Abstract:The pollen development-related gene msLTP from Brassica campestris ssp. chinensis belongs to the lipid transfer protein (LTP) gene family. The promoter of the msLTP gene was isolated by thermal asymmetric interlaced PCR (TAIL-PCR). The functional elements of the promoter (named Bc15) were analysed. Some anther/pollen-specific elements, such as the GGTT box and the GTGA box, were identified in the sequence. In an attempt to confirm the promoter activity, a series of 5'-truncated constructs was fused with the GUS reporter gene to functionally analyse the key regulatory regions of the promoter. Transient expression analysis showed high GUS activity in onion epidermal cells containing the region from -731 to -1 bp. The truncated construct bearing the -731 to -1 bp fragment was stably introduced into Arabidopsis. Histochemical analysis of the transgenic plants showed distinct and specific GUS expression at different stages of anther/pollen development. Transverse sections of flower buds further indicated that the Bc15 promoter activates reporter gene expression in the anther specifically. These findings elucidated the molecular mechanisms of LTP gene regulation during pollen development, and suggested a potential application of the Bc15 promoter in engineering male sterility for hybrid production.

Abstract:The promoter is an important element of gene expression,and tissue specific promoters are very useful in gene engineering.A promoterless UidA containing plasmid derived from pPLGUS was transformed into tritordeum.Histochemical analysis of various tissues in transgenic tritordeum was carried to examine tissue specific expression of GUS activity.A transformant line with anther-specific GUS expression was successfully labeled and isolated.PCR method was used to isolate this anther-specific promoter with DNA extracted from leave as template,using primer pair of rice anther specific promoter,primer P1 as forward primer,UidA gene specific sequence as reverse primer.By sequencing and analyzing the amplified DNA fragment from PCR reaction,a part of UidA gene and a flanking sequence were obtained.Some essential promoter elements were found in this sequence.Therefore the fragment obtained was believed to be a part of anther specific promoter.
The Inductive Activation of the Promoter of Pib Gene

Abstract:A 5.7 kb putative promoter region of pib gene was isolated from the pib genomic clone and substituted for the 35S promoter upstream of gus gene in plasmid pCAMBIA1301 to construct a new plant expression vector pNAR604 (putative pib promoter-GUS + 35S-hpt). From Agrobacterium-mediated transformation and hygromycin selective culture in vitro, hygromycin resistant calli and 36 transgenic rice (Oryza sativa L.) plants were obtained. Histochemical assays of GUS activity showed that no expression was observed in the resistant calli and roots from transgenic rice if cultured under light, but after 24 h dark treatment there was strong GUS staining. Fluorimetric quantitative analysis indicated that GUS expression was organ-specific in transgenic rice. Without the dark treatment, GUS activity in roots and stems were about 7 and 3 times higher than in leaves in which GUS activity was only trace detected. After 24 h dark treatment, GUS activity in roots, stems and leaves of transgenic plants were all promoted and the largest increase was observed in leaves. Twenty-four hours after spraying with 5 mmol/L SA (Salicylic Acid) or 0.3 mol/L NaCl, GUS activity in leaves of the transgenic plants was 2.7 or 3.6 times respectively higher than untreated control. It was confirmed that an inductive promoter was involved in this 5.7 kb upstream region of pib gene, and dark, SA and NaCl treatments were inductive factors for pib promoter.
Production of Brassica Tri-Genomic Vegetable Germplasm by Hybridisation Between Tuber Mustard ( Brassica Juncea ) and Red Cabbage ( B. Oleracea )

