Abstract:Structured illumination microscopy (SIM) is a powerful technique for providing super-resolution imaging, but its reconstruction algorithm, i.e., linear reconstruction structured illumination microscopy (LRSIM) algorithm in the Fourier domain, limits the imaging speed due to its computational effort. Here, we present a novel reconstruction algorithm that can directly process SIM data in the spatial domain. Compared to LRSIM, this approach uses the same number of frames to achieve a comparable resolution but with a much faster processing speed. Our algorithm was verified on both simulated and experimental data using sinusoidal pattern illumination. Moreover, this algorithm is also applicable for speckle pattern illumination. Abstract:The resolution of reconstructed images is significantly limited by the size of captured holograms in digital holography. A multiplane digital holographic algorithm based on extrapolation iterations is proposed herein. Holograms at different axial distances are captured with a shifting imaging sensor and then extrapolated to a larger hologram after padding. Multiple holograms provide multipoint constraints in diffraction calculation. Constraints based on the energy distribution and transmissions of an object are applied to extract the object function from the holograms. The method achieves a higher convergence speed for solving the inverse problem compared with the single-plane extrapolation method. We demonstrate that the reconstruction in multiple planes yields a better resolution than that in one plane.

Abstract:Axial error arises from the error inherent in man-made measurement of distance between object and detector for in-line holography, resulting in image blurring, defocusing and low reconstruction quality. Nevertheless, existing methods for autofocusing and axial correction requires computing parameters with several distances neighboring current distance, which incorporates additional loops into the iterative algorithms. In this study, we propose an approach that updates diffraction distance directly based on statistical information of reconstructed object, thus eliminating the need for additional loops and reducing reconstruction time. Besides, our methods could achieve high reconstruction accuracy, while maintaining high convergence rate, stability and wide initialization space. The effectiveness of our proposed method has been validated through both simulation and experiments.
Iterative Reconstruction of Diffraction Limited Digital Holography

Abstract:Digital holographic imaging has the characteristics of fast and flexible with simple set up, and it can quantitatively obtain the intensity and phase of objects. The resolution of the reconstructed image is greatly limited by the pixelated imaging detectors during the digitization processing of holograms. Due to the limited diffraction bandwidth, the diffracted beam of each object point can be viewed as a diffractive cone. The lack of diffraction components causes the reconstructed quality to decrease when the detector cannot collect a complete diffracted wave field in the inline holography. In this work, we studied the reconstructed quality of holograms under different sampling forms. The losing diffraction components can be reconstructed by using an iterative algorithm based on energy attenuation and support constraints. We propose three iterative methods. The first is the bicubic interpolation and extrapolation iterative method based on a single small detector. The second is multi-plane extrapolation iterative method. The third is a faithful iterative reconstruction based on a sparse sensor array. When the diffraction component recorded by the hologram is missing, the losing diffraction components can be reconstructed by using the principle of the holographic redundancy. The reduction of reconstruction quality caused by the loss of sampling in the small-sized detector and sparse sensor array in the digital holography can be compensated.
Iterative Projection Meets Sparsity Regularization: Towards Practical Single-Shot Quantitative Phase Imaging with In-Line Holography

Abstract:Holography provides access to the optical phase. The emerging compressive phase retrieval approach can achieve in-line holographic imaging beyond the information-theoretic limit or even from a single shot by exploring the signal priors. However, iterative projection methods based on physical knowledge of the wavefield suffer from poor imaging quality, whereas the regularization techniques sacrifice robustness for fidelity. In this work, we present a unified compressive phase retrieval framework for in-line holography that encapsulates the unique advantages of both physical constraints and sparsity priors. In particular, a constrained complex total variation (CCTV) regularizer is introduced that explores the well-known absorption and support constraints together with sparsity in the gradient domain, enabling practical high-quality in-line holographic imaging from a single intensity image. We developed efficient solvers based on the proximal gradient method for the non-smooth regularized inverse problem and the corresponding denoising subproblem. Theoretical analyses further guarantee the convergence of the algorithms with prespecified parameters, obviating the need for manual parameter tuning. As both simulated and optical experiments demonstrate, the proposed CCTV model can characterize complex natural scenes while utilizing physically tractable constraints for quality enhancement. This new compressive phase retrieval approach can be extended, with minor adjustments, to various imaging configurations, sparsifying operators, and physical knowledge. It may cast new light on both theoretical and empirical studies.
Lensless Microscope Based on Iterative In-Line Holographic Reconstruction

