Abstract:DNA was amplified from 11 samples of crude snake drugs by 2 random primers. "Wushaoshe" (Zaocys dhumnades) and its substitutes, Elaphe taeniurus, E. rufodorsata, E. carinata and Sinonatrix annularis, and "Jinqian Baihyashe" (Bungarus multicinctus) and its adulterants, Dinodon rufozonatum and Sinonatrix annularis, can be distinguished by electrophoresis pattern of the amplified products. The results showed that the molecular genetic marker technique can be used as a new method for the identification of crude snake drugs.
Molecular Identification of Some Bursaphelenchus Populations with the Aid of PCR-RFLP of Rdna

Abstract:Six populations of genus Bursaphelenchus isolated from pine wood produced in China, Japan and Korea were identified with the aid of PCR-RFLP of rDNA. It was bound that one of the populations, which had its origin in the forests of Zhenhai, Zhejiang Province, China, and the other, which was derived from imported coniferous wood packages produced in Japan were both identified as B. xylophilus. The other four populations, including populations from Fuyang of the Zhejiang Province, Korea, Hongkong and Japan, were identified as B.mucronatus. All the four restriction enzymes, namely HinfⅠ, HaeⅢ, MspⅠ, and CfoⅠ, with their special sites of cutting could be used to differentiate the two morphologically very similar species.

Abstract:The secretions of the venom glands, dry skin and whole body of several Bufo species have been used as traditional medicines in East Asia to treat heart failure and cancer. Due to the highly similar morphological features of Bufo species and their derived commercial crude drugs, along with the high similarity of chemical composition of the secretions of venom glands, it is very challenging to identify the medicinal toads and the related crude drugs. The cyt-b sequences provide useful information to authenticate medicinal Bufo species. Based on the cyt-b sequences, a simple PCR-RFLP method was established for the identification of the medicinally used Bufo species as well as their derived crude drugs. The 23 specimens from three medicinally used Bufo species, B. bufo gargarizans (Bbg), B. melanostictus (Bm) and B. raddei (Br), were clearly divided into 3 groups according to the sequences of amplified cyt-b regions, which could be digested by specific restriction enzymes NcoI, EcoRV and BstXI, respectively. Then the specific PCR-RFLP method was further used to identify 9 samples of commercial crude drugs even with serious degradation of DNA, and all nine samples were identified as B. bufo gargarizans. The method is suitable for identification of medicinally used Bufo species and the related crude drugs.

[An Authentication Method of Bear Bile Powder Based on the Near Infrared Spectroscopy].

Abstract:A method based on the near infrared (NIR) spectroscopy was established for the authentication of bear bile powder and the quantitative analysis of the proportion in the counterfeits. The NIR spectra of 30 bear bile powder samples from 4 different origins, 6 pig gall powder samples, and 65 mixed samples of bear bile powder and pig gall powder with different proportions range from 0 to 100% were collected. The principal component analysis (PCA) was done and well-marked boundaries were observed between the bear bile and the counterfeits, also the bear bile samples from different origins. Partial least squares discriminant analysis (PLS-DA) was used to differentiate the bear bile and the counterfeits, and the rate of accuracy is 95%. Finally, the PLS calibration model was established for the quantitative analysis of the bear bile proportion in the counterfeits, the coefficient of determination of the prediction set is 0.9759, and the root-mean-square prediction error is 4.25%, which indicated that the result was satisfied. This research contributes to the rapid and undamaged identification of bear bile powder, and also offers reference for the quality identification of other expensive medicinal materials.
Molecular characterisation of some Asian isolates of Bursaphelenchus xylophilus and B. mucronatus using PCR-RFLPs and sequences of ribosomal DNA

