Abstract:Palladium-catalyzed enantioselective C(sp3)-H functionalization reactions has attracted considerable attention due to its ability for the synthesis of enantiomerically enriched molecules and stimulation of novel retrosynthetic disconnections. Understanding the reaction mechanism, especially the stereochemical process of the reaction, is crucial for the rational design of more efficient catalytic systems. Previously, we developed a Pd(II)/sulfoxide-2-hydroxypridine (SOHP) catalytic system for asymmetric C(sp3)-H functionalization reactions. In this study, we focused on unraveling the chemistry of chiral palladacycles involved in the Pd(II)-catalyzed enantioselective C(sp3)-H functionalization. We have isolated key palladacycle intermediates involved in the enantioselective β-C(sp3)-H arylation of carboxylic acids catalyzed by the Pd(II)/SOHP system. These palladacycles, exhibiting ligand-induced chirality, provided a significant opportunity to investigate the stereochemical process and the ligand effect in this asymmetric C-H functionalization. Our investigation provided direct evidence for the C-H palladation step as the enantioselectivity-determining step, which forms diastereomeric palladacycles that exhibited preservation of chirality in the functionalization step. DFT calculations provided insights into the chiral induction in palladacycle formation. This work highlights the value of chiral palladacycle chemistry in offering mechanistic insights into the Pd(II)-catalyzed asymmetric C(sp3)-H functionalization reactions.

Abstract:A direct, efficient, and practical protocol to install a chiral oxazoline unit onto aryl/heteroaryl rings via palladium-catalyzed C-H functionalization of 2-positions of oxazolines with a variety of halides using dppe as the ligand has been developed. Various chiral oxazoline ligands could be synthesized, even in a 10-g scale process. This protocol is a good supplement to traditional methods and for diverse synthesis of chiral oxazoline ligands.

Abstract:In transition metal-catalyzed asymmetric synthesis, enantioselectivity strongly depends on the structures of chiral ligands, so the development of new chiral ligands is crucial. Here, an efficient and highly enantioselective palladium-catalyzed intramolecular hydroarylation has been developed, and a new kind of N-heterocycles, 1H-pyrazolo[5,1-a]isoindol-2(8H)-ones containing a quaternary stereocenter, was prepared in high yields and excellent enantiomeric excess values. The reaction was effectively catalyzed by palladium-diphosphine complexes with numerous functional group tolerance, in which the newly developed axially chiral cyclic diphosphine ligands played key roles in the reactivity and enantioselectivity of the substrates. We believe that the cyclic diphosphine ligands with adjustable dihedral angles will find wide application in asymmetric synthesis.

Abstract:Planar-chiral macrocycles play a pivotal role in host-guest chemistry and drug discovery. However, compared with the synthesis of other types of chiral compounds, the asymmetric construction of planar-chiral macrocycles still remains a forbidding challenge. Herein, we report a sequential palladium and N-heterocyclic carbene catalysis to build planar-chiral macrocycles. This protocol features board scope and good functional group tolerance, and allows a rapid assembling of planar-chiral macrocycles with excellent enantioselectivities.

Abstract:Planar-chiral cycloalkenes are challenging synthetic targets due to the medium-sized ring and the trans-configured olefin. Asymmetric resolution of racemic E-cycloalkenes, UV-induced asymmetric isomerization of Z-cycloalkenes, and asymmetric intramolecular cyclization of olefinic substrates are known as the limited approaches to these scaffolds. However, these methods usually suffer from tedious synthesis, harsh conditions, and insufficient structural diversity. Here, we present a novel palladium-catalyzed asymmetric [7+2] cycloaddition to facilitate the modular synthesis of planar-chiral 9-membered cycloalkenes from readily available chemicals. Excellent selectivity for planar chirality was achieved by retaining the trans-2H configuration of the π-allyl-Pd species throughout the whole process. Mechanistic studies were performed to elucidate the pivotal role of hydrogen bonding from the chiral ligand in enhancing both reaction efficiency and stereocontrol. In addition, synthetic transformations of planar chiral cycloalkene products and their applications in the identification of anticancer agents and selective bioimaging of cancer cells demonstrated the synthetic value of this new methodology. This research provides a promising way to access synthetically valued planar-chiral cycloalkenes.

