Abstract:In this paper, we propose a fluorescent biosensor for the sequential detection of Pb2+ ions and the cancer drug epirubicin (Epn) using the interactions between label-free guanine-rich ssDNA (LFGr-ssDNA), acridine orange (AO), and a metal-phenolic nanomaterial (i.e., nano-monoclinic copper-tannic acid (NMc-CuTA)). An exploration of the sensing mechanism shows that LFGr-ssDNA and AO strongly adsorb on NMc-CuTA through π-π stacking and electrostatic interactions, and this results in the fluorescence quenching of AO. In order to sense the target Pb2+, initially, LFGr-ssDNA specifically binds with Pb2+ ions to form a G4 complex (G-Pb2+-G base pair), which was released from the surface of NMc-CuTA with strong AO fluorescence enhancement (Turn-ON). The subsequent addition of a biothiol, like cysteine (Cys), to the G4 complex decreases the fluorescence, as the Pb2+ ions released from the G4 complex have a higher interaction affinity with the sulfur atoms of Cys; this further induces the unwinding of the G4 complex to form LFGr-ssDNA. Finally, Epn was added to this, which intercalates with LFGr-ssDNA to form a G4 complex via G-Epn-G, resulting in fluorescence recovery (Turn-ON). Accordingly, the Turn-ON fluorescent probe had subsequent limits of detection of 1.5 and 5.6 nM for Pb2+ and Epn, respectively. Hence, the reported NMc-CuTA-based sensing platform has potential applications for the detection of Pb2+ and Epn in real samples with good sensitivity and selectivity.
Guanine-Based DNA Biosensor Amplified with Pt/SWCNTs Nanocomposite as Analytical Tool for Nanomolar Determination of Daunorubicin as an Anticancer Drug: A Docking/Experimental Investigation

Abstract:Daunorubicin is a famous anthracycline anticancer chemotherapy drug with many side effects that is very important to measure in biological samples. A daunorubicin electrochemical biosensor was fabricated in this study using ds-DNA as the biorecognition element and glassy carbon electrode (GCE) amplified by Pt/SWCNTs as a sensor. The synthetization of Pt/SWCNTs was done by the polyol method, and their characterization was accomplished via XRD, EDS, and TEM methods. The results showed a diameter of about 5.0 nm for the Pt nanoparticle decorated at the surface of SWCNTs. The morphological and conductivity properties of Pt/SWCNTs/GCE were investigated by EIS and AFM methods, and the results confirmed that Pt/SWCNTs/GCE had a high surface area and high conductivity. ds-DNA/Pt/SWCNTs/GCE showed an oxidation signal relative to that of the guanine base at the potential of 847 mV and a positive shift after interaction with the daunorubicin anticancer drug. This point confirms the intercalation reaction between the guanine base in the ds-DNA structure and the drug that could be used as an analytical factor for the determination of daunorubicin. Furthermore, molecular docking study is used to predict the interaction site of daunorubicin with DNA. It is found that daunorubicin interacts with guanine bases of DNA via an intercalative mode. Kinetic investigation showed an association equilibrium constant (K a) of about 5.044 × 103 M–1 between ds-DNA and daunorubicin. The differential pulse voltammetric results showed a linear dynamic range of 4.0 nM to 250.0 μM with a detection limit of 1.0 nM for determination of daunorubicin on the surface of ds-DNA/Pt/SWCNTs/GCE. Finally, ds-DNA/Pt/SWCNTs/GCE was successfully used for the determination of daunorubicin in injection samples with a recovery range of 98.27–10313%.

