Abstract:Bacterial colonization on the surface of medical implanted devices and bacterial infection-induced biofilm have been a lethal risk for patients of clinical treatment. While antibacterial coatings fabricated by layer-by-layer (LBL) assembly techniques have been well explored, the facile preparation of substrate-independent smart antibacterial coatings with on-demand antibiotics release profile and excellent antibacterial performance is still urgently needed. In this work, this goal is addressed by LBL assembly fabrication of robust antibacterial coatings using naturally occurring and commercially available building blocks (i.e., aminoglycosides, 5,6-dihydroxyindole, and formylphenylboronic acid) via the subsequentially performed mussel-inspired polymerization and dynamic covalent chemistries. The resulting antibacterial coatings on different substates all presente a dynamic feature (i.e., pH-responsive), on-demand antibiotics release properties, and highly effective antibacterial performance both in vitro and in vivo. It is envisioned that this work can expand the scope of LBL assembly technique toward the next generation of robust and universal antibacterial coating materials by using natural building blocks and readily available chemistries.

Controlling cell adhesion using layer-by-layer approaches for biomedical applications

Abstract:Controlling the adhesion of mammalian and bacterial cells at the interfaces between synthetic materials and biological environments is a real challenge in the biomedical fields such as tissue engineering, antibacterial coating, implantable biomaterials and biosensors. The surface properties of materials are known to profoundly influence the adhesion processes. To mediate the adhesion processes, polymeric coatings have been used to functionalize surfaces to introduce diverse physicochemical properties. The polyelectrolyte multilayer films built via the layer-by-layer (LbL) method, introduced by Moehwald, Decher, and Lvov 20years ago, has led to significant developments ranging from the fundamental understanding of cellular processes to controlling cell adhesion for biomedical applications. In this review, we focus our attention on the modification of surface physicochemical properties, using the LbL approach, to construct films which can either promote or inhibit mammalian/bacterial cell adhesion. We also discuss the emerging field of multifunctional surfaces capable of responding to specific cellular activity but being inert to the others.

Abstract:The widespread occurrence of nosocomial infections and the emergence of new bacterial strands calls for the development of antibacterial coatings with localized antibacterial action that are capable of facing the challenges posed by increasing bacterial resistance to antibiotics. The Layer-by-Layer (LbL) technique, based on the alternating assembly of oppositely charged polyelectrolytes, can be applied for the non-covalent modification of multiple substrates, including medical implants. Polyelectrolyte multilayers fabricated by the LbL technique have been extensively researched for the development of antibacterial coatings as they can be loaded with antibiotics, antibacterial peptides, nanoparticles with bactericide action, in addition to being capable of restricting adhesion of bacteria to surfaces. In this review, the different approaches that apply LbL for antibacterial coatings, emphasizing those that can be applied for implant modification are presented.

Abstract:Although antibiotics have been widely used in clinical applications to treat pathogenic infections at present, the problem of drug-resistance associated with abuse of antibiotics is becoming a potential threat to human beings. We report a biohybrid nanomaterial consisting of antibiotics, enzyme, polymers, hyaluronic acid (HA), and mesoporous silica nanoparticles (MSNs), which exhibits efficient in vitro and in vivo antibacterial activity with good biocompatibility and negligible hemolytic side effect. Herein, biocompatible layer-by-layer (LBL) coated MSNs are designed and crafted to release encapsulated antibiotics, e.g., amoxicillin (AMO), upon triggering with hyaluronidase, produced by various pathogenic Staphylococcus aureus (S. aureus). The LBL coating process comprises lysozyme (Lys), HA, and 1,2-ethanediamine (EDA)-modified polyglycerol methacrylate (PGMA). The Lys and cationic polymers provided multivalent interactions between MSN-Lys-HA-PGMA and bacterial membrane and accordingly immobilized the nanoparticles to facilitate the synergistic effect of these antibacterial agents. Loading process was characterized by dynamic light scattering (DLS), transmission electron microscopy (TEM), thermogravimetric analysis (TGA), and X-ray diffraction spectroscopy (XRD). The minimal inhibition concentration (MIC) of MSN-Lys-HA-PGMA treated to antibiotic resistant bacteria is much lower than that of isodose Lys and AMO. Especially, MSN-Lys-HA-PGMA exhibited good inhibition for pathogens in bacteria-infected wounds in vivo. Therefore, this type of new biohybrid nanomaterials showed great potential as novel antibacterial agents.

