Abstract:In this paper, we propose a fluorescent biosensor for the sequential detection of Pb2+ ions and the cancer drug epirubicin (Epn) using the interactions between label-free guanine-rich ssDNA (LFGr-ssDNA), acridine orange (AO), and a metal-phenolic nanomaterial (i.e., nano-monoclinic copper-tannic acid (NMc-CuTA)). An exploration of the sensing mechanism shows that LFGr-ssDNA and AO strongly adsorb on NMc-CuTA through π-π stacking and electrostatic interactions, and this results in the fluorescence quenching of AO. In order to sense the target Pb2+, initially, LFGr-ssDNA specifically binds with Pb2+ ions to form a G4 complex (G-Pb2+-G base pair), which was released from the surface of NMc-CuTA with strong AO fluorescence enhancement (Turn-ON). The subsequent addition of a biothiol, like cysteine (Cys), to the G4 complex decreases the fluorescence, as the Pb2+ ions released from the G4 complex have a higher interaction affinity with the sulfur atoms of Cys; this further induces the unwinding of the G4 complex to form LFGr-ssDNA. Finally, Epn was added to this, which intercalates with LFGr-ssDNA to form a G4 complex via G-Epn-G, resulting in fluorescence recovery (Turn-ON). Accordingly, the Turn-ON fluorescent probe had subsequent limits of detection of 1.5 and 5.6 nM for Pb2+ and Epn, respectively. Hence, the reported NMc-CuTA-based sensing platform has potential applications for the detection of Pb2+ and Epn in real samples with good sensitivity and selectivity.
Advanced fluorescence-based determination of carboplatin, a potent anticancer agent, with tripeptide-functionalized copper nanoclusters

Abstract:This study introduces a straightforward photoluminescence sensor strategy employing copper nanoclusters stabilized with glutathione (GSH-CuNCs) to determine Pt-containing anticancer drug carboplatin. The interaction between the probe and carboplatin relies on the high affinity between the SH group on the surface of GSH-CuNCs and platinum (Pt) of the carboplatin. This interaction enhances the emission of nanoclusters via aggregation process. The fluorescence signal of GSH-CuNCs at 620 nm demonstrated a linear increase with carboplatin concentration within the range of 0.5–70 μg mL −1 after excitation at 380 nm. Based on a signal to noise = 3, the detection limit (LOD) was calculated as 150 ng mL −1 , signifying the sensitivity of the fluorometric probe. In addition, the probe exhibited good selectivity against potential interfering species. The viability of the approach was illustrated by its effective implementation on human serum samples containing carboplatin. This research significantly contributes to developing a simple and effective sensor for monitoring Pt-containing anticancer drugs during chemotherapy.

Abstract:A new fluorescence sensing approach has been proposed for the precise determination of the anti-cancer drug oxaliplatin (Oxal-Pt). This method entails synthesizing blue-emitting copper nanoclusters (CuNCs) functionalized with bovine serum albumin (BSA) as the stabilizing agent. Upon excitation at 360 nm, the resultant probe exhibits emission at 460 nm. Notably, the fluorescence response of BSA@CuNCs substantially increases upon incubation with Oxal-Pt due to multiple binding interactions between the drug and the fluorescent probe. These interactions involve hydrogen bonding, hydrophobic interaction, and the high affinity between the SH groups (cysteine residues of BSA) and platinum (in Oxal-Pt). Consequently, this interaction induces aggregation-induced emission enhancement (AIEE) of BSA@CuNCs. The probe demonstrates a broad response range from 0.08 to 140.0 μM, along with a low detection limit of 20.0 nM, determined based on a signal-to-noise ratio of 3. Furthermore, the probe effectively detects Oxal-Pt in injections, human serum, and urine samples, yielding acceptable results. This study represents a significant advancement in the development of a straightforward and efficient sensor for monitoring platinum-containing anti-cancer drugs during chemotherapy.