Abstract:In this work, UiO-66-NH2/GO nanocomposite was prepared using a simple solvothermal technique, and its structure and morphology were characterized using field emission scanning electron microscopy (FE-SEM), energy-dispersive X-ray spectroscopy (EDS), and X-ray diffraction (XRD). An enhanced electrochemical sensor for the detection of epirubicin (EP) was proposed, which utilized a UiO-66-NH2/GO nanocomposite-modified screen-printed graphite electrode (UiO-66-NH2/GO/SPGE). The prepared UiO-66-NH2/GO nanocomposite improved the electrochemical performance of the SPGE towards the redox reaction of EP. Under optimized experimental conditions, this sensor demonstrates a remarkable limit of detection (LOD) of 0.003 μM and a linear dynamic range from 0.008 to 200.0 μM, providing a highly capable platform for sensing EP. Furthermore, the simultaneous electro-catalytic oxidation of EP and topotecan (TP) was investigated at the UiO-66-NH2/GO/SPGE surface utilizing differential pulse voltammetry (DPV). DPV measurements revealed the presence of two distinct oxidation peaks of EP and TP, with a peak potential separation of 200 mV. Finally, the UiO-66-NH2/GO/SPGE sensor was successfully utilized for the quantitative analysis of EP and TP in pharmaceutical injection, yielding highly satisfactory results.

Abstract:In this research, glassy carbon electrode (GCE) amplified with single-wall carbon nanotubes (SWCNTs) and ds-DNA was fabricated and utilized for voltammetric sensing of doxorubicin with a low detection limit. In this technique, the reduction in guanine signal of ds-DNA in the presence of doxorubicin (DOX) was chosen as an analytical factor. The molecular docking study revealed that the doxorubicin drug interacted with DNA through intercalation mode, which was in agreement with obtained experimental results. The DOX detection performance of ds-DNA/SWCNTs/GCE was assessed at a concentration range of 1.0 nM–20.0 μM. The detection limit was found to be 0.6 nM that was comparable and even better (in many cases) than that of previous electrochemical reported sensors. In the final step, the ds-DNA/SWCNTs/GCE showed powerful ability for determination of the DOX in injection samples with acceptable recovery data.

Abstract:The use of systemic models in family therapy obliged our team to devise therapeutic tasks involving the entire family. Among these, one was found to be extremely effective: the prescribing of a family ritual. This article gives details of on such example aimed at the destruction of a myth that had been created by three generations of a family. In order that the reader may have a adequate understanding of this ritual, were shall fully describe the story of the family and of the transgenerational evolution of this myth. In the description of the treatment of the family, certain errors were made by the therapists that will come to light-errors that, as usual, were far more instructive than the actual successes. Eventually, it was the very understanding of these errors and their repercussions that led us to the successful prescription of the ritual. Finally, detailed analysis of the substance and aim of the ritual will illustrate and explain exactly what we mean by the term ritual.

Abstract:In this work, the electrochemical performance of the modified carbon paste electrode with tetrahydrodipyrazolo pyridine derivative as an organic modifier, and Cu-MCM-41 nanoparticles have been investigated to determine Capecitabine (CAP), an anticancer drug. CAP has been used for treating some cancers such as rectal, stomach, breast, pancreatic, and colon cancer. It works by inhibiting cancer cells growth. Herein, an organic modifier was prepared using a green biocatalyst in a biomimetic synthesis, a simple method was used for synthesis of Cu-MCM-41 nanoparticles. The electrochemical parameters such as the transfer coefficient and the number of electrons and protons involved in the redox reaction of the modifier were calculated using cyclic voltammetry (CV). Also, an electrocatalytic reaction was observed for CAP oxidation on the modified electrode surface as a result of a reduction in the CAP species oxidation overvoltage. To measure the CAP species by the modified electrode, some optimizations were performed. The Tafel curve was used to determine the transfer coefficient between the CAP and modifier, which was determined as 0.9. The chronoamperometric experiment was also investigated to obtain the numerical value of the CAP diffusion coefficient (D = 7.67 × 10 –6 cm 2 s −1 ). The calibration curve was drawn by the differential pulse voltammetry technique (DPV), and linear ranges of 2.0–80.0 μM and 80.0–800.0 μM, and the detection limit (LOD = 0.144 μM) were obtained. To evaluate the efficiency of the proposed sensor for CAP determination, measurement was done on a real blood serum sample. The proposed sensor displayed good selectivity, repeatability, and stability.