Abstract:Using a combination of an aqueous layer-by-layer deposition technique, nanoparticle surface modification chemistry, and nanoreactor chemistry, we constructed thin film coatings with two distinct layered functional regions: a reservoir for the loading and release of bactericidal chemicals and a nanoparticle surface cap with immobilized bactericides. This results in dual-functional bactericidal coatings bearing both chemical-releasing bacteria-killing capacity and contact bacteria-killing capacity. These dual-functional coatings showed very high initial bacteria-killing efficiency due to the release of Ag ions and retained significant antibacterial activity after the depletion of embedded Ag because of the immobilized quaternary ammonium salts.

Abstract:This paper will update readers on the latest work in the area of antibacterial polymeric systems. There is extensive literature on existing systems. This complexity confines us to the latest antibacterial materials which possess (1) responsive antibacterial activity on their own; (2) anti-biofilm formation and (3) formation of antibacterial polymeric films. The objective of this review is to provide an overview of the antibacterial synthetic polymer field. In this paper, I will analyze the early promise of this technology as well as highlight potential challenges that adopters could face. The primary focus will be the application of materials to the medical industry and to show how these materials can be tailored to create responsive, customized bactericidal materials.

Abstract:In recent years, chitosan-based biomaterials have been continually and extensively researched by using layer-by-layer (LBL) assembly, due to their potentials in biomedicine. Various chitosan-based LBL materials have been newly developed and applied in different areas along with the development of technologies. This work reviews the recent advances of chitosan-based biomaterials produced by LBL assembly. Driving forces of LBL, for example electrostatic interactions, hydrogen bond as well as Schiff base linkage have been discussed. Various forms of chitosan-based LBL materials such as films/coatings, capsules and fibers have been reviewed. The applications of these biomaterials in the field of antimicrobial applications, drug delivery, wound dressings and tissue engineering have been comprehensively reviewed.

Abstract:In recent years, antibacterial coatings have become an important approach in the global fight against bacterial pathogens. Developments in materials science, chemistry, and biochemistry have led to a plethora of materials and chemical compounds that have the potential to create antibacterial coatings. However, insufficient attention has been paid to the analysis of the techniques and technologies used to apply these coatings. Among the various inorganic coating techniques, atomic layer deposition (ALD) is worthy of note. It enables the successful synthesis of high-purity inorganic nanocoatings on surfaces of complex shape and topography, while also providing precise control over their thickness and composition. ALD has various industrial applications, but its practical application in medicine is still limited. In recent years, a considerable number of papers have been published on the proposed use of thin films and coatings produced via ALD in medicine, notably those with antibacterial properties. The aim of this paper is to carefully evaluate and analyze the relevant literature on this topic. Simple oxide coatings, including TiO2, ZnO, Fe2O3, MgO, and ZrO2, were examined, as well as coatings containing metal nanoparticles such as Ag, Cu, Pt, and Au, and mixed systems such as TiO2-ZnO, TiO2-ZrO2, ZnO-Al2O3, TiO2-Ag, and ZnO-Ag. Through comparative analysis, we have been able to draw conclusions on the effectiveness of various antibacterial coatings of different compositions, including key characteristics such as thickness, morphology, and crystal structure. The use of ALD in the development of antibacterial coatings for various applications was analyzed. Furthermore, assumptions were made about the most promising areas of development. The final section provides a comparison of different coatings, as well as the advantages, disadvantages, and prospects of using ALD for the industrial production of antibacterial coatings.

