorithmic prediction of viable routes to a desired compound; the other is implementation of a known reaction sequence on a platform that needs little to no human intervention. Coley et al. now report preliminary integration of these two protocols. They paired a retrosynthesis prediction algorithm with a robotically reconfigurable flow apparatus. Human intervention was still required to supplement the predictor with practical considerations such as solvent choice and precise stoichiometry, although predictions should improve as accessible data accumulate for training. Abstract:Automation and high-throughput techniques provide a solid technical foundation for realizing the deep fusion of artificial intelligence and chemistry as well as the full utilization of their advantages. In recent years, with the unique advantages of low consumption, low risk, high efficiency, high reproducibility, high flexibility and good versatility, intelligent automated platforms for high-throughput chemical synthesis aroused widespread concerns of synthetic chemists. In this review, the automated high-throughput chemical synthesis, automated high-throughput sample treatment and characterization technique, as well as the application of artificial intelligence technique in chemical synthesis are introduced. The characteristics of the systems and platforms based on these techniques, including the iChemFoundry platform developed in the ZJU-Hangzhou Global Scientific and Technological Innovation Center, are introduced. The intelligent automated platforms for high-throughput chemical synthesis will reshape the thinking mode of traditional disciplines, promote the innovation of disruptive techniques, redefine the rate of chemical synthesis, and innovate the way of material manufacturing.

Abstract:Data science has revolutionized chemical research and continues to break down barriers with new interdisciplinary studies. The introduction of computational models and machine learning (ML) algorithms in combination with automation and traditional experimental techniques has enabled scientific advancement across nearly every discipline of chemistry, from materials discovery, to process optimization, to synthesis planning. However, predictive tools powered by data science are only as good as their data sets and, currently, many of the data sets used to train models suffer from several limitations, including being sparse, limited in scope and requiring human curation. Likewise, computational data faces limitations in terms of accurate modeling of nonideal systems and can suffer from low translation fidelity from simulation to real conditions. The lack of diverse data and the need to be able to test it experimentally reduces both the accuracy and scope of the predictive models derived from data science. This Account contextualizes the need for more complex and diverse experimental data and highlights how the seamless integration of robotics, machine learning, and data-rich monitoring techniques can be used to access it with minimal human labor. We propose three broad categories of data in chemistry: data on fundamental properties, data on reaction outcomes, and data on reaction mechanics. We highlight flexible, automated platforms that can be deployed to acquire and leverage these data. The first platform combines solid- and liquid-dosing modules with computer vision to automate solubility screening, thereby gathering fundamental data that are necessary for almost every experimental design. Using computer vision offers the additional benefit of creating a visual record, which can be referenced and used to further interrogate and gain insight on the data collected. The second platform iteratively tests reaction variables proposed by a ML algorithm in a closed-loop fashion. Experimental data related to reaction outcomes are fed back into the algorithm to drive the discovery and optimization of new materials and chemical processes. The third platform uses automated process analytical technology to gather real-time data related to reaction kinetics. This system allows the researcher to directly interrogate the reaction mechanisms in granular detail to determine exactly how and why a reaction proceeds, thereby enabling reaction optimization and deployment.

Abstract:Chemical synthesis always involves laborious and time-consuming operations. Autonomous chemical synthesis systems can liberate experimentalists from those routine tasks, but the design of automatic chemical reaction machines is of great challenge. We describe herein the development of a real-life automatic chemical operation platform by using a coordinated multi-robot system with artificial intelligence and machine learning methods. The capacities of this platform were demonstrated by its application in condensation reaction of 2,4-dinitrophenyldrazine with formaldehyde, and automatic catalyst evaluation of a heterogeneous aza Diels-Alder reaction, suggesting the designed machine is applicable to both homogenous and heterogeneous chemical reactions. Besides, the excellent repeatability and reproducibility of robotic tests on these two model reactions indicate the great promise of massive transfer of manual to automatic operation.

Abstract:Autonomous laboratories can accelerate discoveries in chemical synthesis, but this requires automated measurements coupled with reliable decision-making 1,2 . Most autonomous laboratories involve bespoke automated equipment 3,4,5,6 , and reaction outcomes are often assessed using a single, hard-wired characterization technique 7 . Any decision-making algorithms 8 must then operate using this narrow range of characterization data 9,10 . By contrast, manual experiments tend to draw on a wider range of instruments to characterize reaction products, and decisions are rarely taken based on one measurement alone. Here we show that a synthesis laboratory can be integrated into an autonomous laboratory by using mobile robots 11,12,13 that operate equipment and make decisions in a human-like way. Our modular workflow combines mobile robots, an automated synthesis platform, a liquid chromatography–mass spectrometer and a benchtop nuclear magnetic resonance spectrometer. This allows robots to share existing laboratory equipment with human researchers without monopolizing it or requiring extensive redesign. A heuristic decision-maker processes the orthogonal measurement data, selecting successful reactions to take forward and automatically checking the reproducibility of any screening hits. We exemplify this approach in the three areas of structural diversification chemistry, supramolecular host–guest chemistry and photochemical synthesis. This strategy is particularly suited to exploratory chemistry that can yield multiple potential products, as for supramolecular assemblies, where we also extend the method to an autonomous function assay by evaluating host–guest binding properties.

