Abstract:Tea saponins, found in Camellia plants, are natural non-ionic surfactants that offer obvious beneficial effects in soil remediation. Most tea saponins are extracted from the Camellia oleifera seed meal, with the leaves and flowers of Camellia sinensis as potential sources. Water extraction and ultrasoundassisted water extraction combined with acetone precipitation are recommended for the industrial extraction and purification of tea saponins, considering multiple factors. The detailed physical, chemical and biochemical properties of tea saponins need to be clarified, especially whether tea saponins with slightly different structures from distinct sources have different soil remediation properties. Applied in leaching remediation, phytoremediation and microbial remediation, tea saponins desorb heavy metals from contaminated soil as well as enhancing their bioavailability. Tea saponins improve the accumulation of pollutants by hyperaccumulators as well as the degradation of organic pollutants by microorganisms. Currently the mechanisms of tea saponins are not clear, although they are proven to be effective natural surfactants for the remediation of contaminated soils. This review enriches our understanding of tea saponins from various aspects and encourages further studies of industrial extraction and purification, and the field remediation mechanisms of tea saponins, making better use of Camellia plants and contributing to environmental protection.

Abstract:The residue of Camellia seeds after oil extraction contains many bioactive ingredients, including tea saponin. Tea saponin has many pharmacological effects and is an excellent nonionic surfactant. The development of natural surfactants has become a hot topic in food research. This review gathers the applications of tea saponin as a surfactant in food. It focuses on the application of tea saponin in emulsions, delivery systems, extraction and fermentation, as well as the challenges and development prospects in food applications. Tea saponin shows great potential as a surfactant in food applications, which can replace some synthetic surfactants. The full utilization of tea saponin improves the comprehensive utilization value of Camellia seed residue, contributes to the sustainable development of Camellia industry and avoids resource waste.

Development of a Rapid and Simple Method for Preparing Tea-Leaf Saponins and Investigation on Their Surface Tension Differences Compared with Tea-Seed Saponins

Abstract:The relative overcapacity in China's tea-leaf production and the potential application of tea-leaf saponins in soil remediation encouraged in-depth developments and comprehensive utilizations of tea-leaf resources. Through variables optimizations using Box–Behnken designs for ultrasonic power, temperature as well as ultrasonic treatment time in ultrasonic-assisted water extraction and single-variable experiments for acetone-extraction solution ratio in acetone precipitation, a rapid and simple method was developed for preparing tea-leaf saponins. Tea-leaf saponins with the concentration of 3.832 ± 0.055 mg/mL and the purity of 76.5% ± 1.13% were acquired under the optimal values of 78 w, 60 °C, 20 min and 0.1 ratio of acetone-extraction solution. Both Fourier transform-infrared (FT-IR) spectra and ultraviolet (UV) spectra revealed slight composition differences between tea-leaf saponins and tea-seed saponins, while these differences were not reflected in the critical micelle concentration (CMC) and the surface tension of tea-leaf saponins and tea-seed saponins, indicating there was no need to distinguish them at the CMC. Further research attention on where tea-leaf saponins were in low concentrations is deserved to discover whether they had differences in comparison with tea-seed saponins, which was beneficial to apply them in the phytoremediation of contaminated soils.

Abstract:Background: Tea seed (TS), as the mature fruit and growth source of tea plant, has been characterized with appealing bioactive substances, remarkable health benefits and diverse application prospects. As one of the most significant products of TS, tea seed oil (TSO) has gained recognition as the "oriental olive oil". Tea seed meal (TSM) and tea seed shell (TSS) continue to be underutilized and are usually considered as low-value by-products. Nevertheless, current exploration of the underlying mechanisms is inadequate, and investigations concerning deep processing and potential prospects still remain at their infancy. Consequently, there exists a gap in the comprehensive aspects of TS utilization and the related advancements.Scope and approach: In this review, we have summarized the latest research progresses on TS, specifically focused on the bioactive substances, pharmacological activities, manufacturing process and potential applications. Furthermore, we have also elucidated the current challenges in existing studies, and proposed the research orientations and potential prospects of TS.Key findings and conclusions: TS contain abundant bioactive substances, and exhibit outstanding performance in antioxidant, anti-tumor, anti-obesity, anti-microbe, gastric protection, and so on. These properties contribute to its application potentials as crucial materials in industry, agriculture and ecology, as well as new perspective in drug development and clinical translation. Future researches should concentrate on the specific active in-gredients, intricate structure-activity relationships, bioactive mechanisms, undiscovered health benefits and manufacturing process, which would facilitate the transformation of waste resources into valuable treasures, thus fostering sustainable development and socio-economic growth.

