Abstract:We describe a highly sensitive electrochemical sandwich-type immunosensor for the prostate specific antigen (PSA), a biomarker related to prostate cancer. A glassy carbon electrode was covalently modified with multi-walled carbon nanotubes to which the anti-PSA was immobilized. Following addition of a sample containing PSA, a secondary antibody is added that consists of gold nanoparticles modified with secondary antibody and 6-ferrocenyl hexanethiol acting as the signal molecule. This approach represents a multiple signal amplification strategy in that the functionalized carbon nanotubes improve the electron transfer on the surface of the electrode, while the gold nanoparticles act as carriers for capturing large quantities of Ab2 and ferrocene. The modifying processes were characterized by cyclic voltammetry and electrochemical impedance spectroscopy. Under the optimal conditions, the differential pulse voltammetric signals (acquired at a typical potential of 0.31 V vs. SCE) is linearly related to the concentration of PSA in the 10 pg.mL−1 to 100 ng.mL−1 range. The detection limit is 5.4 pg.mL−1. When applied to the determination of PSA in spiked human serum, the recoveries were between 92.4 and 120.0 %. Abstract:Prostate specific antigen (PSA), as a biomarker, plays important roles in early diagnosis of male prostate diseases, especially prostate cancer. In this study, a novel electrochemical probe polymer of poly(ferriporphyrin-co-acrylamide) was synthesized, by further combined with reduced graphene oxide and gold nanoparticles, and oriented immobilization of PSA antibody, a label-free electrochemical immunosensor was obtained, and then successfully applied in the determination of PSA in 10 % non-antigen serum system. The prepared biosensor expressed high selectivity toward PSA, with a LOD of 0.001 ng mL−1, a linear range of 0.01–110 ng mL−1, and a sensitivity of 15.78 µA mL ng−1. What’s more, it showed good stability, well repeatability and high selectivity in the real PSA sample detection. This work provided certain references for the design of the new electrochemical probes and the detection of tumor markers.

Abstract:A novel and competitive electrochemiluminescence (ECL) aptasensor for prostate specific antigen (PSA) assay was constructed using gold nanorods functionalized graphene oxide (GO@AuNRs) multilabeled with glucose oxidase (GOD) and streptavidin (SA) toward luminol-based ECL system. A strong initial ECL signal was achieved by electrodeposited gold (DpAu) on the electrode because of gold nanoparticles (AuNPs) motivating the luminol ECL signal. The signal probes prepared by loading GOD and SA-biotin-DNA on GO@AuNRs were used for achieving multiple signal amplification. In the absence of PSA, the signal probes can be attached on the electrode by hybridization reaction between PSA aptamer and biotin-DNA. In this state, the GOD loaded on the probe could catalyze glucose to in situ produce H2O2 and then AuNRs catalyze H2O2 to generate abundant reactive oxygen species (ROSs) in luminol ECL reaction. Both the high-content GOD and AuNRs in the signal probe amplified the ECL signal in the ECL system. Moreover, the combination of SA with biotin-DNA further expands ECL intensity. The integration of such amplifying effects in this protocol endows the aptasensor with high sensitivity and good selectivity for PSA detection. This aptasensor exhibits a linear relation in the range of 0.5pgmL-1 to 5.0ngmL-1 with the detection limit of 0.17pgmL-1 (S/N = 3). Besides, the strategy was successfully applied in determination of human serum samples with recovery of 81.4-116.0%.

Abstract:In this work, a novel ratiometric electrochemical aptasensor was designed for prostate specific antigen (PSA) detection based on toehold-mediated strand displacement reaction (TMSDR). For PSA detection, PSA could bind with the aptamer sequence in hairpin probe 1 (HP1), and HP1/PSA complex was obtained. Then, in the presence of S3 DNA, HP1/PSA could trigger two TMSDR processes by hybridizing with Fc-S2/S1 double-stranded DNA, which would generate a large amount of Fc-S2 and recycle HP1/PSA. This enabled the hybridization of Fc-S2 with methylene blue labeled thiolated hairpin probe 2 (MB-HP2) immobilized on the electrode surface, causing MB molecules away from the electrode and Fc close to the electrode. Thus, with the increasing PSA concentrations, the current of MB (I MB ) decreased and the current of Fc (I Fc ) increased. The ratio of I Fc /I MB was shown to be linear with the logarithm of PSA concentration from 1 pg mL −1 to 100 ng mL −1 , with a detection limit of 0.28 pg mL −1 . In addition, the aptasensor had good specificity, reproducibility, repeatability, and stability for PSA detection, and was successfully applied to spiked diluted human serum samples.

