Abstract:In this paper we describe the strategy used in the development and validation of a near infrared spectroscopy method for the rapid determination of baicalin, chlorogenic acid, ursodeoxycholic acid (UDCA), chenodeoxycholic acid (CDCA), and the total solid contents (TSCs) in the Tanreqing injection. To increase the representativeness of calibration sample set, a concentrating-diluting method was adopted to artificially prepare samples. Partial least square regression (PLSR) was used to establish calibration models, with which the five quality indicators can be determined with satisfied accuracy and repeatability. In addition, the slope/bias (S/B) method was used for the models transfer between two different types of NIR instruments from the same manufacturer, which is contributing to enlarge the application range of the established models. With the presented method, a great deal of time, effort and money can be saved when large amounts of Tanreqing injection samples need to be analyzed in a relatively short period of time, which is of great significance to the traditional Chinese medicine (TCM) industries.

Abstract:As a rapid and accurate technique, near infrared (NIR) spectroscopy has been widely used in process monitoring to improve efficiency and ensure quality consistency. In the present work, the application of NIR spectroscopy for on-line quantitative monitoring of the concentration process of Wangbi tablets was investigated. Partial least squares regression (PLSR) models based on the full-range spectra or the key wavelengths selected by the competitive adaptive reweighted sampling (CARS) method were established. The accuracy of the CARS-PLSR method was greater than that of full-spectrum PLSR for three quality parameters (soluble solid contents, paeoniflorin and icariin). For the prediction set samples using the CARS-PLSR models the coefficients of determination (r(2)) and root mean square error [RMSEP (%)] were 0.97 and 1.29 for soluble solid contents, 0.93 and 0.014 for paeoniflorin and 0.85 and 0.009 for icariin, respectively. Overall the results indicated that NIR spectroscopy coupled with CARS-PLSR calibration is a reliable and non-destructive alternative method for on-line monitoring of the concentration process of Wangbi tablets on an industrial scale.

Abstract:To implement calibration transfer between Raman spectrometers, an improved piecewise direct standardization (PDS) is proposed in the present paper. Standard normal variate (SNV) is firstly introduced to reduce the influence of spectral background and intensity corresponding to the master spectrometer and the slave spectrometer; then PDS algorithm is used to eliminate the differences between Raman spectra for a specific sample. Moreover, a new quantitative criterion, called transfer error rate, is proposed to evaluate the performance of calibration model transfer. This improved PDS is applied to Raman spectral analysis of gasoline. The result shows that the proposed algorithm not only needs a small quantity of transfer samples, but also obtains high transfer accuracy and strong model robustness.

Abstract:Calibration transfer (CT) is an effective tool to address spectral inconsistencies and the decline in model prediction capabilities caused by various influences. This study introduced a modified semi-supervised parameterfree calibration enhancement (MSS-PFCE) strategy to enable calibration models extensively applicable across varying temperatures, instruments and orientations. Investigations conducted on three datasets of watermelon juice, corn, and apples were utilized to validate the efficacy of MSS-PFCE. The results showed that MSS-PFCE decreased the average RMSEP for slave spectrum predictions by 96.51%, 75.96%, and 23.04%, respectively. MSS-PFCE significantly outperformed the original SS-PFCE approach and three classical standardization methods (direct standardization (DS), piecewise direct standardization (PDS) and slope and bias correction (SBC)). Furthermore, MSS-PFCE required no standards and needed only 5%-10% of slave samples for CT, maintaining robust reliability across varying sample sizes and cost thresholds (cth). The conclusions were further examined in three additional datasets with these variations. Notably, MSS-PFCE exhibited high accuracy, fitting stability, low sample dependency, and versatile transferability, positioning it as a promising candidate for widespread implementation in visible/near-infrared (Vis/NIR) spectroscopy. This study has significant potential to enhance the applicability of quality analysis, broaden the scope of food quality control, and promote the sound development of the food industry.