Abstract:Brassica is a key agricultural genus, containing oilseed and vegetable crops sharing three major genomes (A, B and C) distributed in diploid (genome: AA, BB and CC) and allotetraploid (genome: AABB, AACC and BBCC) species. Interspecific hybridisation between Brassica species is an important way to combine valuable characters of different species. In the present study, interspecific hybrids between tuber mustard (AABB) and red cabbage (CC) were obtained with the aid of embryo culture. The results showed that ovules at five different maturity levels (7, 9, 10, 13 and 14 days after pollination), which were cultured on Murashige and Skoog medium supplemented with 3 % sucrose, 0.2 % active carbon and 400 mg/L glutamine, showed varying seed survival rates. A total of 15 (5 %) seeds were obtained, with ovules cultured at 14 days after pollination yielding the most seeds. Subsequently, eight (53 %) of the seeds did not germinate into normal plants. Furthermore, the putative hybrids were confirmed by random amplified polymorphic DNA (RAPD) and flow cytometry analysis, and only one (14 %) of the seven plants was confirmed to be a true tri-genomic hybrid (genome: ABC). Then the nodal segments of the true hybrid were treated with liquid medium supplemented with colchicine to obtain an allohexaploid, as confirmed by flow cytometry analysis and morphological observation. The synthesized hybrid showed unique phenotypes in comparison with the parents, such as in stem colour and leaf margin. The synthesized Brassica tri-genomic vegetable germplasm with A, B and C genomes has potential as a source of new crops for agriculture as well as a basis for further investigation of the mechanisms of Brassica evolution and adaptation.

Abstract:The promoter of Brassica campestris Male Fertile 5 ( BcMF5 ), a pollen coat protein member, class A ( PCP-A ) gene family, was isolated from Brassica rapa L. ssp. chinensis Makino (Chinese cabbage-pak-choi) by Thermal Asymmetric Interlaced Polymerase Chain Reaction (TAIL-PCR). Sequence analysis suggested that the 605-bp promoter of BcMF5 appears to be a pollen promoter. In an attempt to confirm the promoter activity of BcMF5 promoter, −609 to +3 bp and −377 to +3 bp fragments of the upstream sequence of BcMF5 were inserted at the site upstream of the coding region of the uid A gene in the sense orientation to construct two deletion expression vectors. Transient expression analysis in onion epidermal cells by particle bombardment showed that both −609 to +3 bp and −377 to +3 bp fragments of BcMF5 promoter were capable of driving β-glucuronidase gene expression. Furthermore, by Agrobacterium-mediated genetic transformation method, Arabidopsis transgenic Kan R plants were obtained. GUS assay analysis revealed that the promoter of BcMF5 induced gene expression at the early stage of anther development and drove high levels of GUS expression in anther walls, upper regions of petals, pollen, and pollen tubes in the middle and late stage of anther development, but did not drive any expression in sepals and pistils.

Abstract:SLR1 gene is a stigma-specific S-locus related gene widespread in Brassica. Primers derived from the SLR1 promoter sequence of B. napus var. westar were used to perform PCR analysis of several B. napus varieties, so as to isolate corresponding promoters of self-compatible B. napus. The fragments achieved by PCR were cloned and sequenced, and then fused to the GFP gene, replaced the 35S promoter in the plant expressed vector pFGC-eGFP. The vector was transformed to Arabidopsis by Agrobacterium-mediated transformation. By sequences analysis and fluorescence detection, the results showed that the SLR1 promoter in self-compatible B. napus had similar structure and function with that in self-incompatible B. oleracea which had been reported previously.

Abstract:Activation tagging is an important strategy in plant genomics by generating gain-of-function mutants. In this work, a library of Arabidopsis mutants was constructed by in planta transformation mediated by Agrobacterium tumefaciens containing an activation tagging vector pSKI015 with herbicide Basta as a selection marker (Fig. 1). Among 20000 independent transformants, 38 lines, i.e. about 0.2% of T(1) progeny, show visible morphological phenotypic variations (Fig. 2). Results of Southern blot analysis revealed that most of the transformants have more than three copies of T-DNA insertion (Fig. 3). Plasmid rescue and TAIL-PCR were used to recover the flanking genomic sequences of mutated target genes as the first step towards mutant gene cloning (Fig. 4, 5).

Abstract:The importance of using tissue-specific promoters in the genetic transformation of plants has been emphasized increasingly. Here, we report the isolation of a novel seed-specific promoter region from peanut and its validation in Arabidopsis and tobacco seeds. The reported promoter region referred to as groundnut seed promoter (GSP) confers seed-specific expression in heterologous systems, which include putative promoter regions of the peanut (Arachis hypogaea L.) gene 8A4R19G1. This region was isolated, sequenced, and characterized using gel shift assays. Tobacco transgenics obtained using binary vectors carrying uidA reporter gene driven by GSP and/or cauliflower mosaic virus 35S promoters were confirmed through polymerase chain reaction (PCR), RT-PCR, and computational analysis of motifs which revealed the presence of TATA, CAAT boxes, and ATG signals. This seed-specific promoter region successfully targeted the reporter uidA gene to seed tissues in both Arabidopsis and tobacco model systems, where its expression was confirmed by histochemical analysis of the transgenic seeds. This promoter region is routinely being used in the genetic engineering studies in legumes aimed at targeting novel transgenes to the seeds, especially those involved in micronutrient enhancement, fungal resistance, and molecular pharming.
Molecular tagging and mapping of a major linked marker for taproot thickness in a Brassica crop

