Abstract:Sulfatase participates in a variety of physiological processes in organisms including hormone regulation, cell signaling, and bacterial pathogenesis. Current sulfatase fluorescent probes can be used to track sulfate esterase overexpression in cancer cells for diagnostic purposes and to understand the pathological activity of sulfate esterase. However, some sulfatase fluorescent probes based on the hydrolysis of the sulfate bond were easily disturbed by the catalytic activity of sulfatase. Herein, we developed the fluorescent probe BQM-NH2 for sulfatase detection, which was based on the quinoline-malononitrile. The probe BQM-NH2 showed a fast response to sulfatase within 1 min and satisfactory sensitivity with a calculated LOD of 1.73 U/L. Importantly, it was successfully used to monitor endogenous sulfate in tumor cells, indicating BQM-NH2 has the potential to monitor sulfatase under physiological and pathological conditions.

Abstract:Sulfite (SO32-) is considered as a monitor of a wide range of physiological processes. However, cells and tissues are adversely affected when the body ingests high level of sulfite. Here, we designed and synthesized a "turn on" fluorescent probe ImiSft-1 with 2-cyano-N-methylacetamide as the specific recognition site of SO32-. This probe predominantly achieved high response intensity to SO32- and desirable properties such as large Stokes shift (similar to 180 nm), fast response time (within 15 s), and high sensitivity (LOD = 0.12 mu M). Importantly, the probe was highly selective for sulfite from other bio-species including biological thiols. Other functional properties included broad pH adaptability (5.0-10.0) and low cytotoxicity. Given these advantages and the fluorescence imaging in living MCF-7 cells, it was demonstrated that probe ImiSft-1 could monitor the changes of sulfite concentration in living cells.

Abstract:We have developed a novel colorimetric and ratiometric fluorescence probe for the selective and sensitive monitoring of hydrogen sulfide based on a dicyanoisophorone platform. An excellent linear relationship of fluorescence intensity ratio (I-637/I-558) (R-2 = 0.9867) versus hydrogen sulfide concentration in the range of 1-12 mu M was obtained. This probe exhibited a remarkable fluorescence response to hydrogen sulfide over other physiological thiols or biological species, which fluoresces in the red region with a large Stokes shift (172 nm). This probe was successfully utilized to monitor H2S under in vitro physiological conditions and for imaging H2S in living cells and living zebrafish in vivo.

Abstract:Aggregation-induced emission luminogens (AIEgens) enable highly sensitive and in situ visualization of sulfatase to benefit the early diagnosis of breast cancer (BC), but current sulfatase AIEgens always emit visible light (<650 nm). Herein, a near-infrared (NIR) AIEgen QMT-SFA is developed for sulfatase imaging in vivo . Hydrophilic QMT-SFA is cleaved by sulfatase to yield hydrophobic QMT-OH, which subsequently aggregates into nanoparticles to turn the AIE fluorescence "on", enabling sensitive sulfatase imaging in 4T1 cells and mouse models.

Abstract:Sulfenylation is one of the reversible post-translational modifications, playing significant roles in cellular redox homeostasis and signaling systems. Herein, small fluorescent probe (CPD and CPDDM) based live-cell labelling technology for the visualization of protein sulfenylation responses in living cells has been developed. This approach enables the detection of protein sulfenylation without the need for cell lysis, fixation or purification, and permits the noninvasive study of protein sulfenylation in live cells through the direct fluorescent readout. This technology also can realize dynamic tracking of protein sulfenylation in situ with minimal perturbation to sulfenylated proteins and less interference with cellular function. Information on the global distribution and dynamic changes of endogenous protein sulfenylation has been obtained.

Abstract:Sulfites are widely used as preservative and antioxidant additives in food and drug. A non-invasive method for in vivo imaging of sulfite represents a powerful tool for estimating its potential effects in living organisms. Herein, we report the design, development, and application of sulfite bioluminescent probes (SBPs) for the analyte-specific detection of sulfite through sulfite-mediated intramolecular cleavage. Among them, SBP-1 exhibited the excellent responsivity, high selectivity and sensitivity. By taking advantage of this probe, the first in vivo imaging of sulfate was successfully carried out, not only to trace exogenous sulfite level in living animal, but also to investigate endogenous sulfite in a sulfite oxidase deficiency model.

