Abstract:Optical imaging plays a central role in biology and medicine but is hindered by light scattering in live tissue. We report the counterintuitive observation that strongly absorbing molecules can achieve optical transparency in live animals. We explored the physics behind this observation and found that when strongly absorbing molecules dissolve in water, they can modify the refractive index of the aqueous medium through the Kramers-Kronig relations to match that of high-index tissue components such as lipids. We have demonstrated that our straightforward approach can reversibly render a live mouse body transparent to allow visualization of a wide range of deep-seated structures and activities. This work suggests that the search for high-performance optical clearing agents should focus on strongly absorbing molecules.

Abstract:In vivo imaging using shortwave infrared light (SWIR, 1000-2000 nm) benefits from deeper penetration depths, decreased background autofluorescence, and high resolution. However, the development of biocompatible contrast agents for these low energy wavelengths has significant challenges. While there have been significant advances in SWIR chromophore scaffolds over the past 5 years, a major barrier for widespread utility of SWIR small molecule fluorophores is their hydrophobicity and tendency to form non-emissive aggregates. Here, we report a platform for generating a panel of soluble and functional dyes for SWIR imaging by late-stage functionalization of a fluorophore intermediate via click chemistry. The resulting fluorophores with sulfonate, ammonium or zwitterion functionalities are all water soluble with bright SWIR fluorescence in serum, allowing for fast imaging in mice. Specifically, the sulfonate-carrying derivative enables clear video-rate imaging of vasculature with as little as 0.05 nmol injected dye, and the ammonium-modified dye shows strong retention in cells that enables tracking of xenograft tumor growth. We further showcase the versatility of this design by incorporating phosphonate functionalities for imaging of bone in awake and moving mice. This modular design of functional SWIR fluorophores in water provides insights for facile derivatization of existing fluorophores to introduce solubility and bioactivity towards bioimaging applications.

Abstract:The field of tissue clearing has experienced rapid advancements in the past decades. Tissue clearing techniques primarily rely on physical or chemical approaches to match the refractive indices of cells and their surrounding medium, thereby reducing light scattering and rendering the tissue optically transparent. By doing so, it becomes possible to utilize fluorescence imaging and 3D reconstruction techniques to observe and analyze the contents and structures of tissues at a cellular level. However, effective research on in vivo tissue clearing remains limited due to the involvement of toxic substances and the removal of water or lipids. A recent article in Science reports a technique that achieves tissue transparency in live animals with absorbing molecules, marking a significant advancement in the field of in vivo tissue clearing. Although this work has great potential for imaging and light-based therapeutics, some labs found it difficult to reproduce the reported results. In this study, we report the successful reproduction of the findings, provide experimental details that we identified as key to reproducibility, present a new transparency effect observed in the back of live mice, which was not shown in the original paper, and offer insights into the future development and applications of in vivo clearing reagents.

Abstract:Dyeing behaviors of polylatic acid blended fabric assisted with ultrasonic wave were studied,the results were compared with those of conventional dyeing.Effects of ultrasonic wave on dye liquor and fabric were analyzed by scanning electron microscopy(SEM),X-ray diffraction(XRD)and laser particle size analyzer.The results showed that ultrasonic treatment could break the micelles of disperse dyes,therefore,dye uptake and K/S value could be increased.After ultrasonic treatment,the fabric had no obvious etching on surface nor changes in α crystalline form,and only a little decrease in strength and intensity of diffraction.

Abstract:Spontaneously blinking fluorophores permit the detection and localization of individual molecules without reducing buffers or caging groups, thus simplifying single-molecule localization microscopy (SMLM). The intrinsic blinking properties of such dyes are dictated by molecular structure and modulated by environment, which can limit utility. We report a series of tuned spontaneously blinking dyes with duty cycles that span two orders of magnitude, allowing facile SMLM in cells and dense biomolecular structures.

