Abstract:Highly regio- and enantioselective alcohol dehydrogenases BDHA (2,3-butanediol dehydrogenase from Bacillus subtilis BGSC1A1), CDDHPm (cyclic diol dehydrogenase from Pseudomonas medocina TA5), and CDDHRh (cyclic diol dehydrogenase from Rhodococcus sp. Moj-3449) were discovered for the oxidation of racemic trans-cyclic vicinal diols. Recombinant Escherichia coli expressing BDHA was engineered as an efficient whole-cell biocatalyst for the oxidation of ()-1,2-cyclopentanediol, 1,2-cyclohexanediol, 1,2-cycloheptane-diol, and 1,2-cyclooctanediol, respectively, to give the corresponding (R)--hydroxy ketones in >99% ee and (S,S)-cyclic diols in >99% ee at 50% conversion in one pot. Escherichia coli (BDHA-LDH) co-expressing lactate dehydrogenase (LDH) for intracellular regeneration of NAD(+) catalyzed the regio- and enantioselective oxidation of (+/-)-1,2-dihydroxy-1,2,3,4-tetrahydronaphthalene to produce the corresponding (R)--hydroxy ketone in >99% ee and (S,S)-cyclic diol in 96% ee at 49% conversion. Preparative biotransformations were also demonstrated. Thus, a novel and useful method for the one-pot synthesis of both vicinal diols and -hydroxy ketones in high ee was developed via highly regio- and enantioselective oxidations of the racemic vicinal diols.
Characterization of Four Diol Dehydrogenases for Enantioselective Synthesis of Chiral Vicinal Diols

Abstract:Enantiopure vicinal diols are important building blocks used in the synthesis of fine chemicals and pharmaceutical compounds. Diol dehydrogenase (DDH) mediated stereoselective oxidation of racemic vicinal is an efficient way to prepare enantiopure vicinal diols. In this study, four new bacterial DDHs (AnDDH from Anoxybacillus sp. P3H1B, HcDDH from Hazenella coriacea, GzDDH from Geobacillus zalihae and LwDDH from Leptotrichia wadei) were mined from the GenBank database and expressed in E. coli T7. The four DDHs were purified and biochemically characterized for oxidation activity toward (R)-1-phenyl-1,2-ethanediol, with the optimal reaction condition of pH9.0 (AnDDH), 10.0 (HcDDH) and 11.0 (GzDDH and LwDDH) and the temperatures at 40 degrees C (AnDDH), 50 degrees C (HcDDH) and 60 degrees C (GzDDH and LwDDH), respectively. The four enzymes were stable at the pH from 7.0 to 9.0 and below 40 degrees C. Kinetic parameters of four DDHs showed that the HcDDH from Hazenella coriacea had high activity toward a broad range of vicinal diols. A series of racemic vicinal diols were successfully resolved by recombinant E. coli (HcDDH-NOX) resting cells co-expression of an NADH oxidase (NOX), affording (S)-diols and (1S, 2S)-trans-diols in >= 99% ee. The synthetic potential of HcDDH was proved by E. coli (HcDDH-NOX) via kinetic resolution of racemic trans-1,2-indandiol on a 100 ml scale reaction, (S, S)-trans-1,2-indandiol was prepared in 46.7% yield and >99% ee. In addition, asymmetric reduction of four alpha-hydroxy ketones (10-300 mmol.L-1) by E. coli (HcDDH-GDH) resting cells resulted in >99% ee and 69-98% yields of (R)-vicinal diols. The current research expands the toolbox of DDHs to synthesize chiral vicinal diols and demonstrated that the mined HcDDH is a potential enzyme in the synthesis of a broad range of chiral vicinal diols. Abstract:As a chiral precursor for the important anticoagulant Edoxaban, enantioselective synthesis of (S)-3-cyclohexene-1-carboxylic acid is of great significance. The complicated procedures and generation of massive solid waste discourage its chemical synthesis, and the alternative biocatalysis route calls for an enzyme capable of asymmetric hydrolysis of racemic methyl-3-cyclohexene-1-carboxylate. To this end, we engineered the E. coli esterase BioH for improved S-enantioselectivity via rational design. By combinatorial modulation of steric and aromatic interactions, a positive mutant Mu3 (L24A/W81A/L209A) with relatively high S-selectivity in hydrolyzing racemic methyl-3-cyclohexene-1-carboxylate was obtained, improving the enantiomeric excess from 32.3% (the wild type) to 70.9%. Molecular dynamics simulation was conducted for both (R)- or (S)- complexes of the wild type and Mu3 to provide hints for the mechanism behind the increased S-selectivity. Moreover, the reaction conditions of Mu3 in methyl-3-cyclohexene-1-carboxylate hydrolysis was optimized to improve the conversion rate to 2 folds.

