Abstract:Objective To investigate the changes of VEGF expression and ultrastructure in hippocampus of adult rats after whole- brain irradiation. Methods The whole brain of female Sprague- Dawley rats was irradiated by 20Gy 6mV X- ray. The ex-pression of VEGF mRNA in hippocampus tissue before and 1d, 1, 2, 4, 8w after ionizing radiation was detected by RT- PCR. The ultra- structural changes of hippocampus were observed by electronic microscope. Results RT- PCR showed a transient de-crease in VEGF mRNA expression within 1d after exposure to a single dose of 20Gy 6mV X- ray ionizing radiation;however, there were no differences in VEGF mRNA expression at 1, 2, 4, 8w after ionizing radiation when compared to controls (P>0.05). Neu-ronal mitochondria edema,vacuolization, endothelial gap increase and lumen narrow were demonstrated by electron microscope at 4w after whole brain irradiation. Conclusion VEGF may be involved in early radiation- induced damage in the hippocampus, which indicates that VEGF might be a novel target for prevention of radiation- induced brain injury.
Effect of enoxaparin and aspirin on hemodynamic disturbances after global cerebral ischemia in rats.

Abstract:Study aim: Whether patients benefit from anticoagulation or anti-platelet agents when subjected to global ischemia is controversial. In this study we assessed the effect of enoxaparin and aspirin on hemodynamic disturbances following global ischemia in rats. Methods: Twenty-minute global ischemia was induced by four-vessel occlusion in Sprague-Dawley rats. Enoxaparin (1.5 mg/kg, iv.) was administered 4 min before or after ischemia, and aspirin (40 mg/kg, i.p.) was administered twice, 18 and 1.5h before ischemia. Parietal cortical cerebral blood flow was measured in real time with a laser Doppler flowmeter, and activated partial thromboplastin time (APTT) and prothrombin time (PT) were measured. Three phenomena of hemodynamic disturbance: no-reflow, hyperperfusion, and hypoperfusion, were assessed after 20-min global ischemia. Results: No significant differences in PT values were found; in contrast, the APTT values were shortened markedly by 20-min ischemia, which indicated activation of the endogenous coagulation system. The shortened APTT was markedly prevented by enoxaparin administered before ischemia. No-reflow was partly mitigated by administration of enoxaparin or aspirin before ischemia. Hypoperfusion was almost fully blocked by aspirin, and the hypoperfusion value was increased and hypoperfusion duration was shortened by enoxaparin administered 4 min before or after ischemia. Conclusion: These results indicate that enoxaparin and aspirin have protective effects against cerebral hemodynamic disturbances induced by global ischemia in rats. (C) 2010 Elsevier Ireland Ltd. Effects of Tetrahydrobiopterin on Cerebral Infarction after Transient Focal Ischemia in Rats.

Abstract:OBJECTIVES:The present study investigated the effects of tetrahydrobiopterin (BH4) on cerebral infarction after transient focal ischemia in rats.METHODS:Focal ischemia (1·5 hours) was created in male Sprague-Dawley rats (250-280 g) by middle cerebral artery occlusion. Some rats were treated with 20 mg/kg tetrahydrobiopterin by intraperitoneal injection 30 minutes before reperfusion. At 2, 6, and 12 hours of reperfusion, the brains were harvested for the nitric oxide synthase (NOS) activity and nitric oxide (NO) level assays. At 12 hours of reperfusion, the brains were harvested for infarct size measurement.RESULTS:NOS activity and NO level were all augmented after reperfusion. BH4 treatment significantly further increased NOS activity and NO level. Cerebral infarct size was significantly bigger in BH4 treatment group compared to that in no treatment group.CONCLUSIONS:The data indicate that BH4 enhances cerebral infarction after transient focal ischemia in rats, through NOS and NO pathway.
Neuroprotective effect of edaravone on focal cerebral ischemia in rats