Abstract:The rational design of effective tools capable of monitoring hydrogen sulfide (H2S) is of great importance to fully understand its physiological and pathological functions. Herein, a self-assembled fluorescent nanoprobe with both aggregation-induced emission (AIE) and excited-state intramolecular proton transfer (ESIPT) characteristics for H2S detection has been successfully developed via a modified nanoprecipitation method. The nanoprobe features high water dispersibility, a large Stokes shift (ca. 100 nm), good biocompatibility as well as high selectivity towards H2S over other putative interferents. Live cell imaging experiments demonstrate that the nanoprobe, with good cell-membrane permeability, can facilitate the visualization of exogenous and endogenous H2S levels. Moreover, the nanoprobe has also been found capable of detecting H2S in zebrafish. This nanoprobe with AIE and ESIPT characteristics can provide a novel method for the development of fluorescent probes for monitoring biological processes.

Abstract:Stimulated Raman Scattering (SRS) coupled with alkyne tags has been an emerging imaging technique to visualize small-molecule species with high sensitivity and specificity. Here we describe the development of a ratiometric Raman probe for visualizing hydrogen sulfide (H2S) species in living cells as the first alkyne-based sensor for SRS microscopy. This probe uses an azide unit as a selective reactive site, and it targets mitochondria with high specificity. The SRS ratiometric images show a strong response to H2S level changes in living cells.

Abstract:Sulfur dioxide (SO2) is an important marker in the human body, and maintaining its balance is very significant for human health. The high concentration of SO2 will cause harm to human health because of the transient nature of SO2. To measure the level of SO2, it is urgent to find a suitable detection method for the real-time monitoring of sulfite. Herein, we designed and exploited a Near-Infrared (NIR) turn-on fluorescent probe L derived from rhodamine for the detection of SO32- in living cells and zebrafish. The addition of sulfite would attack the 4-position of the carbonyl group of levulinic acid, resulting in the cleavage of the ester bond and the formation of the fluorophore. The intramolecular charge transfer (ICT) was restored thus the monitoring system emitted the corresponding long NIR fluorescence at 646 nm. Probe L had high selectivity, a short response period within 10 min, and the detection limit of 0.6 mu M. Besides, Probe L had a long excitation wavelength (>580 nm), which was in favor of more deeply biological imaging properties. Moreover, Probe L had low cell cytotoxicity through MTT assay, thus it was confirmed with good biocompatibility. In addition, probe L could successfully real-time monitor exogenous and endogenous sulfite in living cells, and be applied in zebrafish. The confocal results suggested that probe L had strong potential application prospect of outstanding capabilities for detecting sulfite in clinical diagnostics.

Abstract:A near infrared fluorescent probe YSP for sulfite was synthesized, in which a julolidine fused with a pyran-2-one was employed as the fluorophore and the vinyl activated by an indole salt as the receptor. The introduction of julolidine and indole salt strengthens the electron push-pull effect of the probe and allows it to absorb (597 nm) and emit (681 nm) in red wavelength region. The addition of sulfite to the C˭C bond led to prominent blue-shifts in both absorption (171 nm) and emission (165 nm) spectra, which made it possible for colorimetric and ratiometric fluorescent detection of sulfite. NMR titration results illustrated that the determination of sulfite is a two-step process: nucleophilic addition of sulfite to the unsaturated carbon of C˭N in indole ring followed by intramolecular rearrangement through a four-membered ring to form adduct-B with shorter absorption wavelength. In addition, the cationic feature of YSP enables the probe to be specifically localized in mitochondria, and it could ratiometric bioimaging sulfite in living HepG-2 and L929 cells.

Abstract:A new fluorescent probe 7-(diethylamino)-3-((1e,3e)-5-oxo-5-phenylpenta-1,3-dien-1-yl)-2H-chromen-2-one (SPH), based on Michael addition mechanism, was designed and synthesized for selective detection of sulfite. The probe was constructed by incorporating an α,β-unsaturated ketone conjugated with a C˭C bond into the coumarin fluorophore as a specifical reaction site for sulfite utilizing its nucleophilic property. The extra conjugated C˭C bond induced obvious red-shifts in both absorption and emission maxima, and remarkably promoted the nucleophilic addition rate. Once treated with sulfite, the solution's color changed from orange to yellow companied with a strong blue-green fluorescence. The ratio of fluorescence intensity at 488nm and 630nm (I488/I630) was linear with sulfite concentration over the range of 0-80μM with a detection limit of 0.23μM. High selectivity and good competition of the probe toward sulfite over other anions and thiols enable us to monitor sulfite levels in realistic samples as well as in living HeLa cells.

