Abstract:Injury to mammalian motor nerves can lead to paralysis, but relatively successful regeneration may occur when conditions are favorable. Elucidation of the mechanism upholding successful regeneration is of theoretical and clinical interest. In this study, the hypothesis that insulin-like growth factor II (IGF-II) can stimulate motor nerve regeneration was tested. When IGF-II was infused continuously near a site of crush on the sciatic nerve, the distance of motor axon regeneration was increased significantly in rats. In contrast, spontaneous regeneration was inhibited when an anti-IGF-II antiserum was infused through a "window" in the epineurium. Thus, infused IGF-II can increase, and endogenous IGFs can support, the regeneration of motor axons in lesioned nerves.

Abstract:An extraordinary variety of experimental (e.g., flicker, magnetic fields) and clinical (epilepsy, migraine) conditions give rise to a surprisingly common set of elementary hallucinations, including spots, geometric patterns, and jagged lines, some of which also have color, depth, motion, and texture. Many of these simple hallucinations fall into a small number of perceptual geometries-the Klüver forms-that (via a nonlinear mapping from retina to cortex) correspond to even simpler sets of oriented stripes of cortical activity (and their superpositions). Other simple hallucinations (phosphenes and fortification auras) are linked to the Klüver forms and to pattern-forming cortical mechanisms by their spatial and temporal scales. The Klüver cortical activity patterns are examples of self-organized pattern formation that arise from nonlinear dynamic interactions between excitatory and inhibitory cortical neurons; with reasonable modifications, this model accounts for a wide range of hallucinated patterns. The Klüver cortical activity patterns are a subset of autonomous spatiotemporal cortical patterns, some of which have been studied with functional imaging techniques. Understanding the interaction of these intrinsic patterns with stimulus-driven cortical activity is an important problem in neuroscience. In line with this, hallucinatory pattern formation interacts with physical stimuli, and many conditions that induce hallucinations show interesting interactions with one another. Both types of interactions are predictable from neural and psychophysical principles such as localized processing, excitatory-inhibitory neural circuits, lateral inhibition, simultaneous and sequential contrast, saccadic suppression, and perceptual opponency. Elementary hallucinations arise from familiar mechanisms stimulated in unusual ways.

Abstract:We propose to use bifurcation theory and pattern formation as theoretical probes for various hypotheses about the neural organization of the brain. This allows us to make predictions about the kinds of patterns that should be observed in the activity of real brains through, e.g. optical imaging, and opens the door to the design of experiments to test these hypotheses. We study the specific problem of visual edges and textures perception and suggest that these features may be represented at the population level in the visual cortex as a specific second-order tensor, the structure tensor, perhaps within a hypercolumn. We then extend the classical ring model to this case and show that its natural framework is the non-Euclidean hyperbolic geometry. This brings in the beautiful structure of its group of isometries and certain of its subgroups which have a direct interpretation in terms of the organization of the neural populations that are assumed to encode the structure tensor. By studying the bifurcations of the solutions of the structure tensor equations, the analog of the classical Wilson and Cowan equations, under the assumption of invariance with respect to the action of these subgroups, we predict the appearance of characteristic patterns. These patterns can be described by what we call hyperbolic or H-planforms that are reminiscent of Euclidean planar waves and of the planforms that were used in [1, 2] to account for some visual hallucinations. If these patterns could be observed through brain imaging techniques they would reveal the built-in or acquired invariance of the neural organization to the action of the corresponding subgroups.

Spatial and color hallucinations in a mathematical model of primary visual cortex

Abstract:We study a simplified model of the representation of colors in the primate primary cortical visual area V1. The model is described by an initial value problem related to a Hammerstein equation. The solutions to this problem represent the variation of the activity of populations of neurons in V1 as a function of space and color. The two space variables describe the spatial extent of the cortex while the two color variables describe the hue and the saturation represented at every location in the cortex. We prove the well-posedness of the initial value problem. We focus on its stationary, i.e. independent of time, and periodic in space solutions. We show that the model equation is equivariant with respect to the direct product G of the group of the Euclidean transformations of the planar lattice determined by the spatial periodicity and the group of color transformations, isomorphic to O(2), and study the equivariant bifurcations of its stationary solutions when some parameters in the model vary. Their variations may be caused by the consumption of drugs and the bifurcated solutions may represent visual hallucinations in space and color. Some of the bifurcated solutions can be determined by applying the Equivariant Branching Lemma (EBL) by determining the axial subgroups of G . These define bifurcated solutions which are invariant under the action of the corresponding axial subgroup. We compute analytically these solutions and illustrate them as color images. Using advanced methods of numerical bifurcation analysis we then explore the persistence and stability of these solutions when varying some parameters in the model. We conjecture that we can rely on the EBL to predict the existence of patterns that survive in large parameter domains but not to predict their stability. On our way we discover the existence of spatially localized stable patterns through the phenomenon of "snaking".

