Abstract:Pure chitosan membranes present insufficient mechanical properties and a high swelling ratio, which limits their application in biomedical field. In this study, silk microfibers were obtained by chemical hydrolysis, and a novel type of chitosan/silk microfiber (CS/mSF) blended membrane was reported and its multiple physical properties were evaluated. The mechanical properties were significantly improved after blending silk microfibers with a chitosan matrix, while the swelling ratio was decreased. Observation of the surface microstructures of the blended membranes via scanning electron microscopy showed abundant embedding of mSF into the CS matrix, as well as connections among mSF. In vitro cytocompatibility was also investigated, and the blended membranes exhibited significant cytocompatibility, which was demonstrated by cell proliferation and cell morphology. Furthermore, the in vivo healing effects of the blended membrane as a wound dressing were determined on a full-thickness skin wound model of rats. Animal studies revealed that the membranes containing mSF exhibited increased wound healing efficiency compared with pure CS membranes and treatment without wound dressing. From an examination of histological changes, a higher level of epithelialization and collagen formation was observed with treatment of CS/mSF blended membranes after a 21 day repair period. In conclusion, our results indicated that the blended membranes with CS and mSF might be a potential candidate material for wound healing.

Abstract:<p>Skin damage, especially the extensive full-thickness wound, is seriously affecting people's daily life and health. Meanwhile, wound healing is always challenged by bacterial infection. In this study, for the purpose of developing a disinfectant wound dressing, we designed a novel multi-functional nanofiber mats via electrospinning combining chitosan derivations and stereocomplex crystallite (SC). The SC membrane of poly (lactic acid)/chitosan derivatives were prepared via warming at 80 °C for 1 h. The thermal and mechanical properties of the heated mats were strengthened owing to the formation of SC, which restricted the lactide chains mobility. I<em>n vivo</em> wound healing test revealed that the SC mats have better wound repair ability than the control group with a wound healing rate of 100 % within 15 days. In a word, the biomass-based mats with enhanced thermal and mechanical properties, antibacterial effect and antioxidant activity, providing a potential multi-functional platform for designing of disinfectant wound dressings.</p>

Fabrication of a core-shell nanofibrous wound dressing with an antioxidant effect on skin injury

Abstract:Oxidative stress is one of the obstacles preventing wound regeneration, especially for chronic wounds. Herein, designing a wound dressing with an anti-oxidant function is appealing for wound regeneration. In this study, a biocompatible and degradable nanofiber with a core-shell structure was fabricated via coaxial electrospinning, in which the polycaprolactone (PCL) was applied as the core structure, and a mixture made the shell of silk fibroin (SF) and tocopherol acetate (TA). The electrospun PST nanofibers were proven to have a network structure. The electrospun produced exhibited a diameter distribution with an average of 321 ± 134 nm, and the tensile strength is about 1.55 MPa, the water contact angle of its surface is 124° ± 2°. The PSTs also exhibited good tissue compatibility, which can promote the adhesion and proliferation of L929 cells. Besides, the dissolution of silk fibroin encourages the release of TA, which could play a synergistic effect and regulate the oxidative stress effect in the damaged area, for it promotes the adhesion and proliferation of skin fibroblasts (L929), reduces the cytotoxicity of hydrogen peroxide to cells, and lowers the level of reactive oxygen species. The animal experiment indicated that the PSTs would promote the reconstruction of skin. These nanofibers are expected to repair skin ulcers related to diabetes.

Abstract:Accidents and surgical procedures inevitably lead to wounds, presenting clinical challenges such as inflammation and microbial infections that impede the wound-healing process. This study aimed to address these challenges by developing a series of novel wound dressings known as electrospun biomimetic nanofiber membranes. These membranes were prepared using electrostatic spinning technique, incorporating hydroxypropyl-β-cyclodextrin/dihydromyricetin inclusion complexes. The prepared electrospun biomimetic nanofiber membranes exhibited randomly arranged fiber morphology with average fiber diameters ranging from 200 to 400 nm, resembling the collagen fibers in the native skin. These membranes demonstrated excellent biocompatibility, hemocompatibility, surface hydrophilicity, and wettability, while also releasing dihydromyricetin in a sustained manner. In vitro testing revealed that these membranes, loaded with hydroxypropyl-β-cyclodextrin/dihydromyricetin inclusion complexes, displayed higher antioxidant potential and inhibitory effects against Staphylococcus aureus and Escherichia coli . Furthermore, these membranes significantly reduced the M1 phenotypic transition in RAW264.7 cells, even when stimulated by lipopolysaccharides, effectively restoring M2 polarization, thereby shortening the inflammatory period. Additionally, the in vivo wound healing effects of these membranes were validated. In conclusion, this study introduces a promising nanofiber membrane with diverse biological properties that holds promise for addressing various crucial aspects of the wound-healing process.