Abstract:Five isolates of Bursaphelenchus xylophilus, four of which were collected from Chinese forests and one from Japan, as well as, four isolates of B. mucronatus extracted from pinewood packaging imported from Japan, Korea and from forests in China, were studied using PCR-RFLPs and sequences of the ITS and D2-D3 fragments of rDNA. RFLP analysis of the ITS region with restriction endonucleases revealed that CfoI, HaeIII, HinfI, MspI and TaqI yielded distinct patterns that separated A xylophilus and A mucronatus. Phylogenctic analyses of alignments of published and original sequences of both the ITS I region and ITS1-5.8S-ITS2 region revealed that both A xylophilus and A mucronatus are composed of two groups of populations with different ITS types. The taxonomic status of these groups is discussed.
STUDY ON ALLELE-SPECIFIC DIAGNOSTIC PCR OF THE TRADITIONAL CHINESE MEDICINES OF THE DEERS

Abstract:AIM:To develop a convenient and accurate method of DNA molecular marker for the identification of traditional Chinese medicines made of deers, consisting of pilose antler, penis and testis, tendon and foetus.METHODS:Based on the analysis of DNA sequence of mitochondrial Cyt b gene from original animals of both genuine crude drugs, Cervus nippon and Cervus elaphus, and adulterants, a pair of allele-specific primers named as ILu01-L and ILu01-H were designed for distinguishing geniune crude drugs of deers from their adulterants.RESULTS:The results of diagnostic PCR annealing at 64 degrees C for original animals showed that a 365 bp fragment was only amplified from DNA templates of Cervus nippon and Cervus elaphus. For the identification of medicinal materials total of 43 samples from 6 packages were tested under the same reaction conditions except for DNA templates extracted from these crude drugs. Only 9 samples mentioned above was shown to generate positive amplificon. The result indicate that of 8 samples from 1 package of pilose antler and only 1 sample of deer tendon was genuine crude drug. After that, 3 amplified fragments selected randomly were performed with sequencing analysis with the purpose of verifying the results from diagnostic PCR. Data from sequencing confirmed the reliability of diagnostic PCR identification.CONCLUSION:The diagnostic primers designed in the present study were highly specific for Cervus nippon and Cervus elaphus, and they could be used for the authentication of traditional Chinese medicines made from the deer. The quality of the crude drugs of the deer in the current market is a problem and more effective quality control for these traditional Chinese medicines is urgently needed.
[Identification of Chinese Crude Drug Snake Gallbladder by DNA Molecular Marker].

Abstract:AIM:It is difficult to identify the Chinese crude drug snake gallbladder accurately by morphological and microscopical characteristics or chemical components only. In order to solve the problem, the technique based on DNA molecular marker was introduced into the authentication of snake gallbladder.METHODS:DNA templates were extracted from the membrane or the bile of snake gallbladder, and also from the muscle of the original animal Elaphe schrenckii. About 400 bp DNA fragments of 12S rRNA gene were amplified from the templates and sequenced subsequently.RESULTS:Enough amounts of DNA templates could be extracted from a bit of membrane or bile of snake gallbladder. The sequence of amplicons from the membrane, bile and muscle of the same individual were identical completely.CONCLUSION:The technique of DNA molecular marker could be used for the authentication of snake gallbladder and bile. The results indicate that the technique could be used for the identification of crude drugs from other animal secretion. DNA sequence analysis also demonstrated that the origins of commercial snake gallbladder were complicated and more efficient quality control was necessary for supervising the crude drug in the market.