Abstract:Prostate specific membrane antigen (PSMA)-positive exosomes have the potential to serve as highly sensitive biomarkers for prostate cancer detection. Herein, a sensitive electrochemical biosensor for the ultrasensitive detection of PSMA-positive exosomes has been constructed based on a peptide-templated AgNPs nanoprobe. In this work, PSMA-specific binding peptides immobilized on a gold electrode were responsible for prostate cancer-derived exosomes capturing. Well-designed peptide (CCY- LWYIKC) serves a dual role: as a signal probe and as a recognizer in the exosomes-identification process. Specifically, LWYIKC bind to cholesterol at the exosome membranes, and CCY function as peptide templates to host a large number of silver nanoparticles, leading to a strong electrochemical signal. Thus, the concentration of exosomes can be quantified via electrochemical signal. This innovative method displayed a wide detection range of 10 2 to 10 8 particles/μL and a detection limit as low as 37 particles/μL. Notably, the method has shown outstanding performance when validated using clinical samples, suggesting its potential for clinical applications.

Nanogold-functionalized G-C3n4 Nanohybrids for Sensitive Impedimetric Immunoassay of Prostate-Specific Antigen Using Enzymatic Biocatalytic Precipitation

Abstract:This work reports on a new impedimetric immunosensing strategy for sensitive detection of prostate-specific antigen (PSA) in biological fluids. The assay was carried out on monoclonal anti-PSA capture antibody-modified glassy carbon electrode with a sandwich-type detection format. Gold nanoparticles-decorated g-C3N4 nanosheets (AuNP/g-C3N4), synthesized by the wet-chemistry method, were utilized for the labeling of polyclonal anti-PSA detection antibody and horseradish peroxidase (HRP). Upon target PSA introduction, the sandwiched immunocomplex could be formed between capture antibody and detection antibody. Followed by the AuNP/g-C3N4, the labeled HRP could catalyze 4-choloro-1-naphthol into benzo-4-chlorohexadienone. The as-generated insoluble product was coated on the electrode surface, thus increasing the Faradaic impedance of Fe(CN)6(4-/3)(-) indicator between the solution and the base electrode. Under the optimal conditions, the impedance increased with the increasing target PSA in the sample, and exhibited a wide linear range from 10pgmL(-1) and 30ngmL(-1) with a detection limit of 5.2pgmL(-1). A repeatability and intermediate precision of <14% was accomplished. The specificity and method accuracy in comparison with commercial PSA ELISA kit for analysis of human serum specimens were relatively satisfactory.

Abstract:Prostate specific antigen (PSA) is a widely used marker for the diagnosis of prostate cancer, and the increasing attention has been attracted on the development of rapid assay using biosensing technology. However, it remains challenging for the sensitive and selective detection of PSA in clinical samples. Here, we report a label-free microfluidic paper-based analytical device for highly sensitive electrochemical detection of PSA. The paper device was fabricated with wax printing to generate hydrophobic and hydrophilic layers for the construction of microfluidic channel, followed by screen-printing of three electrodes including working, counter and reference electrode. Gold nanoparticles (AuNPs)/reduced graphene oxide (rGO)/thionine (THI) nano composites were synthesized and characterized, which were coated onto working electrodes for the immobilization of DNA aptamer probe. THI servers as the electrochemical mediator to transduce the biological recognition between DNA aptamer and PSA, and the excellent conductivity of AuNPs and rGO also play a significant role of electron transfer, leading to a sensitive detection for PSA, able to detect PSA as low as 10 pg mL-1, with a linear range from 0.05 to 200 ng mL-1. We demonstrated that our electrochemical sensor for the detection of clinical serum samples, indicating that our sensor would provide a new platform for low cost, sensitive and point-of-care diagnosis of prostate cancer.
DNA walker-amplified signal-on electrochemical aptasensors for prostate-specific antigen coupling with two hairpin DNA probe-based hybridization reaction