Abstract:Dermal substitutes provide a template for dermal regeneration and reconstruction. They constitutes an ideal clinical treatment for deep skin defects. However, rapid vascularization remains as a major hurdle to the development and application of dermal substitutes. Several bioactive factors play an important regulatory role in the process of angiogenesis and an understanding of the mechanism of achieving their effective delivery and sustained function is vital. Nanomaterials have great potential for tissue engineering. Effective delivery of bioactive factors (including growth factors, peptides and nucleic acids) by nanomaterials is of increasing research interest. This paper discusses the process of dermal substitute angiogenesis and the roles of related bioactive factors in this process. The application of nanomaterials for the delivery of bioactive factors to enhance angiogenesis and accelerate wound healing is also reviewed. We focus on new systems and approaches for delivering bioactive factors for enhancing angiogenesis in dermal substitutes.

Abstract:Healing of various skin wounds is a lengthy process and often combined with bacterial infection and scar formation. Biomimetic electrospun nanofibrous wound dressing loaded with materials that possess properties of dual antibacterial and tissue repair would be developed to address this problem. In this study, a composite chitosan electrospun nanofibrous material containing Cur@β-CD/AgNPs nanoparticles composed of silver and curcumin possessed synergic effects on antibacterial activity and wound healing. The developed functionalized silver nanoparticles showed effective activity against both Gram-negative and Gram-positive bacteria. In vivo, Cur@β-CD/AgNPs chitosan dressing displayed enhanced wound closure rates compared to commercial AquacelAg. Moreover, Cur@β-CD/AgNPs chitosan dressing contributed to the most uniform collagen distribution by Masson's trichrome staining. In brief, Cur@β-CD/AgNPs chitosan nanofibers work as a potential wound dressing with antibacterial and antiscarring properties.

Abstract:Developing a novel nanofibrous wound dressing with multi-functional properties, integrating suitable mechanical property, electroactivity, anti-oxidant and inherent antibacterial activity to promote wound healing process is urgently desired, which could meet the increasing requirements of clinical needs. Herein, a series of antibacterial, anti-oxidant and electroactive nanofibrous membranes were fabricated by electrospinning poly(epsilon-caprolactone) (PCL) and quaternized chitosan-graft-polyaniline (QCSP) polymer solutions which combined the good mechanical property of PCL and multi-functionality of QCSP. The nanofibrous wound dressings exhibited electroactivity, similar mechanical properties to soft tissue, free radical scavenging capacity, antibacterial property, and biocompatibility. In particular, PCL/QCSP15 (15 wt% of QCSP in the sample) showed a good balanced ability between antibacterial activity and cell proliferation, which significantly accelerated the healing process of wound in a mouse full-thickness wounds defect model than commercial dressing (Tegader (TM) Film) and pure PCL (PCL/QCSP0) nanofibrous membrane. Moreover, the histopathological examination and immunofluorescence staining showed that the wounds treated by PCL/QCSP15 nanofiber dressing exhibited higher collagen deposition, granulation tissue thickness, and more angiogenesis. In one word, these antibacterial, antioxidant, electroactivity nanofibrous membranes showed promising applications for full-thickness skin repair.

Abstract:Chitosan, a naturally occurring polysaccharide with abundant resources, has been extensively exploited for various biomedical applications, typically as wound dressings owing to its unique biocompatibility, good biodegradability and excellent antibacterial properties. In this work, composite nanofibrous membranes of chitosan (CS) and silk fibroin (SF) were successfully fabricated by electrospinning. The morphology of electrospun blend nanofibers was observed by scanning electron microscopy (SEM) and the fiber diameters decreased with the increasing percentage of chitosan. Further, the mechanical test illustrated that the addition of silk fibroin enhanced the mechanical properties of CS/SF nanofibers. The antibacterial activities against Escherichia coli (Gram negative) and Staphylococcus aureus (Gram positive) were evaluated by the turbidity measurement method; and results suggest that the antibacterial effect of composite nanofibers varied on the type of bacteria. Furthermore, the biocompatibility of murine fibroblast on as-prepared nanofibrous membranes was investigated by hematoxylin and eosin (H&E) staining and MTT assays in vitro, and the membranes were found to promote the cell attachment and proliferation. These results suggest that as-prepared chitosan/silk fibroin (CS/SF) composite nanofibrous membranes could be a promising candidate for wound healing applications.