Abstract:DNA templates were extracted from Rana chensinensis , Bufo gargarizans and Gadus macroccphalu , and 12S rRNA gene fragment was amplified and sequenced with universal primers. Based on the sequence data obtained both from our experiments and the references, a pair of diagnostic primers, HsmL1 and HsmH1, were designed for the authentication of Oviductus Ranae. When PCR was performed using diagnostic primers, positive amplification appeared in the tube with template DNA from R. chensinensis and R. amurensis only. Therefore, HsmL1 and HsmH1 could be used for authentication of Oviductus Ranae. The samples of Oviductus Ranae from 5 different drug stores were examined with the primers, and the results indicate that the sources of material of Oviductus Ranae are still complicated in the markets. Abstract:Freshly-used crude drugs have unique functions and advantages in TCM practice of treating diseases. Jinlong Capsule is a patent traditional Chinese medicine product effective for treatment of hepatocarcinoma, and fresh Jinqian Baihua She (JBS, the body of juvenile Bungarus multicinctus ) is one of its important ingredients. The emergence of counterfeit fresh JBS, often identified as dried JBS with almost identical appearance, poses a difficult problem in the quality control of the product. Herein we report a molecular quantification-based method for differentiation of fresh and dried JBS by determining the copy number of a specific DNA marker in the samples. Using species-specific primers and TaqMan probes, we established a real-time quantitative PCR system for amplification of a fragment in the 658-bp cytochrome oxidase subunit I (COI) region from JBS specimens. The amplicon copy number in the muscle tissues ranged from 1.14×10 7 to 4.83×10 7 copies/mg in fresh JBS samples, as compared with 1.13×10 5 − 8.91×10 6 copies/mg in dried JBS samples. Based upon Fisher discriminant analysis, we used 1.27×10 7 copies/mg as the cut-off value for differentiating fresh and dried JBS, which was validated in the single-blinded validation test of fresh and dried JBS samples. This qPCR system may provide an efficient means for accurate identification of fresh JBS to improve the quality control of the medicinal product.

Authentication of an Animal Crude Drug, Zaocys, by Diagnostic PCR.

Abstract:A pair of diagnostic primers for distinguishing the Chinese crude drug Zaocys (Zaocys dhumandes) from its substitutes was designed based on the sequence data of the original animal of the drug and substitutes. Total DNAs were extracted from genuine crude drug and 5 of its substitutes, as well as from 12 species of original animal of the snake crude drug. Diagnostic PCRs were performed using the primers with these total DNAs as a template, annealing at 60-65 degrees C, Positive amplifications were obtained from all DNA templates of Zaocys, whereas negative amplifications were obtained from that of others, The results indicate that Zaocys samples could be definitely distinguished from its substitutes by diagnostic PCR, and no incorrect discrimination was found under the same reaction conditions. The advantages of the method in the authentication of crude drugs are also discussed in the present paper.
Application of DNA Barcoding for the Identification of a Traditional Chinese Medicine Shedan

Abstract:Abstract Background Shedan has a long history of application in Traditional Chinese medicine (TCM), however, Shedan from different original source has been indiscriminately used. So far, there is still a lack of an effective tool to differentiate the original source of Shedan medicinal materials, which brings great risk to the safety and effectiveness of clinical applications. Hence, it is imperative to develop a practicable approach to identify Shedan medicinal materials. Methods The specificity of two pairs of primers, including Folmer’s universal primers and a pair of originally designed primers COISNFF/COISNFR, was tested to screen the more specific primers for further origin identification of Shedan. A total of 253 fresh snake gallbladder samples from 31 morphologically identified snake species were collected and authenticated. Moreover, 51 fresh snake bile samples and 17 fresh bile samples from five other common domestic poultry and livestock (cattle, chicken, duck, pig and sheep) were collected and distinguished using the more specific primers. Additionally, a total of 195 market Shedan samples randomly selected from 18 batches of Shedan medicinal materials were investigated. Sequence definition was executed by querying sequence similarities in GenBank and the Barcode of Life Data System (BOLD), respectively. Results It turned out that the standard COI barcode obtained by COISNFF/COISNFR primers, rather than Folmer’s universal primers, can distinguish all the testing samples from each other in fresh Shedan samples, and COISNFF/COISNFR primers were also specific to snake species and the other four animal species except duck. In terms of market Shedan, 84.6% (165/195) samples can be attributed to 13 snake species from four families and 4.6% (9/195) can be attributed to adulterated chicken species. Conclusion The COI-based DNA barcoding was practicable for species identification of Shedan used in traditional Chinese medicine. The original source of current market Shedan, including adulterated species, has been preliminarily clarified, which provides a foundation for quality control of Shedan medicinal materials.
Authentication of oviductus ranae and its original animals using molecular marker.

