Abstract:Antibiotic-resistant bacteria pose considerable risks to global health, particularly through transmission in the food chain. Herein, we developed the artificial intelligence-driven quantification of antibiotic-resistant bacteria in food using a color-encoded multiplex hydrogel digital loop-mediated isothermal amplification (LAMP) system. The quenching of unincorporated amplification signal reporters (QUASR) was first introduced in multiplex digital LAMP. During amplification, primers labeled with different fluorophores were integrated into amplicons, generating color-specific fluorescent spots. While excess primers were quenched by complementary quenching probes. After amplification, fluorescent spots in red, green, and blue emerged in hydrogels, which were automatically identified and quantified using a deep learning model. Methicillin-resistant Staphylococcus aureus and carbapenem-resistant Escherichia coli in real fruit and vegetable samples were also successfully detected. This artificial intelligence-driven color-encoded multiplex hydrogel LAMP offers promising potential for the digital quantification of antibiotic-resistant bacteria in the food industry.

Abstract:Some veterinary drug residues in food products and environment have been widely regarded as severe threats to human health. Rapid and simultaneous detection methods are crucial to monitor and control veterinary drug usage. Here, we propose a fluorescence biosensor utilizing immunomagnetic beads(IMBs) and quantum dots(QDs) for the rapid and simultaneous detection of 1-adamantylamine(ADA), enrofloxacin(ENR) and tilmicosin(TIL) in raw chicken meat. A pretreatment method using sodium phosphotungstate–magnesium as extraction reagent was developed to simultaneously extract ADA, ENR and TIL from chicken meat with minor interference in background or response. By adding the IMBs modified with three types of antibodies and the QD-antigens modified with three types of BSA-antigens to sample, IMBs competitively conjugated to target antigens in a sample or QD-antigens. After magnetic separation, the residual QD-antigens were adopted to collect signals using fluorescence spectroscopy. Using QDs with well separated emission peaks, the detection of one type of targets was minorly interfered by the others. Under the optimum conditions, the biosensor exhibited the limit of detection of 0.96,3.32, and 3.17 ng/mL for ADA, ENR and TIL in chicken samples, respectively, as well as good specificity. Due to the way of direct collection of signals in extracts, the tedious and complicated multiple magnetic separation and signal amplification procedures in conventional methods were avoided, thus the procedures were significantly simplified, and the reduction of the operation time of 30 min for sample pretreatment and 40 min for detection part was achieved. The biosensor might be promising in the rapid, in-field and sensitive screening of multiple veterinary drugs to ensure agriculture and food safety.
Core-satellite-structured magnetic nanozyme enables the ultrasensitive colorimetric detection of multiple drug residues on lateral ﬂow immunoassay

Abstract:Background: Excessive use of veterinary drugs causes severely environmental pollution and agricultural pollution, and poses great threat to human health. A simple method for the rapid, highly sensitive, and on-site monitoring of veterinary drug residues in complex samples remains lacking. Results: In this study, we propose a catalytically enhanced colorimetric lateral ﬂow immunoassay (LFA) based on a novel core-satellite-structured magnetic nanozyme (Fe-Au@Pt) that can simultaneously and quantitatively detect three common veterinary drugs, namely, gentamicin (GM), streptomycin (STR), and clenbuterol (CLE), within a short testing time (<30 min). The Fe-Au@Pt nanozyme was simply prepared through the self-assembly of numerous Au@Pt nanoparticles on a large Fe3O4 core via electrostatic adhesion, which exhibited the advantages of high peroxidase-like activity, strong magnetic responsiveness, and multiple catalytic sites. Under the dual-signal amplification effect of magnetic enrichment and catalytic enhancement, the proposed nanozyme-LFA allowed the multiplex detection of STR, CLE, and GM with detection limits of 10.1, 6.3, and 1.1 pg/mL, respectively. Significance: The developed Fe-Au@Pt-LFA achieves direct, simultaneous, and accurate detection of three target drugs in food samples (honey, milk, and pork). The proposed assay shows great potential for application in the real-time monitoring of small-molecule pollutants in complex environment.

