Abstract:In this paper we describe the strategy used in the development and validation of a near infrared spectroscopy method for the rapid determination of baicalin, chlorogenic acid, ursodeoxycholic acid (UDCA), chenodeoxycholic acid (CDCA), and the total solid contents (TSCs) in the Tanreqing injection. To increase the representativeness of calibration sample set, a concentrating-diluting method was adopted to artificially prepare samples. Partial least square regression (PLSR) was used to establish calibration models, with which the five quality indicators can be determined with satisfied accuracy and repeatability. In addition, the slope/bias (S/B) method was used for the models transfer between two different types of NIR instruments from the same manufacturer, which is contributing to enlarge the application range of the established models. With the presented method, a great deal of time, effort and money can be saved when large amounts of Tanreqing injection samples need to be analyzed in a relatively short period of time, which is of great significance to the traditional Chinese medicine (TCM) industries.

Abstract:The existing calibration algorithms for Raman spectral quantitative analysis,such as principal component regression,partial least squares and support vector machine need a large number of training samples and the regression model may have unsatisfactory generalization ability.Indirect hard modeling(IHM)is a recently introduced method for spectral quantitative analysis,which is used to analyze spectra with highly overlapping band and nonlinear variation.This method can be used to build a calibration model with high generalization ability only by a few training samples.However,all the spectra of pure-components in the mixture is required in IHM method.Obviously,this condition is difficult to achieve in practical applications.To overcome the limitation of IHM,a novel method based on Voigt function for spectral quantitative analysis,i.e.,direct hard modeling(DHM),was presented.The novel method did not require the pure-component spectra.By DHM algorithm,the characteristic peaks of every pure component in the spectra of mixture could be fixed,and then a linear calibration model could be built based on the areas of unique peaks and the concentration of pure-components.The DHM algorithm was successfully demonstrated while testing on the quantitative analysis of xylene mixture in a PX unit.Experimental results showed that the DHM algorithm had the advantages of robustness and fewer training samples.

Abstract:A method for rapid quantitative analysis of four kinds of Tanreqing injection intermediates was developed based on Fourier transform near infrared (FT-NIR) spectroscopy and partial least squares (PLS) algorithm. The NIR spectra of 120 samples were collected in transflective mode. The concentrations of chlorogenic acid, caffeic acid, luteoloside, baicalin, ursodesoxycholic acid (UDCA), and chenodeoxycholic acid (CDCA) were determined with the HPLC–DAD/ELSD as reference method. In the PLS calibration, the NIR spectra were pretreated with different methods and the number of PLS factors used in the model calibration was optimized by leave-one-out cross-validation. The performance of the final PLS models was evaluated according to the root mean square error of calibration (RMSEC), root mean square error of cross-validation (RMSECV), root mean square error of prediction (RMSEP), BIAS, standard error of prediction (SEP), and correlation coefficients (R). The R values in the prediction sets were all higher than 0.93, and the SEPs for the 6 compounds are 1.18, 6.02, 2.71, 155, 126, 30.0mg/l, respectively. The established models were used for the liquid preparation process analysis of Tanreqing injection in three batches, and a model updating method was proposed for the long-term usage of the established models. This work demonstrated that NIR spectroscopy is more rapid and convenient than the conventional methods to analyze the intermediates of Tanreqing injection, and the presented method is helpful to the implementation of process analytical technology (PAT) in pharmaceutical industry of Chinese Medicines Injections.

Abstract:Two different multivariate techniques have been applied for the quantitative analysis of caffeine, codeine, paracetamol and p-aminophenol (PAP) in quaternary mixture, namely, Partial Least Squares (PLS-1) and Artificial Neural Networks (ANN). For suitable analysis, a calibration set of 25 mixtures with various ratios of the drugs and PAP impurity were established using a 4-factor 5-level experimental design. The most meaningful wavelengths for the chemometric models were chosen using Genetic Algorithm (GA) as a variable selection technique. By using an independent validation set, the validity of the proposed methods was evaluated. A comparative study was established between the three multivariate models (PLS-1, GA–PLS and GA–ANN). The comparison between the various models revealed that the GA–ANN model was superior at resolving the highly overlapped spectra of this quaternary combination. The drugs were successfully quantified in their pharmaceutical dosage form utilizing the GA–ANN models.