Abstract:The skin, as the largest organ, serves as a protective barrier against external stimuli. However, when the skin is injured, wound healing becomes a complex process influenced by physiological conditions, bacterial infections, and inflammation. To improve the process of wound healing, a variety of wound dressings with antibacterial qualities have been created. Electrospun nanofibers have gained significant attention in wound dressing research due to their large specific surface area and unique structure. One interesting method for creating Janus-structured nanofibers is side-by-side electrospinning. This work used side-by-side electrospinning to make cellulose acetate/gelatin Janus nanofibers. Curcumin and zinc oxide nanoparticles were added to these nanofibers. We studied Janus nanofibers' physicochemical characteristics and abilities to regulate small-molecule medication release. Janus nanofibers coated with zinc oxide nanoparticles and curcumin were also tested for antibacterial activity. The Janus nanofibers with specified physicochemical characteristics were successfully fabricated. Nanofibers released small-molecule medicines in a controlled manner. Additionally, the Janus nanofibers loaded with curcumin exhibited excellent antibacterial capabilities. This research contributes to the development of advanced wound dressings for promoting wound healing and combating bacterial infections.

Abstract:Electrospun nanofibers have been considered as relatively ideal material system for the design and construction of surgical wound dressings in the past few decades. Although there are lots of other biopolymers employed for electrospinning, chitosan is one of the most important biopolymers for the fabrication of electrospun nanofibers, due to its nontoxic, antibacterial, biocompatible as well as biodegradable properties. Electrospun chitosan nanofibers with random or aligned fiber structure can further be cross-linked by physical, chemical and ionic crosslinking methods. The as-crosslinked electrospun chitosan nanofibrous membranes possess enhanced wet stability, improved mechanical property and good biocompatibility, which are necessary for wound healing application. The present work reviews the recent studies on the design and development of electrospun chitosan nanofibers and their applications for wound healing, such as antibacterial materials, hemostatic dressings, angiogenesis and immune regulation.
Novel SA@Ca2+/RCSPs Core–shell Structure Nanofibers by Electrospinning for Wound Dressings

Abstract:Therapeutic drugs remain of great significance for the absorption of wound blood, the closure of wounds and rapid wound healing. Hence, we propose a novel composite nanofiber membrane with the above characteristics as a wound healing material. We utilize the reaction of calcium ion and alginate gel, sodium alginate (SA) and Rana chensinensis skin peptides (RCSPs) extracted from discarded Rana chensinensis skin; these two natural substances were successfully used to prepare composite nanofibers by coaxial electrospinning. The composite nanofibers are named SA@Ca2+/RCSPs nanofibers. SA@Ca2+/RCSPs nanofibers exhibited that the nanofibers contact with the liquid is unmelted, instead become gel, when compared to nanofibers of does not contain calcium ions, and the absorption rate reached 179.87%. SA@Ca2+/RCSPs nanofibers conform to the quasi-first-order dynamics model and the Ritger-Peppas release model. In vivo wound healing experiments showed that the wound-healing rate of SA@Ca2+/RCSPs nanofiber-treated wounds was 46.65% and 97.46% on days 5 and 15, respectively. In addition, SA@Ca2+/RCSPs nanofibers promoted collagen deposition and enhanced epidermal regeneration. The present study showed that composite nanofibers could quickly undergo hemostasis and effectively promote wound healing.
Smart chitosan-based nanofibers for real-time monitoring and promotion of wound healing

Abstract:Timely healing of acute wounds and stopping wound chronicity are current and future priorities in wound therapy. It is urgent and relevant to develop a wound dressing that has antimicrobial and monitors the wound microenvironment in real time. In this study, quaternary ammonium chitosan (HTCC) was selected as the antimicrobial agent and CS/PEO/HTCC nanofiber membranes (CPHs) were prepared by electrostatic spinning technique. The nanofiber membrane (CPH91) with the best antimicrobial performance was screened by the disk diffusion method and drug susceptibility testing by dilution method, and its antimicrobial effect on S. aureus was better than that of E. coli. Subsequently, functional carbon dots (CDs) were synthesized by solvothermal method and doped into CPH91 nanofibers by electrospinning. A good linear relationship between pH value (5.0-8.0) and the fluorescence intensity of CDs was observed. In addition, the nanofibers (CPH91@CDs) had good morphology, hydrophilicity, and biocompatibility. Changes in fluorescence intensity of CPH91@CDs at different pH (5.0-8.0) were monitored and converted into RGB values that were linearly fitted to pH value. Finally, the potential of CPH91@CDs of improving wound healing and instantaneously controlling wound healing process was confirmed by an infected wound model (S. aureus) on the back of SD rats.
Integration of Zn2+, ATP, and bFGF to Nanodressing with Core-Shell Structure Fabricated by Emulsion Electrospinning for Wound Healing

