Abstract:BACKGROUND We describe the antimicrobial susceptibility pattern of 64 blood stream isolates of Salmonella enterica serotypes Typhi and Paratyphi A studied from January 2013 to December 2014 at a tertiary care centre in North India. METHODS Isolates were identified by standard biochemical reactions and confirmed by slide agglutination using specific antisera. Antimicrobial susceptibility testing was performed by Kirby-Bauer disc diffusion method and by E-test. RESULTS In this study, 92% (46/50) of Salmonella Typhi and all Paratyphi A (n=14) isolates were susceptible to ampicillin, chloramphenicol and cotrimoxazole. Eighty percent of Typhi (40/50) and 64% (9/14) of Paratyphi A were intermediately susceptible to ciprofloxacin. Nineteen percent (12/64) of isolates were resistant to ciprofloxacin. No resistance to ceftriaxone and azithromycin was detected. CONCLUSIONS Our study adds to the current knowledge of world-wide reports of multidrug resistance in S. Typhi.

Abstract:THIS little book, originally a thesis for the Ph.D. degree of the University of London, is written, according to the author, to dispel the mists which are gathering around our conception of the phlogiston theory: indeed, definite misstatements with regard to it are becoming increasingly common. In any event, it is useful to have in one book the full story of the wax and wane of the theory through a century of chemical history. At the outset of the eighteenth century, alchemy was flourishing and the philosopher's stone was still eagerly sought. By the end of it, quantitative chemistry had begun, chemical combination was being understood: chemistry had advanced in spite of the phlogiston theory, the work of Lavoisier had overthrown the doctrine of Stahl.

Fire-air and dephlogistication. Revisionisms of oxygen's discovery

Abstract:Americans are taught that Joseph Priestley discovered oxygen in 1774 and promptly brought that news to Lavoisier. Lavoisier proved that air contained a new element, oxygen, which combined with hydrogen to make water. He disproved the phlogiston theory but Priestley called it dephlogisticated air until his death 30 years later. Scandanavians learn that a Swedish apothecary Carl Wilhelm Scheele beat Priestley by 2 years but was deprived of credit because Lavoisier denied receiving a letter Scheele later claimed to have sent in September 1774 describing his 1772 discovery of "fire air". His claim was unconfirmed because Scheele first published his work in 1777. However, Scheele's missing letter was made public in 1992 in Paris, 218 years late, and now resides at the French Academie de Sciences. Lavoisier received it on Oct 15, 1774. His guilt was kept secret in the effects of Madame Lavoisier. He failed on several occasions to credit either Priestley or Scheele for contributing to the most important discovery in the history of science. Priestley was a teacher, political philosopher, essayist, Unitarian minister and pioneer in chemical and electrical science. He discovered 9 gases including nitrous oxide. He invented soda water, refrigeration, and gum erasers for which he coined the term "rubber". He discovered photosynthesis. He was humorless, argumentative, brilliant and passionate, called a "furious free-thinker". While his liberal colleagues Josiah Wedgwood, Erasmus Darwin, James Watts, and others of the Lunar Society were celebrating the 2nd anniversary of the French revolution, a Birmingham mob, supported by the royalists and the established church, destroyed Priestley's home, laboratory and church. Driven from England, he emigrated to Pennsylvania where he built a home and laboratory and collected a 1600 volume library, then among the largest in America. He is regarded as a founder of liberal Unitarian thinking. He was friend and correspondent of Thomas Jefferson. His philosophy and insight persuaded Jefferson to initiate what Americans call a liberal arts education. Scheele was later recognized as a brilliant and productive pioneer in chemistry although he died at age 44 of tasting his own arsenic compounds. In the new time-lapse play "Oxygen" set in Stockholm in both 18th and 21st centuries, in 1774, blame falls on Lavoisier's wife who hid Scheele's letter in hopes of giving her husband sole credit for discovering oxygen. In 2001, four Nobel committee panelists cannot agree which should receive the first "Retro-Nobel Prize" for the greatest discovery of all time: Priestley, Scheele or Lavoisier or all three. The audience is asked to choose.