Abstract:<p>Carbon dots have been rapidly developed as a kind of luminescent nanomaterials. However, methods for the synthesis of long-wavelength-emissive carbon dots are still not mature enough. Meanwhile, developing a fast and highly sensitive method to detect residual veterinary drugs in food is particularly significant for food safety. Herein, we report the facile fabrication of red-emissive carbon dots (R-CDs) for ratiometric sensing of veterinary drugs. With use of sorbitol and o-phenylenediamine as precursors, R-CDs was prepared via a one-pot solvothermal approach. The resulting R-CDs show mainly excitation-independent and dual-emissive photoluminescence (PL) located at 597 and 645 nm. Meanwhile, the PL quantum yield (QY) of R-CDs without further surface treatment is 33% under 540 nm excitation. Interestingly, the R-CDs exhibit fast fluorescence response to malachite green oxalate (MG oxalate), 1-aminohydantoin hydrochloride (AHD) and pefloxacin, which are abused widely and retained easily in animals. Ratiometric sensing of these veterinary drugs were successfully performed taking the advantage of dual-emissive PL property; the detection limits of MG oxalate, AHD and pefloxacin were 0.79, 0.25 and 0.85 μM, respectively.</p>

Abstract:The broad spectrum detection of veterinary drugs is very important for rapid and large-scale safe screen of animal-derived foods. Metal-organic frameworks (MOFs), as a kind of emerged functional porous materials are quite promising in the chemical sensing and molecular detection. In this work, we report the high-performance broad spectrum detection of 15 commonly-used veterinary drugs through the fluorescence quenching in a newly-designed chemically stable Al-based MOF, Al3(μ3-O)(OH)(H2O)2(PPTTA)3/2 (BUT-22). To the best of our knowledge, this is the first systematic investigation for the application of MOFs in the detection/sensing of veterinary drugs through fluorescence quenching method. The quenching efficiencies of the tested veterinary drugs on BUT-22 are all beyond 82%, and the limits of detection (LOD) are low at parts per billion (ppb) levels. Interestingly, BUT-22 also enables the selective detection of nicarbazin (NIC) through the clearly-observed red shift of its maximum fluorescence emission wavelength. Moreover, the fluorescence quenching mechanism was explored with the help of theoretical calculations. Our work indicates that MOFs are favorable materials for the detection of veterinary drugs, being potentially useful in monitoring drug residues of animal-derived foods.

Abstract:The development of efficient, rapid, portable, and accurate analysis of veterinary drug residues in food matrices is in great demand for food safety assessment. Here, we have developed a smartphone-integrated platform for fluorometric quantification of metronidazole (MNZ) residues and constructed a sensor array for discrimination of different nitroimidazole antibiotics (NIIMs). Multicolor CDs (B-CDs, C-CDs, Y-CDs, and R-CD) were prepared and showed different fluorescence response to MNZ. The fluorescence of C-CDs was quenched Because of the inner filter effect (IFE) between the C-CDs and MNZ, while that of R-CDs was enhanced due to the passivation of surface defects by MNZ. Based on the response pattern, the fluorometric quantification of MNZ based on the fluorescence images of C-CD + R-CD system (R/G values) was achieved with a low detection limit of 0.45 μM. By designing a smartphone-integrated platform, the analysis can be completed within 20 min. In addition, a fluorescence sensor array based C-CDs and R-CDs was also developed. The unique fingerprint of each NIIMs was obtained by linear discriminant analysis (LDA) of the response patterns, indicating an effective discrimination of five NIIMs. Moreover, the platform was used for quantification of MNZ in food samples and the recoveries were within 84.0-106.3 % with relative standard deviations 1.2-10.2 %. Therefore, the proposed method shows great potential as a universal platform for rapid detection of veterinary drug residues.