Abstract:Abstract An enzymatic cascade reaction was developed for one‐pot enantioselective conversion of aliphatic dialdehydes to chiral vicinal diols using pyruvate decarboxylases (PDCs) and alcohol dehydrogenases (ADHs). The PDCs showed promiscuity in catalysing the cyclization of aliphatic dialdehydes through intramolecular stereoselective carbon‐carbon bond formation. Consequently, 1,2‐cyclopentanediols in three different stereoisomeric forms and 1,2‐cyclohexanediols in two different stereoisomeric forms could be prepared with high conversion and stereoisomeric ratio from the respective initial substrates, glutaraldehyde and adipaldehyde. Abstract:Cascade biocatalysis via intracellular epoxidation and hydrolysis was developed as a green and efficient method for enantioselective dihydroxylation of aryl olefins to prepare chiral vicinal diols in high ee and high yield. Escherichia coli (SSP1) coexpressing styrene monooxygenase (SMO) and epoxide hydrolase SpEH was developed as a simple and efficient biocatalyst for S-enantioselective dihydroxylation of terminal aryl olefins 1a-15a to give (S)-vicinal diols 1c-15c in high ee (97.5-98.6% for 10 diols; 92.2-93.9% for 3 diols) and high yield (91-99% for 6 diols; 86-88% for 2 diols; 67% for 3 diols). Combining SMO and epoxide hydrolase StEH showing complementary regioselectivity to SpEH as a biocatalyst for the cascade biocatalysis gave rise to R-enantioselective dihydroxylation of aryl olefins, being the first example of this kind of reversing the overall enantioselectivity of cascade biocatalysis. E. coli (SST1) coexpressing SMO and StEH was also engineered as a green and efficient biocatalyst for R-dihydroxylation of terminal aryl olefins 1a-15a to give (R)-vicinal diols 1c-15c in high ee (94.2-98.2% for 7 diols; 84.2-89.9% for 6 diols) and high yield (90-99% for 6 diols; 85-89% for 5 diols; 65% for 1 diol). E. coli (SSP1) and E. coli (SST1) catalyzed the trans-dihydroxylation of trans-aryl olefin 16a and cis-aryl olefin 17a with excellent and complementary stereoselectivity, giving each of the four stereoisomers of 1-phenyl-1,2-propanediol 16c in high ee and de, respectively. Both strains catalyzed the trans-dihydroxylation of aryl cyclic olefins 18a and 19a to afford the same trans-cyclic diols (1R,2R)-18c and (1R,2R)-19c, respectively, in excellent ee and de. This type of cascade biocatalysis provides a tool that is complementary to Sharp less dihydroxylation, accepting cis-alkene and offering enantioselective trans-dihydroxylation.

Firstly, the NaBH4-H2SO4 system converted L-alpha-amino acids to (S)-amino alcohols via amino acid carboxyl reduction. Secondly, the three-enzyme (transaminase, carbonyl reductase and glucose dehydrogenase) cascade biocatalysis system converted amino alcohols to enantiopure vicinal 1,2-diols via amino alcohol deamination, alpha-hydroxy ketone asymmetric reduction and cofactor regeneration. Taking advantage of the two different reaction systems, chiral vicinal 1,2-diols could be obtained from L-alpha-amino acids with high yields (69-90 %) and excellent ee values (91->99 % ee).

Abstract:Although biocatalysis has garnered widespread attention in both industrial and academic realms, the enzymatic synthesis of chiral oxetanes remains an underdeveloped field. Halohydrin dehalogenases (HHDHs) are industrially relevant enzymes that have been engineered to accomplish the reversible transformation of epoxides. In our work, a biocatalytic platform was constructed for the stereoselective kinetic resolution of chiral oxetanes and formation of 1,3-disubstituted alcohols. HheC from Agrobacterium radiobacter AD1 was engineered to identify key variants capable of catalyzing the dehalogenation of γ-haloalcohols (via HheC M1-M3) and ring opening of oxetanes (via HheC M4-M5) to access both (R)- and (S)-configured products with high stereoselectivity and remarkable catalytic activity, yielding up to 49% with enantioselectivities exceeding 99% ee and E>200. The current strategy is broadly applicable as demonstrated by expansion of substrate scope to include up to 18 examples for dehalogenations and 16 examples for ring opening. Additionally, the functionalized products are versatile building blocks for pharmaceutical applications. To shed light on the molecular recognition mechanisms for the relevant variants, molecular dynamic (MD) simulations were performed. The current strategy expands the scope of HHDH-catalyzed chiral oxetane ring constructions, offering efficient access to both enantiomers of chiral oxetanes and 1,3-disubstituted alcohols.