Abstract:Electrochemical aptasensing systems have been developed for screening low-abundance disease-related proteins, but most of them involve multiple washings and multi-step separation during measurements, and thus are disadvantageous for routine use. In this work, an innovative and simple electrochemical aptasensing platform was designed for the voltammetric detection of prostate-specific antigen (PSA) in biological fluids without any washing and separation steps. This system mainly included a PSA-specific aptamer, a DNA walker and two hairpin DNA probes (i.e., thiolated hairpin DNA1 and ferrocene-labeled hairpin DNA2). Introduction of target PSA caused the release of the DNA walker from a partially complementary aptamer/DNA walker hybridization strand. The dissociated DNA walker opened the immobilized hairpin DNA1 on the electrode, accompanying subsequent displacement reaction with hairpin DNA2, thus resulting in the DNA walker step-by-step reaction with numerous hairpin DNA1 probes on the sensing interface. In this case, numerous ferrocene molecules were close to the electrode to amplify the voltammetric signal within the applied potentials. All reactions and electrochemical measurements including the target/aptamer reaction and hybridization chain reaction were implemented in the same detection cell. Under optimal conditions, the fabricated electrochemical aptasensor gave good voltammetric responses relative to the PSA concentrations within the range of 0.001-10 ng mL-1 at an ultralow detection limit of 0.67 pg mL-1. A good reproducibility with batch-to-batch errors was acquired for target PSA down to 11.5%. Non-target analytes did not interfere with the voltammetric signals of the electrochemical aptasensors. Meanwhile, 15 human serum specimens were measured with electrochemical aptasensors, and displayed well-matched results in comparison with the referenced human PSA enzyme-linked immunosorbant assay (ELISA) method. Significantly, this method provides a new horizon for the quantitative monitoring of low-concentration biomarkers or nucleic acids.

Abstract:A colorimetric immunoassay has been reported for prostate-specific antigen (PSA) detection with CuO nanoparticles (CuO NPs) as signal labels. The method is based on Cu2+-catalyzed oxidation of ascorbic acid (AA) by O2 to depress the formation of colored gold nanoparticles (AuNPs). Specifically, HAuCl4 can be reduced by AA to produce AuNPs in situ. In the presence of target, CuO NPs-labeled antibodies were captured via the sandwich-type immunoreaction. After dissolving CuO nanoparticles with acid, the released Cu2+ catalyzed the oxidation of AA by O2, thus depressing the generation of AuNPs. To demonstrate the accuracy of the colorimetric assay, the released Cu2+ was further determined by a fluorescence probe. The colorimetric immunoassay shows a linear relationship for PSA detection in the range of 0.1~10 ng/mL. The detection limit of 0.05 ng/mL is comparable to that obtained by other CuO NPs-based methods. The high throughput, simplicity, and sensitivity of the proposed colorimetric immunoassay exhibited good applicability for assays of serum samples.
An epitope imprinted electrochemical sensor for highly sensitive and selective determination of prostate-specific antigen

Abstract:A simple method for highly selective and sensitive prostate-specific antigen (PSA) detection using a molecularly imprinted electrochemical sensor is presented. The sensor was developed through an epitope imprinted strategy combined with electrochemical measurement techniques. An epitope molecularly imprinted polymer (EMIP) film was constructed on a AuNPs-coated gold electrode surface through electropolymerization, utilizing the C-terminus epitope of PSA (KWIKDTIVANP) as the template molecular and o-phenylenediamine as the functional monomer. The characteristics of EMIP film were investigated by using a scanning electron microscope and electrochemical test methods, including electrochemical impedance spectroscopy and cyclic voltammetry. Key parameters such as electropolymerization cycles, elution and rebinding times, and the molar ratio of template molecular to functional monomer were systematically optimized. The sensor demonstrated a detection limit (LOD) of 0.31 fg/mL and exhibited an excellent linear response towards PSA concentration ranging from 1.0 fg/mL to 0.1 µg/mL. Furthermore, the EMIP sensor showed excellent selectivity against other biological macromolecules, such as bovine serum albumin, human serum albumin, alpha-fetoprotein, and carcinoembryonic antigen. With recoveries between 95.89 and 106.04% for PSA detection in human serums the EMIP/AuNPs/AuE electrochemical sensor showed great potential in real sample analysis.

