See all article types » Following the publication of this article [1], concerns were raised regarding results presented in Figs 1 and 5. Specifically, when levels are adjusted to visualize the background: The corresponding author provided additional methodological information to explain the appearance of vertical discontinuities: Western blots were performed using the mini-protean II multiscreen apparatus (Bio-Rad, Hercules, CA). This technique results in spaces appearing between the raised lanes where there is no contact with the primary antibody, and when the blots are scanned into a computer, the raised effect of each lane is lost and each lane appears as a separate blot (see example images in S1 and S2 Files). In the LJL23 Dog panel in Fig 5, there was no reaction to the LJL23 antigen in the negative control sample in lane 6 and in the LJL13 Human panel of Fig 5, lane 7 had no sample and was used to separate samples on the left from samples on the right. The corresponding author provided the available uncropped underlying images for this article (S3–S7 Files) including the Dog and Fox panels of Fig 1B, and the Dog LJL23 panel of Fig 5, but they were unable to retrieve all original images for the figures included in the article. In evaluating the "-" lane of the Dog panel of Fig 1B, PLOS noted some differences between the published figure and the underlying blot image. Although this issue was not resolved, PLOS concluded that the data provided for Figs 1B and 5 appear to support the results reported in [1]. Specifically: A corresponding author (SR) disagreed about the first and third concerns discussed above. They stated that the published data are correct for lanes 5, 6, and 10; an incorrect image was used during the preparation of lane 8; and the original blot image for lane 7 is no longer available. The PLOS Neglected Tropical Diseases Editors remain concerned that lanes 4 and 7 and lanes 6 and 10 appear more similar than would be expected for independent results. Regarding the concerns in multiple lanes containing similar repeating patterns, the responding corresponding author stated these were likely caused by indentations when the strips were handled with forceps or small debris such as dust. The original underlying data for Fig 1 and repeat experiment data were provided (S1 File). The experiments in Fig 1 were used to demonstrate that the plasmids used in subsequent in vivo experiments drive expression of the expected proteins in COS-7 cells. The PLOS Neglected Tropical Diseases Editors consider that the raw data provided in S1 File support that His-tagged proteins of the indicated sizes were expressed from the plasmids. However, we remain concerned about the integrity of the published Fig 1 regarding additional bands evident in the raw data. Therefore, the PLOS Neglected Tropical Diseases Editors issue this Expression of Concern. Specifically: In response, author JMS stated that the above concerns are artifacts generated through reproduction of the image and that the areas of concern are not relevant to the interpretation of the respective figure. They provided underlying data from the original experiments for Fig 1 (S1–S3 Files) which do not show the similar regions listed above. The PLOS Neglected Tropical Diseases Editors therefore consider these concerns to be resolved. A member of the PLOS Neglected Tropical Diseases Editorial Board stated any differences between S1-S5 Files and the published images would be expected for 2D-gel electrophoresis and 2D-western blots due to the methods used, and the underlying images in S1-S5 Files support the conclusions reported in [1] which do not solely rely on the gel and blot data as the findings were further validated by MS/MS. Author JMS stated that lanes 2 and 3 in S4-S5 Files were cut from Fig 3 as they were controls not relevant to this experiment. An updated version of Fig 3 with the cut indicated by a vertical black line is provided here. The remainder of the data underlying this article are available from author JMS. The His-tagged recombinant protein (r Ti SP14.6) was purified by HPLC (lane 1) and deglycosylated with PNGase F (lane 2), PNGase F and Sialidase A (lane 3) or PNGase F, Sialidase A and O-glycanase (lane 4).