Abstract:The automated chemicals flow in organic electrochemistry will accelerate the scale-up process. In traditional flow electrochemistry, the reaction parameters (concentration, current density, flow rate, temperature, pressure) can be precisely adjusted. However, control over these parameters in the automated electrochemical flow system will be innovative. Multi- objective algorithms that link hardware and digital chemical operations on software can establish the autonomous system for flow electro-organic reactions. Electrochemical transformations are controlled by the application of electrical current, so the chemical process in an electrochemical microreactor can be organised and started or stopped via computer- controlled power supply. Such control over chemical transformations is not possible in conventional chemical reactions. Due to advantage of this unique feature of electrochemical process, new methods for acquisition of real-time reaction data are possible with the help of online analysis and self-optimising machine learning algorithms. Herein, we build our opinions and describe the perspective of automation in flow organic electrochemistry.

Abstract:Clear directions for a robotic platform The chemistry literature contains more than a century's worth of instructions for making molecules, all written by and for humans. Steiner et al. developed an autonomous compiler and robotic laboratory platform to synthesize organic compounds on the basis of standardized methods descriptions (see the Perspective by Milo). The platform comprises conventional equipment such as round-bottom flasks, separatory funnels, and a rotary evaporator to maximize its compatibility with extant literature. Abstract:Recent developments in automated flow chemistry for pharmaceutical compound synthesis have garnered significant attention. Automation in synthesis represents a cutting-edge frontier in the field of chemistry, offering highly efficient, rapid, and reproducible synthetic methods that significantly shorten reaction time and reduce costs. In the realm of pharmaceutical compound synthesis, automated flow chemistry demonstrates unique importance. By utilizing flow chemistry, reactions can be performed under continuous flow conditions, enabling precise reaction control, higher yields, and increased product purity. Additionally, automated flow synthesis overcomes several challenges encountered in traditional batch synthesis, such as decreased generation of chemical waste, optimization of reaction conditions, and enhanced operational safety. This review highlights the recent developments in automated flow synthesis of various pharmaceutical compounds, including large biopharmaceutical molecules, small organic drug molecules, and carbohydrates. It covers automated iterative synthesis and the use of machine learning to enhance synthesis efficiency. Furthermore, it explores the practical application of high-throughput synthesis and screening technologies. Finally, the review offers concise perspectives on potential future developments in the field.

Abstract:Robotic platforms for chemistry are developing rapidly but most systems are not currently able to adapt to changing circumstances in real-time. We present a dynamically programmable system capable of making, optimizing, and discovering new molecules which utilizes seven sensors that continuously monitor the reaction. By developing a dynamic programming language, we demonstrate the 10-fold scale-up of a highly exothermic oxidation reaction, end point detection, as well as detecting critical hardware failures. We also show how the use of in-line spectroscopy such as HPLC, Raman, and NMR can be used for closed-loop optimization of reactions, exemplified using Van Leusen oxazole synthesis, a four-component Ugi condensation and manganese-catalysed epoxidation reactions, as well as two previously unreported reactions, discovered from a selected chemical space, providing up to 50% yield improvement over 25–50 iterations. Finally, we demonstrate an experimental pipeline to explore a trifluoromethylations reaction space, that discovers new molecules.

Abstract:Synthetic chemistry has played a vital role in miscellaneous fields of human civilization over the past century. The synthetic stage yet remains time-consuming and labor-intensive. To overcome these limitations, automation has been introduced to transform synthetic chemistry, leading to the development of high-throughput methods for molecular discovery. Automated flow chemical synthesis (AFCS) has recently emerged as a promising candidate, offering improved efficiency, scalability, and sustainability over the well-known automated solid-phase peptide synthesis. To further advance AFCS, elements like artificial intelligence-based computer-aided structure design and synthesis planning, autonomously assembled compatible synthesis with enhanced automated process control, and autonomous optimization can be considered. This review focuses on recent advances in computer-aided automated flow chemical synthesis (CAAFCS) of polymers in living polymerization and iterative synthesis strategy. The current challenges and outlook are finally discussed for developing more powerful CAAFCS systems and expanding their applicability across numerous fields, potentially providing brand-new perspectives and guidelines for future developments in this field.

Abstract:Modularized synthesis of small organic molecules is transforming our capacity to create medicines and materials. Disruptive acceleration of this molecule building strategy will broadly unlock its functional potential and requires integration of many new assembly chemistries. Recent advances in high-throughput chemistry stand to enable selection of appropriate chemical reaction conditions from the vast range of potential options. However, a disconnect between the rates of exploration and evaluation has limited progress. Here we report how intrinsic fragmentation features of chemical building blocks generalizes their analysis to yield sub-second readouts of reaction outcomes. Central to this advance was identifying that groups typically attached to boron, nitrogen, and oxygen atoms fragment in a specific and selective manner by mass spectrometry, enabling target agnostic analysis. Combining these features with acoustic droplet ejection mass spectrometry we could eliminate slow chromatographic steps and continuously evaluate chemical reaction outcomes in multiplexed formats. This allowed rapid assignment of reaction conditions to molecules derived from ultra-high throughput chemical synthesis experiments.