Abstract:We described the protein engineering of the halohydrin dehalogenase HheG to enhance its R enantioselectivity for the synthesis of chiral 4‐aryloxazolidinones via α‐regioselective ring‐opening of aryl epoxides with cyanate. Additionally, we achieved the inversion of its regioselectivity and enantioselectivity, resulting in a HheG variant tailored for the synthesis of chiral 5‐aryloxazolidinones via β‐regioselective and S‐enantioselective ring‐opening of aryl epoxides with cyanate. Leveraging these engineered mutants, we developed a biocatalytic platform capable of synthesis of both chiral 4‐aryloxazolidinones (up to 47% yield, 98% ee, and 99% regioselectivity) and chiral 5‐aryloxazolidinones (up to 46% yield, >99% ee, and 98% regioselectivity). Furthermore, a collaborative approach utilizing two regio‐ and enantioselective HheG mutants has been demonstrated to enable the simultaneous synthesis of chiral 4‐aryloxazolidinones and chiral 5‐aryloxazolidinones.

Abstract:Abstract The remarkable selectivity, sustainability, and efficiency afforded by biocatalytic strategies position them as complements or alternatives to traditional synthetic methods. Nevertheless, the currently narrow spectrum of enzymatic reactions available imposes limitations on synthesizing diverse desired compounds. Consequently, there continues to be a high demand for developing novel biocatalytic processes to access reactions that were previously unattainable. Herein, we report the discovery and subsequent protein engineering of a unique halohydrin dehalogenase to develop a biocatalytic platform for enantioselective formation and ring-opening of oxetanes. This biocatalytic platform, exhibiting high efficiency, excellent enantioselectivity, and broad scopes, facilitates the preparative-scale synthesis of not only both enantiomers of chiral oxetanes (up to 49% yield, >99 e.e.) but also a variety of chiral γ-substituted alcohols (up to 53% yield, >99 e.e.). Additionally, both the enantioselective oxetane formation and ring-opening processes have been proven scalable for large-scale transformations (20 mmol) at high substrate concentrations (200 mM), and can be integrated efficiently in a one-pot, one-catalyst cascade system. Moreover, useful derivatizations highlight the potential synthetic applications of the biocatalytic platform. This work expands the enzymatic toolbox for non-natural reactions and will promote further exploration of the catalytic repertoire of halohydrin dehalogenases in synthetic and pharmaceutical chemistry.
Engineering of a Novel Carbonyl Reductase with Coenzyme Regeneration in E. Coli for Efficient Biosynthesis of Enantiopure Chiral Alcohols.

Abstract:The novel anti-Prelog stereospecific carbonyl reductase from Acetobacter sp. CCTCC M209061 was successfully expressed in E. coli combined with glucose dehydrogenase (GDH) to construct an efficient whole-cell biocatalyst with coenzyme NADH regeneration. The enzymatic activity of GAcCR (AcCR with a GST tag) reached 304.9U/g-dcw, even 9 folds higher than that of wild strain, and the activity of GDH for NADH regeneration recorded 46.0U/mg-protein in the recombinant E. coli. As a whole-cell biocatalyst, the recombinant E. coli BL21(DE3)pLysS (pETDuet-gaccr-gdh) possessed a broad substrate spectrum for kinds of carbonyl compounds with encouraging yield and stereoselectivity. Besides, the asymmetric reduction of ethyl 4-chloroacetoacetate (COBE) to optically pure ethyl 4-chloro-3-hydroxybutyrate (CHBE) catalyzed by the whole-cell biocatalyst was systematically investigated. Under the optimal reaction conditions, the optical purity of CHBE was over 99% e.e. for (S)-enantiomer, and the initial rate and product yield reached 8.04μmol/min and 99.4%, respectively. Moreover, the space-time yield was almost 20 folds higher than that catalyzed by the wild strain. Therefore, a new, high efficiency biocatalyst for asymmetric reductions was constructed successfully, and the enantioselective reduction of prochiral compounds using the biocatalyst was a promising approach for obtaining enantiopure chiral alcohols.

Abstract:The first cascade biotransformation involving enantioselective reduction of a C═C double bond, Baeyer–Villiger oxidation, and lactone hydrolysis was developed as a green and sustainable tool for synthesizing enantiopure δ-lactones. One-pot cascade biotransformations were achieved with Acinetobacter sp. RS1 containing a novel enantioselective reductase and an enantioselective lactone hydrolase and Escherichia coli coexpressing cyclohexanone monooxygenase and glucose dehydrogenase, converting easily available 2-alkylidenecyclopentanones 1–2 into the corresponding valuable flavors and fragrances (R)-δ-lactones 5–6 in high ee. The one-pot synthesis is better than the reported two-step preparation. This concept is useful in developing other redox cascades with the substrates containing C═C double bond.