Designing a calibration set in spectral space for efficient development of an NIR method for tablet analysis

Abstract:Designing a calibration set is the first step in developing a multivariate spectroscopic calibration method for quantitative analysis of pharmaceutical tablets. This step is critical because successful model development depends on the suitability of the calibration data. For spectroscopic-based methods, traditional concentration based techniques for designing calibration sets are prone to have redundant information while simultaneously lacking necessary information for a successful calibration model. A method for designing a calibration set in spectral space was developed. The pure component spectra of a tablet formulation were used to define the spectral space of that formulation. This method maximizes the information content of measurements and minimizes sample requirements to provide an efficient means for developing multivariate spectroscopic calibration. A comparative study was conducted between a commonly employed full factorial approach to calibration development and the newly developed technique. The comparison was based on a system to quantify a model drug, acetaminophen, in pharmaceutical compacts using near infrared spectroscopy. A 2-factor full factorial design (acetaminophen with 5 levels and MCC:Lactose with 3 levels) was used for calibration development. Three replicates at each design point resulted in a total of 45 tablets for the calibration set. Using the newly developed spectral based method, 11 tablets were prepared for the calibration set. Partial least square (PLS) models were developed from respective calibration sets. Model performance was comprehensively assessed based on the ability to predict acetaminophen concentrations in multiple prediction sets. One prediction set contained similar information to calibration set while the other prediction sets contained different information from calibration set in order to assess the model accuracy and robustness. Similar prediction performance was achieved using the 11-tablet design (spectral space), compared to the 45-tablet full factorial approach. This work demonstrates that a calibration set designed in spectral space provided an efficient means of developing spectroscopic multivariate calibration.

Abstract:A multivariate spectrophotometric method is a potential approach that enables discrimination of spectra of components in complex matrices (e.g., pharmaceutical formulation) serving as a substitution method for chromatography. Four green smart multivariate spectrophotometric models were proposed and validated, including principal component regression (PCR), partial least-squares (PLS), multivariate curve resolution-alternating least squares (MCR-ALS), and artificial neural networks (ANN). The developed chemometric models were compared to resolve highly overlapping spectra of Paracetamol (PARA), Chlorpheniramine maleate (CPM), Caffeine (CAF), and Ascorbic acid (ASC). The four multivariate calibration models were assessed via recoveries percent, and root mean square error of prediction. Hence, the proposed models were efficiently applied with no need for any preliminary separation step. The models were utilized to analyze the studied components in their combined pharmaceutical formulation (Grippostad® C capsules). Analytical GREEnness Metric Approach (AGREE) and eco-scale tools were applied to assess the greenness of the established models and found to be 0.77 and 85, respectively. Moreover, the proposed models have been compared to official ones showing no considerable variations in accuracy and precision. Therefore, these models can be highly advantageous for conducting standard pharmaceutical analysis of the substances researched within product testing laboratories.

Abstract:The quality of over-the-counter (OTC) pain relievers is important to ensure the safety of the marketed products in order to maintain the overall health care of patients. In this study, the multivariate curve resolution-alternating least squares (MCR-ALS) chemometric method was developed and validated for the resolution and quantification of the most commonly consumed OTC pain relievers (acetaminophen, acetylsalicylic acid, ibuprofen, naproxen, and caffeine) in commercial drug formulations. The analytical performance of the developed chemometric methods such as root mean square error of prediction, bias, standard error of prediction, relative error of prediction, and coefficients of determination was calculated for the developed model. The obtained results are linear with concentration in the range of 0.5-7 μg/mL for acetaminophen and 0.5-3.5 and 0.5-3 μg/mL for naproxen and caffeine, respectively, while the linearity ranges for acetyl salicylic acid and ibuprofen were 1-15 μg/mL. High values of coefficients of determination ≥0.9995 reflected high predictive ability of the developed model. Good recoveries ranging from 98.0% to 99.7% were obtained for all analytes with relative standard deviations (RSDs) not higher than 1.62%. The optimized method was successfully applied for the analysis of the studied drugs either in their single or coformulated pharmaceutical products without any separation step. The optimized method was also compared with a reported HPLC method using paired t-test and F-ratio at 95% confidence level, and the results showed no significant difference regarding accuracy and precision. The developed method is eco-friendly, simple, fast, and amenable for routine analysis. It could be used as a cost-effective alternative to chromatographic techniques for the analysis of the studied drugs in commercial formulations.