Abstract:In the current work, a novel and simple electrochemical sensing platform was built utilizing a graphene oxide (GO)/metal–organic framework-235 (MOF-235) nanocomposite, which was successfully synthesized and used as an excellent modifier to determine the simultaneous presence of two key anticancer medications, daunorubicin (DNR), and dacarbazine (DTIC). Fourier transform infrared spectroscopy, energy-dispersive X-ray spectroscopy, field-emission scanning electron microscopy, and scanning electron microscopy (SEM) are used to characterize the GO/MOF-235 nanocomposite. The electrocatalytic properties of the modified sensor based on MOF and GO in phosphate buffer solution (PBS) (0.1 M—pH 7.0) toward DNR redox reaction were thoroughly investigated using electrochemical methodologies including cyclic voltammetry, differential pulse voltammetry, and chronoamperometry. This GO/MOF-235-modified CPE demonstrated a wide linear response of 0.005–400.0 μM under optimized conditions. It was determined that 0.002 μM was the detection limit. This sensor displayed two distinct oxidation peaks at 390 mV for DNR and at 750 mV for dacarbazine (DTIC) (peak potential separation = 360 mV), allowing for the simultaneous detection of the two anticancer medications. Determination of these anticancer medicines in samples (DNR injection and DTIC injection) was effectively accomplished in the final step using the GO/MOF-235/CPE sensor.

A Novel Quantum Dots-Based Fluorescent Sensor for Determination of the Anticancer Dacomitinib: Application to Dosage Forms

Abstract:One of the most promising drugs recently approved for the treatment of various types of cancer is dacomitinib, which belongs to the tyrosine kinase inhibitor class. The US Food and Drugs Administration (FDA) has recently approved dacomitinib as a first-line treatment for patients suffering from non-small cell lung cancer (NSCLC) with epidermal growth factor receptor (EGFR) mutations. The current study proposes the design of a novel spectrofluorimetric method for determining dacomitinib based on newly synthesized nitrogen-doped carbon quantum dots (N-CQDs) as fluorescent probes. The proposed method is simple and does not require pretreatment or preliminary procedures. Since the studied drug does not have any fluorescent properties, the importance of the current study is magnified. When excited at 325 nm, N-CQDs exhibited native fluorescence at 417 nm, which was quantitatively and selectively quenched by the increasing concentrations of dacomitinib. The developed method involved the simple and green microwave-assisted synthesis of N-CQDs, using orange juice as a carbon source and urea as a nitrogen source. The characterization of the prepared quantum dots was performed using different spectroscopic and microscopic techniques. The synthesized dots had consistently spherical shapes and a narrow size distribution and demonstrated optimal characteristics, including a high stability and a high fluorescence quantum yield (25.3%). When assessing the effectiveness of the proposed method, several optimization factors were considered. The experiments demonstrated highly linear quenching behavior across the concentration range of 1.0-20.0 μg/mL with a correlation coefficient (r) of 0.999. The recovery percentages were found to be in the range of 98.50-100.83% and the corresponding relative standard deviation (%RSD) was 0.984. The proposed method was shown to be highly sensitive with a limit of detection (LOD) as low as 0.11 μg/mL. The type of mechanism by which quenching took place was also investigated by different means and was found to be static with a complementary inner filter effect. For quality purposes, the assessment of the validation criteria adhered to the ICHQ2(R1) recommendations. Finally, the proposed method was applied to a pharmaceutical dosage form of the drug (Vizimpro® Tablets) and the obtained results were satisfactory. Considering the eco-friendly aspect of the suggested methodology, using natural materials to synthesize N-CQDs and water as a diluting solvent added to its greenness profile.
Advanced fluorescence-based determination of carboplatin, a potent anticancer agent, with tripeptide-functionalized copper nanoclusters

Abstract:This study introduces a straightforward photoluminescence sensor strategy employing copper nanoclusters stabilized with glutathione (GSH-CuNCs) to determine Pt-containing anticancer drug carboplatin. The interaction between the probe and carboplatin relies on the high affinity between the SH group on the surface of GSH-CuNCs and platinum (Pt) of the carboplatin. This interaction enhances the emission of nanoclusters via aggregation process. The fluorescence signal of GSH-CuNCs at 620 nm demonstrated a linear increase with carboplatin concentration within the range of 0.5–70 μg mL −1 after excitation at 380 nm. Based on a signal to noise = 3, the detection limit (LOD) was calculated as 150 ng mL −1 , signifying the sensitivity of the fluorometric probe. In addition, the probe exhibited good selectivity against potential interfering species. The viability of the approach was illustrated by its effective implementation on human serum samples containing carboplatin. This research significantly contributes to developing a simple and effective sensor for monitoring Pt-containing anticancer drugs during chemotherapy.