Simultaneous Removal of Polycyclic Aromatic Hydrocarbons and Heavy Metal from Co-Contaminated Soils by Saponin, a Plant-Derived Biosurfactant

Abstract:Biosurfactants have great advantages in the remediation of organic contaminated soils as their environmental friendly nature. The performance of saponin,a plant-derived biosurfactant,for the removal of polycyclic aromatic hydrocatbons(PAHs) and heavy metal from co-contaminated soils were studied. The results indicated that saponin can greatly enhance the solubilization of phenanthrene and pyrene in solution by the partition of them into saponin micelle and then the removal of phenanthrene and pyrene from PAHs-contaminated soil. The maximum removal rates of 84.1% and 81.4% were obtained for phenanthrene and pyrene from PAHs-contaminated soil,respectively. Saponin could form complex compound with heavy metals by the external carboxyl groups,which greatly enhanced the removal of zinc and cadmium from heavy metal contaminated soil and the removal rates of 93.0% and 79.6% were obtained for zinc and cadmium with a sapoin concentration of 0.4%,respectively. For the co-contaminated soil with PAHs and heavy metals,87.6% of phenanthrene,83.5% of pyrene,92.3% of zinc and 78.6% of cadmium were simultaneously removed by saponin at a concentration of 0.4%. The removal rates of zinc and cadmium from co-contaminated soil did not show obvious difference with the presence of PAHs,but the addition of heavy metals slightly enhanced the removal of phenanthrne and pyrene from the co-contaminated soil. Thus,saponin had the potential capability to simultaneously remove PAHs and heavy metal from the co-contaminated soils.

Abstract:Tea saponins have high surface-active and biological activities and are widely used in chemicals, food, pharmaceuticals, and pesticides. Tea saponins are usually extracted using ethanol or water, but both methods have their disadvantages, including a negative impact on the environment, high energy consumption, and low purity. In this study, we explored an effective process for extracting tea saponins from tea meal using deep eutectic solvents combined with ultrasonic extraction and enzymatic techniques. The experimental results showed that a high extraction efficiency of 20.93 ± 0.48% could be achieved in 20 min using an ultrasonic power of 40% and a binary DES consisting of betaine and ethylene glycol (with a molar ratio of 1:3) at a material–liquid ratio of 1:35 and that the purity of the tea saponins after purification by a large-pore adsorption resin reached 95.94%, which was higher than that of commercially available standard tea saponin samples. In addition, the extracted tea saponins were evaluated for their antioxidant and bacteriostatic activities using chemical and biological methods; the results showed that the tea saponins extracted using these methods possessed antioxidant properties and displayed significant antibacterial activity. Therefore, the present study developed a method for using deep eutectic solvents as an environmentally friendly technological solution for obtaining high-purity tea saponins from tea meal oil. This is expected to replace the current organic solvent and water extraction process and has great potential for industrial development and a number of possible applications.

Abstract:Tea saponins from Camellia oleifera Abel. seed pomace are new sources of commercial saponins. This study established an eco-friendly and efficient extraction method for tea saponins from C. oleifera seed pomace. A ternary deep eutectic solvent (DES) composed of l-proline, glycerol and sucrose (4:10:1 in molar ratio, abbreviated as PGS-5) achieved the highest extraction yield of tea saponins among all screened DESs. A maximum extraction yield of 23.22 ± 0.28% was obtained using PGS-5 under the optimized extraction time, DES concentration and liquid-solid ratio. Through ultraviolet, Fourier transform infrared spectroscopy and ultrahigh-performance liquid chromatography-Q Exactive HF mass spectroscopy, as well as analyses of antioxidant and antimicrobial activities, it was determined that extracted saponins did not altered during processing. Therefore, PGS-5 can serve as a solvent to obtain stable and beneficial tea saponins from C. oleifera seed pomace.