Abstract:A "post-infiltration and subsequent photo-cross linking layer-by-layer assembly" strategy was, for the first time, introduced to fabricate carboxylated chitosan (CCS) based antibacterial multilayer films with improved stability under alkaline conditions. Precursory polyelectrolyte multilayers were assembled from CCS and poly(allylamine hydrochloride) (PAH). 4,4'-Diazostilbene-2,2'-disulfonic acid disodium salt was then infiltrated to cross-link the multilayers under UV irradiation. Analysis by UV vis spectra showed that absorbance at characteristic wavelengths of the multilayers increased almost linearly with number of CCS/PAH bilayers. AFM images indicated that surfaces of the cross-linked multilayers were rather uniform and stable against basic treatments. Surface wettability of the multilayers was determined by water contact angle measurements. The multilayers exhibited reasonable antibacterial properties against Gram-negative E. coli, demonstrating that the multilayers were active in preventing bacterial growth and represented a novel type of antibacterial films with improved stability under alkaline conditions. These results open new possibilities for building functionalized architectures onto polyelectrolyte multilayer films.

Abstract:Objectives Bacteria adhesion and subsequent biofilm formation are primary causes of implant associated infection. The biofilm makes the bacteria highly resistant to the host defense and antimicrobial treatment. Antibacterial coatings on the surface of titanium implant can prevent biofilm formation effectively, but it is still a challenge to accomplish relatively long lasting antibacterial effects before wound healing or formation of biological seal. The purpose of our work was to construct antibacterial multilayer coatings loaded with minocycline on surface of Ti substrates using chitosan and alginate based on layer-by-layer (LbL) self-assembly technique. Methods In this study, the surfaces of Ti substrates were first hydroxylated and then treated with 3-aminopropyltriethoxysilane (ATPES) to obtain amino-functionalized Ti substrates. Next, the precursor layer of chitosan was covalently conjugated to amino-functionalized Ti substrates. The following alternately coating alginate loaded with minocycline and chitosan onto the precursor layer of chitosan was carried out via LbL self-assembly technique to construct the multilayer coatings on Ti substrates. Results The multilayer coatings loaded more minocycline and improved sustainability of minocycline release to kill planktonic and adherent bacteria. Moreover, surface charge and hydrophilicity of the coatings and antibacterial ability of chitosan itself also played roles in the antibacterial performance, which can keep the antibacterial ability of the multilayer coatings after minocycline release ceases. Conclusions In conclusion, LbL self-assembly method provides a promising strategy to fabricate long-term antibacterial surfaces, which is especially effective in preventing implant associated infections in the early stage. Clinical significance Loading minocycline on the surface of implants based on LbL self-assembly strategy can endow implants with sustained antibacterial property. This can inhabit the immediate colonization of bacteria onto the surface of implants in the process of dental implant surgery, and thereby prevents and reduces the occurrence of periimplantitis.

Abstract:Antibacterial coatings have drawn much attention because of their high potential applications in medicine. However, the weak mechanical property, poor biosafety, and biocompatibility of most established antibacterial coatings restrict their applications. In this study, robust antibacterial coatings were fabricated via a simple organic-inorganic hybrid method. The polymer component provides an excellent antibacterial property to the coatings, and the hybrid silica sol improves the hardness of coatings. After cross-linking, network-structured coatings were formed. The coating surfaces exhibited high transmittance, excellent mechanical property, and good antibacterial effect against Escherichia coli (Gram-negative) and Streptococcus mutans (Gram-positive). Additionally, the robust coatings were noncytotoxic with satisfactory biocompatibility. Such results provide a theoretical basis for their applications in the biomedical field.