Preparation of Bioconjugates of CdTe Nanocrystals for Cancer Marker Detection

Abstract:Highly fluorescent CdTe quantum dots (Q-dots) stabilized by 3-mercaptopropionic acid (MPA) were prepared by an aqueous solution approach and used as fluorescent labels in detecting a cancer marker, carcinoembryonic antigen (CEA), expressed on human colon carcinoma cell line LS 180. Nonspecific adsorptions of CdTe Q-dots on carcinoma cells were observed and effectively eliminated by replacing MPA with a thiolated PEG ( poly( ethylene glycol), Mn = 750) synthesized according to literature. It was unexpectedly found out that the PEG- coated CdTe Q-dots exhibited very strong and specific affinity to anti-CEA monoclonal antibody rch 24 ( rch 24 mAb). The resultant CdTe -(rch 24 mAb) conjugates were successfully used in detections of CEA expressed on the surface of cell line LS 180. Further experiments demonstrated that the fluorescent CdTe Q-dots exhibited much better photostability and a brighter fluorescence than FITC, which consequently led to a higher efficiency in the cancer marker detection.
Ratiometric Fluorescence Nanosensors Based on Core- Shell Structured Carbon/ CdTe Quantum Dots and Surface Molecularly Imprinted Polymers for the Detection of Sulfadiazine

Abstract:Sulfadiazine is an environmental pollutant derived from abuse of antibiotics. Its content in environmental water is closely related to human health. Thus, a novel dual-emission surface molecularly imprinted nanosensor is designed for the specific adsorption and detection of sulfadiazine. In the system, blue emissive carbon quantum dots wrapped with silica served as the internal reference signal for eliminating background interference, while red emissive thioglycolic acid modified CdTe quantum dots (CdTe QDs), which are low dimensional semiconductor materials by the combination of cadmium and tellurium with excellent optical properties, were encapsulated in the imprinted layer to offer recognition signal. The fluorescence of CdTe quantum dots was quenched and the fluorescence quenching degree of carbon quantum dotswas inconspicuous with the increase of concentration of sulfadiazine, thereby reflecting the color change. The detection of sulfadiazine was successfully achieved in a concentration range of 0.25-20 mu mol/L with detection limit of 0.042 mu mol/L and nanosensors had specific recognition for sulfadiazine over its analogues. Compared to single-emission fluorescence sensors, ratiometric fluorescence nanosensors had wider linear range and higher detection accuracy. Furthermore, the nanosensors were also successfully applied for the determination of sulfadiazine in real water and milk samples with acceptable recoveries. The study provides a feasible method for the detection of sulfadiazine and a reference for the detection of sulfonamides.
Ultrathin MXene nanosheet-based TiO2/CdS heterostructure as a photoelectrochemical sensor for detection of CEA in human serum samples

Abstract:To develop highly accurate and ultrasensitive strategies is of great importance for the clinical measurement, in particular, the detection of cancer biomarkers. Herein, we synthesized an ultrasensitive TiO2/MXene/CdS QDs (TiO2/MX/CdS) heterostructure as a photoelectrochemical immunosensor, which favors energy levels matching and fast electron transfer from CdS to TiO2 in the help of ultrathin MXene nanosheet. Dramatic photocurrent quenching can be observed upon incubation of the TiO2/MX/CdS electrode by Cu2+ solution from 96-well microplate, which caused by the formation of CuS and subsequent CuxS (x = 1, 2), reducing the absorption of light and boosting the electron-hole recombination upon irradiation. As a result, the as-prepared biosensor demonstrates a linearly increased photocurrent quenching percentage (Q%) value with CEA concentration ranging from 1 fg/mL to 10 ng/mL, as well as a low detection limit of 0.24 fg/mL. Benefit from its excellent stability, high selectivity and good reproducibility of as-prepared PEC immunosensor, we believe that this proposed strategy might provide new opportunities for clinical diagnosis of CEA and other tumor markers.