Three components, namely paracetamol, chlorzoxazone and ibuprofen were present in tablet dosage form. The linearity studies were carried out by recording zero order spectra and measuring absorbances at 285.0, 282.0 and 220.0 nm for paracetamol, chlorzoxazone and ibuprofen respectively. The drugs exhibited linear response in the concentration range of 6.0 – 18.0, 3.0 – 15.0 and 4.0 – 20.0 μg / mL for paracetamol, chlorzoxazone and ibuprofen respectively. The spectrum of paracetamol was most extended which was subsequently followed by moderately extended spectrum of chlorzoxazone and unextended spectrum of ibuprofen. The ratio spectra were manipulated successfully for quantification of paracetamol, chlorzoxazone, and ibuprofen. The developed method was validated as per ICH guidelines for the parameters of specificity, linearity, precision and accuracy. The percent recoveries for all the three drugs were in the range of 98.0 – 102.0 % with mean recovery of paracetamol, chlorzoxazone and ibuprofen were 99.72, 99.02 and 100.34 % respectively. Additionally the validity of the method is assured by analyzing marketed formulation.

Abstract:Mixtures of acetylsalicylic acid (ASA), paracetamol (PAR), and caffeine (CAF) have been successfully analyzed by constrained multivariate curve resolution-alternating least squares (MCR-ALS). The MCR-ALS methodology adequately exploits the second-order advantage which enables quantitation of analyte in the presence of unknown and uncalibrated interferences. The procedure simultaneously takes into account the spectroscopic and pH-dependent properties of the compounds, which leads to a higher selectivity. Specially, for CAF determination fully protonated or deprotonated forms of CAF are not dominant in the pH range of data acquisition but spectral changes with pH were recorded and used for accurate determination of CAF. Furthermore, quantitative determination of an analyte in a complex mixture is performed using a synthetic solution as standard containing only the analyte of interest. Even in the presence of the rank deficiencies, in most cases accurate quantitation with relative errors in prediction lower than 5 % was obtained. The procedure was successfully applied to the analysis of real samples (pharmaceuticals) using synthetic external standards. Percent relative errors of 4.03, 3.26, and 5.85 were obtained for ASA, PAR, and CAF, respectively, in A.C.A tablets and 4.49 and 2.75 for PAR and CAF, respectively in Novafen capsules.

Abstract:In this manuscript, the effectiveness of multivariate and univariate tools in conjunction with spectrophotometric techniques was evaluated for the concurrent analysis of ciprofloxacin (CI) and ornidazole (OR) in prepared mixtures, tablets, and human serum. The artificial neural network was chosen as the multivariate Technique. Bayesian regularization (trainbr) and Levenberg–Marquardt algorithms (trainlm), were constructed and trained using feed-forward back-propagation learning. The optimal logarithm was determined based on mean recovery, mean square error of prediction (MSEP), relative root mean square error of prediction (RRMSEP), and bias-corrected MSEP (BCMSEP) scores. Trainbr outperformed trainlm, yielding a mean recovery of 100.05% for CI and 99.84% for OR, making it the preferred algorithm. Fourier self-deconvolution and mean-centering transforms were chosen as the univariate Techniques. Fourier self-deconvolution was applied to the zero-order spectra of ciprofloxacin and ornidazole by electing an appropriate full width at half maximum, enhancing peak resolution at 380.1 nm and 314.2 nm for CI and OR, respectively. Mean centering transform was applied to CI and OR ratio spectra to eliminate constant signals, enabling accurate quantification of CI and OR at 272.0 nm and 306.2 nm, respectively. The introduced approaches were optimized and validated for precise CI and OR analysis, with statistical comparison against the HPLC method revealing no notable differences. The sustainability of these approaches was confirmed through the green certificate (modified eco-scale), AGP, and whiteness-evaluation tool, corroborating their ecological viability.

Abstract:Two accurate, precise and robust multivariate chemometric methods were developed for the simultaneous determination of montelukast sodium (MON), rupatadine fumarate (RUP) and desloratadine (DES). These methods provide a cost-effective alternative to chromatographic techniques by utilizing spectrophotometry in pharmaceutical quality control. The proposed approaches, partial least squares-1 (PLS-1) and artificial neural network (ANN), were optimized using genetic algorithm (GA) to select the most influential wavelengths, enhancing model performance. A five-level, three-factor design was employed to construct a calibration set with 25 mixtures, utilizing concentration ranges of 3–19, 5–25, and 4–20 μg.mL −1 for MON, RUP, and DES, respectively. An independent validation set was employed to assess the performance of the models. GA significantly improved the PLS-1 and ANN models for RUP and DES, though minimal enhancement was observed for MON. These methods were successfully applied to the simultaneous quantification of the compounds in pharmaceutical formulations and proved useful as stability-indicating assays for RUP, given that DES is a known degradation product. The developed methods offer a valuable tool for impurity profiling and quality control in pharmaceutical analysis.