Abstract:OBJECTIVE To determine the neuroprotective effect of edaravone on focal cerebral ischemia in rats.METHODS Focal cerebral ischemia was induced by middle cerebral artery (MCA) occlusion for 60 min and followed by 24 h repeffusion. Edaravone (0.1～3.0 mg·kg~(-1)) or vehicle (saline 2 mL·kg~(-1)) was injected intravenously 30 min before MCA ochision and after 2 h reperfusion. The neurological score, infarct volume, neuron density (in cortex, hippocampus, and striatum), brain edema, malondialdehyde (MDA) content, superoxide dismutase (SOD) and lactate dehydrogenase (LDH) activity in brain tissue were determined after 24 h reperfusion. RESULTS Edaravone slightly improved the neurological deficiency, decreased infarct volume and neuron loss at the doses of 0.3, 1.0, 3.0 mg·kg~(-1)(P0.05). The enlargement of the ischemie hemisphere was inhibited at the doses of 0.3, 1.0,3.0 mg·kg~(-1). Edaravone markedly reduced MDA content and LDH activity at the doses of 0.3～3.0 mg·kg~(-1). Edaravone had no effect on SOD activity. CONCLUSION Edaravone displays the pretective effect on focal cerebral ischemia in rats, which is partially attributed to the inhibition of brain edema and the elimination of free radical.
Vinpocetine and coenzyme Q10 combination alleviates cognitive impairment caused by ionizing radiation by improving mitophagy

Abstract:Objective: This research was designed to ascertain the effect and mechanism of vinpocetine (VIN) and coenzyme Q10 (CoQ10) combination on cognitive impairment induced by ionizing radiation (IR). Methods: Cognitive impairment in mice was induced by 9-Gy IR, and they were intraperitoneally injected with VIN, CoQ10, or VIN + CoQ10. Then novel object recognition and Morris water maze tests were used to detect cognitive function. The number of hippocampal neurons and BrdU+Dcx+ cells was observed by Nissl and immunofluorescence staining. Mitochondrial respiratory complex I, adenosine triphosphate (ATP), and mitochondrial membrane potential (MMP) were evaluated, as well as oxidative stress injury. Mitophagy in hippocampal neurons was evaluated by observing the ultrastructure of hippocampal neurons and assessing the expression of mitophagy-related proteins. Results: IR reduced novel object discrimination index, the time for platform crossing, and the time spent in platform quadrant, in addition to neuron loss, downregulated levels of mitochondrial respiratory complex I, ATP, and MMP, aggravated oxidative stress injury, increased expression of LC3 II/I, Beclin1, PINK1, and parkin, and decreased P62 expression. VIN or CoQ10 treatment mitigated cognitive dysfunction, neurons loss, mitochondrial damage, and oxidative stress injury, and enhanced mitophagy in hippocampal neurons. VIN and CoQ10 combination further protected against IR-induced cognitive dysfunction than VIN or CoQ10 alone. Conclusion: VIN combined with CoQ10 improved neuron damage, promoted mitophagy, and ameliorated cognitive impairment in IR mice.

Abstract:BACKGROUND:Gypenosides（ GP） has a protective effect on the brain of experimental rats with cerebralischemia,but whether it has the same effect on rats with vascular dementia(VD) for the rats also have ischemic-hypoxia injury in the neurons of hippocampus? OBJECTIVE： To observe the protective effect of GP on the positive neurons of neuronal nitric oxide synthase（ nNOS） and the nucleic acid in hippocampus of VD rats. DESIGN： A randomized control study. SETTING and MATERIALS： Study were carried out in the Central Laboratory of Medical college of CPAPF.A total of 30 male rats of grade two,with a body mass from 240 g to 260 g,were purchased from Medical Experimental Animal Center of Tianjin. INTERVENTIONS： Thirty male rats weighing 240 g to 260 g were divided by random number table into control group， model group,and drug group. Modified Pulsinelli's 4-vessel occlusion（ 4-VO） method was used to establish VD rat models.Drug group:GP(200 mg/kg) were filled into stomach and operation was made according to the establishing method of VD rat models.Control group:The same operation was made but without burning the vertebral artery and occluding the general artery. MAIN OUTCOME MEASURES：① the expression of nNOS in hippocampus of rats.② the intensity of fluorescence staining of DNA and RNA in hippocampus of rats. RESULTS： In the model group， the numbers of nNOS positive neurons in CA1 and CA3 fields of hippocampus were 20.47± 4.22 and 25.47± 3.52， which were significantly less than those in the control group(24.73± 5.72 and 37.13± 5.10,respectively)（ P< 0.05） and the fluorescent intensity of the DNA and RNA after acridine orange(AO) staining（ reflecting DNA and RNA contents） also decreased significantly.In the drug group， the numbers of nNOS positive neurons in hippocampus CA1 and CA3 fields were 30.00± 3.63 and 38.00± 5.00 respectively, which increased significantly than those in the model group（ P< 0.001),while the fluorescent intensity of the DNA and RNA after AO staining in the drug group was stronger than that of the model group,which was similar to that of the control group. CONCLUSION:GP can increase the expression of nNOS positive neurons in hippocampus of VD rats significantly.It has a protective effect on DNA and RNA injury of neurons in hippocampus.
Puerarin Protects Against Ischemic Brain Injury in a Rat Model of Transient Focal Ischemia