A Bidirectional Link between Brain Oscillations and Geometric Patterns

Abstract:Like hallucinogenic drugs, full-field flickering visual stimulation produces regular, geometric hallucinations such as radial or spiral patterns. Computational and theoretical models have revealed that the geometry of these hallucinations can be related to functional neuro-anatomy. However, while experimental evidence links both visual flicker and hallucinogenic drugs to upward and downward modulations of brain oscillatory activity, the exact relation between brain oscillations and geometric hallucinations remains a mystery. Here we demonstrate that, in human observers, this link is bidirectional. The same flicker frequencies that preferentially induced radial (<10 Hz) or spiral (10-20 Hz) hallucinations in a behavioral experiment involving full-field uniform flicker without any actual shape displayed, also showed selective oscillatory EEG enhancement when observers viewed a genuine static image of a radial or spiral pattern without any flicker. This bidirectional property constrains the possible neuronal events at the origin of visual hallucinations, and further suggests that brain oscillations, which are strictly temporal in nature, could nonetheless act as preferential channels for spatial information.

Abstract:Understanding sensory-induced cortical patterns in the primary visual cortex V1 is an important challenge both for physiological motivations and for improving our understanding of human perception and visual organisation. In this work, we focus on pattern formation in the visual cortex when the cortical activity is driven by a geometric visual hallucination-like stimulus. In particular, we present a theoretical framework for sensory-induced hallucinations which allows one to reproduce novel psychophysical results such as the MacKay effect (Nature, 1957) and the Billock and Tsou experiences (PNAS, 2007).

Abstract:Despite decades of research, we do not definitively know how people sometimes see things that are not there. Eight models of complex visual hallucinations have been published since 2000, including Deafferentation, Reality Monitoring, Perception and Attention Deficit, Activation, Input, and Modulation, Hodological, Attentional Networks, Active Inference, and Thalamocortical Dysrhythmia Default Mode Network Decoupling. Each was derived from different understandings of brain organisation. To reduce this variability, representatives from each research group agreed an integrated Visual Hallucination Framework that is consistent with current theories of veridical and hallucinatory vision. The Framework delineates cognitive systems relevant to hallucinations. It allows a systematic, consistent, investigation of relationships between the phenomenology of visual hallucinations and changes in underpinning cognitive structures. The episodic nature of hallucinations highlights separate factors associated with the onset, persistence, and end of specific hallucinations suggesting a complex relationship between state and trait markers of hallucination risk. In addition to a harmonised interpretation of existing evidence, the Framework highlights new avenues of research, and potentially, new approaches to treating distressing hallucinations.
Cortical origins of MacKay-type visual illusions: A case for the non-linearity

Abstract:To study the interaction between retinal stimulation by redundant geometrical patterns and the cortical response in the primary visual cortex (V1), we focus on the MacKay effect (Nature, 1957) and Billock and Tsou's experiments (PNAS, 2007). We use a controllability approach to describe these phenomena starting from a classical biological model of neuronal field equations with a non-linear response function. The external input containing a localised control function is interpreted as a cortical representation of the static visual stimuli used in these experiments. We prove that while the MacKay effect is essentially a linear phenomenon (i.e., the nonlinear nature of the activation does not play any role in its reproduction), the phenomena reported by Billock and Tsou are wholly nonlinear and depend strongly on the shape of the nonlinearity used to model the response function.

How lateral inhibition and fast retinogeniculo-cortical oscillations create vision: A new hypothesis