Abstract:Catalyzed by Rhodococcus erythropolis AJ270, a nitrile hydratase and amidase containing microbial whole cell catalyst, a number of racemic 1-arylmethyl-and 1-allyl-4-oxoazetidine-2-carbonitriles and carboxamides underwent efficient transformations under very mild conditions to produce enantiopure functionalized S-amide and R-acid products in excellent yields. While the nitrile hydratase showed good enzyme activity but virtually no enantioselectivity, the amidase displayed high R-enantioselectivity against almost all amide substrates tested. The synthetic applications of the resulting functionalized chiral beta-lactam derivatives were demonstrated by the facile preparation of beta-lactam-fused heterocyclic compounds.

Abstract:Chiral vicinal diols are crucial scaffolds frequently used for the preparation of adrenomimetic drugs. Biotransformation of benzaldehyde is one of the most effective strategies to access chiral vicinal diol. Herein, we present a novel and ecofriendly biocatalytic approach for synthesizing vicinal diol in good yield (>97%) and excellent ee (>97%). This method involves employing HeBAL, a benzaldehyde lyase from Herbiconiux sp. SALV-R1, as a catalyst for the crucial benzoin condensation step in synthesizing a ketone precursor. The treatment of benzaldehyde with BAL, in the presence of thiamin-diphosphate (ThDP) and Mg2+, results in the formation of α-hydroxy ketone, which subsequently undergoes biotransformation by carbonyl reductases (CR) to yield vicinal diols as the final product. The mutation of HeBAL highlights the pivotal role played by two specific alanine amino acid residues, Ala392 and Ala476. Furthermore, the catalysis by HeBAL effectively eliminates the formation of self-condensation by-products, which holds great implications for future phenylephrine synthesis.

Abstract:AbstractOptically pure vicinal 1,2‐diols are versatile chiral building blocks in the fine chemical and pharmaceutical industries. L‐α‐amino acid is a good feedstock source for high value‐added product production since it is inexpensive and renewable. However, conversion of L‐α‐amino acids to enantioenriched vicinal 1,2‐diols remains a significant challenge. In this study, combining a simple chemical process and a three‐enzyme cascade biocatalysis system, we have successfully implemented a one‐pot sequential process for the transformation of L‐α‐amino acids into enantiopure vicinal 1,2‐diols in aqueous medium. Firstly, the NaBH4‐H2SO4 system converted L‐α‐amino acids to (S)‐amino alcohols via amino acid carboxyl reduction. Secondly, the three‐enzyme (transaminase, carbonyl reductase and glucose dehydrogenase) cascade biocatalysis system converted amino alcohols to enantiopure vicinal 1,2‐diols via amino alcohol deamination, α‐hydroxy ketone asymmetric reduction and cofactor regeneration. Taking advantage of the two different reaction systems, chiral vicinal 1,2‐diols could be obtained from L‐α‐amino acids with high yields (69–90 %) and excellent ee values (91–>99 % ee).
Exploring the Biocatalytic Scope of a Novel Enantioselective Halohydrin Dehalogenase from an Alphaproteobacterium

Abstract:A gene encoding halohydrin dehalogenase from an alphaproteobacterium (AbHHDH) was identified, cloned and over-expressed in Escherichia coli. AbHHDH was able to catalyze the stereoselective dehalogenation of prochiral and racemic halohydrins. It showed the highest enantioselectivity in the dehalogenation of 20 mM (R,S)-2-bromo-1-phenylethanol, which yielded (S)-2-bromo-1-phenylethanol with 99% ee and 34.5% yield. Moreover, AbHHDH catalyzed the azidolysis of epoxides with low to moderate (S)-enantioselectivity. The highest enantioselectivity (E = 18.6) was observed when (R,S)-benzyl glycidyl ether was used as the substrate. A sequential kinetic resolution catalyzed by HHDH was employed for the synthesis of chiral 1-chloro-3-phenoxy-2-propanol. We prepared enantiopure (S)-isomer with a high enantiopurity of ee > 99% and a yield of 30.7% (E-value: 21.3) by kinetic resolution of 20 mM substrate. The (S)-isomer with 99% ee readily obtained from 40 to 150 mM (R,S)-1-chloro-3-phenoxy-2-propanol. Taken together, the results of this study demonstrate the applicability of this HHDH for the production of optically active compounds.