Abstract:OBJECTIVES:This study examines the efficacy of puerarin, a drug used in traditional Chinese medicine, in attenuating ischemic brain injury after cerebral ischemia and reperfusion, and explores possible mechanisms underlying neuroprotective effects.METHODS:The animal model of ischemia/reperfusion injury was induced by middle cerebral artery occlusion for 2 hours followed by up to 72 hour reperfusion. The rats were randomly assigned into four groups (n=6/group): puerarin at 100, 200 and 400 mg/kg or saline, administered intraperitoneally. Neurological outcome and infarct volume by 2% triphenyl tetrazolium chloride staining were determined 72 hours after reperfusion. Terminal deoxynucleotidyl transferase biotin-dUTP nick end labeling staining was used to detect the cell damage of brains (n=5/group). Erythropoietin activation was detected by enzyme-linked immunosorbent assay (n=5/group).RESULTS:Compared with the vehicle saline group, puerarin decreased infarction volume at doses of 200 mg/kg (p=0.045) and 400 mg/kg (p=0.0002), but not at 100 mg/kg (p=0.387). Functional neurological outcome was improved with puerarin at 400 mg/kg (p=0.015), but not at 100 mg/kg (p=0.68) or 200 mg/kg (p=0.056). Puerarin significantly decreased the terminal deoxynucleotidyl transferase biotin-dUTP nick end labeling staining cells compared with the vehicle group 4, 24 and 72 hours after reperfusion. The erythropoietin activity was higher in puerarin treated group compared with the vehicle group.DISCUSSION:Puerarin has neuroprotection effects in rats at doses of 200 and 400 mg/kg, administered intraperitoneally after transient middle cerebral artery occlusion which may be partly due to activation of erythropoietin activity.
Protective effect of chuanxiongzine-puerarin in a rat model of transient middle cerebral artery occlusion-induced focal cerebral ischemia.

Abstract:Aim The aim of this study was to investigate the protective effect of chuanxiongzine-puerarin on cerebral ischemia-reperfusion damage in a rat model. Methods Rats were randomized into the following three groups: control group (sham operation group), model group (cerebral ischemia-reperfusion damage group), and treatment group (puerarin group, chuanxiongzine-puerarin group, and nimodipine group). The effect of neurological deficit, infarct volume and levels of superoxide dismutase (SOD), malondialdehyde (MDA), nitric oxide (NO), synaptophysin, and interleukin-1 was assessed. The therapeutic effect was also evaluated micro-positive emission tomography with [F-18]fiuoro-2-deoxy-D-glucose. Results After cerebral ischemia-reperfusion damage, the neurologic deficit score and the infarct volume percentage in treatment group were significantly lower than in the model group (P < 0.01). Micro-positive emission tomography imaging showed that glucose metabolism in the right temporal lobe and the right frontal lobe were significantly decreased in the model group in comparison with the sham operation group (P < 0.01). After treatment, glucose metabolism increased significantly in the puerarin group and the chuanxiongzine-puerarin group, compared with the model group (P < 0.05 and < 0.01, respectively). The expression of interleukin-1 beta-mRNA was significantly higher in the model group than in the sham operation group (P < 0.01). After treatment, however, it was significantly decreased in the puerarin group (P < 0.05) and the chuanxiongzine-puerarin group (P < 0.01) compared with the model group. SOD activity in the model group (P < 0.01) was lower than that in the sham operation group, whereas MDA and NO content in the model group was more than that in the sham operation group (P < 0.01). In the chuanxiongzine-puerarin and puerarin treatment groups, SOD activity showed higher level compared with the model group (P < 0.01); however, MDA and NO content was decreased in the treatment group (P < 0.01). Synaptophysin level was significantly lower in the model group compared with the sham operation group (P < 0.01), whereas the chuanxiongzine-puerarin and puerarin groups showed significantly higher values than the model group (P < 0.05). Conclusion This study showed that chuanxiongzine-puerarin played an important protective role against cerebral ischemic reperfusion damage in a rat model. Radioprotective effects of valproic acid, a histone deacetylase inhibitor, in the rat brain.