Abstract:Traditional organic solvent extractions of tea saponins have many drawbacks. This study aimed to establish an environment-friendly and efficient technology based on deep eutectic solvents (DESs) to extract tea saponins from Camellia oleifera seed meal. The solvent consisting of choline chloride and methylurea was screened as optimal DES. Under the optimal extraction conditions obtained by response surface methodology, the extraction yield of tea saponins reached 94.36 mg/g, which increased by 27% compared with ethanol extraction, while the extraction time was reduced by 50%. Analysis of UV, FT-IR, and UPLC-Q/TOF-MS indicated tea saponins did not alter during DES extraction. Surface activity and emulsification evaluation showed that extracted tea saponins could reduce interfacial tension at the oil-water interface with excellent foamability and foam stability, and they could form nanoemulsions (d32 < 200 nm) with excellent stability. This study provides a suitable approach for the efficient extraction of tea saponins.

Enhanced Solubilization of Pyrene by Saponin,a Plant-Derived Biosurfactant

Abstract:Biosurfactants have great advantages in the remediation of organic contaminated soils because of their environmentally friendly nature. The solubilization of pyrene by saponin,a plant-derived biosurfactant,was studied and the results indicated that saponin can greatly enhance the solubilization of pyrene in solution and the weight solubilization ratio (MSR) of saponin to pyrene was greater than those of some representative nonionic surfactants. The enhanced solubilization of pyrene by saponin showed strong dependence on the solution pH and ionic strength. It decreased as the solution pH increased from 4.0 to 8.0,but increased as the NaCl concentration increased from 0.01 mol·L-1 to 1.0 mol·L-1,which can be attributed to the increased or decreased critical micelle concentration (CMC) of saponin solution with the changing pH or ionic strength,respectively. Saponin was more effective in enhancing pyrene solubilization at low pH and high ionic strength. In addition,heavy metal ions (Zn2+ and Cd2+) can also enhance the solubilization of pyrene by saponin.

Abstract:Natural preservatives are substances that are secreted by organisms or present in the body and have bacteriostatic effects, so they are not toxic to humans. Natural preservatives have become a research focus because of their high safety and strong antibacterial effects. Some commodities in the market are added with tea saponins, such as a brand of foaming cleanser, shampoo, and toothpaste rich in tea saponins. In this study, A high concentration of tea saponin was obtained through extraction and purification from Camellia oleifera shells, and its antibacterial activity for the two tested bacteria was determined in vitro. The results revealed that AB-8 macroporous adsorption resin was used in combination with Sephadex G-15 to obtain tea saponin with 82.5 % purity. The optimal purification process conditions were: the loading quantity of sample is 1 bed volume (BV), the loading solution is 25 mg/mL, the loading solution PH is 5, the loading flow rate is 3 BV/h, the elution volume is 4 BV, the elution flow rate is 3 BV/h, the eluent ethanol concentration is 95 %, and the eluent pH is 9. Tea saponin had good bacteriostatic effect on Escherichia coli and Staphylococcus aureus with its minimum inhibitory concentration of 1 and 0.5 mg/mL, respectively. In addition, its minimum bactericidal concentration was 4 mg/mL. Tea saponin could change membrane permeability, destroy cell membrane structure, and inhibit bacterial growth. This study provides basic guidance for the application of tea saponin in the development of natural antibacterial agents, is conducive to the high-value utilization of tea saponin, provides a reference for the economical of camellia tea waste, and plays a benefit in increasing agricultural income.

