Abstract:Electrochemical sensors can be used to detect neurotransmitters. A miniature potentiometric all-solid-state coated wire dopamine sensor was investigated in this work. Conducting polymer poly (3,4- ethylenedioxythiophene) doped with poly (styrenesulfonate) as the solid contact was electropolymerized on one end of a gold wire (diameter 0.25 mm). The conducting polymer was covered with a dopamine-selective membrane containing 12-crown-4-tetraphenylborate as a neutral carrier, 2-nitrophenyloctyl ether as a plasticizer and poly (vinyl chloride) as the membrane matrix. The dopamine sensor covered a wide linear range from 10(-5) M to 10(-1) M with a slope of 53.85 +/- 0.93 mV/decade, while the detection limit was 5.80 +/- 0.31 mu M. The other characteristics of the dopamine sensor were also evaluated. The experimental results showed that the sensor operated properly within the pH range of 5.5-7.5 and displayed good selectivity, fast dynamic response, high reproducibility and long stability. In addition, the dopamine sensor was successfully applied for the detection of dopamine in artificial cerebrospinal fluid by using the standard addition method, indicating that this sensor is promising for applications in dopamine determination in neuroscientific research.

Abstract:The present work reports the development and application of an amperometric biosensor based on carbon paste electrode modified with laccase enzyme, glutaraldehyde, and gold nanoparticles (Lac-Glu-AuNPs/CPE) for the determination of the neurotransmitter dopamine (DA). The materials were characterized morphologically and chemically using scanning electron microscopy (SEM), transmission electron microscopy (TEM), and cyclic voltammetry. Optimization studies were performed in order to determine the optimal amount of enzyme and pH level that can yield the best conditions of analysis. The application of the biosensor in optimal conditions using the amperometric technique yielded a linear concentration range of 8.0 × 10−7–6.2 × 10−5 mol L−1 with a limit of detection of 6.0 × 10−8 mol L−1. The proposed biosensor was successfully applied for the determination of DA in biological and environmental samples. In addition, the application of the biosensor for the conduct of electrochemical measurements showed that the sensing device has good repeatability and stability, and it does not suffer from matrix interference effects. The proposed biosensor exhibited an analytical signal of 85% after 10 days of consecutive use.
A carbon dots-enhanced laccase-based electrochemical sensor for highly sensitive detection of dopamine in human serum.

Abstract:Enzyme-based electrochemical sensor possesses a significant advantage in the highly efficient detection of small molecules, however, the poor electron transport efficiency limits their wide application. In this study, taking advantage of the distinct biocatalytic activity of laccase and the excellent electroconductibility of carbon dots, a carbon dots-enhanced laccase-based electrochemical sensor for the detection of dopamine (DA) is established. Thereinto, laccase can specifically recognize DA and promote its electrocatalytic oxidation on the electrode, while, the carbon dots can be used as the immobilization substrate of laccase and enhance its electron transfer efficiency, thus achieving the highly sensitive detection of dopamine. The electrochemical performance of the modified electrode interface is studied by electrochemical impedance spectroscopy and differential pulse voltammetry. As demonstrated, the electrocatalytic activity of the proposed electrochemical sensor for DA is significantly improved and exhibits a low detection limit (0.08 μM) and a wide linear range (0.25 μM-76.81 μM). The excellent selectivity allows the sensor has the capacity for specific discrimination the DA from other interferents. Furthermore, by analyzing the DA in human serum verifies the practicability of this assay in real sample analysis.
MnO 2 nanosheets based catechol oxidase mimics for robust electrochemical sensor: Synthesis, mechanism and its application for ultrasensitive and selective detection of dopamine