Abstract:For the purpose of detecting prostate specific antigen (PSA), a ratiometric electrochemical aptasensor has been developed based on target-aptamer binding triggered isothermal strand displacement polymerase reaction (ISDPR). A ferrocene (Fc)-modified hairpin probe 2 (Fc-HP2) was self-assembled on the modified electrode surface. One terminus of hairpin probe 1 (HP1) bound to PSA and formed the HP1/PSA complex, and the other hybridized with Fc-HP2. Then, the S1 strand hybridized with the stem of Fc-HP2 and was extended with the aid of phi29 DNA polymerase (phi 29) and deoxyribonucleoside triphosphates (dNTPs), and the HP1/PSA complex could be displaced to trigger a new cycle of polymerization reaction. Finally, numerous double-stranded DNAs (dsDNAs) were produced for methylene blue (MB) intercalation. With the raising of PSA concentrations, the electrochemical responses of Fc (IFc) and MB (IMB) decreased and increased, respectively. When IMB/IFc was used as the response signal for quantitative determination of PSA, the proposed strategy possessed a wide linear range (10 pg mL−1–100 ng mL−1) and a low detection limit of 3.2 pg mL−1. Moreover, the proposed aptasensor displayed good performance for PSA determination in spiked human serum samples, showing great application prospects in bioanalysis.

Abstract:In the present work, a label-free prostate specific antigen (PSA) electrochemical immunosnesor was constructed based on Au nanoparticles/MoS2-graphene aerogels nanocomposite (AuNPs/MoS2-GAs) as sensing platform. MoS2-GAs composite with porous network structure provided large specific area for the immobilization of AuNPs, which further improved the loading of PSA antibodies. In addition, the AuNPs/MoS2-GAs composite has good conductivity, which could accelerate the electron transport of the electrode interface. Based on these advantages of AuNPs/MoS2-GAs material, the prepared immunosensor exhibited wide linearity in the range of 0.00001–50 ng mL−1 and a low detection limit of 0.003 pg mL−1. The immunosensor also exhibited acceptable stability, good selectivity, and reproducibility. Moreover, when applied to analyze PSA in human serum, the immunosensor obtained good recovervries, indicating the method shows a promising application for early diagnosis of PSA.

Abstract:An ultrasensitive luminol electrochemiluminescence (ECL) immunosensor was constructed for the detection of prostate specific antigen (PSA) using glucose oxidase-decorated hemin-graphene oxide-gold nanoflowers ternary nanocomposites as probes. Graphene oxide was first modified with hemin and then with gold nanoflowers through an in situ growth method, which has significantly boosted the catalytic activity of this graphene oxide-based peroxidase mimetics. The biocatalytical activity of this ECL immunosensor was thoroughly investigated to achieve selective recognition of the analyte molecules (PSA) by specific binding between antigens and antibodies. The limit of detection was calculated to be 0.32 pg mL-1 with a signal-to-noise ratio of 3. A broad linear range from 7.5 × 10-4 to 2.5 ng mL-1 was obtained. Such step-by-step assembled biosensor showed controlled nanostructure and exhibited promising application in analysis of human serum samples with a recovery range of 90.6-111.8% and a RSD range of 3.9-5.5%.
Gold interdigitated triple-microelectrodes for label-free prognosticative aptasensing of prostate cancer biomarker in serum