Abstract:We propose a lensless microscopic imaging technique based on iteration algorithm with known constraint for image reconstruction in digital in-line holography. In our method, we introduce a constraint on the sample plane as known part in the lensless microscopy for iteration algorithm in order to eliminate the twin-image effect of holography and thus lead to better performance on microscopic imaging. We evaluate our method by numerical simulation and built a prototype in-line holographic imaging system and demonstrated its capability by preliminary experiments. In our proposed setup, a carefully designed photomask used to hold the sample is under illumination of a coherent light source. The in-line hologram is then recorded by a CMOS sensor. In the reconstruction, the known information of the illumination beam and the photomask is used as constraints in the iteration process. The improvement of image quality because of suppression of twin-images can be clearly seen by comparing the images obtained by direct holographic reconstruction and our iterative method.

Abstract:Digital in-line holography (DIH) is a lensless imaging technique that can be used to build low-cost and compact imaging systems. In DIH, the in-line hologram is recorded by a CMOS or CCD sensor and later used to reconstruct the image of the sample. The imaging resolution is determined by the system numerical aperture provided that the pixel size is smaller than the required Nyquist criteria for sampling distance. In the case of short sample-to-sensor distance, pixel size is often a limiting factor for the resolution. To solve this problem, we propose to use iterative method along with data interpolation for the holographic reconstruction. Proof-of-concept numerical simulations have been done to show the effectiveness of our method. In our algorithm, the optical field is propagated back and forth between the sample plane and the sensor plane while using the measured intensity and a priori information about the sample as constraints, following Gerchberg-Saxton and Fienup's methods. The iteration will converge and we can get both intensity and phase information of the sample. Before the iteration, the intensity data matrix measured by the sensor is interpolated to enlarge the matrix dimension and thus effectively reduce the pixel size. During the iteration, we apply the sensor plane constraints on only the measured intensity location but not the interpolated data location. In our simulation, we observed that during the iteration, the interpolated data will be changed reasonably and we can finally reconstruct the sample image with better resolution.

Abstract:Compared with in-line digital holography, off-axis digital holography can be used to image more kinds of objects without the limitation of weak phase and sparsity, but the resolution is lower. In this paper, we put forward a high-resolution iterative reconstruction algorithm for off-axis holography which requires only the recording of a single off-axis hologram. Interference and inverse interference processes are used to extract the object wave from the off-axis hologram. A frequency domain filter with increasing radius is used to improve the competitive advantage of low frequency information in the early iterations, while retaining the high frequency information. The constraints of total variation and positive absorption are implemented at the object plane to enhance the robustness of the algorithm. Numerical simulations and experiments show that the resolution of our method is higher than traditional off-axis digital holography. In addition, it is suitable for phase image when the object has a strong phase fluctuation, as well as for imaging dynamical objects.

Abstract:Digital holography provides access to quantitative measurement of the entire complex field, which is indispensable for the investigation of wave-matter interactions. The emerging iterative phase retrieval approach enables to solve the inverse imaging problem only from the given intensity measurements and physical constraints. However, enforcing imprecise constraints limits the reconstruction accuracy and convergence speed. Here, we propose an advanced iterative phase retrieval framework for single-shot in-line digital holography that incorporates adaptive constraints, which achieves optimized convergence behavior, high-fidelity and twin-image-free reconstruction. In conjunction with morphological operations which can extract the object structure while eliminating the irrelevant part such as artifacts and noise, adaptive constraints allow the support region to be accurately estimated and automatically updated at each iteration. Numerical reconstruction of complex-valued objects and the capability of noise immunity are investigated. The improved reconstruction performance of this approach is experimentally validated. Such flexible and versatile framework has promising applications in biomedicine, X-ray coherent diffractive imaging and wavefront sensing.
Image Processing in Numerical Reconstruction for Lensless Fourier Transform Hologram