Abstract:Nanozymes have favorable activity and good stability, which are potential alternatives to natural enzymes and widely applied in biosensing. However, nanozymes based electrochemical sensors are fewer, because many nanozymes are not easy to form efficient electrocatalytic interface and experience interference from the possible oxidizable species. Herein, ultrathin MnO 2 nanosheets (MnO 2 NSs) were synthesized by reduction of KMnO 4 in acidic environment by chemical precipitation method. The MnO 2 NSs show excellent catechol oxidase mimicking activity. The steady-state kinetics of MnO 2 NSs shows a higher V max of 3.441 μM s −1 . Additionally, the catalytic mechanism is that in the presence of oxygen, catechol or its analogues are oxidized to quinone or quinoids, while oxygen is reduced to H 2 O. Afterwards, the possible mechanism of dopamine (DA) at MnO 2 NSs-modified electrode is investigated. DA is first catalytically oxidized to dopamine quinone (DQ) by MnO 2 NSs based catechol oxidase, which further undergoes irreversible electrochemical reduction to DA involving two protons and two electrons transfer during the process. On the basis of this, a selective and ultrasensitive amperometric detection of DA at 0.0 V (vs. SCE) was explored, which have two linear parts, 0.01–20 μM and 20–100 μM DA with the LOD of 4.1 nM (S/N = 3). Most importantly, the presence of oxidizable species including ascorbic acid, uric acid and cysteine may coexist with DA in biological systems do not exhibit any appreciable interference. The as-proposed method was applicable to detect DA in human serum samples with good satisfactory accuracy and recovery. This research casts prospect to apply nanozymes for electrochemical sensor with excellent analytical performance.

Abstract:Designing new nanozyme systems as a simple substitute of natural enzymes with higher catalytically active sites and stability has attracted growing research interests for widespread applications especially in environmental remediation. Laccases belong to a group of multicopper-containing enzymes that show significant potential in diverse biotechnological and bioremediation applications. In this paper, a facile strategy is reported for the preparation of an efficient laccase mimic inspired by structural features and electron transfer mechanism of natural laccase, via Cu 2+ ions doping in ZIF8 framework (denoted as Cu-ZIF8). Cu-ZIF8 nanozyme showed laccase-like activity and significant catalytic performance in the oxidation of a wide range of phenolic pollutants such as 2,4-dichlorophenol, phenol, catechol, hydroquinone, and o-nitrophenol. Kinetic studies were carried out to estimate the kinetic parameters of the as-prepared Cu-ZIF8 nanozyme including apparent Michaelis–Menten constant ( K m ) and maximum velocity ( V max ). The K m and V max were calculated to be 0.28 mM and 0.017 mM min −1 , respectively. In addition, Cu-ZIF8 nanozyme was robust over a wide range of temperature (30–90 °C), at extreme pH and high salt concentration. In addition, Cu-ZIF8 nanozyme can catalyze the oxidation of dopamine to a brown product, where an absorption band at 290 nm was observed. Accordingly, a simple colorimetric assay has been established for detection of dopamine in the linear range of 0.01–0.44 mM with a detection limit of 1.1 × 10 −3 mM (S/N = 3). The developed colorimetric method showed good selectivity and high sensitivity in measuring dopamine among potential interfering components.

Abstract:Dopamine is an important neurotransmitter for organisms and affects their function, so monitoring and detecting dopamine levels is essential. Sensitivity and selectivity of electrochemical sensors for the detection of dopamine usually relies on hybrid nanocomposites. In this context, we designed a micro-regionally restricted hybrid nanocomposite by modulating the micro-nano structure. Using functional groups on nitrogen-doped reduced graphene oxide (N-rGO) to restrict the random growth of tungsten disulfide nanosheets, we obtained a 2D layered material with bidirectional structural support to achieve high specific surface and high activity for application in robust and novel dopamine electrochemical sensors. A facile electrochemical sensor for dopamine detection was fabricated by modifying a screen-printed carbon electrode with the nanocomposite. The sensor exhibits excellent performance of dopamine detection in the wide range of 1 μmol/L100 μmol/L with a detection limit of 0.2 μmol/L and a high sensitivity of 0.56 μA/μmol/L. The optimized sensor shows a good selectivity, satisfactory long-term stability and reproducibility towards DA and a separation of peak potential of 142 mV with uric acid. In addition, the prepared sensor provides favorable recovery in human serum and artificial cerebrospinal solution (aSCF), indicating great potentials in electrochemical detection and biological sensing applications.

