Highly Efficient Synthesis of Maleimide-braced Peptide Macrocycles and Their Potential Anti-SARS-CoV-2 Mechanisms

Abstract:Peptidomimetic macrocycles often exhibit superior biological properties and druggability, leading to their versatile application in the pharmaceutical industry. A highly efficient and practical methodology for the functionalization and macrocyclization of Trp and Trp-containing peptides was established via Pd(II)-catalyzed C–H alkenylation at the C4 position. This method provides direct access to a maleimide-braced peptide macrocycle that is challenging to synthesize by conventional strategies. In particular, these unique peptide macrocycles revealed low micro- to sub-micromolar EC50 values with promising anti-SARS-CoV-2 activity in Vero E6 cells. To identify their anti-SARS-CoV-2 target, a computational drug target prediction workflow was designed. Based on surface plasmon resonance assays and site mutagenesis experiments, the nucleocapsid protein, one of the crucial structural components of SARS-CoV-2, was determined to be the binding target of our peptide macrocycles. Collectively, we developed a group of peptide macrocycles with highly efficient synthesis methods that could offer new modalities for anti-SARS-CoV-2 agents.