Abstract:A DNA marker linked to the locus controlling storage-root thickness was identified using bulked segregant analysis (BSA) in an F-2 population derived from a cross between turnip (Brassica rapa L. ssp. rapifera Matzg.) and rapeseed (Brassica rapa L.). Root-thickness in the F-2 plants showed a continuous distribution from 2.2 - 10.7 cm, suggesting the effects of a few major and minor genes. BSA, using three bulks based on root-thickness, suggested that an AFLP marker (EACCMCAG-220) which appeared only in plants with a thickened root, was linked to a locus that controlled root size. This AFLP band was cloned, sequenced, and successfully converted into a simple, dominant, sequence characterised amplified region (SCAR) marker. This marker may help to identify turnip genotypes with thicker taproots in segregating progenies of root crops during marker-assisted selection (MAS) breeding programmes.
Reduced tar, nicotine, and carbon monoxide exposure while smoking ultralow- but not low-yield cigarettes.

Abstract:An unresolved public health issue is whether some modern cigarettes are less hazardous than others and whether patients who cannot stop smoking should be advised to switch to lower-yield cigarettes. We studied "tar" (estimated by urine mutagenicity), nicotine, and carbon monoxide exposure in habitual smokers switched from their usual brand to high- (15 mg of tar), low- (5 mg of tar), or ultralow-yield (1 mg of tar) cigarettes. There were no differences in exposure comparing high- or low-yield cigarettes, but tar and nicotine exposures were reduced by 49% and 56%, respectively, and carbon monoxide exposure by 36% while smoking ultralow-yield cigarettes. Similarly, in 248 subjects smoking their self-selected brand, nicotine intake, estimated by blood concentrations of its metabolite cotinine, was 40% lower in those who smoked ultralow but no different in those smoking higher yields of cigarettes. Our data indicate that ultralow-yield cigarettes do deliver substantial doses of tar, nicotine, and carbon monoxide, but that exposures are considerably less than for other cigarettes.

Abstract:Agrobacterium-mediated transformation of canola (Brassica napus) via hypocotyl segments has been a commonly used method for the past 30 years. While the hypocotyl-based method is well-established, it is not readily adapted to elite germplasm and the prolonged process is not ideal for a production transformation setting. We developed an Agrobacterium-mediated transformation method based on epicotyl and higher stem (internodal) segments that is efficient, rapid and amenable for high-throughput transformation and genome editing. The method has been successfully implemented in multiple canola genotypes. The method appears to be genotype-independent, with varying transformation efficiencies. Internodal segment transformation was used to generate transgenic events as well as CRISPR-Cas9-mediated frameshift gene knockouts.

Abstract:Tissue/organ-specific promoters are important tools in genetic engineering and crop molecular breeding. They are well characterized in dicots, such as Arabidopsis, tobacco, and tomato, but not sufficiently in monocots, especially in wheat. In this study, the genes specifically expressed in seven different tissues, including coleoptile, root, leaf, pistil, anther, embryo, and endosperm were identified through analyzing the public transcriptome data from a wheat microarray using the ROKU method. The expression patterns of selected genes were validated by reverse transcription polymerase chain reaction. The results showed that these selected genes were expressed specifically or preferentially in each representative tissue/organ. Moreover, the function of their promoters was verified by transient expression in wheat or stable transformation in Arabidopsis. The results showed that these promoters can efficiently and predominantly drive uidA (β-glucuronidase) reporter gene expression in different tissues. Due to their tissue-specific nature, these promoters can be used as potential candidates in plant genetic engineering.