Abstract:A simple, single-masked gold interdigitated triple-microelectrodes biosensor is presented by taking the advantage of an effective self-assembled monolayer (SAM) using an amino-silanization technique for the early detection of a prostate cancer's biomarker, the prostate-specific antigen (PSA). Unlike most interdigitated electrode biosensors, biorecognition happens in between the interdigitated electrodes, which enhances the sensitivity and limit of detection of the sensor. Using the Faradaic mode electrochemical impedance spectroscopy (EIS) technique to quantify the PSA antigen, the developed sensing platform demonstrates a logarithmic detection of PSA ranging from 0.5 ng/ml to 5000 ng/ml, an estimated LOD down to 0.51 ng/ml in the serum, and a good sensor's reproducibility. The sensor's detection range covers the clinical threshold value at 4 ng/ml and the crucial diagnosis 'grey zone' of 4-10 ng/ml of PSA in serum for an accurate cancer diagnosis. The selectivity test revealed an excellent discrimination of other competing proteins, with a recorded detection signals at 5 ng/ml PSA as high as 7-fold increase versus the human serum albumin (HSA) and 8-fold increase versus the human glandular kallikrein 2 (hK2). The stability test showed an acceptable stability of the aptasensor recorded at six (6) days before the detection signal started degrading below 10% of the peak detection value. The developed sensing scheme is proven to exhibit a great potential as a portable prostate cancer biosensor, also as a universal platform for bio-molecular sensing with the versatility to implement nanoparticles and other surface chemistry for various applications.

Abstract:A ratiometric electrochemiluminescence (ECL) aptamer-based sensing platform was fabricated for prostate-specific antigen (PSA) determination. Activated CdS nanocrystals/multi-walled carbon nanotubes (CdS/MCNTs) and luminol-Pt/PAMAM nanocomposites (L-Pt/PAMAM NCs) were synthesized and used as cathodic and anodic ECL emitters, respectively. Amino group-modified aptamers were assembled on carboxylated magnetic beads, followed by hybridization with probe DNA functionalized L-Pt/PAMAM NCs. In the presence of PSA, the aptamer would bind specifically to the target PSA, thereby releasing L-Pt/PAMAM NCs. After magnetic separation, the separated L-Pt/PAMAM NCs would hybridize with capture DNA on CdS/MCNTs coated on glassy carbon electrode. This binding would lead to a decrease in cathodic ECL signal of CdS/MCNTs, due to the efficient energy transfer from CdS/MCNTs to L-Pt/PAMAM NCs. Meanwhile, L-Pt/PAMAM brought the anodic ECL signal from luminol. With the increase of PSA concentration, the ECL emission from luminol increased and the ECL emission from CdS/MCNTs decreased. The ratio of ECL intensity of luminol at 0.55 V and CdS/MCNTs at - 1.25 V could be used to quantify the concentration of PSA. This method enables sensitive and reliable detection of PSA over a wide range from 0.05 to 200 ng mL-1, and the detection limit is 0.02 ng mL-1.

Abstract:Prostatic cancer threatens the lives and health of numerous male patients all over the world. As a single-chain glycoprotein, prostate specific antigen (PSA) can be used to perform prostate cancer screening. At present, there are still problems such as time-consuming and low sensitivity in hospital detection. At the same time, electrochemical immunosensor technology has received a lot of attention due to its benefits, including high sensitivity, good selectivity, low usage and cost, and so on. Electrocatalysts are typically used to build electrochemical immunosensors for signal amplification in the recent years to realize sensitive detection of cancer biomarkers such as PSA. In recent years, three-dimensional nanomaterials have been widely used in the study of electrocatalysts, but the study of the sensitivity for PSA detection is still very scarce. The present work reported the application of a 3D graphene aerogel(GA)/AuNPs/Nafion electrochemical platform for highly sensitive PSA quantification. The electrochemical immunosensor has been designed by immobilizing aptamers on the screen-printed electrode (SPE) surface modified by GA/AuNPs/Nafion. Scanning electron microscopy (SEM), Raman spectra, and electrochemical characterization demonstrated the successful combination of GA/AuNPs/Nafion/SPE. Based on electrochemical impedance spectroscopy (EIS), the proposed immunosensor has demonstrated excellent electrochemical performance, exhibiting sensitive and stable responses for electrochemical detection of PSA ranging from 0.05 to 50 ng∙mL −1 with a limit of detection (LOD) of 0.0306 ng∙mL −1 . This study has great potential application in PSA detection and provides a valuable reference for point-of-care testing (POCT) of prostate cancer.