Abstract:The article focuses on how the chemical concept of phlogiston functions in the so-called economy of fire from the eighteenth to twenty-first centuries, in view of a shift in “pyropolitics” (politics of fire) in their relation to the economic paradigms (cameralism, industrial capitalism, postindustrial digital economy), and theories of chemical flame processes (phlogistics, oxygen theory,theory of detonation and deflagration). The phlogiston concept is explored as the key substantial notion of the phlogictic chemical theory of the Enlightenment (regarded also as a natural “cameralistic science” in terms of metallurgy), the epistemological metaphor of ignorance in nineteenth-century Marxist discourse. Phlogiston circulates in the economics of fire discourse as a signifier for surplus value (and some other Marxist terms). “Fire as equivalent to money” becomes, in petropolitical studies, a means for turning petropolitics into pyropolitics with the radical metaphor of “PyroGaia” (Nigel Clark) in the Anthropocene period.
Interference Screw Fixation Strength of a Quadrupled Hamstring Tendon Graft Is Directly Related to Bone Mineral Density and Insertion Torque

Abstract:The purpose of this study was to determine whether bone mineral density of the host bone, measured using conventional dual photon absorptiometry techniques, and insertion torque can predict part of the ultimate failure strength of interference screw fixation of quadrupled hamstring tendon grafts. The semitendinosus and gracilis tendons were harvested from 10 human cadaveric knees, mean age 66.5 years (range, 53 to 81). The bone tunnel was sized within 0.5 mm of the graft. The graft was fixed with a biodegradable screw (7 25 mm for the femur, and 9 25 mm for the tibia) directly against the tendon and at the joint surfaces. Tibial fixation and femoral fixation were tested to failure using a materials testing system. Bone mineral density was measured in the metaphyseal region of the tibia and femur. The results of multiple regression analyses showed that both insertion torque and bone mineral density were related to the maximum load the graft withstood. These two variables explained 77.1% of the maximum load observed. We concluded that bone mineral density measurements of the host bone site are an important determinant of postoperative graft strength and thus have an important, but previously unrecognized, clinical role in establishing individual postsurgery rehabilitation protocols. Insertion torque in this study was a useful predictor of graft fixation strength.

Abstract:The phlogiston theory was established around 1700 and lasted for about one hundred years. According to the Phlogiston Theory, phlogiston is released during heating processes, and the remaining material becomes lighter. The demise of this theory started with Lavoisier's new insights into the phenomena of chemical reactions in general and combustion in particular, as well as about the composition of air. The rise and fall of the Phlogiston theory is a good example to the process of the replacement of one theory by another, due to new facts and new discoveries. In addition, it stresses the advantages and limitations of scientific models and theories, as well as the nature of science. A brief program, planned for two lessons, was developed around the Phlogiston Theory, in the framework of teaching and learning the "Science: An Ever-Developing Entity" program. Semi-structured interviews with teachers and students were conducted after the completion of the Phlogiston topic. Based on the findings, it is suggested that the brief program, reached its goals. The students, who studied the program, learned more about the scientists – their curiosity and their boldness, as well as about the scientific endeavor, consisting of discoveries, models and theories.