Abstract:Real-time imaging of fluctuations in intracellular glutathione (GSH) concentrations is critical to understanding the mechanism of GSH-related cisplatin-resistance. Here, we describe a ratiometric fluorescence probe based on a reversible Michael addition reaction of GSH with the vinyl-functionalized boron-dipyrromethene (4,4-difluoro-4-bora-3a,4a-diaza-s-indacene or BODIPY) 1. The probe was applied for real-time monitoring of the fluctuations in GSH levels in cells under cisplatin treatment. Notably, in cellular cisplatin-sensitive A549 cells, GSH concentrations rose until cell death, while in cisplatin-resistant cell lines, GSH levels first rose to the maximum then fell back to the initial concentration without significant apoptosis. These results indicate that different trends in GSH fluctuation can help distinguish cisplatin-resistant from cisplatin-sensitive cells. As such, this study has shown that probe 1 may potentially be used for real-time monitoring of intracellular GSH levels in response to therapeutics.
Novel CMC-CdTe / ZnS QDs Nanosensor for the Detection of Anticancer Drug Epirubicin

Abstract:Epirubicin (EPI) is one of the standard anticancer drugs that apply for various cancers treatment. However, the accumulation of EPI in the human body can be highly toxic, and it causes inevitable harm to organs. As a result, the evaluation of low concentrations of this drug in body samples requires sensitive, rapid, and accurate analysis methods. The fluorescence method is an efficient way in comparison of the traditional methods such as liquid chromatography, capillary electrophoresis, and electrochemical methods. Herein, we synthesized a novel fluorescence nanosensor named CMC-CdTe/ZnS based on using quantum dots (QDs). The structure of the prepared nanosensor is confirmed by different analysis methods such as FT-IR, TGA, and TEM. Besides that, the fluorescence intensity response of CMC-CdTe/ZnS QDs in the presence of Epirubicin drug is investigated. Based on obtained results, not only this nanosensor developed, but also the fluorescence quenching was explained by the typical Stern-Volmer equation. The best linear quenching equation for entitled nanosensor in the presence of Epirubicin is F0/F = 0.0346Q + 1.08 (R2 = 0.99), and the detection limit of Epirubicin is around 0.04 × 10-6 mol/L at 25 °C. All of the results display that this method could be reliable and suitable approach for determination of Epirubicin in commercial samples as well.

Abstract:A molecular brush based on conjugated polyelectrolyte (CPE) grafted with dense poly(ethylene glycol) (PEG) chains was successfully complexed with an anticancer agent, cisplatin, to form cisplatin-loaded nanoparticles (CPE-PEG-Pt). The obtained nanoparticles have high far-red/near-infrared fluorescence and are able to release the drug in a continuous and slow manner. These nanoparticles have not only been used to visualize HepG2 cancer cells, but also served as an in vivo fluorescent imaging probe that simultaneously tracks the in vivo drug distribution in nude mice upon intravenous administration.

Abstract:A novel chemiluminescence resonance energy transfer (CRET) system was developed and combined with a structure-switching aptamer for the highly sensitive detection of platinum. Platinum was chosen as a model analyte to demonstrate the generality of the new CRET system. This aptameric platform consisted of a streptavidin labeled aptamer against platinum and a streptavidin-coated magnetic bead for the selective separation of platinum-bound aptamer. The platinum–aptamer probe contained several guanine (G) bases bound to the 3,4,5-trimethoxyphenyl-glyoxal (TMPG) donor group at the 5′ end, a fluorescent acceptor (6-carboxy-2′,4,7,7′-tetrachlorofluorescein, TET) at the 3′ end, and a streptavidin aptamer sequence in which several base pairs were replaced by the G-G mismatch to induce the platinum-oligonucleotide coordination. The chemiluminescence (CL) generated by TMPG/G bases is transferred to the acceptor (TET). In the presence of platinum, the platinum–aptamer probe was folded such that the G bases at the 5′ end and TET at the 3′ were in close proximity. The complex was separated using streptavidin-coated magnetic beads by the addition of TMPG to form the TMPG/G bases complex. The ultraweak CL from the TMPG/G bases was strongly enhanced by TET. This novel CRET-based method can be easily performed with high limit of detection (50 ng·mL−1) and selectivity over other metal ions. This technique provides a novel method for simple, fast, and convenient point-of-care diagnostics for monitoring proteins and metal ions.