Abstract:Basic fibroblast growth factor (bFGF) plays an important role in active wound repair. However, the existing dosage forms in clinical applications are mainly sprays and freeze-dried powders, which are prone to inactivation and cannot achieve a controlled release. In this study, a bioactive wound dressing named bFGF-ATP-Zn/polycaprolactone (PCL) nanodressing with a "core-shell" structure was fabricated by emulsion electrospinning, enabling the sustained release of bFGF. Based on the coordination and electrostatic interactions among bFGF, ATP, and Zn2+, as well as their synergistic effect on promoting wound healing, a bFGF-ATP-Zn ternary combination system was prepared with higher cell proliferation activity and used as the water phase for emulsion electrospinning. The bFGF-ATP-Zn/PCL nanodressing demonstrated improved mechanical properties, sustained release of bFGF, cytocompatibility, and hemocompatibility. It increased the proliferation activity of human dermal fibroblasts (HDFs) and enhanced collagen secretion by 1.39 and 3.45 times, respectively, while reducing the hemolysis rate to 3.13%. The application of the bFGF-ATP-Zn/PCL nanodressing in mouse full-thickness skin defect repair showed its ability to accelerate wound healing and reduce wound scarring within 14 days. These results provide a research basis for the development and application of this bioactive wound dressing product.
Collagen Scaffolds Functionalized by Cu2+-Chelated EGCG Nanoparticles with Anti-Inflammatory, Anti-Oxidation, Vascularization, and Anti-Bacterial Activities for Accelerating Wound Healing

Abstract:Skin injury is a common health problem worldwide, and the highly complex healing process poses critical challenges for its management. Therefore, wound dressings with salutary effects are urgently needed for wound care. However, traditional wound dressing with a single function often fails to meet the needs of wound repair, and the integration of multiple functions has been required for wound repair. Herein, Cu2+-chelated epigallocatechin gallate nanoparticles (EAC NPs), with radical scavenging, inflammation relieving, bacteria restraining, and vascularization accelerating capacities, are adopted to functionalize collagen scaffold, aiming to promote wound healing. Radical scavenging experiments verify that EAC NPs could efficiently scavenge radicals. Additionally, EAC NPs could effectively remove Escherichia coli and Staphylococcus aureus. H2O2 stimuli-responsive EAC NPs show slow and sustained release properties of Cu2+. Furthermore, EAC NPs exhibit protective effects against H2O2-induced oxidative-stress damage and anti-inflammatory activity in vivo. Physicochemical characterizations show that the introduction of EAC NPs does not disrupt the gelation behavior of collagen, and the composite scaffolds (CS) remain porous structure similar to collagen scaffold. Animal experiments demonstrate that CS could promote wound healing through improving the thickness of renascent epidermis and number of new vessels. CS with multiple salutary functions is a promising dressing for wound care.

Abstract:The complicated wound repair process caused by microbial infection is still a clinical problem due to antibiotic resistance. Therefore it is necessary to employ the incorporating bioactive molecules in the dressing to solve this problem. Herein, a multifunctional nanocomposite hydrogel (CS-HCA-Icps) with the pathological pH-responsive drug release has been developed to promote the infection-impaired wound healing. CS-HCA-Icps nanocomposite hydrogel composed of catechol-grafted chitosan (CS-HCA) and a curcumin-Fe 3+ coordination nanoparticles (Icps, CurFe 3+ ) exhibits the favorable activities in free radical scavenging, anti-bacterial and anti-inflammatory. The favorable biocompatibility is also demonstrated both in vitro and in vivo experiments. These demonstrate the promoting efficacy of hydrogel in wound healing. In this study, Chitosan (CS) shows excellent biocompatibility and antibacterial properties for tissue repair. After functional modification with HCA, the catechol groups are beneficial to improve antioxidant capacity for wound repair, Moreover, Icps nanomedicine are able to enhance the loaded Cur release in response to the pathological acidic microenvironment at the inflammatory stage of wounds. Thus, the pathological pH-responsive hydrogel integrating anti-bacterial, antioxidant, and anti-inflammatory functions may represent a promising strategy for safe and efficient wound healing, in particular for potential clinical use.

