Abstract:The synthesis of novel spirobenzazepinoindole derivatives has been achieved through a highly efficient and synthetic route. The approach involves a two‐step reaction, utilizing indole derivatives, 2‐amino benzyl alcohol, and ninhydrin as key starting materials under mild reaction conditions. The reaction proceeds via a sequential cascade process involving cyclization, condensation and spiro‐annulation, leading to the formation of the spirobenzazepinoindole core structure in good to excellent yields. The method offers broad substrate scope, high atom economy, and operational simplicity. The synthesized spirobenzazepinoindoles were fully characterized by spectroscopic techniques, including NMR (1H & 13C), IR and mass spectrometry. The methodology provides a valuable tool for the rapid generation of structurally complex spirobenzazepinoindoles, which could serve as scaffolds for the development of new therapeutic agents.

Abstract:Although there have been various reviews on the asymmetric construction of C3-spirooxindoles, there is a scarcity of reviews focusing on the asymmetric organocatalytic synthesis of C3-spiro-cyclopentaneoxindole derivatives. This particular scaffold has garnered significant attention from synthetic chemists due to its relevance in medicinal chemistry. In this review, we provide an overview of recent advancements in the asymmetric organocatalytic synthesis of various C3-spiro-cyclopentaneoxindoles using organic catalysts. The work is divided into sections according to the type of catalysis, including covalent catalysis (amine catalysis and N-Heterocyclic carbenes catalysis) and non-covalent catalysis (chiral phosphoric acid catalysis, hydrogen bond catalysis, and phase transfer catalysis). Furthermore, we discuss existing challenges and future directions within this field. It is our belief that this review will serve as an informative resource for researchers engaged in synthesizing C3-spiro-cyclopentaneoxindoles and inspire further advancements in this area.

Abstract:The ubiquitous presence of spirooxindole architectures with several functionalities and stereogenic centers in bioactive molecules has been appealing for the development of novel methodologies seeking their preparation in high yields and selectivities. Expansion and refinement in the field of asymmetric organocatalysis have made possible the development of straightforward strategies that address these two requisites. In this review, we illustrate the current state-of-the-art in the field of spirooxindole synthesis through the use of non-covalent organocatalysis. We aim to provide a concise overview of very recent methods that allow to the isolation of unique, densely and diversified spirocyclic oxindole derivatives with high structural diversity via the use of cascade, tandem and domino processes.

Abstract:The introduction of fluorine-containing groups into organic molecules either changes or improves the characteristics of the target compounds. On the other hand, spirocyclic-oxindoles featuring C-3 functionalized sp3-hybridized carbon atoms of three-dimensional orthogonally shaped molecules were well recognized in the core structure of varied natural products and synthetic pharmaceutical targets. Consequently, the construction of spirooxindoles by an elegant synthetic approach with efficient stereocontrol has received tremendous interest over the past decades. In this context of the synergic combination of the features associated with fluorine-containing compounds and the synthetic and medicinal efficiency associated with spirooxindoles, the stereodivergent installation of CF3 groups embedded with spirooxindoles is of increasing academic and scientific interest. This mini-review article is dedicated to demonstrating a critical overview of the recent stereoselective synthesis of spirocyclic-oxindoles featuring trifluoromethyl groups by employing the reactivity of N-2,2,2-trifluoroethylisatin ketimines as an efficient and easily prepared synthon, and covers the literature reports from 2020 to date. Besides exploring the advancements accomplished in this area, we also investigate the limitations associated with reaction discovery, mechanistic rationalization, and future applications.

Abstract:Herein, we report the discovery of the ipso-selective, dearomatizing spirocyclization of indole-tethered epoxides as a fundamentally new approach for constructing spiroindolenines equipped with three contiguous stereogenic centers under complete diastereocontrol (dr >99:1) and in high yields. Promoted by hexafluoroisopropanol, the protocol features a broad substrate scope, easy scale-up, and versatile transformations of the synthesized spiroindolenines.

Abstract:Spiroheterocycles are regarded as a privileged framework because of their wide distribution in various natural products and synthetic molecules and promising bioactivities. This review focuses on the recent advances in the synthesis of spiroheterocycles by using the strategy of N-heterocyclic carbene (NHC) organocatalysis, and is organized based on the stereoselectivity and the reactive intermediates. According to the stereochemistry, this review was divided into two main parts, covering racemic and enantioselective versions. In each part, we firstly describe the synthetic transformations using nucleophilic Breslow intermediates, and then discuss the reactions that employ electrophilic acylazolium or radical cation intermediates. With those distinct catalytic activation modes of NHC organocatlysis, we expect this synthetic protocol will possibly produce new molecules with structural novelty and complexity, which may warrant further research in the field of drug discovery.