Abstract:Drug resistance poses an enormous challenge for successful chemotherapy. Glutathione S-transferase (GST) has been confirmed to be involved in the progression of drug resistance to some anticancer drugs, thus revealing that the role of GST in anticancer drug resistance is necessary. Herein, by taking advantage of frequency upconversion luminescence (FUCL) technology, we reported an FUCL probe (NRh-NDs) that can detect GST based on a rhodamine derivative structure decorated with a 2,4-dinitrobenzenesulfonyl group (NDs). The NRh-NDs showed excellent sensitivity and high selectivity for GST and released the emissive dye NRh–NH2, which showed emission and excitation wavelengths in vitro of 820 nm and 850 nm, respectively. The NRh-NDs probe successfully tested endogenic GST in U87, MCF-7 and A549 cells. The cell data showed that the increased levels of GST were positively related to cisplatin resistance but not to 5-fluorouracil resistance. These results suggested that the probe could be used as a visual tool to reveal the cause of drug resistance for cisplatin resistance in cancer treatment. Furthermore, it may serve as an effective tool to confirm the mechanism of antitumor drug resistance.

Abstract:It is still a huge challenge for concurrent highly efficient loading of chemotherapeutic agent and photosensitizer into single nanocarrier via stimuli-responsive linkages due to their different physicochemical properties and pharmacokinetics. Herein, based on the discovery of unique cisplatin-hydrazide and cisplatin-indocyanine green (ICG) coordination reactions, a multifunctional coordination nanoprodrug, cisplatin/ICG co-loaded hydrazided hyaluronan/bovine serum albumin (HBCI) nanoparticles, was developed by a desolvation-dual coordination process. The nanoprodrug exhibited ultrahigh drug loading efficiency and glutathione/NIR light dual-responsive drug release behavior. In vitro cellular studies demonstrated efficient internalization and apoptosis-inducing ability of the nanoprodrug in HepG2 cells. In vivo results confirmed the efficacious tumor accumulation and biosafety of HBCI nanoprodrug and synergistic effect of HBCI-based combined photodynamic chemotherapy on inhibiting tumor growth. Overall, this work not only provides a novel dual coordination approach for highly efficient loading of cisplatin and ICG but also verifies the therapeutic potential of HBCI nanoprodrug in combating hepatocellular carcinoma.

Abstract:A new ratiometric fluorescence strategy for sensitive and selective detection of cobalt ions (Co 2+ ) and topotecan (TOP) is proposed. The dual-probe system consists of red-emissive nitrogen and sulphur doped carbon dots (R-NS@CDs) and TOP. For Co 2+ detection, the fluorescence of R-NS@CDs at 680 nm is enhanced upon addition of Co 2+ , while complexed TOP emission peak at 545 nm remains constant. This enables ratiometric detection of Co 2+ over a range of 5.0–160.0 ng mL −1 . The mechanism of R-NS@CDs fluorescence enhancement by Co 2+ is elucidated using FTIR, fluorescence spectroscopy, zeta potential measurements, and TEM imaging. For the detection of TOP, a ratiometric probe system comprising R-NS@CDs and Co 2+ was utilized. TOP forms a complex with Co 2+ bounded to R-NS@CDs, quenching the R-NS@CDs-Co 2+ fluorescence and simultaneously the native TOP fluorescence is enhanced. This allows ratiometric quantification of TOP from 1.0-90.0 ng mL −1 . The method provides high selectivity and low detection limits of 1.51 ng mL −1 for Co 2+ and 0.37 ng mL −1 for TOP. Practical applicability is demonstrated through selective detection of Co 2+ in environmental water samples and TOP in real plasma samples. The built-in self-calibration enabled by dual analyte modulation of R-NS@CDs makes this a simple and powerful analytical approach.

Abstract:Mesoporous silica nanoparticles (MSNs) have been synthesized and loaded with both aluminum chloride phthalocyanine (AlClPc) and cisplatin as combinatorial therapeutics for treating cancer. The structural and photophysical properties of the MSN materials were characterized by different spectroscopic and microscopic techniques. Intracellular uptake and cytotoxicity were evaluated in human cervical cancer (HeLa) cells by confocal laser scanning microscopy (CLSM) and 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium (MTS) assays, respectively. The CLSM experiments showed that the MSN materials can be readily internalized in HeLa cells. The cytotoxic experiments demonstrated that, after light exposure, the combination of both AlClPc and cisplatin compounds in the same MSN platform potentiate the toxic effect against HeLa cells in comparison to the control AlClPc-MSN and cisplatin-MSN materials. These results show the potential of using MSN platforms as nanocarriers for combination photodynamic and chemotherapies to treat cancer.
Optimized Two-Photon Imaging by Stimuli-Responsive Peptide Self-Assembly Facilitates Self-Assisted Counteraction of Cisplatin-Resistance in Cancer Cells

