Abstract:The reaction of o-aminoacetophenones and arylboronic acids catalyzed by copper(II) salts in the presence of pyridine under an O2 atmosphere provides a general and efficient one-pot preparation of biologically interesting acridones. This relay reaction comprises an intermolecular Suzuki cross-coupling, intramolecular oxidative C(sp3)-H amination, and C(sp2)-H activation with simultaneous rearrangement of the generated isatin intermediates. This strategy tolerates both electron-donating and -withdrawing functionalities to afford various acridones in good to excellent yields.

Abstract:A novel Pd-catalyzed intermolecular dehydrogenative annulation of aryl iodides and aryl carbamic chlorides for the efficient synthesis of phenanthridinone derivatives was developed. Simple aryl iodides and carbamic chlorides readily made from various anilines, a broad substrate scope with hetero/polycycles, as well as high-value products, make this direct dehydrogenative annulation approach very practical and attractive.

Abstract:An unprecedented Ir(III)-catalyzed C-H activation/amination/annulation of 2-phenyloxazoles with anthranils for the highly selective preparation of acridone derivatives in one-pot under controlled conditions is reported. This protocol is characterized by atom economy and high regioselectivity. A wide range of anthranils with 2-phenyloxazoles were well tolerated and afforded the desired products in moderate to good yields, in which the anthranil serves as a convenient amination reagent.

Abstract:A multiple synthetic strategy is employed to furnish the synthesis of 6,12,18-triaryldibenzo[b, j]quinoline[2,3-f][1, 7]phenanthroline. The key intermediate 6,12,18-trichlorobenzotriquinoline was prepared by Buchwald-Hartwig reaction of 1,3,5-tribromobenzene with methyl anthranilate, followed by hydrolysis and POCl3-promoted ring closure reaction. The palladium-catalyzed Suzuki coupling was carried out with a novel catalyst Pd-132 to form fourteen C-3-symmetric 9-aryl acridine derivatives. The reactions can be performed at low catalyst loading (0.1 mol%, ca. 0.03 mol% per site).

Abstract:A facile palladium-catalyzed synthesis of acridines has been realized by consecutive C=C double bond formation and CN cross-coupling. A variety of functionalized acridines can be accessed from easily available o-dihalobenzenes and N-tosylhydrazones in a single operation. This one-pot protocol has a wide scope with respect to both coupling partners, and provides an efficient route to functionalized acridine derivatives, which are generally difficult to synthesize by previously known methods.

Abstract:An efficient aerobic oxidative annulation of cyclohexanones and 2-aminophenyl ketones approach to substituted acridines, a structural motif for a large number of pharmaceuticals and functional materials is described. The key feature of this method is the use of oxygen as the sole oxidant and Pd catalyst, which resulting in the high regioselectivity with unsymmetrical meta-substituted cyclohexanones. The electron gap of the global redox condensation process is filled and the reaction efficiency is significantly promoted by O-2 as a redox moderator. This protocol possesses many advantages such as using O-2 as a cheap and nonhazardous oxidant, high regioselectivity and water as the only by-product, which meet the principle of green chemistry.

Abstract:Herein, we report a novel one-pot method for synthesizing polysubstituted pyrrole derivatives via three-component reactions of alkenyl bromides, amines, and isocyanides. The conversion process is catalyzed by a Pd catalyst without additional ligands. The key features of this methodology are the orderly insertion of three isocyanide molecules which is directed by the intramolecular imine group. The method also provides an alternative way to synthesize the cyano compounds from isocyanides. DFT calculation results support a part of the proposed reaction mechanism.

Abstract:Pyrimidine scaffold is one of the important six-membered heterocyclic compounds,which possess important biological and pharmaceutical activities.A convenient one-pot procedure for the synthesis of 2,4,6-trisubstituted pyrimidines from acid chlorides, phenylacetylene and formamidine catalyzed by Pd(PPh3)2Cl2/CuI was described through a tandem coupling and cyclocondensation sequence.Acid chlorides coupled with phenylacetylene to give α,β-unsaturated ynones,and in situ converted into pyrimidines by the cycloaddition of formamidine.The desired pyrimidines were obtained with 33~50%isolated yields.The protocol was compatible to other substrates and easy work up procedures making it feasible for heterocycle synthesis.