Abstract:Two pairs of diagnostic primers, IHm01-L/IHm01-H and IHm02-L/IHm02-H, for distinguishing the Chinese crude drug Oviductus Ranae from its substitutes were designed based on sequences of Cyt b gene fragment of the original animals of the drug and substitutes. Total DNAs were extracted from crude drugs purchased from five drugstores in different regions, as well as from original animals of the drug, Rana chensinensis, and seven species of related ranid species. Diagnostic polymerase chain reactions (PCRs) were performed using the two pairs of primers with the total DNAs of the original animals as a template. The result showed that a 240 bp DNA segment was clearly amplified from all templates of Rana chensinensis using primers IHmO1-L and IHm01-H, whereas no DNA band appeared from other templates. While using primers IHm02-L and IHm02-H, we got a clear 140 bp DNA band from all the templates of R. huanrenensis and 3 oviducts of the same species, no PCR product was observed from the other samples. A set of PCR reactions was employed to identify crude drugs from the five drugstores using the two pairs of primers together with HsmL1 and HsmH1 reported in our previous study. The results show that only 20% of the Oviductus Ranae currently sold in markets are qualified products and the rest are not.
Authentication of Chinese Crude Drug, Gecko, by Allele-Specific Diagnostic PCR

Abstract:Based on the sequences of the mitochondrial 12S rRNA gene fragment of 17 samples from Gekkonidae, Salamandridae, Agamidae and Hynobiidae, respectively, a pair of allele-specific primers was designed for differentiating the Chinese medicinal material Gecko from its adulterants by PCR. The results of amplification with the primers indicate that amplicons from the templates of Gekko gecko were clearly revealed by agarose gel electrophoresis, whereas no evident amplicons were found from other species. The primers were employed to identify crude drug samples from different sources. Among a total of 9 samples, 3 were diagnosed as genuine Gecko. This result is consistent with morphological identification and DNA sequence analyses.
Molecular authentication of the animal crude drug Sailonggu (bone of Myospalax baileyi).

Abstract:Two pairs of allele-specific diagnostic primers (SL1L/SL1H and SL2L/SL2H) for distinguishing the Chinese crude drug Sailonggu (bone of plateau zokor, Myospalax baileys) from its substitutes were designed based on complete sequences of mitochondrial 12S rRNA and cytochrome b genes of the original animals of Myospalacinae, bamboo rat Rhizomys sinensis and black lipped pika Ochotona curzoniae. Total DNA was extracted from crude drug samples and original animals. Allele-specific diagnostic PCRs were performed using these primers with the total DNA as a template annealing at 65degreesC. Positive amplifications were obtained from all DNA templates of Sailonggu and M. baileyi, whereas negative amplifications resulted from those of other zokors, the bamboo rat and black lipped pika. These results indicate that Sailonggu samples can be definitely distinguished from their substitutes by diagnostic PCR, and no incorrect discrimination was found under the same reaction conditions. Each of the two diagnostic primer pairs can be used to distinguish crude drug Sailonggu from its substitutes or adulterants. The three Sailonggu samples studied were diagnosed as genuine Sailonggu. In addition, the results of sequence alignment and phylogenetic analysis are congruent with that of the allele-specific diagnostic PCR.
Factors Affecting Birth Weights of Swine.

Abstract:The data were taken from the breeding and farrowing records of the Animal Husbandry Section of the IOWA GRICULTURAL EXPERIMENT STATION during the years 1915 and 1917 to 1930 inclusive. The data were collected in connection with other experiments2 not planned expressly to make this study possible. Yet no practice was followed which seems likely to have affected the birth weights in a way which would have invalidated this analysis. The data analyzed are the birth weights of 506 litters containing 3639 pigs, all of which were produced by gilts about one year old at the time of farrowing. The time of weighing ranged from immediately after birth in some cases to a little more than twelve hours after birth in a few cases. Some pigs had suckled before being weighed but others had not. All litters were farrowed in the months of April and May, except nine in June, one in July and three in August. The breeds included purebred or high grade Poland Chinas, Tamworths, Hampshires and six different crosses. Over 71 percent of all these litters were purebred or high grade Poland Chinas. To determine the importance of each factor affecting birth weight, FISHER'S (1930) method of analyzing variance was used. The percentage of the variance which disappears when a certain variable is held constant may be used to express the closeness of the relation between that variable and birth weight (table 3). For those accustomed to think in terms of correlation coefficients, it is easy to understand such a proportion as equivalent to a squared primary correlation coefficient between birth weight
Identification and differentiation of Asian seabass and mangrove red snapper fillets by CYTB sequence-based PCR analysis