Abstract:Radiotherapy is commonly used in the treatment of brain tumors but can cause significant damage to surrounding normal brain. The radioprotective effects of valproic acid (VPA) on normal tissue in the rat brain were evaluated following irradiation. Male Wistar rats were used in the present study and 48 rats were randomly divided into four groups consisting of 12 rats each. The whole-brain irradiation (WBI) was delivered by X-ray and the rats received the following treatment once a day for 5 days. The control group (sham-exposed group) received sham irradiation plus physiological saline. The VPA group received sham irradiation plus 150 mg VPA/kg. The X-ray group received WBI plus physiological saline. The combined group received WBI plus 150 mg/kg intraperitoneally VPA. A total of 6 months post-irradiation, the rats were sacrificed and the brains were harvested. Cell apoptosis in the cortex was determined by immunohistochemistry 24 h post-irradiation using an antibody for protein caspase-3. Transmission electron microscope (TEM) analyses were used to assess the effects of VPA on the radioprotection of rat normal brain cells 6 months post-irradiation. The weights of the animals in the TEM group measured over the two weeks after the first injection of VPA were also observed. Histological findings demonstrated that apoptosis occurred on the cortex 1 day after treatment, peaking in the X-ray group. The cells of the combined group showed a moderate caspase-3 staining compared to the X-ray group. There was a trend towards a lower body weight of the X-ray group following irradiation compared to either no-irradiation or rats of the combined group, although there was no significant difference in the average weight between the combined group and irradiated rats. Mild swelling of the capillary endothelial cells in the irregular lumen was observed the combined group, whereas the X-ray group showed a severe structural disorder. In conclusion, VPA supplementation during radiotherapy may be beneficial for radioprotection following WBI by reducing normal brain cell injury.
Comparison of neuroprotective effects in ischemic rats with different hypothermia procedures.

Abstract:Objective: The neuroprotective effect of hypothermia has long been recognized. The aim of this work was to compare the neuroprotective effect of systemic, head and local vascular cooling hypothermia procedures in ischemic rats. Methods: Stroke in Sprague-Dawley rats (n=64) was induced by a 3 hour right middle cerebral artery occlusion using an intraluminal filament. Before reperfusion, ischemic animals (n=16 in each group) received hypothermia (systemic, head or local vascular) or no treatment. Brain temperature, infarction volume (n=8 in each group) and functional outcome (n=8 in each group) were compared. Results: Regarding brain temperature, vascular cooling significantly reduced the temperature of ischemic territory in cortex from 37.2 +/- 0.1 to 33.4 +/- 0.4 degrees C and in striatum from 37.5 +/- 0.2 to 33.9 +/- 0.4 degrees C within 5 minutes. This hypothermic condition remained for up to 60 minutes after reperfusion. However, systemic cooling reduced brain temperature at a similar level for six times longer. In the head cooling group, the target temperature was reached in 15 minutes, but returned to normal within 5 minutes. Although all hypothermia procedures induced neuroprotection, ischemic rats with vascular cooling showed significantly (p<0.001) better neuroprotection with 10.7 +/- 2.6% infarction, compared to 54.2 +/- 1.9% (no treatment), 37.1 +/- 1.0% (head cooling) and 29.1 +/- 3.4% (systemic cooling). Significantly (p<0.001) better effects on motor function were also detected in vascular cooling groups at 14 and 28 days. Conclusion: Vascular cooling appears to be the most effective in reducing infarct volume and improving functional outcome than the other two hypothermia methods in a rat ischemia/reperfusion model.
The dose-effectiveness of intranasal VEGF in focal cerebral ischemia/reperfusion injury of rats