Utilizing Copper Nanoclusters as a Fluorescent Probe for Quantitative Monitoring of Doxorubicin Anticancer Drug

Abstract:Monitoring the amount of chemotherapeutic drugs in biological fluids is extremely important for dose adjustment or control of side effects during the treatment process. In this study, copper nanoclusters (Cu NCs) were synthesized via a one-pot method using ammonium citrate as the reducing agent. Cu NCs exhibited bright blue fluorescence, good optical properties and outstanding photostability. The produced Cu NCs were characterized in detail by UV‒vis absorption, fluorescence spectroscopy and transmission electron microscopy (TEM). The produced Cu NCs showed a high quantum yield of 0.97. A fluorescence system was used for doxorubicin (DOX) determination using Cu NCs as a nanoprobe. The presence of DOX decreased the fluorescence intensity of the CuNCs at 445 nm but increased the fluorescence intensity of the CuNCs at 619 nm. As a result, quantitative detection of DOX can be achieved by measuring the ratio of fluorescence intensities at 445 and 619 nm (F619/F445). The fluorescence quenching activity of the Cu NCs was determined to have a linear relationship with the amount of DOX anticancer drug in the range of 1–15 ppb, and the usability of the Cu NCs as a sensor for detection in biological fluids was demonstrated. It was determined that this method can be used to measure the amount of DOX in biological samples effectively.

Abstract:The accurate and efficient quantification of nanodrug dosage is crucial for early anticancer therapy. The enzyme-linked immunosorbent assay (ELISA) has emerged as a robust tool for detecting anticancer nanodrug dosage; however, the development of sensing elements to quantify anticancer nanodrugs still poses a challenge. To overcome this problem, we utilize polysuccinimide-loaded curcumin (CUR @PSIOAm) as a model to employ an ELISA based on peroxidase nanozyme Pt-SiO2 Janus nanoparticles (Pt-SiO2 JNPs) for the indirect quantitative analysis of intracellular anticancer nanodrug dosage. This novel approach employs an immunoassay to indirectly quantify the dosage of anticancer nanodrugs while preserving its structural integrity. The silica components of Pt-SiO2 JNPs adsorb intermediates, while the Pt NP components exhibit high catalytic activity. Pt-SiO2 JNPs are functionalized with anti-PSIOAm antibody (Pt-SiO2 JNPs-Ab) to serve as an immunosensor capable of specific recognition of CUR @PSIOAm. Additionally, we employed cytotoxicity assays and confocal imaging techniques to demonstrate the excellent biocompatibility of CUR @PSIOAm, as well as its specific uptake by cancer cells. According to the experimental results, the limit of detection (LOD) for the immunoassay of Pt-SiO2 JNPs as a marker for detecting CUR @PSIOAm is approximately 4.5-fold lower than that of horseradish peroxidase. Therefore, by optimizing the conditions, we established a direct competitive ELISA using Pt-SiO2 JNPs as colorimetric indicators for the quantitative detection of intracellular CUR @PSIOAm. The LOD for this ELISA was determined to be 0.01 ng/mL, while the loaded CUR amount calculated from the drug loading capacity was found to be 0.22 pg/mL. Furthermore, the recoveries obtained from this established ELISA ranged between 94.0 and 108%, demonstrating excellent accuracy. Consequently, the peroxidase mimic Pt-SiO2 JNPs-based ELISA exhibits significant potential for precise quantification of intracellular anticancer nanodrug dosages.