Abstract:Levofloxacin (LVFX) as a representative drug of quinolone antibiotics is widely used in clinical, and its residues enriched in water bodies and sideline products seriously damage human health. It is imperative to develop a real-time/on-site sensing method for monitoring residual antibiotics. Here, we report a portable sensing platform by utilizing a composite fluorescent nanoprobe constructed by the cerium ions (Ce 3+ ) coordination functionalized CdTe quantum dots (QDs) for the visual and quantitative detection of LVFX residues. This fluorescent probe provides a distinct color variation from red to green, which shows a good linear relationship to LVFX residues concentrations in the range of 0-6.0 μmol/L with a sensitive limit of detection (LOD) of 16.3 nmol/L. The smartphone platform with Color Analyzer App installed, which could accomplish quantified detection of LVFX in water, milk, and raw pork with a LOD of 27.9 nmol/L. The facile sensing method we proposed realizes rapid visualization of antibiotics residual in the environment and provides a practical application pathway in food safety and human health.

Multicomponent mesofluidic system for the detection of veterinary drug residues based on competitive immunoassay

Abstract:An automated multicomponent mesofluidic system (MCMS) based on biorecognitions carried out on meso-scale glass beads in polydimethylsiloxane (PDMS) channels was developed. The constructed MCMS consisted of five modules: a bead introduction module, a bioreaction module, a solution handling module, a liquid driving module, and a signal collection module. The integration of these modules enables the assay to be automated and reduces it to a one-step protocol. The MCMS has successfully been applied toward the detection of veterinary drug residues in animal-derived foods. The drug antigen-coated beads (varphi250 microm) were arrayed in the PDMS channels (varphi300 microm). The competitive immunoassay was then carried out on the surface of the glass beads. After washing, the Cy3-labeled secondary antibody was introduced to probe the antigen-antibody complex anchored to the beads. The fluorescence intensity of each bead was measured and used to determine the residual drug concentration. The MCMS is highly sensitive, with its detection limits ranging from 0.02 (salbutamol) to 3.5 microg/L (sulfamethazine), and has a short assay time of 45 min or less. The experimental results demonstrate that the MCMS proves to be an economic, efficient, and sensitive platform for multicomponent detection of compound residues for contamination in foods or the environment.
Quantum confined peptide assemblies in a visual photoluminescent hydrogel platform and smartphone-assisted sample-to-answer analyzer for detecting trace pyrethroids

Abstract:Quantum confinement (QC) effect-related materials have been extensively studied as photoluminescent probes for agricultural, food, and environmental analyses, with the advantage of simple-to-synthesize, reusable, nontoxic, and environmentally friendly. Herein, we propose a strategy to dimerize aromatic cyclo-dipeptides, namely cyclo-ditryptophan (cyclo-WW), cyclo-diphenylalanine (cyclo-FF), and cyclo-dihistidine (cyclo-HH), into quantum dots as basic building blocks for the self-assembly of QC supramolecular structures with excellent photoluminescent properties in aqueous solutions. In particular, through coordination with Zn(II), the bandgap can be tuned to change the photo-absorption and luminescence properties of the cyclo-dipeptide-based QC assemblies. The fluorescence quantum yield of cyclo-WW+Zn(II) was 16.9%. Such a good luminous effect makes it applicable to the detection of LC. A good linear relationship between fluorescence response of cyclo-WW+Zn(II) and LC concentration was observed in the range of 5–350 μg/L, with a low limit of detection of 2.9 μg/L and good spiked recovery of 90.72%–104.3%. A visual platform using the cyclo-WW+Zn(II)-based photoluminescent hydrogel and smartphone-assisted sample-to-answer analyzer were developed, which showed good responsiveness to LC. The developed fluorescence method, validated using traditional HPLC, is a biocompatible alternative for the rapid detection of trace pollutants with the advantages of portability and simple operation.

Abstract:A bead-based microfluidic device was developed and demonstrated to achieve rapid and sensitive enzyme-linked immunosorbent assay ( ELISA) with quantum dots as the labeling fluorophore for virus detection. In comparison to standard ELISA performed on the same virus, the minimal detectable concentration of the target virus was improved from 360 to 22 ng mL(-1), the detection time was shortened from >3.25 h to <30 min, and the amount of antibody consumed was reduced by a factor of 14.3.