Abstract:A rapid and simultaneous quantification of the main active compounds of the in-process extract solutions is beneficial to the process monitoring as well as ensuring quality consistency of the end products during the production of traditional Chinese medicine. Here we present a near infrared (NIR) spectroscopy-based method for rapid analysis of extract solutions in the production of compound E Jiao oral liquid. The partial least-squares regression (PLSR) models for four quality indicators (viz. total flavonoids, total saponins, total saccharides and soluble solid contents) were established and validated. The results showed that all of the four models exhibited satisfactory fitting and predictive capacity. The root mean squares error of prediction (RMSEP) was 0.0384 mg mL−1, 0.0154 mg mL−1, 3.80 mg mL−1 and 0.199% for total flavonoids, total saponins, total saccharides and soluble solid contents, respectively. This work here demonstrated that NIR spectroscopy coupled with PLSR calibration can offer a reliable and non-destructive alternative in the routine monitoring of the extraction process in the production of compound E Jiao oral liquid. The presented approach is expected to be equally applicable to the mixed decoction of other herbal medicines.

Abstract:In recent years, transmission Raman spectroscopy (TRS) has emerged as a potent new tool for rapid, nondestructive quantitation in pharmaceutical manufacturing. In order to expand the applicability of TRS and enhance its use in product quality monitoring during drug production, we aimed, in the present study, to apply partial least-squares (PLS) approaches to build a model consisting of 150 handmade tablets and covering 15 levels through the use of a multifactor orthogonal design of experiment (DOE), which was used to predict concentrations of validation tablets made by hand. The difference between results according to HPLC and TRS were negligible. The model was used to predict the active pharmaceutical ingredient (API) content in four random commercial paracetamol tablets, and corrected with the spectra of the commercial tablets to obtain four corresponding models. The results show that the content relative error in the model’s predictions after correction with commercially available tablets was significantly lower than that before correction. The corrected model was used to make predictions for 20 tablets from the brand Panadol. Compared with the HPLC results, the prediction relative error was basically less than 4.00%, and the relative standard deviation (RSD) of the content was 0.86%.

Abstract:A method for rapid quantitative analysis of four kinds of Tanreqing injection intermediates was developed based on Fourier transform near infrared (FT-NIR) spectroscopy and partial least squares (PLS) algorithm. The NIR spectra of 120 samples were collected in transflective mode. The concentrations of chlorogenic acid, caffeic acid, luteoloside, baicalin, ursodesoxycholic acid (UDCA), and chenodeoxycholic acid (CDCA) were determined with the HPLC–DAD/ELSD as reference method. In the PLS calibration, the NIR spectra were pretreated with different methods and the number of PLS factors used in the model calibration was optimized by leave-one-out cross-validation. The performance of the final PLS models was evaluated according to the root mean square error of calibration (RMSEC), root mean square error of cross-validation (RMSECV), root mean square error of prediction (RMSEP), BIAS, standard error of prediction (SEP), and correlation coefficients (R). The R values in the prediction sets were all higher than 0.93, and the SEPs for the 6 compounds are 1.18, 6.02, 2.71, 155, 126, 30.0mg/l, respectively. The established models were used for the liquid preparation process analysis of Tanreqing injection in three batches, and a model updating method was proposed for the long-term usage of the established models. This work demonstrated that NIR spectroscopy is more rapid and convenient than the conventional methods to analyze the intermediates of Tanreqing injection, and the presented method is helpful to the implementation of process analytical technology (PAT) in pharmaceutical industry of Chinese Medicines Injections.

Abstract:Designing a calibration set is the first step in developing a multivariate spectroscopic calibration method for quantitative analysis of pharmaceutical tablets. This step is critical because successful model development depends on the suitability of the calibration data. For spectroscopic-based methods, traditional concentration based techniques for designing calibration sets are prone to have redundant information while simultaneously lacking necessary information for a successful calibration model. A method for designing a calibration set in spectral space was developed. The pure component spectra of a tablet formulation were used to define the spectral space of that formulation. This method maximizes the information content of measurements and minimizes sample requirements to provide an efficient means for developing multivariate spectroscopic calibration. A comparative study was conducted between a commonly employed full factorial approach to calibration development and the newly developed technique. The comparison was based on a system to quantify a model drug, acetaminophen, in pharmaceutical compacts using near infrared spectroscopy. A 2-factor full factorial design (acetaminophen with 5 levels and MCC:Lactose with 3 levels) was used for calibration development. Three replicates at each design point resulted in a total of 45 tablets for the calibration set. Using the newly developed spectral based method, 11 tablets were prepared for the calibration set. Partial least square (PLS) models were developed from respective calibration sets. Model performance was comprehensively assessed based on the ability to predict acetaminophen concentrations in multiple prediction sets. One prediction set contained similar information to calibration set while the other prediction sets contained different information from calibration set in order to assess the model accuracy and robustness. Similar prediction performance was achieved using the 11-tablet design (spectral space), compared to the 45-tablet full factorial approach. This work demonstrates that a calibration set designed in spectral space provided an efficient means of developing spectroscopic multivariate calibration.