Abstract:Microfluidic Electrochemical biosensors present promising means for many critical biosensing applications such as neurotransmitter monitoring, pathogen detection, and molecular diagnostics. However, such biosensors are prone to many challenges like enzyme degradation being the major concern with electrode materials while testing multiple times, hindered detection limit range, low sensitivity due to direct contact of enzyme with the environment, and poor shelf life of the electrodes patterned by conventional techniques. Herein, a microfluidic multi-sensitive biosensor is developed using a layer-by-layer process, incorporating a novel Laccase/MXene/LIG (L-Ti 3 C 2 -G) composite designed for selective detection of dopamine and other biochemicals with applicability in human blood serum and synthetic urine. 2D nano-material, MXene, gave an excellent conductivity, more volumetric capacity, flexibility, surface hydrophilicity, and temperature stability, leading to its use in various applications. It was found that laccase immobilized electrode (L-Ti 3 C 2 -G) showcased a significant electrocatalytic activity towards biomolecules such as uric acid (0.12 V), L-cysteine (0.62 V), xanthine (0.7 V), ascorbic Acid (0.1 V), and dopamine (0.42 V). For dopamine alone, the proposed biosensor exhibits a LOD of 0.47 nM with a linear concentration range of 1 nM – and 6.37 mA nM −1 cm −2 sensitivity. Moreover, real sample analysis indicated that spiked dopamine can be determined accurately by the electrode with a recovery ratio of more than 97% in synthetic urine and human blood serum samples. The fabricated bioelectrode had high stability and reproducibility with an exceptional selectivity and negligible interference with various biochemicals.

Abstract:Dopamine is one of the significant neurotransmitters and its monitoring in biological fluids is a critical issue in healthcare and modern biomedical technology. Here, we have developed a dopamine biosensor based on surface plasmon resonance (SPR). For this purpose, the carboxymethyl dextran SPR chip was used as a surface to immobilize laccase as a bioaffinity recognition element. Data analysis exhibited that the acidic pH value is the optimal condition for dopamine interaction. Calculated kinetic affinity (K D ) (48,545 nM), obtained from a molecular docking study, showed strong association of dopamine with the active site of laccase. The biosensor exhibited a linearity from 0.01 to 189 μg/ml and a lower detection limit of 0.1 ng/ml (signal-to-noise ratio (S/N) = 3) that is significantly higher than the most direct dopamine detecting sensors reported so far. Experiments for specificity in the presence of compounds that can co-exist with dopamine detection such as ascorbic acid, urea and l -dopa showed no significant interference. The current dopamine biosensor with high sensitivity and specificity, represent a novel detection tool that offers a label-free, simple procedure and cost effective monitoring system.

Abstract:This paper presented an electrochemical aptasensor for dopamine detection with high sensitivity and selectivity. The electrochemical signal was amplified by electrochemical-chemical redox cycling with tris(2-carboxyethyl)phosphine (TCEP) as the reducing reagent. Specifically, dopamine captured by the aptamer-covered gold nanoparticles-modified carbon glass electrode was cycled by TCEP after its electrochemical oxidization, enabling an increase in the anodic current. Because of the high specificity and strong binding affinity of aptamer to dopamine, the sensor is fairly selective in not responding to common interferences. The results of chronoamperometry indicated that this aptasensor allowed for the detection of dopamine in a linear concentration range of 5nM to 0.5μM. The detection limit was estimated to be 1.8nM. Additionally, the aptasensor was successfully applied to determine dopamine spiked in the serum sample and gave recoveries ranging from 94.6 to 107.2%. This work would also be valuable for development of new types of electrochemical sensors for determination of other electroactive substances by marrying specific receptors and effective reducing or oxidizing reagents.
Hemin-doped metal-organic frameworks based nanozyme electrochemical sensor with high stability and sensitivity for dopamine detection