Abstract:Infections associated with medical devices have become a major concern. The adhesion of bacteria to the devices' surfaces during the initial 24 h is believed to be a "decisive period" for implant-associated infections, which pose key challenges to optimal antiadhesion of microbes in this period. Herein, we have designed and constructed a (heparin/chitosan)(10)-(polyvinylpyrrolidone/poly(acrylic acid))(10) [(HEP/CHI)(10)-(PVP/PAA)(10)] multilayer film by layer-by-layer self-assembly. Assembly of the underlying (HEP/CHI)(10) multilayer film is based on electrostatic interactions, showing the properties of contact killing of bacteria. Deposition of the top (PVP/PAA)(10) multilayer film is based on hydrogen bond interactions. The PAA molecules are then cross-linked to form anhydride groups by thermal treatment at 110 degrees C for 16 h. Therefore, it shows a top-down degradable capability in the determined period, leading to almost no adhesion of bacteria in 24 h. Our system combining the adhesion resistance and the contact killing properties shows an enhanced antibacterial capability through targeting the "decisive period" of implantation may have great potential for applications in medical implants, tissue engineering, etc.

Abstract:The high incidence of bacterial infection and the growing resistance of bacteria to conventional antibiotics have resulted in the strong need for the development of new generation of antibiotics. Nano-sized particles have been considered as novel antibacterial agents with high surface area and high reactivity. The overall antibacterial properties of antimicrobial nanostructures can be significantly enhanced compared with conventional antibacterial agents not in a regular nanostructure, showing a better effect in inhibiting the growth and reproduction of microbials such as bacteria and fungi, etc. In this review, recent advances in the research and applications of antimicrobial polymeric nanostructures have been highlighted, including silver-decorated polymer micelles and vesicles, antimicrobial polymer micelles and vesicles, and antimicrobial peptide-based vesicles, etc. Furthermore, we proposed the current challenges and future research directions in the field of antibacterial polymeric nanostructures for the real-world biomedical applications.

Abstract:<p>Hydrogels have emerged as promising antimicrobial materials due to their unique three-dimensional structure, which provides sufficient capacity to accommodate various materials, including small molecules, polymers and particles. Coating substrates with antibacterial hydrogel layers has been recognized as an effective strategy to combat bacterial colonization. To prevent possible delamination of hydrogel coatings from substrates, it is crucial to attach hydrogel layers <em>via</em> stronger links, such as covalent bonds. To date, various surface chemical strategies have been developed to introduce hydrogel coatings on different substrates. In this review, we first give a brief introduction of the major strategies for designing antibacterial coatings. Then, we summarize the chemical methods used to fix the antibacterial hydrogel layer on the substrate, which include surface-initiated graft crosslinking polymerization, anchoring the hydrogel layer on the surface during crosslinking, and chemical crosslinking of layer-by-layer coating. The reaction mechanisms of each method and matched pretreatment strategies are systemically documented with the aim of introducing available protocols to researchers in related fields for designing hydrogel-coated antibacterial surfaces.</p>

Abstract:Multilayered polymer particles with a wide range of properties can be precisely engineered via the layer-by-layer technique using a multitude of templates and materials. Such multilayered particles are emerging as a powerful platform for biomedical applications. In this short review, we provide an overview of developments in the engineering of multilayered polymer particles, and describe recent progress in their utilization in biological systems. Emphasis is placed on engineering strategies that achieve different biological objectives, ranging from triggered intracellular release of therapeutics to induction of protective immunity in vivo through particle-enabled vaccination. Finally, we provide a critical outlook of the key issues associated with the continued development of multilayered particles for biological applications.

Abstract:Healthcare-acquired infections as well as increasing antimicrobial resistance have become an urgent global challenge, thus smart alternative solutions are needed to tackle bacterial infections. Antibacterial materials in biomedical applications and hospital hygiene have attracted great interest, in particular, the emergence of surface design strategies offer an effective alternative to antibiotics, thereby preventing the possible development of bacterial resistance. In this review, recent progress on advanced surface modifications to prevent bacterial infections are addressed comprehensively, starting with the key factors against bacterial adhesion, followed by varying strategies that can inhibit biofilm formation effectively. Furthermore, "super antibacterial systems" through pre-treatment defense and targeted bactericidal system, are proposed with increasing evidence of clinical potential. Finally, the advantages and future challenges of surface strategies to resist healthcare-associated infections are discussed, with promising prospects of developing novel antimicrobial materials.