Enhanced Soil Washing of Phenanthrene by A Plant-Derived Natural Biosurfactant, Sapindus Saponin

Abstract:Biosurfactants have great advantages for using in environmental remediation as their environmental friendly nature. In this study, a plant-derived natural biosurfactant of Sapindus saponin was extracted from the fruit pericarps of Sapindus mukorossi (Chinese soapberry), and the properties of this biosurfactant in enhancing washing for phenanthrene contaminated soil were investigated and compared with those of Tween 80, a representative synthetic nonionic surfactant. Sapindus saponin showed a linear solubilization enhancement for phenanthrene and the weight solubilization ratio was 0.0312 and about one third of that of Tween 80. However, Sapindus saponin can effectively remove phenanthrene from contaminated soil with a maximum removal percentage of about 87.4%, which was only slightly less than that of Tween 80 (about 91.5%) because the sorption of Sapindus saponin onto soil was much less than that of Tween 80. The solubilization enhancement of Sapindus saponin for phenanthrne and the sorption of Sapindus saponin onto soil both decreased with increasing solution pH, the net results of which resulted in that the maximum washing efficiency of Sapindus saponin for phenanthrene contaminated soil was obtained at pH around 6.0. Sapindus saponin solutions can be recovered for their reusage after soil washing by using organo-bentonite to remove phenanthrene from the washed surfactant solutions with an appropriate dosage. Sapindus saponin showed a favorable performance in enhancing soil washing and should have the potential application in soil washing technology for the remediation of organic contaminated soils and groundwater. Positive Effects and Optimal Ranges of Tea Saponins on Phytoremediation of Cadmium-Contaminated Soil

Abstract:Confirming positive effects and offering optimal ranges of tea saponins on improving the efficiency of phytoremediation on cadmium is a prerequisite for applying tea saponins in field remediation. Existing studies qualitatively tested the feasibility of tea saponins on promoting the absorption of cadmium by hyperaccumulators in pots experiments, while this study investigated the effects of tea saponins on increasing the proportion of cadmium available fraction in contaminated soil quantitatively and confirmed tea saponins promoted the absorption by Portulaca oleracea in cadmium-contaminated water by independent soil experiments and hydroponic experiments. The results showed that for acquiring a higher proportion of cadmium available fraction, the concentration of tea saponins was negatively correlated with the concentration of cadmium contained in the soil, and the optimal treatment time of tea saponins was between 3–9 days depending on the cadmium concentration in contaminated soil. Using tea saponins could enhance the absorption of cadmium by Portulaca oleracea in a relatively short time to decrease the concentration of cadmium left in the contaminated water. The above findings help to deepen the understanding of tea saponins’ effects and use ranges on phytoremediation of cadmium both in soil and water and conduce studies on phytoremediation of other heavy-metal-contaminated soil and water with the help of tea saponins.

Abstract:Due to the benign features of biosurfactants such as their low toxicity and desirable biodegradability, there has been growing interest in developing these materials for environmental remediation applications. In this work, the relationship between the temperature and the solubilization of tea saponin was investigated via critical micelle concentration (CMC), particle size, and thermodynamics measurements. In addition, the aqueous solubility enhancements of naphthalene provided by micellar solutions of biosurfactants (tea saponin) and a synthetic surfactant (Tween 80) were measured and compared. Results indicated that the solubilization effect of tea saponin increased as the temperature was elevated from 298 to 313 K, which thus indicated that this effect had a strong dependence on temperature. An increase in the temperature also led to a lower CMC value, an increase of the particle diameter, and an enhancement of the molecular activity, which thus led to the enhanced solubilization effect of tea saponin. Moreover, the solubilization effect of tea saponin was better than that provided by Tween 80. The results confirmed the potential applicability of tea saponin for enhanced remediation of naphthalene.