Abstract:Variable selection methods can simplify the modelling process and improve the accuracy of models constructed for rapid monitoring using near-infrared (NIR) spectroscopy. In this study, NIR spectroscopy was applied in combination with chemometrics to determine the icariin, salvianolic acid B, and puerarin content, as well as the relative density through the concentration process of a Yangxinshi tablet (YXST) water extraction solution. Partial least squares with several variable selection algorithms were used for the modelling. Variable selection methods, including synergy interval partial least squares (Si-PLS), a genetic algorithm, competitive adaptive reweighted sampling (CARS), and their combination, were comparatively applied to calibrate the regression model. The performances of the models obtained were systematically evaluated according to the correlation coefficients of prediction (Rp), the relative standard error of prediction (RSEP), and the residual predictive deviation (RPD). For icariin, salvianolic acid B, and puerarin, the Si-PLS model showed optimal results. The Rp, RSEP, and RPD of Si-PLS were 0.9905, 7.84%, and 7.38 for icariin; 0.9781, 9.48%, and 4.80 for salvianolic acid B; and 0.9668, 12.81%, and 3.92 for puerarin, respectively. For the relative density, the CARS-Si-PLS acquired the best prediction results. The Rp, RSEP, and RPD of CARS-Si-PLS were 0.9725, 0.47%, and 4.31 for the relative density, respectively. This study demonstrated that variable selection methods can simplify the modelling process and improve the accuracy of the models. In addition, the methods developed in this study are suitable for a quality monitoring of the concentration process of YXST water extraction solution, and they provide reference for real-time monitoring of the concentration processes used in the preparation of other types of traditional Chinese medicine.
On-line Monitoring of Traditional Chinese Medicinal Powder Blending Process by Using Near-Infrared Spectroscopy

Abstract:OBJECTIVE To establish a rapid method based on near-infrared diffuse reflectance spectroscopy(NIR) to monitor the blending process of traditional Chinese medicinal powder on-line.METHODS NIR spectra were collected during the blending processes of mixtures consisting of Red Peony Root powder and defatted Spine Date Seed powder.Multivariate calibration models based on PLS regression were developed to correlate the collected spectra with the content of Red Peony Root powder.The optimal calibration model was applied to monitor the blending process in real time with noninterference.RESULTS After spectral pretreatment using multiplicative signal correction(MSC),first derivative and Savitzky-Golay filter,Red Peony Root powder contents were quantified using PLS algorithm in the spectral ranges of 5 900-5 950 cm-1,6 700-6 750 cm-1 and 6 950-7 100 cm-1.The correlation coefficient of the calibration model was 0.999 0.The RMSEC,RMSEP and RMSECV were 1.14,1.16 and 1.26,respectively.CONCLUSION It was demonstrated that a miniature near-infrared analyzer could be used to monitor the contents of the constituents and the blending homogeneity of traditional Chinese medicinal powder in real time,and to rapidly determine the end-point of blending process.In addition,it can be used to judge whether the physical state of powder mixture conforms to the standard.Therefore,NIR spectroscopy is considered to be an appropriate PAT tool for end-point control of blending process of traditional Chinese medicinal powder.
Nondestructive Qualitative and Quantitative Analysis of Yaobitong Capsule Using Near-Infrared Spectroscopy in Tandem with Chemometrics.