Abstract:Mathematical manipulation technique has proven to be a very powerful tool for efficient processing and handling of highly overlapped spectra. This work introduced two green and sustainable approaches for the successful recovery of the ternary mixture, Tylosin tartarate (TYL), Sulfadimdine (SLD), and Trimethoprim (TRI). The approaches were constructed using three different methods, derivative ratio spectrum zero-crossing method (DRSZ), double divisor ratio spectra derivative method (DDRD), and factorized derivative ratio method coupled with spectrum subtraction (FDRM-SS). The two approaches succeeded in recovering the three drugs (Linearity range achieved were 0.5–5 μg/mL for TYL, 0.3–1.3 μg/mL for SLD, and 0.3–5 μg/mL for TRI), giving convenient standard deviations and satisfactory recovery percentages. The recommended methods have been verified in accordance with (ICH) guidelines. When the results were statistically compared to the official methods, no significant difference was found. Both AGREE—Analytical GREEnness Metric Approach and Software, and White Analytical Chemistry (WAC) RGB model gave scores of 0.93 and 97.2%, respectively, which proved that the approaches were eco-friendly and abiding by the sustainability principles.

Abstract:The purpose of our research was to investigate efficient procedures for generating multivariate prediction vectors for quantitative chemical analysis of solid dosage forms using terahertz pulse imaging (TPI) reflection spectroscopy. A set of calibration development and validation tablet samples was created following a ternary mixture of anhydrous theophylline, lactose monohydrate, and microcrystalline cellulose (MCC). Spectral images of one side of each tablet were acquired over the range of 8 cm−1 to 60 cm−1. Calibration models were generated by partial least-squares (PLS) type II regression of the TPI spectra and by generating a pure-component projection (PCP) basis set using net analyte signal (NAS) processing. Following generation of the calibration vectors, the performance of both methods at predicting the concentration of theophylline, lactose, and MCC was compared using the validation spectra and by generating chemical images from samples with known composition patterns. Sensitivity was observed for the PLS calibration over the range of all constituents for both the calibration and the validation datasets; however, some of the calibration statistics indicate that PLS overfits the spectra. Multicomponent prediction images verified the spatial and composition fidelity of the system. The NAS-PCP calibration procedure yielded accurate linear predictions of theophylline and lactose, whereas the results for MCC prediction were poor. The poor sensitivity for MCC is assumed to be related to the relative lack of phonon absorption bands, which concurs with the characterization of MCC as being semi-crystalline. The results of this study demonstrate the use of TPI reflection spectroscopy and efficient NAS-PCP for the quantitative analysis of crystalline pharmaceutical materials.
On-line Monitoring of Traditional Chinese Medicinal Powder Blending Process by Using Near-Infrared Spectroscopy

Abstract:OBJECTIVE To establish a rapid method based on near-infrared diffuse reflectance spectroscopy(NIR) to monitor the blending process of traditional Chinese medicinal powder on-line.METHODS NIR spectra were collected during the blending processes of mixtures consisting of Red Peony Root powder and defatted Spine Date Seed powder.Multivariate calibration models based on PLS regression were developed to correlate the collected spectra with the content of Red Peony Root powder.The optimal calibration model was applied to monitor the blending process in real time with noninterference.RESULTS After spectral pretreatment using multiplicative signal correction(MSC),first derivative and Savitzky-Golay filter,Red Peony Root powder contents were quantified using PLS algorithm in the spectral ranges of 5 900-5 950 cm-1,6 700-6 750 cm-1 and 6 950-7 100 cm-1.The correlation coefficient of the calibration model was 0.999 0.The RMSEC,RMSEP and RMSECV were 1.14,1.16 and 1.26,respectively.CONCLUSION It was demonstrated that a miniature near-infrared analyzer could be used to monitor the contents of the constituents and the blending homogeneity of traditional Chinese medicinal powder in real time,and to rapidly determine the end-point of blending process.In addition,it can be used to judge whether the physical state of powder mixture conforms to the standard.Therefore,NIR spectroscopy is considered to be an appropriate PAT tool for end-point control of blending process of traditional Chinese medicinal powder.