Abstract:In his classic work The Mind and its Place in Nature published in 1925 at the height of the development of quantum mechanics but several years after the chemists Lewis and Langmuir had already laid the foundations of the modern theory of valence with the introduction of the covalent bond, the analytic philosopher C. D. Broad argued for the emancipation of chemistry from the crass physicalism that led physicists then and later—with support from a rabblement of philosophers who knew as much about chemistry as etymologists—to believe that chemistry reduced to physics. Here Broad’s thesis is recast in terms more familiar to chemists. In the hard sell of particle physics, several prominent figures in chemistry—Hoffmann, Primas, and Pauling—have had their views interpreted to imply that they were sympathetic to greedy reductionism when in fact they were not. Indeed, being chemists without physicists as alter egos, they could not but side with Broad’s contention that chemistry, as a science that deals primarily in emergent phenomena which are beyond the purview of physicalism, owes no acquiescence to particle physics and its ethereal wares. Historically, among the most widely used expediencies in chemistry and materials science are additivity or mixture rules and their cohort transferability, all of which are devised and used under the mantle of naive reductionism. Here it is argued that while the transfer of functional groups between molecules works empirically to an extent, it is strictly outlawed by the no-cloning theorem of quantum mechanics. Several illustrative examples related to chemistry’s irreducibility to physics are presented and discussed. The failure of naive reductionism exhibited by the deep-inelastic scattering of leptons by A > 2 nuclei is traced to the same flawed reasoning that was the original basis of Moffitt’s ‘atoms in molecules’ hypothesis, the neglect of context, nuclei in the case of high-energy physics and molecules in the case of chemistry. A non-exhaustive list of other contexts from physics, chemistry, and molecular biology evidencing similar departures from the ideal of additivity or reductionism is provided for the perusal of philosophers. Had the call by the mathematician J. T. Schwartz for developments in mathematical linguistics possessed of a less single, less literal, and less simple-minded nature been met, perhaps it might have persuaded scientists to abandon their regressive fixation with unphysical reductionism and to adapt to new methodologies that engender a more nuanced handling of ubiquitous emergent phenomena as they arise in Nature than is the case today.

The revolution in physics of the early Nineteenth century revisited in the context of science-and-society interaction

Abstract:The radical changes in the concepts and approach in Physics at the turn of the Nineteenth century were so deep, that is acknowledged as a revolution. However, in 1970 Thomas Kuhn's careful reconstruction of the researches on the black body problem, the concept itself of the revolution seemed to vanish in his diluted discussion of every details. In the present paper, after an examination of the limitations of Kuhn's response to his critics, I put forward the idea, although it is not new, that these changes in Physics cannot be reduced to a point-like event, but happened instead through multiple successive (and even contradictory) changes in the course of decades. Such as the old quantum hypothesis, wave mechanics, orthodox quantum mechanics. In fact, the innovative perspectives started in the 1980s have been considered as a third quantum revolution. My basic argument is that these changes, in order to be really understood, must be interpreted not as mere specific changes in Physics, but framed in the context of the deep social, cultural, and economic changes during those turbulent years. Abstract:In 1661, Robert Boyle published his <i>Essay on Nitre</i>. In this famous essay, the author of <i>The Sceptical Chymist</i> (1661) introduces and promotes his new Corpuscular Philosophy. Central to this paper was the so-called redintegration or reconstitution experiment with saltpeter. However, this article shows that Boyle borrowed this experiment from Johann Rudolph Glauber, who had given it an alchemical interpretation. By contrast, opening the way to modern chemistry, Boyle gave it a new interpretation within the conceptual framework of his own Mechanical Philosophy. The redintegration experiment is not only important for the history of chemistry. It is very likely that the experiment also inspired Newton in his views on the composition of white light.

Abstract:Relatedness, the central concept of Hamilton's (1964a, 1964b) theory of kin selection, refers to similarity in genetic structure among groups of individuals, and plays a key role in the evolution of interactions that affect the fitness of genetically related individuals. Such interactions include, but are not limited to, interactions that are considered "social" in the usual sense of the word. For example, relatedness represents an important component of reproductive value in populations evolving sex ratio modification (Hamilton, 1972; Trivers and Hare, 1976; Uyenoyama and Bengtsson, 1981, 1982) and parthenogenesis (Williams, 1971, 1980; Maynard Smith, 1974; Uyenoyama, 1984). Relatedness within groups is determined by the rules of group formation (assortation of interacting phenotypes into demes (Hamilton, 1971)), and in the particular case of groups defined with respect to genealogical relationship it depends on the mating system. Numerous definitions of relatedness in inbred populations appear in the kin selection literature (Hamilton, 1971, 1972; Flesness, 1978; Bartz, 1979; Michod, 1979; Michod and Anderson, 1979; Aoki, 1981; Seger, 1981). Abstract:Bruno Latour’s distinctive historiographic standpoint on the origins of scientific revolutions and the multifarious accounts of their reconstructions are elicited. It is contended that, in spite of their highly iconoclastic character, Latour’s views can be welcomed for their innovative approach to the inquiries of social facets of science. In particular, they can elucidate the foundations of the lucid mature theory change model proffered in our preceding writings. Correspondingly, the Copernican Revolution is envisaged in the wayward context of intense interaction and interpenetration of Aristotelean and Ptolemaic sophisticated research practices. Eventually, the Aristotle – Ptolemy pagan cosmology could not help but be exposed to repeated cogent attacks during the Middle Ages since it apparently confronted the renowned principles of monotheism, not admitting the impervious demarcation line between the celestial and mundane realms. All the opposite worlds should have one and the same Creator. Commencing with the unification, Copernicus, in effect, paved the way for the descent of mathematics from Heaven to Earth and the spread of natural philosophy from Earth to Heaven.