Preparation and performance evaluation of environmental-friendly, temperature-resistant and salt-resistant tea saponin foaming agent

Abstract:Camellia oleifera produced many wastes after being squeezed oil and the wastes were made into tea seed cake that contains rich tea saponin. A green tea saponin foam agent was developed to improve the recovery of crude oil. In this study, ethanol method was used to extract and isolate tea saponins from tea seed cake. Response surface methodology was used to optimize the tea saponin extraction process and improve production. The effect of ethanol concentration, material-to-liquid ratio, extraction temperature, and extraction time were used as independent variables. The results showed that highest yield of tea saponin was 21.55% under optimal experimental conditions of ethanol concentration (76.21%), material-to-liquid ratio (1:12.91 g/mL), extraction temperature (75.92°C), and extraction time (3.13 h). The foam properties showed that tea saponin foam had good foam stabilization and resistance to pH, temperatures, and salinity. About 0.1 wt% tea saponin foam indicated good foam performance at pH of 5–6.5, Na + (10,000–20,000 mg/L), Ca 2+ (400–800 mg/L) and Mg 2+ (400–800 mg/L), long-term temperature (120°C, 96 h) still has good foam performances. Subsequently, the experiments were tested the plugging ability by researching the differential pressure of the foam agents at different saponin concentrations. Experimental results illustrated great plugging effects when the pressure reached 1.54 MPa at 0.2 wt% saponin. Saponin foam could adapt to the formation environment and had good tolerance to temperature, salinity, and acidity. In addition, tea saponin had a good application prospect in the field of EOR. Abstract:Batch experiments were conducted to evaluate the performance of saponin, a plant-derived biosurfactant, for simultaneously removing phenanthrene and cadmium from the combined contaminated soils. Results showed that phenanthrene was desorbed from the contaminated soils by saponin with the partition of phenanthrene into surfactant micelle, meanwhile cadmium was effectively removed from the contaminated soils by the complexation of cadmium with the external carboxyl groups of saponin micelle. The efficiencies of saponin for the removal of phenanthrene and cadmium from the contaminated soils were greater than that of Triton X100 and citric acid, respectively. At concentration of 3750 mg/L, saponin has a removal rate of 87.7% and 76.2% of cadmium and phenanthrene, respectively, from the combined contaminated soil. The removals of cadmium and phenanthrene from the soils were not obviously constrained each other. Abstract:HYPOTHESIS: This study compared the interfacial and emulsification properties of tea saponins, quillaja saponins, and Tween 80. We hypothesized that tea saponins are an effective and sustainable source of plant-based emulsifiers that could replace synthetic or animal-based emulsifiers in many commercial applications.EXPERIMENTS: Interfacial tension measurements were used to characterize the behavior of the three surfactants at an oil-water interface. The emulsifying properties of the surfactants were determined by preparing oil-in-water emulsions containing 10 wt% medium chain triglycerides (MCT) and varying surfactant levels (0.1-2 wt%) using high-pressure homogenization (pH 7). The impact of surfactant type on emulsion formation and stability was determined by measuring particle size, zeta-potential, microstructure, and creaming stability.FINDINGS: The tea saponins were capable of producing nano-scale droplets (d32 < 200 nm) at low surfactant-to-oil ratios (SOR < 0.1). Emulsions containing tea saponins remained stable to droplet aggregation when exposed to various temperatures (30-90 °C), salt levels (0-200 mM NaCl), and pH values (3-9). However, droplet flocculation and/or coalescence occurred under highly acidic (pH 2) and high ionic strength (300-500 mM NaCl) conditions. Tea saponin-coated oil droplets appeared to be mainly stabilized by a combination of electrostatic and steric repulsion. The tea saponins behaved similarly or better than the other two emulsifiers under most conditions. These results suggest that tea saponins are effective plant-based surfactants that may have applications in commercial products.

Abstract:Camellia seeds have been traditionally used as oil raw materials in Asia, and are known for a wide spectrum of applications. Oleanane-type triterpene saponins are the major specialised metabolites in Camellia seeds, and more than seventy saponins have been isolated and characterized. These natural compounds have caught much attention due to their various biological and pharmacological activities, including modulation of gastrointestinal system, anti-cancer, anti-inflammation, anti-microorganism, antioxidation, neuroprotection, hypolipidemic effects, foaming and detergence, as well as helping the accumulation of pollutants by plants. These compounds have a promising application in medicine, agriculture, industry and environmental protection. The present paper summarized the information from current publications on Camellia seed saponins, with a focus on the advances made in chemical structures, determination methods, bioactivities and toxicity. We hope this article will stimulate further investigations on these compounds.