Abstract:The purpose of the study is to present a nondestructive qualitative and quantitative approach of hard-shell capsule using near-infrared (NIR) spectroscopy combined with chemometrics. The Yaobitong capsule (YBTC) was used for demonstration of the proposed approach and the NIR spectra were collected using a handheld fiber probe (FP) without the damage of capsule shell. By comparing the differences and similarities of the NIR spectra of capsule shells, contents and intact capsules, a preliminary conclusion can be drawn that the NIR spectra contained the information of the contents. Characteristic variables were selected by competitive adaptive weighted resampling (CARS) method, and least squares support vector machine (LSSVM) method based on particle swarm optimization (PSO) algorithm was applied to the construction of quantitative models. The relative standard error of prediction (RSEP) values of five saponins including notoginsenoside R1, ginsenoside Rg1, Re, Rb1, and Rd were 3.240%, 5.468%, 5.303%, 5.043%, and 3.745%, respectively. In addition, for qualitative model, three different types of adulterated capsules were designed. The model established by data driven version of soft independent modeling of class analogy (DD-SIMCA) demonstrated a satisfactory result that all adulterated capsules were identified accurately after an appropriate number of principal components (PCs) were chosen. The results indicated that although the NIR spectra collection was affected by capsule shell, sufficient content information can be obtained for quantitative and qualitative analysis after combining with chemometrics. It further proved that acquired NIR spectra do contain the effective component information of the capsule. This study provided a reference for the rapid nondestructive quality analysis of traditional Chinese medicine (TCM) capsule without damaging capsule shell.
Feasibility of an NIR spectral calibration transfer algorithm based on optimized feature variables to predict tobacco samples in different states.

Abstract:The prediction accuracy of calibration models for near-infrared (NIR) spectroscopy typically relies on the morphology and homogeneity of the samples. To achieve non-homogeneous tobacco samples for non-destructive and rapid analysis, a method that can predict tobacco filament samples using reliable models based on the corresponding tobacco powder is proposed here. First, as it is necessary to establish a simple and robust calibrated model with excellent performance, based on full-wavelength PLSR (Full-PLSR), the key feature variables were screened by three methods, namely competitive adaptive reweighted sampling (CARS), variable combination population analysis-iteratively retaining informative variables (VCPA-IRIV), and variable combination population analysis-genetic algorithm (VCPA-GA). The partial least squares regression (PLSR) models for predicting the total sugar content in tobacco were established based on three optimal wavelength sets and named CARS-PLSR, VCPA-IRIV-PLSR and VCPA-GA-PLSR, respectively. Subsequently, they were combined with different calibration transfer algorithms, including calibration transfer based on canonical correlation analysis (CTCCA), slope/bias correction (S/B) and non-supervised parameter-free framework for calibration enhancement (NS-PFCE), to evaluate the best prediction model for the tobacco filament samples. Compared with the previous two transfer algorithms, NS-PFCE performed the best under various wavelength conditions. The prediction results indicated that the most successful approach for predicting the tobacco filament samples was achieved by VCPA-IRIV-PLSR when coupled with the NS-PFCE method, which obtained the highest determination coefficient ( = 0.9340) and the lowest root mean square error of the prediction set (RMSEP = 0.8425). VCPA-IRIV simplifies the calibration model and improves the efficiency of model transfer (31 variables). Furthermore, it pledges the prediction accuracy of the tobacco filament samples when combined with NS-PFCE. In summary, calibration transfer based on optimized feature variables can eliminate prediction errors caused by sample morphological differences and proves to be a more beneficial method for online application in the tobacco industry.
Development and Validation of a Near-Infrared Spectroscopy Method for Multicomponent Quantification During the Second Alcohol Precipitation Process of Astragali Radix

