Abstract:Dressings are necessary during the process of wound healing. Since the early 1980s, several types of wound dressings have been produced, but they cannot always take into account some effects include antibacterial effect, wound healing promotion, and other properties. In this study, we would like to develop an effective dressing with the above properties, especially accelerating wound healing effect. A chitosan‐calcium alginate dressing (CCAD) was developed by coating mixture of chitosan with high‐low molecular weight on calcium alginate dressing (CAD). We investigated the structural characteristics of CCAD with Fourier‐transform infrared spectroscopy (FTIR) and electron microscopy. The cytotoxicity and antibacterial property were evaluated in vitro using CCK‐8 and inhibition zone method. Moisture retention was tested on the skin of Sprague‐Dawley (SD) rats, and wound healing studies were performed on a full‐thickness skin wound model in SD rats. CCAD showed good moisturizing and antibacterial properties with no cytotoxicity. CCAD could inhibit inflammation by decreasing IL‐6, and it could also promote angiogenesis by increasing VEGF, resulting in better wound healing than CAD. CCAD is a better choice in wound care due to its antibacterial property, biocompatibility, moisture retention, healing promotion, and non‐cytotoxicity characteristics.

Abstract:Immediate hemostatic control and anti-infection are critical for wounds and have received much attention in the pharmacological fields. Whereas the development of effective wound dressings has faced the challenge with the desired combination of properties, including antimicrobial agent selection, adhesion to tissue, hemostatic performance, biocompatibility, ease of use, etc. It is known that hyaluronic acid (HA) derivatives are beneficial to promoting coagulation and wound healing, and have many functional groups in the structure, which are easy to be chemically modified. ε-Poly-(L-lysine) (ε-PL) has long-acting and broad-spectrum antibacterial properties and can bind to the aldehyde group by Schiff base reaction. Choline-based bio-ionic liquids (Bio-ILs) are structural precursors of phospholipid bilayers in cell membranes, and the adhesive strength of the polymers could be improved by conjugated with them. In addition, the humid environment also facilitates wound healing. Herein, we developed a novel wound dressing, which was a multifunctional hydrogel interwoven with two networks, one network was constructed from conjugation of Bio-ILs with methacrylated hyaluronic acid (MeHA) and self-coupling of MeHA, and the other network was formed by oxidized hyaluronic acid (OHA) and ε-PL via Schiff base reaction. In vitro studies indicated that hydrogels exhibited a three-dimensional porous structure with good swelling, antibacterial and biocompatibility properties. In addition, the rat tail amputation and wound healing experiments expressed that hydrogels had the ability of rapid hemostasis and promoting wound healing. In conclusion, Bio-IL/MeHA/OHA/ε-PL hydrogel is a naturally-derived, biocompatible and promising wound dressing.

Abstract:Over millions of years, nature has created countless unique features frequently used in the manufacture of adhesive formulations for wound healing. The bioinspired adhesives could accommodate dynamic tissue movement with excellent biocompatibility to facilitate external and internal wound healing. In this review, we summarise two types of bioinspired adhesives used for wound healing, including chemical-rationale bioinspired adhesives, such as mussel-inspired injectable hydrogel and physical-rationale bioinspired adhesives, such as octopus-inspired adhesive patches. Herein, we mainly focus on fundamental design, multi-faceted bionic strategies and potential clinical utility in surgical wound closure, haemostasis and tissue regeneration. We discuss the feasibility of incorporating nature-inspired properties into formulation design to offer a new reference for the application of bioinspired wound dressings.

Abstract:Wound healing presents a formidable challenge for global healthcare systems. We aimed to address this challenge by designing a multifunctional wound dressing tailored to meet diverse therapeutic needs. Arginine (Arg), selected for its ability to promote wound healing, is grafted onto aldehyde-modified regenerated cellulose (DAC) via Schiff base bonds for a reversible controlled release. At the same time, DAC provides hemostatic function, while Zn2+ plays an antibacterial role and strengthens cross-linking within the dressing matrix. The hydrogels were characterized by FTIR, XRD, SEM, and EDS. Mechanical strength, adhesion, swelling, water retention, oxygen permeability, hemostasis, antioxidant capacity, and antibacterial activity were all rigorously evaluated to demonstrate the superior properties of the dressing, which promotes accelerated wound healing. The skin of injured mice has been shown to recover almost completely within 13 days of dressing treatment. These findings highlight the potential of this innovative multifunctional wound dressing to address complex wound management challenges.