Abstract:Planar chiral ferrocenyl phosphines have been employed as highly valuable ligands in metal-catalyzed asymmetric reactions. However, their preparation remains a formidable challenge due to the requirement for intricate, multistep synthetic sequences. In addressing this issue, we have developed a groundbreaking enantioselective C-H activation strategy facilitated by P(III) directing groups, enabling the efficient construction of planar chiral ferrocenyl phosphines in a single step. Our innovative approach entails the combination of a palladium catalyst, a parent ferrocenyl phosphine, and a chiral phosphoramidite ligand, leading to exceptional reactivity and enantioselectivity. Remarkably, these novel ligands exhibit remarkable efficacy in silver-catalyzed asymmetric 1,3-dipolar cycloadditions. We carried out a combination of experimental and computational studies to obtain a more comprehensive understanding of the reaction pathway and the factors contributing to enantioselectivity.

Abstract:The merging of C-H and C-C bond cleavage into one single chemical process remains a daunting challenge, especially in an asymmetric manner. Herein, a Pd(II)-catalyzed enantioselective tandem C-H/C-C activation for the synthesis of axially chiral biaryls is described. Two types of simple cyclopropanes, such as vinylcyclopropanes and cyclopropanols, were used as efficient and readily available coupling partners. This catalytic system features good functional group compatibility, excellent enantiocontrol (up to >99% ee) and the first use of palladium catalyst in this process. The synthetic utility of this protocol was demonstrated by gram-scale synthesis and further synthetic transformations to access various axially chiral biaryls with high enantiopurity. Two distinct but closely related C-C cleavage pathways of cyclopropanes were achieved in the enantioselective C-H/ C-C activation process, which represents a novel platform to further utilize the ring-opening attribute of cyclopropanes in asymmetric catalysis. Preliminary mechanistic studies provide insights into the role of cyclopropanols, which may pave the way for the development of novel catalytic transformations.
Enantioselective Construction of Axially Chiral Cyclohexylidene Scaffolds Via Pd-catalyzed Asymmetric Coupling Reaction

Abstract:Axially chiral cyclohexylidenes exhibit unique chirality because of the restricted rotation of the C=C bond. Compared with other axially chiral compounds, they aremuch less explored, and their efficient construction remains a synthetic challenge. Herein, we report the first palladium-catalyzed asymmetric coupling reaction of aryl iodides with 4-substituted cyclohexanol-derived alpha-diazocarbonyl compounds to give tetrasubstituted cyclohexylidenes in good yields (up to 92%) with excellent enantioselectivities (up to 96% ee) under mild conditions. This protocol provides a general access to axially chiral cyclohexylidenes. The utility of this unique scaffold was demonstrated by the carboxylic acid serving as a useful chiral ligand in transition-metal-catalyzed C-H bond activation, which showed great promise as a new type of olefin ligand.

Abstract:Asymmetric cycloaddition reactions are the most powerful tool to the expeditious construction of enantioenriched cyclic motifs in organic chemistry. In sharp contrast to well-developed cycloaddition reactions via the palladium-trimethylenemethane (Pd-TMM) intermediate, hetero (3 + 2) cycloadditions of the heteroallyl cations remain rare, largely due to their thermally forbidden nature. To the best of our knowledge, there is no example of asymmetric version leading to enantioenriched heterocycles reported so far. Herein we enabled the first example of catalytic asymmetric (3 + 2) cycloaddition of electrophilic palladium-heteroallyl zwitterion intermediate (Pd-OTMM or Pd-NTMM) with cyclic or acyclic 1,3-dienes via a pathway terminated with C-N or C-O bond formation, delivering the highly substituted or fused pyrrolidine and tetrahydrofuran rings in high yields with excellent regio-, diastereo-, and enantioselectivity. Engineering the PC-Phos, one of the chiral sulfinamide phosphine (Sadphos) type ligands, by introducing the di-tert-butyl or/and 3,5-difluorophenyl group is a vital component in achieving excellent catalytic reactivity and enantioselectivity.