Abstract:This study reports a new type of artificial nanozyme based on Hemin-doped-HKUST-1 (HKUST-1, also referred to as MOF-199; a face-centered-cubic MOF containing nanochannels) as a redox mediator for the detection of dopamine (DA). Hemin-doped-HKUST-1 was successfully synthesized by one-pot hydrothermal method, which was combined with reduced graphene oxide (rGO) modified on a glassy carbon electrode (GCE) to construct a sensor (Hemin-doped HKUST-1/rGO/GCE). The morphology and structure of Hemin-doped-HKUST-1 were characterized by X-ray diffraction (XRD), scanning electron microscope (SEM), transmission electron microscope (TEM) and infrared spectra (IR) techniques. The Hemin-doped HKUST-1/rGO nanozyme showed an excellent electrocatalytic activity for DA oxidation, which is due to the enhanced Hemin activity through the formation of a metal-organic framework (MOFs) and the synergy between the Hemin-doped HKUST-1 and rGO in nanozyme. The resulted sensor exhibited a high sensitivity of 1.224 μA μM-1, with a lower detection limit of 3.27 × 10-8 M (S/N = 3) and a wide linear range of 0.03-10 μM for DA detection. In addition, due to the stabilizing effect of MOFs on heme, the sensor showed satisfactory stability and has been successfully applied to the detection of DA in serum samples, indicating that this work has potential value in clinical work.
Dopamine levels determined in synthetic urine using an electrochemical tyrosinase biosensor based on ZnO@Au core-shell

Abstract:This work presents a biosensor based on core-shell nanostructure formed by zinc oxide (ZnO) nanoparticles coated with gold (Au). The core-shell nanostructure served as a support for the immobilisation of tyrosinase on screen-printed carbon electrodes to measure dopamine using differential pulse voltammetry. While ZnO is a semiconductor with good electrical conductivity, Au offers high stability and biocompatibility, which is beneficial for maintaining enzyme activity. Atomic force microscopy (ATM), ultraviolet (UV) and infrared (IR) spectroscopy measurements confirmed that the core-shell was successfully formed. The biosensor comprising of ZnO@Au core-shell nanostructures with immobilised tyrosinase allowed the detection of dopamine in real samples with remarkable selectivity and accuracy with a relative error of 3.8%. The limit of detection and dynamic range of the biosensor for dopamine in real samples were 86 nmol L-1 and 0.1 to 500 μmol L-1, respectively. Thus, the results indicate that the proposed miniaturized biosensor device is promising for the monitoring of dopamine in real samples and can be used for disease diagnosis and prognosis. Furthermore, the reported electrochemical biosensor is of low-cost when compared to conventional techniques.

Abstract:Dopamine is one of the most important neurotransmitters and plays a crucial role in various neurological, renal, and cardiovascular systems. However, the abnormal levels of dopamine mainly point to Parkinson's, Alzheimer's, cardiovascular diseases, etc. Hydroxyapatite (HAp), owing to its catalytic nature, nanoporous structure, easy synthesis, and biocompatibility, is a promising matrix material. These characteristics make HAp a material of choice for doping metals such as cobalt. The synthesized cobalt-doped hydroxyapatite (Co-HAp) was used as a colorimetric sensing platform for dopamine. The successful synthesis of the platform was confirmed by characterization with FTIR, SEM, EDX, XRD, TGA, etc. The platform demonstrated intrinsic peroxidase-like activity in the presence of H 2 O 2 , resulting in the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB). The proposed sensor detected dopamine in a linear range of 0.9–35 μM, a limit of detection of 0.51 μM, limit of quantification of 1.7 μM, and an R 2 of 0.993. The optimization of the proposed sensor was done with different parameters, such as the amount of mimic enzyme, H 2 O 2 , pH, TMB concentration, and time. The proposed sensor showed the best response at 5 mg of the mimic enzyme, pH 5, 12 mM TMB, and 8 mM H 2 O 2 , with a short response time of only 2 min. The fabricated platform was successfully applied to detect dopamine in physiological solutions.