Abstract:Based on the characteristics of numerical reconstruction for lensless Fourier transform hologram, a set of digital image processing algorithm is presented to eliminate zero diffraction component and twin image. This method is different from the former one and consists of three steps: hologram contrast enhancement, bandpass filtering and V-filtering, and reconstructed image smoothing. The hologram contrast enhancement pretreatment, which can be achieved by highpass filtering, is adopted to improve the diffraction efficiency of the recorded digital hologram and suppress zero diffraction component of the reconstructed image. Using bandpass filtering and V-filtering to the reconstructed image with background noise, the noise can be eliminated and the viewfield is made homogeneous. Furthermore, smooth processing such as wavelet filtering and median filtering can also enhance the signal-noise ratio of the reconstructed image. It is shown that the method need record hologram only once and these three simple processing steps are effective for improving the reconstructed image quality and obtaining the satisfying results. It is especially applicable for the numerical reconstruction of lensless Fourier transform hologram, and more practical.

Abstract:In this paper, we analyze the influence of two kinds of deviation errors on sectional image reconstruction for an optical scanning holography system using a random-phase pupil. The first deviation occurs in the lateral pixel position while the second occurs in the pixel value of the decoding function. Theoretical analysis and numerical simulation show that these two deviations may lead to noise in the reconstructed image. Additional discussions include the signal-to-noise ratio of the reconstructed image.

Abstract:We propose a digital inline holographic reconstruction method based on learned sparsifying transform. An iterative algorithm which includes the steps of updating background, phase, and image, as well as a step of sparse coding, is adopted to optimize the designed penalized least squares object function with regularization based on a sparsifying transform learned from sample images. A lensless inline holographic microscope (LIHM) was built and used to image a U.S. air force target and a pumpkin stem sample. The proposed method was applied to reconstruct the sample images. Compared with conventional holographic reconstruction method or the L1-norm-based penalized least squares reconstruction method, the imaging results show that the proposed method could better suppress the twin-image disturbance, staircase edges and block artifacts, thus enhance the reconstructed image quality.

Abstract:A dual-plane in-line digital holographic imaging method incorporating volume holographic microscopy (VHM) is presented to reconstruct objects in a single shot while eliminating zero-order and twin-image diffracted waves. The proposed imaging method is configured such that information from different axial planes is acquired simultaneously using multiplexed volume holographic imaging gratings, as used in VHM, and recorded as in-line holograms where the corresponding reference beams are generated in the fashion of Gabor's in-line holography. Unlike conventional VHM, which can take axial intensity information only at focal depths, the proposed method digitally reconstructs objects at any axial position. Further, we demonstrate the proposed imaging technique's ability to effectively eliminate zero-order and twin images for single-shot three-dimensional object reconstruction.
Accurate Reconstruction in Digital Holographic Microscopy Using Fresnel Dual-Tree Complex Wavelet Transform

Abstract:Digital holography is a promising measurement method in the fields of bio-medicine and micro-electronics. But the captured images of digital holography are severely polluted by the speckle noise because of optical scattering and diffraction. Via analyzing the properties of Fresnel diffraction and the topographies of micro-structures, a novel reconstruction method based on the dual-tree complex wavelet transform (DT-CWT) is proposed. This algorithm is shift-invariant and capable of obtaining sparse representations for the diffracted signals of salient features, thus it is well suited for multiresolution processing of the interferometric holograms of directional morphologies. An explicit representation of orthogonal Fresnel DT-CWT bases and a specific filtering method are developed. This method can effectively remove the speckle noise without destroying the salient features. Finally, the proposed reconstruction method is compared with the conventional Fresnel diffraction integration and Fresnel wavelet transform with compressive sensing methods to validate its remarkable superiority on the aspects of topography reconstruction and speckle removal.

Abstract:Holographic techniques are indispensable tools for modern optical engineering. Over the past two decades, research about incoherent digital holography has continued to attract attention. Optical scanning holography (OSH) can obtain incoherent holograms using single-pixel detection and structured illumination with Fresnel zone patterns (FZPs). Particularly by changing the size of a detector, OSH can also obtain holograms under coherently illuminated conditions. Since 1979, OSH has continuously evolved. According to the evolution of semiconductor technology, spatial light modulators (SLMs) come to be useful for various imaging fields. By using SLM techniques for OSH, the practicality of OSH is improved. These SLM-based OSH methods are termed computational OSH (COSH). In this review, the configurations, recording and reconstruction methods, and proposed applications of COSH are reviewed.

