Characterisation of xylan-type polysaccharides and associated cell wall components by FT-IR and FT-Raman spectroscopies

Abstract:Xylan models with (1⃗4), (1⃗3), (1⃗2) α or β glycosidic links were studied by FT-IR and FT-Raman spectroscopy. The results indicate the unique spectral properties of samples with the (1⃗3) glycosidic link or O3 branching point. The possibility of identifying other plant cell wall constituents in complex xylan samples, e.g. pectic polysaccharides, protein, lignin, and phenolics, was also investigated. The diagnostic bands of phenolics (lignin, ferulic acid) and proteins in the 1700–1500cm−1 region may overlap if both are present, but their band intensities vary greatly between FT-IR and FT-Raman spectra. In certain cases the use of these two complementary spectroscopic techniques can give valuable information about the complex samples, but lignin containing samples suffered from strong fluorescence in the NIR-Raman. The characteristic spectral features can be used to identify these substances in crude xylan samples without the need of physical separation.