Abstract:The objective of this study was to develop and validate a near-infrared (NIR) spectroscopy based method for in-line quantification during the second alcohol precipitation process of Astragali radix. In total, 22 calibration experiments were carefully arranged using a Box–Behnken design. Variations in the raw materials, critical process parameters, and environmental temperature were all included in the experimental design. Two independent validation sets were built for method evaluation. Validation set 1 was used for optimization. Different spectral pretreatments were compared using a “trial-and-error” approach. To reduce the calculation times, the full-factorial design was applied to determine the potential optimal combinations. Then, the best parameters for the pretreatment algorithms were compared and selected. Partial least squares (PLS) regression models were obtained with low complexity and good predictive performance. Validation set 2 was used for a thorough validation of the NIR spectroscopy method. Based on the same validation set, traditional chemometric validation and validation using accuracy profiles were conducted and compared. Conventional chemometric parameters were used to obtain the overall predictive capability of the established models; however, these parameters were insufficient for pharmaceutical regulatory requirements. Then, the method was fully validated according to the ICH Q2(R1) guideline and using the accuracy profile approach, which enabled visual and reliable representation of the future performances of the analytical method. The developed method was able to determine content ranges of 8.44–39.8% at 0.541–2.26 mg/mL, 0.118–0.502 mg/mL, 0.220–0.940 mg/mL, 0.106–0.167 mg/mL, 0.484–0.879 mg/mL, and 0.137–0.320 mg/mL for total solid, calycosin glucoside, formononetin glucoside, 9, 10-dimethoxypterocarpan glucopyranoside, 2′-dihydroxy -3′, 4′-dimethoxyisoflavan glucopyranoside, astragloside II, and astragloside IV, respectively. These ranges were specific to the early and middle stages of the second alcohol precipitation process. The method was confirmed to be capable of achieving an in-line prediction with a very acceptable accuracy. The present study demonstrates that accuracy profiles offer a potential approach for the standardization of NIR spectroscopy method validation for traditional Chinese medicines (TCMs).
A Feasibility Study on the Non-Invasive Analysis of Bottled Compound E Jiao Oral Liquid Using Near Infrared Spectroscopy

Abstract:The aim of this study was to explore the feasibility of near infrared (NIR) spectroscopy for the quantitative analysis of Compound E Jiao oral liquid, a popular oral TCM preparation, through intact glass vials. Transmission spectra were acquired with a sample holder tailored to the instrument's sample compartment. Total flavonoids, total saponins and soluble solid contents were considered as quality indicators of this oral preparation and calibration models were built using partial least-squares regression (PLSR) algorithm. The sample set was comprised of production samples as well as laboratory samples prepared by a concentrating-diluting method so as to extend the concentration range beyond the production variation. The developed models gave root mean square error of prediction (RMSEP) of 0.084 mg mL(-1) for total flavonoids, 0.014 mg mL(-1) for total saponins and 0.21% for soluble solid contents. To better evaluate the prediction performance, an independent set of ten production samples was introduced for the external validation. The models are compared with those built with the transflectance spectra collected in the destructive way. The results of paired t-test and F-test at a significance level of 0.05 revealed that the two sets of models exhibited comparable model performance. The overall results demonstrate that the proposed non-destructive method can be used as an advantageous alternative in the quality inspection of Compound E Jiao oral liquid production. Abstract:Handheld Raman spectroscopy is actually booming. Recent devices improvements aim at addressing the usual Raman spectroscopy issues: fluorescence with shifted-excitation Raman difference spectroscopy (SERDS), poor sensitivity with surface enhanced Raman scattering (SERS) and information only about the sample surface with spatially offset Raman spectroscopy (SORS). While qualitative performances of handheld devices are generally well established, the quantitative analysis of pharmaceutical samples remains challenging. The aim of this study was to compare the quantitative performances of three commercially available handheld Raman spectroscopy devices. Two of them (TruScan and IDRaman mini) are equipped with a 785 nm laser wavelength and operate in a conventional backscattering mode. The IDRaman has the Orbital Raster Scanning (ORS) option to increase the analyzed surface. The third device (Resolve) operates with an 830 nm laser wavelength both in backscattering and in SORS modes. The comparative study was carried out on ibuprofen-mannitol-microcrystalline cellulose ternary mixtures. The concentration of ibuprofen ranged from 24 to 52% (w/w) while the proportions of the two excipients were varied to avoid cross-correlation as much as possible. Analyses were performed either directly through a glass vial or with the glass vial in an opaque polypropylene flask, using a validated FT-NIR spectroscopy method as a reference method. Chemometric analyses were carried out with the Partial Least Squares Regression (PLS-R) algorithm. The quantitative models were validated using the total error approach and the ICH Q2 (R1) guidelines with ± 15% as acceptance limits.
An Approach Combining Real-Time Release Testing with Near-Infrared Spectroscopy to Improve Quality Control Efficiency of Rhizoma Paridis