Abstract:In this paper, A label-free electrochemical immunosensor for sensitive detection of prostate antigen (PSA) was developed based on NiFe PBA/AuNPs composite. The prostate antigen antibody was immobilized and the immunosensor was constructed by using a glassy carbon electrode modified with a nanocomposite consisting of nickel-iron Prussian blue analog (NiFe PBA) and gold nanoparticles (AuNPs). The good biological affinity of AuNPs for biomolecules, as well as the porous nanostructure and regular shape of NiFe PBA, the NiFe PBA/AuNPs nanocomposites significantly improve the electron transport rate, while achieving the excellent performance of the sensor. Due to the interaction between the antibody and the antigen on the modified electrode, the current signal of the NiFe PBA itself is reduced due to the redox changes in Fe2 + and Fe3 +, which can be determined by differential pulse voltammetry (DPV). Therefore, the monitoring of prostate antigen detection is realized. Under optimal experimental conditions, the immunosensor exhibited excellent detection performance with a dynamic response range from 0.5 pg/mL to 1000 pg/mL for PSA concentration and a detection limit of 0.23 pg/mL (S/N = 3). In addition, the PSA aptasensor has good selectivity, high stability, and satisfactory reproducibility and has broad potential in clinical research and diagnostic applications.

Abstract:This study describes a novel dual-mode immunosensor that combines electrochemical (EC) and surface-enhanced Raman scattering (SERS) techniques for the detection of prostate-specific antigen (PSA), a biomarker associated with prostate cancer. The sensor consists of a nanocomposite of gold nanoparticles (AuNPs) deposited on two-dimensional (2D) molybdenum disulfide (Au@MoS2) modified on a working carbon electrode of a screen-printed electrode (SPE). Subsequently, the primary antibody (Ab1) is immobilized on the modified electrode, creating Ab1/Au@MoS2/SPE for specific recognition of the target PSA. In parallel, AuNPs are conjugated with a secondary antibody (Ab2) and a probe molecule, 3,3',5,5'-tetramethylbenzidine (TMB), leading nanotags (TMB/Ab2/AuNPs) formation exhibiting strong SERS and EC responses. Upon the presence of the target, sandwich immunocomplexes can be formed through antigen-antibody interactions (Ab1-PSA-Ab2). The differential pulse voltammetry (DPV) technique is employed for EC detection mode, while a handheld Raman spectrometer with a 785 nm excitation laser is utilized to collect SERS signals. The developed system demonstrates excellent selectivity and sensitivity, with low limits of detection (LODs) of 3.58 pg mL-1 and 4.83 pg mL-1 for EC and SERS sensing, respectively. Importantly, the dual-mode immunosensor proves effective quantifying PSA protein in human serum samples with good recovery. Given its high sensitivity and proficiency in analyzing biological samples, this proposed immunosensor holds promise as an alternative tool for the early diagnosis of cancers.

Abstract:A novel sandwich-type electrochemical aptasensor based on supramolecularly immobilized affinity bioreceptor was prepared via host–guest interactions. This method utilizes an adamantane-modified, target-responsive hairpin DNA aptamer as a capture molecular receptor, along with a perthiolated β-cyclodextrin (CD) covalently attached to a gold-modified electrode surface as the transduction element. The proposed sensing strategy employed an enzyme-modified aptamer as the signalling element to develop a sandwich-type aptasensor for detecting prostate-specific antigen (PSA). To achieve this, screen-printed carbon electrodes (SPCEs) with electrodeposited reduced graphene oxide (RGO) and gold nanoferns (AuNFs) were modified with the CD derivative to subsequently anchor the adamantane-modified anti-PSA aptamer via supramolecular associations. The sensing mechanism involves the affinity recognition of PSA molecules on the aptamer-enriched electrode surface, followed by the binding of an anti-PSA aptamer–horseradish peroxidase complex as a labelling element. This sandwich-type arrangement produces an analytical signal upon the addition of H2O2 and hydroquinone as enzyme substrates. The aptasensor successfully detected the biomarker within a concentration range of 0.5 ng/mL to 50 ng/mL, exhibiting high selectivity and a detection limit of 0.11 ng/mL in PBS.