Abstract:Objective To study the dose-effectiveness of intranasal(IN) vascular endothelial growth factor(VEGF)in focal cerebral ischemia/reperfusion injury of rats.Methods Sprague-Dawley rats were randomized into four groups as IN low(100 μg/mL),IN middle(200 μg/mL) and IN high(500 μg/mL) VEGF-treated group,and IN saline-treated group(n=12),given recombinant human VEGF or saline intranasally.Focal cerebral ischemia/reperfusion was induced by transient(90 minutes) middle cerebral artery occlusion(MCAO) method.Behavioral neurological deficits were assessed 1 d,7 d and 14 d after the onset of MCAO.Rats were euthanized at 14 d,the brain sections were stained and an image analysis system was used to calculate the infarct volume and calculated the number of cerebral vessels.Results Compared to IN saline,infarct volume after MCAO significantly reduced(P<0.01) in rats which received the middle(200 μg/mL) dose of IN VEGF.VEGF with middle dose significantly improved behavioral recovery(P<0.01).No significant differences in behavioral recovery and infarct volume after MCAO were seen between the IN saline group and in low and high VEGF-treated group(P>0.05).Compared to IN saline,VEGF with middle and high dose significantly increased the number of cerebral vessels(P<0.01).No significant differences in the number of cerebral vessels were seen between the IN saline group and low VEGF-treated group(P>0.05).Conclusion The middle dose(200 μg/mL)of IN VEGF was most effective at reducing infarct volume,improving behavioral recovery and increasing the number of vessels,which can be used in the following treatments to further evaluate the effect of VEGF.
Effect of vinpocetine against acrylamide‐induced nephrotoxicity in rats

Abstract:Abstract Vinpocetine (VIN) is a synthetic drug derived from the natural alkaloid vincamine. The antioxidation and anti‐inflammation effects of VIN allow it to be used for multiple therapeutic purposes. So, the research aims to discover the possibility of using VIN to improve the nephrotoxicity of acrylamide (ACR). Twenty‐four male albino rats were used in the trial: rats in the control group received 0.5 mL of oral saline, rats in the VIN group received an oral dose of VIN (5 mg/kg), rats in the ACR group received an oral dose of ACR (38.27 mg/kg), and rats in the VIN + ACR group received VIN and then ACR 1 h later. Rat blood and kidneys were collected 10 days after the experiment began to assess biochemical parameters and to examine both renal histopathological and immunohistochemistry. The ACR‐treated rats showed high levels of serum kidney function biomarkers (creatinine, urea, and uric acid), serum protein biomarkers (total protein, albumin, and globulin), renal kidney injury molecule (KIM)−1, renal malondialdehyde (MDA), and renal caspase‐3 immunoexpression. Moreover, ACR lowed both renal superoxide dismutase (SOD) activity and renal glutathione (GSH) level and caused renal histological alterations. While administration of VIN improved serum kidney function biomarkers, serum protein biomarkers, renal KIM‐1, renal oxidative stress biomarkers (MDA, SOD, and GSH), renal caspase‐3 immunoexpression, and renal histological alterations induced by ACR. The study confirmed the ability of VIN to reduce the nephrotoxic effects of ACR, which was evident through the results of biochemical parameters and histological and immunohistochemical examinations of the kidney tissues.