Abstract:The role of the physiological processes involved in human vision escapes clarification in current literature. Many unanswered questions about vision include: 1) whether there is more to lateral inhibition than previously proposed, 2) the role of the discs in rods and cones, 3) how inverted images on the retina are converted to erect images for visual perception, 4) what portion of the image formed on the retina is actually processed in the brain, 5) the reason we have an after-image with antagonistic colors, and 6) how we remember space. This theoretical article attempts to clarify some of the physiological processes involved with human vision. The global integration of visual information is conceptual; therefore, we include illustrations to present our theory. Universally, the eyeball is 2.4cm and works together with membrane potential, correspondingly representing the retinal layers, photoreceptors, and cortex. Images formed within the photoreceptors must first be converted into chemical signals on the photoreceptors' individual discs and the signals at each disc are transduced from light photons into electrical signals. We contend that the discs code the electrical signals into accurate distances and are shown in our figures. The pre-existing oscillations among the various cortices including the striate and parietal cortex, and the retina work in unison to create an infrastructure of visual space that functionally "places" the objects within this "neural" space. The horizontal layers integrate all discs accurately to create a retina that is pre-coded for distance. Our theory suggests image inversion never takes place on the retina, but rather images fall onto the retina as compressed and coiled, then amplified through lateral inhibition through intensification and amplification on the OFF-center cones. The intensified and amplified images are decompressed and expanded in the brain, which become the images we perceive as external vision. Summary: This is a theoretical article presenting a novel hypothesis about the physiological processes in vision, and expounds upon the visual aspect of two of our previously published articles, "A unified 3D default space consciousness model combining neurological and physiological processes that underlie conscious experience", and "Functional representation of vision within the mind: A visual consciousness model based in 3D default space." Currently, neuroscience teaches that visual images are initially inverted on the retina, processed in the brain, and then conscious perception of vision happens in the visual cortex. Here, we propose that inversion of visual images never takes place because images enter the retina as coiled and compressed graded potentials that are intensified and amplified in OFF-center photoreceptors. Once they reach the brain, they are decompressed and expanded to the original size of the image, which is perceived by the brain as the external image. We adduce that pre-existing oscillations (alpha, beta, and gamma) among the various cortices in the brain (including the striate and parietal cortex) and the retina, work together in unison to create an infrastructure of visual space thatfunctionally "places" the objects within a "neural" space. These fast oscillations "bring" the faculties of the cortical activity to the retina, creating the infrastructure of the space within the eye where visual information can be immediately recognized by the brain. By this we mean that the visual (striate) cortex synchronizes the information with the photoreceptors in the retina, and the brain instantaneously receives the already processed visual image, thereby relinquishing the eye from being required to send the information to the brain to be interpreted before it can rise to consciousness. The visual system is a heavily studied area of neuroscience yet very little is known about how vision occurs. We believe that our novel hypothesis provides new insights into how vision becomes part of consciousness, helps to reconcile various previously proposed models, and further elucidates current questions in vision based on our unified 3D default space model. Illustrations are provided to aid in explaining our theory.
Random wiring, ganglion cell mosaics, and the functional architecture of the visual cortex

Abstract:The architecture of iso-orientation domains in the primary visual cortex of placental carnivores and primates apparently follows species invariant quantitative laws. Dynamical optimization models assuming that neurons coordinate their stimulus preferences throughout cortical circuits linking millions of cells specifically predict these invariants. This might indicate that V1's intrinsic connectome and its functional architecture adhere to a single optimization principle with high precision and robustness. To validate this hypothesis, it is critical to closely examine the quantitative predictions of alternative candidate theories. Random feedforward wiring within the retino-cortical pathway represents a conceptually appealing alternative to dynamical circuit optimization because random dimension-expanding projections are believed to generically exhibit computationally favorable properties for stimulus representations. Here, we ask whether the quantitative invariants of V1 architecture can be explained as a generic emergent property of random wiring. We generalize and examine the stochastic wiring model proposed by Ringach and coworkers, in which iso-orientation domains in the visual cortex arise through random feedforward connections between semi-regular mosaics of retinal ganglion cells (RGCs) and visual cortical neurons. We derive closed-form expressions for cortical receptive fields and domain layouts predicted by the model for perfectly hexagonal RGC mosaics [...] We conclude that V1 layout invariants are specific quantitative signatures of visual cortical optimization, which cannot be explained by generic random feedforward-wiring models. *See pdf for the full abstract.*

Spatiotemporal resonance in mouse primary visual cortex

Abstract:Human primary visual cortex (V1) responds more strongly, or resonates, when exposed to ~10, ~15-20, ~40-50 Hz rhythmic flickering light. Full-field flicker also evokes perception of hallucinatory geometric patterns, which mathematical models explain as standing-wave formations emerging from periodic forcing at resonant frequencies of the simulated neural network. However, empirical evidence for such flicker-induced standing waves in the visual cortex was missing. We recorded cortical responses to flicker in awake mice using high spatial resolution widefield imaging in combination with high temporal resolution glutamate-sensing fluorescent reporter (iGluSnFR). The temporal frequency tuning curves in the mouse V1 were similar to those observed in humans, showing a banded structure with multiple resonance peaks (8 Hz, 15 Hz, 33 Hz). Spatially, all flicker frequencies evoked responses in V1 corresponding to retinotopic stimulus location, but some evoked additional peaks. These flicker-induced cortical patterns displayed standing wave characteristics and matched linear wave equation solutions in an area restricted to the visual cortex. Taken together, the interaction of periodic traveling waves with cortical area boundaries leads to spatiotemporal activity patterns that may affect perception.