Abstract:Spiro-heterocyclic indolenines are privileged scaffolds widely present in numerous indole alkaloids. Here, we develop a novel approach for the one-pot multistep synthesis of different spiro[indole-isoquinolines]. The protocol proposed involves the visible light mediated oxidation of N -aryl tertiary amines using bromochloroform with the generation of a reactive iminium species, which reacts with an isocyanide and an electron-rich aniline in a three-component Ugi-type reaction to give an α-aminoamidine. This compound might undergo an additional visible light-mediated oxidation to furnish a second iminium intermediate, which acts as electrophile in an intramolecular electrophilic aromatic substitution giving the final spiro-indolenine. The scope of the process has been investigated with respect to all three components. Simple operations, mild conditions, and good yields make this strategy a convenient and sustainable way to obtain novel spiro-indolenine derivatives. Beilstein J. Org. Abstract:This review centers on the synthetic strategies employed with indolyl‐ynones. Indolyl‐ynone, owing to its highly reactive ynone moiety, frequently partakes in dearomatizing spirocyclization reactions and subsequent rearrangment reactions when subjected to various reaction conditions. Hence, various heterocycles such as densly functionalized spiroindolenines, carbazoles, quinolines and carbo‐and hetero‐cycle substituted indoles are synthesized from indolyl‐ynones. In addition to this, the versatile spirocyclic indole scaffold exhibits the capability to produce tetracyclic and polyclic structures through intricate skeletal rearrangement processes. Moreover, the total synthesis of natural products using the indolyl‐ynone moiety was also compiled in this review.

Abstract:Abstract Spirooxindoles are privileged scaffolds in diverse bioactive natural products and pharmaceuticals, significant achievements for the construction of these molecules have thus been made in the past years. Among them, organocatalysis, in particular, nucleophilic Lewis base catalysis, has recently emerged as an efficient and reliable method for the preparation of diverse and valuable functionalized spirooxindoles. According to different kinds of nucleophilic catalysts, we summarize and classify three catalytic strategies; these are tertiary amine‐catalyzed cycloadditions, NHC‐catalyzed cycloadditions, and tertiary phosphine‐catalyzed cycloadditions, respectively. Through these methods, potential bioactive spirooxindole skeletons owning various functional groups can be produced to enrich the small organic molecule library. In this review, we describe a comprehensive and updated advances of nucleophilic Lewis base catalytic cycloaddition reactions for the construction of spirooxindoles. Meanwhile, the related mechanism and the application of these annulation strategies in natural product total synthesis will be highlighted in detail.

Abstract:One‐pot three‐component synthesis of biologically promising various indenoquinoxaline fused spiroheterocycles starting from 11H‐indeno[1,2‐b]quinoxalin‐11‐one or its derivatives under diverse reaction conditions. Heterocycles are very common in many naturally occurring bioactive compounds as well as in the commercially available drug molecules. Very recently spiroheterocycles have gained considerable attention due to their wide range of pharmacological applicability. Among many others, 11H‐indeno[1,2‐b]quinoxalin‐11‐one in recent time have gained significant attention as an important building block to prepare structurally diverse biologically promising spiroheterocycles under various reaction conditions. On the other hand, it was found that one‐pot three‐component reaction strategy plays an important role in the preparation of spiroheterocycles starting from 11H‐indeno[1,2‐b]quinoxalin‐11‐one as it has many advantages including high atom economy, operational simplicity, easy workup procedures, reduction of byproducts, energy and reaction step efficiency etc. In this review, we have discussed all the recent developments related to the one‐pot three‐component synthesis of biologically promising indenoquinoxaline fused spiroheterocycles starting from 11H‐indeno[1,2‐b]quinoxalin‐11‐ones under diverse reaction conditions.

Abstract:Comprehensive Summary Herein we report a condition‐controlled divergent synthesis of spiro indene‐2,1'‐isoindolinones and spiro isochroman‐3,1'‐isoindolinones through cobalt‐catalyzed formal [4 + 1] and [4 + 1 + 1] spirocyclization of aromatic amides with 2‐diazo‐1H‐indene‐1,3(2H)‐dione. When the reaction is carried out under air in ethyl acetate, spiro indene‐2,1'‐isoindolinones are formed through Co(II)‐catalyzed C−H/N−H [4 + 1] spirocyclization. When the reaction is run under O2 in CH3CN, on the other hand, spiro isochroman‐3,1'‐isoindolinones are generated through Baeyer‐Villiger oxidation of the in situ formed spiro indene‐2,1'‐isoindolinones with O2 as a cheaper and environmental‐friendly oxygen source. In general, these protocols have advantages such as using non‐precious and earth‐abundant metal catalyst, no extra additive, high efficiency and regioselectivity. Abstract:A direct spirocyclization approach for the chemical synthesis of spiro[cyclohexane-2-indoline] alkaloids is reported. The absolute stereochemistry was introduced by a desymmetrizing Dieckmann condensation, and the relative stereochemistry was controlled by the manipulation of the kinetic and thermodynamic pathways of the spirocyclization. By contrast to the biogenetic proposal involving the diester-type alkaloid as the precursor, we demonstrate that chemically a common lactone-type intermediate could bridge the chemical synthesis of this class of natural products.

Abstract:<p>We report a 2-iodoxybenzoic acid (IBX)-mediated intarmolecular oxidative spiro-fused tandem cyclization reaction of tryptophan analogs bearing an N-arylamides side-chain to rapidly afford polycyclic spiroindolines featuring multiple stereocenters including a quaternary stereocenters under mild reaction conditions. Among them, a novelty azaphosphol idine-containing spiroindoline compound is synthesized for the first time. It may open the door to azaphos pholidine-containing spiroindoline compound of potential interest in synthetic and medicinal chemistry. A plausible mechanism is proposed.</p>
