Poly(phosphonium)-Functionalized Double-Armed β-CD Antimicrobial Material via RAFT

Abstract:Bacterial infections have emerged as the primary adversary, causing severe health complications and incalculable economic losses. To tackle this issue, it is imperative to develop novel and highly effective antimicrobial agents. In this study, a series of double-armed antibacterial polymers (CD-QPS) were synthesized via Steglich esterification and reversible addition–fragmentation chain transfer polymerization (RAFT) polymerization. Notably, this represents a significant breakthrough in the field of antibacterial polymers derived from the quaternary phosphonium salt ionic liquid (QPSIL) and β-cyclodextrin (β-CD) via RAFT polymerization. Leveraging the controllable polymerization degree offered by RAFT and the functional group density resulting from its three-dimensional topological structure, CD-QPS exhibited exceptional antibacterial activity against Escherichia coli and Staphylococcus aureus with bacteriostatic rates reaching 100 and 99.9%, respectively. What is even more exciting is that CD-QPS demonstrated minimum inhibitory concentrations (MICs) as low as 20 μg mL–1 for E. coli and 40 μg mL–1 for S. aureus, surpassing those reported for other antibacterial materials as well as the standard drug ampicillin (32/62.5 μg mL–1). Furthermore, preservation tests conducted on oranges confirmed that CD-QPS exerted long-term antibacterial effects for an astonishing duration of 14 days. Additionally, the cytotoxicity test verified good biocompatibility of CD-QPS. In brief, we not only provide a new strategy for constructing double-armed polymers known as CD-QPS but also introduce a novel antimicrobial agent with high efficiency and long-term performance.