Abstract:In understanding how visual scene is processed in visual cortex, it has been an intriguing problem for theoretical and experimental neuroscientists to examine the relationship between visual stimuli and the induced responses of visual cortex. In particular, it is less explored whether and how the collective responses of visual neurons are patterned to reflect the geometrical regularities. In this paper, through a computation model and statistical analysis, we show that the orientation preference maps induced from correlated visual stimuli exhibit geometrical regularities similar as observed in natural images.
Episodic Visual Hallucinations, Inference and Free Energy

Abstract:Understandings of how visual hallucinations appear have been highly influenced by generative approaches, in particular Friston's Active Inference conceptualization. Their core proposition is that these phenomena occur when hallucinatory expectations outweigh actual sensory data. This imbalance occurs as the brain seeks to minimize informational free energy, a measure of the distance between predicted and actual sensory data in a stationary open system. We review this approach in the light of old and new information on the role of environmental factors in episodic hallucinations. In particular, we highlight the possible relationship of specific visual triggers to the onset and offset of some episodes. We use an analogy from phase transitions in physics to explore factors which might account for intermittent shifts between veridical and hallucinatory vision. In these triggered forms of hallucinations, we suggest that there is a transient disturbance in the normal one-to-one correspondence between a real object and the counterpart perception such that this correspondence becomes between the real object and a hallucination. Generative models propose that a lack of information transfer from the environment to the brain is one of the key features of hallucinations. In contrast, we submit that specific information transfer is required at onset and offset in these cases. We propose that this transient one-to-one correspondence between environment and hallucination is mediated more by aberrant discriminative than by generative inference. Discriminative inference can be conceptualized as a process for maximizing shared information between the environment and perception within a self-organizing nonstationary system. We suggest that generative inference plays the greater role in established hallucinations and in the persistence of individual hallucinatory episodes. We further explore whether thermodynamic free energy may be an additional factor in why hallucinations are temporary. Future empirical research could productively concentrate on three areas. Firstly, subjective perceptual changes and parallel variations in brain function during specific transitions between veridical and hallucinatory vision to inform models of how episodes occur. Secondly, systematic investigation of the links between environment and hallucination episodes to probe the role of information transfer in triggering transitions between veridical and hallucinatory vision. Finally, changes in hallucinatory episodes over time to elucidate the role of learning on phenomenology. These empirical data will allow the potential roles of different forms of inference in the stages of hallucinatory episodes to be elucidated.
Thalamocortical interactions reflecting the intensity of flicker light-induced visual hallucinatory phenomena

Abstract:The thalamus has a critical role in the orchestration of cortical activity. Aberrant thalamocortical connectivity occurs together with visual hallucinations in various pathologies and drug-induced states, highlighting the need to better understand how thalamocortical interactions may contribute to hallucinatory phenomena. However, concurring symptoms and physiological changes that occur during psychopathologies and pharmacological interventions make it difficult to distil the specific neural correlates of hallucinatory experiences. Flicker light stimulation (FLS) at 10 Hz reliably and selectively induces transient visual hallucinations in healthy participants. Arrhythmic flicker elicits fewer hallucinatory effects while delivering equal amounts of visual stimulation, together facilitating a well-controlled experimental setup to investigate the neural correlates of visual hallucinations driven by flicker rhythmicity. In this study, we implemented rhythmic and arrhythmic FLS during fMRI scanning to test the elicited changes in cortical activation and thalamocortical functional connectivity. We found that rhythmic FLS elicited stronger activation in higher-order visual cortices compared to arrhythmic control. Consistently, we found that rhythmic flicker selectively increased connectivity between ventroanterior thalamic nuclei and higher-order visual cortices compared to arrhythmic control, which was also found be positively associated with the subjective intensity of visual hallucinatory effects. As these thalamic and cortical areas do not receive primary visual inputs, it suggests that the thalamocortical connectivity changes relate to a higher-order function of the thalamus, such as in the coordination of cortical activity. In sum, we present novel evidence for the role of specific thalamocortical interactions with ventroanterior nuclei within visual hallucinatory experiences. Importantly, this can inform future clinical research into the mechanistic underpinnings of pathologic hallucinations.