Abstract:A photoelectrochemical immunosensor based on multi-electrode array was developed for simultaneous and sensitive determination of veterinary drug residues. In this system, poly(dimethyldiallylammonium chloride) (PDDA), Au nanoparticles (Au NPs) and thioglycolic acid (TGA)-capped CdS quantum dots (QDs) were layer-by-layer assembled onto the home-made Au electrode array. The assembling process of the (CdS/PDDA/Au NPs/PDDA)n multilayer was characterized by electrochemical impedance spectroscopy. And then the antibodies for clenbuterol (CB), ractopamine (RAC) and chloramphenicol (CAP) were covalently immobilized onto the Au electrode array by 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide (EDC) coupling reaction, respectively. The concentrations of CB, RAC and CAP were measured based on the photoelectrochemical effects of CdS QDs. Under the optimal conditions, the limits of detection (LOD) for CB, RAC and CAP were 25, 50 and 2.2 pg/mL (3?), respectively, with acceptable recovery over the range of 95.40%105.5% in pig liver samples. All results indicate that the immunosensor array system has potential application for practical, effective and high throughput analysis of veterinary drugs residues.

Abstract:Oxytetracycline (OTC) is a widely used antibiotic in veterinary practices because of its broad-spectrum antibacterial activity and low cost. However, drug abuse has triggered adverse effects on human health, which brings a growing demand for on-site diagnosis of OTC residues in animal-derived foods. In this study, we demonstrate the combined use of interdigital electrode microarray and electrochemiluminescence (ECL) imaging for parallel multiplex measurement of OTC. Well-ordered and vertically aligned mesoporous silica nanochannels modified on microarray substrate could exert a strong electrostatic attraction to the ECL luminophores and accelerate their mass transport to generate enhanced ECL signal. The performance of the integrated ECL microarray sensor was fully validated with respect to linearity (0.5 to 50 μM), sensitivity (limit of detection 0.26 μM), accuracy (recovery rate between 96.78% and 106.1%), low operating sample volume (480 nL), short assay time (1.5 min), and antifouling ability toward complex media. The multiplex microarray platform can serve as a simple, fast, and low-cost tool for the detection of a wide spectrum of antibiotics in the field of food safety.

Abstract:In the evolving field of food safety, rapid and precise detection of antibiotic residues is crucial. This study aimed to tackle this challenge by integrating advanced inkjet printing technology with sophisticated microfluidic paper-based analytical devices (µPADs). The µPAD design utilized "green" quantum dots synthesized via an eco-friendly hydrothermal method using green white mulberry leaves as the carbon source, serving as the key fluorescent detection material. The action mechanism involved a photoinduced electron transfer system using red carbon dots (CDs) as electron donors and blue CDs combined with two-dimensional layered molybdenum disulfide (MoS2) nanosheets as electron acceptors. This system could quickly detect antibiotics within 10 min in pork and water samples, demonstrating high sensitivity and recovery rates: 6.5 pmol/L at 99.75%-110% for sulfadimethoxine, 3.3 pmol/L at 99%-105% for sulfamethoxazole, and 8.5 pmol/L at 98.5%-105% for tetracycline. It achieved a relative standard deviation under 5%, ensuring reliability and reproducibility. The fabricated sensor offered a promising application for the rapid and efficient on-site detection of antibiotic residues in food.
Laser-Printed Paper-Based Microfluidic Chip Based on a Multicolor Fluorescence Carbon Dot Biosensor for Visual Determination of Multiantibiotics in Aquatic Products

Abstract:Excessive use of antibiotics in aquaculture severely endangers human health and ecosystems, which has raised significant concerns in recent years. However, conventional laboratory-based approaches regularly required time or skilled manpower. Herein, we propose a point-of-care-testing (POCT) biosensor detection device for the simultaneous determination of multiantibiotics without complex equipment or professional operators. A laser-printed paper-based microfluidic chip loaded with multicolor fluorescence nanoprobes (mCD-μPAD) was developed to rapidly detect sulfamethazine (SMZ), oxytetracycline (OTC), and chloramphenicol (CAP) on-site. These "fluorescence off" detection probes composed of carbon dots (CDs) conjugated with aptamers (donor) and MoS2 nanosheets (acceptor) (CD-apt-MoS2) were based on Förster resonance energy transfer. Upon the addition of target antibiotics, the significantly recovered fluorescence signal on the μPAD can be sensitively perceived by employing a 3D-printed portable detection box through a smartphone. Under optimal conditions, this μPAD allowed for a rapid response of 15 min toward SMZ, OTC, and CAP with considerable sensitivities of 0.47, 0.48, and 0.34 ng/mL, respectively. In shrimp samples, the recoveries were 95.2-101.2, 96.4-105, and 96.7-106.1% with RSD below 6%. This paper-based sensor opens an avenue for on-site, high-throughput, and rapid detection methods and can be widely used in POCT in food safety.