Abstract:Raw material examination is a critical process in the industrial production of traditional Chinese medicine (TCM); high accuracy and minimal time consumption are both required. In this study, near-infrared (NIR) spectroscopy was applied to improve the quality control efficiency of Rhizoma paridis. Partial least squares regression (PLSR) was first used to develop quantitative calibration models, and the discriminant analysis model was established to qualitatively discriminate the qualified samples from the unqualified samples. These two established NIR models were applied for real-time release testing (RTRT) of R. paridis. R. paridis saponins (RPS)≥0.6% and moisture ≤12% were used as the quantitative releasing criteria of RTRT according to the Chinese Pharmacopoeia. Qualified samples classified by the discriminant analysis model were deemed to meet the qualitative releasing criterion of RTRT. Using the established quantitative model, 24 samples were allowed to be released to the subsequent production processes with 100% accuracy. For the qualitative RTRT analysis, three samples were misclassified as the unqualified class and were released unsuccessfully, the accuracy of the qualitative RTRT was 90%. Therefore, the quantitative RTRT was more feasible for actual manufacturing processes. Based on this study, a rapid and effective quantitative NIR spectroscopic method was proposed for the RTRT of R. paridis. The combination of RTRT and NIR spectroscopy could be a potential tool to improve the quality control efficiency of R. paridis.
Quantitative Real-Time Release Testing of Rhubarb Based on Near-Infrared Spectroscopy and Method Validation

Abstract:The quality control of the Traditional Chinese medicine (TCM) are important links in the production and application of pharmaceuticals. In this study, the quantitative analysis of rhubarb was realized by near-infrared spectroscopy (NIRs), including the rapid detection of free anthraquinone and total anthraquinone content. Combined with method validation of NIR quantitative model and release limit, a reliable real-time release testing (RTRT) method for rhubarb was constructed. The competitive adaptive weighted resampling (CARS) method was applied for characteristic variables selection, and the quantitative model was established based on partial least squares regression (PLSR) method and particle swarm optimization based least square support vector machines (PSO-LSSVM) method. The relative standard error of prediction (RSEP) values of free anthraquinone and total anthraquinone models were 10.66% and 4.95%, respectively. The accuracy profile (AP) was introduced to validate and evaluate the performance of the optimized model at different concentration levels. The relative bias, precision and linearly of the two quantitative models were all within the acceptable range. Based on the results of the method validation, the minimum release control limit for total anthraquinone content was set as 1.569% to make sure accurate release, slightly higher than the pharmacopoeia standard. The constructed RTRT system for rhubarb based on the NIR model validation can improve the efficiency and accuracy of quality control.

Abstract:Calibration transfer without standard samples is essential for practical applications of near infrared spectroscopy because, sometimes, it is difficult or even impossible to obtain the standard samples for measuring their spectra on the secondary instrument. In this work, a modified linear model correction method is proposed for improving the transfer accuracy and computational efficiency. The constraint of linear model correction was replaced by a robust convex equation to restrict the model similarity in the optimization. The near infrared dataset of pharmaceutical tablet measured with different instruments are used to test the performance of the method. The result shows that a modified linear model correction achieves a high efficiency of the transfer while the computational complexity can be considerably reduced. The method may provide a robust way in practical application.

Abstract:Compatibility and in-use stability screening studies are required for dosing vehicle selection based on the FDA's guidance, "Use of Liquids and/or Soft Foods as Vehicles for Drug Administration: General Considerations for Selection and In Vitro Methods for Product Quality Assessments." One of the major analytical challenges in these studies is sample preparation because extracting active pharmaceutical ingredient (API) from the drug product mixed into viscous soft-food matrices (e.g., yogurt or apple sauce) is laborious, prone to human errors, and time-consuming. Additionally, observed in-solution degradation caused by dosing vehicle ingredients causes analytical error. In our study, NIR- and Raman-based non-destructive tests have been explored and developed with drug product powder formulation prepared in dosing vehicles. A transmission Raman chemometrics model was developed and calibrated with samples varying in API content, water content, and milled extrudate particle size distribution. The method was proven to be accurate, linear, selective, and robust. Our work with non-destructive tests eases the laboratory burdens to perform in-use stability studies with dosing vehicles for all phases of development that need to cover all application scenarios of clinical preparation and usage.