Abstract:The cell surface protein epithelial cell adhesion molecule (EpCAM) has oncogenic effects, and its overexpression is strongly correlated with tumor development. Here, an electrochemical method is proposed to detect and identify EpCAM at the level of purified proteins and living cells. The core concept of this method is the use of DNA nanotetrahedron (NTH)-based capture probes and aptamer-modified quasi-ZIF-67@Au@methylene blue (QZIF-67@Au@MB@APT) as signal probes. In our design, NTH-assisted targeted immobilization can greatly improve the accessibility of nucleotide-containing artificial aptamers to suspended targets and the specificity of the target EpCAM. Moreover, following partial deligandization, ZIF-67 was converted to QZIF-67 with an extensively improved porous structure that significantly enhanced the loading efficiency of signaling molecules and thus led to a remarkable increase in detection sensitivity. QZIF-67@Au@MB can be integrated with an aptamer to act as a signal reporter with an electrochemical sensing platform for EpCAM detection. By tracing the electrochemical signals from the QZIF-67@Au@MB@APT, the method demonstrated the detection of targets as low as 3 pg/mL within a wide linear range from 0.005 to 100 ng/mL. We have successfully applied this aptasensor for in situ characterization of EpCAM on the cellular surface and monitoring alterations in EpCAM expression during drug treatment, which provides a low-cost but robust alternative to highly expensive and experience-dependent flow cytometry. Overall, electrochemical characterization may provide promising technical support for the precise management of tumors.

Abstract:For the first time, a high-efficient and low-cost electrochemiluminescence (ECL) biosensor was constructed for ultrasensitive detection of breast cancer related human epidermal growth factor receptor-2 (HER-2) using a novel binary luminophore and low concentration of coreactant. Firstly, Au nanoparticles decorated graphite-like carbon nitride (g-C3N4@Au) was designed as sensing platform, which can generate ECL signal in the potential range from −1.2 V to 0 V when peroxydisulfate (S2O82−) was used as the coreactant. Secondly, amino-modified capture probes (CP) were immobilized on the surface of g-C3N4@Au via Au-N bonds. In addition, Zr-based metal organic framework (Zr-MOF) modified bulk boron carbon oxynitride (BCNO), named ZMB, exhibited remarkable ECL intensity at the potential window of −2.0 V to 0 V with 0.8 mM S2O82−. ZMB possessed a large specific surface area and abundant amino groups to anchor Au@Pt nanoparticles (Au@PtNPs). Therefore, HER-2 binding aptamers (HBA) could be anchored on Au@PtNPs to form the tracer label. Under optimal conditions, the proposed ECL biosensor displayed a wide linear range from 10 fg mL−1 to 100 ng mL−1 with a limit of detection (LOD) as low as 0.2 fg mL−1. Importantly, this work utilized ZMB/S2O82− as new ECL system with satisfactory detection performances, which provided a potential approach to improve the sensitivity in clinical diagnosis.

Abstract:A new bimetallic CuCo Prussian blue analogue (CuCo PBA) loaded with carbon dots (CDs) was prepared (represented by CD@CuCoPBA) and developed as a scaffold for anchoring the epidermal growth factor receptor (EGFR) aptamer to detect EGFR and living EGFR-overexpressed cancer cells. The basic characterizations revealed CuCo PBA exhibited nanocube shape and still remained its nanostructure and physical/chemical properties after coupling with large amounts of CDs. As compared with the pristine CuCo PBA, the CD@CuCoPBA displayed good electrochemical activity, strong binding interaction toward aptamer, and high stability of aptamer-EGFR G-quadruplex in aqueous solution. As such, the results of electrochemical impedance spectroscopy measurements indicated that the CD@CuCoPBA-based aptasensor displayed an ultra-low detection limit toward EGFR (0.42 fg mL(-1)) and living EGFR-overexpressed MCF-7 cancer cells (80 cell per mL), as well as high selectivity, good reproducibility, high stability, repeatability, and acceptable applicability. Consequently, the constructed CD@CuCoPBA-based aptasensor can be extended to be a promising universal method for early diagnosis of cancers.
Sensitive Electrochemical Aptasensor for Detecting EpCAM with Silica Nanoparticles and Quantum Dots for Signal Amplification