Abstract:Non-destructive analysis of chlorpheniramine maleate (CPM), pharmaceutical tablets, and granules was conducted by chemometrics-assisted attenuated total reflectance infrared spectroscopy (ATR-IR). For tablets, an optimum PLSR model with eight latent factors was obtained from area-normalized and standard normal variate (SNV) pretreated ATR-IR spectral data with correlation coefficients (R2) of calibration and cross-validation of 0.9716 and 0.9602, respectively. The model capability for the 42 test set samples was proven with R2 between the reference and model prediction values of 0.9632, and a root-mean-square error of prediction (RMSEP) of 1.7786. The successive PLSR model for granules was constructed from SNV and first derivative pretreated ATR-IR spectral data with two latent factors and correlation coefficients (R2) of calibration and cross-validation of 0.9577 and 0.9450, respectively.

Abstract:As a rapid and accurate technique, near infrared (NIR) spectroscopy has been widely used in process monitoring to improve efficiency and ensure quality consistency. In the present work, the application of NIR spectroscopy for on-line quantitative monitoring of the concentration process of Wangbi tablets was investigated. Partial least squares regression (PLSR) models based on the full-range spectra or the key wavelengths selected by the competitive adaptive reweighted sampling (CARS) method were established. The accuracy of the CARS-PLSR method was greater than that of full-spectrum PLSR for three quality parameters (soluble solid contents, paeoniflorin and icariin). For the prediction set samples using the CARS-PLSR models the coefficients of determination (r(2)) and root mean square error [RMSEP (%)] were 0.97 and 1.29 for soluble solid contents, 0.93 and 0.014 for paeoniflorin and 0.85 and 0.009 for icariin, respectively. Overall the results indicated that NIR spectroscopy coupled with CARS-PLSR calibration is a reliable and non-destructive alternative method for on-line monitoring of the concentration process of Wangbi tablets on an industrial scale.

Abstract:The use of near-infrared spectroscopy for quality control of nonsteroidal anti-inflammatory drugs (NSAIDs) in blister packs is demonstrated. NSAIDs with the active ingredients ibuprofen, diclofenac, naproxen, etoricoxib, acetylsalicylic acid and meloxicam have been studied. IR spectra have been recorded using a Frontier FTIR spectrometer with a NIRA attachment, used to collect diffuse reflectance spectra of solids. Processing and visualization of the study results has been carried out using chemometric algorithms – principal component analysis (PCA), hierarchical cluster analysis (HCA) and partial least squares regression (PLS). It is shown that using these algorithms it is possible to identify drugs by their manufacturer and determine the concentration of active substances. In the PCA method, the points corresponding to the samples of the analyzed tablets are located in separate quadrants depending on their manufacturer. The use of hierarchical cluster analysis (HCA) made it possible not only to differentiate the analyzed tablets by manufacturer, but also to determine the content of the active substance in them. “Measured-predicted” calibration models have been constructed, the values of the root mean square residual of calibration (RMSEC) did not exceed 6.28 mg. The relative discrepancies between the values obtained during the analysis and those assigned correspond to the permissible deviations established by order of the Ministry of Health of the Russian Federation dated October 26, 2015 no. 751n. The duration of the analysis was 7–10 min.

Abstract:Terahertz (THz) radiation is the electromagnetic spectrum lying between the microwave and infrared portions, and covers wavelength between 3mm and 30um. Terahertz spectroscopy has been used in qualitative and quantitative analysis since molecular vibrations and rotations of a great large number of materials lie in this region. Many techniques, such as principal component regression (PCR) and partial least square regression (PLSR), have been applied in quantitative analysis of terahertz time domain spectroscopy. However, those "soft-modeling" methods, though effective in many cases, all require a large number of samples and have poor extrapolation capability, and the utilization is also restricted in some cases (samples lack or requirement of extrapolation). A parametric method of spectrum, Indirect Hard Modeling (IHM), was proposed to resolve terahertz spectral matrix and quantify the mixtures. Savitzky-Golay method was used to eliminate the influence of noise. Moreover, the features of terahertz spectra were considered and the artificial baseline was also resolved as a signal spectrum to improve spectrum extraction accuracy. Subsequently, the analysis of IHM was carried out, and the ability of prediction and accuracy of the model in the case of low training samples were discussed. In order to validate the capability of the proposed method, quaternary systems of Rifampicin, Lactose monohydrate, microcrystalline cellulose and magnesium stearate were tested. The regression correlation coefficient (R) and root mean square error (RMSE) were employed to estimate the performance of the model. Finally, the comparison of IHM and PLSR were carried out, and theoretical analysis and experimental results indicated that the number of samples required by IHM modeling could be reduce to two during calibration, at the same time, the accuracy of quantitative and extrapolation capabilities were also improved.