Abstract:A new ratiometric fluorescence probe E1 based on an excited-state intramolecular proton transfer (ESIPT) mechanism for detection of hydrogen sulfide (H2S) is reported. E1 responds to H2S quickly and showed a 30-fold fluorescence enhancement in 2 minutes. Moreover, E1 can detect H2S quantitatively with a detection limit as low as 0.12 mu M in aqueous solution. Its potential for biological applications was confirmed by employing it for fluorescence imaging of H2S in living cells.

Abstract:In this work, a ratiometric and chromogenic fluorescent probe 1 was synthesized for the detection of SO32-. The probe 1 at PBS (10 mM, pH = 7.4) presented a marked emission band at 661 nm. Upon addition of SO32- ions, a highly emissive adduct with a marked fluorescence at 471 nm were obtained through a Michael addition. The probe 1 displayed a noticeable fluorescence ratiometric response with a large shift (190 nm) in emission wavelength. The probe can quantitatively detect SO32- with high specificity, fast response (within 130 s) as well as low detection limit (13 nM), and a large Stokes shift (139 nm). Fluorescence imaging of HeLa cells indicated that 1 could be used for monitoring the intrinsically generated intracellular SO32- in living cells by ratiometric fluorescence imaging. Furthermore, 1 could be application in real water and sugar samples with high sensitivity and good recoveries.

Abstract:Ratiometric fluorescent probes hold great promise for in vivo imaging; however, stimuli-activatable ratiometric probes with fluorescence emissions in near-infrared (NIR) region are still very few. Herein, we report a hydrogen sulfide (H2S)-activatable ratiometric NIR fluorescent probe (1-SPN) by integrating a H2S-responsive NIR fluorescent probe 1 into a H2S-inert poly[2,6-(4,4-bis-(2-ethylhexyl)-4H-cyclopenta[2,1-b;3,4-b’]dithiophene)-alt-4,7(2,1,3-benzothiadiazole)] (PCPDTBT)-based NIR semiconducting polymer nanoparticle (SPN). 1-SPN shows “always on” PCPDTBT fluorescence at 830 nm and weak probe 1 fluorescence at 725 nm under excitation at 680 nm. The ratio of NIR fluorescence intensities between 725 and 830 is small. Upon interaction with H2S, the fluorescence at 725 nm is rapidly switched on, resulting in a large enhancement of I725/I830, which is allowed for sensitive visualization and quantification of H2S concentrations in living cells. Taking advantage of enhanced tissue penetration depth of NIR fluorescence, 1-SPN is also applied for real-time ratiometric fluorescence imaging of hepatic and tumor H2S in living mice. This study demonstrates that activatable ratiometric NIR fluorescent probes hold great potential for in vivo imaging.

Abstract:In daily life, sulfite and bisulfite have been extensively used as antimicrobial agent and antioxidant for food products, beverages and pharmaceutical products, but, SO3 2−/HSO3 − are fantastically harmful to human health, so it is meaningful to develop an effective probe for these two anions. In this work, a new fluorescent material probe 1 was designed and constructed, which was able to sense sulfite and bisulfite through fluorescence “turn-on” manner. Compared with correlative fourteen other anions used, probe 1 showed great selectivity toward sulfite or bisulfite. Furthermore, the probe can detect SO3 2−/HSO3 − with fast response (within 4min) as well as low detection limit (8.8nM) compared with the previous work. We characterized the adduct through NMR and X-ray diffraction analysis and further confirmed the detection mechanism of probe 1. Finally, fluorescence imaging of MCF-7 cells indicated that 1 could be used for monitoring SO2 derivatives in living cells with low cytotoxicity.

Abstract:Aqueous sulphides, including hydrogen sulphide, have important roles in biological signalling and metabolic processes. Here we develop a selective sulphide-trapping strategy involving sulphide addition to an aldehyde; the resulting hemithioacetal undergoes a Michael addition with an adjacent unsaturated acrylate ester to form a thioacetal at neutral pH in aqueous solution. Employing this new strategy, two sulphide-selective fluorescent probes, SFP-1 and SFP-2, were synthesized on the basis of two different fluorophore templates. These probes exhibit an excellent fluorescence increase and an emission maximum shift (SFP-1) in response to Na 2 S and H 2 S in a high thiol background as found under physiological conditions. We show the utility of the probes for the selective detection of sulphides, and the capacity of our probes to monitor enzymatic H 2 S biogenesis and image free sulphide in living cells.