Abstract:Pd-catalyzed annulation of aryne and aryl ketone O-acetyloxime via C–H bond activation was realized. Through the C–H bond activation/insertion/cyclization/elimination reaction sequence, phenanthridines are successfully constructed, providing an attractive strategy to approach substituted heterocycle without preactivation of starting materials.

Abstract:In this paper, a concise and practicable route to the synthesis of multisubstituted 2‐pyridones via a four‐component one‐pot stepwise cyclization catalyzed by Cs2CO3 has been developed. During this stepwise transformation, two new C‐C bonds and two new C‐N bonds were generated via an intermolecular condensation/aza‐Michael reaction/intermolecular Michael addition/intramolecular cyclization sequence. The alternative protocol offers a complementary route to provide a series of structurally diverse polysubstituted 2‐pyridones in good to excellent yields from easily available raw materials under mild conditions. A concise and practicable route to the synthesis of multisubstituted 2‐pyridones via a four‐component one‐pot stepwise cyclization catalyzed by Cs2CO3 has been developed. During this stepwise transformation, two new C‐C bonds and two new C‐N bonds were generated through an intermolecular condensation/aza‐Michael reaction/intermolecular Michael addition/intramolecular cyclization sequence. The alternative protocol offers a complementary route to provide a series of structurally diverse polysubstituted 2‐pyridones (21 examples) in good to excellent yields (52–81%) from easily available raw materials arylamines, dialkyl acetylenedicarboxylate, 1,3‐dicarbonyl compounds, and trialkyl orthoformate under mild conditions. It might provide new opportunities for the discovery of 2‐pyridones‐containing drugs.

Abstract:In this paper, a palladium-catalyzed [3+2] annulation of allenyl carbinol acetates with azomethine imines has successfully been developed under mild reaction conditions, affording biologically interesting tetrahydropyrazoloisoquinoline derivatives in high to excellent yields and with excellent stereoselectivity. The reaction follows a tandem [3+2] cycloaddition/allylation/elimination of AcOH pathway. Allenyl carbinol acetates also reacted well with in situ generated azomethine imine under cocatalysis of Ag(i)/Pd(0) catalysts in a similar reaction pathway.

Abstract:A copper catalyzed annulation-aromatization of benzyl trifluoromethyl ketimines with 3-acryloyloxazolidin-2-ones for the synthesis of 3-fluoropyridines through double C-F bond cleavages has been developed. In this approach, the annulation occurred between the in situ formed dienes from trifluoromethyl ketimines via the first C-F bond cleavage and 3-acryloyloxazolidin-2-ones. Then the aromatization afforded 3-fluoropyridines in moderate yields through the second C-F bond cleavage. The 3-fluoropyridine products could be further hydrolyzed to multi-substituted 3-pyridinecarboxylic acids.

Abstract:A novel synthesis method has been developed for the construction of functionalized 2,4-diarylpyridines. The reaction involves several key processes, including a reduction reaction, Michael reaction, decarboxylation, intramolecular cyclization, oxidation, ring-opening and aromatization reaction. All of these processes are facilitated by heating a mixture of the starting materials in the environmentally benign solvent, propylene carbonate (PC), with the oxidant (air) and the catalyst Cu(OAc)2. This cascade reaction not only forms two new bonds but also cleaves three existing bonds in a single pot. The protocol presented in this study is a valuable tool for synthesizing functionalized unsymmetrical 2,4-diarylpyridines.

Abstract:A Pd-catalyzed three-component synthesis of 1,4-dihydropyridines is described. Starting from simple or easily available aromatic aldehydes, aryl amines and terminal olefins, the reaction proceeds smoothly under mild conditions and an O2 atmosphere to afford 2,6-unsubstituted 1,4-dihydropyridines in moderate to good yields. The process constitutes an efficient modified Hantzsch reaction. However, the reaction yields were greatly decreased when the process was conducted in laboratory air or at a lower temperature.