Abstract:Accurate diagnosis and effective treatment of tumors remain significant clinical challenges. While fluorescence imaging is essential for tumor detection, it has limitations in terms of specificity, penetration depth, and emission wavelength. Here, we report a novel glutathione (GSH)-responsive peptide self-assembly excimer probe (pSE) that optimizes two-photon tumor imaging and self-assisted counteraction of the cisplatin resistance in cancer cells. The GSH-responsive self-assembly of pSE induces a monomer-excimer transition of coumarin, promoting a near-infrared redshift of fluorescence emission under two-photon excitation. This process enhances penetration depth and minimizes interference from biological autofluorescence. Moreover, the intracellular self-assembly of pSE impacts GSH homeostasis, modulates relevant signaling pathways, and significantly reduces GSTP1 expression, resulting in decreased cisplatin efflux in cisplatin-resistant cancer cells. The proposed self-assembled excimer probe not only distinguishes cancer cells from normal cells but also enhances the efficacy of cisplatin chemotherapy, offering significant potential in tumor diagnosis and overcoming cisplatin-resistant tumors.

Abstract:Multimode bioimaging probes with complementary advantages of imaging sensitivity and spatial resolution in a single drug delivery system have significant potential applications for clinical cancer diagnosis and chemotherapy. In this work, a polyethyleneimine-driven self-assembled lanthanide-doped CaF2 nanocluster was synthesized and used for fluorescence/MR imaging guided cancer chemotherapy. This nanocluster has an ordered pore structure and good cisplatin loading capacity. The release of cisplatin from this nanoplatform was triggered by acidic pH. In addition, this nanoplatform showed a high relaxation rate (4.91 mM(-1) s(-1)) and fluorescence signal. The cell assay results indicated that the nanoplatform was effectively internalized by HeLa cells and showed a good fluorescence/MR imaging ability and cell inhibition effect. In vivo experiment, the nanoplatform displayed excellent fluorescence/MR imaging and anticancer activity. In addition, the side effects of cisplatin on vital organs were significantly mitigated. Thus, this multifunctional nanoplatform is an excellent candidate for fluorescence/MR imaging guided cancer chemotherapy.
A sequential dual-locked luminescent copper nanocluster probe for tumor cell imaging and killing

Abstract:A sequential dual-locked luminescent copper nanoclusters (CuNCs) probe was designed and synthesized for the specific imaging and selective killing of tumor cells. This nanoprobe was prepared by first forming a Fe3+-coupled tannic acid (TA)-stabilized CuNCs (CuNCs-FeIII), which is in quenching state due to the electron transfer between CuNCs and Fe3+, and then coating a protectable layer of hyaluronic acid (HA) on the surface of CuNCs-FeIII to form the final dual-locked nanoprobe (CuNCs-FeIII@HA). When the nanoprobe of CuNCs-FeIII@HA target enter the tumor cells through CD44-HA receptor, HAase will first digest the HA layer of the nanoprobes, and then, GSH over-expressed in tumor cells will reduce Fe3+ to Fe2+, thus restoring the fluorescence emission of CuNCs and at the same time killing the tumor cells with the hydroxyl free radicals (∙OH) produced by the Fenton reaction between Fe2+ and H2O2. This sequential dual-locked luminescent nanoprobe of CuNCs-FeIII@HA has been successfully used for the specific imaging and selective killing of tumor cells.