Abstract:OBJECTIVE:To conduct a systematic evaluation on the efficacy and safety of vinpocetine injection for acute ischemic stroke.METHODS:The randomized controlled trials(RCTs) on vinpocetine for acute ischemic stroke was retrieved.Data collection and quality of the included RCTs were performed using Cochrane systematic evaluation method with qualified RCTs screened,then meta-analyses were conducted by using RevMan 5.0 software.RESULTS:A total of 15 RCTs involving 1 526 cases were included with 770 cases in the treatment group and 756 in the control group.The baseline data of each included RCT have comparability.Randomization was reported in all yet random digits table was used in only 1 case.The blinding method and allocation concealment were unreported.The studies in which there was report of endpoint indicator were failed to be retrieved.Mata-analysis on surrogate indicators demonstrated that there were statistical significances between vinpocetine injection and other medications or conventional therapy in effective rate(OR=2.58,95%CI=1.96-3.40) and in the improvement of neurologic impairment scores(WMD=1.58,95%CI=1.13-2.02).The adverse drug reaction was extremely less frequent and mild thus vinpocetine injection was safe to use.CONCLUSION:The systematic evaluation on vinpocetine injection revealed that vinpocetine is quite effective for acute ischemic stroke and it can improve the nerve dysfunction.This conclusion should be treated with caution because of the poor quality of the included RCTs which may impact the power of the proof.More randomized double-blind control trials with high quality are needed to provide high-quality evidence.

Control Study of CT Perfusion Imaging and Histology of Rat VX2 Implanted Cerebral Tumor: Before and after Radiation Therapy

Abstract:Objective With correlative study of CT perfusion parameter change and tumor micro-vascular density change of rat VX2 implanted cerebral tumor before and after radiation therapy, this study was supposed to provide experimental basis for evaluation of CT perfusion imaging in measurement of effect assessment of radiation therapy of brain tumor. Methods Nine rats with VX2 tumor were involved, and all rats were divided into treatment group (n=4) and non-treatment group (n=5). The rats in treatment group underwent brain directional radiation therapy. CT enhanced and perfusion scans were performed one week both before and after the radiation for the two groups. Brain tissue was collected to perform HE stain and CD34 immuno-histo-chemical stain after all rats were sacrificed to death. Significant analysis of CT perfusion parameters was applied between before and after the radiation therapy of treatment and non-treatment group respectively. Correlation study was also made between CBV value and MVD value of the tumor region in treatment group after radiation therapy and in the non-treatment group as well. Results All the CT perfusion parameters including CBF, CBV and PS decreased after radiation in treatment group with statistic significance (P0.05). All the CT perfusion parameters had no obvious decrease twice in non-treatment group(P0.05). Significant correlation was observed between CBV and MVD value after CD34 immuno-histo-chemical stain in tumor region(r=0.896,P0.01). Conclusion CT perfusion imaging is able to reflect hemodynamic changes of brain tumors after radiation therapy. Cerebral blood volume value can be taken as an index to the micro-vascular of tumor in-vivo as it has significant correlation with microvascular density.
Vinpocetine mitigates DMH-induce pre-neoplastic colon damage in rats through inhibition of pro-inflammatory cytokines

Abstract:Colorectal cancer (CRC) is currently recognized as the third most prevalent cancer worldwide. Vinpocetine is a synthetic derivative of the vinca alkaloid vincamine. It has been found effective in ameliorating the growth and progression of cancerous cells. However, its pharmacological effect on colon damage remains elusive. Hence, in this study, we have shown the role of vinpocetine in DMH-induced colon carcinogenesis. At first, male albino Wistar rats were administered with DMH consistently for four weeks to induce pre-neoplastic colon damage. Afterward, animals were treated with vinpocetine (4.2 and 8.4 mg/kg/day p.o.) for 15 days. Serum samples were collected to assess the physiological parameters, including ELISA and NMR metabolomics. Colon from all the groups was collected and processed separately for histopathology and western blot analysis. Vinpocetine attenuated the altered plasma parameters; lipid profile and showed anti-proliferative action as evidenced by suppressed COX-2 stimulation and decreased levels of IL-1β, IL-2, IL-6, and IL-10. Vinpocetine is significantly effective in preventing CRC which may be associated with its anti-inflammatory and antioxidant potential. Accordingly, vinpocetine could serve as a potential anticancer agent for CRC treatment and thus be considered for future clinical and therapeutic research.