Abstract:A pair of novel palladium complexes {[(1S,2S)-L]PdBr2} and {[(1R,2R)-L]PdBr2} of chiral di-N-heterocyclic carbene ligands derived from chiral 1,2-cyclohexanediamine have been prepared in moderate to good yields and characterized by NMR, HRMS and X-ray single crystal diffraction studies. The obtained chiral NHC–Pd compound was able to catalyze the asymmetric Suzuki–Miyaura couplings of aryl bromides with arylboronic acids in good yields and moderate enantioselectivities (up to 61% ee).

Abstract:Bipyridines represent a class of ligands renowned for their versatility and efficacy in numerous transition metal-catalyzed reactions. Chiral bipyridine ligands are noted for their distinctive reactivity and stereoselectivity. In this work, we have designed and synthesized a class of bipyridine ligands endowed with an axially chiral scaffold. These ligands feature: (a) a precisely aligned coplanar bipyridine framework that is beneficial for metal chelation; (b) a spacious environment around the metal-bipyridine complexes, capable of hosting substrates with substantial steric bulk; (c) a chiral pocket induced by the axially chiral architecture. These atropochiral planar bipyridine ligands were successfully applied in copper-catalyzed ring-opening reactions of cyclic diaryliodoniums with bulky secondary amines, achieving high efficiency and stereoselectivity that were unsuccessful in our previous efforts.

Abstract:Axially chiral biaryls exist widely in bioactive natural products and auxiliary ligands for homogeneous catalysis. The particular spatial arrangement of the two aromatic rings in the specific space determines their unique properties; therefore, how to effectively control their absolute configuration is the key issue. Within the last decade, the synthesis of axially chiral biaryls has been a research topic of great interest. Asymmetric biaryl coupling controlled by chiral palladium catalysts was first reported in 2000 and has greatly advanced the chemistry of transition-metal-catalyzed cross-coupling reactions. Most notably, some novel chiral auxiliary ligands such as ferrocene- or biaryl backbone-containing monophosphine, bisphosphines, bis-hydrazones, hydrazone-phosphines, diene and NHC ligands have been applied to this reaction. At the same time a broad range of aryl electrophiles such as phenols, aryl ethers, esters, carbonates, carbamates, sulfamates, phosphates, phosphoramides, phosphonium salts, and fluorides can now be coupled efficiently with boron reagents to afford functionalized axially chiral biaryl products with good to excellent yields and enantioselectivities in the presence of chiral palladium catalysts. N,N′-Bisaryl Substituted Chiral Linker-Bridged Bis(N-Heterocyclic Carbene) Palladium Complexes: Design, Synthesis, and Catalytic Properties

Abstract:A series of novel N,N'-bisaryl bis(NHC) (NHC = N-heterocyclic carbene) precursors 5a-e, 12a-d, and 13a-c and their palladium complexes 14a-e, 15a-d, and 16a,b with (1R,2R)-cyclohexene and (1R,2R)-diphenylethylene linkers have been designed, synthesized in good yields, and fully characterized by NMR, HRMS, and elemental analysis. The X-ray single crystal analysis of 15a showed that these complexes adopted a slightly distorted square-planar geometry around the Pd(II) center. These N,N'-bisaryl-bis(NHC)-Pd complexes exhibit excellent catalytic activity in asymmetric aryl aryl cross-coupling reactions of arylboronic acids and aryl halides. The axial chiral biaryl compounds could be achieved in high yields (up to 74%) and good enantioselectivities (up to 60% ee) within 24 h. The results show that for this kind of bis(NHC) palladium catalysts the structure characteristics of the chiral linkers have decisive effect on the enantioselectivities of cross-coupling reactions.

Abstract:Chiral half-sandwich cyclopentadienyl rhodium(III) (CpRhIII) complexes are potent catalysts for asymmetric C–H activation. They are primarily produced from chiral Cp ligands bearing diverse chiral backbones. However, it remains a big challenge to develop planar-chiral CppxRhIII catalysts with prochiral Cp ligands (Cppx) due to the lack of design guidelines and ideal chiral resolution methods. To solve this problem, we present herein a feasible design concept and a robust chiral resolution strategy. Notably, a type of planar-chiral CppxRhIII catalyst bearing prochiral tetrasubstituted Cppx ligands with the formula C5HMe2PhAr has been successfully developed by routine silica-gel chromatographic separation of related diastereomeric chiral diene derivatives. This catalyst proved to be highly effective for asymmetric C–H activations, including the reaction of N-methoxybenzamides with 1,4-benzoquinone (up to 92% yield, 96% ee) and the reaction of O-Boc hydroxamates with norbornene (up to 92% yield, 94% ee).