Abstract:A simple process, rich information, and intelligent response are the goals pursued by cancer diagnosis and treatment. Herein, we developed a core-shell plasmonic nanomaterial (Au@MnO2-DNA), which consisted of a AuNP core with an outer shell MnO2 nanosheet decorated with fluorophore modified DNA, to achieve the aforementioned aims. On the basis of the unique optical properties of plasmonic nanoparticles and the oxidability of the shell MnO2, scattering signal and fluorescence (FL) signal changes were both related to the expression level of glutathione (GSH), for which a dual-mode imaging analysis was successfully achieved on single optical microscope equipment with one-key switching. Meanwhile, the product of Mn2+ from the reaction between MnO2 and GSH not only served as a smart chemodynamic agent to initiate Fenton-like reaction for achieving chemodynamic therapy (CDT) of cancer cells but also relieved the side effect of intracellular GSH in cancer therapy. Therefore, the core-shell plasmonic nanomaterials with dual modal switching features and diagnostic properties act as excellent probes for achieving bioanalysis of aberrant levels of intracellular GSH and simultaneously activating the CDT of cancer cells based on the in situ reactions in cancer cells.

Abstract:Nanomaterial selection is critical for photoelectrochemical (PEC) sensing. In this report, a novel cathodic photoelectrochemical (PEC) strategy was proposed for the detection of doxorubicin hydrochloride (Dox) and gentamicin sulfate (CN). The photocathode was synthesized by noncovalently coupling cadmium sulfide (CdS) to the porphyrin-derived metal-organic framework (CdS@PCN-224). This type of assembly created a pleasant interface for the combination of doxorubicin hydrochloride and gentamicin sulfate, resulting in a good CdS@PCN-224 donor-acceptor system. When compared to a single optoelectronic material, its photocurrent is enhanced by unprecedented nine times. This research could pave the way for the realization of PCN-224's enormous potential in PEC sensing.

Abstract:Ramos cells are human Burkitt's lymphoma cells, which are a kind of cancer cells to facilitate the monitoring of the relevant biological processes of cancers. Sensitive and accurate detection of Ramos cells using emerging ratiometric ECL biosensing technology shows increasing importance, however, the target analytes of current ratiometric ECL biosensors are mainly limited to DNA/RNA or proteins. In this study, we proposed a dual-potential ratiometric sensing strategy for the electrochemiluminescence detection of Ramos cells based on two types of electrochemiluminescence (ECL)-responding molecular. Au nanosucculent films (AuNFs) were electrodeposited on the fluorine doped tin oxide (FTO) electrode to increase the effective area of the electrode for more efficient assembly of DNA and effectively improving the conductivity of the sensing interfaces. In the presence of Ramos cells, aptamers capped with would conjugate with Ramos cells and then remove from AuNFs, accompanying the decrease of ECL signal from l. Then, Au–DNA was captured and alternately hybridized with DNA-modified CdS nanocrystals (NCs) on the surface of AuNFs with the formation of a super reticulate structure. The reticulate structure not only raised another identified ECL signal from CdS NCs but also greatly promoted its ECL intensity from the surface plasmon resonance originating from Au NPs. The value of log (ECLCdS/ECLluminol) and the logarithm of the number of cells exhibit considerable linear relation ranging from 80 to 8 × 105 cells mL−1 with a low detection limit of 20 cells mL−1 (S/N = 3). The selectivity and specificity of this dual-potential ECL sensor showed good performance and indicated considerable promise in avoiding false-positive results in detection.

Abstract:Inspired by dual-wavelength fluorescence ratiometric method which could reduce the influence from the environmental change, here, we present a novel dual-potential electrochemiluminescence (ECL) ratiometric sensing approach. CdS nanocrystal (NC) and luminol as two different ECL emitters are employed. ECL from CdS NCs coated on glassy carbon electrode at -1.25 V (vs SCE) could be quenched by closely contacted Pt nanoparticles (NPs) via a biological binding event, while ECL from luminol at +0.45 V (vs SCE) could be enhanced by the same Pt NPs, in the presence of their common coreactant of H₂O₂. Thus, the quenching of ECL from CdS NCs and the enhancement of ECL from luminol could indicate the same biological binding event. With the mp53 oncogene as a model DNA molecule, a molecular beacon (MB) containing a 20-base loop, which is complementary with the mp53 oncogene, is immobilized on CdS NCs/GCE first; Pt NPs are then captured on CdS NCs surface by DNA hybridization between the MB and mp53 oncogene labeled on Pt NPs. By measuring the ratio of ECL intensities at two excitation potentials, this approach could sensitively detect the concentration of target DNA in a wide range from 5.0 fM to 1.0 pM. The sensing scheme is general and can be utilized for many other biological binding events.