Chinese traditional herbal medicine has a long history in skin wound healing, and its merging with novel approaches (nanotechnology) has resulted in more promising results. The current study aimed to combine the biological properties of a long-lasting Chinese traditional herbal medicine (Thyme) with cutting-edge technology (electrospinning) to the fabricated interactive and bioactive wound dressing. The extract of Thyme was obtained and added into the polymeric solution and converted to the nanofibrous wound dressing. The SEM analysis revealed that the fabricated nanofibers were intact without deformity with an acceptable nanometric diameter. The release kinetics evaluation showed that 80±4% of the extract was released from the nanofibers during the first 24 h. Hemolysis lower than 8% for all nanofibers revealed hemocompatibility in the fabricated wound dressings. The in vitro studies confirmed the cytocompatibility of the nanofibers. The applied animal studies exhibited that the Thyme-loaded nanofibrous dressing enhanced the wound-healing process in a dose-dependent manner. These findings demonstrate the combination of Chinese traditional herbal medicine with modern cutting-edge technology, resulting in an interactive nanofibrous mat with promising potential as the wound dressing material.

Chitosan–Collagen Electrospun Nanofibers Loaded with Curcumin as Wound-Healing Patches

Abstract:Composite chitosan–collagen nanofibrous mats embedded with curcumin were prepared via a single-step electrospinning procedure and explored as wound-healing patches with superior biological activity. A mild crosslinking protocol consisting of a short exposure to ammonia vapor and UV radiation was developed to ensure proper stability in physiological-like conditions without affecting the intrinsic biocompatibility of chitosan and collagen. The fabricated composite patches displayed a highly porous, homogeneous nanostructure consisting of fibers with an average diameter of 200 nm, thermal stability up to 200 °C, mechanical features able to ensure protection and support to the new tissues, and water-related properties in the ideal range to allow exudate removal and gas exchange. The release kinetic studies carried out in a simulated physiological environment demonstrated that curcumin release was sustained for 72 h when the mats are crosslinked hence providing prolonged bioactivity reflected by the displayed antioxidant properties. Remarkably, combining chitosan and collagen not only ensures prolonged stability and optimal physical–chemical properties but also allows for better-promoting cell adhesion and proliferation and enhanced anti-bacteriostatic capabilities with the addition of curcumin, owing to its beneficial anti-inflammatory effect, ameliorating the attachment and survival/proliferation rates of keratinocytes and fibroblasts to the fabricated patches.

Abstract:Bacterial infections and inflammation are two main factors for delayed wound healing. Coaxial electrospinning nanofibrous patches, by co-loading and sequential co-delivering of anti-bacterial and anti-inflammation agents, are promising wound dressing for accelerating wound healing. Herein, curcumin (Cur) was loaded into the polycaprolactone (PCL) core, and broad-spectrum antibacterial tetracycline hydrochloride (TH) was loaded into gelatin (GEL) shell to prepare PCL-Cur/GEL-TH core-shell nanofiber membranes. The fibers showed a clear co-axial structure and good water absorption capacity, hydrophilicity and mechanical properties. In vitro drug release results showed sequential release of Cur and TH, in which the coaxial mat showed good antioxidant activity by DPPH test and excellent antibacterial activity was demonstrated by a disk diffusion method. The coaxial mats showed superior biocompatibility toward human immortalized keratinocytes. This study indicates a coaxial nanofiber membrane combining anti-bacterial and anti-inflammation agents has great potential as a wound dressing for promoting wound repair.

Abstract:Chronic wound healing is a common clinical challenge, characterized by bacterial infection, protracted inflammatory response, oxidative stress, and insufficient neovascularization. Nanozymes have emerged as a promising solution for treating skin wounds due to their antioxidant, antibacterial, and angiogenic properties. In recent years, combining nanozymes with hydrogels to jointly promote wound healing has attracted increasing research interest. However, most of the current nanocomposite hydrogels are still not effective in simultaneously controlling inflammatory, oxidative stress and bacterial invasion in wound healing. Improving the therapeutic functional diversity and efficacy of nanocomposite hydrogels remains a problem that needs to be addressed. In this study, we prepared nanocomposite hydrogels (GelMD-Cur@ZHMCe) by combining methylacrylated gelatin modified with dopamine (GelMD) with Zinc-doped hollow mesoporous cerium oxide nanoparticles loaded with curcumin (Cur@ZHMCe). The resulting hydrogels exhibited excellent water absorption, adhesion, and biocompatibility. In vitro and in vivo studies have demonstrated that GelMD-Cur@ZHMCe has excellent antioxidant, antibacterial, anti-inflammatory and vasculature-promoting properties, which enable it to rapidly promote wound repair. The wound healing rate of the rat total skin defect infection model treated with GelMD-Cur@ZHMCe reached 98.5±4.9 % after 14 days of treatment. It was demonstrated that this multifunctional nanocomposite hydrogel provides a promising therapeutic strategy for skin repair.
