Abstract:An intramolecular organocatalytic cascade dearomatizing spirocycloaddition reaction of indole-ynone compounds containing O-silyl-naphthol substituents has been developed with the use of a chiral bifunctional thiourea. This process was able to provide various structurally diverse polycyclic spiroindolines in high yields (up to 98%) with excellent stereoselectivities (>20:1 dr, up to 98% ee) involving the formation of carbonylvinylidene ortho-quinone methide intermediates.

Abstract:<p>We report a 2-iodoxybenzoic acid (IBX)-mediated intarmolecular oxidative spiro-fused tandem cyclization reaction of tryptophan analogs bearing an N-arylamides side-chain to rapidly afford polycyclic spiroindolines featuring multiple stereocenters including a quaternary stereocenters under mild reaction conditions. Among them, a novelty azaphosphol idine-containing spiroindoline compound is synthesized for the first time. It may open the door to azaphos pholidine-containing spiroindoline compound of potential interest in synthetic and medicinal chemistry. A plausible mechanism is proposed.</p>

Abstract:A strategy for the synthesis of 6,9-dihydropyrido[1,2-a]indoles through a cascade iodocyclization of 4-(3-methyl-1H-indol-1-yl)-1,1-diphenylbut-2-yn-1-ol derivatives is presented. This reaction was conducted under very mild conditions and in a short time. The reactions are metal-free, are environmentally friendly and give up to 94% yield. Moreover, the obtained halides allow functional group diversification by palladium-catalyzed coupling reactions, which could act as potential intermediates for the synthesis of valuable compounds.

Abstract:A cascade reaction between 2‐styrylbenzaldehydes and indoles has been developed for the synthesis of 1H‐indenes. An iodine‐catalyzed nucleophilic addition of indoles to an aldehyde group of 2‐styrylbenzaldehydes leads to the formation of carbocation intermediates that undergo 4π‐electrocyclization to give indenes with a trisubstituted double bond. These indenes are transformed into thermodynamically more stable products with a fully substituted double bond under the reaction conditions. Minor quantities of indolylbenzo[b]carbazoles are also produced as by‐products.

Abstract:The development of green and efficient methods for construction of azepinoindole skeletons remains highly desirable yet challenging. Described herein is the EtOH-mediated cascade C(sp3)‒H alkylation reactions of 4-dialkylamino-indole-3-carbaldehydes for green and divergent synthesis of spirocyclic azepino[4,3,2-cd]indole derivatives. This protocol proceeded through an in situ assembly of pre-aromatics/aromatization-driven [1,6]-hydride transfer/cyclization sequence, which exhibited many advantages such as green bio-sourced EtOH as the reaction medium, metal-free and redox-neutral conditions, high step-/atom-economy, water as waste, high yields, excellent diastereoselectivites (up to ˃20:1 dr), wide substrate scope, and diverse transformations.

Abstract:Comprehensive Summary Herein we report a condition‐controlled divergent synthesis of spiro indene‐2,1'‐isoindolinones and spiro isochroman‐3,1'‐isoindolinones through cobalt‐catalyzed formal [4 + 1] and [4 + 1 + 1] spirocyclization of aromatic amides with 2‐diazo‐1H‐indene‐1,3(2H)‐dione. When the reaction is carried out under air in ethyl acetate, spiro indene‐2,1'‐isoindolinones are formed through Co(II)‐catalyzed C−H/N−H [4 + 1] spirocyclization. When the reaction is run under O2 in CH3CN, on the other hand, spiro isochroman‐3,1'‐isoindolinones are generated through Baeyer‐Villiger oxidation of the in situ formed spiro indene‐2,1'‐isoindolinones with O2 as a cheaper and environmental‐friendly oxygen source. In general, these protocols have advantages such as using non‐precious and earth‐abundant metal catalyst, no extra additive, high efficiency and regioselectivity. Abstract:A novel electrochemical multicomponent cascade reaction of indole‐tethered alkenes with CF3SO2Na, n‐Bu4NI, and H2O has been developed, which proceeds via oxidation of CF3SO2Na, coupling with alkene, spirocyclization, oxidation of sulfinate, iodide substitution, and water coupling enabling the rapid assembly of iodomethyl spiropyrrolidinyl‐oxindoles in good yields. Comprehensive Summary A novel electrochemical multicomponent cascade reaction of indole‐tethered alkenes with CF3SO2Na and n‐Bu4NI has been developed, which enables the rapid assembly of spiropyrrolidinyl‐oxindoles in good yields. The experimental results and DFT calculations suggest that this reaction proceeds through the oxidation of CF3SO2Na, radical coupling with alkene, spirocyclization, oxidation of sulfinate, iodide substitution, and water coupling. This strategy features mild reaction conditions, easy‐to‐handle reactants, and good chemical yields. This finding not only enriches the research contents of indole‐tethered alkenes but also provides a green strategy for the construction of spiropyrrolidinyl‐oxindoles compared with the existing methodologies.

Abstract:A cost-effective, highly regioselective and metal-free version for the synthesis of indolizine derivatives by means of Tf2O-mediated cascade reaction of pyridyl-enaminones and thiophenols/thioalcohols under mild reaction conditions has been reported. Diverse electron-rich indolizine derivatives could be obtained in up to 94% yield via the selective 1,4-addition of vinyl iminium triflate tandem cyclization/aromatization, which allowed the simultaneous construction of C-N and C-S/and one example of C-Se bonds.

Abstract:Spiro-heterocyclic indolenines are privileged scaffolds widely present in numerous indole alkaloids. Here, we develop a novel approach for the one-pot multistep synthesis of different spiro[indole-isoquinolines]. The protocol proposed involves the visible light mediated oxidation of N -aryl tertiary amines using bromochloroform with the generation of a reactive iminium species, which reacts with an isocyanide and an electron-rich aniline in a three-component Ugi-type reaction to give an α-aminoamidine. This compound might undergo an additional visible light-mediated oxidation to furnish a second iminium intermediate, which acts as electrophile in an intramolecular electrophilic aromatic substitution giving the final spiro-indolenine. The scope of the process has been investigated with respect to all three components. Simple operations, mild conditions, and good yields make this strategy a convenient and sustainable way to obtain novel spiro-indolenine derivatives. Beilstein J. Org.