Abstract:Overall quality attributes between deskinned Asian seabass and mangrove red snapper fillet are similar. This may lead to the substitution of Asian seabass, a cheaper species, with the latter. Therefore, the authentication method for distinguishing these two species could conquer adulteration. Four primers were designed, based on mitochondrial cytb gene alignment. Polymerase chain reaction (PCR) was performed using the DNA of both species with 35 cycles of amplification. The primers provided the amplicons with the size of 480, 396, 185, and 268 for primers 1, 2, 3, and 4, respectively. All primers successfully differentiated Asian seabass from mangrove red snapper both raw and cooked fillets from at least 30 mg of the sample weight. Moreover, primer 4 showed the highest specificity when tested with other 5 species of fish generally sold in the Thai market. The limit of detection was around 6 ng of Asian seabass DNA in a total of 120 ng mangrove red snapper DNA. Therefore, the PCR-based method using primer 4 was a useful tool for the identification of Asian seabass adulterated in mangrove red snapper fillets or slices. This primer could be implemented for authentication of fish and fish products to prevent mislabeling for customers.

Abstract:Penis et testis cervi is one of the precious traditional Chinese medicines (TCMs) in China, which, when authentic, is sourced from species of Cervi nippon Temmink (C. Nippon) and Cervi elaphus Linnaeus (C. elaphus L). Authentication of species of Penis et testis cervi remains technically challenging. This study attempted to modify the species-specific polymerase chain reaction (PCR) and PCR-restriction fragment length polymorphism (PCR-RFLP) method using restriction enzyme Hpy188III for the identification of Penis et testis cervi species. First, the cytochrome b (Cyt b) and cytochrome c oxidase subunit I (COI) genes in mitochondrial DNA from two authentic species and their counterfeits or adulterants were downloaded and analyzed with bioinformatics tools to establish the phylogenetic tree, then a series of primers were designed, and the restriction enzyme sites were confirmed. Genomic DNA was extracted from all samples by the improved SDS method. Species-specific PCR reactions were optimized to differentiate authentic Sika deer and Red deer Penis et testis cervi from their counterfeits or adulterants. PCR-RFLP using single restriction enzyme Hpy188III was performed in Cyt b regions to identify the species of Sika deer and Red deer. Simultaneously, recombination plasmid carrying the cyt b fragment could be cloned, sequenced, and blasted. The assay specificity and sensitivity were evaluated, and 30 batches of commercial samples were validated. The phylogenetic tree showed that the physico-chemical properties of Sika deer and Red deer variants were similar. Sufficient DNA extracts were extracted from all samples used in the improved SDS method. Establishment and optimization of a species-specific PCR reactions and PCR-RFLP system were undertaken to identify deer species in Penis et testis cervi samples. A specific PCR-RFLP pattern with three distinct products of sizes 155, 113, and 204 bp was exhibited in Sika deer species, and two distinct products of 204 and 268 bp in size were evident in Red deer species, which were significantly different from the pattern of the counterfeits or adulterants. The distinctive PCR-RFLP fingerprint pattern using the restriction enzyme Hpy188III is a rapid and reliable assay for identification of the Penis et testis cervi species.