Abstract:Development of hybrid nanozymes with high catalytic efficiency is of critical importance for enzyme mimics in biological catalytic fields. Herein, we reported a polydopamine (PDA) surface modification-assisted pyrolysis strategy for preparing CoO nanoparticles/N-doped carbon sheets/reduced graphene oxide composites (CoO/N-CS-rGO). In this strategy, PDA with abundant amines and imines could serve as an eco-friendly N-contained precursor and a connecting agent between GO matrix and CoO. Due to the superior conductivity of N-CS-rGO, synergistic catalysis between CoO nanoparticles (NPs) and N-CS-rGO, and intrinsic oxidase-like characteristic, the CoO/N-CS-rGO was used as an ultrasensitive signal amplification platform for simultaneously analyzing dopamine (DA) and uric acid (UA) in a neutral solution. Significantly, the fabricated electrochemical sensor showed ultrahigh sensitivities of 1378 μA mM–1 for DA and 1393 μA mM–1 for UA. The linear range of 0.5–110 μM for DA and 1–125 μM for UA could be obtained by this sensor. Furthermore, the stability and selectivity of the CoO/N-CS-rGO-based hybrid biosensor were evaluated. The results showed that the biosensor performed good stability and brilliant selectivity against interfering analytes. Also, the CoO/N-CS-rGO/GCE was adopted to monitor two analytes in human serum samples with high accuracy. This work provides an efficient PDA-assisted synthetic strategy to prepare hybrid nanomaterials and shows broad prospects for biosensing, biotechnology, and clinical diagnosis.

Abstract:New polydopamine (PDA)-laccase (Lac)-Fe3O4 magnetic polymeric bionanocomposites (MPBNCs) were prepared through one-pot Lac-catalyzed oxidation of dopamine in an aqueous suspension containing Lac, magnetic Fe3O4 nanoparticles and dopamine. A PDA-Lac-Fe3O4/Au electrode was prepared by facile and efficient magnetic separation/immobilization of the MPBNCs on a magnetic Au electrode for biosensing of hydroquinone (HQ), giving a sensitivity of 374μAmmol−1Lcm−2, a limit of detection of 30nmolL−1, as well as good selectivity and stability. The biosensing electrode was also used for HQ analysis in real sample matrices with satisfactory results.

Abstract:Carbon nanotube-encapsulated nickel selenide composite nanostructures were used as nonenzymatic electrochemical sensors for dopamine detection. These composite nanostructures were synthesized through a simple, one-step, and environmentally friendly chemical vapor deposition method, wherein the CNTs were formed in situ from pyrolysis of a carbon-rich metallo-organic precursor. The composition and morphology of these hybrid NiSe2-filled carbon nanostructures were confirmed by powder X-ray diffraction, Raman, X-ray photoelectron spectroscopy, and high-resolution transmission electron microscopy images. Electrochemical tests demonstrated that the as-synthesized hybrid nanostructures exhibited outstanding electrocatalytic performance toward dopamine oxidation, with a high sensitivity of 19.62 μA μM–1 cm–2, low detection limit, broad linear range of 5 nM–640 μM, and high selectivity. The synergistic effects of enhanced electrochemical activity of nickel selenide along with the enhanced conductivity of carbon nanotubes led to the high electrocatalytic efficiency for these nanostructured composites. The high sensitivity and selectivity of this nanostructured composite could be exploited to develop simple, selective, and sensitive electrochemical sensors to detect and quantify dopamine in human tear samples with high reliability. This nanotube-encapsulated sensor, hence, paves the way for discoveries in the development of dopamine sensors with low cost and high stability, which can be used for noninvasive dopamine detection in peripheral bodily fluids.

Abstract:As a biomarker of endocrine gland diseases, the rapid and accurate determination of dopamine has a great significance. The aim of this study was to improve the dopamine biosensing performance of the Fe3O4@CS-Au-Lac (laccase immobilized on Fe3O4@CS-Au nanocomposites) by using the photothermal effect of magnetic carriers. Electrochemical impedance spectroscopy measurements revealed that the Fe3O4@CS-Au nanocomposites enhanced the electron transfer between laccase and the electrode. The Fe3O4@CS-Au-Lac biosensor presented an ultra-fast response to substrates, accurately detected dopamine within 0.2 s. Dopamine biosensing assays demonstrated that the Fe3O4@CS-Au-Lac biosensor possessed high sensitivity, remarkable tolerances towards organic compounds and metal ions, and excellent selectivity and stability. Amperometric curves indicated that the illumination brought a higher current to the biosensor compared to the dark environment. The Fe3O4@CS-Au-Lac biosensor held a low detection limit of 0.79 μM (S/N = 3) and a broad linear range of 1–1000 μM under the irradiation. Most significantly, the Fe3O4@CS-Au-Lac biosensor provided reliable results in the photothermal analysis of dopamine in synthetic urine samples. This finding paves the way for using the Fe3O4@CS-Au-Lac biosensor for photothermal detection of dopamine in biomedical applications.

