Abstract:The effect of solute, sodium deoxycholate bile salt (DOCNa), on the cooperativity of ordered-fluid, fosfolipid phase transition has been investigated by determining of the transition widths in multibilayer dispersions of the dipalmitoylphosphatidylcholine (DPPC)-DOCNa mixtures. The DT1/2 values were determined by differential scanning calorimetry. The van't Hoff values of the main transition enthalpies were calculated using an approximative expression, deduced from Zimm and Bragg theory [Sturtevant,J.M.(1982), <a class="link-external link-http" href="http://Proc.Natl.Acad.Sci.USA" rel="external noopener nofollow">this http URL</a>,79:3963-3967]. The bile salt decreases the cooperativity (size of cooperative unit) of DPPC bilayers.
What problem does this paper attempt to address?
This paper aims to study the effect of cholate (DOCNa) on the phase - transition cooperativity of dipalmitoylphosphatidylcholine (DPPC) bilayer membranes. Specifically, the authors measured the phase - transition width ($\Delta T_{1/2}$) of DPPC - DOCNa mixtures at different molar ratios in a multilamellar vesicle dispersion system by differential scanning calorimetry (DSC) to evaluate the effect of cholate on the phase - transition cooperativity of DPPC bilayer membranes.
### Research Background
The phase - transition cooperativity of phospholipid bilayer membranes refers to the degree of synergy between molecules during the phase - transition process. According to the Zimm and Bragg theory, lipid molecules exist in different states (ordered, fluid, and interfacial states) during the phase - transition process. The cooperativity parameter $\sigma$ of the phase - transition can be calculated by the following formula:
\[ \theta = \frac{1}{2} \left[ 1 + \frac{s - 1}{\sqrt{(s - 1)^2 + 4\sigma s}} \right] \]
where:
- $\theta$ is the average fraction of molecules in the fluid state,
- $s$ is the statistical weight of the post - phase - transition state,
- $\sigma$ is the cooperativity parameter.
The cooperativity parameter $\sigma$ is related to free energy and can be expressed by the following formulas:
\[ s = \exp\left(\frac{\Delta S_t}{R}\right) \exp\left(-\frac{\Delta H_t}{RT}\right) \]
\[ \sigma = \exp\left(\frac{S_i}{R}\right) \exp\left(-\frac{H_i}{RT}\right) \]
where:
- $\Delta H_t$ and $\Delta S_t$ are the phase - transition enthalpy and entropy, respectively,
- $H_i$ and $S_i$ are the interfacial enthalpy and entropy, respectively.
The smaller the cooperativity parameter, the stronger the phase - transition cooperativity. The size of the cooperative unit $n$ is defined as:
\[ n = \frac{1}{\sqrt{\sigma}} \]
### Experimental Method
The authors used differential scanning calorimetry (DSC) to measure the phase - transition temperature $T_t$ and phase - transition width $\Delta T_{1/2}$ of DPPC - DOCNa mixtures at different molar ratios. The experimental results showed that as the DOCNa concentration increased, the phase - transition temperature $T_t$ of the DPPC bilayer membrane decreased, and the phase - transition width $\Delta T_{1/2}$ increased, indicating that cholate reduced the phase - transition cooperativity of the DPPC bilayer membrane.
### Results and Discussion
Based on the data obtained by DSC measurement, the authors calculated the cooperativity parameter $\sigma$ and the cooperative unit size $CU$ at different molar ratios. The results showed that as the DOCNa concentration increased, the cooperativity parameter $\sigma$ increased, and the cooperative unit size $CU$ decreased, indicating a decrease in the phase - transition cooperativity. For example, in the case of DPPC:DOCNa = 1:1, the cooperativity parameter $\sigma$ increased significantly, resulting in a significant decrease in the cooperative unit size $CU$.
### Conclusion
This study shows that cholate (DOCNa) significantly reduces the phase - transition cooperativity of DPPC bilayer membranes. This may be because cholate molecules insert into the phospholipid bilayer membrane, disrupting the van der Waals forces between phospholipid molecules, thereby reducing the phase - transition cooperativity. This finding helps to understand the mechanism of action of cholate in biological membranes and its effect on membrane fluidity.