[Molecular identification of animal species of Bufonis Venenum from secretions]

Abstract:The animal species is one of the key factors affecting the quality of Bufonis Venenum. The quality of Bufonis Venenum derived from Bufo bufo gargarizans is significantly higher than that from B. melanostictus. Since Bufonis Venenum is from secretions, the conventional identification methods are difficult to identify the animal species due to the lack of the appearance and morphology of the animals. The rapid development of molecular identification technology has provided new methods for the identification of Bufonis Venenum. However, because of the low content and serve degradation of residual DNA in secretions, the research on the molecular identification of Chinese medicinal materials from secretions remains to be carried out. To understand the animal species of Bufonis Venenum, this study collected 83 samples of Bufonis Venenum, including 7 commercially available samples, 5 reference medicinal materials, and 71 animal samples from which Bufonis Venenum was prepared according to the method in the 2020 edition of the Chinese Pharmacopoeia. Different DNA extraction methods were used and compared, and the mitochondrial 16S rRNA gene fragments were amplified, on the basis of which the phylogenetic trees were built. Finally, molecular identification of the animal species of the samples was performed. The results showed that the DNA extracted from Bufonis Venenum by the reagent kit had good quality, and 16S rRNA sequences were successfully amplified from 80 out of the 83 samples. In addition, 71 16S rRNA sequences of the animal species of Bufonis Venenum were downloaded from GenBank. The phylogenetic trees constructed based on the neighbor-joining(NJ) method and the Bayesian inference(BI) method showed that the samples derived from B. bufo gargarizans and B. melanostictus were clustered into separate monophyletic clades, with the support of 100%(NJ) and 1.00(BI), respectively. The animal species of both commercially available samples and reference medicinal materials were B. bufo gargarizans. In conclusion, DNA can be extracted from Bufonis Venenum derived from secretions, and the 16S rRNA gene sequences can be amplified, which can be used for molecular identification of the animal species of Bufonis Venenum. The findings provide a reference for the quality control of Bufonis Venenum and the identification of animal species of medicinal materials derived from secretions.
A specific SNP-based multiplex PCR assay for the simultaneous identification of two biological ingredients for the Chinese patent medicine, Danggui Buxue pill

Abstract:Background: An increasing number of Chinese patent medicines (CPM) have been widely used in East Asian and North American countries, and the safety and efficacy of CPM have highly attracted public attention. However, it is difficult to supervise the authenticity of multiple biological ingredients within CPM based on microscopic inspection and physical and chemical detection. The raw materials may have similar characteristics of tissue structures and ergastic substances or similar chemical composition and contents when substitutes and/or adulterants are added. DNA molecular markers have been used to distinguish the biological ingredients within CPM based on conventional PCR assay. However, it was proved to be time- and labor-consuming and reagent-wasting, as multiple PCR amplification strategies were required for identifying the complex species composition within CPM. Here, we took the CPM (Danggui Buxue pill) as an example and aimed to establish a specific SNP-based multiplex PCR assay and simultaneously determine the authenticity of the two biological ingredients (Angelicae Sinensis Radix and Astragali Radix) within this CPM. Methods: We, respectively, designed the species-specific primers based on highly variable nrITS for discriminating Angelicae Sinensis Radix and Astragali Radix from their common substitutes and adulterants. The specificity of the primers was checked through conventional PCR assay and multiplex PCR assay. Furthermore, we used a handcrafted Danggui Buxue pill sample (DGBXP) to optimize annealing temperatures for the primers with multiplex PCR, and the sensitivity was also assessed. Finally, fourteen batches of commercial Danggui Buxue pills were used to verify the stability and practicability of the established multiplex PCR assay. Results: Two pairs of highly species-specific primers for amplifying Angelicae Sinensis Radix and Astragali Radix were screened, and our established multiplex PCR assay showed high specificity and sensitivity (lowest detection concentration: 4.0 × 10 −3 ng/μL) at an optimal annealing temperature of 65°C. The method could simultaneously identify both biological ingredients within the Danggui Buxue pill. Conclusion: The specific SNP-based multiplex PCR provided a simple, time-, and labor-saving method for the simultaneous identification of the two biological ingredients within Danggui Buxue pills. This study was expected to provide a novel qualitative quality control strategy for CPM.