Abstract:Abstract Vinpocetine is considered as neuroprotectant drug and used for treatment of brain ischemia and cognitive deficiencies for decades. A number of enzymes, channels and receptors can bind vinpocetine, however the mechanisms of many effects’ are still not clear. The present study investigated the effects of vinpocetine from the mitochondrial bioenergetic aspects. In primary brain capillary endothelial cells the purinergic receptor-stimulated mitochondrial Ca 2+ uptake and efflux were studied. Vinpocetine exerted a partial inhibition on the mitochondrial calcium efflux. In rodent brain synaptosomes vinpocetine (30 μM) inhibited respiration in uncoupler stimulated synaptosomes and decreased H 2 O 2 release from the nerve terminals in resting and in complex I inhibited conditions, respectively. In isolated rat brain mitochondria using either complex I or complex II substrates leak respiration was stimulated, but ADP-induced respiration was inhibited by vinpocetine. The stimulation of oxidation was associated with a small extent of membrane depolarization. Mitochondrial H 2 O 2 production was inhibited by vinpocetine under all conditions investigated. The most pronounced effects were detected with the complex II substrate succinate. Vinpocetine also mitigated both Ca 2+ -induced mitochondrial Ca 2+ -release and Ca 2+ -induced mitochondrial swelling. It lowered the rate of mitochondrial ATP synthesis, while increasing ATPase activity. These results indicate more than a single mitochondrial target of this vinca alkaloid. The relevance of the affected mitochondrial mechanisms in the anti ischemic effect of vinpocetine is discussed.

Abstract:Manganese (Mn) is required for many essential biological processes as well as in the development and functioning of the brain. Extensive accumulation of Mn in the brain may cause central nervous system dysfunction known as manganism, a motor disorder associated with cognitive and neuropsychiatric deficits similar to parkinsonism. Vinpocetine, a synthetic derivative of the alkaloid vincamine, is used to improve the cognitive function in cerebrovascular diseases. It possesses antioxidant and antiinflammatory properties. The present work was designed to explore the potential neuroprotective mechanisms exerted by vinpocetine in the Mn-induced neurotoxicity in rats. Rats were allocated into four groups. First group was given saline. The other three groups were given MnCl2; two of them were treated with either l-dopa, the gold standard antiparkinsonian drug, or vinpocetine. Rats receiving MnCl2 exhibited lengthened catalepsy duration in the grid and bar tests, motor impairment in the open-field test and short-term memory deficit in the Y-maze test. Additionally, histological examination revealed structural alterations and degeneration in different brain regions. Besides, striatal monoamines and mitochondrial complex I contents were declined, apoptotic biomarker caspase-3 expression and acetylcholinesterase activity were elevated. Moreover, oxidative stress and inflammation were detected in the striata. l-dopa or vinpocetine exerted protective effects against MnCl2-induced neurotoxicity. It could be hypothesized that modulation of monoamines, upregulation of mitochondrial complex I, antioxidant, antiinflammatory, and antiapoptotic activities are significant mechanisms underlying the neuroprotective effect of vinpocetine in the Mn-induced neurotoxicity model in rats.

Effects of delayed puerarin treatment in long-term neurological outcomes of focal ischemic stroke in rats

Abstract:OBJECTIVE: The present study aimed at investigate the therapeutic effects of delayed puerarin treatment in neurological outcomes after middle cerebral artery occlusion (MCAO) in rats.MATERIALS AND METHODS: Male Wistar rats were subjected to MCAO for 120 min followed by reperfusion for 14 days. Puerarin (0, 50, 100, 200 mg/kg, intra-peritoneally) was administered at 24 h after stroke onset and repeated daily for 14 days. Neurological deficits were evaluated at 1, 4, 7, 14 days after stroke. Brain infarct volume and peri-infarct context vessel density were examined at 14 days after stroke.RESULTS: Puerarin significantly improved neurological functions up to 14 days after stroke and decreased the infarct volume with doses of 50 mg/kg and 100 mg/kg compared with saline controls. Puerarin treatment also significantly increased peri-infarct context vessel density at 14 days after stroke.CONCLUSIONS: Delayed treatment of puerarin initiated at 24 h after stroke is beneficial with improved long-term neurological outcomes and reduced infarction volume in focal ischemic stroke in rats. Enhanced vascular remodeling by puerarin might at least partially contribute to its beneficial effects.