Abstract:Abstract Background Brassica napus is an important oilseed crop which offer a considerable amount for global vegetable oil production. The establishment of an efficient genetic transformation system with a convenient transgenic-positive screening method is of great importance for gene functional analysis and molecular breeding. However, to our knowledge, there are few such aforementioned systems available for efficient application in B. napus .Results Based on the well-established genetic transformation system in Brassica napus , five vectors companied with red fluorescence protein from Discosoma sp. ( DsRed ) were constructed and transformed via agrobacterium -mediated hypocotyl transformation. The average of 59.1% tissues were marked with red fluorescence by visual screening method in tissue culture medium, and 96.1% of which were amplified with the exogenous genes in eight different rapeseed varieties on average. In addition, the final transgenic-positive efficiency of the rooted plantlets was up to 90.7% from the red fluorescence marked tissues, which was much higher than that of the previous reports. Besides, DsRed also could be applicable to the seedlings, including of seed coats, roots, hypocotyls and cotyledons during seed germination. These results indicate that the high-efficiency genetic transformation system combining with the transgenic-positive visual screening method presented here will help us to efficiently and conveniently obtain transgenic-positive rapeseed plantlets.Conclusion A novel method was developed for rapidly, conveniently and high-efficiently obtaining the transgenic lines, which would help to obtain the higher proportion of transgenic positive regenerated plantlets, thereby avoiding the long period plant regeneration. This will benefit the gene functional study especially in high throughput molecular biology research.

Abstract:γ-TMT Gene in Brassica napus was obtained by PCR technique and cloned into pMD18-Tvector.The recombinant clone was detected by PCR technique and analyzed by the restriction enzyme.A full length of cDNA gene was sequenced.The results showed that the length of the clone sequence was 1 044 bp.γ-TMT gene in Brassica napus was cloned into 35S promoter downstream of plant over-expression vector pCambia1300 in orientation.

Abstract:A pair of primers was designed according to the sequence of phloem protein 2 (PP2), which shows phloem-specificity from Cucubita maxima, and a 966bp fragment of PP2 gene in the promoter region was amplified by polymerase chain reaction (PCR) using the genomic DNA of a local C. maxima cv.variety as the template. The fragment was cloned into pUCm-T vector, and a new recombined vector named pUCm-PSP was obtained after analyzed with PCR and restriction digestion. The results of sequence analysis indicated that this fragment had 95% and 98% homology compared with two reported promoters, respectively. This result suggested that it might have the same function as other PSP. A new plant expression vector named pHZ03 in which the reporter gene GFP is droved by PSP was constructed after cutting two vectors pUCm-PSP and pCAMBIA1302 with two restriction enzymes Pst I and Bgl II, subsequently recovering the small PSP fragment from pUCm-PSP recombined vector and the long fragment excised CaMV35S promoter in the upper (region) of GFP reporter gene from pCAMBIA1302 plant expression vector, and then ligation, transformation and identification. The recombined plant expression vector was transferred into Agrobacterium tumefaciens strains of LBA4404, GV3101, EHA105 and A.rhizogenes strain of 15834 by using cell competent method, and the foundation has been set up for further research work in expression and function of this promoter.

Abstract:Background: Transcription factors GATAs are involved in plant developmental processes and respond to environmental stresses through binding DNA regulatory regions to regulate their downstream genes. However, little information on the GATA genes in Brassica napus is available. The release of the reference genome of B. napus provides a good opportunity to perform a genome-wide characterization of GATA family genes in rapeseed. Results: In this study, 96 GATA genes randomly distributing on 19 chromosomes were identified in B. napus , which were classified into four subfamilies based on phylogenetic analysis and their domain structures. The amino acids of BnGATAs were obvious divergence among four subfamilies in terms of their GATA domains, structures and motif compositions. Gene duplication and synteny between the genomes of B. napus and A. thaliana were also analyzed to provide insights into evolutionary characteristics. Moreover, BnGATAs showed different expression patterns in various tissues and under diverse abiotic stresses. Single nucleotide polymorphisms (SNPs) distributions of BnGATAs in a core collection germplasm are probably associated with functional disparity under environmental stress condition in different genotypes of B. napus . Conclusion: The present study was investigated genomic structures, evolution features, expression patterns and SNP distributions of 96 BnGATAs. The results enrich our understanding of the GATA genes in rapeseed.
T-DNA Insertional Mutagenesis for Activation Tagging in Rice