Abstract:The sensitive on-site monitoring of multiple agricultural drugs in complex real media is critical to human health but remains a challenge. In this paper, we reported the design of film-like dual-mode nanolabel (MTQD), using molybdenum disulfide (MoS 2 ) nanosheet-loaded numerous quantum dots (QDs) to generated co-enhanced colorimetric-fluorescence signals, and demonstrated its powerful capabilities in competitive lateral flow immunoassay (LFA) for flexible, ultrasensitive, and multiplex detection of four kinds of common agricultural drugs. Three layers of red and green QDs were continuously coated onto the surface of MoS 2 to suppress the inner filter effect of two-dimensional nanomaterials effectively and greatly improve the specificity of the LFA system for multitarget analysis. The dual-mode LFA combined with a low-cost dual-fluorescence reading instrument can flexibly and simultaneously monitor two pesticides (clothianidin, carbendazim) and two veterinary drugs (kanamycin, chloramphenicol) in complex food and experimental samples with low detection limits of 0.1 ng/mL level in the observation of colorimetric signals and showed at least a 46-fold improvement in sensitivity in the sensing of fluorescence signals. This colorimetric dual-fluorescence platform greatly broadens the application range of existing LFA techniques without increasing testing time, thus showing great application prospects for the point-of-care testing of small-molecule pollutants in different situations.

Abstract:Florfenicol (F), an antimicrobial agent exclusive to veterinary use within the chloramphenicol class, is extensively applied as a broad-spectrum remedy for animal diseases. Despite its efficacy, concerns arise over potential deleterious residues in animal-derived edibles, posing threats to human health. This study pioneers an innovative approach, introducing a quantum dot fluorescence-based immunoassay (FLISA) for the meticulous detection of F residues in animal-derived foods and feeds. This method demonstrates heightened sensitivity, with a detection limit of 0.3 ng/mL and a quantitative detection range of 0.6-30.4 ng/mL. Method validation, applied to diverse food sources, yields recoveries from 90.4 % to 109.7 %, featuring RSDs within 1.3 % to 8.7 %, the results showed high consistency with the national standard HPLC-MS/MS detection method. These findings underscore the method's accuracy and precision, positioning it as a promising tool for swift and reliable F residue detection, with substantial implications for fortifying food safety monitoring.
A novel fluorescence platform for specific detection of tetracycline antibiotics based on [MQDA-Eu3+] system

Abstract:Tetracycline antibiotics (TCs) are extensively used in clinical medicine, animal husbandry, and aquaculture because of their cost-effectiveness and high antibacterial efficacy. However, the presence of TCs residues in the environment poses risks to humans. In this study, an inner filter effect (IFE) fluorescent probe, 2,2'-(ethane-1,2-diylbis((2-((2-methylquinolin-8-yl)amino)-2-oxoethyl)azanediyl))diacetic acid (MQDA), was developed for the rapid detection of Eu3+ within 30 s. And its complex [MQDA-Eu3+] was successfully used for the detection of TCs. Upon coordination of a carboxyl of MQDA with Eu3+ to form a [MQDA-Eu3+] complex, the carboxyl served as an antenna ligand for the effective detection of Eu3+ to intensify the emission intensity of MQDA via "antenna effect", the process was the energy absorbed by TCs via UV excitation was effectively transferred to Eu3+. Fluorescence quenching of the [MQDA-Eu3+] complex was caused by the IFE in multicolor fluorescence systems. The limits of detection of [MQDA-Eu3+] for oxytetracycline, chlorotetracycline hydrochloride, and tetracycline were 0.80, 0.93, and 1.7 μM in DMSO/HEPES (7:3, v/v, pH = 7.0), respectively. [MQDA-Eu3+] demonstrated sensitive detection of TCs in environmental and food samples with satisfactory recoveries and exhibited excellent imaging capabilities for TCs in living cells and zebrafish with low cytotoxicity. The proposed approach demonstrated considerable potential for the quantitative detection of TCs.
Quantum dot-based fluorescence-linked immunosorbent assay for the rapid detection of lomefloxacin in animal-derived foods