Abstract:The epithelial cell adhesion molecule (EpCAM) is a known biomarker of circulating tumor cells that plays an important role in tumor metastasis. The detection of EpCAM is vital for personalized diagnosis and therapy but is challenging because of its extremely low concentration in peripheral blood. In our work, an electrochemical aptasensor has been developed to quantitatively detect EpCAM at low concentrations. A gold electrode modified with an aptamer can capture EpCAM, which can then be further recognized by a second aptamer acting as a signal reporter. To amplify the signal, a silica nanoparticle/CdSe complex has been designed to enhance the detection system. First, silica nanoparticles were synthesized to act as carriers for loading many CdSe quantum dots which are capable of binding with the second aptamer by biotin-streptavidin interactions. A molecule of EpCAM can bind with numerous CdSe quantum dots (QDs) to realize significant signal amplification. By stripping square wave voltammetry (SSWV), concentrations down to 10 aM EpCAM can be successfully detected and quantified based on the newly developed aptasensor. The detection linear range is from 10 aM to 100 pM. This principle can also be extended to other biomarkers when a suitable aptamer or antibody is available. This aptasensor is expected to be a potential tool for the diagnosis, therapeutic evaluation and personalized medical care of cancer patients. (C) 2019 Elsevier B.V. All rights reserved.
Alginate-derived carbon dots for "turn off-on" anti-neoplastic 5-fluorouracil sensing in biological samples

Abstract:As a chemotherapy drug, 5-fluorouracil (5-FU) has been used for colon cancer for decades. Excessive levels of 5-FU in the human body can lead to notable adverse effects, including severe diarrhea, infection, mouth sores, skin peeling, skin inflammation, and ulcers, which are important and relatively common digestive side effects. In addition, 5-FU is an analog of uracil and also has similarities to pyrimidines. Therefore, it is not easy to separate them. This research presented a sensor capable of detecting drugs in minimal amounts. An alginate-derived carbon dot (CD) was synthesized by unique optical properties that obey an on-off fluorescence mechanism for 5-FU sensing. Introducing copper (Cu(I)) to CDs results in fluorescence quenching through electron transfer. However, when 5-FU is added to the system as an oxidizing agent, a redox reaction occurs on the surface of the CDs, which leads to the restoration of fluorescence as Cu(I) is altered to Cu(II). Experimental results showed a strong linear correlation (R2 = 0.99) in the concentration range of 1.00-45.00 nM, with the following linear regression, and revealed the relative standard deviation (RSD%) and detection limit of 2.57%, and 1.00 nM, respectively. These results validated the excellent detection capability of the proposed method even at low concentrations of 5-FU and in the presence of other drugs and interfering substances. Also, the recovery of 5-FU (varies from 100.46% to 113.7%, with RSD equal to 1.89-3.63) in serum samples indicates the absence of matrix interference in the determination of 5-FU. In summary, this novel approach to developing a cost-effective and sensitive sensor holds great potential for future applications in healthcare and related fields.

Abstract:Alpelisib, an anti-cancer drug with great potential, targets the phosphoinositide 3-kinase (PI3K) pathway, which is commonly dysregulated in various cancer types. Accurate and sensitive quantification of Alpelisib is crucial for optimizing treatment strategies and monitoring therapeutic outcomes. In this study, we have developed the first electrochemical sensor utilizing a modified electrode to precisely determine Alpelisib concentrations. Cyclic voltammetry (CV), differential pulse voltammetry (DPV), and Electrochemical impedance spectroscopy (EIS) techniques were extensively employed to investigate the electrochemical behavior of Alpelisib. The modified electrode was constructed by immobilizing multi-walled carbon nanotubes (MWCNTs), iron oxide (Fe3O4), and copper aluminum spinel (CuAl2O4) onto a glassy carbon electrode (GCE). This modification scheme aimed to improve the electrocatalytic activity and selectivity of the electrode towards Alpelisib. Subsequently, the developed electrode was successfully applied for the detection of Alpelisib in pharmaceutical formulations and biological fluids, thus demonstrating its applicability in real-world scenarios. The limit of detection (LOD) was determined to be 0.036 μM, affirming the sensor’s high sensitivity in detecting low concentrations of Alpelisib.