Abstract:This article addresses the construction of a dopamine electrochemical sensor based on a molecularly imprinted polymer on carbon electrodes, for instance, glassy carbon and highly oriented pyrolytic graphite electrodes with the orientation of edge and basal plane by the formation of thiophene acetic acid-dopamine thin film using electropolymerization. The electrochemical deposition of Pt nanoparticles was then carried out at the modified electrode surface, followed by the extraction of dopamine as a template molecule from the generated layer was performed via elution brought on by chemicals. The variables that affect the performance of the imprinted polymer-based sensor, such as monomer-template ratio, immersion time, and the number of electropolymerization cycles, were examined and optimized. To confirm the changes in the oxidation peak current of DA and to investigate the electrochemical behavior of the MIP sensor, cyclic voltammetry, chronoamperometry, and differential pulse voltammetry (DPV) tests were performed. The differential pulse voltammetry studies revealed that, under ideal circumstances, the limit of detection values of the proposed MIP sensor decorated with Pt nanoparticles was found to be 14.40 nmol L −1 , 42.50 nmol L −1 , and 0.671 μmol L −1 for glassy carbon, edge, and basal plane electrodes, respectively. In the presence of interferents with comparable structural chemicals, such as ascorbic acid, uric acid, glucose, o-phenylenediamine, and glycine, the proposed sensor exhibits noticeable selectivity. Dopamine analysis in a human fluid such as blood serum was conducted with success using the devised sensor, which was shown to possess impressive stability and reproducibility.

Abstract:A novel electrochemical sensor using the molecularly imprinted (MIP) oxygen-containing polypyrrole (PPy) decorated carbon nanotubes (CNTs) composite was proposed for in vivo detection of dopamine (DA). The prepared sensor exhibits a remarkable sensitivity of (16.18μA/μM) with a linear range of 5.0×10−11–5.0×10−6M and limit of detection as low as 1.0×10−11M in the detection of DA, which might be due to the plenty cavities for binding DA through π–π stacking between aromatic rings and hydrogen bonds between amino groups of DA and oxygen-containing groups of the novel PPy.

Abstract:The studies on a nano-molar sensitive disposable biosensor based on the screen-printing basal electrode and the supramolecular assembly of dual mediators, tetramethylbenzidine and ferrocene, by beta-cyclodextrin inclusion complexation have been reported in this paper. This biosensor shows the high quality of analytical performance because of the complexation and the microenvironment in the supramolecular structure. The extracted protein of mushroom, which contains abundant polyphenol oxidase, was immobilized in the membrane of cross-linked cyclodextrin film. This biosensor responds to dopamine with high selectivity and sensitivity. Its response range on the concentration of dopamine is from 1.0 x 10(-9) to 1.0 x 10(-6) M with the detection limit as low as 5.0 x 10(-10) M. The response time reaching to 90% of its steady value is less than 60 s. The biosensor can keep its activity for at least 15 days.

Abstract:A novel electrochemical sensing platform based on nitrogen-doped graphene (N-G)/nickel tetrasulfonated phthalocyanine (NiTsPc) nanocomposites has been developed for selective determination of dopamine (DA). NiTsPc was immobilized on N-G matrix via π-π interaction, while N-G provided a compatible microenvironment for NiTsPc to enhance electron transfer and to retain its electrocatalytic activity as well. The N-G/NiTsPc nanocomposite was characterized by using transmission electron microscopy (TEM), electrochemical impedance spectroscopy (EIS), X-ray photoelectron spectroscopy (XPS) and cyclic voltammetry (CV). The proposed modified electrode presented superior catalytic efficiency for DA in the presence of ascorbic acid (AA) and uric acid (UA) owing to the synergistic effect of N-G and NiTsPc. The sensors also exhibited a wide linear range from 1×10−7 to 2×10−4M and a limit of detection as low as 100nM with high sensitivity of 88.9μAmM−1.