Abstract:Abstract We have developed a new T-DNA vector, pGA2715, which can be used for promoter trapping and activation tagging of rice (Oryza sativa) genes. The binary vector contains the promoterlessβ-glucuronidase (GUS) reporter gene next to the right border. In addition, the multimerized transcriptional enhancers from the cauliflower mosaic virus 35S promoter are located next to the left border. A total of 13,450 T-DNA insertional lines have been generated using pGA2715. Histochemical GUS assays have revealed that the GUS-staining frequency from those lines is about twice as high as that from lines transformed with the binary vector pGA2707, which lacks the enhancer element. This result suggests that the enhancer sequence present in the T-DNA improves the GUS-tagging efficiency. Reverse transcriptase-PCR analysis of a subset of randomly selected pGA2715 lines shows that expression of the genes immediately adjacent to the inserted enhancer is increased significantly. Therefore, the large population of T-DNA-tagged lines transformed with pGA2715 could be used to screen for promoter activity using the gusreporter, as well as for creating gain-of-function mutants.

Abstract:PSG076 is a pollen-specific gene isolated from wheat. The 1.4-kb promoter upstream of the ATG start codon was isolated by inverse-PCR (IPCR). To determine its activity, the PSG076 promoter was fused with the β-glucuronidase (GUS) reporter gene and introduced into tobacco. Histochemical analysis in transgenic tobacco showed that GUS activity was detected in late bicellular pollen grains and increased rapidly in mature pollen. GUS activity was also detected in pollen tubes of transgenic tobacco. No GUS activity was found in other floral and vegetable tissues. These results indicate that the PSG076 promoter directs pollen-specific activity at late stages of pollen development and pollen tube growth. Deletion analysis showed that a 0.4 kb fragment of the promoter was enough to confer pollen-specific expression.

Abstract:Helicases are molecular motor proteins that perform a variety of cellular functions including transcription, translation, DNA replication and repair, RNA maturation, ribosome synthesis, nuclear export and splicing processes. The p68 is an evolutionarily conserved protein which plays pivotal roles in all aspect RNA metabolism processes. It is well established that helicases provides abiotic stress adaptation in plants but analysis of cis-regulatory elements present in the upstream regions is still infancy. Here we report isolation and functional characterization of the promoter of a DEAD-box helicase Psp68 in response to abiotic stress and hormonal regulation. The promoter of Psp68 was isolated by gene walking PCR from pea genomic DNA library constructed in BD genome walker kit. In silico analysis revealed that promoter of Psp68 contained a TATA, a CAAT motif and also harbors some important stress and hormone associated cis regulatory elements, including E-box, AGAAA, GATA-box, ACGT, GAAAA and GTCTC. Functional analyses were performed by Agrobacterium-mediated transient assay in tobacco leaves. Very high level of GUS activity was observed in agroinfiltrated tobacco leaves by the construct carrying the Psp68 promoter::GUS, subjected to abiotic stress and exogenous hormonal treatments. Stress-inducible nature of Psp68 promoter opens possibility for the study of the gene regulation under stress condition. Therefore, may be useful in the field of agriculture and biotechnology.

Abstract:Plants are proven effective bioreactors for the production of heterologous proteins including those desired by the biopharmaceutical industry. However, the potential of plants as bioreactors is limited by the availability of characterized plant promoters that can drive target gene expression in relatively distant plant species. Seeds are ideal for protein storage because seed proteins can be kept stably for several months. Hence, a strong promoter that can direct the expression and accumulation of target proteins within seeds represents a powerful tool in plant biotechnology. Toward this end, an effort was made to identify such a promoter from Vigna radiata (mung bean) to drive expression in dicot seeds. A 784-bp 5'-flanking sequence of the gene encoding the 8S globulin alpha' subunit (8SG alpha') of the V. radiata seed storage protein was isolated by genome walking. When the 5'-flanking region was analyzed with bioinformatics tools, numerous putative cis-elements were identified. The Green Fluorescent Protein (GFP) regulated by this promoter was observed to be transiently expressed in protoplasts derived from V. radiata cotyledons. Finally, transgenic Arabidopsis plants expressing the beta-glucuronidase (GUS) reporter gene driven from the 8S globulin alpha' promoter showed strong GUS expression in transgenic embryos in both histochemical and quantitative GUS assays, confirming high expression within seeds. Therefore, the V. radiata 8S alpha' promoter has shown potential in directing expression in seeds for bioreactor applications.