Abstract:In this study, rare earth doped infrared (IR) to visible, up-converting particles along-with efficient ultraviolet (UV) to visible fluorescent molecules were imbedded in proteins and were used as a mean for increasing the human vision range to infrared and ultraviolet wavelengths. Stilbene-420 fluorescent molecules which convert near UV light to blue light, were chosen for the strong overlap of their fluorescence emission with the sensitivity region of blue cone cells and for the ability of blue light to increase the regeneration of bleached visual pigments. Our data show that the up-conversion efficiency of the IR up-converting particles and blue fluorescing molecules efficiencies remained unchanged when these material are imbedded in proteins compared to their efficiencies in water solutions. Addition of up-converting particles to rod visual cells resulted in the bleaching of the visual pigments, rods, when irradiated with infrared light (980 nm) whereas no bleaching was observed, under the same conditions, without the presence of up-converting particles. This suggests that the up-converted green light induced visual process in the rod visual pigments. In addition, we describe the design and present data of a novel optical device, which can be used as eye glasses, utilizing up-converting particles that allows the wearer to see rather intense infrared light.

Abstract:Developing chemical tools to detect and influence biological processes is a cornerstone of chemical biology. Here we combine two tools which rely on orthogonality- perfluorocarbons and multiplexed shortwave infrared (SWIR) fluorescence imaging- to visualize nanoemulsions in real time in living mice. Drawing inspiration from fluorous and SWIR fluorophore development, we prepared two SWIR-emissive, fluorous-soluble chromenylium polymethine dyes. These are the most red-shifted fluorous fluorophores- "fluorofluorophores"-to date. After characterizing the dyes, their utility was demonstrated by tracking perfluorocarbon nanoemulsion biodistribution in vivo. Using an excitation-multiplexed approach to image two variables simultaneously, we gained insight into the importance of size and surfactant identity on biodistribution.

Abstract:Wool pretreated with dimethyl sulfoxide (DMSO) was characterized and its dyeing behavior in non-aqueous green solvents was investigated. Reactive dyeing of wool in deep shades is challenging because the mandatory alkaline aftertreatment to match the fastness of mordant dyes inevitably causes damage to wool keratin. The current study showed that the colorfastness-integrity dilemma could be solved by replacing water with organic solvents as the dyeing medium. Covalent fixation is predominantly favored in solvent dyeing so that excellent colorfastness is achievable at any given shade without alkali aftertreatment. Compared with aqueous dyeing, solvent dyeing was found to give 30% higher covalent fixation on average, which translated to better colorfastness (0.5–1 grade) at higher color depth (Δ K/ S = 1.3–4.9). In addition, scanning electron microscopy, X-ray diffraction and attenuated total reflection-infrared spectroscopy results indicated that DMSO pretreatment induced morphological and structural changes of wool in favor of dyeability. The findings are crucial to the phasing-out of metal-containing dyes for eco-friendly wool dyeing.

Abstract:Acid mordant dye is one of the important dyes for wool dyeing,but waste-water produced by dyeing process generally contains ions of heavy metal,which causes pollution,studying on dyeing abilities and dyeing processes is beneficial for developing environment-friendly dyeing.In this paper,with two novel naphthoquinone pigments synthesized by our research team,we investigated the influence of concentration,pH value,temperature and time on dye-uptake of wool fabric,and adapted environment-friendly mordant such as Fe2+,Fe3+ and Al3+ for sulfonated pigments.It was concluded that sulfonated naphthoquinone can be used for wool dyeing,Fe3+ had the most obvious effect on mordant dyeing and dyed wool fabric had superior scrub fastness.

Abstract:Five potentially environmentally friendly and light-stable hemicyanine dyes were designed based on integrated consideration of photo, environmental, and computational chemistry as well as textile applications. Two of them were synthesized and applied in dyeing polyacrylonitrile (PAN), cotton, and nylon fabrics, and demonstrated the desired properties speculated by the programs. The computer-assisted analytical processes includes estimation of the maximum absorption and emission wavelengths, aquatic environmental toxicity, affinity to fibers, and photo-stability. This procedure could effectively narrow down discovery of new potential dye structures, greatly reduce and prevent complex and expensive preparation processes, and significantly improve the development efficiency of novel environmentally friendly dyes.