Abstract:The chemico-medical essay "De sale lactis essentiali" is a thesis presented in Leyden in 1756 by a physician from Lausanne, M.-L. Vullyamoz, to obtain the medical degree. It shows that chemistry has become a university science connected with medicine and that combustion was explained at that time by G. E. Stahl's theory of the phlogiston. It reminds us that the hypotheses of this German physician, which were part of the animistic doctrine, were widely adopted in Europe. In chemistry they began to fade in 1789 after the publication of Lavoisier's work on oxydation. Nevertheless, they have contributed to establish a modern science. In medicine Stahl's animism evolved towards vitalism, which survived in several forms. Vullyamoz's thesis, which presents chemical experiences intended to analyse and promote a popular medicine, is a testimony of the spirit of Enlightenment, which rejects dogmatism and tries to understand facts through observation and the use of reason.

Abstract:The alchemical concepts of chemical symbolism, nomenclature, and affinity underwent fundamental changes between the 1770s and the 1820s, roughly simultaneously with the Chemical Revolution (ca. 1772-89), i. e. the replacement of the phlogiston theory with Lavoisier's New Chemistry. Using the old, alchemical symbols, Bergman devised a system of formulas to describe virtually all known inorganic chemistry, and he influenced Guiton de Morveau's Mémoire sur les Dénominations Chimiques, and the subsequent Méthode de nomenclature. Hassenfratz and Adet devised a new artificial sign language which, however, was too complicated and unintuitive to gain widespread acceptance. Bergman refined the concept of affinity, but his belief in phlogiston rapidly made the system obsolete. Wenzel realized that the dissolution of metals in acids is not just a question of affinity but rather of concentration, and he and Berthollet separately formulated early versions of the Law of Mass Action thereby making attempts to quantify affinity redundant. Richter formulated a principle that became known as the Law of Equivalent Proportions, describing acid-base reactions and double decompositions of salts, but continuing to use Bergman-style alchemical formulas. Only John Dalton's atomic theory with little globules denoting atoms and their combination into molecules made a definite break with the alchemical symbols.

Abstract:This paper elaborates on the following correspondence theorem (which has been defended and formally proved elsewhere): if theory T has been empirically successful in a domain of applications A, but was superseded later on by a different theory T* which was likewise successful in A, then under natural conditions T contains theoretical expressions $${\varphi}$$ which were responsible for T’s success and correspond (in A) to certain theoretical expressions $${\varphi}^{*}$$ of T*. I illustrate this theorem at hand of the phlogiston versus oxygen theories of combustion, and the classical versus relativistic theories of mass. The ontological consequences of the theorem are worked out in terms of the indirect reference and partial truth. The final section explains how the correspondence theorem may justify a weak version of scientific realism without presupposing the no-miracles argument.

Identification of an upstream region that controls the transcription of the human autocrine motility factor receptor.