Abstract:Wound healing constitutes a formidable challenge within the healthcare system, attributable to infection risks and protracted recovery periods. The pressing need for innovative wound healing methods has spurred the urgency to develop novel approaches. This study sought to advance wound healing by introducing a novel approach employing a composite sponge dressing. The composite sponge dressing, derived from LFL-ZnO (synthesized through the green methodology utilizing Lactobacillus plantarum ZDY2013 fermentation liquid), polyvinyl alcohol (PVA), and sodium alginate (SA) via a freeze–thaw cycle and freeze-drying molding process, demonstrated notable properties. The findings elucidate the commendable swelling, moisturizing, and mechanical attributes of the SA/LFL-ZnO/PVA composite sponge dressing, characterized by a porous structure. Remarkably, the dressing incorporating LFL-ZnO exhibited substantial inhibition against both methicillin-resistant Staphylococcus aureus (MRSA) and Staphylococcus aureus (S. aureus). Hemolysis and cytotoxicity tests corroborated the excellent biocompatibility of the sponge dressing. In vivo evaluation of the therapeutic efficacy of the 1 mg/mL LFL-ZnO composite dressing on scald wounds and S. aureus-infected wounds revealed its capacity to accelerate wound healing and exert pronounced antibacterial effects. Consequently, the composite sponge dressings synthesized in this study hold significant potential for application in wound treatment.

Abstract:Bacterial infections, excessive reactive oxygen species (ROS) accumulation and a persistent inflammatory response severely impede the wound healing process. In this study, we developed a novel multifunctional hydrogel dressing (LCPN) based on lipoic acid modified chitosan (LAMC), polypyrrole nanoparticles (PPy NPs) and nicotinamide mononucleotide (NMN) for accelerated healing of infected wounds. The synthesized LCPN hydrogel has several properties. Modification of lipoic acid significantly enhances the water solubility of chitosan making it easier to dissolve and absorb wound secretions. Interestingly, owing to the breaking and restructuring of disulfide bonds, LCPN hydrogel can be quickly bonded under UV light without relying on photoinitiators. In addition, the incorporation of PPy NPs not only enhances the electrical conductivity of LCPN hydrogel, but also confers photothermal antimicrobial capability to LCPN hydrogel. More importantly, the sustained release of NMN in LCPN hydrogel can significantly enhance cell proliferation, migration and antioxidant capacity, which is conducive to accelerated wound healing. In vitro and in vivo experiments have shown that LCPN hydrogel has excellent biocompatibility and the ability to promote wound healing. Therefore, the prepared multifunctional hydrogel is expected to be used as a novel dressing to accelerate wound healing.

Abstract:Timely healing of acute wounds and stopping wound chronicity are current and future priorities in wound therapy. It is urgent and relevant to develop a wound dressing that has antimicrobial and monitors the wound microenvironment in real time. In this study, quaternary ammonium chitosan (HTCC) was selected as the antimicrobial agent and CS/PEO/HTCC nanofiber membranes (CPHs) were prepared by electrostatic spinning technique. The nanofiber membrane (CPH91) with the best antimicrobial performance was screened by the disk diffusion method and drug susceptibility testing by dilution method, and its antimicrobial effect on S. aureus was better than that of E. coli. Subsequently, functional carbon dots (CDs) were synthesized by solvothermal method and doped into CPH91 nanofibers by electrospinning. A good linear relationship between pH value (5.0-8.0) and the fluorescence intensity of CDs was observed. In addition, the nanofibers (CPH91@CDs) had good morphology, hydrophilicity, and biocompatibility. Changes in fluorescence intensity of CPH91@CDs at different pH (5.0-8.0) were monitored and converted into RGB values that were linearly fitted to pH value. Finally, the potential of CPH91@CDs of improving wound healing and instantaneously controlling wound healing process was confirmed by an infected wound model (S. aureus) on the back of SD rats.

Abstract:Chronic wounds and the accompanying inflammation are ongoing challenges in clinical treatment. They are usually accompanied by low pH and high oxidative stress environments, limiting cell growth and proliferation. Ordinary medical gauze has limited therapeutic effects on chronic wounds, and there is active research to develop new wound dressings. The chitosan hydrogel could be widely used in biomedical science with great biocompatibility, but the low mechanical properties limit its development. This work uses polyacrylamide to prepare double-network (DN) hydrogels based on bioadhesive catechol-chitosan hydrogels. Cystamine and N, N′-Bis(acryloyl)cystamine, which can be cross-linking agents with disulfide bonds to prepare redox-responsive DN hydrogels and pH-responsive nanoparticles (NPs) prepared by acetalized cyclodextrin (ACD) are used to intelligently release drugs against chronic inflammation microenvironments. The addition of catechol groups and ACD-NPs loaded with the Resolvin E1, promotes cell adhesion and regulates the inflammatory response at the wound site. The preparation of the DN hydrogel in this study can be used to treat and regulate the inflammatory microenvironment of chronic wounds accurately. It provides new ideas for using inflammation resolving factor loaded in DN hydrogel of good biocompatibility with enhanced mechanical properties to intelligent regulate the wound inflammation and promote the wound repaired.