Universality in visual cortical pattern formation

Abstract:During ontogenetic development, the visual cortical circuitry is remodeled by activity-dependent mechanisms of synaptic plasticity. From a dynamical systems perspective this is a process of dynamic pattern formation. The emerging cortical network supports functional activity patterns that are used to guide the further improvement of the network's structure. In this picture, spontaneous symmetry breaking in the developmental dynamics of the cortical network underlies the emergence of cortical selectivities such as orientation preference. Here universal properties of this process depending only on basic biological symmetries of the cortical network are analyzed. In particular, we discuss the description of the development of orientation preference columns in terms of a dynamics of abstract order parameter fields, connect this description to the theory of Gaussian random fields, and show how the theory of Gaussian random fields can be used to obtain quantitative information on the generation and motion of pinwheels, in the two dimensional pattern of visual cortical orientation columns.

Abstract:The map of orientation columns in primary visual cortex (V1) is known to show strong local distortions, with a generally smooth progression of orientation preference across extended regions of cortex, interrupted by sharp jumps (fractures) and point singularities. The map of visual space on V1, in contrast, has been assumed to be locally smooth and isotropic. We find, on the contrary, that the map of visual space on cat V1 shows strong and systematic local distortions in register with inhomogeneities in the orientation map, with the rate of receptive field movement across cortex being largely proportional to the local rate of change of orientation. This suggests possible systematic local variations in the functional connectivity of short-range lateral connections that underlie local cortical processing.

Abstract:While our understanding of the way single neurons process chromatic stimuli in the early visual pathway has advanced significantly in recent years, we do not yet know how these cells interact to form stable representations of hue. Drawing on physiological studies, we offer a dynamical model of how the primary visual cortex tunes for color, hinged on intracortical interactions and emergent network effects. After detailing the evolution of network activity through analytical and numerical approaches, we discuss the effects of the model's cortical parameters on the selectivity of the tuning curves. In particular, we explore the role of the model's thresholding nonlinearity in enhancing hue selectivity by expanding the region of stability, allowing for the precise encoding of chromatic stimuli in early vision. Finally, in the absence of a stimulus, the model is capable of explaining hallucinatory color perception via a Turing-like mechanism of biological pattern formation.

A mathematical model of the visual MacKay effect

Abstract:This paper investigates the intricate connection between visual perception and the mathematical modeling of neural activity in the primary visual cortex (V1). The focus is on modeling the visual MacKay effect [D. M. MacKay, Nature, 180 (1957), pp. 849--850]. While bifurcation theory has been a prominent mathematical approach for addressing issues in neuroscience, especially in describing spontaneous pattern formations in V1 due to parameter changes, it faces challenges in scenarios with localized sensory inputs. This is evident, for instance, in MacKay's psychophysical experiments, where the redundancy of visual stimuli information results in irregular shapes, making bifurcation theory and multiscale analysis less effective. To address this, we follow a mathematical viewpoint based on the input-output controllability of an Amari-type neural fields model. In this framework, we consider sensory input as a control function, a cortical representation via the retino-cortical map of the visual stimulus that captures its distinct features. This includes highly localized information in the center of MacKay's funnel pattern "MacKay rays". From a control theory point of view, the Amari-type equation's exact controllability property is discussed for linear and nonlinear response functions. For the visual MacKay effect modeling, we adjust the parameter representing intra-neuron connectivity to ensure that cortical activity exponentially stabilizes to the stationary state in the absence of sensory input. Then, we perform quantitative and qualitative studies to demonstrate that they capture all the essential features of the induced after-image reported by MacKay.

From hallucinations to synaesthesia: A circular inference account of unimodal and multimodal erroneous percepts in clinical and drug-induced psychosis

Abstract:Psychedelics distort perception and induce visual and multimodal hallucinations as well as synaesthesia. This is in contradiction with the high prevalence of distressing voices in schizophrenia. Here we introduce a unifying account of unimodal and multimodal erroneous percepts based on circular inference. We show that amplification of top-down predictions (descending loops) leads to an excessive reliance on priors and aberrant levels of integration of the sensory representations, resulting in crossmodal percepts and stronger illusions. By contrast, amplification of bottom-up information (ascending loops) results in overinterpretation of unreliable sensory inputs and high levels of segregation between sensory modalities, bringing about unimodal hallucinations and reduced vulnerability to illusions. We delineate a canonical microcircuit in which layer-specific inhibition controls the propagation of information across hierarchical levels: inhibitory interneurons in the deep layers exert control over priors, removing descending loops. Conversely, inhibition in the supragranular layers counterbalances the effects of the ascending loops. Overall, we put forward a multiscale and transnosographic account of erroneous percepts with important theoretical, conceptual and clinical implications.