Abstract:We have isolated from a human placenta cosmid library a 0.7 kb genomic clone that contains the 5' terminal portion of the autocrine motility factor receptor (hAMFR) coding region. Chloramphenicol Acetyl Transferase (CAT) reporter gene assays have identified this region as the promoter of the hAMFR gene. A single transcription initiation site (+1) has been mapped to 129 bp upstream of the ATG start codon by primer extension. DNA sequence analysis and CAT assay revealed a TATA element at the position -485/-468 which was able to conduct only a marginal transcription (less than 5% of the total activity). The majority of the hAMFR promoter's activity is contributed by a transcription initiator (Inr) element overlapping the initiation site (+1) which independently controls the transcription of the hAMFR gene. Gel mobility shift assays showed that DNA-binding proteins in HeLa cells nuclear extract can bind specifically to both promoter's elements. DNA-binding proteins were found to be differentially expressed by sparse and dense cultured normal fibroblasts. The nuclear-binding protein expressed by sparse NIH-3T3 cells induced a DNA mobility shift similarly to the nuclear protein of HeLa cells, while a different DNA-protein complex size was observed with nuclear proteins extracted from dense cultured NIH-3T3 cells. Also CAT-reporter gene analysis revealed a significant lower activity in dense NIH-3T3 cells as compared with the sparse-cultured counterparts. These results help to explain the previously observed cell-cell contact regulation of AMFR expression in normal cells and its consecutive expression in tumor cells.

Abstract:THE work of a teacher of chemistry is becoming more difficult and more perplexing every day. The mass of facts of primary importance both to the science and to technology is now so great that the amount of time that can reasonably be devoted to the business of lecturing during an ordinary College course is wholly inadequate to overtake them. The chemical student of to-day is naturally expected to have a wider range of knowledge, and a far higher standard of acquirement, than his brother of five-and-twenty years ago. The wonder is, that one small head can carry all that he is required to know. One inevitable result of this mass of material is seen in the specialization, both in work and in teaching, which is becoming increasingly apparent. There are chemists to whom the chemistry of the carbon compounds is rapidly becoming a sort of “Dark Continent,” and who begin to regard the intricacies of a structural formula with much the same feelings as they would look upon the tangled vegetation of a jungle; and, on the other hand, there are men to whom the sesquipedalian names of organic chemistry are as familiar as household words, but who are oblivious to the most ordinary facts of mineral or physical chemistry. Specialism is of course inevitable. The field is far too big to be ranged over by one man if he means to do his fair share of the work of cultivating it. But the question remains, What to teach, and how to teach it? The truth is, that as chemistry is too frequently taught to-day, the facts obscure the view of the principles. We pile up the deck-load when we ought to jettison half the cargo. What we want is, a stricter subordination of facts to principles. We need to import the methods of the statistician into our procedure. Could anything be more deadly dull, or intellectually more depressing, than the courses of so-called “advanced chemistry” professed in some of our Colleges, in which the only stimulus to mental exertion on the part of the teacher and the taught comes from the spur of the inevitable examination at the end? Not one teacher in ten seems to recognize that his first duty is to be interesting. His first duty, he will tell you, is to pass his men; and as our systems of examination are at present ordered, the passing is more a question of the facts than of the principles. And yet no one who has listened to the lectures of such men as Liebig or Hofmann or Victor Meyer can doubt for a moment that the teaching of even the most “advanced” chemistry is capable of affording a high intellectual enjoyment. But then, such men are not the slaves of a Syllabus; they are not held in bondage in Burlington Gardens. They are free to develop their own methods and to stamp their own individuality on their work. The revolt in the Nineteenth Century, the other day, might have been more successful if it had been more judiciously fought. The chemist who knows, can afford to smile at Mr. Frederic Harrison's sneer at the value of the knowledge of the number of the isomeric amyl alcohols. Of course, the bare fact of the number is not of cardinal importance, but it is evi dently not givan to Mr. Harrison to know all that is implied by that fact. In this respect at least, Mr. Harrison is a degenerate disciple. The Master's knowledge of chemistry was not bounded by the limits of a volume in the “International Scientific Series”: Comte had dabbled sufficiently deep in the science to have appreciated the real worth even of the fact, could he have lived to acquire it. But, although Mr. Harrison may shoot badly, the circumstance that he should have gone to the barricades at all is significant; and every teacher who has a soul above that of a crammer must share in his growing impatience with the present condition of things.