Abstract:Slightly off-axis digital holographic microscopy (DHM) has recently gained considerable attention due to its unique ability to improve the space-bandwidth product (SBP) of the imaging system while separating the object information from the background intensity to a certain extent. In order to obtain a decent image reconstruction, the spectral aliasing problem still needs to be addressed, which, however, is difficult to be achieved by the conventional linear Fourier domain filtering. To this end, in this paper, we propose a high-throughput artifact-free slightly off-axis holographic reconstruction method based on Fourier ptychographic microscopy (FPM). Inspired by the nonlinear optimized phase reconstruction algorithm of FPM, we perform constrained updates between the real and Fourier domains in an iterative manner to reconstruct the complex amplitude by the hologram intensity. Experimental results on live HeLa cell samples show that the proposed method can provide higher reconstruction accuracy and better image quality compared with the conventional Fourier method and the Kramers–Kronig (KK) relation-based method.

Abstract:Since the pupil function is defined as a time-invariant system, the traditional partially coherent imaging model is time-consuming to calculate the effect of spatially varying wavefront aberrations on the scanning image. A fast reconstruction method of the aberrated scanning aerial image is presented for the scanning projection lithographic tool. In the proposed method, the principal components (PCs) are used to decompose and reconstruct the aberrated aerial image. Due to the exact quadratic relationship between the PC coefficients and the Zernike coefficients, the integration of the PCs in the intensity domain can be transformed into the integration of quadratic Zernike vectors when reconstructing the scanning aerial image. An integral transfer function is introduced to describe this process. This method can not only reconstruct the aberrated scanning image quickly but can also obtain the explicit relationship between the Zernike coefficients and the aberrated scanning aerial image.
Fast iterative method empowered by GPU acceleration for Fourier-domain optical coherence tomography image reconstruction

Abstract:Iterative methods could provide high-quality image reconstruction for Fourier-domain optical coherence tomography (FD-OCT) by solving an inverse problem. Compared with the regular IFFT-based reconstruction, a more accurate estimation could be iteratively solved by integrating prior knowledge, however, it is often more time-consuming. To deal with the time problem, we proposed a fast iterative method for FD-OCT image reconstruction empowered by GPU acceleration. An iterative scheme is adopted, including a forward model and an inverse solver. Large-scale parallelism of OCT image reconstruction is performed on B-scans. We deployed the framework on Nvidia GeForce RTX 3090 graphic card that enables parallel processing. With the widely used toolkit Pytorch, the inverse problem of OCT image reconstruction is solved by the stochastic gradient descent (SGD) algorithm. To validate the effectiveness of the proposed method, we compare the computational time and image quality with other iterative approaches including ADMM, AR, and RFIAA method. The proposed method could provide a significant speed enhancement of 1,500 times with comparable image quality to that of ADMM reconstruction. The result indicates a potential for high-quality real-time volumetric OCT image reconstruction via iterative algorithms.

Abstract:The image reconstruction for Fourier-domain optical coherence tomography (FD-OCT) could be achieved by iterative methods, which offer a more accurate estimation than the traditional inverse discrete Fourier transform (IDFT) reconstruction. However, the existing iterative methods are mostly A-line-based and are developed on CPU, which causes slow reconstruction. Besides, A-line-based reconstruction makes the iterative methods incompatible with most existing image-level image processing techniques. In this paper, we proposed an iterative method that enables B-scan-based OCT image reconstruction, which has three major advantages: (1) Large-scale parallelism of the OCT dataset is achieved by using GPU acceleration. (2) A novel image-level cross-domain regularizer was developed, such that the image processing could be performed simultaneously during the image reconstruction; an enhanced image could be directly generated from the OCT interferogram. (3) The scalability of the proposed method was demonstrated for 3D OCT image reconstruction. Compared with the state-of-the-art (SOTA) iterative approaches, the proposed method achieves higher image quality with reduced computational time by orders of magnitude. To further show the image enhancement ability, a comparison was conducted between the proposed method and the conventional workflow, in which an IDFT reconstructed OCT image is later processed by a total variation-regularized denoising algorithm. The proposed method can achieve a better performance evaluated by metrics such as signal-to-noise ratio (SNR) and contrast-to-noise ratio (CNR), while the speed is improved by more than 30 times. Real-time image reconstruction at more than 20 B-scans per second was realized with a frame size of 4096 (axial) × 1000 (lateral), which showcases the great potential of the proposed method in real-world applications.