Abstract:Lomefloxacin (LMF), a third-generation fluoroquinolone antibacterial agent, is often used to treat bacterial and mycoplasma infections. However, due to its prolonged half-life and slow metabolism, it is prone to residues in animal-derived foods, posing a potential food safety risk. Therefore, it is particularly urgent and important to establish a method for detecting lomefloxacin. In this study, direct and indirect competitive fluorescence-linked immunosorbent assay (dc-FLISA and ic-FLISA) based on quantum dots (QDs) was established for the detection of LMF. As for dc-FLISA, the half-maximal inhibitory concentration (IC50) and limit of detection (LOD) were 0.84 ng/mL, 0.04 ng/mL, respectively, the detection ranges from 0.08 to 9.11 ng/mL. The IC50 and LOD of ic-FLISA were 0.43 ng/mL and 0.03 ng/mL, respectively, meanwhile the detection ranges from 0.05 to 3.49 ng/mL. The recoveries of dc-FLISA and ic-FLISA in animal-derived foods (milk, fish, chicken, and honey), ranged from 95.8% to 105.2% and from 96.3% to 103.4%, respectively, with the coefficients of variation less than 8%. These results suggest that the dc-FLISA and ic-FLISA methods, which are based on QD labelling, are highly sensitive and cost-effective, and can be effectively used to detect LMF in animal-derived foods.

Abstract:The biocompatible semiconductor quantum dots (QDs) have unique photophysical properties, which provide important advantages over organic dyes and lanthanide probes in fluorescence labeling applications. In this work, multicolor quantum QD-encoded microspheres have been prepared via the layer-by-layer (LbL) assembly approach. Polystyrene microspheres of 3mum diameter were used as templates for the deposition of different sized CdTe QDs/polyelectrolyte multilayers via electrostatic interactions. Two kinds of biofuntional multicolor microspheres with two different antibodies, anti-human IgG and anti-rabbit IgG were prepared. Human IgG and rabbit IgG can be detected as target antigens in the multiplexed fluoroimmunoassays. Furthermore, a novel microfluidic on-chip device was developed to detect two kinds of antigen-conjugated multicolor QD-encoded microspheres; the microspheres can be distinguished from each other based on their fluorescence signals.

Abstract:A simple smartphone-integrated ratiometric fluorescent sensing system for visual determination of copper ions (Cu2+) and fluoroquinolones (FQs) was developed based on carbon dots (CDs) which were synthesized through the high-temperature pyrolysis of citric acid. In this system, with the fluorescence resonance energy transfer effect between CDs and 2,3-diaminophenazine (oxOPD), the detection of Cu2+ and ciprofloxacin (CIP, an example for FQs) was realized. Cu2+ catalyzes the oxidation of OPD to form oxOPD with yellow fluorescence, resulting in the quenching of CDs. In addition, CIP can inhibit the catalytic activity of Cu2+ and induce the recovery of CDs fluorescence. Under the excitation of 400 nm, the changes of CDs fluorescence at 472 nm and oxOPD fluorescence at 556 nm were monitored. The detection results showed that the sensing system exhibited good selectivity and sensitivity to Cu2+ and CIP with the limit of detection of 2.32 × 10−8 mol L−1 and 0.2 ng mL−1, respectively. In addition, a smartphone was developed as a portable analyzer to capture the change of fluorescence color and quickly analyze the concentration of Cu2+ and CIP. The proposed smartphone-based sensing platform has satisfactory sensitivity, and it has application prospects for detecting Cu2+ and FQs in food safety monitoring.