Abstract:Asymmetric synthesis of chiral allenes starting from prochiral substrates under mild reaction conditions promoted by Pd-SYNPHOS catalyst is reported. This protocol provides an efficient access to various enantioenriched aryl- and alkyl- substituted allenes, which are versatile building blocks of high utility to both organic and medicinal chemists, in excellent isolated yields (up to 96%) and high enatiomeric ratio values (up to 95:5). In addition, a comparative study using several C-2-symmetric atropisomeric diphosphine ligands revealed the overwhelming impact of the steric and electronic properties of the ligands for the catalytic efficiency of this process.

Abstract:An unusual hybrid palladium catalyst containing an anionic chiral CoIII complex and a chiral phosphoramidite ligand shows a high capacity for catalyzing asymmetric thioamide-directed C(sp3 )-H arylation and delivers excellent yield and enantioselectivity (up to 99 % yield, 99 % ee). Significant synergy between the chiral ligand and the anion in terms of stereochemical control was observed. Mechanistic investigations have revealed both the nature of the C-H activation and the origin of the enantioselectivity.

Abstract:A palladium-catalyzed asymmetric synthesis of planar chiral metallocene compounds is reported. The reaction stereoselectively functionalized one of the ortho C-H bonds of Cp rings by intramolecular cyclization to form indenone derivatives in high yields with excellent enantioselectivity. The mild set of reaction conditions allowed a wide variety of chiral metallocene compounds to be synthesized with broad functional group tolerance. The influences of preinstalled chiralities on the other Cp-ring were also investigated.

Abstract:Over the past few decades, there have been major developments in transition metal-catalyzed asymmetric cyclization reactions, enabling the convenient access to a wide spectrum of structurally diverse chiral carbo- and hetero-cycles, common skeletons found in fine chemicals, natural products, pharmaceuticals, agrochemicals, and materials. In particular, a plethora of enantioselective cyclization reactions have been promoted by chiral palladium catalysts owing to their outstanding features. This review aims to collect the latest advancements in enantioselective palladium-catalyzed cyclization reactions over the past eleven years, and it is organized into thirteen sections depending on the different types of transformations involved.

Abstract:The series of chiral 1,2-cyclohexane-bridged bis-N-heterocyclic carbene ligand precursors H-2[(1R,2R)-(1a- i)]Br with different substituent groups and their neutral and cationic diaqua palladium complexes, namely {Pd[(1R,2R)-(1a -i)]Br-2) (2a-i), {Pd[(1R,2R)-(1a)]X-2} (X = Cl (3), OAc (4-OAc), OC(O)CF3 (4-OCH(O)CF3)), and {Pd[(1R,2R)(L-OMe)] (OH2)(2)}X-2 = (X = OTf (5-OTf), SbF6 (5-SbF6)) have been prepared in moderate to good yields. These chiral palladium complexes were fully characterized by elemental analysis, high-resolution mass spectra, H-1 and C-13 NMR, and optical rotation determinations. The crystal structures of the chiral complexes 2a and 5-OTf were further confirmed to adopt a distorted-square-planar coordination geometry around the palladium center. The obtained chiral NHC-Pd compounds were able to catalyze the asymmetric Suzuki-Miyaura couplings of aryl halides with arylboronic acids in good yields (up to 96%) and moderate enantioselectivities (up to 64% ee). The coligand and steric effects were studied carefully. The coligands, including Br-, Cl-, AcO-, CF3COO(-), and water molecules, have little influence on the catalytic results. However, a strong steric effect of the two aromatic substituents R on the enantiocontrol has been proved in the catalytic asymmetric Suzuki-Miyaura coupling reaction. The highest enantioselectivity of 64% ee could be achieved under the standard reaction conditions.