Abstract:Optical clearing with high-refractive-index (high-n) reagents is essential for 3D tissue imaging. However, the current liquid-based clearing condition and dye environment suffer from solvent evaporation and photobleaching, causing difficulties in maintaining the tissue optical and fluorescent features. Here, using the Gladstone-Dale equation [(n-1)/density=constant] as a design concept, we develop a solid (solvent-free) high-n acrylamide-based copolymer to embed mouse and human tissues for clearing and imaging. In the solid state, the fluorescent dye-labeled tissue matrices are filled and packed with the high-n copolymer, minimizing scattering in in-depth imaging and dye fading. This transparent, liquid-free condition provides a friendly tissue and cellular environment to facilitate high/super-resolution 3D imaging, preservation, transfer, and sharing among laboratories to investigate the morphologies of interest in experimental and clinical conditions.

Abstract:In vivo fluorescent imaging represents a potent means for real-time probe quantification, facilitating insights into disease pathophysiology and therapeutic responses. Nonetheless, accurate signal quantification remains challenging due to inherent factors like light scattering and tissue absorption. Existing imaging systems, though sophisticated, often entail high costs and are typically restricted to well-funded laboratory settings. This study introduces BrightMice, an innovative in vivo fluorescent imaging system that harnesses 3D printing and consumer-grade digital cameras. Tailored for various fluorophores such as EYFP and E2-crimson, the system showcases both adaptability and effectiveness in detecting in vivo fluorescent signals in several reporter mouse strains. Comparative analyses against commercial instruments confirm BrightMice's sensitivity and underscore its potential to democratize in vivo fluorescence imaging. By providing a cost-effective and accessible solution, BrightMice stands to benefit diverse research environments.

Abstract:The thermochromic and mechanochromic fluorescence of diphenyldibenzofulvenes is investigated. Emission is boosted and blue-shifted upon crystallization. Yellow emissive crystals of the material transform to green fluorescent crystals upon heating before melting. Reversible switching of the emission color and efficiency are achieved by repeated amorphization and crystallization of dye molecules by a pure thermal process or grinding-heating cycles.

Abstract:Biological tissue transparency combined with light-sheet fluorescence microscopy is a useful method for studying the neural structure of biological tissues. The development of light-sheet fluorescence microscopy also promotes progress in biological tissue clearing methods. The current clarifying methods mostly use liquid reagent to denature protein or remove lipids first, to eliminate or reduce the scattering index or refractive index of the biological tissue. However, denaturing protein and removing lipids require complex procedures or an extended time period. Therefore, here we have developed acrylate resin with a high refractive index, which causes clearing of biological tissue directly after polymerization. This method can improve endogenous fluorescence retention by adjusting the pH value of the resin monomer.

Abstract:In order to observe and quantify long-range neuronal connections in intact mouse brain by light microscopy, it is first necessary to clear the brain, thus suppressing refractive-index variations. Here we describe a method that clears the brain and preserves the signal from proteinaceous fluorophores using a pH-adjusted non-aqueous index-matching medium. Successful clearing is enabled through the use of either 1-propanol or tert-butanol during dehydration whilst maintaining a basic pH. We show that high-resolution fluorescence imaging of entire, structurally intact juvenile and adult mouse brains is possible at subcellular resolution, even following many months in clearing solution. We also show that axonal long-range projections that are EGFP-labelled by modified Rabies virus can be imaged throughout the brain using a purpose-built light-sheet fluorescence microscope. To demonstrate the viability of the technique, we determined a detailed map of the monosynaptic projections onto a target cell population in the lateral entorhinal cortex. This example demonstrates that our method permits the quantification of whole-brain connectivity patterns at the subcellular level in the uncut brain.

Abstract:Inspired by the remarkable adhesive property of marine mussels, a novel method was developed for wool coloration at room temperature. In this method, dopamine was utilized as a biomimetic precursor for wool coloration due to its structure similar to 3,4-dihydroxyphenylalanine, an important component responsible for mussel adhesive. By using dopamine, wool coloration could be easily achieved at room temperature through a simple dip-coating process. Moreover, the obtained color appearance of wool fabrics could be tuned in a controllable way by the addition of other chemical components to the coloration bath. The dyed wool fabrics showed good color fastness properties.
